Description
懸濁され得、そして経口、直腸及び尿道適用又は生物中
への注入のためにたとえば超音波検査及び他の医学的な
用途のためにこの形で使用され得る有機ポリマーエンベ
ロープにより包まれた空気又はガス充填マイクロカプセ
ル又はマイクロバルーンに関する。

マイクロバルーンは存在する類似する生成物にわたって
改良された性質を有する懸濁液を付与するために水性液
体キャリヤーにすぐに分散され得る。従って、投与のた
めに容易なキャリヤー液体中、マイクロバルーンの懸濁
液もまた、本発明の一部である。

ルーンのような微小球体は、超音波検査のために非常に
効果的な超音波レフレクターであることは良く知られて

不溶性固体と前記液体とを混合することにより生成され

れるべきであり;実際、バブルはしだいに消出するの
で、その濃度は、その期間の後、使用のためには低過ぎ
る。
ロバブルに関するもう1つの問題は大きさである。通常
許容される場合、小さな血管を通しての容易な透過を可
能にするために有用な大きさのマイクロバブルは約0.5
〜10μmの範囲であり;大きなバブルに関しては、凝集
及び連続的なエンボリの危険性が存在する。たとえば、
US−A−4,466,442(Schering) に開示されるバブル懸
濁液においては、界面活性剤、たとえばレシチン、脂肪
酸及び脂肪アルコールとポリオキシエチレン及びポリオ
キシエチル化ポリオール、たとえばソルビトール、グル
コール及びグリセロールとのエステル及びエーテル、コ
レステロール又はポリオキシ−エチレン−ポリオキシプ
ロピレンポリマーの水溶液が、粘度上昇性及び安定性化
合物、たとえば単糖類及び多糖類(グルコース、ラクト
ース、スクロース、デキストラン、ソルビトール);ポ
リオール、たとえばグリセロール、ポリグリコール;及
びポリペプチド、たとえばタンパク質、ゼラチン、オキ

振盪され、ここで単に約50%のマイクロバブルが、多く

50μm以下である。

製造された実体的なエンベロープ、たとえば明確な機械
強度を有するポリマー膜を有する。言い換えれば、それ

微小球体がUS−A−4,718,433(Feinstein)に開示され
ている。これらの微小球体は、5%の血清アルブミンの

り製造され、そして2〜20μm、主に2〜4μmの有径

トロピックゼラチン化によるポリマー被覆された微小球
体の調製を報告している。その引例は、微小球体を生成
するためのいくつかの技法を言及し;1つの場合、アル
ギネート溶液が、1.2%の水性CaCl2 の槽において硬化
される発生期空気充填カプセルの噴霧を生成する針を通
してエアジェットで強制された。ガス及び液体の同時押
出を包含する第2の場合、ガスバルブがトリプル−ベア
ールドヘッドにより発生期カプセル中に導入され、すな
わち空気が中央毛細管中に注入され、ここでアルギネー
ト溶液がその毛細管と同軸に配置された大きな方の管を

ば、それらの剛性のために、膜は、微小球体が血流を通

並びに続く請求項に定義される本発明のマイクロバルー
ンにより達成される。さらに、本発明の微小球体は一般
的に比較的短い寿命であり、すなわち選択されたタイプ
のポリマーを用いることによって前記代謝問題を処理す
るために生物分解性に対して敏感なように製造され得る
が、この性質(実際、加工パラメーターにより調節され
る)は、マイクロバルーンが、不明確に安定している乾
燥条件下で貯蔵され、そして輸送され得又はその膜がキ
ャリヤー液体に対して実質的に耐湿性にされ(劣化は注

い。第1の場合、乾燥粉末形で供給されるマイクロバル
ーンは、使用の前、ある割合の水性相キャリヤーと共に
単純に混合され、ここで前記割合は必要に依存して選択
される。これは、濃度が随意に選択され得、そして前記
108 /mLをはるかに超える、すなわち105 〜1010の範囲
の初値が容易に許容できるので、従来技術の生成物以上
の追加の利点であることを注目すべきである。本発明の
方法(この後に開示される)は、広範囲の多孔度の調節
を可能にし;従って、実質的に耐湿性の膜を有するマイ

