天然(野生型)または改変IFN(例えば、IFN-α)に付着したターゲティング部分(例えば、抗腫瘍マーカー抗体)を含むキメラ構築物を用いて、ターゲティング部分が向けられるマーカーを発現する、またはそのマーカーを会合する標的癌細胞の成長および/または増殖を効果的に抑制することができることは、驚くべき発見であった。特定の実施形態において、ターゲティング部分は、インターフェロンに化学的にコンジュゲートされ、一方、他の実施形態において、ターゲティング部分は、IFN-αとの融合タンパク質として発現している。融合タンパク質として産生される場合、ターゲティング部分(例えば、抗体)構成要素は、IFN-αに直接融合することができ、またはペプチドリンカー(例えば、(Gly4Ser)3(配列番号31)リンカー、GlyGlyGlyGlySer(配列番号32)リンカー、AEAAAKEAAAKA(配列番号33)など)によって付着することができる。
様々な実施形態において、ターゲティング部分は、標的細胞(複数可)によって発現した(例えば、標的細胞の表面上の)、または標的細胞に会合したマーカーを特異的または優先的に結合する分子である。本質的に任意の細胞を標的とすることができるが、特定の好ましい細胞として、細胞の過剰増殖によって特徴づけられる病態(すなわち、過剰増殖性障害)に関連した細胞が挙げられる。例示的な過剰増殖性障害には、乾癬、好中球増加症、赤血球増加症、血小板増加症、および癌が挙げられるが、それらに限定されるわけではない。
特定の実施形態において、ターゲティング部分は、腫瘍マーカーを特異的または優先的に結合する抗体、UniBody、またはAffibodyを含むことができる。腫瘍マーカーを特異的または優先的に結合する抗体は、当業者によく知られている。このように、例えば、ヒトB細胞上に発現するCD22抗原を結合する抗体として、HD6、RFB4、UV22-2、Tol5、4KB128、ヒト化抗CD22抗体(hLL2)が挙げられる(例えば、Liら、(1989) Cell. Immunol. 111:85〜99頁; Masonら、(1987) Blood 69頁:836〜40頁; Behrら、(1999) Clin. Cancer Res. 5:3304s〜3314s頁; Bonardiら、(1993) Cancer Res. 53:3015〜3021頁参照)。
UniBodyは、安定な、より小さい抗体形式を生じる抗体テクノロジーであり、ある特定の小さい抗体形式より予想される治療濃度域が広い。特定の実施形態において、UniBodyは、IgG4抗体からその抗体のヒンジ領域を除去することによって作製される。完全なサイズのIgG4抗体と違って、半分の分子断片は、非常に安定であり、UniBodyと名付けられている。IgG4分子を半減させて、標的に結合できるUniBody上の1つの領域のみを残す。UniBodyを作製する方法は、全体として参照により本明細書に組み入れられているPCT公開WO 2007/059782に詳細に記載されている(Kolfschotenら、(2007) Science 317:1554〜1557頁も参照)。
Affibody分子は、ブドウ球菌プロテインAのIgG結合ドメインの1つに由来する58個のアミノ酸残基のタンパク質ドメインに基づいた親和性タンパク質の類である。この3ヘリックスバンドルドメインは、コンビナトリアルファージミドライブラリーの構築のためのスキャフォールドとして用いられており、そのライブラリーから、所望の分子を標的とするAffibody変異体が、ファージディスプレイテクノロジーを用いて選択することができる(例えば、Nordら、(1997) Nat. Biotechnol. 15:772〜777頁; Ronmarkら、(2002) Eur. J. Biochem. 269:2647〜2655頁参照)。Affibodyの詳細および作製方法は当業者に知られている(例えば、全体として参照により本明細書に組み入れられている、米国特許第5,831,012号参照)。
様々な実施形態において、本発明のキメラ成分は、ターゲティング部分(例えば、抗HER2/neu抗体)に連結したインターフェロン(例えば、IFN-α)を含む。インターフェロンは、完全長野生型インターフェロン(例えば、IFN-α、IFN-β、IFN-γなど)、インターフェロン断片(例えば、IFN-α断片)、および/または突然変異型インターフェロンであり得る。典型的には、インターフェロン断片は、野生型インターフェロンより、好ましくは少なくとも80%、より好ましくは少なくとも90%または95%、最も好ましくは少なくとも98%、99%、100%のレベル、またはそれ以上のレベルでの内因性活性を有するものである。
一般的に言えば、ターゲティング部分(例えば、抗HER2/neu抗体および抗CD20抗体など)は、任意の順序で連結することができる。したがって、例えば、抗体は、インターフェロンのアミノ末端かまたはカルボキシ末端かのいずれかに連結することができる。抗体はまた、インターフェロンの内部領域に連結することもでき、または逆に、インターフェロンが、その付着が抗体のその標的マーカー(例えば、HER2/neu受容体)への結合に干渉しない限り、抗体の内部の位置もしくは任意の末端に連結することができる。
特定の実施形態において、ターゲティング部分(例えば、C6.5、C6MH3-B1、G98A、ML3-9、H3B1、B1D2などの抗HER2/neu抗体)は、インターフェロン(例えば、IFN-α)分子に化学的にコンジュゲートされる。分子を化学的にコンジュゲートする手段は当業者によく知られている。
