Description
ウイルス及び細菌、特に多数の血清型又は高率の変異を有するものに対する中和抗体及び/又は細胞媒介防御免疫応答の誘起が望ましいが、それらに対するワクチンの開発に対する主要な障害は、異なる単離体又は株間に存在する外部タンパク質の多様性である。マウスとヒトの両方での細胞障害T−リンパ球(CTL)は保存された内部ウイルスタンパク質由来のエピトープを認識でき(J.W.Yewdellら、Proc.Natl.Acad.Sci.(USA)82, 1785 (1985);A.R.M.Townsendら、Cell 44, 959(1986);A.J.McMichaelら、J.Gen.Virol. 67, 719(1986);J.Bastinら、J.Exp.Med. 165, 1508(1987);A.R.M.Townsend及びH.Bodmer,Annu.Rev.Immunol. 7, 601(1989))、ウイルスに対する免疫応答で重要であると考えられるので(Y.-L.Lin及びB.A.Askonas, J.Exp.Med. 154, 225(1981);I.Gardnerら、Eur.J.Immunol. 4, 68(1974);K.L.Yap及びG.L.Ada, Nature 273, 238(1978);A.J.McMichaelら、New Engl.J.Med. 309, 13(1983);P.M.Taylor及びB.A.Askonas, Immunol. 58, 417(1986))、異なるウイルス株に対する異種防御を提供できるCTLワクチンの開発への努力がなされた。
CTLは、CTLのT細胞レセプターがMHCクラスI及び/又はクラスII分子と結合した外来のペプチドを認識すると、ウイルス又は細菌に感染した細胞を殺すことが知られている。これらのペプチドは、該タンパク質の病原体中存在場所又は機能に関係なく、外来タンパク質から内部合成される。保存されたタンパク質からのエピトープの認識によって、CTLは異種防御を提供することができる。細胞内細菌の場合、細菌から分泌又は放出されたタンパク質はプロセシングを受け、MHCクラスI及びII分子によって提示され、感染を減少させる又は除去することに関与しうるT−細胞応答を生ぜしめる。
CTL応答を生じさせる大部分の努力において、細胞内でタンパク質抗原を産生させる複製ベクターを使用するか(J.R.Benninkら、上記311, 578(1984);J.R.Bennink及びJ.W.Yewdell, Curr.Top.Microbiol.Immunol. 163, 153(1990);C.K.Stoverら、Nature 351, 456(1991);A.Aldovini及びR.A.Young, Nature 351, 479(1991);R.Schferら、J.Immunol. 149, 53(1992);C.S.Hahnら、Proc.Natl.Acad.Sci.(USA) 89, 2679(1992))、又は細胞質ゾルの中にペプチドを導入することに焦点をあててきた(F.R.Carbone及びM.J,Bevan, J.Exp.Med. 169, 603(1989);K.Deresら、Nature 342, 561(1989);H.Takahasiら、同書344, 873(1990);D.S.Collinsら、J.Immunol. 148, 3336(1992);M.J.Newmanら、同書148,2357(1992))。これらのアプローチの両方とも、ワクチンとしての有用性を減少させうる限界を有する。レトロウイルスベクターは、組換えウイルスの複製能力を保持しながら融合タンパク質として発現できるポリペプチドのサイズと構造に限界を有するし(A.D.Miller, Curr.Top.Microbiol.Immunol. 158,1(1992))、結果として起こる免疫化のための、ワクシニアなどのベクターの有効性は、ワクシニアに対する免疫反応によって低下しうる(E.L.Cooneyら、Lancet 337, 567(1991))。また、ウイルスベクターと改変病原体は、ヒトにおけるそれらの使用を妨げうる固有のリスクを有する(R.R.Redfieldら、New Engl. J. Med. 316, 673(1987);L.Mascolaら、Arch.Intern.Med. 149, 1569(1989))。更に、提示されるペプチドエピトープの選別は、個々人のMHC抗原の構造に依存していて、それ故、ペプチドワクチンは、異系交配された集団におけるMHCハプロタイプの多様性のため有効性は限られたものでありうる。
マウスの腹腔内(i.p.)、静脈内(i.v.)、又は筋肉内(i.m.)に導入されたCaCl2沈殿DNAは発現されることができることをBenvenisty.N.及びReshef,L.(PNAS 83, 9551-9555(1986))は示した。マウスでのDNA発現ベクターの筋肉内(i.m.)注射により筋肉細胞によるDNAの取込みとそのDNAによりコードされたタンパク質の発現が起こることが証明された(J.A.Wolffら、Science 247,1465(1990);G.Ascadiら、Nature 352, 815(1991))。プラスミドはエピソームとして維持されることが知見され、プラスミドは複製しなかった。次に、ラット、魚、霊長類の骨格筋、及びラットの心筋でのi.m.注射の後、永続性発現が観察された(H.Linら、Circulation 82, 2217(1990);R.N.Kitsisら、Proc.Natl.Acad.Sci.(USA) 88, 4138(1991);E.Hansenら、FEBS Lett. 290, 73(1991);S.Jiaoら、Hum. Gene Therapy 3, 21(1992);J.A.Wolffら、Human Mol. Genet. 1, 363(1992))。治療薬として核酸を使用する技術がWO90/11092(1990年10月4日)で報告されたが、その中で、裸のポリヌクレオチドを使用して脊椎動物にワクチン接種することが報告された。
腫瘍の除去のためにCTLを活性化する際に、抗原提示細胞の表面のB7と主要組織適合性複合体(MHC)のエピトープの提示が同格の役割を果たすことが最近、総説として報告された(Edgington, Biotechnology 11, 1117-1119, 1993)。抗原提示細胞(APC)の表面のMHC分子がT−細胞レセプター(TCR)にエピトープを提示すると、同じAPCの表面上に発現されたB7がCTLA−4又はCD28に結合することによって、第2のシグナルとして作用する。その結果、CD8+T−細胞が増殖しAPCを殺すように、信号を送るCD4+ヘルパーT−細胞が急速に分裂する。
免疫化が筋肉内であることは、方法の成功に必要ない。ウシ成長ホルモン(BGH)をコードするDNAでコートされた金微小弾丸をマウスの皮膚に導入すると、マウスで抗BGH抗体が産生されることをTangら[Nature, 356, 152-154(1992)]は開示した。ジェット注射器を使用して、生きている動物の皮膚組織、筋肉組織、脂肪組織、乳腺組織をトランスフェクトすることができることをFurthら[Analytical Biochemistry, 205, 365-368(1992)]は示した。核酸を導入する種々の方法が最近、総説として記載された[(Friedman,T., Science, 244, 1275-1281(1989)]。鳥類のインフルエンザDNAをニワトリにim,ip,iv投与すると、致死的誘発に対する防御を与えることを主張しているRobinsonら[Abstracts of Papers Presented at the 1992 meeting on Modern Approaches to New Vaccines, Including Prevention of AIDS, Cold Spring Harbor, p92;Vaccine 11, 957(1993)]をも参照。マウスにおける、DNA:カチオンリポソーム複合体の静脈内注射はZhuら[Science 261, 209-211(1993年7月9日);WO93/24640(1993年12月9日)をも参照]によって示され、クローンされたトランス遺伝子の全身的発現が起った。インフルエンザウイルスタンパク質をコードするDNAの注射による、インフルエンザウイルス感染に対する異種防御について、最近Ulmerら[Science 259, 1745-1749(1993)]が報告した。