てまたそれ自体市販され得る)が容易に製造され得るこ
とが注目されるべきである。

なわち乳化剤の性質(界面活性剤がより効果的なほど、
すなわち親油性バランスに対する親水性バランスが大き
いほど、より小さな液滴が得られる)及び攪拌条件(よ
り早く且つよりエネルギッシュな攪拌が、より小さな液
滴を生成する)を変えることによって調節され得る。

マー包封微小ビーズの懸濁液の形でナフタレンを回収
し、そして最後に、微小ビーズを昇華にゆだねることに

ロバルーンが生成される。他の例も存在し、ここでポリ
マーが揮発性疎水性有機溶媒及び水溶性有機溶媒を含ん

ロバルーンエンベロープの微孔性構造(数nm〜数百nmの
孔サイズ又は50〜500nm の範囲の厚さのマイクロバルー
ンエンベロープ)はレジリエンスの要因であり、すなわ
ち微小球体は破壊を伴わないで圧力変化を容易に受け入
れることができる。好ましい孔サイズの範囲は約50〜20
00nmである。

発明のマイクロバルーンのエンベロープにおける微小孔
の存在は、超音波検査応答にも関係しているように思わ
れ、すなわちすべての他の要因は一定である場合、微孔
性小胞は対応する非多孔性小胞よりも一層効果的な超音
波検査用シグナルを提供することが注目されるべきであ
る。その理由は知られていないが、しかしガスが密閉構
造において共鳴して存在する場合、その構造体の制動性
は、それが多孔性か又は非多孔性かのいづれかで異なる
ことが予測され得る。

いて疎水性溶媒として便利である。これらは、炭化水
素、たとえばn−オクタン、シクロオクタン、デメチル
シクロヘキサン、エチル−シクロヘキサン、2−、3−
及び4−メチル−ヘプタン、3−エチル−ヘキサン、ト
ルエン、キシレン、2−メチル−2−ヘプタン、2,
2,3,3−テトラメチルブタン及び同様のものを包含
する。エステル、たとえばプロピル及びイソプロピルブ
チレート及びイソブチレート、ブチル−ホルメート及び
同様のものもまた、この範囲において便利である。凍結
乾燥の他の利点は、空気の代わりにガスの減圧下で操作
することであり、それによってガス充填マイクロバルー

疎水性相の選択によるこの調節は、マイクロバルーンの
形成の間、水蒸発の速度を調節するために、安定剤の選
択により及びその濃度を調整することによりさらに改良

行なわれ、そしてさらに論じる必要はない。

ましくは例3及び4の方針に従って主に調製され得る。

寿命は約30分又はそれ以上であった。非生物分解性であ
るポリスチレンは注入超音波検査法のためには好ましく
ないが、しかし消化管の検査のためには有用である。こ
の例は、本発明の方法の実施可能性を明確に確立する。

を、CHCl3 7.5mLに溶解し、溶液(1)を得た。
パラフィン−ワックス(M.P. 54〜56度C)20mg を含む溶
液(2)を調製し、そして1.2gのCHCl3 もまた含むPl
uronic F-108(酸化エチレン及び酸化プロピレンのブ
ロックコポリマー)の水溶液(0.13重量%)150mLに乳化
した。乳化を、Polytronヘッドを用いて 7000rpmで1分
間、室温で行なった。次に溶液(1)を攪拌(7000rpm)

螺施攪拌機(500rpm)と取り換え、そして攪拌を室温(22
度C)で約3時間続けた。その懸濁液を50μmのスクリ
ーンに通し、そしてブロックに凍結し、続いて−20〜0
度Cの温度で高い真空下で(−60〜−80度Cに達する)
蒸発せしめた。乾燥状態で安定する空気充填のマイクロ
バルーン0.264 g(88%)を得た。

