Abstract
Description
[1]ヒト化K型CpGオリゴデオキシヌクレオチド及びポリデオキシアデニル酸を含む、オリゴデオキシヌクレオチドであって、ポリデオキシアデニル酸が、ヒト化K型CpGオリゴデオキシヌクレオチドの3’側に配置されている、オリゴデオキシヌクレオチド。
[2]ヒト化K型CpGオリゴデオキシヌクレオチドが、10ヌクレオチド以上の長さであり、且つ式:
で表されるヌクレオチド配列を含む、[1]に記載のオリゴデオキシヌクレオチド。
[3]ヒト化K型CpGオリゴデオキシヌクレオチドが、配列番号1で表されるヌクレオチド配列からなる、[1]又は[2]に記載のオリゴデオキシヌクレオチド。
[4]オリゴデオキシヌクレオチドのリン酸ジエステル結合の一部又は全てがホスホロチオエート結合で置換されている、[1]〜[3]のいずれかに記載のオリゴデオキシヌクレオチド。

本発明は、K型CpGオリゴデオキシヌクレオチド及びポリデオキシアデニル酸(dA)を含む、オリゴデオキシヌクレオチド(以下、本発明のオリゴデオキシヌクレオチドと称する。)を提供するものである。
本発明のオリゴデオキシヌクレオチドにはリン酸ジエステル結合が修飾(例えば、一部又は全てのリン酸ジエステル結合がホスホロチオエート結合により置換)されているものを含む。
本発明のオリゴデオキシヌクレオチドは薬学的に許容可能な塩を含む。
なお、本明細書においてオリゴデオキシヌクレオチドとODNは同義である。また、「ヒト化K型CpGオリゴデオキシヌクレオチド(CpG ODN)」と、「ヒト化K型CpGオリゴデオキシヌクレオチド(CpG ODN)残基」とは末尾の用語「残基」の有無に関わらず同義であり、交換可能に用いられる。更に、ポリデオキシアデニル酸とポリデオキシアデノシン酸(残基)は同義である。用語「残基」は、より大きな分子量の化合物の部分構造を意味するが、本明細書中、「ヒト化K型CpGオリゴデオキシヌクレオチド(CpG ODN)」が、独立した分子を意味するか、より大きな分子量の化合物の部分構造を意味するかは、当業者であれば、文脈から容易に理解可能である。「ポリデオキシアデニル酸」等、本発明のオリゴデオキシヌクレオチドに含まれる他の部分構造に関する用語についても、同様である。

本発明は、上記本発明のオリゴデオキシヌクレオチド及びβ−1,3−グルカンを含有する複合体(以下、本発明の複合体と称する。)を提供するものである。
レンチナンは活性化マクロファージ、キラーT細胞、ナチュラルキラー細胞及び抗体依存性マクロファージ仲介性細胞障害作用(ADMC)活性の増強作用を有する(Hamuro, J., et al.:Immunology, 39, 551-559, 1980、Hamuro, J., et al.:Int. J. Immunopharmacol., 2, 171, 1980、 Herlyn, D., et al.:Gann, 76, 37-42, 1985)。動物実験においては同系腫瘍及び自家腫瘍に対して化学療法剤との併用投与により腫瘍増殖抑制作用ならびに延命効果が認められている。また、レンチナンの単独投与によっても腫瘍増殖抑制作用ならびに延命効果が認められている。臨床試験においては手術不能又は再発胃癌患者に対して、テガフール経口投与との併用により生存期間の延長が認められ(医薬品インタビューフォーム「レンチナン静注用1mg「味の素」」)、本邦で承認されている。レンチナンの単独投与による効果は現在のところ確認されていない。
本発明は、上記本発明のオリゴデオキシヌクレオチド又は上記本発明の複合体を含む、医薬組成物を提供するものである。本発明の医薬組成物は、上記本発明のオリゴデオキシヌクレオチド又は上記本発明の複合体を常套手段に従って製剤化することにより得ることができる。本発明の医薬組成物は、本発明のオリゴデオキシヌクレオチド又は複合体と薬理学的に許容され得る担体を含む。また、本医薬組成物は抗原を更に含んでいても良い。このような医薬組成物は、経口又は非経口投与に適する剤形として提供される。
本発明のオリゴデオキシヌクレオチド及び複合体は、優れた免疫賦活活性を有するので、本発明のオリゴデオキシヌクレオチド、複合体及び医薬組成物は、免疫賦活剤として用いることができる。本発明のオリゴデオキシヌクレオチド、複合体又は医薬組成物を哺乳動物(ヒト等の霊長類、マウス等のげっ歯類等)に投与することにより、該哺乳動物における免疫反応を惹起することができる。特に、本発明の複合体は、D型CpG ODNの活性特性を有し、末梢血単核球を刺激して、I型インターフェロン(Pan-IFN-α、IFN-α2等)及びII型インターフェロン(IFN-γ)の両方を大量に産生させるので、I型インターフェロン産生誘導剤、II型インターフェロン産生誘導剤、I型及びII型インターフェロン産生誘導剤として有用である。I型及びII型インターフェロンの両方の産生を誘導することから、本発明の複合体及びこれを含有する医薬組成物は、I型及びII型インターフェロンのいずれか一方又は両方が有効な疾患の予防又は治療に有用である。I型インターフェロンが有効な疾患としては、ウイルス感染症(例えば、C型肝炎ウイルス(HCV)、ヘルペスウイルス、パピローマウイルス、RSウイルス、インフルエンザウイルス等)、癌等を挙げることが出来る。II型インターフェロンが有効な疾患としては、アレルギー疾患、細胞内寄生性の原虫(リーシュマニア等)や細菌(リステリア、結核菌等)等の感染症等を挙げることが出来る。RSウイルスやインフルエンザウイルスを含む急性ウイルス感染症に対しては、I型インターフェロン及びII型インターフェロン共に、ウイルス排除に関する免疫応答を高めるので、本発明の複合体及びこれを含有する医薬組成物は、急性ウイルス感染症に対する有効性が期待できる。