クローンされたゲノム(スプライスされていない)HIV遺伝子の筋肉内接種によって、HIVに対するマウスの免疫応答の誘起について、Wangら[P.N.A.S. USA 90, 4156-4160(1993年5月)]が報告した。しかし、達成された免疫応答のレベルは非常に低く、システムはマウス乳癌ウイルス(MMTV)ロングターミナルリピート(LTR)プロモーターの部分及びシミアンウイルス40(SV40)プロモーターとターミネーターの部分を使用した。SV40は、恐らく宿主細胞DNAへの挿入によって、細胞を形質転換することが知られている。従って、Wangらによって記載されたシステムはヒトへの投与(これは本発明の目的の一つである)のために、全体として不適切である。
WO93/17706は、担体粒子が遺伝子構築物でコートされ、コートされた粒子が動物の細胞内に加速されて入ることを特徴とする、ウイルスに対し動物にワクチン接種をする方法を記載する。
レポーター遺伝子をコードするプラスミドDNAの筋肉内注射によって、注射部位及びその近辺の伸縮性細胞でその遺伝子が発現することを、Wolffら(上記)の研究により初めて証明された。インフルエンザA赤血球凝集素(Montgomery,D.L.ら、1993, Cell Biol., 12, pp.777-783)又はヌクレオプロテイン(Montgomery,D.L.ら、上記;Ulmer,J.B.ら、1993, Science, 259, pp.1745-1749)をコードするプラスミドの注射により、インフルエンザに対する、マウスの免疫化が成功したことを、最近の報告が証明した。ヘルペスウイルスのDNA免疫化の最初の使用が報告された(Coxら、1993, J.virol., 67, pp.5664-5667)。ウシヘルペスウイルス1(BHV−1)糖タンパク質gIVをコードするプラスミドの注射により、マウスと子ウシで抗gIV抗体ができた。BHV−1による鼻内誘発の際、免疫化した子ウシは症状が軽く、対照より実質的に少ないウイルスが生成した。
結核(TB)は、病原体マイコバクテリウム・ツバーキュロシス(Mycobacterium tuberculosis)によって起る肺の慢性感染症である。TBは、世界中で最も臨床的に重大な感染の一つであって、毎年3百万の死と1千万の新しい患者がある。世界の人口の1/3もの多くが感染していると推定されるし、発展途上国では、活性TBの5千5百万の患者が報告されている。今世紀の半ばまで、TBは米国における第1の死因であった。しかし、衛生状態が改善されたこと、及び抗微生物薬の出現により、死亡者数は、2000年までにこの病気が撲滅されるであろうと予測されるまでに着実に減少した。しかし、発展途上国の大部分で、活性TBの患者数は1980年半ば以来毎年増加している。この復活は、部分的に移住と免疫低下HIV−感染患者数の増加に帰せられる。減少しなければ、TBによって、次の10年で3千万以上のヒトの命が失われることが予測される。これらの数字は驚くべきものであるようだが、M.tuberculosisの多剤耐性(MDR)株が出現したことはより大きな関心事である。これらのMDR株は伝統的薬剤治療で扱いにくく、これらのMDR株のために、最近の幾つかのTBの流行、特に都会の中心での流行が起った。そのため、長期間でのTBの管理のキーとなる事の一つは有効なワクチンであろう(総説として、Bloom及びMurray, 1993, Science 257, 1055を参照)。
M.tuberculosisは、マクロファージに感染する細胞内病原体であり、このタイプの細胞のファゴリソソームの厳しい環境内で生き残ることができる。大部分の吸入された桿菌は活性化した肺胞マクロファージで破壊される。しかし、生存する桿菌はマクロファージ内で増殖でき、細胞の死の際に放出される。細胞の死は、リンパ球、単球、マクロファージが部位に来るように合図する。桿菌を含むマクロファージの溶解は遅延型超過敏性反応(DTH)に媒介され、感染細胞の区域を囲む凝固乾酪結核結節を発達させる。DTHが続くと結核結節を液体化し、含有する桿菌を放出させる。細胞外の桿菌が大量に入ると、更なるDTHを引起こし、気管支に打撃を与え、リンパ性播種、血行性播種、気管支性播種を引起し、最後に感染桿菌を呼吸によって広がらせる。
TBに対する免疫には幾つかのタイプのエフェクター細胞が必要である。インターフェロン−γなどのサイトカインによるマクロファージの活性化は、細胞内マイコバクテリウム増殖を最小化する有効な方法である。しかし、この方法では該桿菌の完全な撲滅はしばしばは達成されない。TBに対する防御の獲得にはTリンパ球が必要である。これらの内、CD8+とCD4+T細胞が重要であるように考えられる(Ormeら,1993, J.Infect.Dis. 167, 1481)。これらの細胞タイプはマイコバクテリウムに反応してインターフェロン−γを分泌するが、これはTh1免疫応答の特徴であり、これらの細胞タイプはマイコバクテリウムパルスの標的細胞に対する細胞毒性活性を有する。β−2ミクログロブリン欠失・CD8欠失マウスを用いる最近の研究によると、CTL反応はM.tuberculosisに対する防御を付与するのに重要であることが知見された(Flynnら、1992, Proc. Natl. Acad. Sci. USA 89, 12013;Flynnら、1993, J. Exp. Med. 178, 2249;Cooperら、1993, J. Exp. Med. 178, 2243)。対照的にBリンパ球は関与しているようには考えられず、抗マイコバクテリウム抗体の受動移入は防御を与えない。そのため、TBに対する効果的ワクチンは細胞媒介免疫応答を生ぜしめなければならない。
T細胞の抗原刺激にはMHC分子による提示が必要である。マイコバクテリア抗原が抗原提示経路に関与するために、該抗原は該細菌から放出されなければならない。感染マクロファージでは、分泌又は細菌溶解でこのことを達成できる。マイコバクテリアは多数の可能性のあるT−細胞抗原を有し、そのうち幾つかは現在同定されている(Andersen 1994, Dan.Med.Bull. 41, 205)。これらの抗原のうち幾つかは該細菌によって分泌される。TBに対する免疫は、これらの分泌抗原に対するCD8+とCD4+T細胞に媒介されると一般的に考えられている。TBのマウスモデルとモルモットモデルにおいて、減少した体重損失によって測定された、細菌攻撃からの防御は、分泌マイコバクテリウム抗原の混合物を使用して達成された(Pal及びHorowitz, 1992, Infect.Immunity 60, 4781;Andersen 1994, Infect.Immunity 62, 2536;Collins, 1994, Veterin.Microbiol. 40, 95)。
