本発明は、多様な医療用途並びに工業的及び環境的衛生用途を含む一般的な衛生用途における使用のための殺菌組成物、方法及びシステム、に関する。本発明の組成物は、抗微生物、抗真菌、抗ウイルス及び抗アメーバ特性を有しており、また抗凝血剤としても作用することができる。エチレンジアミン四酢酸(EDTA)(C10H12N2Na4O8)の特定の塩及び組成物が特定の濃度及びpHレベルで用いられる。典型的な用途は、表面上または溶液中の微生物及び/または真菌生物、またはバイオフィルム等の複雑な形態中の微生物及び/または真菌生物、の存在を阻害し、低減しまたは除去することを含む。典型的な方法は、感染症の発生率を低減するために対象物の表面に抗菌コーティングを備えること、及び、微生物群の増殖を阻害し、または微生物群を低減または除去するために、洗い流す、浸す及び/またはすすぐことによって対象物及び/または表面を殺菌溶液に接触させることを含む。
医療分野等の衛生条件が重要である多くの分野において感染症は重大な問題である。問題となる感染症は細菌、真菌、アメーバ、原生動物及び/またはウイルスによってもたらされる。かつて確立されていた、感染症を予防すること及び/または感染症を低減または除去すること、の両面において難問に直面している。感染する環境は、対象物の表面、流動体及び流動管、及び/またはヒトまたは動物を含む。
本発明は、生理学的pHより高いpH及び処方される濃度において、EDTAの一つまたはそれより多い塩を含む、本質的にEDTAの一つまたはそれより多い塩からなる、または、EDTAの一つまたはそれより多い塩からなる、殺菌組成物を提供する。発明者らは、あるEDTA組成物が強力な殺菌活性を有し、広いスペクトルの抗細菌剤として作用し、そして病原性酵母の多くの菌株に対して抗真菌剤として作用するということを発見した。本発明のEDTA組成物は、病原性バイオフィルム生物を死滅させること、既存のバイオフィルムを減少及び撲滅すること、及びバイオフィルム形成を予防することについて非常に有効である。本発明のEDTA組成物及び組み合わせは、また、抗原生動物活性及び抗アメーバ活性を示す。既存の報告を基にすると、本発明のEDTA組成物にはさらに、抗ウイルス活性が期待できる。
多くの組成物において、安定剤または保存剤としての他の活性成分との組み合わせで、EDTAは低濃度で使用される。本発明の殺菌組成物は、通常、高い濃度のEDTAを含む。好ましくは本発明の殺菌組成物は、溶液の体積分の質量(w/v)として、少なくとも0.01%のEDTA塩を含み、そして15%(w/v)までのEDTA塩を含むことができる。多くの用途に対して、本発明の殺菌組成物は、好ましくは、少なくとも0.1%(w/v)のEDTA塩を含みそして10%(w/v)より少ないEDTA塩を含み、より好ましくは0.1%(w/v)乃至8.0%(w/v)のEDTA塩を含み、最も好ましくは0.1%(w/v)乃至6.0%(w/v)のEDTA塩を含む。後述の例となるな組成物は、水溶液中3.6乃至4.4%(w/v)のEDTA塩を含み、または、水とエタノールの混合物中0.01乃至0.2%(w/v)のEDTAを含む。
寒天希釈法を使用した、EDTAの異なった製剤に対する生物の最小阻止濃度(MIC)及び最小殺菌濃度(MBC)データ
以下に述べる寒天希釈法を使用して、EDTAのいくつかの異なった製剤に対する多様なグラム陽性菌、グラム陰性菌及び酵母生物の最小阻止濃度(MIC)及び最小殺菌濃度(MBC)を確立した。また、EDTA塩の組み合わせについても、多様な生物に対するMIC及びMBCの試験を行った。
寒天の作成
・2リットルの寒天培地をスチームバスに入れ、約1時間(溶けるまで)そのままにした。
・その寒天を50℃に冷却した。