特定の実施形態において、キメラのターゲティング部分-インターフェロン融合タンパク質は、組換えDNA方法を用いて合成される。一般的に、これは、融合タンパク質をコードするDNA配列を作製する段階と、そのDNAを特定のプロモーターの制御下の発現カセットに配置する段階と、宿主においてタンパク質を発現させる段階と、発現したタンパク質を単離する段階と、必要に応じて、タンパク質を再生する段階を含む。
特定の実施形態において、本発明は、インターフェロンを標的細胞にターゲティングするための多価、好ましくは、三価、四価、五価、またはそれ以上のターゲティング部分(例えば、抗HER2/neu抗体、抗CD20抗体など)の使用を企図する。
本発明のキメラ構築物は、標的細胞(例えば、癌細胞)の成長および/または増殖を抑制するのに有用である。様々な実施形態において、キメラ成分は、疾患進行を抑制する、腫瘍サイズを縮小させる、および/または退縮/寛解を安定化するために用いることができる。
本発明の方法を行うために、本発明の1つまたは複数の活性物質(キメラ成分)が、例えば、癌を有すると診断された個体に投与される。活性物質(複数可)は、「天然」の形で、または、必要に応じて、塩、エステル、アミド、プロドラッグ、誘導体などの形で投与することができるが、ただし、塩、エステル、アミド、プロドラッグ、または誘導体が、本方法において薬理学的に適している、すなわち、効果的であるという条件である。活性物質の塩、エステル、アミド、プロドラッグ、および他の誘導体は、合成有機化学分野の業者に知られた、および例えば、March (1992) Advanced Organic Chemistry; Reactions, Mechanisms and Structure、第4版、N.Y. Wiley-Interscienceに記載の標準手順を用いて調製することができる。
特定の実施形態において、本発明は、原発性癌の処置のための、および/または補助治療においてのキットを提供する。キットは、典型的には、本発明のキメラ成分(例えば、抗HER2/neu-IFNα、抗CD20-IFN-αなど)を含む容器を含む。キメラ成分は、薬理学的に許容される賦形剤中に存在することができる。
以下の実施例は、例示するために提供されるが、主張された発明を限定するものではない。
抗Her2/neu IgG3とIFN-αの融合タンパク質は、B細胞リンパ腫に対する強力なアポトーシス活性および抗腫瘍活性を示す
本研究において、本発明者らは、C6MH3-B1の可変領域(20)を有する抗HER2/neu-IgG3およびIFN-αからなる融合タンパク質を構築し、ヒトHER2/neuを発現するマウスB細胞リンパ腫38C13(38C13/HER2)へのその効果を調べた。本発明者らは、このB細胞リンパ腫のIFN-αへの応答性を仮定して(21)、このモデルにおけるIFN-αの腫瘍へのターゲティングを評価するために選択した。IFN-αの抗体への融合は、そのインビボの半減期を有意に増加させた。抗HER2/neu-IgG3-IFN-αは、38C13/HER2の小腫瘍および樹立腫瘍の両方のインビボでの成長を抑制するのに効果があることが見出され、有効量において全身毒性の徴候は観察されなかった。抗HER2/neu-IgG3-IFN-αは、38C13/HER2細胞において成長を抑制し、アポトーシスを誘導した。これらの結果より、IFN-αの腫瘍特異的抗体への融合が、B細胞リンパ腫の処置に効果的な作用物質をもたらすことを示している。
細胞系および培養条件
38C13は、C3H/HeNマウス由来の極めて悪性のマウスB細胞リンパ腫である。ヒトHER2/neuを発現する38C13(38C13/HER2)の構築および特徴づけは、以前に記載されている(6)。38C13および38C13/HER2の両方を、2mMのL-グルタミン、10U/mlのペニシリン、10マイクロg/mlのストレプトマイシン(GPS; Sigma-Aldrich)、および10%仔ウシ血清(Atlanta Biologicals)を追加したIMDM(Irvine Scientific)中で培養した。マウス骨髄腫P3X63Ag8.653(American Type Culture Collection)および抗HER2 IgG3-IFN-αまたはIgG3-IFN-αを発現するそれの誘導体を、10%仔ウシ血清を追加したIMDM中で成長させ、GPS. L929線維芽細胞(American Type Culture Collection)を、5%仔ウシ血清およびGPSを含むIMDM中で培養した。ヒトHER2/neuを過剰発現するマウス結腸腺癌細胞系であるCT26/HER2の構築および特徴づけは、以前に記載されている(6)。CT26/HER2を、5%仔ウシ血清およびGPSを含むIMDM中で培養した。