幾つかの可能性のある防御T細胞抗原がM.tuberculosisで同定され、これらのうち幾つかはワクチン標的として研究されている。最近の研究によると、優勢なT−細胞抗原は、マクロファージ内にあるときマイコバクテリウムによって分泌されるタンパク質である。例えば、i)タンパク質の抗原85複合体(85A,85B,85C)(Wiker及びHarboe, 1992, Microbiol.Rev. 56, 648)、ii)ESAT−6という名の6kDaタンパク質(Andersen 1994, Infect.Immunity 62, 2536)、iii)PhoSと相同性を有する38kDaリポタンパク質(Young及びGarbe, 1991, Res.Microbiol. 142, 55;Andersen, 1992, J.Infect.Dis. 166, 874)、iv)65kDa GroEL熱ショックタンパク質(Siva及びLowrie, 1994, Immunol. 82, 244)、v)プロリンとスレオニンに富む55kDaタンパク質(Romainら、1993, Proc.Natl.Acad.Sci. USA 90, 5322)、vi)19kDaリポタンパク質(Faithら、1991, Immunol. 74, 1)である。
3種の抗原85タンパク質(A、B、C)の各々の遺伝子がクローンされ、配列決定された(Borremansら、1989, Infect. Immunity 57, 3123;Contentら、Infect.Immunity 59,3205;DeWitら、1994, DNA Seq.4, 267)。更に、これらの構造的に関連するタンパク質は、感染とワクチン接種後、強力なT−細胞応答の標的である(Huygenら、1988, Scand.J.Immunol. 27, 187;Launoisら、1991, Clin.Exp.Immunol. 86, 286;Huygenら、1992, Infect..Immunity 60, 2880;Munkら、1994, Infect.Immunity 62, 726;Launoisら、1994, Infect.Immunity 62, 3679)。それ故、抗原85タンパク質は良好なワクチン標的であると考えられる。
本発明の概要
M.tb病の予防でDNA免疫化の効力を試験するために、M.tbタンパク質をコードするDNA配列を真核発現ベクターにクローンした。これらのDNA構築物は、動物に注入すると免疫応答を誘起する。該遺伝子(又は他のM.tb遺伝子)による直接的DNA免疫化が病気から免疫化された動物を防御することができるかどうかを評価するために、免疫化動物をマイコバクテリウムで感染させる。それ故、注射又は他の方法で動物組織に直接導入すると、M.tbタンパク質のin vivo発現を誘起できる、DNA構築物及びRNA転写体を含む核酸を開示する。これらの核酸の注射は、次の段階の攻撃に対し防御的である、M.tb抗原の特異的な細胞障害Tリンパ球(CTL)を産生させ、M.tb特異的ヘルパーTリンパ球応答を生じさせる免疫応答を誘起できる。これらの核酸は、M.tbに対して免疫を誘起するワクチンとして有用であり、感染を予防でき、及び/又はM.tb関連疾患を軽減できる。

発明の詳細な説明
本発明は、ヒトなどの哺乳動物を含む脊椎動物へのin vivoで直接導入によって、該動物内でコードされたタンパク質の発現を誘起するポリヌクレオチドを提供する。本明細書で使用するポリヌクレオチドは、生きている脊椎動物細胞に導入されると、ポリヌクレオチドを含む遺伝子によりコードされる翻訳産物を細胞マシナリーにより産生させることができるような必須の制御要素を含む核酸である。本発明の一実施態様では、ポリヌクレオチドは、転写プロモーターと機能を有するように結合したMycobacterium tuberculosis(M.tb)遺伝子を含むポリヌクレオチドである。本発明の別の実施態様では、ポリヌクレオチドワクチンは、真核細胞マシナリー(リボソーム、tRNA、及び他の翻訳因子)により翻訳されるM.tb遺伝子をコードするポリリボ核酸を含む。ポリヌクレオチドによってコードされるタンパク質が、M.tbのタンパク質などの、病理状態の場合を除き通常は動物では存在しないものであるとき(即ち異種タンパク質)、動物の免疫システムは活性化され、防御免疫応答が誘起される。これらの外因性タンパク質は動物自身の組織で産生されるので、実際のM.tb感染が起きるときと同様な方法で、発現タンパク質は主要組織適合システム(MHC)でプロセシングを受ける。本明細書で示す結果は、M.tbに対する免疫応答の誘導である。コードされたタンパク質に対する免疫応答を生じせしめる目的のためのポリヌクレオチドを、本明細書ではポリヌクレオチドワクチン又はPNVという。
当業者が明細書から理解できる、本発明の多数の実施態様がある。即ち、異なる転写プロモーター、異なるターミネーター、異なるキャリアベクター、又は異なる特定の遺伝子配列を使用しても成功できる。
本発明は、哺乳動物組織に導入すると、in vivoでポリヌクレオチドの発現を誘起し、コードされたタンパク質を産生するポリヌクレオチドを使用する方法を提供する。コードされたタンパク質のアミノ酸配列を変える、タンパク質をコードするヌクレオチド配列の変種体又は変異体を産生できることは、当業者に容易に明らかである。改変した発現タンパク質は改変アミノ酸を有しうるが、マイコバクテリウムタンパク質と反応する免疫応答を依然として誘起するので、機能性等価物と考えられる。更に、完全長のタンパク質の一部をコードする、完全長の遺伝子のフラグメントも構築できる。これらのフラグメントは、マイコバクテリウムタンパク質と反応する抗体を誘起するタンパク質又はペプチドをコードしうるので、機能性等価物と考えられる。
本発明の一実施態様では、M.tb遺伝子産物をコードする遺伝子が発現ベクターに挿入される。ベクターは、真核RNAポリメラーゼによって認識される転写プロモーター、及びM.tb遺伝子コード配列の終りに転写ターミネーターを含む。好適実施態様では、プロモーターは、イントロンA配列(CMV−intA)を有するサイトメガロウイルスプロモーターであるけれども、強力イムノグロブリンプロモーター又は他の真核遺伝子プロモーターなどの任意の多数の他の公知のプロモーターを使用できることを、当業者は認識するであろう。好適な転写ターミネーターはウシ成長ホルモンターミネーターである。CMVintA−BGHターミネーターの組合わせが好適である。更に、原核細胞でのポリヌクレオチドの調製を助けるために、抗生物質耐性マーカーをも、適切な原核プロモーターの転写制御下の発現ベクターに任意に含ませる。アンピシリン耐性遺伝子、ネオマイシン耐性遺伝子、任意の他の適切な抗生物質耐性マーカーを使用できる。本発明の好適実施態様では、抗生物質耐性遺伝子はネオマイシン/カナマイシン耐性の遺伝子産物をコードする。更に、原核生物の生長によるポリヌクレオチドの高レベル産生を助けるために、ベクターが原核複製起点を含み、高コピー数であることが有利である。多数の市販の原核クローニングベクターの任意のものは、これらの要素を提供する。本発明の好適実施態様では、これらの機能は、pUCシリーズとして知られる市販のベクターによって提供される。しかし、必須でないDNA配列を除去するのが望ましいであろう。従って、pUCのlacZ及びlacIコード配列を除去しうる。ベクターは真核細胞で複製できないことも望ましい。このことは、レシピエントのゲノムにポリヌクレオチドワクチン配列の挿入の危険を最小化する。