・20個の無菌ガラスビン(125mL)に、それぞれ100mLの寒天培地を配分した。200mg/mLの保存溶液を使用して、これらに0.5、1.0、1.5、2.0、4、6、8、10、15、20、25、30、40、50、60、70、80、90及び100mg/mLの四ナトリウムEDTA(または試験される他のEDTA塩またはEDTA塩の組み合わせ)を加えた。
・無菌のペトリ皿に20mLの寒天を注ぎ、放置した。さらに三つのプレートに注いだ。それらが含んでいるEDTAの濃度をプレートにラベルした。これを各濃度について行った。
・必要になるまで、これらのプレートを4℃の冷蔵庫で保存した。
プレートへの接種
・栄養培養液中で23のグラム陽性菌及び19のグラム陰性菌の培養物を、一晩成長させた。
・リン酸緩衝食塩水(PBS)を用いて、各培養物を106cfu/mLに希釈した。
・自動プレート接種機を用いて、各プレートに21の菌を接種した。
・プレートを37℃で培養した。
・翌日、成長について+または−で記録した。
・MBCを決定するために、無菌ろ紙を使用して、成長したものを最初のプレートから新しいCled寒天プレートに移動した。
・そのレプリカプレートを37℃で一晩、培養した。
・翌日、成長について+または−で記録した。MIC及びMBCを、成長が無かった最も低い濃度として記載した。
修飾したカルガリー装置法(Calgary device method)を使用した、四ナトリウムEDTAに対する生物の最小バイオフィルム撲滅濃度(MBEC)データ
細菌汚染において、バイオフィルム形成は重要な要因である。実効的な殺菌組成物は、好ましくはバイオフィルムの増殖を低減する能力、またはバイオフィルムの形成を予防または阻害する能力を有している。そのため、発明者らは、候補の四ナトリウムEDTA殺菌溶液がバイオフィルムの形成を予防または阻害するかどうかを決定するための試験を行った。四ナトリウムEDTAに対する多様な生物の最小バイオフィルム撲滅濃度(MBEC)を修飾したカルガリー装置法を使用して測定した。カルガリー法についてはOlsen et al.のCanadian Journal of Veterinary Research,66:86−92(2002)及び米国特許6599714に記載されている。方法と結果を以下に述べる。
バイオフィルムの形成
・必要とする生物のミュラーヒントンの一晩の培養液(Muller Hinton overnight broth)100mLを使用した。
・96ウエルのマイクロタイタートレイの総てのウエルに200μlをピペットで加えた。96のピンを持った蓋を設置した。200rpmの速度、37℃で24時間、オービタルシェイカー(orbital shaker)で培養した。
・上記で形成したバイオフィルムを使用した。
・必要な濃度の試験用薬剤250μlを含んだ新しい96ウエルのマイクロタイタートレイに蓋(ピンを持った)を設置した。37℃で1乃至24時間培養した(シェイカーを使用しない)。
・1、3、6及び24時間の間隔で、ねじ回しを挿入してピンをウエルの中に折ることによって、各濃度において蓋から四つのピンを除いた。
・各濃度において、三つのピンをPBSの5ml洗浄液に入れ、そして一度反転させた。
・その三つのピンを3mLのPBSに入れ、そして15分間超音波をあてた。2μlを3xCLEDプレート上に移し、無菌のプラスチック製スプレッダーを使用して引き延ばした。37℃で一晩培養した。翌日、コロニーの数を記録した。
・SEMのために、残っているルーズピン(loose pin)を4%生理食塩水600μlに置いた。
患者の陽性なカテーテルに対するインビトロカテーテルロック処理手順
