本発明は、治療ポリペプチド組成物およびそれを必用としている生物に投与して免疫機能をモデュレーションするための方法に関する。特に、本発明は、生物に投与されたとき改良されたインビボ半減時間および有効性を示す、リンホカインポリペプチド部分およびリンホカインレセプター部分を含む有効量の治療タンパク質の投与に関する。
リンパ球は、免疫システム調節に関与するタイプの白血球である。2つの広い種類のリンパ球、即ちT細胞およびB細胞がある。T細胞は細胞性免疫の責任を担い。これに対して、B細胞は体液性免疫(抗体に関する)の責任を担っている。T細胞は、これらのリンパ球が胸腺で成熟するのでこのように名付けられ、そしてB細胞は骨髄で成熟するのでこのように名付けられる。リンパ球は、リンパ系システムにおいてはるかに多く行き渡っておりそしてB細胞、T細胞、キラーT細胞およびナチュラルキラー細胞を含む。B細胞は、病原体に結合して病原体を破壊することができる抗体を作る。CD4+(ヘルパー)T細胞は免疫応答を調整する(それらはHIV感染において欠損するものである)。CD8+(細胞傷害性)T細胞およびナチュラルキラー(NK)細胞は、ウイルスにより感染されているかまたは抗原性配列を示す身体の細胞を殺すことができる。
は、共有されたIL−2RbおよびgCとのヘテロトリマー複合体を形成すると考えられる。IL−15と同様に、IL−15Raは、広く多様な細胞型により発現されるが、必ずしもIL−2RbおよびgCと共にではなく発現されると考えられる。IL−15Ra、IL−2RbおよびgC鎖は、ヘテロトリマーレセプターとして会合すると考えられるけれども、これが生理学的に適切な形態のIL−15レセプターであるかどうかは、推測の域を出ないままである。例えば、IL−15Ra鎖は、IL−15の存在下にIL−2Rb/gCと共沈殿しない。
本発明は、一般に、治療ポリペプチド組成物およびそれを必用としている個体にそれを投与する方法に関する。本発明は、核酸およびそれによりコードされたポリペプチドならびに、本発明の核酸を含有する核酸ベクターを含む関連した組成物、本発明の核酸を含有する細胞系および本発明の治療ポリペプチドに結合する抗体(例えばポリクローナル、モノクローナル、キメラ等)に関する。本発明は、少なくとも1つのリンホカインレセプターもしくはその一部とのプレカップリングさせた複合体において少なくとも1つのリンホカインもしくはその一部を含む治療剤を発生させるための方法にも関する。驚くべきことに且つ意外にも、本発明のプレカップリングされた組み合わせは、IL−15単独の投与で観察されたよりもインビボでのより長い半減時間および高い治療有効性を示すことが観察された。
好ましい態様の組成物および方法の例では、治療ポリペプチドの有用な且つ有利な発生が提供される。好ましい態様では、本発明は、リンホカインもしくはその一部およびリンホカインレセプターもしくはその一部を含む治療ポリペプチド複合体に関する。用語「リンホカインレセプター」または「インターロイキンレセプター」は、それぞれのリンホカインまたはインターロイキンに対する膜貫通レセプターを指し、そしてある態様では、該リンホカインまたはインターロイキンに結合することができる抗体を含むことができる。この状況では、抗体は、リンホカインまたはインターロイキンポリペプチドに対する「レセプター」として有効に機能する。

本発明は、インターロイキンの治療有効性は、インターロイキンをインターロイキンレセプターまたはその可溶性部分にプレカップリングまたは複合体化することにより増強させることができるという驚くべき且つ予想外の発見に基づいている。ある態様では、本発明は、本発明の治療ポリペプチド複合体を形成するための方法を含む。1つの態様では、この方法は、適当な量の少なくとも1つのリンホカインポリペプチドまたはその一部を提供し、適当な量の少なくとも1つのリンホカインレセプターポリペプチドまたはその一部を提供し、適当なpHおよびイオン条件化下に、複合体形成を可能とするのに十分な期間、リンホカインおよびリンホカインレセプターポリペプチドを混合し、そして場合により複合体を濃縮または精製することを含む。複合体のポリペプチドは、例えば、標準方法に従うペプチド合成機を使用し;細胞または細胞抽出物において別々に各成分ポリペプチドを発現させ、次いでポリペプチドを単離しそして精製することにより、形成されうる。