れを−40度Cで凍結し、そして約1トル下で凍結乾燥せ
しめた。残留物、すなわちひじょうに細かな粉末1.32g
を、蒸留水40mLに取り、手で3分間攪拌した後、粒子分
析器(MalvernからのMastersizer)を用いて測定される場
合、平均サイズ4.5μmのマイクロバルーンのひじょう
に均質な分散液を得た。マイクロバルーンの濃度は2×
109 /mLであった(Coulter Counter) 。この懸濁液は、
約1時間持続する強い超音波検査用シグナルを付与し
た。

MEG 0.1g、THF 70mL、シクロオクタン1mL及びPluron

は高分子量炭化水素は添加されなかった。乳濁色のエマ
ルジョンを、残留物が約 100mLになるまで27度C/10ト
ルで蒸発せしめ、次にそれを50μmメッシュ上でスクリ
ーンし、そして凍結した。凍結ブロックの蒸発を、乾燥
まで0.5〜1トルで行なった。その収量は、界面活性剤
の存在のために0.18gであった。これを蒸留水10mLに分
散し、そしてCoulter Counter により計数した。測定さ
れた濃度は1.43×109 個のマイクロカプセル/mLであ
り、粒子分析器(MalvernからのMastersizer)により測定
される場合、5.21μmの平均粒子を有することが見出さ
れた。その分散液を 100倍、すなわち約1.5×107 個の
マイクロカプセル/mLに希釈し、そして超音波検査性に
ついて測定した。超音波シグナルの振幅は2.25MHz より
も7.5MHzで5倍大きかった。これらのシグナルは、長期

キシグラス検体用ホルダー(直径30mm)、一定温度の水
浴に含浸されたトランスジューサーホルダー、40dBの一
定増量での外部プレー増幅器及び−40〜+40dBに調節で

ccutron M3010JS)及び互換性の13mmの末焦点トランスジ
ューサーから成るパルス−音響システムにより超音波検
査を行なった。
酸−ラクトンコポリマーにより交換される場合〔ここで
ラクトンはδ−ブチロラクトン、δ−ラレロラクトン又
はε−カプロラクトンである(Fukuzakiなど., J.Bioch
emical Mater.Res. 25 (1991),315〜328 を参照のこ
と)〕、類似する好ましい結果が得られた。また類似す
る関係において、ポリアルキルシアノアクリレート及び
特に90:10のコポリマーであるポリ(DL−ラクチド−
コグリコリド)が満足する結果を付与した。最後に、好
ましいポリマーは、“Resomer R-206”又はResomer R
-207として売られているCampany Boehringer Ingelheim
からのポリ(DL−ラクチド)である。
分散液(1.43×109 /mL)0.1〜2mLを実験用犬の末梢
静脈に注入した後、Acuson-128装置を用いて行なわれ
た。右心臓の通常予測されるコントラスト増強イメージ
ングの後、心内膜の明確な外形でもって左心臓のはっき
りした及び永続的なシグナル増強が観察され、それによ
って、ポリ−POMEG により製造されたマイクロバルーン

法。 - 【請求項26】 添加物が、リン脂質などの両親媒性化
合物、又は低分子量ポリマーのような両親媒性化合物を
含み、膜ポリマー中でのその存在が、水性液体へのマイ
クロバルーンの透過性を高めるであろう、請求項24に
記載の方法。 - 【請求項27】 水性相中での乳化に供された疎水性相
が、水溶性溶媒をも含み、この水溶性溶媒が、乳化の間
に上記水性相中に希釈される間に、滴のサイズの減少を
助け、そして段階(4)が行なわれる前にポリマーの界
面沈殿を誘導するであろう、請求項18に記載の方法。 - 【請求項28】 超音波検査画像形成で使用されるため
の請求項1〜14のいずれか1項に記載のマイクロバル