抗ヒトHER2/neu scFvである、C6MH3-B1のH鎖可変領域およびL鎖可変領域を、ヒトγ3H鎖(pAH4802)およびκL鎖(pAG4622)発現ベクターへ、それぞれ挿入し(22)、それらを用いて、この特異性のキメラIgG3を作製した。抗ヒトHER2/neu-IgG3(C6MH3-B1)-IFN-α融合タンパク質を構築するために、まずPCRを用いて、BALB/cマウスのゲノムDNAからフォワードプライマー5'-CGC GGA TCC TGT GAC CTG CCT CAG ACT C-3(配列番号55)およびリバースプライマー5'-GCT CTA GAT CAT TTC TCT TCT CTC AGT CTT C-3(配列番号56)を用いるPCRによって増幅された成熟マウスIFN-α遺伝子の上流にBamH1制限酵素部位、および下流にXbaI制限酵素部位を導入した。最終PCR産物を、TAベクターにライゲーションした。シーケンシング後、生じたベクターをBamH1およびXbaIで消化し、DNA断片を遊離させ、それを、C6MH3-B1 H鎖可変領域およびCH3の末端のGGGGSGGGGSGGGGS(配列番号57)ペプチドリンカーと共にIgG3定常領域を含むベクターpAH9612に挿入した。最終PCR産物、pAH9616は、抗HER2/neu-IgG3、続いて、GGGGSGGGGSGGGGS(配列番号57)ペプチドリンカーおよびマウスIFN-αを含んだ。
IFN-αに融合したC6MH3-B1可変領域を含むIgG3 H鎖をコードするプラスミドを、960μFd電気容量および0.2Vのパルスでのエレクトロポレーションによって、抗HER2/neu-IgG3-IFN-αを産生するためにC6MH3-B1可変領域(23)を含むL鎖を発現するP3X63Ag8.653細胞へトランスフェクションするか、またはIgG3-IFN-αを産生するために非特異的L鎖(4D5; Genentech)(6)を発現するP3X63Ag8.653細胞へトランスフェクションした。抗HER2/neu(C6MH3-B1)-IgG3、抗HER2/neu(C6MH3-B1)-IgG3-IFN-α、またはIgG3-IFN-αを産生するトランスフェクタントを、以前に記載されているように(6)、選択し、特徴づけた。抗HER2/neu(C6MH3-B1)-IgG3を、Sepharose 4B fast flow(Sigma-Aldrich)上に固定化されたプロテインGを用いて培養上清から精製し、抗HER2/neu(C6MH3-B1)-IgG3-IFN-αおよびIgG3-IFN-αを、Sepharose 4B fast flow(Sigma-Aldrich)上に固定化されたプロテインAを用いて培養上清から精製した。純度および完全性を、SDS-PAGEにより分離されたタンパク質のクーマシーブルー染色によって評価した。National Institute of Healthによって提供されたマウスIFN-αについての国際参照基準を用いて、融合タンパク質のIFN活性を決定した。rIFN-αについては、PBL Biomedical Laboratoriesから入手した。
融合タンパク質が溶液中に単量体および/または多量体として存在するのかを決定するために、内部対照を提供するための400μgのOVAと混合した100μgのIgG3-IFN-αを、PBSおよび0.5ml/分流速を用いる高速タンパク質液体クロマトグラフィー(FPLC)に付属した30×1.5cm Superose 6カラムによるゲル濾過によって分析した。分子量標準を提供するために、350kDaの分子量をもつ主に二量体抗体として存在するIgA2m、および分子量150kDaのMiles IgGと分子量45kDaのOVAの混合物の同じカラムによるゲル濾過を用いた。
様々な抗HER2/neu融合タンパク質のCT26/HER2細胞との反応性を検出するために、1×106個の細胞を、10pMの融合タンパク質と4℃で1時間、インキュベートした。いくつかの実験については、融合タンパク質を、CT26/HER2細胞とのインキュベーション前に900Uのヘパリンと4℃で17時間、あらかじめインキュベートした。その後、細胞を、1/100希釈されたビオチン化ラット抗ヒトIgG(BD Biosciences)と反応させた。結合したビオチン化抗体を、1/1500希釈されたPE標識ストレプトアビジン(BD Biosciences)で検出し、細胞をFACScan(BD Biosciences)を用いるフローサイトメトリーによって分析した。
水疱性口内炎ウイルス(VSV)感染に感受性があるL-929線維芽細胞系を用いて、IFN-αの生物活性を定量化した。L-929細胞を96ウェル組織培養プレート(Falcon; BD Biosciences)に4×104個の細胞/ウェルの密度で蒔き、5%CO2雰囲気下、37℃で一晩、インキュベートした。その後、種々のIFN-α融合タンパク質または基準IFN-α(マウスIFN-αについての国際参照基準; National Institutes of Health、Bethesda、MD)の段階希釈を加え、プレートを37℃で24時間、インキュベートした。その後、4000PFUのVSVを各ウェルに加え、37℃でさらに48時間、インキュベートした。生存している接着細胞を、50μlのクリスタルバイオレット(20%エタノール中0.05%)で10分間、染色した。プレートを水で洗浄し、残存する色素を、100μlの100%メタノールの添加によって可溶化した。プレートを、ELISAリーダーを用いて595nmで読み取った。
簡単には、38C13または38C13/HER2細胞を、96ウェル組織培養プレートに1.25×104個の細胞/ウェルの密度で蒔き、種々の融合タンパク質の段階希釈を加えた。その後、プレートを5%CO2雰囲気下、37℃で48時間、インキュベートした。プレートを、20μlのMTS溶液(Promega)の添加によって発色させ、ELISAリーダーを用いて490nmで分析した。増殖の阻害(パーセント)を以下のように計算した: [(ODexp-ODblank)/(ODmedium-ODblank)]×100。