場合により、本発明の治療ポリペプチド複合体は、同じ細胞または細胞抽出物において本発明の複合体の両ポリペプチド成分を発現させ、次いで例えば、クロマトグラフィー技術、例えば、リンホカイン部分に対する抗体、リンホカインレセプター部分に対する抗体または複合体に対する抗体によるアフィニティークロマトグラフィーを使用して、複合体を単離および精製することにより、形成されうる。更に、本発明は、フレーム内の(in-frame)且つインターロイキンレセプターまたはその一部と連続した、インターロイキンを含むキメラまたは融合タンパク質の発現を含む。
前記したとおり、本発明は、キメラタンパク質または融合タンパク質も提供する。本明細書で使用された、「キメラタンパク質」または「融合タンパク質」は、他のポリペプチド、例えば、配列番号5〜12またはその一部から選ばれた1つ以上のポリペプチド、に作用的に連結されたポリペプチドを含む。これに対して、配列番号5〜12から選ばれたポリペプチドは少なくとも30%の相同性を有するアミノ酸配列を有するポリペプチドを含む。融合タンパク質内では、ポリペプチドは、配列番号5〜12から選ばれたポリペプチドのすべてまたは一部に相当することができる。1つの態様では、融合タンパク質は、タンパク質の少なくとも1つの生物学的に活性な部分を含む。他の態様では、融合タンパク質は、配列番号5〜12から選ばれた少なくとも1つのタンパク質の少なくとも2つの生物学的に活性な部分を含む。更に他の態様では、融合タンパク質は、配列番号5〜12から選ばれた少なくとも1つのタンパク質の少なくとも3つの生物学的に活性な部分を含む。融合タンパク質の範囲内で、用語「作用的に連結された」は、別個のポリペプチドがN末端またはC末端でお互いにフレーム内で融合されていることを示すことを意図する。

本発明に従えは、本発明の抗原性タンパク質に対して特異的な単一鎖抗体の産生に技術を適合させることができる(例えば、U.S.Pat.No.4,946,778参照)。更に、方法をFab発現ライブラリーの構築のために適合させて((例えば、Huse, et al., 1989 Science 246: 1275-1281)、タンパク質またはその誘導体、フラグメント、アナログまたは相同体に対する所望の特異性を有するモノクローナルFabフラグメントの迅速な且つ有効な同定を可能とすることができる。タンパク質抗原に対するイディオタイプを含有する抗体フラグメントは、(i)抗体分子のペプシン消化により産生されたF(ab’)2フラグメント;(ii)F(ab’)2フラグメントのジスルフィドブリッジを還元することにより発生させたFabフラグメント;(iii)ハパインおよび還元剤による抗体分子の処理により発生させたFabフラグメントおよび(iv)Fvフラグメントを含むがそれらに限定されない当技術分野で知られた技術による産生させることができる。
二重特異的抗体は、少なくとも2つの異なる抗原に対する結合特異性を有するモノクローナル、好ましくはヒトまたはヒト化抗体である。この場合に、結合特異性の1つは、本発明の抗原性タンパク質に対してである。第2結合ターゲットは任意の他の抗原であり、そして有利には細胞表面タンパク質またはレセプターまたはレセプターサブユニットである。
ヘテロコンジュゲート抗体も、本発明の範囲内にある。ヘテロコンジュゲート抗体は、2つの共有結合により結合された抗体からなる。このような抗体は、例えば、免疫系細胞を望まれない細胞にターゲティングするため(U.S.Pat.No.4,676,980)およびHIV感染の処置のため(WO91/00360; WO92/200373; EP03089)に提供された。該抗体は、架橋剤を含む合成タンパク質化学を含む、合成タンパク質化学における既知の方法を使用してインビトロで調製されうることを意図する。例えば、免疫毒素は、ジスルフィド交換反応を使用してまたはチオエーテル結合を形成することにより構築することができる。この目的に適当な試薬の例は、イミノチオレートおよびメチル−4−メルカプトブチルイミデートおよび例えばU.S.Pat.No.4,676,980に開示された試薬を含む。