本明細書において、アジュバントとは免疫応答を促す補助剤のことであり、抗原とともに生体に投与されたとき、その抗原に対する免疫応答を非特異的に増強させる物質を意味する。
動物及び試薬
Tlr9欠損マウス及びDectin-1欠損マウスの作成については、以前に記載した(Koyama, S., et al., Science translational medicine 2, 25ra24 (2010); Saijo, S., et al., Nature immunology 8, 39-46 (2007))。C57BL/6Jマウスは、日本クレアから購入した。
全ての動物実験は、医薬基盤研究所及び大阪大学動物施設の機関ガイドラインに従って実施した。以下のCpG ODNsは、(株)ジーンデザインで合成された(下線はホスホロチオエート結合を示す)。
7.22 mgのK3-dA40を水(3.7mL)に溶解した。SPG(三井製糖) 15 mgを0.25 N NaOH (1 mL)に溶解した。1 mLの330 mM NaH2PO4をDNA溶液に加え、次にSPG溶液をDNA/NaH2PO4溶液に加え、4℃にて一晩維持することにより複合体化を完了した。モル比(MSPG/MDNA)は、0.27に固定した。複合体の形成は、マイクロチップ電気泳動装置(SHIMADZU:MultiNA)によって確認した。
レンチナン(LNT:味の素株式会社、ロット番号:2D8X1)を50mg/mlとなるように、0.25N NaOHに溶解し、室温で一晩放置した。各種CpG ODN(表2,3)は100mMとなるように、注射用水に溶解した。LNT水溶液と各種CpG水溶液(K3-dA20(配列番号9)、K3-dA25(配列番号10)、K3-dA30(配列番号11)、K3-dA35(配列番号12)、K3-dA40(配列番号2))を表4に示す比率で混合し、続いて、添加したLNTと同体積の330 mM NaH2PO4を加え、4℃にて一晩維持することにより複合体化を完了した。複合体の形成は、サイズ排除クロマトグラフィーを用い、CpG ODNの高分子量側へのシフトを、260nmにおける吸収をモニタリングすることによって確認した(System:Agilent 1100series、Column:Asahipak GF7M-HQ (Shodex) 2本連結、Flow rate:0.8mL/min、Buffer: 10mM EDTA PBS, pH7.4、Temperature:40°C)。
PBMCsは、3人の健康な成人男性ボランティア(30〜40 歳)から得た。ヒトPBMCsを用いる全ての試験は、医薬基盤研究所の施設内治験審査委員会により認可された。Ficollを用いてPBMCsを調製後、1 x 107 個/mlの濃度で播いた。PBMCsを、コンプリートRPMI(10% FCS、ペニシリン、及びストレプトマイシンが添加されたRPMI 1640)中で維持した。PBMCsを、K3 (0.24、0.74、2.2、6.6、20 μg/mL) 、K3-dA40、K3-SPG (K3-dA40の複合体)、dA40-K3、SPG-K3(dA40-K3の複合体)、D35、CpG21798、CpG21889、CpG2395、又はM362で24時間刺激した。上清をpan-IFN-α (Mabtech)、IL-6 (R&D)、及びMilliplex (Millipore)用のELISAに付した。
染色する前に、試料をホルムバール−カーボンコートグリップ上に滴下した。陰性染色のため、2% 酢酸ウラニウム(pH 4.0)の液滴をグリップ上に載せ、風乾させた。グリップを電子顕微鏡(Hitachi H-7650)上で40,000倍の拡大率にて観察した。
水溶液中の平均ナノ粒子サイズを、80℃にて、Malvern Instruments Zeta Sizer上で動的光散乱法を用いて測定した。
6週齢C57BL/6Jマウス、Tlr9欠損マウス、及びDectin-1欠損マウスから、マウス脾臓を採取した。脾細胞を懸濁後、ACK溶解緩衝液で赤血球(RBCs)を溶解し、細胞をコンプリートRPMI中で維持した。細胞を1 x 107cells/mLの濃度で播いた。マウス骨髄由来細胞を、ヒトFlt3L (Peprotech) (100 ng/mL)と共に7日間培養することにより、骨髄由来DCsを作成した。細胞を1 x 107cells/mLの濃度で播いた。マウスM-CSF (Peprotech) (20 ng/mL)と共に7日間培養することによりBMDMsを作成した。これらの細胞は、コンプリートRPMI中で維持した。
