본 발명은 노화가 지연되는 쌀밥을 짓는 방법에 관한 것이다. 특히, 본 발명은 최대 7일 동안 저장한 후에도 개선된 질감을 갖는 쌀밥을 짓는 방법에 관한 것이다.

본 발명은 노화가 지연되는 쌀밥을 짓는 방법에 관한 것이다. 특히, 본 발명은 저장 후에도 개선된 질감을 갖는 쌀밥을 짓는 방법에 관한 것이다.
쌀밥은 식품 노점 및 편의점에서 판매되는 다수의 대중적인 즉석 식품, 예를 들어 일본식 식품, 예를 들어, 마끼(maki) 및 니기리(nigiri)와 같은 스시(sushi), "오니기리(Onigiri)" 또는 "오무스미(Omusubi)"와 같은 주먹밥에서 사용된다. 고객의 관심을 끌기 위해서는 이러한 쌀밥 식품은 품질이 일정해야 하고, 소비 시까지 이 품질을 유지해야 한다.
그러나, 실온에 두거나 냉장시켰을 때, 쌀밥은 시간이 경과됨에 따라 굳어진다. 쌀밥의 이러한 열화를 방지하기 위하여, 효소, 예를 들어 알파-아밀라제, 베타-아밀라제, 글루코아밀라제, 글루코즈 옥시다제, 펙티나제, 트레할로즈 트랜스글루타미나제 및 프로타아제를 첨가하는 것과 같은 방법들을 이용하는 시도를 하여 왔다(EP 1108366, EP 2340732).
빵에 말토게닉 알파-아밀라제을 사용하는 것에 대한 다수의 보고가 있었고, 말토게닉 알파-아밀라제가 빵 및 발효시켜 구운 다른 식품의 저장 수명을 개선하는 효과를 갖는다는 것이 알려져 있다(WO 1991/04669).
그러나, 쌀밥은 빵과는 완전히 다른 식품이고, 따라서, 쌀밥에 말토게닉 알파-아밀라아제를 첨가하여 얻어지는 효과는 빵에 동일한 것을 첨가하여 얻어지는 효과와는 크게 다를 것으로 예상된다.
본 발명의 목적은 최대 수 동안 실온에 저장하거나 냉장시킨 후에 개선된 질감을 갖는 쌀밥 식품을 제공하는 것이다.
상기 기술된 목적을 해결하기 위한 집중적인 연구의 결과, 본 발명자들은 예를 들어, 말토게닉 알파-아밀라아제(maltogenic alpha-amylase) 또는 분지 효소(branching enzyme)를 첨가함으로써, 저장에 의해 야기되는 질감에 있어서 쌀밥 식품의 품질 열화를 종래의 방법에 의해 방지할 수 있는 것보다 훨씬 큰 정도로 방지할 수 있다는 것을 발견하였다.
따라서, 본 발명은 (a) 말토게닉 알파-아밀라아제, 분지 효소 및 이들의 혼합물로 이루어진 그룹으로부터 선택된 효소의 수용액과 쌀을 접촉시키는 단계; 및 (b) 쌀밥을 짓는 단계를 포함하는, 노화가 지연되는 쌀밥을 짓는 방법을 제공한다. 또한, 본 발명의 방법에 의해 만들어진 쌀밥 식품을 제공한다.
본 발명의 방법에서 이용된 쌀은 쌀밥을 짓는데 적절한 임의의 쌀 종류 및 임의의 변종일 수 있다. 오리자 종(Oryza sp.) 또는 지자니아 종(Zizania sp.)의 쌀이 선호되고, 바람직하게는 오리자 사티바(Oryza sativa)(아시아 쌀) 또는 오리자 글라베리마(O. glaberrima)(아프리카 쌀), 지자니아 아쿠아티카( Zizania aquatica)(야생 쌀), 지자니아 팔루스트리스(Z. palustris)(북쪽(Northern) 야생 쌀), 및 지자니아 라티폴리아(Z. latifolia)(만주 야생 쌀) 중 어느 하나이다. 오리자 사티바 아종(subspecies), 자포니카 및 아종, 인디카의 쌀이 가장 선호된다. 가장 바람직한 쌀은 도정미(polished rice)이다. 그러나, 이 방법은 보리, 밀 및 호밀 중 어느 하나를 포함하는 임의의 겉껍질을 벗긴 곡물의 식품에 동일하게 적용될 수 있지만, 이에 한정되는 것은 아니다.