本発明は、化学療法剤、毒素(例えば、バクテリア、真菌、植物または動物起源の酵素的に活性な毒素、またはそのフラグメント)または放射性同位元素(即ち、放射性コンジュゲート)などの細胞傷害剤にコンジュゲーションされた抗体を含むイムノコンジュゲートにも関する。抗体と細胞傷害剤のコンジュゲートは、種々の二官能性タンパク質カップリング剤、例えば、N−スクシンイミジル−3−(2−ピリジルジチオール)プロピオナート(SPDP)、イミノチオラン(IT)、イミドエステルの二官能性誘導体(例えばジメチルアジピミデートHCl)、活性エステル(例えばジスクシンイミジルスベレート)、アルデヒド(例えばグルタルアルデヒド)、ビス−アジド化合物(例えば、ビス(p−アジドベンゾイル)ヘキサンジアミン)、ビス−ジアゾニウム誘導体(例えば、ビス−(p−ジアゾニウムベンゾイル)−エチレンジアミン)、ジイソシアナート(例えば、トリレン2,6−ジイソシアナート)、およびビス−活性フッ素化合物(例えば1,5−ジフルオロ−2,4−ジニトロベンゼン)を使用して作られる。例えば、リシン免疫毒素は、Vitetta et al., Science, 238:1098(1987)に記載の如く調製されうる。炭素14標識された1−イソチオシアナトベンジル−3−メチルジエチレントリアミンペンタ酢酸(MX−DTPA)は、抗体への放射性核種のコンジュゲーションのための例示的キレート化剤である。WO94/11026参照。
本明細書に開示された抗体をイムノリポソームとして処方することもできる。抗体を含有するリポソームは、Epstein et al., Proc. Natl. Acad. Sci. USA, 82: 3688(1985); Hwang et al., Proc. Natl. Acad. Sci. USA, 77: 4030(1980);およびU.S.Pat.Nos.4,485,045および4,544,545に記載の如き当技術分野で知られている方法により調製される。増強された循環時間を有するリポソームは、U.S.Pat.No.5,013,556に開示されている。

本発明の核酸およびタンパク質は、代謝障害、糖尿病、肥満、感染疾患、食欲不振、癌、神経変性障害、アルツハイマー病、パーキンソン病、免疫障害、造血障害および種々の異常脂血症(dyslipidemias)、肥満と関連した代謝障害、慢性疾患および種々の癌と関連した代謝症候群Xおよび廃棄障害、心筋症、アテローム性動脈硬化症、高血圧、先天性心臓欠陥、大動脈弁狭窄症、心房中隔欠損(atrial septal defect)(ASD)、房室(A−V)管欠損、動脈管(ductus arteriosus)、肺狭窄症、サブ大動脈狭窄症、心室中隔欠損(VSD)、弁疾患、結節状硬化症、強皮症、エリテマトーデス(lupus erythematosus)、肥満、移植、副腎白質異栄養症、先天性副腎過形成(congenital adrenal hyperplasia)、前立腺癌、新生物(neoplasm)、腺癌、リンパ腫、子宮癌、受精能、白血病、血友病、凝固亢進、特発性血小板減少性紫斑病、免疫不全、移植片対ホスト病、AIDS、気管支喘息、リウマチ様疾患および骨関節症、クローン病、多発性硬化症、オーブライト遺伝性骨ジストロフィーおよび他の疾患、障害および状態などを含むが、それらに限定されない種々の疾患に関係する潜在力のある予防および治療用途において有用である。
本発明の方法で使用されるトランスジェニック細胞または動物は、例えば、インターロイキンとインターロイキンレセプターを含むキメラポリペプチドのコピーをコードするトランスジーンを含むことができる。トランスジーンは、ヒトタンパク質を含む、トランスジェニック細胞または動物に対して普通外因性であるタンパク質をコードすることができる。トランスジーンは、異種またはネイティブプロモーターに連結されうる。
IL−15およびIL−15Raの共投与は、インビボでのCD8メモリーT細胞およびNK細胞増殖を駆動する。
