内容積が50mLのビーカーに2つの銅電極を2.5cm離して取り付けた。このビーカーにデキストランおよびゼラチンを含有する液状熱可逆性ヒドロゲルの水溶液を充填した。電解槽全体に18Vの電位差をかけた。アノードの表面上には共有結合により架橋した熱可逆性ヒドロゲル調製物からなるヒドロゲル複合体が直ちに形成され、そして時間の経過に伴って膜厚が増加した。無菌ヒドロゲルは37℃では非水溶性であり、基礎にある基質に付着し、アノード金属の表面へ適合した。
それに架橋したヒドロゲルマトリックスを有するために適切な活性化生体材料表面は、そのうちの一方が保護されている二官能基を含有するモノマーの共重合によって調製することができる。例えば、モノマーであるグリシジルメタクリレートは、ヒドロゲルおよびヒドロゲル膜を形成するために他のアクリレートとのフリーラジカル開始を使用して共重合することができる。ポリ(2−ヒドロキシエチルメタクリレート−co−グリシジルメタクリレート)−ポリ(HEMA−GMA)ヒドロゲル膜は、好ましくは25℃の不活性大気下で、開始剤としてα,α’−アゾイソブチロニトリル(AIBN)を用いたUV開始光重合によって調製できる。ヒドロゲル膜のエポキシド含量は、HEMA対GMAの相対比率を変化させることによって変動させることができる。例えば、高密度のエポキシドを備える膜は、0.2mLのHEMA、0.8mLのGMA、1mLのイソプロピルアルコール、10mgのAIBN(重合開始剤として)、および3.0mLの0.1Mリン酸緩衝液(pH=7.0)を混合することにより調製できる。結果を生じた混合液を撹拌し、恒温水浴中において15分間にわたり25℃で平衡化させる。この混合液を型の中へ注ぎ込み、20分間にわたり長波紫外線に曝露させることができる。重合後、ポリ(HEMA−GMA)膜を蒸留水で数回洗浄し、生検用パンチを用いて円形ピースを切り取ることができる。上記のように形成したエポキシ官能基を有するポリ(HEMA−GMA)膜ディスク(湿重量10g、直径=1.0cm)を2時間にわたりリン酸緩衝液(50mM、pH=8.0)中で平衡化させ、39℃で保持した熱可逆性ヒドロゲルを保持する容器へ移す。生体材料膜の表面への熱可逆性ヒドロゲルの固定化は、頻回に撹拌しながら39℃で実施できる。熱可逆性ヒドロゲルでコーティングしたポリ(HEMA−GMA)膜を取り外して洗浄すると、非共有結合したヒドロゲル材料を除去することができる。

- 金属、生体活性ガラス、セラミック、サンゴ質材料、加工処理組織、合成熱可塑性ポリマー、およびそれらの混合物、複合体、またはサブアセンブリーからなる群より選択される材料で構築された表面を有する基質と;
前記基質の前記表面上に設けられ、前記基質の前記表面上に固定化された生物活性ヒドロゲルマトリックス層であって、前記マトリックスがポリグリカン成分およびポリペプチド成分、さらに極性アミノ酸、無傷コラーゲン、および二価カチオンのキレート剤からなる群から選択される少なくとも1つの増強剤を含んでおり、
前記ポリグリカン成分および前記ポリペプチド成分の少なくとも1つが、前記いずれかの材料で構築された前記基質の前記表面に共有結合により付着しているか、または、前記合成熱可塑性ポリマーでできた基質の開いた、膨潤した表面内に捕捉されており、共有結合による付着が、前記ポリグリカン成分および前記ポリペプチド成分の少なくとも1つに保持された反応性基と、前記基質の前記表面に保持された反応性基との反応により生じる、生物活性ヒドロゲルマトリックス層と、

金属、生体活性ガラス、セラミック、サンゴ質材料、加工処理組織、合成熱可塑性ポリマー、およびそれらの混合物、複合体、またはサブアセンブリーからなる群より選択される材料で構築された表面を有する基質を供するステップと;
前記第一高分子量成分を前記いずれかの材料でできた前記基質の前記表面に共有結合により付着させる、または前記第一高分子量成分を前記合成熱可塑性ポリマーで構築された基質の開いた、膨潤した表面内に捕捉することにより、前記基質の前記表面上に前記第一高分子量成分を固定化するステップであって、前記共有結合による付着が、前記第一高分子量成分に保持された反応性基と、前記基質の前記表面に保持された反応性基との反応により生じるステップと;
