Description
Claims (1)
- 【特許請求の範囲】 1.核酸増幅反応の進行をリアルタイムでモニタリングする装置において、 増幅生成物を合成すべく核酸増幅反応を行なうための反応混合物と、 光ファイバにより伝達される励起ビームがレンズにより反応混合物の体積中 に合焦されるように、レンズと同心状に配置された光ファイバからなる試料イン ターフェースと、 第1蛍光信号を発生できる第1蛍光指標とを有し、第1蛍光信号の強度は励 起ビームにより照射される反応混合物の体積中の増幅生成物の量に比例し、 反応混合物の全体に亘って均質に分散され且つ励起ビームにより照射される 反応混合物の体積に比例する第2蛍光信号を発生できる第2蛍光指標を更に有し 、 試料インターフェースのレンズが、第1及び第2蛍光信号の一部を収集し且 つ該部分を光ファイバ上に合焦させ、光ファイバが前記部分を検出/分析手段に 伝達することを特徴とする装置。 2.前記第1蛍光信号が或る強度を有し且つ前記第2蛍光信号が或る強度を有し 、前記検出/分析手段が、前記第1蛍光信号の強度と第2蛍光信号の強度との比 からなる読取りを与えることを特徴とする請求の範囲第1項に記載の装置。 3.前記第1蛍光指標が錯体形成染料であることを特徴とする請求の範囲第2項 に記載の装置。 4.前記装置は複数の前記試料インターフェースからなり、該試料インターフェ ースの前記各光ファイバが、前記レンズと同心状に配置される第1端部及び光フ ァイバマルチプレクサに連結される第2端部を備えていることを特徴とする請求 の範囲第3項に記載の装置。 5.前記反応混合物は閉鎖反応容器内に収容され、前記レンズは、前記励起ビー ムを閉鎖反応容器の壁の部分を通して前記反応混合物中に合焦させ、前記壁の部 分は加熱されて、該部分に前記反応混合物の成分が凝縮することを防止すること を特徴とする請求の範囲第3項に記載の装置。 6.前記閉鎖反応容器がつや消しチューブであることを特徴とする請求の範囲第 5項に記載の装置。 7.前記装置が複数の前記第1蛍光指標を有し、各第1蛍光指標が異なる前記増 幅生成物に対応することを特徴とする請求の範囲第3項に記載の装置。 8.前記増幅反応がポリメラーゼ鎖反応であることを特徴とする請求の範囲第3 項に記載の装置。 9.前記増幅反応がリガーゼ鎖反応であることを特徴とする請求の範囲第3項に 記載の装置。 10.前記増幅反応がポリメラーゼ鎖反応であり、前記第1及び第2蛍光指標が、 前記増幅生成物の鎖の部分を補完するヌクレオチド配列を備えたオリゴヌクレオ チドに共有結合され、前記第2蛍光指標が前記第1蛍光指標の蛍光を消光するこ とを特徴とする請求の範囲第2項に記載の装置。 11.前記装置が複数の前記試料インターフェースを有し、該試料インターフェー スの前記各光ファイバが、前記レンズと同心状に配置される第1端部及び光ファ イバマルチプレクサに連結される第2端部を備えていることを特徴とする請求の 範囲第10項に記載の装置。 12.前記反応混合物は閉鎖反応容器内に収容され、前記レンズは、前記励起ビー ムを閉鎖反応容器の壁の部分を通して前記反応混合物中に合焦させ、前記壁の部 分は加熱されて、該部分に前記反応混合物の成分が凝縮することを防止すること を特徴とする請求の範囲第10項に記載の装置。 13.前記閉鎖反応容器がつや消しチューブであることを特徴とする請求の範囲第 12項に記載の装置。 14.前記装置が複数の前記第1蛍光指標を有し、各第1蛍光指標が異なる前記増 幅生成物に対応することを特徴とする請求の範囲第10項に記載の装置。