IL−15およびリコンビナントマウスIL−15Ra−Fc(rmIL−15Ra−Fc)の共投与がインビボでIL−15活性を媒介することができるかどうかを決定するために、我々は、養子移入モデルを利用して、CD8+T細胞の増殖に対するIL−15の効果を測定した。CD45.1CFSE標識され濃縮された脾臓CD8+T細胞(CD45.1 CFSE labeled enriched splenic CD8+T cells)を正常なCD45.2マウスおよびrmIL−15Ra−Fc(約15μg)に移入した。IL−15単独による処理の4日後、ドナーCD8+T細胞集団の約11%は我々の以前の結果と合致して分裂した(図1a、上部パネル)。極めて対照的に、rmIL−15Ra−Fcに結合したIL−15の同じ量の共投与は、ドナーCD8+T細胞の約60%の増殖をもたらした(図1)。更に、IL−15単独に対応するCD8T細胞の大多数は一度分裂したが、組み合わせ処理に対応する細胞は5〜7回の分裂を受け、細胞数の実質的な増化をもたらした(データは示されていない)。分裂している細胞の大部分は、高いレベルのCD44を発現し、対応する細胞が主としてメモリーCD8+T細胞であるかまたは、CD44がアップレギュレーションされた(図1a、下部パネル)ことを示唆する。重要なことであるが、rmIL−15Ra−Fc単独の投与は、CD8+T細胞の増殖を誘導しなかった(データは示されていない)。注目すべきことに、可溶性形態のrmIL−15RaのIL−15との共投与も、ドナーCD8+T細胞の増強された増殖をもたらしたが、IL−15単独およびrmIL−15Ra−Fcと組み合わされたIL−15に対して中間のレベルをもたらした(データは示されていない)。真正のメモリーCD8T細胞に対する組み合わせた治療の作用を試験するために、我々は、OVAを発現するリコンビナント水泡性口内炎ウイルス(VSV−OVA)による感染により発生させた、CFSE標識されたオブアルブミン(OVA)特異的CD8+メモリーT細胞を養子移入した。上記結果と同様に、組み合わせたIL−15/IL−15Ra−Fc処理に対応する抗原特異的メモリーCD8+T細胞は、IL−15単独で得られた程度よりはるかに高い程度に増殖した(図1b)。
複合体化されたIL−15/IL−15RaはインビボでIL−15活性を大いに増強する
我々は、次いでrmIL−15Ra−FcのIL−15との共投与に対する増殖応答の早期速度論を検討した。CFSE希釈は処理の1日後では無視できるものであったが、2日までにドナーCD8+T細胞集団の約36%が分裂し、第3および第4回の分裂においで認識できる数の細胞があった(図3)。3日までにドナーCD8+T細胞の約59%が分裂し、多くの細胞は5〜6回の分裂にあり、4日までに約73%が分裂し、いくらかの細胞は第7回の分裂にあった。これらの結果および他の結果は、単一用量のIL−15/IL−15Ra−Fcの最大効果は処理の約4日後に達成され、続いてドナーCD8+T細胞がメモリーCD8+T細胞の特徴的な長引く増殖率(protracted rate of proliferation)に入ったことを示した(データは示されていない)。
IL−15Ra−FcのIL−15との共投与は、IL−15効能を顕著に増強する。−1日目に、マウスは、約1.5×106のコンジェニックな、CFSE標識された、CD8濃縮されたリンパ球を静脈内に受け取り、そして0日目にPBS(示されていない)、IL−15(約5μg)または変化する用量のIL−15+IL−15Ra−Fc(約2.5μg+15μg、約0.5μg+3μg、約0.1μg+0.6μg、または約0.02μg+0.12μg)を腹腔内に受け取った。−1日目に、各マウスは、約4.5×106の、コンジェニックな、CFSE標識された、CD8濃縮されたリンパ球を静脈内に受け取り、そして0日目にPBS(示されていない)、IL−15(約0.5μg)+IL−15Ra−Fc(約3μg)または変化する用量のIL−15(約12.5μg、25μg、または37.5μg)を腹腔内に受け取った。CD8+脾細胞をフローサイトメトリーによりCFSE希釈について4日目に解析した。
複合体化されたIL−15/IL−15Raは、IL−15Rbを必用とするトランス提示を介して作用を及ぼす。