候補の40mg/ml(4%w/v)四ナトリウムEDTA溶液を用いたカテーテルロック処理手順を発展させ、そして、様々な微生物感染について陽性であったサンプルの患者の血液透析カテーテルに対して使用した。微生物感染が確認されたカテーテルに対して四ナトリウムEDTAを用いたカテーテルロック処理を行い、そして様々な時間でコロニー数を数えた。第一の実験で、総てのカテーテルを4%w/v四ナトリウムEDTA溶液で処理した。一方、第二の実験では、カテーテルを様々な濃度の四ナトリウムEDTA溶液で処理した。四ナトリウムEDTA溶液は、実施例1及び2で述べたようにして調製、保存した。手順と結果を以下に示す。
・1mLの無菌のリン酸緩衝食塩水で各管腔を洗い流すことによって、感染の疑いのために取り除かれた腎臓血液透析カテーテルを選別した。血液寒天プレート上に広げられた1μL及び10μLのアリコートを用いて定量培養を行い、そして培養した。
・選別の後まで4℃でカテーテルを保存し、そして外部管腔をアルコールワイプで消毒した。
・ロック処理試験の前に、バイオフィルムの生育を確実にし、そして感染生物による総ての管腔内表面の全体的なコロニー化を確実にするために、選別された陽性のカテーテルを5mLシリンジを用いて栄養培養液でロックし、そして37℃で一晩培養した。
・一晩培養後、各カテーテル管腔5mLの無菌食塩水で洗い流し、その末端から二つの1cmの片を切り取り、そして各片を1mLの1M無菌塩化カルシウム中に置いた(薬剤の中和のため)。片の一つは走査型電子顕微鏡(SEM)のため、そして他の片は無菌のユニバーサル(universal)容器内での培養のためである。
・培養手順のため、そのユニバーサル容器を室温で15分間、超音波処理用バスに設置し、その後20秒間、ボルテックス撹拌(vortexed)した。
・1μl及び10μlのアリコートを血液寒天プレート上に加え、無菌のプラスチック製L形ロッド(rods)で引き延ばし、そして37℃で一晩培養して定量培養を行った。そして、翌日、コロニーを数えた。
・適当な濃度の四ナトリウムEDTAロック溶液でカテーテルを洗い流し、ロックし、そして37℃で18時間培養した。
・3時間、6時間及び18時間培養後に、カテーテル末端から二つの1cmの片を切り取り1mLの1M無菌塩化カルシウム溶液で中和した。
・各時間間隔で、上記のようにして定量的に数を数えた。そして、一つの片はSEMのために保存した。
四ナトリウムEDTAのアカントアメーバに対する効果、及び四ナトリウムEDTA処理クレブシエラ(Klebsiella)のアカントアメーバに対する効果
数種類のアカントアメーバがヒトに感染する。ヒトに接触するコンタクトレンズ及び他の医療器具の不適切な保存の結果として、しばしばアカントアメーバ感染症が起こる。アカントアメーバは細菌個体を餌にしており、多くの処置に対して耐性である。上記のようにして調製した四ナトリウムEDTAのアカントアメーバ個体群に対する効果を以下のようにして試験した。また、溶媒としてページ食塩水(Pages saline)及び生理食塩水を使用して四ナトリウムEDTA組成物を調製した。クレブシエラと処理した四ナトリウムEDTAのアカントアメーバに対する効果についても、以下の方法によって実験的に試験した。
方法:
・試験前に、Klebsiella edwardsiiを持った新しい血液寒天プレートを37℃で18時間培養した。
・四ナトリウムEDTA(100mg/mL)の保存溶液を用いてページ食塩水中で22mg/mL及び44mg/mLの濃度の溶液を調製した。
・各濃度の溶液9mLを無菌ガラス試験管に入れた。コントロールとして別の無菌ガラス試験管に無菌のページ食塩水9mLを入れた。
・無菌のページ食塩水6mLにKlebsiella edwardsiiを懸濁した。マクファーランド標準(McFarland standard)5に調節した。