용어 "노화가 지연되는(retarded staling)"은 최대 12 시간, 최대 24 시간, 최대 36 시간 또는 최대 48 시간(2일) 동안 또는 보다 오랫동안 저장한 이후에 경도 및/또는 점착성이 검출가능한 정도로 감소하는 것을 가리킨다. 경도의 감소는 재료 및 방법 섹션에서 기술된 바와 같은 질감 분석기(Texure Analyzer)를 사용하여 측정될 수 있다. 바람직하게는 1일 및/또는 2일 후 경도가 대조군 대비 5%, 10%, 15%, 20% 또는 심지어 25% 이상 감소한다.
용어 "말토게닉 알파-아밀라제"는 EC 3.2.1.133 내의 아밀라제를 포함한다. 말토게닉 알파-아밀라제는 쇄(chain)의 비-환원 말단으로부터 연속적인 α-말토즈 잔기를 제거하기 위하여 폴리사카라이드에서 (1→4)-α-D-글루코시딕 연결의 가수분해를 촉진한다.
말토게닉 알파-아밀라제는 바실러스 스테아로써모필러스(Bacillus stearothermophilus), 예를 들어, EP 120693에 기술된 균주 NCIB 11837로부터 얻을 수 있거나, 또는 그것은 WO 1999/043794, WO 2006/032281 또는 WO 2008/148845에 기술된 변이체일 수 있다.
말토게닉 알파-아밀라제는 알리사이클로바실러스 폴리에(Alicyclobacillus pohliae), 예를 들어 임페리오 등(Imperio et al.)의 문헌[(2008) int J Syst Evol Microbiol 58, 221-225]에 기술된 균주 NCIMB 14276으로부터 얻을 수 있다. 말토게닉 알파-아밀라제는 US 8,426,182에 개시된 효소일 수 있다.
말토게닉 알파-아밀라제 제제는 바실러스 스테아로써모필러스의 균주로부터, 또는 적절한 박테리아 생성 유기체의 유전자 조작된 균주로부터 얻을 수 있다.
바람직하게는 말토게닉 알파-아밀라제는 서열번호 2의 아미노산 서열과 75% 이상, 80% 이상, 85% 이상, 90% 이상, 95% 이상, 96% 이상, 97% 이상, 98% 이상, 99% 이상, 또는 심지어 100% 동일한 아미노산 서열을 갖는다.
말토게닉 알파-아밀라제는 전형적으로 쌀의 건조 고형물 kg당 200 내지 50000 MANU, 특히 500 내지 10000 MANU/kg의 용량으로 사용된다.
용어 "분지 효소(branching enzyme)"는 E.C. 2.4.1.18 내의 효소를 포함한다. 분지 효소는 (1→4)-α-D-글루칸 쇄의 단편을 유사한 글루칸 쇄 내의 1차 하이드록시 그룹에 전달한다.
분지 효소는 WO 2000/58445에서 서열번호 2로서 개시된 분지 효소와 같은 로도테르무스 오바멘시스(Rhodothermus obamensis), 또는 로도테르무스 마리누스(R. marinus)로부터 얻을 수 있다. 바람직하게는, 분지 효소는 서열번호 3(WO 2000/58445에서의 서열번호 2와 동일)에 적어도 75% 이상, 80% 이상, 85% 이상, 90% 이상, 95% 이상, 96% 이상, 97% 이상, 98% 이상, 99% 이상, 또는 심지어 100% 동일한 아미노산 서열을 갖는다.
다른 적절한 분지 효소는 JP 2000316581에서의 서열번호 1에 개시된 분지 효소와 같은 아퀴펙스 아에오리쿠스(Aquifex aeolicus)로부터 또는 EP 418945에 개시된 분지 효소와 같은 바실러스 스테아로써모필러스(Bacillus stearothermophilus)로부터 얻을 수 있다.
쌀은 조리 전에 침지될 수 있다. 침지 단계의 기간은 10분 내지 24 시간, 바람직하게는 1 내지 12 시간일 수 있다. 침지 단계는 바람직하게는 5 내지 30℃의 온도에서, 예를 들어 약 20℃의 온도에서 수행될 수 있다.