別の実施態様では、ラウス肉腫ウイルス(RSV)ロングターミナルリピート(LTR)をプロモーターとして使用する発現ベクターpnRSVを使用する。更に別の実施態様では、CMVプロモーター及びBCG転写ターミネーターがクローンされた変異pBR322ベクターであるV1を使用する。本発明の好適実施態様では、V1とpUC19の要素を一緒にしてV1Jと命名した発現ベクターV1Jを製造する。
V1J、V1JtPA、又は別の望ましい発現ベクターに、抗原85複合体遺伝子の一つなどのM.tb遺伝子、又は抗M.tb免疫応答(CTL、ヘルパーTリンパ球及び抗体)を誘起できる任意の他のM.tb遺伝子をクローンする。別の実施態様では、アンピシリン耐性遺伝子をV1Jから除去し、ネオマイシン耐性遺伝子で置換え、V1J−neoを作製し、その中に多数の別のM.tb遺伝子の任意のものを、本発明の使用のためにクローンできる。更に別の実施態様では、ベクターはV1Jnsであるが、単一のSfi1制限部位がV1J−neoの位置2114の単一のKpn1部位に細工されたことを除いて、V1JnsはV1Jneoと同じである。ヒトゲノムDNAのSfi1部位の頻度は非常に低い(100,000塩基当たり約1部位)。従って、このベクターにより、宿主DNAへの発現ベクター挿入を、抽出ゲノムDNAをただSfi1消化するだけで注意深いモニターができる。更なる実施態様では、ベクターはV1Rである。このベクターでは、できるだけ多くの非必須DNAを除去し、高度にコンパクトなベクターを製造する。このベクターでは、所望でない配列をコードするという心配なしに、より大きなインサートを使用でき、特定のウイルス遺伝子をコードする構築物が廻りの組織に導入されるときに、細胞による取込みを最適化できる。前述のベクター改変をするのに使用される方法及び開発方法は、当業者公知の方法により達成できる。
本発明の有用性の一つが、M.tb配列の多様性とCTL及びT−細胞増殖応答との関連、並びに他のパラメーターとの関連を明らかにできるような、in vivoとin vitroでの試験及び解析のシステムを提供することであることを、当業者は、本研究から認識するであろう。これらの種々の遺伝子の単離とクローニングを、当業者公知の方法により達成できる。本発明は更に、ワクチン製造のための、M.tbの株及び配列の系統的同定の方法を提供する。M.tb株の一次単離株からの遺伝子の導入は、該生物の臨床単離株に対する免疫応答を誘起する免疫原を提供し、当分野で未達成の必要性を満たす。更に、もし毒性の単離株が変化するならば、必要なら、新規の配列を反映するように免疫原を改変できる。
本発明の一実施態様では、M.tbタンパク質をコードする遺伝子は直接、転写プロモーターに結合している。組織特異的プロモーター又はエンハンサー(例えば筋肉クレアチンキナーゼ(MCK)エンハンサー要素)は、特定の組織タイプにポリヌクレオチドの発現を制限するために望ましいであろう。例えば、伸縮性細胞は、分裂しない最終的に分化した細胞である。染色体への外来DNAの挿入には、細胞分裂とタンパク質合成の両方が必要であるようである。従って、伸縮性細胞などの非分裂細胞にタンパク質発現を限定することは好ましいであろう。しかし、CMVプロモーターの使用は、PMVが導入される多くの組織で発現を達成するために適当である。
好ましくは、M.tbと他の遺伝子を、ポリヌクレオチドワクチン化のために特別に最適化した発現ベクターに連結する。要素には、本明細書記載の、転写プロモーター、免疫原エピトープ、免疫増強遺伝子又は免疫調節遺伝子をコードする更なるシストロン、並びにそれら自身のプロモーター、転写ターミネーター、細菌の複製起点、抗生物質耐性遺伝子がある。必要に応じて、ベクターは、ポリシストロンmRNAの発現のための内部リボソームエントリー部位(IRES)を含みうる。対応するDNAによってコードされる多シストロンmRNAを産生するためにin vitroで転写されたRNAは、本発明の範囲内であることを当業者は理解するであろう。この目的のために、転写プロモーターとして、T7又はSP6プロモーターのような強力なRNAポリメラーゼプロモーターを使用し、線状化したDNA鋳型を用いてin vitroのrun−on転写を行うことは望ましい。これらの方法は当業界周知である。
次の段階の攻撃に対するポリヌクレオチドM.tb免疫原の防御効力は、本発明のDNAで免疫化することにより証明される。感染体を含まず、細菌のアッセンブリー/複製は必要なく、抗原決定基選択ができるので、上記方法は利点を有する。更に、マイコバクテリウム遺伝子産物の配列は、M.tbの種々の株間で保存されうるので、M.tbの別の株による次の段階での攻撃に対して防御できる。
抗原85A、B、又はCをコードするDNA発現ベクターの注入により、次の段階での攻撃に対するかなりの防御免疫を発生させることができる。特に、特異的CTL応答と特異的ヘルパーTリンパ球応答を発生させることができる。
M.tb遺伝子産物は、M.tbの種々の株間での高程度の保存を示し、免疫応答は、細胞内発現とMHCプロセシングに反応して発生するので、多数の別のM.tbPNV構築物が交差反応性免疫応答を生ぜしめることが期待される。
本発明は、自己複製体又はアジュバントの必要がなく、異種防御免疫を誘起する方法を提供する。ウイルスタンパク質DNAとヒト成長ホルモンDNAの注射後、発現タンパク質に対する高力価抗体の発生(Tangら、Nature 356, 152, 1992)は、保存された抗原に対する細胞障害性T−リンパ球ワクチンとヘルパーTリンパ球ワクチンを別々に、又は組合わせて用いて、抗体ベースのワクチンを作製する容易で高度に有効な方法であることを示す。
容易にDNA構築物を産生し、精製することができ、このことは伝統的タンパク質精製にまさるとも劣らず、組合わせワクチンの産生を容易にする。従って、多重構築物、例えば抗原85複合体遺伝子及び非M.tb遺伝子をも含む任意の他のM.tb遺伝子をコードする多重構築物を製造し、混合し、共投与できる。更に、DNA注入後、タンパク質発現は維持され(H.Linら、Circulation 82, 2217(1990);R.N.Kitsisら、Proc.Natl.Acad.Sci.(USA)88, 4138(1991);E.Hansenら、FEBS Lett. 290, 73(1991);S.Jiaoら、Hum.Gene Therapy 3, 21(1992);J.A.Wolffら、Human Mol. Genet. 1, 363(1992))、B−細胞とT−細胞の記憶の永続性は増強されることができ(D.Grray及びP.Matzinger, J.Exp.Med. 174, 969(1991);S.Oehenら、同書176, 1273(1992))、長寿命の液性免疫と細胞媒介免疫を誘起する。
ワクチンレシピエントに導入される発現可能なDNA又は転写されたRNAの量は、非常に広い投与範囲を有し、使用される転写及び翻訳プロモーターの強さによる。更に、免疫応答の大きさは、タンパク質発現のレベルと発現された遺伝子産物の免疫原性による。一般的に、DNAの約1ng〜5mg、100ng〜2.5mg、1μg〜750μg、好ましくは約10μg〜300μgの効果的投与範囲を直接、筋肉組織に投与する。皮下注射、真皮導入、皮膚圧痕、及び腹腔内送達、静脈内送達、又は吸入送達などの他の型の投与も適している。追加ワクチンをすることも考えられる。M.tbポリヌクレオチド免疫原によるワクチン投与後、抗原85複合体遺伝子産物などのM.tbタンパク質免疫原で追加免疫することも考えられる。本発明のPNVの非経口導入と同時に、又はその後に、インターロイキン−12タンパク質(又は他のサイトカイン、例えばGM−CSF)の静脈内投与、筋肉内投与、皮下投与、又は投与の他の方法などの非経口投与は、利点を有する。