・連続した各希釈液及びコントロールに懸濁液1mLを加えた。Klebsiella懸濁液での希釈のため、各濃度は20mg/mL及び40mg/mLである。コントロールは四ナトリウムEDTAを含んでいない。生理食塩水についても総ての濃度で行った。
・ボルテックスで撹拌した。各試験管はマクファーランド0.5でKlebsiellaの懸濁を含んでいる。
・アカントアメーバプレート全体の表面を削り取り、1.5mLのページ食塩水に懸濁し、ボルテックスで撹拌した。
・アカントアメーバ懸濁液200μlを各連続した希釈液及びコントロールに加えた。
・試験管を24時間、30℃の培養器に置いた。
・培養後、各ユニバーサル(universal)を3000rpmで10分間遠心分離した。
・上澄みを捨て、沈殿物を再懸濁した。
・各希釈液及びコントロール10μlを、二組、Klebsiella地の非栄養寒天プレートに加えた。移動を防ぐために各プレートの中心に溝を作り、そして各サイドに試験を行う希釈液10μlを加えた。
・各培養の個所に黒のマーカーペンで印をした。
・プレートを30℃で72時間培養した。
・各培養個所で、10倍の接眼レンズの光学顕微鏡を用いたプレートの直接可視化によってアカントアメーバの成長を観察した。
方法:
・試験前に、Klebsiella edwardsiiを持った新しい血液寒天プレートを37℃で18時間培養した。
・四ナトリウムEDTA(100mg/mL)の保存溶液を用いてページ食塩水中で22mg/mL及び44mg/mLの濃度の溶液を調製した。
・各濃度の溶液9mLを無菌ガラス試験管に入れた。コントロールとして別の無菌ガラス試験管に無菌のページ食塩水9mLを入れた。
・無菌のページ食塩水6mLにKlebsiella edwardsiiを懸濁した。マクファーランド標準5に調節した。
・連続した各希釈液及びコントロールに懸濁液1mLを加えた。Klebsiella懸濁液での希釈のため、各濃度は20mg/mL及び40mg/mLである。コントロールは四ナトリウムEDTAを含んでいない。生理食塩水についても総ての濃度で行った。
・ボルテックスで撹拌した。各試験管はマクファーランド0.5でKlebsiellaの懸濁を含んでいる。
・試験管を37℃で一晩培養した。
・翌日、300rpmで10分間、遠心分離した。上澄みを捨て、新しい10mlの生理食塩水またはページ食塩水を加え、再懸濁し、そして再び遠心分離した。上澄みを捨て、1mlの生理食塩水またはページ食塩水に懸濁した。
・アカントアメーバプレート全体の表面を削り取り、1.5mLのページ食塩水に懸濁し、ボルテックスで撹拌した。
・アカントアメーバ懸濁液200μlを食塩水9mlの入った試験管3本及びページ食塩水3mlの入った試験管3本に加えた。Klebsiellaとの培養で使用されたEDTA濃度のように、各試験管に印を付けた。
・Klebsiellaの再懸濁液1mlをアカントアメーバを含む適切な試験管に加えた。
・試験管を30℃で24時間、培養器中に置いた。
・試験管の別のセットを準備し、上述のようにして、37℃で一晩、EDTAとKlebsiellaを培養した。
・培養後、アカントアメーバを含む各試験管を3000rpmで10分間遠心分離した。
・上澄みを捨て、沈殿物を再懸濁した。
・各希釈液及びコントロール10μlを、二組、Klebsiella(EDTAと培養していない)地の非栄養寒天プレートに加えた。移動を防ぐために各プレートの中心に溝を作り、そして各サイドに試験を行う希釈液10μlを加えた。
・各培養の個所に黒のマーカーペンで印をした。
・プレートを30℃で培養した。
・各培養個所で、10倍の接眼レンズの光学顕微鏡を用いたプレートの直接可視化によってアカントアメーバの成長を観察した。
・アカントアメーバ懸濁液の残りを、新しい生理食塩水または新しいページ食塩水の入った試験管の新しいセットに加えた。
