Description
図2は、セリンを添加しない培地での指数増殖期への進入を確認するために、ハイフォミクロビウム属SWB−P91(KCTC12695BP)菌株の生育程度を比較したグラフである。
完全平板培地(メタノール0.2%、硫酸アンモニウム0.3%、リン酸水素カリウム0.14%、リン酸二ナトリウム0.21%、セリン0.02%、硫酸マグネシウム0.1%、第1鉄クエン酸塩0.003%、塩化カルシウム0.003%、硫酸マンガン0.0001%、硫酸亜鉛0.002%、硫酸銅0.00001%、寒天1.5%、pH 7.0)で30℃、72時間培養されたハイフォミクロビウム属母菌株Hyphomicrobium denitrificans(ATCC51888)を完全液体培地に接種し、30℃で48時間培養した。本明細書で培地の濃度において「%」は、重量%を意味する。培養後、12000rpmで15分間遠心分離した後、生理食塩水で2回水洗した。同一の液で菌体濃度が約1 OD(600nm)となるように適切に希釈した後、N−メチル−N'−ニトロ−N−ニトログアニチン(NTG)250μg/mlとなるように加えて、30℃で30分から80分間処理した。

発酵生産培地(メタノール1%、硫酸アンモニウム0.3%、リン酸水素カリウム0.14%、リン酸二ナトリウム0.21%、セリン0.02%、硫酸マグネシウム0.1%、第1鉄クエン酸塩0.003%、塩化カルシウム0.003%、硫酸マンガン0.0001%、硫酸亜鉛0.002%、硫酸銅0.00001%、pH 7.0)1.8Lを5L規模の小型発酵槽に入れ、121℃、20分間殺菌を実施した後、同一培地で30℃、120rpm、48時間培養された種菌培養液200mlを接種し、500rpm、1vvmの条件で30℃で150時間発酵を行った。発酵液においてpHは、アンモニア水で7に調節し、追加メタノールは、発酵中に添加する流加式発酵工程で菌株のピロロキノリンキノン生産性を測定した。流加式培養において培養液内のメタノール濃度は、0.5%に維持し、この際、発酵時間150時間後に、最終菌体生育度及びピロロキノリンキノン生産性を母菌株と比較した。母菌株は、流加式培養において培地内のメタノール濃度が0.2%以上で生育が阻害を受けるため、0.1%に維持し、同一条件で発酵を進行した。表3のように、発酵実験で本発明の変異菌株は、母菌株に比べて培地中メタノール濃度0.5%で生育が維持さ、母菌株に比べて生産性に優れた安定的な菌株であることを確認した。
培地成分にセリンを添加することによって生育誘導期(lag phase)から指数増殖期への進入が安定的に進行されるかを調べる比較実験を行った。このために、完全液体培地(メタノール0.2%、硫酸アンモニウム0.3%、リン酸水素カリウム0.14%、リン酸二ナトリウム0.21%、硫酸マグネシウム0.1%、第1鉄クエン酸塩0.003%、塩化カルシウム0.003%、硫酸マンガン0.0001%、硫酸亜鉛0.002%、硫酸銅0.00001%、pH 7.0)にセリンを添加しない培地と、セリンを0.002%で添加した培地それぞれ30個のフラスコにハイフォミクロビウムSWB−P91菌株を10%の量で種菌接種し、30℃で30時間120rpmで培養し、生育程度を比較した。
実施例4−1:ダイヤイオンHP−20(DIAION HP−20)によるピロロキノリンキノンの吸着
実施例2の発酵培養液でピロロキノリンキノン精製を進行した。培養液0.5Lを遠心分離機を利用して菌体を除去し、上清液を5N HClを利用してpH 1.8に調整した。これをダイヤイオンHP−20(DIAION HP-20) 樹脂100mlが充填されたカラムに流して、ピロロキノリンキノンを樹脂に吸着させた。吸着が完了したカラムにpH 1.5に調節された蒸留水を樹脂量の3倍数で流して洗浄を実施した後、0.2Nアンモニア水を利用して脱離を進行した。アンモニア水が樹脂に通過しつつピロロキノリンキノンの赤色の帯が脱離液に沿って移動することが認められ、赤色部分が溶離される部分を分取した。その後、1Nアンモニア水を利用して追加脱離を進行したが、これ以上ピロロキノリンキノンは溶離されなかった。また、100mlのDEAEセファロース(DEAE sepharose)樹脂で充填されたカラムに培養上清液(pH7.0)0.5Lを流して吸着を実施した。その後、0.2M塩化ナトリウム溶液3倍数で洗浄を実施した後、0.65M塩化ナトリウム溶液で脱離を進行した。赤色ピロロキノリンキノン部分が溶離される部分を分取した。その後、1M塩化ナトリウム溶液で再溶離を進行したが、これ以上ピロロキノリンキノンは溶離されなかった。比較実施したDEAEセファロース(DEAE-sepharose)樹脂を利用したピロロキノリンキノン分離精製した結果は、表4に示された通りである。
実施例4−2でダイヤイオンHP−20(DIAION HP-20)樹脂を利用して吸着したピロロキノリンキノン及び0.2Nアンモニア水で溶離されたピロロキノリンキノン溶液に対して減圧濃縮を実施した。減圧濃縮によって全量のアンモニア水を除去することができ、約210mgのピロロキノリンキノンを回収することができた。回収率は、68.9%を示した。

Claims (6)
- ピロロキノリンキノン(pyrrolo-quinoline quinone)高生産能を有するハイフォミクロビウム(Hyphomicrobium sp.)属変異菌株SWB−P91(KCTC12695BP)。
- ハイフォミクロビウム(Hyphomicrobium sp.)属変異菌株SWB−P91(KCTC12695BP)を培養する段階と、
前記培養発酵液から吸着樹脂を利用して発酵液内のピロロキノリンキノンを吸着する段階と、
前記吸着されたピロロキノリンキノンを溶離液で脱離する段階と、
前記脱離されたピロロキノリンキノン溶液を減圧濃縮し、ピロロキノリンキノンを回収する段階と、
を含む、ピロロキノリンキノンの大量生産方法。 - 前記培養は、流加式培養で行うことを特徴とする、請求項2に記載のピロロキノリンキノンの大量生産方法。
- 前記流加式培養を通じて培養液のメタノール濃度を0.1〜0.5重量%に維持させることを特徴とする、請求項3に記載のピロロキノリンキノンの大量生産方法。
- 前記溶離液は、アンモニア水であることを特徴とする、請求項2に記載のピロロキノリンキノンの大量生産方法。
- 前記吸着樹脂は、ダイヤイオン(登録商標)HP−20(DIAION(登録商標) HP-20)樹脂であることを特徴とする、請求項2に記載のピロロキノリンキノンの大量生産方法。