본 발명에 따른 쌀밥 식품을 만드는 방법에서, 임의의 종래의 조리 과정, 예를 들어 쌀을 물에 침지하고, 이를 뚜껑이 있는 솥, 밥솥, 또는 쌀 보일러(rice boiler)에서 끓이는 과정이 사용될 수 있다.

추가 효소들

본 발명에서 사용하기에 적절한 글루코아밀라제는 아스퍼질러스 니거(A. niger) G1 또는 G2 글루코아밀라제(Boel et al.(1984), EMBO J. 3 (5), p. 1097-1102), WO 1984/02921에 개시된 아스퍼질러스 아와모리(A. awamori) 글루코아밀라제, 또는 아스퍼질러스 오리제(A. oryzae) 글루코아밀라제(Agric. 쌀밥의 사용
본 발명에 따라 지어진 쌀밥은 임의의 종류의 쌀 기반 즉석 식품, 예를 들어, 마끼 및 니기리와 같은 스시, 오니기리 또는 오무스비와 같은 주먹 밥, 벤토(bento) 및 에키 벤(eki ben) 등과 같은 도시락을 생산하는 데에 사용될 수 있다. 추가로, 본 발명은 이들 식품을 냉동, 무균 포장, 레토르트 파우치-포장(retort pouch-packing), 건조 또는 통조림화하여 생산된 쌀밥 식품을 그의 범위 내에 포함한다.

말토게닉
알파-아밀라제 활성의 측정
1 MANU(말토게닉 아밀라제 노보 유니트, Maltogenic Amylase Novo Unit)는 37℃에서 pH 5.0의, 0.1 M 시트레이트 버퍼 ml 당 10 mg의 말토트리오즈 기질(≥95% 순도, 예를 들어 Sigma M 8378) 농도에서 30분 동안 분당 1 μ몰의 말토즈를 방출하는 데에 요구되는 효소의 양으로 정의될 수 있다.
분지 효소 활성의 측정
분지 효소 활성은 EP 418 945에 기술된 바와 같이 측정될 수 있다.
질감 분석
쌀 한 알의 질감 분석은 쌀알을 90% 압축한 상태에서 TA-XT2i(SMS)를 사용하여 수행하였고, 프로브(Probe): 10 mm 직경 실린더(delrin). 시험 이전 속도: 1 mm/sec, 시험 속도: 0.2 mm/sec, 시험 후 속도:0.2 mm/sec 이다.
효소들
서열번호 2에 나타난 아미노산 서열을 갖는, 바실러스 스테아로써모필러스로부터 얻어진 말토게닉 알파-아밀라제를 포함하는 효소 제제. 효소 제제는 10000 MANU/g의 활성을 갖는다.
서열번호 3(WO 2000/58445에 있는 서열번호 2와 동일)에 나타난 아미노산 서열을 갖는 로도테르무스 오바멘시스(Rhodothermus obamensis)로부터 얻어진 분지 효소를 포함하는 효소 제제.
실시예

시험 과정:
160 g의 생쌀(오리자 사티바 아종, 자포니카)을 수돗물에서 세척하고, 190 ml 물 또는 쌀에 대해 0.1% 용량(w/w)의 효소 제제를 갖는 190 ml 물에 실온에서 밤새 침지하였다. 침지된 쌀을 밥 솥에 옮겨 조리했다. 조리 후, 쌀밥을 4 또는 20℃에서 저장했다. 0 시간에 그리고 1일 및 2일 저장 후, 한 알의 손상되지 않은 쌀밥의 질감을 평가하였다. 질감 분석은 각 처리 동안 10번의 반복으로 실온에서 수행하였다. 결과가 표 1a, 표 1b 및 표 2에 나타나 있다.

Claims (15)
- (a) 말토게닉 알파-아밀라제, 분지 효소 및 이들의 혼합물로 이루어진 그룹으로부터 선택된 효소를 포함하는 효소 수용액과 쌀을 접촉시키는 단계; 및 (b) 밥을 짓는 단계를 포함하는, 노화가 지연되는 쌀밥 식품을 만드는 방법.
- 제1항에 있어서, 쌀을 효소 수용액과 접촉시키는 것이 밥을 짓는 동안 수행되는, 방법.