・事前にEDTAと一晩培養したKlebsiellaを洗浄し、再懸濁し、そしてアカントアメーバの入った適切な試験管に加えた。
・試験管を30℃で一晩培養した。
・培養後、各ユニバーサルを3000rpmで10分間遠心分離した。
・上澄みを捨て、そして沈殿物を再懸濁した。
・各希釈液及びコントロール10μlを、二組、Klebsiella(EDTAと培養していない)地の非栄養寒天プレートに加えた。移動を防ぐために各プレートの中心に溝を作り、そして各サイドに試験を行う希釈液10μlを加えた。
・各培養の個所に黒のマーカーペンで印をした。
・プレートを30℃で培養した。
・各培養個所で、10倍の接眼レンズの光学顕微鏡を用いたプレートの直接可視化によってアカントアメーバの成長を観察した。
四ナトリウムEDTA組成物が、微生物のシリコンチューブへの付着または密着を予防するかどうかの確認を行った。シリコンチューブへの微生物の付着及び密着を予防することができれば、バイオフィルムの形成を低減することができる。実験方法と得られた結果を以下に示す。
・管腔内部をふさぐために1cm片のシリコンチューブを溶融したワックスで満たし、そして4℃で固めた。
・コントロールとして、無菌のリン酸緩衝食塩水(PBS)30mLに4片のシリコンチューブを入れた。4%四ナトリウムEDTA30mLに8片のシリコンチューブを入れた。
・30分後、PBS中の4片及び4%四ナトリウムEDTA中の4片を熱ブロック上のきれいな容器に移し、乾燥した。
・残りの4片を30mLの無菌PBSに移して洗浄し、そしてこれまでのように空気乾燥した。
・乾燥後、12片総てを105cfu/mLの生物の混合物(栄養培養液中、37℃でKlebsiella pneumoniae及びCNS細胞を一晩培養したもの)に移し、37℃で培養した。
・30分後、各型の2片を分離し、30mLの無菌PBSで2回洗浄した。これまでのように空気乾燥した。各型の汚染を予防するために、別々に洗浄し別々の乾燥容器を使用した。
・各片を遠心分離管の1mLのPBSに移し、超音波水浴で15分間超音波処理した。
・各試験管を、二組、自動プレート培養器上、プレートに加えた、対数希釈の50μl
・1/10希釈の各試験管を、二組、プレートに加えた
・プレートを37℃で一晩培養した。自動プレート読取器ProtoCOLでコロニー数を読み取った。6時間後についても行った。
四ナトリウムEDTAをエタノールと組み合わせた場合のMBC値の変化
単独のEDTA溶液、単独のアルコール溶液及びEDTA/アルコール溶液の効果を試験するために、四ナトリウムEDTAの濃度(0、0.1、0.5、1、2、3、4及び8mg/ml、w/v)の溶液を水及びエタノールを用いて(最終のエタノール濃度が、水中、0、0.1、0.5、1、5、10、20及び40%になるように)調製した。蒸留水で四ナトリウムEDTAの高濃度の保存溶液を調製し、そして適当なエタノール濃度になるようにその高濃度の保存水溶液にエタノールを加えた。
・栄養培養液中、37℃で一晩、生物を培養した。
・アルコールと四ナトリウムEDTAの保存溶液で96ウエルプレート(一つの培養につき一つ)のグリッドパターンを満たした。0、0.1、0.5、1、5、10、20及び40%のアルコール濃度(v/v)でイソプロピルアルコールを含む水溶液中で、0、0.1、0.5、1、2、3、4及び8mg/mlの四ナトリウムEDTA濃度(w/v)のEDTA溶液を使用した。
・各ウエルは各希釈液150μlと50μlの1x108cfu/mLの生物を含む。
・5分、6時間及び24時間後、96ピンの蓋をプレート上(そして各ウエル内に)に置き、そしてその蓋を各ウエルに300μLの新しい栄養培養液が入った96ウエルに移動することによって、各ウエルを培養した。37℃で一晩培養した。その培養の間、微生物を接種した各プレートを37℃で培養した。
