増強されたmPGES−1発現又は活性により媒介された症状を予防又は抑制するための治療に使用される化合物に関する。

a)急性及び慢性炎症;皮膚炎、湿疹、乾癬、バムス(bums)、尋常性ざ瘡、化膿性汗腺炎及び組織外傷などの皮膚疾患;炎症性腸疾患、クローン病、潰瘍性大腸炎、憩室炎、過敏性腸疾患(IBS)、消化性潰瘍、膀胱炎、(慢性)前立腺炎、膵炎又は腎炎などの内臓疾患;インフルエンザ、鼻炎、咽頭炎、扁桃炎、結膜炎、虹彩炎、強膜炎、耳炎及びぶどう膜炎などの耳、鼻、口及びのどの疾患;ウイルス及び細菌感染;炎症関連食欲不振;アレルギー;骨盤内炎症性疾患;再灌流傷害;移植片拒絶;腱炎、血管炎及び静脈炎;
b)急性疼痛、慢性疼痛、神経障害性疼痛、侵害受容性疼痛、痛覚過敏、中枢性感作に関連する疼痛、アロディニア炎症性疼痛、内臓痛、がん性疼痛、外傷性疼痛、歯又は手術痛、術後痛、分娩陣痛(delivery pain)、産痛(childbirth ache)、持続痛、末梢媒介性疼痛(peripheral mediated pain)、中枢媒介性疼痛(central mediated pain)、慢性頭痛、片頭痛、副鼻腔炎性頭痛、緊張型頭痛、幻肢痛、末梢神経傷害化学療法性疼痛及びがん性疼痛;
c)関節炎、変形性関節症、若年性関節炎、関節リウマチ、強直性脊椎炎、痛風、リウマチ熱、滑液包炎、全身性エリテマトーデス(SLE)及び多発性硬化症などの自己免疫性疾患;
d)喘息、慢性閉塞性肺疾患(COPD)、サルコイドーシス及び肺線維症などの呼吸障害又は肺疾患;
e)脳がん、前立腺がん、腎がん、肝がん、膵がん、胃がん、乳がん、肺がん、頭頸部がん、甲状腺がん及、神経膠芽種、黒色腫、リンパ腫、白血病、皮膚T細胞リンパ腫及び皮膚B細胞リンパ腫などのがん;
f)糖尿病性血管傷害、糖尿病性神経障害及び糖尿病性網膜症を含む糖尿病合併症;
g)アルツハイマー病、パーキンソン病、ハンチントン病及び筋萎縮性側索硬化症などの神経変性傷害、並びに
h)アテローム性動脈硬化症、血栓症、卒中及び冠状動脈性心疾患などの心血管疾患
からなる群から選択される疾患又は状態の治療において、増強されたmPGES−1発現又は活性により媒介される症状の予防又は抑制のために使用される。

Tは、コアクロマニル又はクロマニルキノンフレームワークを有する及び2位で置換されているカルボン酸部分を有する水溶性ビタミンE誘導体であり、ここでTは、アミド部分を形成するように、カルボン酸部分を介して窒素に接続し、
Lは、炭素、窒素及び酸素から選択される1〜10個の任意選択で置換されている主鎖原子を含む、アミド窒素原子と遠位窒素原子の間にあるリンカーであり、

方法及び材料
化学薬品及び抗体
cPGES、mPGES−1及びmPGES−2に向けられる抗体は、Cayman Chemicalsから購入した。COX−1、COX−2及びアクチンに向けられる抗体は、それぞれR&D Systems、Thermo Fisher Scientific及びSigma−Aldrichから購入した。COX阻害剤のセレコキシブ及びインドメタシン、並びにリポ多糖はSigma−Aldrichから購入した。mPGES−1阻害剤のMK866及びPF9184、並びにPGH2はCayman Chemicalsから購入した。IL−1βは、Cell Signaling Technologiesから購入した。
RAW264.7細胞(Sigma−Aldrich,St−Louis)を、10%FBS(Greiner Bio−one)及び抗体(100U/mlのペニシリン、100μg/mlのストレプトマイシン)を含有するDMEM(Thermo Fisher Scientific)に5%CO2の加湿雰囲気下、37℃で維持した。細胞を80%の集密に増殖させ、削り落とし、次いで実験の前に、96ウエルプレート(2×104細胞/ウエル)又は6ウエルプレート(4×105細胞/ウエル)のいずれかで6〜24時間培養した。