簡単には、1×106個の細胞を、種々の融合タンパク質で72時間、処理した。その後、細胞を氷冷PBSで洗浄した。アネキシンV/ヨウ化プロピジウム(PI)アッセイを、Vybrant Apoptosis Assay Kit 2(Molecular Probes)を用いて製造会社により推奨された手順に従って行った。
簡単には、1×106個の細胞を、2.5μM CFSE(Molecular Probes)と37℃で10分間、インキュベートした。その後、細胞を、1nMの種々の融合タンパク質で48時間、処理し、CellTrace CFSE Cell Proliferation Kit(Molecular Probes)を用いて製造会社により推奨された手順に従ってフローサイトメトリーによって分析した。
Taconic Farmsから入手した週齢6〜8週間の雌C3H/HeNマウスを用いた。動物を、経木の寝具を含む加圧滅菌したポリカーボネート製ケージを用いる施設で飼育した。動物は、随意に食物および水を受け取った。人工光は、12/12時間の明/暗サイクル下で提供された。施設の温度は20℃であり、1時間あたり10〜15回、換気した。
マウスrIFN-α(PBL Biomedical Laboratories)、IgG3-IFN-α、および抗HER2/neu-IgG3-IFN-αを、Iodo-Beads(Pierce)を用いる125Iで、製造会社のプロトコールに従って、10μCi/μgまでヨウ素化した。マウスに66μCiの125I標識タンパク質を腹腔内に注射した。125I標識rIFN-α、IgG3-IFN-α、または抗HER2/neu-IgG3-IFN-αの注射後の様々な間隔において、残存放射能を、マウス全身カウンター(Wm. B. Johnson and Associates)を用いて測定した。
C3H/HeNマウスは1000個の38C13/HER2腫瘍細胞を皮下に受けた。処置は、腫瘍負荷後1日目、3日目、および5日目か、または12日目、13日目、および14日目かのいずれかで腹腔内注射によって施された。腫瘍を1日おきに測定し、腫瘍容積(立方ミリメートル)を以下の式を用いて近似した:[長さ(mm)×幅(mm)×幅(mm)]/2 (24)。皮下腫瘍の長さが15mmに達するまで、またはいずれかのマウスが苦しんでいるのが観察され、もしくは瀕死の状態であるように見えるまで、動物を観察した。これらの条件下の動物を、機関の方針に従って人道的に安楽死させた。
簡単には、38C13/HER2細胞を、種々の融合タンパク質で示された時間、処理し、氷冷PBSで洗浄し、溶解緩衝液(0.125% Nonidet P-40、0.875% Brij 97、10mM Tris-HCl(pH 7.5)、2mM EDTA、0.15M NaCl、0.4mM Na3VO4、0.4mM NaF、1mM PMSF、2.5μMロイペプチン、および2.5μMアプロチニン)中、10分間、氷上で溶解した。細胞可溶化液を、10,000×gで4℃で10分間、清澄化した。その後、タンパク質試料を、8%SDS-PAGEゲル上での分離前に試料緩衝液中で煮沸し、ポリフッ化ビニリデン微孔性膜(Millipore)上へ移した。150mM NaCl、50mM Tris-HCl(pH 7.6; TBS)中3%BSAで室温で1時間、ブロッキングした後、ブロットを、示された一次抗体で4℃で一晩、探索した。その後、ブロットをTBS中0.05%Tween 20で室温、3回、洗浄し、HRPを結合した適切な二次抗体とインキュベートし、ペルオキシダーゼ触媒ECL検出システム(ECL; Pierce)によって検出した。ポリクローナルウサギ抗ホスホSTAT1についてはCell Signaling Technologyから入手した。ポリクローナルHRP結合ロバ抗ウサギIgGはAmersham Biosciencesから入手した。ポリクローナルウサギ抗GAPDHはAbcamから入手した。
インビトロ研究については両側スチューデントt検定を、動物生存曲線についてはログランク(Mantel-Cox)解析を用いて、統計解析を行った。
抗HER2/neu-IgG3-IFN-αの作製および特徴づけ
C6MH3-B1(20)可変領域を有する抗HER2/neu-IgG3の構築および発現は、以前に記載されている(23)。成熟マウスIFN-αのアミノ末端を、抗HER2/neu-IgG3のカルボキシル末端に、可動性[(Gly4)Ser]3(配列番号31)リンカーで隔てて、融合させた(図2A)。HER2/neu特異性を欠く同一の融合タンパク質、IgG3-IFN-αを、C6MH3-B1 L鎖を4D5(rhuMab HER2、Herceptin; Genentech)L鎖と置換することによって構築した。プロテインGを用いて培養上清から精製したタンパク質を、非還元および還元条件下でのSDS-PAGEによって分析した(図2B)。還元剤の非存在下で、抗HER2/neu-IgG3(図2B、レーン1)は、170kDaの分子量で移動し、一方、抗HER2/neu-IgG3-IFN-α(図2B、レーン2)およびIgG3-IFN-α(図2B、レーン3)は210kDaであり、2分子のマウスIFN-αが付着した完全なIgG3(図2A)について予想されるサイズである。還元剤での処理後、25kDaの分子量で移動するL鎖が、これらのタンパク質について見られる(図2B、レーン4〜6)。しかしながら、抗HER2/neu-IgG3は、60kDaの分子量をもつH鎖を有し(図2B、レーン4)、一方、IgG3-IFN-α(図2B、レーン5)および抗HER2/neu-IgG3-IFN-α(図2B、レーン6)は、予想通り、80kDaの分子量をもつH鎖を有する。レーン1(図2B)における下部のバンドは、プロテインGカラムにも結合したウシIgGである;ウシH鎖およびL鎖もまたレーン4(図2B)に見られ、より少ない程度で、レーン5および6(図2B)に見られる。FPLC分析により、IgG3-IFN-α融合タンパク質が溶液中、単量体として存在することが示された(データ示さず)。
