Description
が、バイオリファイナリーの出発中間体の有望な化合物として挙げられており、今後その利用の拡大が大いに見込まれている。
下記の工程(1)〜(3)を含む、3−ヒドロキシ酪酸又はその塩の製造方法:
(1)ハロモナス属に属する好塩菌を無機塩と、単一若しくは複数の有機炭素源を含む培地で好気培養する工程1、
(2)工程1の培養条件を好気培養から微好気培養に変更して前記菌体を培養し、培養液にて3−ヒドロキシ酪酸又はその塩を生産する工程2、
(3)工程2で得られる培養液から、3−ヒドロキシ酪酸又はその塩を回収する工程3。
前記好塩菌がハロモナス・エスピー(Halomonas sp.)KM−1株(FERM BP−10995)である、上記項1〜3のいずれか一項に記載の方法。
(2)工程1の培養条件を好気培養から微好気培養に変更して前記菌体を培養し、培養液にて3−ヒドロキシ酪酸又はその塩を生産する工程2、
(3)工程2で得られる培養液から、3−ヒドロキシ酪酸又はその塩を回収する工程3。
本発明に係る3−ヒドロキシ酪酸又はその塩の製造方法における工程1は、ハロモナス属に属する好塩菌を無機塩と、単一若しくは複数の有機炭素源を含む培地で好気培養する工程である。
本発明の製造方法工程1にて用いる好塩菌は、下記の(i)又は(ii)のいずれかによって示されるハロモナス属に属する好塩菌を用いればよい。
(i)無機塩と単一若しくは複数の有機炭素源を含む培地にて好気的に増殖し、3−ヒドロキシ酪酸又はその塩を菌体外の培地中に生産させることを特徴とする好塩菌。
(ii)無機塩と単一若しくは複数の有機炭素源を含む培地にて好気的に増殖し、PHBを自らの菌体内にて蓄積した後、微好気条件下で3−ヒドロキシ酪酸又はその塩を菌体外の培養液にて生産することを特徴とする好塩菌。
工程1にて用いる培地は、無機塩と単一若しくは複数の有機炭素源に追加した培地である。培地のpHは特に限定されないが、上述した好塩菌の生育条件を満たすpHであることが好ましく、具体的にはpH5〜12程度にすればよい。より好ましくはpH8.8〜12の培地である。アルカリ性の培地を用いれば、他の菌のコンタミネーションをより効果的に防止することができるので好ましい。
工程1における上述のハロモナス属に属する好塩菌の培養は、好気培養を採用する。工程1における好気培養は、当該菌体が増殖し、且つ、該菌体内にPHBが著量蓄積するような条件となる好気培養である限り、特に限定はされないが、例えば、5ml程度の培地に当該好塩菌を植菌し、通常30〜37℃程度、攪拌速度は120〜180rpm程度で1晩振盪しながら前培養を行う。
本発明に係る3−ヒドロキシ酪酸又はその塩の製造方法における工程2は、上記工程1の終了後、前記菌体を微好気培養して、培養液にて3−ヒドロキシ酪酸又はその塩を生産する工程である。
工程2における微好気培養とは、培地中又は培養環境を完全に嫌気条件下にするのではなく、積極的に酸素の通気を行わない方法で培養することである。
本発明に係る3−ヒドロキシ酪酸又はその塩の製造方法における工程3は、上記工程2にて得られた培養液から、3−ヒドロキシ酪酸又はその塩を回収する工程である。ここで、回収とは工程2によって得られる培養液中に3−ヒドロキシ酪酸又はその塩が存在している時に上述の工程2の培養を停止し、3−ヒドロキシ酪酸又はその塩を含む培養液と、上記好塩菌体を分離することである。
培養液にて生産される3−ヒドロキシ酪酸又はその塩を測定するため、非特許文献5に記載されたポリヒドロキシアルカノエート(PHAs)分析の手法を応用し、以下の実験を行った。
菌体内に蓄積されたPHBの蓄積量を測定するため、非特許文献2に記載の手法を用い以下の実験を行った。
ハロモナス属に属する好塩菌(ハロモナス・エスピーKM−1株、ハロモナス・パンテラリエンシス〔ATCC 700273〕及びハロモナス・カンピサリス〔ATCC 700597〕)を、プレート培養より、16.5mm径の試験管に5mlの上記SOT5改培地(この場合、炭素源として1w/v%グルコース等を含むものを用いた)を加え、37℃で1晩振盪培養した。
プレ培養した各種ハロモナス属に属する好塩菌体0.2mlを、100ml容の三角フラスコに入れた上記SOT改5培地20mlに混合して植菌し、シリコセンをした。これを、33℃で撹拌速度を200rpmとなる条件にて振盪培養し、24時間後から、およそ12時間おきに培養液を0.5mlずつ回収して、OD600、乾燥菌体重量、PHB含有量、及び上清中の3−ヒドロキシ酪酸又はその塩の量を測定した。
また、培養時の窒素源については、ある程度の濃度範囲であればPHBの蓄積に差がないことは、特許文献3,5に示すとおりである。
さらに、別のハロモナス属に属する好塩菌においても同様の傾向を示すことは、図6,7,8,9,10に示すとおりである。
Claims (4)
- 下記の工程(1)〜(3)を含む、3−ヒドロキシ酪酸又はその塩の製造方法:
(1)ハロモナス属に属する好塩菌を無機塩と、単一若しくは複数の有機炭素源を含む培地で好気培養する工程1、
(2)工程1の培養条件を好気培養から微好気培養に変更して前記菌体を培養し、培養液にて3−ヒドロキシ酪酸又はその塩を産生する工程2、
(3)工程2で得られる培養液から、3−ヒドロキシ酪酸又はその塩を回収する工程3。 - 工程2にて得られる培養液1L当り、3g以上の3−ヒドロキシ酪酸又はその塩が含まれることを特徴とする請求項1に記載の方法。
- 有機炭素源がグリセロールまたは廃グリセロールである請求項1又は2のいずれか一項に記載の方法。
- 前記好塩菌がハロモナス・エスピー(Halomonas sp.)KM−1株(FERM BP−10995)である、請求項1〜3のいずれか一項に記載の方法。