細胞を、10%FBS(#758093、Greiner Bio−one)、100IU/mlのペニシリン及び100μg/mlのストレプトマイシン(#30−002−CI、Corning)を含有するM199,HEPES(Thermo Fischer Scientific)に5%CO2の加湿雰囲気下、37℃で維持した。細胞を、継代数20に達するまで、4〜5日毎にトリプシン処理して継代させ、次いで廃棄した。細胞を80%の集密に増殖させ、トリプシン処理し、次いで実験の前に、96ウエルプレート(4×103細胞/ウエル)で24時間培養した。
培養培地中のPGE2、PGD2及び6−ケト−PGF1αの濃度は、PGE2高感度EIAキット(Enzo Life Science)、プロスタグランジンD2−MOX ELISAキット(Cayman Chemicals)及び6−ケトプロスタグランジンF1αELISAキット(Cayman Chemicals)を製造会社の使用説明書に従ってそれぞれ使用して、酵素結合免疫吸着測定法(ELISA)によって決定した。各ウエルから培養培地の試料(100μl)を採取し、アッセイ緩衝液で希釈した。それぞれのプロスタノイドの値は、標準曲線を使用して計算し、図の説明に示されているように正規化した。
処理した後、細胞を削り落として採取し、放射性免疫沈降検定緩衝液(50mMのトリス(Tris)−HCl、pH8.0、150mMのNaCl、0.2%のトリトン(Triton)、1×プロテアーゼ阻害剤及び0.1mg/mlのDNアーゼ)に溶解した。試料のタンパク質濃度をBradfordアッセイにより決定した。各試料からタンパク質の等量をSDS−ポリアクリルアミドゲル電気泳動(SDS−PAGE)により分離し、PVDF膜に電子移動させた。膜を、オデッセイ(Odyssey)遮断緩衝液(Li−Cor)により室温で1時間遮断し、次いでTBST中においてmPGES−1(1:200希釈)、mPGES−2(1:200希釈)、cPGES(1:200希釈)、COX−1(1:250希釈)、COX−2(1:500希釈)又はβ−アクチン(1:10000希釈)に対する一次抗体と共に4℃で一晩インキュベートした。膜をTBSTで3回洗浄した後、二次抗体(ヤギ抗ウサギIgG(H+L)二次抗体アレクサフルオール(Alexa Fluor)488又はヤギ抗マウスIgG(H+L)二次抗体アレクサフルオール647、Thermo Fisher Scientific)を、1:10000の希釈により室温で1時間加えた。ブロットの蛍光走査を、オデッセイ赤外線画像化システム(Odyssey Infrared Imaging System)(Li−Cor)を使用して実施した。
PGES活性は、PGH2からPGE2への変換を評価することによって測定した。簡潔には、1μg/mLのLPSと24時間インキュベートした後、細胞を採取し、300μlの1MトリスHCl(pH8.0)中における超音波処理(1分間の間隔を置いて、それぞれ10秒間で3回)によって溶解した。溶解産物を12,000gにより4℃で10分間遠心分離した後、上澄みを回収し、100,000gにより4℃で1時間更に遠心分離した。ペレット(ミクロソーム膜)を100μlの0.1MトリスHCl、pH8.0(プロテアーゼ阻害剤を含有)に再懸濁し、ミクロソームPGES活性の測定に付した。PGES活性の測定では、50μgのタンパク質に同等の各試料のアリコートを、試験化合物の不在下又は存在下で、2mMのグルタチオン(Sigma−Aldrich)及び14μMのインドメタシンを含有する0.1mlの1MトリスHCl中において2μgのPGH2と共に24℃で60秒間インキュベートした。反応を、100mMのFeCl2の添加により停止させ、20〜25℃で15分間更にインキュベートした。反応混合物を遠心分離した後、上澄みのPGE2濃度を、PGE2高感度EIAキット(Enzo Life Science)により製造会社の使用説明書に従って測定した。
