以下の実施例は、例示を目的とするものに過ぎず、非限定的な態様である。従って、前記の教示を考慮すれば、本開示の多くの改変物、等価物、および変形物が可能であり、添付の特許請求の範囲の範囲内で、本開示が、具体的に記載された以外に実施され得ることが理解されるべきである。
HIV-1のTATタンパク質形質導入ドメインのインフレームN末端10アミノ酸の配列を含有している順方向プライマー(MRKKRRQRRR(SEQ ID NO:7))、および終止コドンを除去する逆方向プライマーを使用した、ヒトMYCのコード領域のPCR増幅によって、プラスミドpTAT−MYC−V5−6×Hisを作製した。PCR産物を、C末端V5エピトープタグおよび6ヒスチジンタンパク質タグを含むpET101/D-Topo(Invitrogen)ベクターにクローニングした。
AGGコドン、AGAコドン、AUAコドン、CUAコドン、CCCコドン、GGAコドンのためのtRNAを発現するBL21 Rosetta細胞(Novagen)から単離されたpRARE(CamR)を用いてBL-21 Star(商標)大腸菌株(Invitrogen)を形質転換することによって、BL-21 RARE細胞を作出した。
発酵槽接種材料を調製するために、TAT-MYC Master Cell Bank(MCB)のバイアルを解凍し、LB培地を含有しており、選択のための抗生物質(40μg/mLカナマイシン)が補足されたシェーカーフラスコに接種するために使用した。細胞を、オービタルシェーカー/インキュベータにおいて、14〜16時間増大させた。次いで、増大した培養物を、発酵槽培養物に接種するために使用した。細胞が対数増殖期(約4.0のOD)にある時に、IPTG(およそ0.5〜1mM)を培養物に添加することによって、37℃で発酵を誘導した。DO(溶存酸素)スパイクが観察されるまで、約3〜6時間、細胞を誘導した。誘導後、細胞ペーストを遠心分離によって採集し、さらなる加工まで-70℃またはそれ未満で保存した。封入体に含有されていたタンパク質の大部分が、TAT-MYCである。
実施例1からのQセファロースフロースループール中のTAT-MYCタンパク質のリフォールディングを、UFDF(限外濾過/ダイアフィルトレーション)膜を使用したタンジェンシャルフロー濾過に基づくリフォールディング方法を使用して達成した。リフォールディング過程は、一連の3回のリフォールディング工程を含んでいた。第1のリフォールディング工程は、約120分間のリフォールド緩衝液1(3M尿素、50mMリン酸、500mM NaCl、10%グリセロール、5mM GSH(還元型グルタチオン)、1mM GSSG(酸化型グルタチオン))の交換を含んでいた。第2のリフォールディング工程は、およそ120分間のリフォールド緩衝液2(1.5M尿素、50mMリン酸、500mM NaCl、10%グリセロール、5mM GSH(還元型グルタチオン)、1mM GSSG(酸化型グルタチオン))の交換、およびその後のおよそ120分の再循環を含んでいた。第3のリフォールディング工程は、およそ120分間のリフォールド緩衝液3(50mMリン酸、500mM NaCl、10%グリセロール、5mM GSH(還元型グルタチオン)、1mM GSSG(酸化型グルタチオン))の交換、およびその後の12時間の再循環からなっていた。第3のリフォールディング工程が完了した後、リフォールド3溶液を0.2μm膜で濾過し、タンパク質濃度を調整した。
以下の通りにTAT-MYC組成物を活性について試験した。
実施例2に記載されたように調製され、かつ実施例3に記載されたように生物学的活性について試験された、ナノ粒子TAT-MYCタンパク質の組成物は、(a)本発明の技術の方法によって調製されたTAT-MYCタンパク質が、驚くべきことに、予想外に、控え目なサイズ範囲を有するナノ粒子を形成すること;(b)この範囲内のナノ粒子の一部分のみが、生物活性、例えば、MYC活性を有すること;(c)活性が粒子サイズおよび1種類または複数種類の翻訳後修飾の両方に関係することを証明するために、数種類の多様な技法を使用して特徴決定された。
試料中の全成分の量およびサイズ分布に関する情報を提供するために、F01およびF02と呼ばれるTAT-MYCタンパク質の2種類の異なる調製物を、2つの異なる温度で、非対称フロー式フィールドフローフラクショネーション(AF4)および多角度レーザー光散乱(MALLS)によって評価した。F01は、実施例2に従ってリフォールディングされた。F02は、後述されるように調製された。AF4-MALLS分析は、F01が控え目なサイズ範囲を有する粒子の単一の集団を主として含むこと、粒子サイズが、経時的に、変動する温度において、安定していることを例示した。
