[1] 米糠タンパク質を含有することを特徴とする脂質代謝異常改善組成物、

[4] 脂質代謝異常が、高脂血症、肥満及びII型糖尿病から選択される少なくとも1の疾患に起因するものであることを特徴とする前記[1]〜[3]のいずれかに記載の組成物。

1.試験方法
4週齢のウィスター系雄ラット(日本クレア(株))を3日間予備飼育した後、1群8匹として以下の5群に群分けして実験に用いた。各群に常法により調製した表1に示す飼料をそれぞれ摂食させた。実験期間中、飲水は自由摂取させた。
試験群:
A1群:通常食(タンパク源:カゼイン20質量%);
A2群:高コレステロール食(タンパク源としてカゼイン20質量%含有;コントロール群);
B群:高コレステロール食(タンパク源としてカゼイン10質量%及び実施例1の米糠タンパク質を含有する米糠抽出物10質量%含有);
C群:高コレステロール食(タンパク源としてカゼイン5質量%及び実施例1の米糠タンパク質を含有する米糠抽出物15質量%含有);
D群:高コレステロール食[タンパク源としてカゼイン10質量%及び分離大豆タンパク質(不二製油株式会社製;タンパク質含量86質量%)10質量%含有]。
表2に試験結果を示す。数値は各試験群の平均値±標準誤差で表している。A2群(コントロール群)では、A1群(通常食群)に比べ有意に血中及び肝臓における総コレステロール及び中性脂肪が上昇し、脂質代謝異常を示した。B群とC群の米糠タンパク質を含有する米糠抽出物を摂食させた群において、血中及び肝臓のコレステロールと中性脂肪がA2群(コントロール群)のそれらと比べ有意に低下し、改善されていた。また、血中アディポネクチンは高コレステロール食食餌により減少した(A2群)が、米糠タンパク質を含有する米糠抽出物を摂食させたB群、C群及び分離大豆タンパク質を摂食させたD群では該血中アディポネクチンの減少を抑制した。
1.試験方法
離乳後のSD系雄ラット(日本クレア(株))を、1群6匹として以下の3群:
コントロール群:表3のコントロール群の飼料群(タンパク源としてカゼイン20質量%含有);
米糠タンパク質投与群:表3の米糠タンパク質投与群の飼料群[タンパク源として糠タンパク質を実施例1と同様に抽出した米糠タンパク質を含有する米糠抽出物(タンパク質量:66.1質量%)をタンパク質換算で20質量%含有];
大豆タンパク質投与群:表3の大豆タンパク質(不二製油株式会社製)投与群の飼料群[タンパク源として分離大豆タンパク質(不二製油株式会社製;タンパク質含量86質量%)をタンパク質換算で20質量%含有];
に群分けし、各群に常法により調製した表3に示す飼料をそれぞれ7週間摂食させた。食餌は制限食とし、1〜4日目は9g、5〜7日目は11g、8〜13日目は15g、14〜21日目は18g、22〜27日目は19g、28〜32日目は20g、33 〜35日目は21g、36〜42日目は22g、43〜49日目は23gずつとした。実験期間中、飲水は自由摂取させた。経時的に体重、糞重量(48時間糞)、糞中窒素量及び尿中窒素量を測定した。糞中窒素量及び尿中窒素量は、ケルダール法によって測定した。7週間後にエーテル麻酔を施し、開腹して腹部下大動脈より採血し、脱血死させた。脱血死後肝臓、ヒラメ筋、内臓脂肪組織(副睾丸脂肪組織、腎臓周囲脂肪組織)及び盲腸を摘出し、各組織重量を測定した。
(1)7週間経過後の成長量、各組織重量比及び血中もしくは肝臓脂質を表4に示す。米糠タンパク質投与群及び大豆タンパク質投与群における副睾丸脂肪組織、腎臓周囲脂肪組織及び腹腔内脂肪組織重量比は、コントロール群の各脂肪組織重量比に比べていずれの組織においてもその重量比を減少させる効果が認められた。その効果は、米糠タンパク質投与群の方が大豆タンパク質投与群よりも大きかった。また、副睾丸脂肪組織及び腎臓周囲脂肪組織を電子顕微鏡で観察したところ、米糠タンパク質投与群では、副睾丸脂肪組織及び腎臓周囲脂肪組織におけるそれぞれの脂肪細胞の大きさも小さかった。このことは、米糠タンパク質を含有する米糠抽出物は、分離大豆タンパク質に比べ内臓脂肪を減少させる効果に優れていることを示す。また、米糠タンパク質投与群及び大豆タンパク質投与群において、コントロール群に比し盲腸重量比が増加し、盲腸内容物のpHが低下した。米糠タンパク質投与群の盲腸の増大は、大豆タンパク質投与群の盲腸の増大や盲腸内容物のpHの低下より有意に大きく、盲腸内容物のpHの減少度合も大きかった。また、米糠タンパク質投与群でヒラメ筋の重量比が高くなる傾向があった。つまり、米糠タンパク質を含有する米糠抽出物は、分離大豆タンパク質に比べ内臓脂肪を減少させ、逆に筋肉を増大させる効果に優れていることを示す。該未消化物の増加は、腸内での糖質や脂質の吸収を抑制することになり、肥満の予防に役立つほか、食後の血糖の上昇を抑制しインスリンの分泌を抑制できることにもなり得る。実際に、糞便中の脂質含量を測定したところ、米糠タンパク質投与群で糞便中の脂質含量が高くなることを確認している。
1.試験方法
試験例1において、試験群の群分けを以下とする以外は試験例1と同様に行い、血清を分離し、肝臓を摘出した。血清及び肝臓中の総コレステロール及び中性脂肪、血清中のGOT(gulutamic-oxaloacetic transaminase)、GPT(gulutamic-pyruvic transaminase)、及びレプチンを測定した。総コレステロール及び中性脂肪は実施例1と同様に行った。GOT及びGPTはトランスアミナーゼCII−テストワコー(和光純薬工業株式会社製)で測定した。レプチンはモリナガラットレプチン測定キット(森永生化学研究所製)で測定した。
試験群:
A群:高コレステロール食(表1のA2群と同じ;コントロール群);
B群:高コレステロール食[表1のB群において、米糠抽出物を実施例1と同様に抽出した米糠タンパク質を含有する米糠抽出物(タンパク質量:75質量%)をタンパク質換算で10質量%に変える以外は表1のB群と同じ];
C群:高コレステロール食[表1のB群において、米糠抽出物を実施例1と同様に抽出した米糠タンパク質を含有する米糠抽出物(タンパク質量:65質量%)をタンパク質換算で10質量%)に変える以外は表1のB群と同じ];
D群:高コレステロール食[表1のD群と同じ]。
結果を表6に示した。試験例1の結果と一致して、コレステロール摂食ラットにおける血清コレステロールや肝臓の脂質含量は、米糠タンパク質を含有する米糠抽出物で抑制された。さらに、肝臓障害の指標となる血清GOTやGPT活性が米糠タンパク質を含有する米糠抽出物により抑制された。このことにより、肝臓に脂質が蓄積し、肝臓機能が低下するような場合に、米糠タンパク質を含有する米糠抽出物は肝臓機能を改善し、脂質代謝異常を予防又は改善させることが期待できる。