・24時間後、各ウエルの濁度を記録した。
エタノール中の四ナトリウムEDTAの溶解度及びpHに及ぼす影響
エタノール中の四ナトリウムEDTAの溶解度が試験され、そしてアルコール溶媒中の多様な四ナトリウムEDTA溶液のpHが測定された。
・1.5mLサイズのエッペンドルフチューブに10乃至100mgの四ナトリウムEDTAを、二組、量り分けた。74%エタノール1mLを各チューブに加えそして30秒管ボルテックスで撹拌した。
・量り分けた四ナトリウムEDTAの二組のセットに、0.5ml無菌蒸留水を加えボルテックスで撹拌し、そして74%エタノール0.5mlを加えた。
・四ナトリウムEDTAのチューブのそれぞれで、溶解度が観察された時点のpHの試験を行った。

四ナトリウムEDTA溶液に対する121℃でのオートクレーブ処理の影響
使用前に四ナトリウムEDTA溶液を無菌化するためにオートクレーブを使用できるかどうかを判断するために、四ナトリウムEDTA溶液に対するオートクレーブ処理の影響についての試験を行った。
・無菌水中の0、20、80及び100mg/mLの四ナトリウムEDTA溶液、及び50℃の無菌溶融寒天培地中の0、20、80及び100mg/mLの四ナトリウムEDTA溶液を、二組、作成した。
・一組を室温に置き(加熱しない)、他方をオートクレーブした(加熱した)。
・翌日、総ての寒天容器を蒸し器に設置し、40分間で溶融した。
拡散区域の測定:
・穿孔器を使用して、新しい16の血液寒天プレートに二つの穴を開けた。
・0.5マクファーランドのCNSの懸濁液を調製し、無菌綿棒でプレート上に引き延ばして培地を作成した。
・それぞれの四ナトリウムEDTA溶液150μlを、二組、打ち抜いた穴に入れ、37℃で一晩培養した。
・翌日、拡散した区域を測定し、記録した。
EDTAの異なった製剤に対する121℃でのオートクレーブ処理の影響
使用前に多様なEDTA溶液を無菌化するためにオートクレーブを使用できるかどうかを判断するために、EDTA溶液の異なった製剤に対するオートクレーブ処理の影響についての試験を行った。使用された方法と結果を以下に示す。
寒天の作成
・50mLの寒天培地溶液を7つの100mL無菌ガラスボトルに加えた。
・第一のボトルにはEDTA粉末を加えない(0とラベルした)。
・第二のボトルに2000mgのEDTA粉末を加えた(40mg/mL オートクレーブとラベルした)。
・第三のボトルに4000mgのEDTA粉末を加えた(80mg/mL オートクレーブとラベルした)。
・第四のボトルに5000mgのEDTA粉末を加えた(100mg/mL オートクレーブとラベルした)。
・第五乃至七のボトルにはEDTAを加えない(しかし40、80及び100mg/mL 非オートクレーブとラベルした)。室温に置いた。
・これらの各EDTA製剤について試験した。オートクレーブとラベルした総てのボトルを、121℃で20分間、オートクレーブ処理した。
・翌日、総てのボトルをスチームバスに入れ、寒天を注げるように溶融した。
・溶融後、50℃に冷却し、そして非オートクレーブとラベルしたボトルに適切な量のEDTAを加え、総てのボトルについて試験の準備をした。
拡散区域の測定
・穿孔器を使用して、新しい7の血液寒天プレートに二つの穴を開けた。
・0.5マクファーランドのCNSの懸濁液を調製し、無菌綿棒でプレート上に引き延ばして培地を作成した。
・各ボトルの150μlを、2つの別々の打ち抜いた穴に入れ、37℃で一晩培養した。
・各EDTA製剤について実施した。
・翌日、拡散した区域を測定し、記録した。二組の穴を使用し、区域当たり二つの測定を行った。
EDTA塩、塩化カルシウム及びクエン酸ナトリウムのpH値