BMDMsを5 x 107 cells/mLで播き、Alexa 488-K3 (1 μM) 及びAlexa 647-K3-SPG (1 μM)、又はAlexa 488-D35 (1 μM) 及びAlexa 647-K3-SPG (1 μM)により3時間刺激した。細胞をHoechst33258で30分染色することにより核を可視化し、次に細胞を固定し、蛍光顕微鏡を用いて解析した。
6週齢のC57BL/6Jマウス、Tlr9欠損マウス、及びDectin-1欠損マウスの尾の付け根に、
OVA (0.1、1、10、又は100 μg);
OVA (0.37、1.1、3.3、又は10 μg)及びK3(538 pmol);
OVA (0.37、1.1、3.3、又は10 μg)及びK3-dA40 (538 pmol);又は

を、0日目及び10日目に投与した。17日目に採血し、抗原特異的血清抗体力価をELISAにより測定した(図10、15a、16、25、28a、29、43a)。17日目にマウス脾臓を採取し、脾細胞を上述の方法で調製した。細胞を

により24又は48時間再刺激した。上清を、マウスIFN-γのためのELISAに付した(図11、15b、17、26、28b、30、43b)。テトラマーアッセイのため、脾細胞をH-2Kb OVAテトラマー (MBL)、抗CD8α (KT15)、抗TCRβ(H57-597)、抗CD62L (MEL-14)、及び抗CD44 (IM7)抗体、並びに7-AADで染色した。OVAテトラマー+ CD44+CD8α+ TCRβ+細胞数をFACSにより測定した(図12、15c、27、28c、31)。
6週齢のC57BL/6Jマウスの尾の付け根に、
OVA (100 μg);
OVA及びK3 (3.3 μg);
OVA及びK3-dA40 (3.3 μg);又は

を、0日目に投与した。免疫接種後7日目に、ナイーブC57BL/6J脾細胞を、異なる濃度のCFSE(5又は0.5 μM)により、10分間、37℃にて標識した。染色した高濃度の細胞をOVA257 (10 μg/mL)で90分間、37℃にてパルスした。培地で2回洗浄後、標識した細胞を混合し、静脈内投与により免疫したマウスへ移入した。移入から24時間後、脾細胞を採取し、CFSEで標識した細胞の割合をFACSにて測定した。
C57BL/6Jマウスを、
OVA257 (10 μg);
OVA257及びK3 (10 μg);
OVA257及びK3-dA40 (10 μg);又は
OVA257及びK3-SPG (10 μg)
で免疫した。免疫から7日後に、脾細胞を調製し、H-2Kb OVAテトラマー、抗CD8α、抗TCRβ、抗CD62L、及び抗CD44抗体で染色した。OVAテトラマー+ CD44+ CD8α+ TCRβ+細胞数をFACSにより解析した(図14左)。調製した脾細胞をインビトロにてOVA257 (10 μg/mL)及びGolgi Plugにより4時間、再刺激した。細胞を抗IFN-γ、抗CD8α、及び抗CD3e抗体で染色し、IFN-γ+ CD8α+ CD3e+細胞数をFACSにて測定した(図14右)。
HEK293を、Lipofectamine 2000を用いて、空、Dectin-1、又はDectin-2発現プラスミドでトランスフェクトした。トランスフェクション後48時間で、細胞を、FITC-SPG (0.5 μM)、Alexa 488-labeled K3-dA40 (0.5 μM)、又はAlexa 488-labeled K3-SPG (0.5 μM)で、37℃にて、60分間処理した。処理後、細胞を集め、SPG又はCpG ODN陽性細胞をFACSにより解析した。
C57BL/6Jマウス、Tlr9欠損マウス、又はDectin-1欠損マウスからの脾細胞及びFL-DCsをK3-SPG (0.014、0.03、0.04、0.08、0.12、0.25、0.37、0.74、1.1、2.2、3.3、6.7、10、又は20 μg/mL)、ザイモザン (3.7、11.1、33.3、又は100 μg/mL)、カードラン、Zymosan-Depleted、又は SPGで24時間刺激した。他の試験においては、C57BL/6Jマウス又はDectin-1欠損マウスからの脾細胞を、D35 (1 μM)の存在下又は非存在下で、Zymosan-Depleted (100、33.3、又は11.1 μg/mL)又はSPG (100、33.3、又は11.1 μg/mL)で、24時間刺激した。上清をIFN-α(PBL)、IL-6 (R&D)、IL-12 p40 (R&D)、IL-12 p70 (R&D)、及びBioplex (BIO-RAD)のためのELISAに付した。
C57BL/6Jマウスの付け根に、
DQ-OVA (10 μg);
Alexa 488-K3-SPG (10 μg);
Alexa 647-K3-SPG (10 μg);又は

を投与した。iLNsを採取後、クリオスタットを用いて凍結切片を調製した。凍結切片を4%パラホルムアルデヒドで10分間固定し、抗Siglec-1 (MOMA-1)、抗MARCO(ED31)、抗CD3e (145-2C11)、又は抗CD11c (N418)抗体とインキュベートした。画像化の結果をImageJにより解析した。