ポリヌクレオチドは裸でよい、即ちレシピエントの免疫システムに影響を与えない任意のタンパク質、アジュバント又は他の薬剤と一緒になっていなてもよい。この場合、ポリヌクレオチドは生理的に許容できる溶液、例えば滅菌生理食塩水又は滅菌緩衝化生理食塩水であるが、それに限定されない溶液中にあることが望ましい。あるいは、DNAは、DNA−リポソーム混合物として、当業界公知のレシチンリポソーム又は他のリポソームなどのリポソームと一緒でありうるし、又は免疫応答を増強するために、当業界公知の、タンパク質又は他の担体などのアジュバントと一緒でもよい。それに限定されないがカルシウムイオンなどの、DNAの細胞内取込みの助けとなる薬剤も使用できる。一般的に、これらの薬剤を、トランスフェクションを容易にする薬剤又は医薬として許容可能な担体と本明細書でいう。ポリヌクレオチドで微小弾丸をコートする技術は当業界公知であり、この技術も本発明との関連で有用である。ヒトでの使用のDNAの場合、最終DNA産物が医薬として許容可能な担体又は緩衝溶液中にあることは有用である。医薬として許容可能な担体又は緩衝溶液は当業界公知であり、Remington著のPharmaceutical Sciencesなどの種々のテキストに記載のものを含む。
別の実施態様では、本発明は、in vivoで真核組織にポリヌクレオチドを導入して遺伝子産物を産生するために、発現できる隣接の核酸配列を含むポリヌクレオチドである。好ましくは、コードされた遺伝子産物は、免疫刺激剤又は免疫応答を生ぜしめることができる抗原として作用する。従って、この実施態様の核酸配列はM.tb免疫原エピトープ、及び必要に応じてB7ファミリータンパク質の一員などの、サイトカイン又はT−細胞共刺激性要素をコードする。
遺伝子産物よりむしろ遺伝子で免疫化する幾つかの利点がある。第1は、天然又は天然に近い抗原を免疫システムに提示できる比較的単純性である。細菌、酵母、又は哺乳動物細胞でさえ、その中で組換え的に発現された哺乳動物タンパク質では、しばしば、適切な抗原性のために広範な処理を必要とする。DNA免疫化の第2の利点は、免疫原がMHCクラスI経路に入り、細胞障害性T細胞応答を誘起する可能性である。インフルエンザAヌクレオブロテイン(NP)をコードするDNAでマウスを免疫化すると、fluの異種株による攻撃に対しマウスを防御する、NPへのCD8+応答が発生した(Montgomery,D.L.ら、上記;Ulmer,J.ら、上記)。
細胞媒介免疫がM.tb感染を抑制することにおいて重要であるという強力な証拠がある(Ormeら、1993, J.Infect.Dis. 167, 1481;Cooperら、1993, J.Exp.Med. 178, 2243;Flynnら、1993, J.Exp.Med. 178, 2249;Ormeら、1993, J.Immunol. 151, 518)。DNA免疫化は液性及び細胞媒介免疫応答の両方を誘起できるので、最も大きい利点は、ワクチンとしての可能性のために、多数のM.tb遺伝子を調査する比較的簡単な方法をDNA免疫化が提出することでありうる。
DNA注入による免疫化によりまた、上記のように、多成分のサブユニットワクチンのアッセンブリを容易にできる。複数のインフルエンザ遺伝子による同時免疫化が最近報告された(Donnelly,J.ら、1994, Vaccines, pp55-59)。遺伝子産物が免疫システムの異なる武器を活性化するが、それらの遺伝子がM.tbワクチンに含まれることにより、次の段階の攻撃からの防御もできる。
本発明のワクチンは、ヒトと同様に家畜又は農業用動物への投与に有用である。それに限定されないが、マイコバクテリウム感染しやすい乳牛などの農業用動物の感染の予防及び/又はその感染との戦いに本発明のワクチンを使用できる。これらのワクチンを、動物及びヒトに投与する技術は、動物とヒトの健康分野のそれぞれの当業者に公知である。
本発明を説明するために以下の実施例を示すが、本発明は実施例に限定されない。
実施例1
ワクチン製造用ベクター
A)V1発現ベクター
発現ベクターV1をpCMVIE−AKI−DHFR[Y.Whangら、J.virol. 61, 1796(1987)]から構築した。AKI遺伝子とDHFR遺伝子を、EcoRIでベクターを切断し、自己連結させて除去した。このベクターはCMVプロモーター中にイントロンAを含まないので、それを、内部SacI部位[B.S.Chapmanら、Nuc.Acids Res. 19, 3979(1991)での番号1855]を欠失したPCRフラグメントとして加えた。PCR反応のために使用した鋳型は、pCMV6a120からのHindIIIとNheIフラグメント(B.S.Chapmanら、上記参照)(hCMV−IE1エンハンサー/プロモーター及びイントロンAを含む)を、pBL3のHindIIIとXbaI部位に連結して作製したpCMVintA−Luxであり、pCMVIntBLを作製した。RSV−Lux(J.R.de Wetら、Mol.Cell Biol. 7, 725, 1987)からの1881塩基対のルシフェラーゼ遺伝子フラグメント(HindIII−SmaIクレノーフィルイン)を、クレノーフィルインとホスファターゼ処理したpCMVIntBLのSalI部位にクローンした。
イントロンAにかかるプライマーは以下の通りである。

PCRフラグメントを、SacIとBglIIで切断し、同じ酵素で切断したベクターに挿入した。
B)V1J発現ベクター
V1Jを作製する目的は、ベクターV1のプロモーターと転写終結要素をより規定されたコンテクストに置き、よりコンパクトなベクターを作製し、プラスミド精製収率を改善するために、ベクターV1からプロモーターと転写終結要素を除去することであった。
V1JはベクターV1と市販のプラスミドであるベクターpUC18から得られる。2つのDNAフラグメントを産生するSspIとEcoRI制限酵素でV1を消化した。異種遺伝子の発現を制御するCMVintAプロモーターとウシ成長ホルモン(BGH)転写終結要素を含む、上記フラグメントの小さい方のフラグメントをアガロース電気泳動ゲルから精製した。それからこのDNAフラグメントの末端を、別の“平滑末端”DNAフラグメントへのこのDNAフラグメントの連結を容易にするために、T4DNAポリメラーゼ酵素を使用して“平滑”にした。
発現ベクターの“骨格”を提供するために、pUC18を選択した。pUC18は高収率のプラスミドを産生することが知られており、それは配列と機能によりよく特徴付けがなされており、小サイズである。HaeII制限酵素での部分消化により、このベクターから完全なlacオペロンを除去した。残っているプラスミドをアガロース電気泳動ゲルで精製し、T4DNAポリメラーゼで平滑末端とし、子ウシ腸アルカリ性ホスファターゼで処理し、上記のCMVintA/BGH要素に連結した。pUC骨格内のプロモーター要素の2つの可能な方向の両方のプラスミドを得た。これらのプラスミドの一つは、大腸菌でDNAのずっと高い収率を示し、V1Jと命名した。このベクターの構造を、接合部領域の配列解析で確認し、次にV1と比較して異種遺伝子が同等又はより高く発現されることが証明された。
C)V1Jneo発現ベクター