試験化合物の不在下又は存在下で、1μg/mLのLPSにより6時間処理した後、全RNAをトリゾール(Trizol)試薬(Thermo Fisher Scientific)により細胞から単離した。RNAを、第1鎖cDNA合成キット(Thermo Fisher Scientific)によりcDNAに転写した。cDNAを95℃で10分間変性させた。COX−1、COX−2、mPGES−1、mPGES−2、cPGES及びPPIAの特異的DNAフラグメントを、95℃で15秒間、60℃で60秒間にわたり40サイクルでSYBRグリーン(Green)(Roche Life Science)サイクラー(Cycler)を用いるPCRにより増幅した。COX1に使用したオリゴヌクレオチドプライマーは、5’−GATTGTACTCGCACGGGCTAC−3’(順方向)及び5’−GGATAAGGTTGGACCGCACT−3’(逆方向)であり、COX2には、5’−AGGACTCTGCTCACGAAGGA−3’(順方向)及び5’−TGACATGGATTGGAACAGCA−3’(逆方向)であり、mPGES−1には、5’−AGCACACTGCTGGTCATCAA−3’(順方向)及び5’−CTCCACATCTGGGTCACTCC−3’(逆方向)であり、mPGES−2には、5’−GCTGGGGCTGTACCACAC−3’(順方向)及び5’−GATTCACCTCCACCACCTGA−3’(逆方向)であり、cPGESには、5’−GGTAGAGACCGCCGGAGT−3’(順方向)及び5’−TCGTACCACTTTGCAGAAGCA−3’(逆方向)であり、PPIAには、5’−AGGGTGGTGACTTTACACGC−3’(順方向)及び5’−GATGCCAGGACCTGTATGCT−3’(逆方向)であった。PCR増幅を、陰性対照としてcDNAを有さない試料においても実施した。COX−1、COX−2、mPGES−1、mPGES−2及びcPGESの相対的遺伝子発現は、ビヒクル及び処理群における発現を比較するΔΔ(デルタデルタ)CT方法によって決定した。PPIAを基準遺伝子として使用した。
化合物I−IVb−XはPGE2のレベルを選択的に減少する。
RAW264.7マクロファージ細胞を、炎症性刺激LPS単独若しくは化合物I−IVb−X(一般構造(I)の化合物、式中、Tは、S,R配置の一般構造(IVb)のものであり、化合物Xとしては、以下が適用される:L=L19;R1=H;R2−R2’=L3;R3=H)の増加濃度との組合せ、NSAIDインドメタシン又はコキシブのセレコキシブで処理した。24時間のインキュベーションの後、PGE2、PGD2及び6−ケトPGF1aのレベルを細胞上澄みにおいてELISAにより定量化した。予想したとおり、LPSは、3つのプロスタグランジンの産生を効率的に誘導し(図2A〜2C)(Ikeda−Matsuo et al.,2005)、2つのCOX阻害剤のインドメタシン及びセレコキシブが全て用量依存的に低減することができた(図2E〜2F)。予想外なことに、化合物I−IVb−Xは、PGE2のレベルを用量依存的に低減することができたが、他の2つのプロスタグランジンPGD2及び6−ケトPGF1aに対して効果を有さず、PGI2の安定した代謝産物を通常PGI2の代替物として測定した(図2D)。同様の結果を、ヒト一次皮膚線維芽細胞における化合物I−IVb−Xから得た。線維芽細胞と炎症性刺激LPS又はIL−1βのいずれかとの24時間のインキュベーションの後、上澄みにおけるPGE2及びPGD2レベルは強く増加した(図3A及び3B)。化合物I−IVb−Xは、LPS(図3C)又はIL−1β(図3D)のいずれかで処理された細胞の上澄みにおいてPGE2のレベルを効率的に低減することができたが、PGD2ではできなかった。