複合体化されたIL−15+IL−15Ra−Fcの活性はIL−2Raを必用とするが、応答する細胞によるIL−15Ra発現を必要としない。−1日目に、マウスは、コンジェニックな、CFSE標識された、CD8濃縮されたIL−15Ra−/−リンパ球を静脈内に受け取り、そして0日目にPBS、IL−15(約2.5μg)またIL−15(約2.5μg)+IL−15Ra−Fc(約15μg)で腹腔内で処理された。4日目にCD8+ドナー脾細胞を、CFSE蛍光およびCD122発現について解析した。−1日目に、正常なマウスは、コンジェニックな、CFSE標識された野生型またはIL−2/IL−15Ra−/−脾細胞を静脈内に受け取り、そして0日目に、PBSまたはIL−15(約2.5μg)+IL−15Ra−Fc(約15μg)で腹腔内で処理された。CD8+ドナー脾細胞を、フローサイトメトリーにより4日目にCFSE希釈について解析した。
複合体化されたIL−15/IL−15Raの効果は、対応する細胞型に対する直接または間接の効果により媒介されうる。もし直接ならば、その場合にはターゲット細胞はIL−15R成分(1つまたは複数)を発現することか必要であろうということが予想されうる。これを試験するために、我々は、CFSE標識されたIL−15Ra−/−CD8+T細胞をIL−15Ra−/−ホストに移入しそしてマウスをIL−15または複合体化されたIL−15/IL−15Raで処理した。IL−15は、IL−15Raの不存在下ではトランスプレゼンテーションされることができず、そして増殖を誘導しなかった(図5a)。他方、IL−15/rmIL−15Ra−Fc処理されたマウスからのドナーCD8+T細胞は甚だしく増殖した。更に、主としてナイーブ表現型CD8+T細胞からなるIL−15Ra−/−ドナー細胞は、分裂と共にCD44およびCD122のそれらの発現を漸次に増加させた。応答するT細胞は、複合体化されたIL−15/IL−15Raに応答するためにIL−15Raを必用としなかったので、我々はこの効果の媒介におけるIL−15Rb(CD122)の役割を調べた。このために、我々は、CFSE標識されたCD122+/+またはCD122−/−CD8+T細胞を正常なマウスに移入しそして処理の4日後にCFSE希釈についてドナー細胞を解析した。コントロール細胞は、IL−15/IL−15Ra−Fc処理に応答して活発に増殖したが、CD122−/−ドナーCD8+T細胞は共投与に応答して増殖しなかった(図5b)。これらの結果は、一緒になって、IL−15/IL−15Ra−Fcは、多分γCとの共同においてIL−15Rbとの相互作用を通じて直接トランスプレゼンテーションを介して作用を及ぼすことを示した。
強制されたIL−15トランスプレゼンテーションにより誘導された増殖は、MHCクラスIを必用とするが、IL−7または樹状細胞を必要としない
CD8+T細胞のプレカップリングされたIL−15+IL−15Ra−Fcで推進された増殖は、MHCクラスI発現を必要とするが、IL−7またはDCを必要としない。−1日目に、B6およびβ2m−/−マウスは、正常なB6およびナイーブOT−I−RAG−/−CFSE標識されたCD8+T細胞の混合物を受け取り、そして0日目に、PBSまたはIL−15(約2.5μg)+IL−15Ra−Fc(約15μg)で処理された。−1日目に、IL−7+/−またはIL−7−/−マウスは、コンジェニックな、CFSE標識されたCD8濃縮されたリンパ球を受け取った。0日目に、マウスは、IL−15(約2.5μg)+IL−15Ra−Fc(約15μg)を腹腔内に受け取った。−1日目にB6またはCD11c−DTR骨髄で産生されたキメラは、碑細胞を静脈内に受け取り、そして0日目に、PBSまたはIL−15Ra−Fc(約15μg)+IL−15(約2.5μg)で腹腔内処理された。すべてのマウスを、0、1および3日目にDTで処理した。すべての場合に、ドナーCD+脾細胞を4日目にCFSE希釈について解析した。

IL−15/IL−15Ra免疫治療は、ナイーブT細胞活性化およびエフェクター機能を誘導する