- 제1항 또는 제2항에 있어서, 단계 (a)가 밥을 짓기 전에 쌀을 효소 수용액에 침지시키는 것을 포함하는, 방법.
- 제1항 내지 제3항 중 어느 한 항에 있어서, 상기 침지 수가 밥을 짓기 위해 사용되는, 방법.
- 제1항 내지 제4항 중 어느 한 항에 있어서, 상기 쌀을 효소 수용액에 침지시키는 것이 10분 내지 24 시간, 바람직하게는 1 내지 12 시간 사이에 수행되는, 방법.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 상기 쌀을 효소 수용액에 침지시키는 것이 5 내지 30℃, 예를 들어 약 20℃의 온도에서 수행되는, 방법.
- 제1항 내지 제6항 중 어느 한 항에 있어서, 상기 말토게닉 알파-아밀라제는 서열번호 2의 아미노산 서열과 75% 이상, 80% 이상, 85% 이상, 90% 이상, 95% 이상, 96% 이상, 97% 이상, 98% 이상, 99% 이상, 또는 심지어 100% 동일한 아미노산 서열을 갖는, 방법.
- 제1항 내지 제7항 중 어느 한 항에 있어서, 상기 분지 효소는 서열번호 3의 아미노산 서열과 75% 이상, 80% 이상, 85% 이상, 90% 이상, 95% 이상, 96% 이상, 97% 이상, 98% 이상, 99% 이상, 또는 심지어 100% 동일한 아미노산 서열을 갖는, 방법.
- 제1항 내지 제8항 중 어느 한 항에 있어서, 하나 이상의 다른 추가의 효소가 수용액 내에 존재하고, 상기 다른 효소(들)는 아밀라제, 글루카나제, 갈락타나제, 만나나제, 아미노펩티다제, 알파-아밀라제, 말토게닉 알파-아밀라제, 베타-아밀라제, 카복시펩티다제, 카탈라제, 키티나제, 쿠티나제, 사이클로덱스트린 글리코실트랜스퍼라제, 데옥시리보뉴클리아제, 에스테라제, 알파-갈락토시다제, 베타-갈락토시다제, 글루코아밀라제, 알파-글루코시다제, 베타-글루코시다제, 헤미셀룰라제, 할로페록시다제, 인버타제, 락카제, 리파제, 포스포리파제, 만노시다제, 옥시다제, 펙티노리틱 효소, 펩티도글루타미나제, 페록시다제, 피타제, 글루코즈 옥시다제, 폴리페놀옥시다제, 프로테아제, 리보뉴클레아제, 트레할로즈 및 트랜스글루타미나제로 이루어진 그룹으로부터 선택되는 방법.
- 제1항 내지 제9항 중 어느 한 항에 있어서, 상기 쌀은 오리자(Oryza) 종, 또는 지자니아(Zizania) 종 중 임의의 것, 바람직하게는 오리자 사티바(Oryza sativa)(아시아 쌀), 또는 오리자 글라베리마(O. glaberrima)(아프리카 쌀), 지자니아 아쿠아티카(Zizania aquatica)(야생 쌀), 지자니아 팔루스트리스(Z. palustris)(북쪽 야생 쌀), 지자니아 라티폴리아(Z. latifolia)(만주 야생 쌀) 중 임의의 것, 가장 바람직하게는 오리자 사티바 아종 자포니카(japonica) 및 아종 인디카(indica) 중 임의의 것인, 방법.
- 제1항 내지 제10항 중 어느 한 항에 있어서, 상기 쌀이 도정미인 방법.
- 제1항 내지 제11항 중 어느 한 항에 있어서, 상기 쌀이 데쳐진(parboiled) 쌀인 방법.
- 제1항 내지 제12항 중 어느 한 항에 있어서, 1일 및/또는 2일 후 경도가 대조군에 비해 5%, 10%, 15%, 20% 또는 심지어 25% 이상 감소된, 방법.
- 제1항 내지 제13항 중 어느 한 항의 방법에 의해 제조된 조리된 쌀밥 식품.
- 제14항에 있어서,
쌀 기반 즉석 식품, 예를 들어, 마끼 또는 니기리와 같은 스시, 오니기리 또는 오무스비와 같은 주먹 밥, 벤토 및 에키 벤 등과 같은 도시락인, 쌀밥 식품.