C57BL/6Jマウス、Tlr9欠損マウス、又はDectin-1欠損マウスの尾の付け根に、DQ-OVA (10 μg)、Alexa 488-K3 (10 μg)、又はAlexa 488-K3-SPG (10 μg)を投与した。iLN採取の30分前に、マウスの尾の付け根にPE-抗MARCO抗体、又はPE-抗Siglec-1抗体を投与した。抗原及びアジュバントの投与後1時間に、iLNsを採取し、二光子顕微鏡(Olympus)による画像解析に付した。ピアソン相関を、Volocity共局在解析を用いて計算した。
C57BL/6Jマウスの尾の付け根に、

を投与した。投与の24時間後に、iLNsを採取した。単一細胞懸濁液を調製するため、iLNsをコラゲナーゼD (1 mg/mL)及びDNase I (0.1 mg/mL)と37℃にて30分間インキュベートした。調製した細胞を抗B220 (RA3-6B2)、抗CD8α (56-6.7)、及び抗CD11c抗体とインキュベートし、異なるDC集団を分離した。OVA及びCpG ODNsの細胞内取込みをFACSにより解析した。
免疫接種の5日又は2日前のいずれかに、6週齢のC57BL/6Jマウスの尾の付け根に、クロドロネートリポソームを投与した。0日目に、OVA及びK3-SPGにより、マウスを尾根部にて免疫した。8日目に血液及び脾臓を採取し、血清抗体力価及びT細胞反応をELISAにより測定した。
C57BL/6Jマウス又はTlr9欠損マウスの尾の付け根に、K3 (10 μg)又はK3-SPG (10 μg)を投与した。投与から24時間後に、iLNsを上述の方法で調製した。細胞を抗CD11c、抗mPDCA-1 (JF05-1C2.4.1)、抗CD8α、及び抗CD40 (3/23) 抗体とインキュベートし、FACSにて解析した。
6週齢のC57BL/6Jマウスに、
スプリットワクチン(0.1 μg);
スプリットワクチン及びK3-SPG (10 μg);又は
WIV(0.2 μg)
を、0日目及び14日目に投与した。免疫接種から2週間後に、血清抗体力価をELISAにより測定し(図39)、マウスに2.3 x 103 pfu (10 LD50)のインフルエンザウイルス A/PR/8/34を鼻腔内へ接種した。接種後20日間に亘り、マウスの体重及び死亡率をモニターした(図40)。
カニクイザルの皮下へ、
インフルエンザスプリットワクチン(5 μg)及びK3 (5 nmol);又は
スプリットワクチン及びK3-SPG (5 nmol)
を、0日目及び14日目に投与した。血液試料を、-2、2、4、6、8及び110週目に採取し、血清抗体力価をELISAにより測定した。
群間の統計学的有意性(P < 0.05)は、Student’s t-test又はMann-Whitney U testを用
いて決定した。
1)K3-SPGの竿状ナノサイズ粒子は、K型及びD型CpG ODNの2つの特性を獲得する。
図1に示すように、CpG ODNとSPGとの間のよりよい複合体化効率には、変性−復元手順を通して、5’末端又は3’末端においてポリdA40のPS骨格の追加的配列を要する(Shimada, N., et al., Bioconjugate chemistry 18, 1280-1286 (2007); Minari, J., et al., Bioconjugate chemistry 22, 9-15 (2011))。ヒト化CpG ODNによる複合体形成の最適化の過程で、本発明者らは、CpG ODNの5’末端及び3’末端の免疫賦活性への影響を調べた。複合体化効率は同等であるにもかかわらず、SPGと複合体化した5’-K3-dA40-3’は、ヒト末梢血単核球(PBMCs)を活性化し、多量のIFN-αを産生させたが、5’-dA40-K3-3’では、そうではなかった(図2、図3)。K3、K3-dA40及びdA40-K3は、ヒトPBMCsを活性化してIL-6のような他のサイトカインを産生させることができるが、IFN-αは産生させることが出来なかった(図2、図3)。これらの結果は、新規のTLR9アゴニストとしては、3’-CpGよりも、5’-CpGの方がより望ましいことを示す。
本発明者らは、マウス免疫化モデルにおける、K3、K3-dA40、及びK3-SPGのアジュバント効果を比較した。野生型マウスを、OVA単独で、或いは各K3由来アジュバントと共にOVAで免疫すると、K3-SPGが、K3により誘導されるものよりも、有意に高い液性免疫反応(図10)、及びより強力なT細胞反応(図11)を誘導した。注目すべきは、K3-SPGによって、より多数のOVA特異的CD8T細胞が誘導されることが、テトラマーアッセイにより明らかになった(図12)。共有結合を何らすることなく共投与したタンパク質抗原に対するきわめて強力なインビボCTL活性も認められた(図13)。このK3-SPGによる強力なCTL誘導は、ペプチドワクチン接種によっても再現され(図14)、用量依存的であった(図15)。K3-SPGの抗原節約活性は極めて強く、同等の抗体及びCD4T細胞反応は100倍量のOVA抗原を用いることにより達成された(図16、17)。これらの結果は、K3-SPGが、K3単独よりも、卓越したアジュバントであることを明確に示す。