抗HER2/neu-IgG3および抗HER2/neu-IgG3-IFN-αの両方が、高レベルのヒトHER2/neuを発現するCT26/HER2細胞を結合したが、IgG3-IFN-αはCT26/HER2を弱く結合した(図2C)。IL-1、IL-2、IL-6(25)、およびIFN-α(26)を含む多くのサイトカインは、ヘパリンと相互作用することが示されている。IgG3-IFN-αとCT26/HER2との間の弱い相互作用がヘパリン結合のせいであるかどうかを決定するために、タンパク質を、CT26/HER2への添加前にヘパリンとインキュベートした。ヘパリンは、IgG3-IFN-αのCT26/HER2細胞への結合を阻害したが、抗HER2/neu-IgG3および抗HER2/neu-IgG3-IFN-αの結合を阻害しなかった(図2C)。
抗HER2/neu-IgG3-IFN-αのインビボ抗腫瘍活性を測定するために、同系のマウスに1×103個の38C13/HER2腫瘍細胞を皮下に接種し、腫瘍負荷後1日目、3日目、および5日目に異なる用量のタンパク質の腹腔内投与によって処置した(図3A〜3B)。2.5μgのIgG3-IFN-αで処置されたマウスは、腫瘍成長のいくらかの退縮を示し、8匹のマウスのうちの1匹(13%)が50日後、生きていた(図3A)。しかしながら、腫瘍特異的抗体を用いるIFN-αの腫瘍へのインビボターゲティングは、その抗腫瘍効果を劇的に向上させた。2.5μg(図3A)の抗HER2/neu-IgG3-IFN-αで処置された全てのマウスは、腫瘍負荷後50日目において、腫瘍がないままであり(PBS対照と比較して、p=0.0048)、処置されたマウスのいずれも毒性の徴候を示さなかった。このように、IFN-αの腫瘍細胞表面へのターゲティングは、非特異的抗体に連結したIFN-αと比較して著しい抗腫瘍活性を生じた(p=0.007)。ターゲティング化抗HER2/neu-IgG3-IFN-αは、より低い用量が用いられた場合も、強力な抗腫瘍活性を示し続けた。1μg(図3B)の抗HER2/neu-IgG3-IFN-αで処置された8匹のマウスのうちの7匹(88%)が、50日後、腫瘍がないままであった。好対照的には、このより低い用量において、IgG3-IFN-αで処置されたマウスは、PBSで処置されたマウスと類似した腫瘍成長を示し(p=0.183)、8匹のマウスのうちの1匹のみ(13%)が生き残った。処置を5μgの3回投与まで増加させた場合、抗HER2/neu-IgG3-IFN-αおよびIgG3-IFN-αの両方は、腫瘍成長を阻止するのに効果があり(データ示さず)、疑う余地無く、38C13細胞がIFN-α処置に感受性がある(21、27、28)という事実を反映している。5μgの抗HER2/neu-IgG3抗体で処置されたマウスにおける腫瘍成長は、PBS対照と同じであり、抗体単独ではインビボで抗腫瘍効果を生じないことを示唆している(データ示さず)。これらの結果により、腫瘍特異的抗体によるIFN-αの腫瘍細胞へのターゲティングが、その効果を劇的に増強し得ることが示され、それは、低用量が投与された場合に最も明らかに見られた。重要なことには、この抗腫瘍活性は、いかなる明らかな毒性もなく、達成することができる。
上記のように、本発明者らは、非腫瘍特異的抗体に融合したIFN-αが抗腫瘍活性を示すことを見出した。その抗腫瘍活性を可溶性rIFN-αのそれと比較するために、マウスに1×103個の38C13/HER2腫瘍細胞を皮下に接種し、腫瘍負荷後1日目および3日目に9600U(4μg)のIgG3-IFN-αまたは9600UのrIFN-αの腹腔内投与によって処置した(図4A)。9600UのIgG3-IFN-αで処置された全てのマウスは腫瘍成長の遅延を示し、マウスの75%が、腫瘍負荷後50日目で、腫瘍がないままであった(p=0.027)。対照的に、同数のユニットのrIFN-αで処置されたマウスは、それらの腫瘍成長パターンにおいてPBS対照と統計的に異ならなかった。
IFN-αは、免疫応答の活性化および腫瘍に対する直接的な細胞傷害性を含む複数の活性を有する。抗HER2/neu-IgG3-IFN-αまたはIgG3-IFN-αのいずれを用いても見られる抗腫瘍効果の可能性のある機構を調べるために、腫瘍がないままであった8匹のマウス(図3A参照)に、1×103個の38C13/HER2腫瘍細胞を負荷した。驚くべきことに、全てのマウスは未処置マウスと似て、すぐに巨大腫瘍を発生した(データ示さず)。これらの結果は、低腫瘍量のこれらの実験条件下で、IFN-α融合タンパク質は防御適応免疫応答を惹起しなかったが、代わりに、IFN-α融合タンパク質の強力な抗腫瘍活性が、自然免疫系によるか、または腫瘍細胞への直接的な細胞傷害効果によるかのいずれかで媒介されることを意味している。