第一高分子量成分を、ポリグリカンおよびポリペプチドからなる群から選択される第二高分子量成分と接触させるステップであって、該接触させるステップが前記固定化するステップの前、間または後のいずれかに行われ、それによって基質の表面上に固定化生物活性ヒドロゲルコーティングを形成するステップであって、第一および第二高分子量成分の1つがポリグリカンであり、その他がポリペプチドであるステップと;
第一高分子量成分を少なくとも1つの増強剤と接触させるステップであって、前記接触させるステップが前記固定化するステップの前、間、後に行われ、増強剤が、極性アミノ酸、無傷コラーゲンおよび二価カチオンのキレート剤からなる群から選択されるステップをさらに含み、
前記ポリペプチド成分が、ラミニン、ニドゲン、フィブリン、フィブリリン、コラーゲン、ゼラチン、ケラチン、デコリン、アグレカン、および糖タンパク質からなる群から選択された組織由来ポリペプチドまたは合成ポリペプチドであり、
前記ポリグリカン成分が、グリコサミノグリカン、グルコサミノグリカン、デキストラン、ヘパラン、ヘパリン、ヒアルロン酸、アルギン酸塩、アガロース、カラゲナン、アミロペクチン、アミロース、グリコーゲン、デンプン、セルロース、キチン、ヘパラン硫酸、コンドロイチン硫酸、デキストラン硫酸、デルマタン硫酸およびケラタン硫酸からなる群から選択される多糖または硫酸化多糖である方法。 - 前記固定化するステップの前に、第一高分子量成分を化学的に修飾し、共有結合に参加できる反応性部位をその上に形成するステップをさらに含む請求項30の方法。
- 前記修飾するステップが、第一高分子量成分を酸化するステップを含む請求項31の方法。
- 前記修飾するステップが、過ヨウ素酸の塩で第一高分子量成分を処理するステップを含む請求項32の方法。
- 前記固定化するステップの前に、基質の表面を修飾し、共有結合に参加できる反応性部位をその上に形成するステップをさらに含む請求項30の方法。
- 前記修飾するステップが、前記基質の表面上に複数の反応性アミン基を形成するステップを含む請求項34の方法。
- 基質が医療用具である請求項30の方法。
- 医療用具が、能動型医療用具および受動型医療用具からなる群から選択される請求項36の方法。
- 医療用具が、肝臓、腎臓もしくはその他の器官支持システムのためのエクスビボ・バイオリアクター、カテーテル、人工動脈、人工器官、組織片を含有する医療用具、細胞を含有する医療用具、人工靱帯、人工骨、グルコースセンサー、心ペースメーカー、ラップバンド、モニター、人工喉頭、プロテーゼ、脳刺激器、膀胱ペースメーカー、シャント、ステント、チューブ、除細動器、電気的除細動器、心臓弁、人工関節、固定具、眼内レンズ、人工内耳、人工乳房、神経刺激器、骨成長刺激器、人工血管、筋刺激器、左室補助装置、圧センサー、迷走神経刺激器、薬物送達システム、縫合糸、ステープル、および細胞スカフォールディング材料からなる群から選択される請求項36の方法。
- 基質の表面が、アクリレート、ポリグリコール酸−ポリ乳酸コポリマー、ポリヒドロキシブチレート、ポリエステル、延伸ポリテトラフルオロエチレン(ePTFE)、生体活性ガラス、セラミック、サンゴ質材料、加工処理組織、ポリカーボネート、ポリウレタン/ポリカーボネートコポリマー、金属、およびそれらの混合物、複合体またはサブアセンブリーからなる群から選択される材料で構築されている請求項30の方法。
- 第一高分子量成分がポリグリカンであり、第二高分子量成分がポリペプチドである請求項30の方法。
- ポリグリカンが、2,000〜8,000,000Daの分子量範囲を有する請求項40の方法。
- ポリペプチドが、粘膜下組織、動脈、声帯、胸膜、気管、気管支、肺胞隔膜、靱帯、耳介軟骨、腹筋膜、肝臓、腎臓、神経鞘、クモ膜、硬膜、および軟膜からなる群から選択される組織由来である請求項40の方法。
- ポリペプチドが、3,000〜3,000,000Daの分子量範囲を有する請求項40の方法。
- ポリグリカンがデキストランであり、ポリペプチドがゼラチンである、請求項40の方法。