Dectin-1は、ザイモザンのようなβグルカンの受容体であることが示されているので(Herre, J., et al., Blood 104, 4038-4045 (2004))、SPG及びK3-SPGの細胞取込、及びそれに引き続くSPG及びK3-SPGによる活性化におけるDectin-1の役割について調べた。フローサイトメトリーを用いて、Dectin-1を発現するHEK293細胞において、ODNの存在に係わらず、インビトロにおけるSPG又はK3-SPGの取り込みが上昇したが、Dectin-2やコントロールベクターでは上昇しないことを見出した(図18、19)。最近、可溶型のβ−グルカンはDectin-1シグナリングを活性化しないことが報告された(Goodridge, H.S., et al., Nature 472, 471-475 (2011))。更に、Dectin-1シグナリングは、サイトカインシグナル抑制因子1(SOCS1)誘導を介して、TLR9媒介サイトカイン産生を抑制する(Eberle, M.E. & Dalpke, A.H., Journal of immunology 188, 5644-5654 (2012))。そこで、SPGのアゴニスト活性を調べた。脾臓細胞をZymosan-Depletedで刺激すると、用量依存的及びDectin-1依存的なTNF-α及び他のサイトカインの産生が認められたが(図20)、SPG刺激では認められなかった(図21)。ザイモザンおよびカードランによるサイトカイン産生は、Dectin-1非依存的であった。Zymosan-Depletedは、CpG ODNにより誘導されたIFN-αを阻害したが、この阻害はDectin-1欠損により軽減された(図22)。対照的に、SPGはCpG ODNにより誘導されたIFN-αを阻害しなかった(図23)。これらの結果は、SPGはDectin-1のリガンドであるがアゴニストではないこと;従ってSPGはTLR9媒介IFN-α産生を妨げないことを示す。
K3-SPGは、CpG ODNとβ−グルカンの複合体であるので、TLR9(Hemmi, H., et al., Nature 408, 740-745 (2000))及びDectin-1(Saijo, S., et al., Nature immunology 8, 39-46 (2007))の役割を、受容体ノックアウトマウスを用いて調べた。Tlr9欠損マウス及びDectin-1欠損マウスからの脾細胞及びFlt3リガンド誘導骨髄由来DCs (FL-DCs)をK3-SPGで刺激すると、サイトカイン産生は完全にTLR9依存的であったが、Dectin-1については、そうではなかった(図24)。インビトロの結果と一致して、K3-SPG及びOVAによるTlr9欠損マウスの免疫の結果、液性反応及びT細胞反応が減弱した(図25-27)。OVA 及び10μgのK3-SPG でマウスを免疫すると、Dectin-1欠損マウスは、野生型マウスと同等の免疫反応を示した(図28)。Dectin-1欠損マウスをOVA及び1μgのK3-SPGで免疫すると、抗体産生及びT細胞からのサイトカイン産生については有意な変化は認められないが(図29、図30)、テトラマーアッセイにおいて減弱したCD8 T細胞反応を示した(図31)。これらの結果は、K3-SPGのアジュバント効果はTLR9シグナリングに依存することを示唆する。SPG及びK3-SPGはDectin-1シグナリングを刺激しないが、K3-SPGの効果は、インビボにおいて、なお部分的にDectin-1に依存する。
K3-SPGは、単純な抗原混合物の免疫接種を通じて、インビボにおいて強力なアジュバント効果をもたらすので、抗原及びK3-SPGの両方を捕捉する細胞が、アジュバント効果の媒介において重要な役割を果たすと仮定した。蛍光標識OVA及びK3-SPGのインビボ分布を調べるため、蛍光顕微鏡及び二光子顕微鏡を用いた。尾の付け根に注入後1時間以内に、抗原及びアジュバントは流入領域リンパ節(iLNs)に到達した(図32、33、35)。24時間後、いくらかのK3-SPGがCD3e+ T細胞領域に移動し、DQ-OVAと共局在した。K3-SPG及びDQ-OVAの両方を含有する、iLNのT細胞領域におけるこれらの細胞は、CD11c+DCsであった。
本発明者らの知見は、ナノ微粒子K3-SPGの大部分は、注入後にiLNsにおけるMARCO+マクロファージにより取り込まれるが、アジュバント効果はDCsによりコントロールされることを示唆する。そこで、iLNsにおけるDC集団による抗原及びアジュバントの取込に焦点をあてた。投与後24時間において、DC集団による抗原及びアジュバントの取込をフローサイトメトリーにより解析した。3つのDCサブセット(pDCs、CD8α+ DCs、CD8α- DCs)におけるCpG陽性の頻度はK3-SPG注入後に、K3のときよりも有意に増加した。対照的に、OVA陽性DCの頻度は、K3注入後とK3-SPG注入後とで同等であった。抗原及びアジュバントの両方とも陽性のDCsに焦点をあてると、K3-SPGにおいてK3よりもかなりの増加が認められた。注入後24時間において、iLNs におけるpDCs及びCD8α陽性DCsの双方とも、K3-SPGにより強力に活性化されたが、K3によってはされず、これは全体的にTLR9に依存した(図38)。これらの結果から、pDCs及びCD8α+ DCsが、成熟化のための非微粒子K3ではなく、優先的にナノ微粒子K3-SPGを捕捉し、アジュバント効果を発揮することが示された。