RAW264.7マクロファージ細胞を、炎症性刺激LPS単独により又は化合物I−IVb−Xの増加濃度とのみ合わせにより処理した。24時間のインキュベーションの後、mPGES−1、mPGES−2、cPGES、COX−1及びCOX−2タンパク質のレベル、並びにRNAのレベルを、細胞において、ウエスタンブロット又はqPCRにより、それぞれ定量化した。予想したとおり、LPSは、タンパク質における2つの誘導性酵素mPGES−1及びCOX−2の発現(図4A〜4C)、並びにRNA(図5A及び5B)のレベルを効率的に誘導したが、タンパク質において構成的に発現した酵素mPGES−2、cPGES及びCOX−1(図4A、4D〜4F)、並びにRNA(図5C〜5E)のレベルに対して効果を有さなかった。化合物I−IVb−XはmPGES−1タンパク質(図4B)及びRNA(図5A)のLPS誘導発現を用量依存的に低減することができたが、COX−2発現(図4C及び5B)に対して効果を有さなかった。化合物I−IVb−Xは、他の3つの構成的酵素mPGES−2、cPGES及びCOX−1(図4D〜4F及び図5C〜5E)に対して効果を有さなかった。これらの結果は、化合物I−IVb−Xが、炎症性刺激LPSにより誘導されたmPGES−1酵素の発現を選択的に阻害できることを示し、PGE2のみを低減し、他のプロスタグランジンを低減しない選択性を説明している。
RAW264.7マクロファージ細胞を炎症性刺激LPSで処理して、mPGES−1の発現を増加させた。24時間のインキュベーションの後、ミクロソームを単離し、増加濃度の化合物I−IVb−X又は単一濃度の前記のmPGES−1阻害剤MK866及びPF9184に30分間暴露した。次いでmPGES−1の活性を、PGH2からPGE2への変換としてミクロソーム画分においてアッセイした。結果は、化合物I−IVb−Xにより又は2つの陽性対照MK866及びPF9184により処理した精製ミクロソームにおいて、mPGES−1活性が減少したことを示す(図6A)。
実施例1に記載された方法を使用して、追加の化合物を試験した。表1は、試験した化合物の概要を、対応するIC50値と共に提示する。図6B、6C及び6Dは、I−IVb−AE、I−IVb−A−HCl及びI−IVb−Iそれぞれの対応する阻害曲線を示す。

R1及びR2は、水素(H)、C1〜C6アルキル若しくはC1〜C6アルケニルからそれぞれ独立して選択され、又はR1及びR2は、一緒に結合し、それによって前記アミド窒素原子と前記遠位窒素原子の間に第2のリンカーを形成し、又はR1は、環状構造におけるリンカーLの主鎖原子と結合し、及び/若しくはR2は、環状構造における前記リンカーLの主鎖原子と結合し、

により表される、請求項3に記載の使用のための化合物。 - 増強されたmPGES−1発現又は活性により媒介される前記症状が少なくとも、炎症、疼痛、腫脹、発熱、血管新生及び食欲不振のうちの1つ又は複数を含む、請求項1〜4のいずれか一項に記載の使用のための化合物。
- 前記化合物が、
a)急性及び慢性炎症;皮膚炎、湿疹、乾癬、バムス、尋常性ざ瘡、化膿性汗腺炎及び組織外傷などの皮膚疾患;炎症性腸疾患、クローン病、潰瘍性大腸炎、憩室炎、過敏性腸疾患(IBS)、消化性潰瘍、膀胱炎、(慢性)前立腺炎、膵炎又は腎炎などの内臓疾患;インフルエンザ、鼻炎、咽頭炎、扁桃炎、結膜炎、虹彩炎、強膜炎、耳炎及びぶどう膜炎などの耳、鼻、口及びのどの疾患;ウイルス及び細菌感染;炎症関連食欲不振;アレルギー;骨盤内炎症性疾患;再灌流傷害;移植片拒絶;腱炎、血管炎及び静脈炎;
b)急性疼痛、慢性疼痛、神経障害性疼痛、侵害受容性疼痛、痛覚過敏、中枢性感作に関連する疼痛、アロディニア炎症性疼痛、内臓痛、がん性疼痛、外傷性疼痛、歯又は手術痛、術後痛、分娩陣痛、産痛、持続痛、末梢媒介性疼痛、中枢媒介性疼痛、慢性頭痛、片頭痛、副鼻腔炎性頭痛、緊張型頭痛、幻肢痛、末梢神経傷害化学療法性疼痛及びがん性疼痛;