V1Jを有する細菌の抗生物質選別のために使用されるampr遺伝子を除去することが必要であった。アンピシリンは大規模発酵で望ましくない可能性があるからである。V1JのpUC骨格からのampr遺伝子を、SspIとEam11051制限酵素で消化して除去した。残っているプラスミドをアガロース電気泳動ゲルで精製し、T4DNAポリメラーゼで平滑末端とし、子ウシ腸アルカリ性ホスファターゼで処理した。トランスポゾン903由来で、pUC4Kプラスミドに含まれる市販のkanr遺伝子を、PstI制限酵素を使用して切出し、アガロース電気泳動で精製し、T4DNAポリメラーゼで平滑末端とした。このフラグメントをV1J骨格と連結し、両方の方向のkanr遺伝子を有するプラスミドを得て、それらをV1Jneo#1と#3と命名した。これらのプラスミドの各々を、制限酵素消化解析、接合領域のDNA配列決定により確認し、これらのプラスミドの各々がV1Jと同様の量のプラスミドを産生することが知見された。これらのV1Jneoベクターの異種遺伝子産物発現はまた、V1Jと同等であった。発現構築物としてV1Jのampr遺伝子と同じ方向のkanr遺伝子を含むV1Jneo#3(以後、V1Jneoと呼ぶ)を選択した。
D)V1Jns発現ベクター
挿入研究を容易にするために、SfiI部位をV1Jneoに加えた。ベクターのBGH配列内のKpnI部位で市販の13塩基対SfiIリンカー(New England BioLabs)を加えた。V1JneoをKpnIで線状化し、ゲルで精製し、T4DNAポリメラーゼで平滑化し、平滑SfiIリンカーに連結した。クローン化された単離体を、制限地図で選択し、リンカーを通る配列決定により確認した。新しいベクターをV1Jnsと命名した。V1Jns(SfiIを有する)での異種遺伝子発現は、V1Jneo(KpnIを有する)での同じ遺伝子の発現と同等であった。
E)V1Jns−tPA
分泌及び/又は膜タンパク質に異種リーダーペプチド配列を与えるために、ヒト組織特異性プラスミノーゲン活性化因子(tPA)リーダーを含むようにV1Jnsを改変した。2種の合成相補的オリゴマーをアニールし、それからBglII消化したV1Jnに連結させた。センスとアンチセンスオリゴマーは、5′−GATC ACC ATG GAT GCA ATG AAG AGA GGG CTC TGC TGT GTG CTG CTG CTG TGT GGA GCA GTC TTC GTT TCG CCC AGC GA−3′(配列番号5)、及び5′−GAT CTC GCT GGG CGA AAC GAA GAC TGC TCC ACA CAG CAG CAG CAC ACA GCA GAG CCC TCT CTT CAT TGC ATC CAT GGT−3′(配列番号6)であった。Kozak配列にはセンスオリゴマーで下線を引いてある。これらのオリゴヌクレオチドは、BglII−開裂配列に連結するために適した突出した塩基を有する。連結後、上流BglII部位を破壊し、下流BglIIを次の連結のために保持する。接合部位と完全tPAリーダー配列の両方を、DNA配列決定により確認した。更に、コンセンサス最適化ベクターV1Jns(=SfiI部位を有するV1Jneo)と適合させるために、T4DNAポリメラーゼによるKpnI部位を平滑化し、SfiIリンカー(New England Biolabs、カタログ#1138)と連結することによって、V1Jn−tPAのBGHターミネータ一領域内のKpnI部位にSfiI制限部位を置いた。この改変は、制限消化とアガロースゲル電気泳動で確認した。
F)pGEM−3−X−IRES−B7(X=任意の抗原遺伝子)
免疫原をコードする遺伝子と免疫刺激タンパク質をコードする遺伝子を同等に発現させる2シストロンワクチン構築物の例として、マウスB7遺伝子をBリンパ腫細胞系列CH1(ATCCから得られた)からPCR増幅した。B7は、主要組織適合性複合体IとIIのコンテクストの中で、抗原による必須の共刺激T細胞活性化をするタンパク質ファミリーの一員である。CH1細胞は、B7mRNAの良好な源である。何故ならば、CH1細胞は、構成的に活性化されているという表現型を有し、B細胞とマクロファージなどの活性化抗原提示細胞によりB7は第1に発現されるからである。これらの細胞は更に、cAMP又はIL−4を使用してin vitroで刺激され、mRNAは標準グアニジウムチオシアネート方法を使用して調製された。このmRNAを用い、GeneAmp RNA PCRキット(Perkin-Elmer Cetus)及びB7翻訳読み枠のB7位置の下流に特異的なプライマーオリゴマー(5′−GTA CCT CAT GAG CCA CAT AAT ACC ATG−3′、配列番号7)を使用して、cDNA合成を行った。以下のセンス及びアンチセンスPCRオリゴマー:5′−GGT ACA AGA TCT ACC ATG GCT TGC AAT TGT CAG TTG ATG C−3′(配列番号8)及び5′−CCA CAT AGA TCT CCA TGG GAA CTA AAG GAA GAC GGT CTG TTC−3′(配列番号9)をそれぞれ用いて、B7をPCR増幅した。これらのオリゴマーは、インサートの末端にBglII制限酵素部位、NcoI制限部位を含むKozak翻訳開始配列、及び3′−末端BglII部位の直前に位置する更なるNcoI部位を提供する。NcoI消化は、NcoIで消化したpGEM−3−IRESにクローニングするのに適したフラグメントを与える。得られたベクターpGEM−3−IRES−B7は、V1Jns−X(Xは抗原をコードする遺伝子を表す)に簡単に移すことができるIRES−B7カセットを含む。
G)pGEM−3−X−IRES−GM−CSF(X=任意の抗原遺伝子)
このベクターは、免疫刺激サイトカインGM−CSFの遺伝子をB7よりむしろ使用するということを除いて、上述の項目Cで記載したものと類似のカセットを含む。GM−CSFは、in vivoで強力な抗腫瘍T細胞活性を誘起することが知見されたマクロファージ分化及び刺激サイトカインである(G.Dranoffら、Proc.Natl.Acad.Sci. USA, 90, 3539(1993))。
H)pGEM−3−X−IRES−IL−12(X=任意の抗原遺伝子)
このベクターは、免疫刺激サイトカインIL−12の遺伝子をB7よりむしろ使用するということを除いて、上述の項目Cで記載したものと類似のカセットを含む。IL−12は、液性応答とは反対の細胞性T細胞−支配経路の方に免疫応答をシフトすることに有力な役割を有することが証明された(L.Alfonsoら、Science, 263, 235, 1994)。
実施例2
ベクターV1R製造
基本的ワクチンベクターの最適化を続ける努力の中で、V1Jnsの誘導体であるV1Rと命名したベクターを製造した。このベクター構築の目的は、不必要のDNA配列をふくまない最小サイズのワクチンベクターであって、V1J及びV1Jnsが与える全体の最適化異種遺伝子発現の特徴と高プラスミド収率を依然として保持するベクターを得ることである。(1)大腸菌複製起点を含む、pUC骨格内の領域を、細菌からのプラスミド収率に影響を与えることなく除去できる;(2)カナマイシン読み枠に続くkanr遺伝子の3′−領域を、もし細菌ターミネーターがその場所に挿入されるならば除去できる;(3)BGHターミネーターの3′−半分からの約300塩基対を、その制御機能に影響を与えることなく除去できる(BGH要素内の元のKpnI制限酵素部位に続く)ということが文献と実験から決定された。