K3-SPGのアジュバント効果を、マウス及び非ヒト霊長類において、より臨床的に関連するインフルエンザワクチン接種モデルを用いて調べた。マウスをエーテル処理ヘムアグルチニン抗原濃縮ビリオン不含スプリットワクチン(SV)及び表示したアジュバントで免疫接種し、抗体反応及びT細胞反応を比較すると、K3-SPGは、K3よりも優れたアジュバント効果を示した(図43)。より重要なことに、SVとK3-SPGによる免疫接種により、ビルトインアジュバントとしてウイルスRNAを含有する(Koyama, S., et al., Science translational medicine 2, 25ra24 (2010))、全(ビリオン)不活化ワクチン(WIV)(0.2 μg/マウス)を用いたワクチン接種と比較しても、100倍大きな抗体反応が生じた(図39)。SV(0.1 μg/マウス)及びK3-SPGで免疫接種したマウスは、WIVで免疫化したマウスよりも体重の減少が少なかった(図40)。驚くべきことに、WIVワクチン接種マウスでは僅か10%のみ生存をもたらす用量において、K3-SPGは、致死的なPR8ウイルス負荷に対して100%防御をもたらした(図40)。これらの結果は、マウスモデルにおいて、K3-SPGがタンパク質又はタンパク質ベースのワクチンの強力なアジュバントとして機能するとの考えを強力に支持した。この知見を、カニクイザルを用いた非ヒト霊長類モデルに拡張することとした。0日目及び14日目に、各群3匹のカニクイザルをK3又はK3-SPGと共にSVで免疫接種した。血清抗体力価を8週間に亘ってモニターした。SV及びK3-SPGにより、免疫接種の2週間後に有意に高い抗体力価が誘導され、この力価は少なくとも更に6週間に亘って高いままであった(図41)。免疫接種から2年(110週)後において、K3-SPG群は、K3群よりも高い抗体力価を示した(図42及び43)。まとめると、これらの結果から、K3-SPGが非ヒト霊長類モデルにおいても卓越したワクチンアジュバントであることが示唆された。
ヒトPBMCs(Lonza社、Cat# CC-2702、Lot# 0000396517)を用いて、K3(K3-dA30, K3-dA35, K3-dA40)単独、K3-LNT複合体(K3-dA30-LNT, K3-dA35-LNT, K3-dA40-LNT)及びK3-SPG複合体(K3-dA40-LNT)のpan-IFN-a(hIFNa)とIL-6(hIL-6)の産生誘導能を評価した。
結果を図44に示す。低用量の刺激において、pan-IFN-aとIL-6産生は、K3とSPGとの複合体であるK3-SPG、およびK3とLNTとの複合体であるK3-LNTにおいて、K3単独に比べて高いことが認められた。また、dA tail長(dA30, dA35, dA40)が異なるK3-LNTによって誘導される炎症性サイトカイン産生能は、ほぼ同等である可能性が示唆された。更に、K3-SPGとK3-LNTの炎症性サイトカイン産生能を比較すると、K3-SPGに比べて、K3-LNTによるpan-IFN-a産生誘導能が高い可能性が示唆された。一方、IL-6産生量においては、K3-SPGとK3-LNTは同等であることが認められた。
7週齢のC57BL/6マウスの尾根部に、一匹あたりRSV F抗原0.5 μgと各種アジュバント(K3単独(K3-dA30, K3-dA35, K3-dA40)、K3-LNT複合体(K3-dA30-LNT, K3-dA35-LNT, K3-dA40-LNT)及びK3-SPG複合体(K3-dA40-SPG)、リン酸アラム)10 μgを2週間隔で2回免疫した。最終免疫から1週後に末梢血の回収と血清の調製を行い、評価試料とした。血清中のRSV Fワクチン抗原に結合する抗体価は、ELISA法を用いて測定した。図45に示されるとおり、RSV F抗原特異的なTotal IgG誘導は、RSV F抗原単独接種群(F)に比べて、アジュバント添加によって増強されており、K3単独(F+K3-dA30, F+K3-dA35, F+K3-dA40)、K3-LNT(F+K3-dA30-LNT, F+K3-dA35-LNT, F+K3-dA40-LNT)及びK3-SPG複合体(F+K3-dA40-SPG)とリン酸アラム(F+Alum)のアジュバント効果は同等である可能性が示唆された。Th2型アジュバントであるリン酸アラム(F+Alum)を接種したマウス群では、IgG1サブクラス誘導能がRSV F抗原単独接種群に比べて高いことが認められた。一方、IgG2cサブクラス抗体において、K3単独(F+K3-dA30, F+K3-dA35, F+K3-dA40)、K3-LNT(F+K3-dA30-LNT, F+K3-dA35-LNT, F+K3-dA40-LNT)、K3-SPG(F+K3-dA40-SPG)を接種した免疫群では、リン酸アラム(F+Alum)に比べて高い結果が示されており、K3-LNT複合体はK3-SPGと同様のTh1型アジュバントである可能性が示唆された。