IFN-αシグナル伝達は、いくつかの腫瘍細胞系においてアポトーシスを誘導することができる。アポトーシスが、本発明者らが観察した抗増殖効果に寄与したかどうかを決定するために、種々のタンパク質で処理した38C13/HER2細胞を、アネキシンV親和性アッセイを用いて細胞膜の内層から外層へのホスファチジルセリンの転位置についてアッセイした(30)。破壊された細胞膜を有する細胞に入り、DNAに結合するPIによって、死細胞が染色された。PBS対照と比較して、抗HER2/neu-IgG3での処理後、死細胞(アネキシンV/PIブライト(bright)、2%)または初期アポトーシス細胞(アネキシンVブライト、3%)の数の増加はなかった(図5C)。対照的に、細胞をIgG3-IFN-αで処理した場合、死細胞(21%)および初期アポトーシス細胞(6%)の数の有意な増加があった。抗HER2/neu-IgG3-IFN-αでの処理は、死細胞(33%)および初期アポトーシス細胞(16%)の数の両方のさらなる増加を生じた。これらの結果により、IFN-αが38C13/HER2腫瘍細胞のアポトーシスを誘導できること、およびIFN-αを腫瘍細胞にターゲティングすることがこの効果を顕著に増加させ得ることが示された。
IFN-α受容体の結合は複数のSTATタンパク質の活性化を惹起することができるが、STAT1は、IFN-α依存性シグナル伝達を媒介するにおいて必須の役割を果たしている(32)。IFN-α融合タンパク質が38C13/HER2においてIFN-αシグナル伝達を惹起するかどうか、およびIFN-αを腫瘍にターゲティングすることがこの効果を増大することを研究するために、処理後のSTAT1のリン酸化を調べた。図6A〜6Cに示されているように、抗HER2/neu-IgG3-IFN-αおよびIgG3-IFN-αは、38C13/HER2において頑強なSTAT1リン酸化を惹起し、STAT1リン酸化は10分間で8倍に増加した。しかしながら、抗HER2/neu-IgG3-IFN-αにより誘導されたSTAT1のリン酸化は、より長い時間、持続し、抗HER2/neu-IgG3-IFN-αで処理された細胞において30分間目、60分間目、および90分間目に、さらに多いSTAT1リン酸化が見られた。これらの結果により、IFN-α融合タンパク質が、38C13リンパ腫細胞においてIFN-αシグナル伝達を誘導することができ、IFN-αを腫瘍細胞にターゲティングすることがこの効果を増大することが示された。
抗HER2/neu-IgG3-IFN-αが、38C13/HER2腫瘍細胞に対して強力な細胞傷害性を示したため、本発明者らは、抗HER2/neu-IgG3-IFN-αが38C13/HER2樹立腫瘍に対して効果があるかどうかを調べた。同系のマウスに1×103個の38C13/HER2腫瘍細胞を皮下に接種し、腫瘍負荷後12日目、13日目、および14日目に、5μg(図7)の示されたタンパク質での処置を腹腔内に施した。12日目における平均腫瘍サイズは100mm3であり、PBSまたは10μgの抗HER2/neu-IgG3(データ示さず)での処置は腫瘍成長を抑制しなかった。5μgのIgG3-IFN-αでの処置は、腫瘍成長を抑制することにいくらかの効果を示した;しかしながら、全てのマウスは巨大腫瘍を発生し、腫瘍負荷後32日目において、それらのいずれも生き残れなかった。対照的に、5μgの抗HER2/neu-IgG3-IFN-αで処置された全てのマウスは、腫瘍成長を遅らせ、8匹のマウスのうち3匹が完全な腫瘍退縮を有し、腫瘍負荷後50日目において腫瘍がないままであった(抗HER2/neu-IgG3-IFN-α対PBS、p=0.0001;抗HER2/neu-IgG3-IFN-α対IgG3-IFN-α、p=0.063)。このように、IgG3-IFN-αおよび抗HER2/neu-IgG3-IFN-αの両方が抗腫瘍活性を示したが、抗HER2/neu-IgG3-IFN-αが、腫瘍成長を遅らせるのにより効果的であり、完全な腫瘍退縮が、抗HER2/neu-IgG3-IFN-αで処置されたマウスにおいてのみ観察された。処置用量が10μgの融合タンパク質まで増加した場合、抗HER2/neu-IgG3-IFN-αかまたはIgG3-IFN-αのいずれかで処置されたほとんど全てのマウスは、完全な腫瘍退縮を有し、50日後、腫瘍がないままであった。
rIFN-αはB細胞リンパ腫および多発性骨髄腫に対して活性を示しているが、不定の効力および全身毒性がその有用性を制限している(33)。本研究は、IFN-αを抗体に融合させることが腫瘍に対するその効力を向上させ、IFN-αが腫瘍特異的抗体によって腫瘍細胞にターゲティングされた場合、さらなる向上が見られることを実証している。この抗腫瘍効力は、いかなる明らかな毒性もなく、観察される。これらの研究により、IFN-αの腫瘍特異的抗体との融合が、B細胞リンパ腫の処置に効果的な生物学的作用物質を生み出す可能性があることを示唆している。
抗CD20-IFN-α融合タンパク質
序論
