Description
本願は、米国特許仮出願第61/486,523号(2011年5月16日出願)及び米国特許仮出願第61/441,716号(2011年2月11日出願)に優先権を主張する米国特許出願第13/367,747号(2012年2月7日出願)一部係属出願である。本願はまた、米国特許仮出願第61/815,073号‘2013年4月23日出願)及び米国特許仮出願第61/880,294号(2013年9月20日出願)にも優先権を主張する。これら先行出願の各々は、その全体が参照により含まれる。
本発明は、腸溶性被覆されたマルチ粒子組成物に、より詳しくはペパーミントオイルを含有する腸溶性被覆されたマルチ粒子組成物に係る。
これらの原則を具体化するマルチ粒子組成物は、複数の個々の腸溶性被覆されたコアを有する。前記コアは、微結晶性セルロース系ゲル内に分散されたペパーミントオイルを含有する疎水性相と、ヒドロゲルを含有する親水性相とを包含する。微結晶性セルロースは、ペパーミントオイル用放出制御ポリマーとして機能し、服用量がダンプしないようし、且つコアが処理されている際、ペパーミントオイルを安定化する。
微結晶性セルロースにより形成されたゲル内に分散されたペパーミントオイルを含有する疎水性相と、ヒドロキシプロピルメチルセルロース及び水を含有する親水性相とを包含する湿塊を形成するため、ペパーミントオイル,微結晶性セルロース,ポリマーを形成するヒドロゲル,及び水を混合するステップ;
押出物を形成するため、前記湿塊を押し出すステップ;
前記押出物を個々の湿ったコアに分割するステップ;
乾燥コアを形成するため、前記湿ったコア内の親水性相から水を除去するステップ;及び
前記乾燥コアに腸溶性コーティングを塗布するステップ
を有する。
前記乾燥コアを液体タンパク質性材料で被覆するステップ、及び
前記液体タンパク質性材料を乾燥するステップ、
をさらに有する。液体タンパク質性材料は、ゼラチンを有するかもしれない。液体タンパク質性材料の特定の例は、少なくとも約50%のゼラチンを含有する溶液である。

AVICEL(登録商標)PH 102(FMC Corp., Philadelphia, PA)の名称で市販されている微結晶性セルロース(MCC),METHOCEL(登録商標)A15LV(Dow Chemical Co., Midland, MI)の名称で市販されているメチルセルロース,蒸留されたペパーミントオイル,及び米国薬局方精製水を使用して、コアが調製された。
ゼラチンサブコーティングは、高温で長期間にわたり貯蔵された場合にペパーミントオイルがコアから蒸発及び脱離するのを防いだことを確実にするため、実施例1に記載されたマルチ粒子組成物を引き続き試験した。
Claims (23)
- 複数の個々の腸溶性被覆されたコアを有するマルチ粒子組成物であって、前記コアは、微結晶性セルロース系ゲル内に分散されたペパーミントオイルを含有する疎水性相と、ヒドロゲルを含有する親水性相とを包含する、マルチ粒子組成物。
- 前記個々のコアを被覆し、且つ前記個々のコアをこれらのそれぞれの腸溶性コーティングから分離する連続タンパク質性サブコーティング層をさらに有する、請求項1に記載のマルチ粒子組成物。
- 前記連続タンパク質性サブコーティングは、コアに接着したゼラチンフィルムを有する、請求項2に記載のマルチ粒子組成物。
- 前記連続タンパク質性サブコーティングは、乾燥タンパク質性ゲルを有する、請求項2に記載のマルチ粒子組成物。
- 前記連続タンパク質性サブコーティングは、ペパーミントオイルが腸溶性コーティングと混ざらないように適合される、請求項2に記載のマルチ粒子組成物。
- 前記腸溶性コーティングは、ペパーミントオイルの標準沸点より高いガラス転移温度を有する、請求項2に記載のマルチ粒子組成物。

- 前記腸溶性被覆されたコアは球状であり、直径が3mm以下である、請求項1に記載のマルチ粒子組成物。
- 15%w/w乃至40%w/wのペパーミントオイル,35%w/w乃至75%w/wの微結晶性セルロース,及び2%w/w乃至15%w/wのメチルセルロースを含有する複数の個々の腸溶性被覆されたコアを有するマルチ粒子組成物であって、前記%w/wは、腸溶性被覆されたコアについての%w/wである、マルチ粒子組成物。
- 前記コアを被覆し、且つ前記コアをこれらのそれぞれの腸溶性コーティングから分離する連続タンパク質性サブコーティング層をさらに有する、請求項9に記載のマルチ粒子組成物。

- 前記腸溶性被覆されたコアは球状であり、直径が3mm以下である、請求項9に記載のマルチ粒子組成物。
- マルチ粒子組成物を作成する方法であって、
微結晶性セルロースゲル内に分散されたペパーミントオイルを含有する疎水性相と、ポリマーを形成するヒドロゲル及び水を含有する親水性相とを包含する湿塊を形成するため、ペパーミントオイル,微結晶性セルロース,ポリマーを形成するヒドロゲル,及び水を混合するステップ;
押出物を形成するため、前記湿塊を押し出すステップ;
前記押出物を個々の湿ったコアに分割するステップ;
乾燥コアを形成するため、前記湿ったコア内の親水性相から水を除去するステップ;及び
前記乾燥コアに腸溶性コーティングを塗布するステップ
を有する、方法。 - 腸溶性コーティングを塗布するステップの前に、サブ被覆されたコアを形成するため、
前記乾燥コアを液体タンパク質性材料で被覆するステップ、及び
前記液体タンパク質性材料を乾燥するステップ、
をさらに有する、請求項17に記載の方法。 - 前記液体タンパク質性材料はゼラチンを有する、請求項18に記載の方法。
- 前記液体タンパク質性材料は、少なくとも50%のゼラチンを含有する溶液である、請求項18に記載の方法。
- 乾燥コアを液体タンパク質性材料で被覆するステップは、液体タンパク質性材料を前記乾燥コア上にスプレーするステップを有する、請求項18に記載の方法。
- 前記腸溶性被覆されたコアは球状であり、且つ直径が3mm以下である、請求項17に記載の方法。
- 乾燥コアを形成するため、前記湿ったコア内の親水性相から水を除去するステップは、ペパーミントオイルを実質的に除去することなく達成される、請求項17に記載の方法。