それぞれCMVintAプロモーター/BGHターミネーター、複製起点、カナマイシン耐性要素を表す、V1JnsからのDNAの3種のセグメントを合成するために、PCRを用いてV1Rを構築した。各々のセグメントで唯一の制限酵素を、PCRオリゴマーを使用して各セグメント末端に加えた。CMVintA/BGH用にSspIとXhoI、kanr用にEcoRVとBamHI、orir用にBclIとSalIIである。これらの酵素部位のために、PCR由来DNAセグメントの各々の直接的連結ができ、次に各部位の欠失ができるので、これらの酵素部位を選択した。EcoRVとSspIは、連結に適した平滑末端DNAを残し、BamHIとBclIは,SalIとXhoIのように相補的突出しを残す。PCRによってこれらのセグメントを得た後、各セグメントを上記の適切な制限酵素で消化し、それから3種全部のセグメントを含む唯一の反応混合液で一緒に連結させた。T2ρ独立ターミネーター配列がカナマイシン耐性遺伝子の終結情報を与えることができるように、orirの5′−末端がT2ρ独立ターミネーター配列を含むように設計した。T2ρ独立ターミネーター配列は、この領域に通常存在する。連結接合部の制限酵素消化(8種を超える酵素)及びDNA配列決定により、連結産物を確認した。DNAプラスミド収率及びV1R内でウイルス遺伝子を使用する異種発現はV1Jnsと同様であるように考えられる。達成されたベクターサイズの正味の減少は1346塩基対であった(V1Jns=4.86kb;V1R=3.52kb)。

実施例3
細胞培養とトランスフェクション
M.tb抗原を発現する、安定にトランスフェクトされた細胞系列の調製のために、RD細胞(ヒト横紋筋肉腫ATCC CCL136)を、10%熱不活化ウシ胎児血清、20mM HEPES、4mM L−グルタミン、ペニシリンとストレプトマイシン各100μg/mLを補充したDulbecco改変Eagle培地(DMEM)で、5%CO2下、37℃で生育させた。100mm2プレート当たり1.5×106細胞を植菌し、18時間生育させた。Cellphectキット(Pharmacia)を使用して、プレート当たりTB構築物10μgと共トランスフェクトCat構築物10μgで、細胞をトランスフェクトし、DNAを細胞に添加後5時間でグリセロールショックを与えた(PBS中15%グリセロール、pH7.2、2.5分)。トランスフェクション後72時間で、プレートを冷PBS、pH7.2(2×−10mL)で洗浄し、冷TEN緩衝液(40mM TRIS−Cl、pH7.5、1mM EDTA、150mM NaCl)5mLを加え、擦り取って、培養物を回収した。タンパク質発現の解析のために、Single Detergent Lysis緩衝液(50mM Tris−Cl、pH8.0、150mM NaCl,0.02%NaN3、1%Nonidet P−40、100mM PMSF、2μg/mLアプロチニン、2μg/mLロイペプチン、1μg/mLペプスタチンA)50μLに細胞ペレットを溶解し、氷上音波処理した(2−15秒処理)。溶解液を、13,000×g、4℃で10分間遠心した。タンパク質濃度をBradford法で測定し、レーン当たり細胞抽出タンパク質20μgを、10%TRIS−グリシンポリアクリルアミドゲル(Novex)にかけ、その後ImmobilonP(Millipore)膜に転移した。イムノブロットを、マウスモノクローナル抗体TD17−4(Huygenら、1994, Infect.Immunity 62, 363)の1:20希釈液と一晩反応させ、その後ヤギ抗マウスIgGFcペルオキシダーゼ(Jackson)の1:1000希釈液と1.5時間反応させた。ECLキット(Amersham)を使用して、プロットを発色させた。
実施例4
クローニングとDNA調製
1.V1Jns−tPA−85A(成熟Ag85AとtPAシグナル配列を含む)の構築を、次のプライマーを使用して行った:

Borremansら、1989(Infect.Immunity 57, 3123)の図2からの1600bpのHindIII−SphIフラグメントに800bpのHindIIIを連結して調製したプラスミドp85A.tubから、M.tuberculosisからのAg85Aを増幅させた。得られた2400bpインサートを、BlueScribeM13+のHindIIIとSphI部位にサブクローンした。BlueScribeM13+(VCS/Stratagene)の完全コード配列と隣接領域を、以下の条件下で、記載したプライマーを用いてPCR増幅させた。各100μl反応液は、以下の酵素(Stratagene)を補充した反応緩衝液中にクローン化されたPfuDNAポリメラーゼ(Stratagene)2.5ユニット、200mM dNTP、各プライマー0.5μg、鋳型DNA250ngを含む。Hybaid熱反応器を次のように、プログラムした:94℃で5分の変性、次に(94℃1分、55℃で2分、72℃で3分)が25サイクル、最後に72℃の伸長10分。
増幅DNAを、プロテイナーゼ(Boehringer Mannheim)50μg/mlで37℃30分間消化し、95℃で10分熱処理し、2回のフェノール(クロロホルム−イソアミルアルコール)抽出し、イソプロパノール1容量で沈殿させ、70%エタノールで2回洗浄し、乾燥させ、H2O20μlに溶解した。増幅DNA3μgを、BglII(Boehringer Mannheim)40ユニットで消化し、907bpフラグメント(85A−C1の場合)を1%アガロースゲルで単離し、製造業者の指示に従い“Prep a Gene(BioRad)”で抽出した。
このフラグメント5ngを、連結緩衝液中にT4DNAリガーゼ(Amersham)2.5ユニットを含む反応液10μl中で16時間14℃で、BglII消化−脱リン酸化V1Jns.tPAベクター20ngに連結し、コンピテントDH5大腸菌(BRL)を形質転換し、カナマイシン(50μg/ml)含有LB寒天培地にプレートした。形質転換体を拾い上げ、プラスミドDNAをBglII(インサートの存在を確認するため)とPvuIIで制限消化し、その方向を確認した。
2.以下のプライマーを使用して、V1Jns−85A[C−2](成熟Ag85Aを含むが、シグナル配列を含まない)の構築を行った。

制限解析後、全ての構築物のベクター接合部を通って部分配列決定を行う。大規模DNA調製は、本質的にMontgomery,D.L.ら、(前出)に記載の通り行った。
プラスミド構築物の特徴付けを、ベクター−インサート接合部の制限地図と配列決定で行った(図1−6)。結果は、公表されたM.tb配列データと一致し、開始コドンは、各構築物で完全であることを示した(図7)。クローニングの結果として挿入されたM.tbAg85に無関係の種々の付加的アミノ酸残基も示す。
実施例5
V1Jns.tPAプラスミドからのM.tbタンパク質の発現
10%熱不活化ウシ胎児血清、2mM L−グルタミン、25mM HEPES、50U/mlペニシリン、50μg/mlストレプトマイシン(全てBRL−Gibco)で補充した高グルコースDMEM中の6穴組織培養クラスターに、9.5cm2当たり1.2×106細胞密度で、使用1日前に横紋筋肉腫細胞(ATCC CCL136)を移植した。RD細胞の各9.5cm2穴当たり5−15μgを使用することを除いて、キット指示に従ってPharmacia CellPhect試薬を使用して、フェノール:クロロホルム抽出塩化セシウム精製プラスミドDNAを、リン酸カルシウムで沈殿させた。リン酸カルシウム−DNA沈殿の添加後6時間で、培養物にグリセロールショックを与えた。再フィード後、培養物を2日間インキュベートし、それから回収した。
1μMロイペプチン、1μMペプスタチン、300nMアプロチニン、10μM TLCKを補充した1×RIPA(0.5%SDS、1.0%トリトンX−100、1%デオキシコレートナトリウム、1mM EDTA、150mM NaCl、25mM TRIS−HClpH7.4)中で、トランスフェクト培養物の溶解液を調製し、簡単に音波処理し、粘度を減少させた。10%Tricineゲル(Novex)での電気泳動で、溶解液を分離し、それからニトロセルロース膜に転移させた。M.tbモノクローナル抗体17/4と32/15(Huygenら、1994, Infect.Immunity 62, 363)を用いてイムノブロットの処理をし、ECL検出キット(Amersham)で発色させた。