7週齢のC57BL/6マウスの尾根部に、一匹あたりRSV F抗原0.5 μgと各種アジュバント(K3単独(K3-dA30, K3-dA35, K3-dA40)、K3-LNT(K3-dA30-LNT, K3-dA35-LNT, K3-dA40-LNT)及びK3-SPG複合体(K3-dA40-SPG)、リン酸アラム)10 μgを2週間隔で2回免疫した。最終免疫から1週後に脾臓を回収し、脾臓細胞を調製した。96 well培養プレートに播種した脾臓細胞を、RSV F抗原のMHC class I拘束性エピトープペプチド、MHC class II拘束性エピトープペプチド、ワクチン抗原タンパク質それぞれで刺激し、24時間もしくは48時間、培養した。RSV F抗原特異的サイトカイン産生能は、培養上清を試料とし、サイトカインELISA法を用いて評価した。本検討では、Th1型サイトカインであるIFN-g、活性化T細胞から産生されるIL-2、Th2型サイトカインであるIL-13の3種のサイトカインについて評価した。
その結果、図46に示されるとおり、RSV F抗原特異的なIFN-g産生誘導とIL-2産生誘導の増強効果は、K3-LNT(F+K3-dA30-LNT, F+K3-dA35-LNT, F+K3-dA40-LNT)とK3-SPG(F+K3-dA40-SPG)で同等である可能性が示唆された。Th2型アジュバントであるリン酸アラムアジュバント接種群(F+Alum)では、IFN-g産生はいずれの刺激においても検出限界以下であった。一方、IL-13産生増強効果は、Th2型アジュバントであるリン酸アラム接種群で高く認められ、Th1型アジュバントであるK3-SPG接種群では低い結果となった。興味深いことに、K3-LNT接種群(F+K3-dA30-LNT, F+K3-dA 35-LNT, F+K3-dA 40-LNT)においては、同じTh1型アジュバントであるK3-SPG接種群(F+K3-dA40-SPG)に比べ、高いIL-13産生増強効果を示した。このことから、K3-LNT複合体は、K3-SPGが有する高いTh1応答増強効果に加え、Th2応答増強能も有することが示唆された。
6〜7週齢のコットンラットの尾根部に、一匹あたりRSV F抗原1 μgと各種アジュバント(K3-LNT(K3-dA35-LNT, K3-dA40-LNT)及びK3-SPG複合体(K3-dA40-SPG)、リン酸アラム)10 μgを2週間隔で2回免疫した。最終免疫から2週後に、RSV血清型A(Long株)をコットンラットに経鼻接種を用いて攻撃感染させ、その3日後に肺内ウイルス量を計測した。Synagis(palivizumab)投与群は、Synagis 2.5 mg /kgを攻撃感染一日前に筋肉内投与し、上記と同様に感染防御能の評価を行った。中和抗体価は、攻撃感染直前に麻酔下でラット頚静脈から採血し、得られた血清を用いて検討を行った。
図47の左に肺内ウイルス量の結果を、右に中和抗体価の結果を示す。肺内ウイルス量の結果から、PBS-投与群では約105pfu/lungのウイルス量が認められるのに対し、リン酸アラム添加RSV Fワクチンを投与した群(F+Alum)では、平均値で約100倍もの低いウイルス量に抑えられていた。一方、K3-SPG添加ワクチン投与群(F+K3-dA40-SPG)においても、感染防御誘導能が認められた。最も良好な感染防御効果を示したのは、K3-LNT添加ワクチン群(F+K3-dA35-LNT, F+K3-dA40-LNT)であり、リン酸アラムやK3-SPGに比べて、高い感染防御能に寄与する新規ワクチンアジュバント候補である可能性が示唆された。
中和抗体誘導能においては、リン酸アラム添加ワクチン投与群(F+Alum)において、シナジスと同等の血中中和活性が認められた。一方、K3-SPG添加ワクチン投与群(F+K3-dA40-SPG)においては、4匹中3匹で中和抗体がほとんど誘導されていないことから、K3-SPGが寄与する感染防御効果は中和抗体非依存的な機作によるものと考察された。また、K3-LNT投与群(F+K3-dA35-LNT, F+K3-dA40-LNT)においては、K3-SPGに比べて、高い中和抗体が認められていることから、K3-LNTアジュバントはK3-SPGと異なる特性、例えばTh2応答増強能、を有している可能性が示唆された。
Claims (37)
- ヒト化K型CpGオリゴデオキシヌクレオチド及びポリデオキシアデニル酸を含む、オリゴデオキシヌクレオチドであって、ポリデオキシアデニル酸が、ヒト化K型CpGオリゴデオキシヌクレオチドの3’側に配置されている、オリゴデオキシヌクレオチド。
- ヒト化K型CpGオリゴデオキシヌクレオチドが、10ヌクレオチド以上の長さであり、且つ式:
