実施例1−1:GLP−1に対してcAMP反応を示す細胞株の作製
ヒトGLP−1受容体遺伝子のcDNA(OriGene Technologies, Inc. USA)においてORF(open reading frame)に相当する部分を鋳型とし、HindIII切断部位とEcoRI切断部位をそれぞれ含む下記正方向及び逆方向プライマーを用いたPCRを行い、PCR産物を得た。

実施例2−1:オキシントモジュリン誘導体の合成
オキシントモジュリン誘導体のin vitro活性を測定するために、下記アミノ酸配列を有するオキシントモジュリン誘導体を合成した(表1)。
実施例2−1で合成したオキシントモジュリン誘導体の抗肥満効能を測定するために、実施例1−1及び1−2で製造した細胞株を用いてin vitroで細胞活性を測定す
る方法を行った。
オキシントモジュリン誘導体のin vivo肥満治療活性を測定するために、ob/obマウスにおいて、対照薬物として天然型オキシントモジュリンを用い、オキシントモジュリン誘導体投与による飼料摂取量の変化を測定した。

- 前記ペプチドが抗肥満効果を有する、請求項1〜5のいずれかに記載のペプチド。
- 請求項1〜7のいずれかに記載のペプチドをコードするポリヌクレオチド。
- 請求項1〜7のいずれかに記載のペプチドを有効成分として含む肥満予防又は治療用の薬学的組成物。
- さらに薬学的に許容される担体を含む、請求項9に記載の組成物。
- 前記薬学的組成物が、単独で若しくは、他の肥満予防又は治療効果を示す薬学的製剤と併用して投与される、請求項9又は10に記載の組成物。
- 前記肥満予防又は治療効果を示す薬学的製剤が、GLP−1受容体アゴニスト、レプチン(Leptin)受容体アゴニスト、DPP−IV阻害剤、Y5受容体アンタゴニスト、MCH(Melanin-concentrating hormone)受容体アンタゴニスト、Y2/3受容体アゴニスト、MC3/4受容体アゴニスト、胃/膵臓リパーゼ阻害剤、5HT2cアゴニスト、β3A受容体アゴニスト、アミリン(Amylin)受容体アゴニスト、グレリン(Ghrelin)アンタゴニスト又はグレリン受容体アンタゴニストである、請求項11に記載の組成物。
- 請求項1〜7のいずれかに記載のペプチド又は請求項9〜12のいずれかに記載の組成物をヒト以外の個体に投与する段階を含む、肥満を予防又は治療する方法。
- 肥満の予防又は治療用医薬の製造における、請求項1〜7のいずれかに記載のペプチド又は請求項9〜12のいずれかに記載の組成物の使用。