本発明者らの最初の研究により、IFN-αに連結した抗HER2/neuに関する融合タンパク質はHER2/neu発現リンパ腫の処置に効果的な治療用物質であることが示された。本発明者らは、IFN-αの抗CD20との融合がCD20発現リンパ腫を処置するための効果的な治療用物質であろうことを示すためにこれらの研究を広げることにした。CD20は、事実上全てのリンパ腫上に存在する。しかしながら、HER2/neuが多くの癌に発現していることに留意すべきであり、抗HER2/neu融合タンパク質がこれらを処置するのに効果的であろうことが予想されるであろう。抗CD20融合タンパク質において、本発明者らは、融合タンパク質におけるIFN-αが、腫瘍細胞に対して直接的な細胞傷害効果を発揮すること、および抗腫瘍免疫応答を誘発するのを助けることの両方を予想している。
ヒトκ軽鎖およびγ3重鎖を有するキメラ抗体の産生のために、抗CD20(リツキシマブ)についての可変領域を増幅し、発現ベクターにクローン化した。タンパク質が産生され、CD20を認識するその能力を、フローサイトメトリーおよびヒトB細胞系Daudiを用いて調べた。図8に示されているように、組換えタンパク質は、組換えIgG1であるリツキシマブと同じくらいよく結合する。
a. 融合タンパク質の設計
融合タンパク質を作製する本発明者らの最初の試みにおいて、本発明者らは、(Gly4Ser)3(配列番号31)からなる可動性グリシン-セリンリンカーを用いてIFN-αをヒトIgG3遺伝子のカルボキシ末端に連結した。重鎖は図9に図式的に示されている。
融合タンパク質がCD20を認識するかどうかを決定するために、CD20を発現するヒト細胞系DaudiをRituxan、抗DNS/IgG3-hu-IFN-α、または抗CD20/IgG3-hu-IFN-αとインキュベートした。抗CD20/IgG3-hu-IFN-αは、Rituxanより良く結合した(図12)。いずれのCD20特異的タンパク質より低かったとはいえ、抗DNS/IgG3-hu-IFN-α融合物もまたいくらかの結合を示した。本発明者らは、抗DNS/IgG3-hu-IFN-αの結合、およびRituxanと比較した抗CD20/IgG3-hu-IFN-αの結合の増強は、hu-IFN-α部分が、Daudi細胞上に発現したIFN受容体に結合するためであると仮定する。
hu-IFN-αタンパク質の抗ウイルス活性を評価するために、HeLa細胞を2×105個の細胞/mlで播種し、融合タンパク質またはRoferon(組換えヒトインターフェロン2a)の2倍段階希釈で24時間、処理した。その後、細胞に、4000pfu/100μlの濃度のVSV(水疱性口内炎ウイルス)を感染させた。72時間後、細胞を0.1%クリスタルバイオレットで染色した。ウイルス感染に対する防御は、0.1%クリスタルバイオレットで染色し、スポット濃度計を用いて各ウェルにおける色素の量を測定することにより感染後生存している細胞を定量化することによるか、またはプラークの数をカウントすることによるかのいずれかで、決定した。両方のアッセイにおいて、融合タンパク質は、有意なIFN-α活性を有したが、Roferonと比較して約100分の1に低下していた。
融合タンパク質によるCD20を発現するリンパ腫細胞の成長抑制/死滅を評価するために2つの方法を用いた。これらの実験について、天然でCD20を発現するヒト細胞系、Daudiを用いたことに留意されたい。第一アプローチにおいて、Daudi細胞を、様々な濃度のIFN-α、抗体、または融合タンパク質と72時間、インキュベートし、CellTiter 96 AQueous細胞増殖アッセイを用いて成長抑制を評価した(図14)。抗ウイルスアッセイにおいてはより低いIFN-α活性を示したが、抗CD20/IgG3-hu-IFN-αおよびRoferonは、リンパ腫成長を抑制する同じような能力を示し、IFN-αをターゲットすることがその細胞傷害効果を増強することを示唆した。抗CD20/IgG3+Roferonは、Roferon単独と比較して活性の増強を示さなかった。抗DNS/IgG3-hu-IFN-α、Rituxan、および抗CD20/IgG3のみは、用いられた最高の濃度でいくらかの成長抑制を示した。融合タンパク質は、このアッセイで細胞成長を防ぐことにおいて、Rituxanより活性が高いことは注目されるべきである。
簡単に上記で述べられているように、John Timmerman博士の研究室では、ヒトCD20を発現し、同質遺伝子的なC3H/HeJマウス中で成長するだろう、マウスリンパ腫、38C13-CD20を開発している。この細胞系を利用できることによって、本発明者らの融合タンパク質のインビボ効力を調べることが可能になる。38C13-CD20細胞を、様々な抗体および融合タンパク質と48時間、インキュベートした。その後、アネキシンVおよびPIで細胞を染色し、フローサイトメトリーを用いてそれらを調べることによって、死滅およびアポトーシスを測定した。タンパク質を100pMの濃度で用いた場合(データ示さず)、組換えmIFN-αおよび抗CD20-IgG3-mIFN-αの両方は、アポトーシスを引き起こすのに非常に効果的であり、抗CD20-IgG3-mIFN-αが組換えmIFN-αより幾分、効果が高かった。38C13-CD20細胞を抗DNS-IgG3-mIFN-αまたはRituxanで処理することによっていくらかのアポトーシスが誘導された。この濃度での抗CD20/IgG3での処理は細胞生存率に効果を生じなかった。処理濃度を10pMまで低下させた場合(図16)、組換えmIFN-αおよび抗CD20/IgG3-mIFN-αは、アポトーシスを引き起こすのに有効であり続け、抗CD20/IgG3-mIFN-αが組換えmIFN-αより効果が高かった。抗DNS-IgG3-mIFN-αでの処理後、アポトーシスが少しだけ見られ、抗CD20-IgG3-mIFN-αを用いるIFN-αのターゲティングが、より効果的な治療剤をもたらすことを示した。この濃度でのRituxanは、ほとんどアポトーシスを引き起こさず、抗CD20-IgG3/mIFN-α融合タンパク質の非融合型抗CD20抗体に対する優位性を示した。先と同様に、抗CD20/IgG3での処理は、細胞生存率に効果を生じなかった。1pMの処理用量において、抗CD20-IgG3-mIFN-αだけが、38C13-CD20にアポトーシスを誘導した(データ示さず)。0.1pMの用量において、いずれの処理もアポトーシスを誘導しなかった(データ示さず)。