で表されるヌクレオチド配列を含む、請求項1に記載のオリゴデオキシヌクレオチド。 - ヒト化K型CpGオリゴデオキシヌクレオチドが、配列番号1で表されるヌクレオチド配列からなる、請求項1又は2に記載のオリゴデオキシヌクレオチド。
- オリゴデオキシヌクレオチドのリン酸ジエステル結合の一部又は全てがホスホロチオエート結合により置換されている、請求項1〜3のいずれかに記載のオリゴデオキシヌクレオチド。
- オリゴデオキシヌクレオチドのリン酸ジエステル結合の全てがホスホロチオエート結合により置換されている、請求項4のオリゴデオキシヌクレオチド。
- ポリデオキシアデニル酸の長さが、20〜60ヌクレオチド長である、請求項1〜5のいずれかに記載のオリゴデオキシヌクレオチド。
- 請求項1〜6のいずれかに記載のオリゴデオキシヌクレオチド及びβ−1, 3−グルカンを含有する複合体。
- β−1, 3−グルカンが、レンチナン、シゾフィラン、スクレログルカン、カードラン、パーキマン、グリホラン、又はラミナランである、請求項7に記載の複合体。
- β−1, 3−グルカンがレンチナン、シゾフィラン、又はスクレログルカンである、請求項8に記載の複合体。
- 下記(i)に記載のオリゴデオキシヌクレオチド、及び(ii)に記載のβ−1, 3−グルカンからなる複合体。
(i)配列番号1で表されるヌクレオチド配列からなるオリゴデオキシヌクレオチドの3’側に20〜60ヌクレオチド長のポリデオキシアデニル酸が結合し、かつリン酸ジエステル結合の全てがホスホロチオエート結合で置換されているオリゴデオキシヌクレオチド
(ii)レンチナン又はシゾフィラン - 三重螺旋構造状のものである、請求項7〜10のいずれかに記載の複合体。
- B細胞を活性化してIL-6を産生させる活性、及び樹状細胞を活性化してIFN-αを産生させる活性を有する、請求項7〜11のいずれかに記載の複合体。
- 請求項1〜6のいずれかに記載のオリゴデオキシヌクレオチド、或いは請求項7〜12のいずれかに記載の複合体を含む、医薬組成物。
- ウイルス感染症、癌、アレルギー疾患、細胞内寄生性原虫又は細菌感染症の予防又は治療のための、請求項13に記載の医薬組成物。
- ウイルス感染症の予防又は治療のための、請求項14に記載の医薬組成物。
- ウイルス感染症がRSウイルス、又はインフルエンザウイルス感染症である、請求項15に記載の医薬組成物。
- 請求項7〜12のいずれかに記載の複合体を含む、I型及び/又はII型インターフェロン産生誘導剤。
- 請求項1〜6のいずれかに記載のオリゴデオキシヌクレオチド、或いは請求項7〜12のいずれかに記載の複合体を含む、免疫賦活剤。
- ワクチンアジュバントである、請求項18の免疫賦活剤。
- [1]〜[6]のいずれかに記載のオリゴデオキシヌクレオチド、或いは請求項7〜12のいずれかに記載の複合体を含む、ウイルス感染症、癌、アレルギー疾患、細胞内寄生性原虫又は細菌感染症の予防又は治療剤。
- ウイルス感染症がRSウイルス、又はインフルエンザウイルス感染症である請求項20に記載の予防又は治療剤。
- 医薬組成物の製造のための、請求項1〜6のいずれかに記載のオリゴデオキシヌクレオチド、或いは請求項7〜12のいずれかに記載の複合体の使用。
- 医薬組成物が、ウイルス感染症、癌、アレルギー疾患、細胞内寄生性原虫又は細菌感染症の予防又は治療のための医薬組成物である、請求項22に記載の使用。
- ウイルス感染症がRSウイルス、又はインフルエンザウイルス感染症である、請求項23に記載の使用。
- 請求項1〜6のいずれかに記載のオリゴデオキシヌクレオチド、或いは請求項7〜12のいずれかに記載の複合体の薬理的有効量を温血動物に投与することを含む、当該温血動物における疾患の治療もしくは予防のための方法。
- 疾患が、ウイルス感染症、癌、アレルギー疾患、細胞内寄生性原虫又は細菌感染症である、請求項25に記載の方法。
- ウイルス感染症が、RSウイルス、又はインフルエンザウイルス感染症である、請求項26に記載の方法。
- 温血動物が、ヒトである、請求項25〜27のいずれかに記載の方法。
- 請求項1〜6のいずれかに記載のオリゴデオキシヌクレオチド、或いは請求項7〜12のいずれかに記載の複合体の薬理的有効量を温血動物に投与することを含む、当該温血動物における防御免疫反応を誘導する方法。
- ウイルス感染症、癌、アレルギー疾患、細胞内寄生性原虫又は細菌感染症の治療又は予防において使用するための、請求項1〜6のいずれかに記載のオリゴデオキシヌクレオチド、或いは請求項7〜12のいずれかに記載の複合体。
- ウイルス感染症が、RSウイルス、又はインフルエンザウイルス感染症である、請求項30に記載のオリゴデオキシヌクレオチド又は複合体。
- (a)請求項1〜6のいずれかに記載のオリゴデオキシヌクレオチド、或いは請求項7〜12のいずれかに記載の複合体、及び
(b) 抗原
を含む、医薬組成物。 - 該抗原に対する免疫反応を誘導するための、請求項32に記載の組成物。
- 抗原が病原体由来の抗原である、請求項33に記載の組成物。
- 病原体の感染症の予防又は治療用である、請求項34に記載の組成物。
- 病原体がウイルスである、請求項35に記載の組成物。