溶媒として蒸留水を用い、特定の濃度において、多様なEDTA塩、塩化カルシウム及びクエン酸ナトリウムのpH値を測定した。結果を以下に示す。

EDTA溶液の抗凝血特性の確認
以下の方法によってEDTA溶液の抗凝血特性を確認した。
・pHが11.0乃至11.6に調節された、濃度0.5乃至100mg/mLの四ナトリウムEDTA溶液または二ナトリウムEDTA溶液の100μlアリコートをプラスチック製キャップ付チューブに加えた。
・健康なボランティアから採取した新鮮な血液900μLをEDTA溶液の各アリコートに加え、一定間隔でチューブを反転させることによって穏やかに混合した。
四ナトリウム塩懸濁液の浸透圧
標準的な実験室技術を用いて、水及び生理食塩水中の多様な濃度の四ナトリウムEDTA溶液の浸透圧と赤血球溶解の試験を行った。各濃度のEDTA溶液2mlに50μlの血液を2時間加えることによって赤血球溶解の試験を行った。血漿浸透圧は275乃至295m/osmolであった。
人工尿結晶(Artificial Urine Crystal:AUC)の溶解に対する三つのEDTA塩の効果
尿道カテーテルについての一つの問題は、カテーテル表面に尿結晶が堆積することである。尿結晶の堆積物は微生物のコロニー化及び/またはバイオフィルムの形成、並びにカテーテルの流量の減少、を促進し得る。そのため、尿道カテーテルに関連して、尿結晶の形成を低減する消毒組成物を使用することが好ましい。標準的な実験室技術を用いて、人工尿結晶の溶解に対する三つのEDTA塩溶液の効果について試験を行った。
・25mlプラスチック製ユニバーサル容器内に人工尿及びウレアーゼを加えて45℃で7日間培養した。
・100mg/mlの二アンモニウムEDTA、二カリウムEDTA及び四ナトリウムEDTAの溶液。
・4000rpm、2分間で人工尿結晶を遠心分離する。
・上澄みをデカントし、結晶を水で洗浄し、そして遠心分離した。
・結晶を水1mlに再懸濁し、そして200μlのアリコートを四つのユニバーサル容器に加えた。
・100mg/mlの各EDTAの溶液4ml及びコントロールとしての水4mlを各ユニバーサル容器に室温で加えた。
・1、2、及び3時間後にコントロールに対する結晶の溶解を目視観察した。
上述の明細書において本発明はある好ましい実施態様との関係で記述し、そして多くの詳細を例証目的で説明しているが、本発明はその他の実施態様を許容し、そしてここで記述したある種の詳細を本発明の基本原理から外れることなく大きく変えることができるということが、当業者には明らかであろう。
Claims (26)
- 溶液中に少なくとも一つのエチレンジアミン四酢酸(EDTA)の塩を含む殺菌組成物であって、
前記少なくとも一つのEDTA塩は、少なくとも0.01%w/vであり且つ15%w/v未満の濃度の四ナトリウムEDTAを含み、
前記殺菌組成物は広いスペクトルの細菌に対して抗細菌作用を有し、少なくとも9.5のpHを有し、そして無菌及び発熱物質を含まない形態で包装され、ヒトの血流への接触及び生体内組織への接触に対して安全であり、生体適合性であり、抗細菌活性及び/または抗真菌活性を有する他の活性物質を含まない、前記組成物。 - 前記組成物が9.5乃至11.5のpHを有する、請求項1に記載の組成物。
- 前記組成物が10.5乃至11.5のpHを有する、請求項2に記載の組成物。
- 前記少なくとも一つのEDTAの塩が、0.2%w/v乃至10.0%w/vの濃度で存在している、請求項1に記載の組成物。
- 前記少なくとも一つのEDTAの塩が、0.2%w/v乃至6.0%w/vの濃度で存在している、請求項4に記載の組成物。
- 前記少なくとも一つのEDTAの塩が、0.2%w/v乃至4.0%w/vの濃度で存在している、請求項5に記載の組成物。