1. サイズ排除クロマトグラフィおよび高速液体クロマトグラフィによって生物活性TAT-MYC調製物中に控え目な粒子サイズが存在することが確証される
TAT-MYCタンパク質の機能が控え目なサイズのナノ粒子に関係することを確証するために、TAT-MYCの3個の異なる調製物を評価した。(「C12」および「C13」と呼ばれる)2個の機能性調製物、および1個の非機能性調製物(「R147」)を、以下の通りのサイズ排除クロマトグラフィ後の高速液体クロマトグラフィによって特徴決定した。
カラムとの予想外の分子相互作用について制御してこれを排除するために粒子サイズ(分子量)を確認するために、SEC-多角度静的光散乱(MALS)も使用して同一の3個の試料を分析した。
生物活性製剤C2Aならびに3個の付加的な調製物C12、C13(いずれも活性)およびR149(不活性)を、粒子サイズを測定するためにアッセイした。機能性のC12およびC13は、実施例2に概説された手法によって作製された。R149は、C12およびC13に類似したランにおいて生成されたが、TFFリフォールド工程が、6時間実施される透析に置き換えられた。機能性のC2Aは、0080〜0082に概説された手法によって作製されたが、最終製剤緩衝液が250mM Argを含んでいた。

ナノ粒子トラッキング分析(NTA)は、ブラウン運動の速度を粒子サイズと関係付ける、液体中の粒子を可視化し分析する方法である。移動の速度は、液体の粘度および温度にのみ関係する;それは粒子密度または屈折率によって影響を受けない。NTAは、液体懸濁物中のおよそ10〜1000ナノメートル(nm)の直径を有する小さい粒子のサイズ分布プロファイルの決定を可能にする。
前記の生化学的結果を、電子顕微鏡法によってさらに確認した。実施例2に記載されるように調製された「C2B」と呼ばれるTAT-MYC製剤を、生物活性を確認するために、実施例3のT細胞アッセイ法において試験した。C2Bは、以下の通りに電子顕微鏡法によって可視化された。
活性TAT-MYC調製物と不活性TAT-MYC調製物とをさらに区別するために、ペプチドマッピングを実施した。ペプチドフィンガープリンティングとしても公知のペプチドマッピングは、タンパク質の部分加水分解後の電気泳動およびクロマトグラフィによる(ペーパーまたはゲル上での)ペプチドの二次元パターンの形成の技法である。作製されたペプチドパターン(またはフィンガープリント)は、特定のタンパク質に特徴的であり、その技法は、ペプチドの混合物を分離するために使用され得る。
MYC含有ナノ粒子製剤の一次構造、二次構造、三次構造、および四次構造を、標準的な技法を使用して、下記表に概説されるような一連の生化学的特徴決定、生物物理学的特徴決定、および機能的特徴決定の技法を使用して評価した。
飛行時間型(TOF)分析計によるエレクトロスプレーイオン化(ESI)質量分析を追加して、前記チャートに記載されたように、myc含有ナノ粒子製剤を分析した。MSランにおいてはUV検出が行われなかったので、観察された質量をそれぞれのUVピークに割り当てるためにMSトータルイオンクロマトグラム(TIC)およびUVクロマトグラムの視覚的アライメントを実施した。
逆相HPLC(RP-HPLC)法を、翻訳後修飾および産物関連不純物の定量化のために用いた。試験試料を、変性緩衝液(7.5MグアニジンHCl、0.0625MトリスHCl、pH7.3)で希釈し、Agilent AdvanceBio RP-mAb C4カラム(2.1×150mm、ソリッドコアビーズ3.5μm、450Å)を使用して分離した。カラムを50℃の温度で平衡化した。0.5mL/分の流速、ならびに下記表に示されるような100%H20中の0.1%TFA(w/v)(溶出液A)および100%ACN中の0.1%TFA(w/v)(溶出液B)の直線勾配を適用することによって、溶出を達成した。検出を280nmで実施した。
ナノ粒子製剤C13の構造を、「遠紫外」スペクトル領域(190〜250nm)および「近紫外」スペクトル領域(250〜350nm)の両方において円偏光二色性分光法(CD)を使用して評価した。遠紫外領域において、発色団はペプチド結合であり、それが通常のフォールディング環境に位置する場合に、シグナルが発生する。近紫外領域は、三次構造のある種の局面に感受性であり得る。これらの波長において、発色団は、芳香族アミノ酸およびジスルフィド結合であり、それらが生じるCDシグナルは、タンパク質の三次構造全体に感受性である。