a. 抗CD20-IgG1-mIFNαおよび抗CD20-IgG1-hIFNα
最初のタンパク質については、IFN-αがヒトIgG3バックボーンに融合しているように作製した。RituxanはIgG1である。免疫グロブリンバックボーンが融合タンパク質の性質に影響するかどうかを決定するために、今度は、m-IFN-αおよびhu-IFN-αがIgG1に融合している融合タンパク質を作製した。それらは、予想された分子量であった。
GlySerリンカーが、配列A(EAAAK)2A(配列番号33)を有するリンカーと置換されている融合タンパク質を作製した。この配列は、αヘリックスとして折り畳まれるよう企てられる。
ヒトCD20を発現するようにTimmerman研究室によって形質導入された38C13リンパ腫をこれらの研究に用いた。38C13は、同質遺伝子的C3H/HeJマウスで成長する侵襲性リンパ腫である。形質導入体、38C13-CD20は、同じ成長特性を示す。したがって、免疫コンピテント動物において融合タンパク質媒介性防御を調べることが可能である。
0日目、マウス(4匹の群)に5000個の38C13-CD20細胞を皮下注射した。1日目、2日目、および3日目、それらに、HEPES緩衝生理食塩水(HBSS)または0.4μg、2μg、もしくは10μgの抗CD20-m-IFN-αでの処置を静脈内に施し、腫瘍成長をモニターした。20日目までに、HBSSで処置された動物の全部が大きな腫瘍を有し、屠殺するほかなかった。対照的に、10μgの融合タンパク質で処置された動物において腫瘍成長は見られなかった;20日目の後、0.4μgの融合タンパク質で処置された4匹の動物のうち3匹、および2μgで処置されたマウスの1匹において腫瘍が成長し始めた。その結果より、抗CD20/IFN-α融合タンパク質が、インビボの腫瘍成長を抑制することにおいて、および生存を増加させるにおいて、非常に効果的であることが示された(例えば、図22参照)。
0日目、C3H/HeJマウスに5000個の38C13-CD20細胞を接種した。5日目、6日目、および7日目、それらを、HBSS、または10μgの抗CD20-IgG1(293T細胞で産生)、抗CD20-IgG3、リツキシマブ、もしくは抗CD20-IgG3-mIFNαで処置した。それらを、腫瘍成長および生存についてモニターした(例えば、図23参照)。抗CD20/IgG3-mIFNαは、中程度のサイズの腫瘍の成長を防ぐにおいて、リツキシマブ、抗CD20/IgG3、または抗CD20/IgG1よりずっと効果が高かった。
0日目、C3H/HeJマウスに5000個の38C13-CD20細胞を接種し、5日目、6日目、および7日目、10μgの抗CD20-IgG3、10μgの抗CD20-IgG3+mIFN-α(融合タンパク質内と同じモルであるように選択された用量)、抗DNS-IgG3-mIFNα、または抗CD20-IgG3-mIFNαで処置し、腫瘍成長および生存について追跡した(例えば、図24参照)。抗CD20-IgG3-IFNαは、腫瘍成長を有意に遅らせ、生存を促進し、抗体の結合部位を用いてIFNαを腫瘍にターゲティングすることが、融合したIFNαをターゲットしない融合タンパク質(抗DNS-IgG3-IFNα)、または共有結合していないIFNαと共での抗CD20の注射(抗CD20-IgG3+mIFN-α)のどちらよりもそれを効果的な治療用物質にさせることを示した。
8匹のC3H/HeJマウスの群に、5000個の38C13-CD20細胞を接種し、8日目、9日目、および10日目、100μgの抗CD20-mIFNαまたはHBSSで処置した。マウスを腫瘍成長(図25参照)および生存(図26参照)についてモニターした。抗CD20-mIFNαを接種されたマウスは、生存の向上を示す(図26)。
Claims (22)
- 癌細胞上の抗原に結合する全長抗体に連結したインターフェロンαを含むキメラ構築物であって、前記全長抗体が、アミノ酸配列AEAAAKEAAAKAGSからなるタンパク質分解抵抗性ペプチドリンカーによって前記インターフェロンαに連結した抗CD20抗体であるか、または前記全長抗体が、アミノ酸配列SGGGGSからなるタンパク質分解抵抗性ペプチドリンカーによって前記インターフェロンαに連結した抗HER2抗体であり、癌細胞と接触した際に、前記癌細胞の殺傷または成長若しくは増殖の抑制を引き起こす、キメラ構築物。
- 前記抗体が全長抗CD20抗体であり、前記ペプチドリンカーがアミノ酸配列AEAAAKEAAAKAGSからなる、請求項1に記載の構築物。
- 前記抗体がリツキシマブ(登録商標)の相補性決定領域を含む、請求項2に記載の構築物。
- 前記抗体がリツキシマブ(登録商標)の可変領域を含む、請求項2に記載の構築物。
- 前記抗体がリツキシマブ(登録商標)である、請求項2に記載の構築物。