先の実験で、我々は、CD44低ポリクローナルCD8T細胞およびナイープTCRトランスジェニックT細胞が、IL−15Ra−Fcと共投与されるときのIL−15に対して応答することに留意した。ホメオスタティックな条件下に、CD8メモリーT細胞は、ナイーブCD8+T細胞よりもはるかに高いIL−15に対する応答を示すことを考慮して、我々は、複合体化されたIL−15/IL−15Ra−Fcに対するこれらの2つのサブセットの応答を直接比較することを望んだ。そうするために、CFSE標識されたメモリーOT−IおよびナイーブOT−ICD8+T細胞を同じコンジェニックC57BL/6ホストに養子移入しそして増殖をIL−15/IL−15Ra−Fcによる処理の4日後に解析した。驚くべきことに、ナイーブOT−ICD8T細胞は、メモリーOT−ICD8+T細胞と殆ど同じく増殖した。ナイーブOT−I細胞も、コントロール複合体に応答して〜10倍エキスパンションしそしてCD44をアップレギュレーションした。ナイーブT細胞において誘導された活発な増殖に照らして、エフェクター機能が付随的に誘導されたかどうかを確立することは興味深いことであった。この問題を試験するために、我々は、ナイーブOT−ICD8+T細胞をコンジェニックC57BL/6ホストに養子移入し、そしてインビトロキリングアッセイを使用して、IL−15/IL−15Ra−Fcによる処理の4日後または比較のためにオブアルブミンを発現するリコンビナント水泡性口内炎ウイルス(VSV−OVA)による感染の後に抗原特異的溶解活性を測定した。興味深いことに、IL−15/rmIL−15Ra−Fc処理は、ウイルス感染により得られたレベルに類似した、活発な抗原特異的溶解活性の誘導をもたらした。溶解活性に加えて、IL−15/IL−15Ra−FcまたはVSV−OVA感染により活性化されたナイーブOT−ICD8+T細胞の大多数は、ペプチドによるインビトロ再刺激に続いて、高いレベルのIFNgを産生した。この結果は、コントロール(PBS)およびIL−15処理したマウスからIFNgを産生するOT−I細胞の無視できる頻度と対照的であった。従って、IL−15Ra−FcのIL−15との共投与によるナイーブCD8+T細胞におけるエフェクター機能の誘導は、感染により得られた活性化と同様であった。
複合体化されたIL−15/IL−15Ra−FCによるナイーブT細胞の処理は、メモリーCD8+T細胞を発生する。
プレカップリングされたIL−15+IL−15Ra−Fc処理は、ナイーブCD8+T細胞からメモリー細胞を発生させる。1日目に、B6マウスは、約6×106CFSE標識されたナイーブOT−I−RAG−/−細胞を受け取りそして0日目に、PBSまたはIL−15(約2.5μg)+IL−15Ra−Fc(約15μg)で腹腔内処理された。44日後、碑細胞をドナーOT−ICD8+T細胞の百分率(上部パネル)およびCD44およびCD122のOT−I発現(中部および下部パネル)について解析した。
ナイーブT細胞はトランスプレゼンテーションされたIL−15に応答してエフェクター細胞に成長したけれども、これが一過性の効果であるかまたはメモリーT細胞発達(memory T cell development)をもたらしたかどうかは、まだ確められていない。従って、我々は、ナイーブOT−IT細胞移入およびIL−15/IL−15Ra−Fc処理の44日後OT−IT細胞の数および表現型を解析した。この時点で、非処理マウスに比較してIL−15/IL−15Ra投与の後〜5倍高い百分率のOT−I細胞が存在していた。更にこれらの細胞の殆どすべてが高いレベルのCD44およびCD122を発現した。従って、抗原の不存在下ですら、IL−15/IL−15Ra−Fc処理は、メモリーCD8+T細胞の発達を誘導することができた。
マウス。C57BL/6−Ly5.1マウスは、The Jackson Laboratory(Bar Harbor,ME)から購入された。C57BL/6−Ly5.2マウスは、Charls Riverから購入された。OT−Iマウス系は、寛大にもDr.W.R.Heath(WEHI,Parkville,Australia)およびDr.F.Carbone(Monash Medical School,Prahan,Victoria,Austrakia)により提供されそしてRAG−/−バックグラウンド上のC57BL/6−Ly5.2系として維持された。

マウスにおけるIL−15+IL−15Raタンパク質複合体により減少した肝臓癌量。約1×105B6−F1メラノーマ細胞を脾臓内に注射した(肝臓に腫瘍を指向する)。1および7日後に、マウスをPBS(コントロール)、2.5μgIL−15または2.5μgIL−15+IL−15Ra複合体で処理した。接種の14日後、腫瘍を肝臓において計数し、そして脾臓の重量を計った。