波数1700〜1500cm-1のフーリエ変換赤外分光法(FTIR)スペクトルは、タンパク質の構造的特性を決定するために使用され得る。このスペクトル領域における分析は、慣習的にアミドIおよびアミドIIと呼ばれ、それぞれ、波数1700〜1600cm-1および1600〜1500cm-1に存在する2個の吸収帯をもたらす。アミド1バンドは、二次構造(αヘリックス、βシートなど)によってモジュレートされるペプチド結合のC=O伸縮振動による。測定されたスペクトルを、既知の二次構造を有するタンパク質について得られたスペクトルと比較することによって、二次構造含量を得ることができる。ナノ粒子製剤C14についてのFTIR分光法を、MCT検出器およびBioATR Cell IIと共にBruker Optics Vertex 70を使用して実施した。試料は、分析前に調製を必要としなかった。分析についての試験パラメータは、下記表に示される。
沈降速度分析用超遠心分離(SV-AUC)による溶液中のタンパク質の沈降挙動に基づき、ナノ粒子製剤C13のより高次の構造を調査した。SV-AUCは、分子が遠心力に応答して沈殿する速度を測定する。この沈降速度は、試料中に存在する分子の分子量に関する情報を提供する。
ナノ粒子製剤C13を、変性サイズ排除クロマトグラフィ-HPLC(dSEC-HPLC)によって分析した。TSKgel G3000SWxl;5μmを用いて、還元条件および非還元条件の両方で、変性条件下で試料を実行した;試料中のより大きい分子量種からの、モノマーの、サイズに基づく分離を達成するために、ステンレス鋼7.8mm×30cmカラムを使用した。
ナノ粒子製剤C2Bの3次元構造の画像の収集のために、電子顕微鏡法を実施した。ナノ粒子製剤の50マイクロリットルの液滴(100ug/mL)を、パラフィルムにスポットし、次いで、グロー放電カーボン・フォルムバールコート銅透過型電子顕微鏡法(TEM)グリッド(G400銅;EM Science)に吸収させた。グリッドをKimwipe(Kimberly-Clark)上で乾かした後、酢酸ウラニル(2% [wt/vol])およびTEM(Philips CM10)によって80kVで染色した。番号100nm粒子を4,800倍で観察した。図7に示されるように、得られた画像は、自己組み立てスフェアの高度に規則正しい複合体を確認する。
本実施例においては、変異がTAT-MYCナノ粒子製剤の安定性に影響するか否かを判定するために、MYC機能において役割を果たす部位におけるMYCの点変異を調査した。前記の通りに、SEQ ID NO:1に示された配列を有するTAT-MYC融合タンパク質は、野生型ヒトMYCタンパク質(c-MYC)とインフレームで融合したHIV-1 TATのタンパク質形質導入ドメイン(PTD)と、その後の2種類のタグ:V5および6×Hisを組み合わせた融合タンパク質である。TAT-3AMYCは、3個のアミノ酸置換:T358A、S373A、およびT400Aを含有していることを除き、TAT-MYCと同一である。これらの3個のアミノ酸が変異のために最初に選択されたのは、これらの3個の残基におけるリン酸化が、全て、Max結合を阻止するか、またはDNAとの結合に直接干渉することによって、MYCのDNAとの会合を阻害することによって、MYC機能を低下させることが示されているためである(Huang et al.Mol Cell Biol.24(4):1582-1594(2004))。以下に詳述されるように、TAT-3AMYCの点変異は、形成された複合体を不安定化し、融合タンパク質調製物を非機能性にすると考えられる。
前記の実施例1および2に記載されたように、TAT-MYCタンパク質およびTAT-3AMYCタンパク質を調製した。融合タンパク質を変性条件下で可溶化し、次いで、塩、グリセロール、および還元剤(即ち、50mMリン酸、500mM NaCl、10%グリセロール、5mM GSH(還元型グルタチオン)、1mM GSSG(酸化型グルタチオン))を含有している最終製剤へとリフォールディングした。
次いで、TAT-MYCおよびTAT-3AMYCを含むナノ粒子製剤を、サイズ排除クロマトグラフィHPLC(SEC-HPLC)を使用して、分子量プロファイルについて分析した。サイズバリアントを、サイズ排除カラム上で均一濃度の移動相(4M GdnHCl、10mM NaOAc、pH4.65)によって分離し、280nMでの紫外(UV)検出によってダイオードアレイ検出器でモニタリングした。使用されたパラメータは、以下の通りであった:流速:0.75ml/分;最大圧力限界:70.0バール;ラン時間:30分;インジェクション体積:20μL。
