니트릴히드라타제 활성을 갖는 폴리헵티드를 코팅하는 DNA단편, 이것을 함유하는 형질 전환체 및 이 전환체를 사용하여 아미드류를 제조하는 방법
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 로도코카스(Rhodococcus)로도크로우스(rhodochrous) J-1에 의해서 제조되는 2종류의 니트릴히드라타제의 α 및β서브유니트의 N-말단 아미노산 배열, 제2도는 DNA스포브로서 사용한 로도코카스 SP, N-774의 니트릴히드라타제유전자의 DNA배열, 제3도는 제조합 플라스미드, PNHJ10및 PNHJ20L의 제한 효소지도.

Claims (9)

  1. 하기와 같은-서브 유니트와-서브 유니트의 아미노산 배열을 갖고 니트릴히드라타제활성을 갖는 폴리펩티드를 코딩하는단편-서브 유니트.

  2. 하기와 같은-서브 유니트와-서브 유니트의 아미노산배열을 갖고 니트릴히드라타제활성을 갖는 폴리펩티드를 코딩하는-단편-서브 유니트

  3. 제1항에 있어서,-서브 유니트와-서브 유니트의 다음DNA배열을 갖는- 단편-서브 유니트 DNA배열 :

  4. 제2항에 있어서,-서브 유니트와-서브 유니트의 다음 DNA배열을 갖는-단편-서브 유니트의 DNA배열

  5. 제1∼4항 기재의또는를 백터중에 포함하는 재조합체 DNA.
  6. 제5항 기재의 재조합체 DNA로 형질 전환된 형질전환체.
  7. 제6항 기재의 형질변환체를 배양하는 공정과, 그 배양물로부터 니트릴히드라타제를 회수하는 공정을 포함하는 니트릴히드라타제를 제조하는 방법.
  8. 제6항 기재의 형질전환체의 배양물로 부터 얻어지는 니트릴히드라타제를 사용하여 니트릴류를 수화시키는 공정을 포함하는 아미드류를 제조하는 방법.
  9. 제6항 기재의 형질전환체를 배양하는 공정과 얻어진 배양물로 분리균제, 균제처리물 및 그들의 공정화물을 사용하여 니트릴류를 수화하여 아미드류화는 공정을 포함하는 아미드류를 제조하는 방법.