- 前記溶液が、水、アルコール、及びそれらの組み合わせ、からなる群から選択される、請求項1に記載の組成物。
- 前記溶液が、水とエタノールの組み合わせである、請求項7に記載の組成物。
- 前記溶液が、0.1%乃至10%w/vのエタノールを含む、請求項8に記載の組成物。
- 前記組成物が、プランクトン性形態または固着形態の細菌に対して抗細菌活性を有する、請求項1に記載の組成物。
- 前記組成物が:
(a)抗凝血活性;
(b)真菌性病原菌に対する抗真菌活性;
(c)原生動物感染症に対する阻害活性;
(d)アメーバ感染症に対する阻害活性;及び
(e)グラム陽性菌及びグラム陰性菌に対する阻害活性
を有する、請求項1に記載の組成物。 - 創傷への局所使用に適した、安全であり、生体適合性である請求項1に記載の組成物の製剤。
- 長期間にわたって殺菌活性を示す、請求項1に記載の組成物の徐放性製剤。
- 溶液中に少なくとも一つのエチレンジアミン四酢酸(EDTA)の塩を含むロックフラッシュ組成物であって、
前記少なくとも一つのEDTA塩は、少なくとも0.01%w/vであり且つ15%w/v未満の濃度の四ナトリウムEDTAを含み、
前記ロックフラッシュ組成物は、少なくとも9.5のpHを有し、無菌及び発熱物質を含まない形態で包装され、そして、留置アクセスカテーテル、尿道カテーテル、鼻腔チューブ及び気管チューブでの使用について安全で生体適合性であり、そして、ヒトの血流への接触及び生体内組織への接触に対して安全であり、生体適合性であり、抗細菌活性及び/または抗真菌活性を有する他の活性物質を含まない、前記組成物。 - 少なくとも0.01%w/vであり且つ15%w/v未満の濃度で、溶液中に三ナトリウムエチレンジアミン四酢酸(EDTA)及び四ナトリウムエチレンジアミン四酢酸(EDTA)を含む殺菌組成物であって、
前記殺菌組成物は、広いスペクトルの細菌に対して抗細菌作用を有し、少なくとも9.5のpHを有し、そして無菌及び発熱物質を含まない形態で包装され、そして、ヒトの血流への接触及び生体内組織への接触に対して安全であり、生体適合性であり、抗細菌活性及び/または抗真菌活性を有する他の活性物質を含まない、前記組成物。 - 前記組成物が9.5乃至11.5のpHを有する、請求項15に記載の組成物。
- 前記組成物が10.5乃至11.5のpHを有する、請求項15に記載の組成物。
- 前記少なくとも一つのEDTAの塩が、0.02%w/v乃至10.0%w/vの濃度で存在している、請求項15に記載の組成物。
- 前記少なくとも一つのEDTAの塩が、0.02%w/v乃至6.0%w/vの濃度で存在している、請求項18に記載の組成物。
- 前記少なくとも一つのEDTAの塩が、0.02%w/v乃至4.0%w/vの濃度で存在している、請求項19に記載の組成物。
- 前記溶液が、水、食塩水、アルコール、及びそれらの組み合わせ、からなる群から選択される、請求項15に記載の組成物。
- 前記溶液が、水とエタノールの組み合わせである、請求項21に記載の組成物。
- 前記溶液が、0.1%乃至10%w/vのエタノールを含む、請求項21に記載の組成物。
- 前記組成物が、プランクトン性形態または固着形態の細菌に対して抗細菌活性を有する、請求項15に記載の組成物。
- 表面または対象物を請求項1乃至24のいずれか一項に記載の組成物と接触させることを含む、表面または対象物上若しくは対象物内部の少なくとも一つの望ましくない微生物の個体群の成長及び増殖を阻害するための方法であって、前記表面または対象物が、カテーテル;医療用チューブ及び導管;血管内器具;ポート;埋め込まれた医療用器具;コンタクトレンズ、からなる群から選択される、方法。
- 請求項1乃至24のいずれか一項に記載の組成物に浸漬されている、傷治療における使用のためのカバー。