RD細胞の一過性トランスフェクションで、M.tb抗原85複合体遺伝子の発現を証明した。トランスフェクトされた、又は偽のトランスフェクトを行った細胞の溶解液を、SDS PAGEで分離し、イムノブロティングで解析した。V1Jns.tPA−85A(C1)、V1Jns.tPA−85A(C2)、V1Jns.tPA−85A(C3)、及びV1Jns.tpA−85B(C1)をトランスフェクトしたRD細胞は、見かけの分子量約30−32kDaを有する免疫反応生成物タンパク質を発現することを図8は示す。
実施例6
PNVによる免疫化及びin vivoでの抗原85タンパク質の発現
生理食塩水中の5mgケタミンHCl(Aveco, Fort Dodge, IA)と0.5mgキシラジン(Mobley Corp., Shawnee, KS)の混合液の腹腔内(i.P.)注射で、5〜6週齢の雌のBALB/cとC57BL/6マウスを麻酔した。後脚を70%エタノールで洗浄した。生理食塩水中に懸濁したDNA(2mg/ml)100μl(各脚毎に50μl)を3回動物に注射した。免疫化後17−18日で、血清サンプルを集め、抗Ag85抗体の存在を分析した。図9は、Ag85DNA注射マウス(C1)からの血清の特異的イムノブロット反応性を示すが、遺伝子インサートを含まない対照DNA(V1J)を注射されたマウスではそうではないことを示す。精製抗原85A300ngに対して少なくとも1:160血清希釈まで反応性が検出された(図9B)。Ag85DNAを注射すると、Ag85DNAがin vivo発現し、それを利用して、BALB/cとC57BL/6(B6)マウスの両方で抗体応答が発生することを上記は証明する。
実施例7
抗原85−特異的T−細胞応答
Huygenら、1992(Infect. Immunity 60, 2880)に記載されているように、特異的再刺激に対する応答でのサイトカイン分泌を、ワクチン注射されたマウスからの脾臓細胞で分析した。特に、M.bovisBCGからの培養物濾液(CF)タンパク質、精製抗原85A、又はC57BL/6マウスの公知のT−細胞エピトープに対応する20−merペプチド(p25)(アミノ酸241−260)と、脾臓細胞をインキュベートした。V1Jns.tPA85A(C1)(100μg)を用いて、3週の間隔で3回マウスを免疫化し、最終注射後17日で分析した。IL−2、インターフェロン−γ(IFN−γ)、IL−6の場合、サイトカインをバイオアッセイを使用してアッセイし、IL−4、IL−10の場合、ELISAでアッセイした。V1Jns.tPA85A(C1)でワクチン接種したBALB/cとC57BL/6マウスの両方で、実質的なIL−2とIFN−γ産生が観察された(図10−13)。更に、C57BL/6マウスはまた、H−2b−制限T−細胞エピトープにも反応した(図13)。IL−4、IL−6、IL−10レベルは、V1Jns.tPAワクチン接種マウスで増加しなかった(図14−16)。これらの結果は、ヘルパーT−細胞応答のTh1タイプが、DNAワクチンで起ったことを示す。
実施例8
マイコバクテリウム攻撃からの防御
M.tbDNAワクチンの効力を試験するために、生きているM.bovisBCG(0.5mg)の静脈内注射で、マウスを攻撃し、脾臓と肺でBCG増殖を分析した。対照として、攻撃されたワクチン接種されていないマウス(一次感染)とDNA注射の時にBCGでワクチン接種された攻撃されたマウス(二次感染)でBCG増殖を測定した。V11Jns.tPA85A(C1)−ワクチン接種されたマウスの肺でのコロニー形成単位(CFU)数は、一次感染マウス又は対照DNAV1Jでワクチン接種されたマウスと比較して実質的に減少した。C57BL/6マウスでは、CFUは攻撃後8日で83%減少し(図17)、BALB/cマウスでは、CFUは20日に65%減少した(図18)。BALB/cマウスの脾臓では、攻撃後20日でCFUは約40%減少し(図19)、C57BL/6マウスの脾臓では、攻撃後8日で同じだけ減少した(図20)。それ故、M.tbDNAワクチン注射後観察された免疫応答は、生きたM.bovis攻撃モデルでの防御を示した。

Claims (19)
- マイコバクテリウム・ツバーキュロシス(Mycobacterium tuberculosis)またはマイコバクテリウム・ボビス(Mycobacterium bovis)曝露から哺乳動物を保護するためのワクチンであり、医薬上許容される担体とともに、転写プロモーターと作動可能なように結合しているマイコバクテリウム・ツバーキュロシス抗原85Aタンパク質、マイコバクテリウム・ツバーキュロシス抗原85Bタンパク質またはマイコバクテリウム・ツバーキュロシス抗原85Cタンパク質をコードするDNAポリヌクレオチド配列の少なくとも1つを含み、当該DNA配列が発現ベクター内に含まれることを特徴とするワクチン。
- 前記プラスミドが2シストロンである請求項2に記載のワクチンであって、前記プラスミドがさらに免疫調整または免疫刺激遺伝子をコードする追加のヌクレオチド配列を含む、請求項2に記載のワクチン。
- 前記の追加のヌクレオチド配列がGM−CSF、IL−12、インターフェロンおよびT細胞共刺激性タンパク質のB7ファミリーの一員からなる群から選択される請求項3に記載のワクチン。
- 前記の転写プロモーターがサイトメガロウイルスプロモーターであり、前記発現ベクターがさらにイントロンA配列およびウシ成長ホルモン転写終結要素を含む請求項1に記載のワクチン。
- 前記ヌクレオチド配列がさらに前記タンパク質に作動可能なように結合しているシグナル配列をコードする請求項1に記載のワクチン。
- 前記シグナル配列がヒト組織特異性プラスミノーゲン活性化因子をコードする遺伝子由来の真核生物性シグナル配列である請求項6に記載のワクチン。
- マイコバクテリウム・ツバーキュロシスまたはマイコバクテリウム・ボビスによる感染に対して哺乳動物を免疫するためのDNAワクチンの製造に用いるプラスミドであって、転写プロモーターと作動可能なように結合しているマイコバクテリウム・ツバーキュロシス抗原85Aタンパク質、マイコバクテリウム・ツバーキュロシス抗原85Bタンパク質またはマイコバクテリウム・ツバーキュロシス抗原85Cタンパク質をコードするDNAポリヌクレオチド配列の少なくとも1つを含むプラスミド。
- 2シストロンであり、さらに免疫調整または免疫刺激遺伝子をコードする追加のヌクレオチド配列を含む、請求項10に記載のプラスミド。
- 前記の追加のヌクレオチド配列がGM−CSF、IL−12、インターフェロンおよびT細胞共刺激性タンパク質のB7ファミリーの一員からなる群から選択される請求項12に記載のプラスミド。
- 前記の転写プロモーターがサイトメガロウイルスプロモーターであり、当該プラスミドがさらにイントロンA配列およびウシ成長ホルモン転写終結要素を含む請求項10に記載のプラスミド。
- 前記ヌクレオチド配列がさらに前記タンパク質に作動可能なように結合しているシグナル配列をコードする請求項10に記載のプラスミド。

- 請求項10ないし18のいずれか1項に記載のマイコバクテリウム・ツバーキュロシスまたはマイコバクテリウム・ボビスによる感染に対して哺乳動物を免疫するためのDNAワクチンの製造に用いるプラスミドであって、当該哺乳動物が家庭の動物または家畜であるプラスミド。
