ことを特徴とする液体組成物を提供する。
・Rは水素、CH3、CX3、CHX2又はCH2X(Xはハロゲン原子である)であり;そして
・B1は単一アミノ酸残基である
ことを特徴とするペプチド化合物を提供する。
「アミノ酸残基」は、N末端が左側に書かれ、C末端が右側に書かれた−NH−CHR−CO−のような構造を有する基を示す。
B1及びB2基中の各アミノ酸残基は、構造−NH−(CH(R))n−C(=O)−〔ここでn=1〜2(好ましくは1)であり、そしてRは次の基:C1〜C6アルキル;フェニル;C7〜C9アルキルアリール;C4〜C8シクロアルキルから選択された、直鎖状又は分枝状及び/又は環状、置換又は非置換構造から選択される〕を含む天然型の又は非天然型のα−もしくはβ−アミノ酸であることができる。L型とD型の両方が含まれる。
チロシンのようなOH置換フェニルアラニンは比較的親水性のアミノ酸であり、ペプチド中にそれが存在すると、一般に、フェニルアラニン、ロイシン、アラニン、システイン、イソロイシン、メチオニン及びバリンのようなより疎水性のアミノ酸〔これらは全てチロシンの負のハイドロパシー指標(hydropathy index)に比較して正のハイドロパシー指標を有する〕に比較して、ペプチドの溶解性が増加するだろう(Kyte & Doolittle (1982), J. Mol. Biol. 157 (1), pp 105-132)(ハイドロパシー指標が大きければ大きいほどより疎水性のアミノ酸になる)。

N末端保護基は、ペプチド合成に使用することができる任意のアミノ末端保護基であってよい。Gross & Meinhoffer編、The Peptides, 第3巻;3-88 (1981), Academic Press, New York, 1981は、多数の適当なアミン保護基を開示しており、その目的のため参考として本明細書に組み込まれる。
好ましい態様では、本発明のペプチド化合物は液体組成物中のサブチリシンを安定化するため又は阻害するために使用され、該液体組成物は界面活性剤と別の酵素とを更に含んでもよい。
サブチリシンは動物、植物又は微生物起源のものであってよく、化学的に又は遺伝学的に修飾された変異体を包含する。それはセリンプロテアーゼ、好ましくはアルカリ性細菌プロテアーゼであってよい。その例は、Siezen他(Protein Engineering, 1991, 第4巻,第7号,719-737頁)により定義されたI−S群からのサブチリシン型プロテアーゼである。サブチリシンの例は、バチルス由来のもの、例えばサブチリシンノボ(Novo)、サブチリシンカールスバーグ(Carlsberg)、サブチリシンBPN’、サブチリシン309、サブチリシン147、サブチリシン168である(WO 89/06279に記載)。例はWO 1998/020115, WO 01/44452, WO 01/58275, WO 01/58276, WO 2003/006602及びWO 2004/099401中に記載されている。
サブチリシンに加えて、液体組成物はアミラーゼ類、リパーゼ類、セルラーゼ類、マンナナーゼ類、オキシドレダクターゼ類及びリアーゼ類から成る群から選択された第二の酵素を含んで成ることができ;特に好ましいのは第二の酵素がリパーゼである液体組成物である。
様々なペプチドアルデヒドが一般のペプチド合成会社により、全て>80%の純度で製造された。ペプチドアルデヒドを使用前に10 mg/mlの濃度になるようにDMSO中に溶かした。
洗剤A:
次の酵素を含有する参照洗剤を調製した:
洗剤L:
ペプチドアルデヒドZ−GAF−H、Z−GAL−H及びZ−GAY−Hを全て純度>80%で、ペプチド合成により製造した。該ペプチドアルデヒドを使用前に10 mg/mlの濃度になるようDMSO中に溶解した。
酵素を含む次の洗剤Nを調製した:
40℃で1週間後の%残留プロテアーゼ活性:
Claims (12)
- 式B2−B1−B0−Hのペプチドアルデヒドであって、ここで
Hは水素であり;
B0はTyr残基であり;
B1は1個のアミノ酸残基であり;そして
B2はN末端保護基が結合している1又は2個のアミノ酸残基である
ことを特徴とするペプチドアルデヒド。 - B1がAla又はVal残基である、請求項1記載のペプチドアルデヒド。
- B2がN末端保護基と結合している単一のGly, Arg又はLeu残基である、請求項1又は2記載のペプチドアルデヒド。
- B2がベンジルオキシカルボニル(Cbz)と結合している単一残基である、請求項1〜3のいずれか1項記載のペプチドアルデヒド。
- B2がアセチル(Ac)N末端保護基を含んで成る2個のアミノ酸残基から成る、請求項1又は2記載のペプチドアルデヒド。
- B2の1個の残基がPhe, Gly又はLeuであり、B2の2番目の残基がPhe, Gly, Leu, Tyr又はTrpである、請求項5記載のペプチドアルデヒド。
- B1がAla又はVal残基であり;そして
B2がベンジルオキシカルボニル(Cbz)と結合している単一のGly, Arg又はLeu残基である
ことを特徴とする請求項1記載のペプチドアルデヒド。 - B1がAla又はVal残基であり;そして
B2がアセチル(Ac)N末端保護基を含んで成る2個のアミノ酸残基から成り、1個の残基がPhe, Gly又はLeuであり、2番目の残基がPhe, Gly, Leu, Tyr又はTrpである
ことを特徴とする請求項1記載のペプチドアルデヒド。 - 式Z−RAY−H,Ac−GAY−H,Z−GAY−H,Z−RVY−H,Z−LVY−H,Ac−LGAY−H,Ac−FGAY−H,Ac−YGAY−H,Ac−FGVY−H又はAc−WLVY−H(ここでZはベンジルオキシカルボニルでありそしてAcはアセチルである)で表される、ペプチドアルデヒド。
- ペプチドアルデヒドZ−RAY−H,Z−GAY−H,Z−RVY−H又はZ−LVY−H(ここでZはベンジルオキシカルボニルである)である、請求項9記載のペプチドアルデヒド。
- サブチリシンと請求項1〜10のいずれか1項記載のペプチドアルデヒドとを含んでなる液体組成物。
- 界面活性剤を更に含んで成る液体洗剤組成物である、請求項11記載の液体組成物。