c)関節炎、変形性関節症、若年性関節炎、関節リウマチ、強直性脊椎炎、痛風、リウマチ熱、滑液包炎、全身性エリテマトーデス(SLE)及び多発性硬化症などの自己免疫性疾患;
d)喘息、慢性閉塞性肺疾患(COPD)、サルコイドーシス及び肺線維症などの呼吸障害又は肺疾患;
e)脳がん、前立腺がん、腎がん、肝がん、膵がん、胃がん、乳がん、肺がん、頭頸部がん、甲状腺がん、神経膠芽種、黒色腫、リンパ腫、白血病、皮膚T細胞リンパ腫及び皮膚B細胞リンパ腫などのがん;
f)糖尿病性血管傷害、糖尿病性神経障害及び糖尿病性網膜症を含む糖尿病合併症;
g)アルツハイマー病、パーキンソン病、ハンチントン病及び筋萎縮性側索硬化症などの神経変性傷害、並びに
h)アテローム性動脈硬化症、血栓症、卒中及び冠状動脈性心疾患などの心血管疾患
からなる群から選択される疾患又は状態の治療において、増強されたmPGES−1発現又は活性により媒介される症状の予防又は抑制のために使用される、請求項1〜5のいずれか一項に記載の使用のための化合物。 - 投与される総1日用量が、約5〜2000mg、好ましくは約20〜800mgの範囲であり、より好ましくは、前記総1日用量が約30〜400mgの間の範囲であり、最も好ましくは、前記総1日用量が約150〜250mgの範囲である、請求項1〜6のいずれか一項に記載の使用のための化合物。
- 前記化合物が経口投与される、請求項1〜7のいずれか一項に記載の使用のための化合物。
- 前記化合物が固体形態又は液体形態で投与され、好ましくは、前記化合物が投与前に水溶液と混合され、より好ましくは、前記水溶液が等張水溶液であり、さらにより好ましくは、前記等張水溶液が食塩水である、請求項1〜8のいずれか一項に記載の使用のための化合物。
- 前記化合物が少なくとも1日に2回投与され、好ましくは、前記化合物が1日2回投与され、より好ましくは、前記化合物が2つの同様の又は等しい用量で1日2回投与される、請求項1〜9のいずれか一項に記載の使用のための化合物。
- 2回の投与の間隔が、少なくとも約1、2、3、4、5、6、7、8、9、10、11又は12時間である、請求項10に記載の使用のための化合物。
- 治療される対象が霊長類であり、好ましくは、前記対象がヒトである、請求項1〜11のいずれか一項に記載の使用のための化合物。
- 増強されたPGES−1発現又は活性に媒介される又は関連する疾患又は状態を治療する方法であって、有効量の請求項1〜4のいずれか一項に定義された化合物を、前記疾患又は状態を患っている対象に投与するステップを含む、前記方法。
- 増強されたPGES−1発現又は活性に媒介される又は関連する前記疾患又は状態が、好ましくは、a)急性及び慢性炎症;皮膚炎、湿疹、バムス、尋常性ざ瘡、化膿性汗腺炎及び組織外傷などの皮膚疾患;潰瘍性大腸炎、憩室炎、過敏性腸疾患(IBS)、消化性潰瘍、膀胱炎、(慢性)前立腺炎又は腎炎などの内臓疾患;インフルエンザ、鼻炎、咽頭炎、扁桃炎、結膜炎、虹彩炎、強膜炎、耳炎及びぶどう膜炎などの耳、鼻、口及びのどの疾患;ウイルス及び細菌感染;炎症関連食欲不振;アレルギー;骨盤内炎症性疾患;移植片拒絶;腱炎、血管炎及び静脈炎;b)急性疼痛、慢性疼痛、神経障害性疼痛、侵害受容性疼痛、痛覚過敏、中枢性感作に関連する疼痛、アロディニア炎症性疼痛、内臓痛、がん性疼痛、外傷性疼痛、歯又は手術痛、術後痛、分娩陣痛、産痛、持続痛、末梢媒介性疼痛、中枢媒介性疼痛、慢性頭痛、片頭痛、副鼻腔炎性頭痛、緊張型頭痛、幻肢痛、末梢神経傷害化学療法性疼痛及びがん性疼痛;c)強直性脊椎炎、痛風、リウマチ熱、滑液包炎、並びにd)糖尿病性血管傷害、糖尿病性神経障害及び糖尿病性網膜症を含む糖尿病合併症からなる群から選択される、請求項13に記載の方法。