TAT-MYC融合タンパク質およびTAT-3AMYC融合タンパク質を、活性化CD4+ T細胞をアポトーシスから救済する能力、ブラスティング(blasting)表現型を保持する能力、および抗原刺激後に増殖を継続する能力を試験することによって、有効性についても査定した。アッセイ法のためのT細胞は、マウスから脾臓およびリンパ節を採集することによって得られた。C10培養培地中の単一細胞懸濁物を生成するために、脾臓およびリンパ節をメッシュスクリーンを通して粉砕した。細胞をコニカルチューブに移し、5分間の1200RPMでの遠心分離によってペレット化した。細胞を、5mlの無菌TAC緩衝液(135mM NH4Cl、17mMトリス、pH7.65)に再懸濁させ、赤血球を溶解するために、1〜2分間、TAC緩衝液中に放置した。細胞を5分間1200RPMで遠心分離し、10mlのC10培地によって洗浄し、4mlのC10培地に再懸濁させた。
ミドリザルTAT-MYCの構築および精製
C.サベウス(ミドリザル)MYCのコード領域をPCR増幅し、実施例1のヒトTAT-MYCベクター内のヒトMYC配列をコードする核酸を、2アミノ酸だけヒトMYC配列と異なるSEQ ID NO:8をコードするミドリザルMYC配列の一部分と置き換えることによって、プラスミドpTAT−MYC(ミドリザル)−V5−6×Hisを作製した。タンパク質の作製および精製は、実施例1に記載されるように実施された。ナノ粒子TAT-MYC組成物の調製は、実施例2に記載されるように実施された。
リフォールディングされたナノ粒子組成物を、実施例4(B)(3)に記載された動的光散乱(DLS)法によって、ナノ粒子サイズ分布について査定した。結果は、ミドリザルTAT-MYCナノ粒子組成物についてのDLSトレースを示す図18に示される。ミドリザルTAT-MYCの平均粒子サイズ(直径)は、約80nmであった。
逆相HPLC(RP-HPLC)法を、ミドリザルTAT-MYCナノ粒子組成物中の翻訳後修飾および産物関連不純物の定量のために用いた。試験試料を、変性緩衝液(7.5MグアニジンHCl、0.0625M TrisHCl、pH7.3)で1mg/mlに希釈した。2μlの0.5M TCEP(トリス(2-カルボキシエチル)ホスフィン)を試料に添加し、37℃で30分間インキュベートし、次いで、2〜8℃に冷却した。試料を、分析前に、最長5日間、2〜8℃で保存した。試料(50μl)(およそ5μg)を、Agilent AdvanceBio RP-mAb C4カラム(2.1×150mm、ソリッドコアビーズ3.5μm、450Å)を使用して分離した。カラムを50℃の温度で平衡化した。溶出は、0.5mL/分の流速、ならびに下記表に示される100%H20中の0.1%TFA(w/v)(溶出液A)および100%ACN中の0.1%(w/v)TFA(溶出液B)の直線勾配を適用することによって達成された。検出は215nmで実施された。参照ヒトTAT-MYCナノ粒子組成物と比較したミドリザルTAT-MYCナノ粒子組成物についての結果が、図19に示される。
ミドリザルTAT-MYCナノ粒子組成物も、サイズ排除クロマトグラフィ後の高速液体クロマトグラフィによって特徴決定した。試料中のより大きい分子量種からの、モノマーの、サイズに基づく分離を達成するために、TSKgel G3000SWxl;5μm;ステンレス鋼7.8mm×30cmカラムを用いて、変性非還元条件下で試料を実行した。溶出は、均一濃度配置の移動相として、4M Gdn HCl、0.1M酢酸ナトリウム、pH4.65を使用して達成された。試料を移動相で1/10希釈し、混合するため10秒間ボルテックスした。次いで、試料を37℃で30分間インキュベートした後、4℃のオートサンプラー内に置いた。
アポトーシスから活性化CD4+ T細胞を救済する能力、ブラスティング表現型を保持する能力、および抗原刺激後に増殖し続ける能力を試験することによって、ミドリザルTAT-MYCナノ粒子組成物および参照ヒトTAT-MYCナノ粒子組成物も有効性について査定した。実施例6に記載された有効性アッセイ法によって、生物活性について組成物をアッセイした。
Claims (60)
a. 該生物活性ナノ粒子の平均直径が約80nm〜約150nmであり;
b. 該製剤のpHが少なくとも約pH6.0でありかつ約pH8以下であり;かつ
c. T細胞増殖に適した条件下で抗CD3活性化または抗CD28活性化T細胞をMYC含有ポリペプチドナノ粒子組成物と接触させることによって、該T細胞の活性化、生存、または増殖のうちの一つまたは複数が、該MYCポリペプチド含有組成物と接触していない抗CD3活性化または抗CD28活性化T細胞と比較して増大する、該組成物。
タンパク質形質導入ドメイン−X−MYC配列
を有するMYC融合ペプチドを含み、
ここで、−X−が、該タンパク質形質導入ドメインと該MYC配列とを連結する分子である、請求項1に記載の組成物。
を含む、造血幹細胞移植(HSCT)後の生着を増強するために造血幹細胞を予備刺激する方法。
(a)可溶化されたMYC含有ポリペプチドを提供するために、ある濃度の変性剤を含む可溶化溶液中にMYC含有ポリペプチドを可溶化する工程;
(b)第1のポリペプチド混合物を提供するために、約0.35〜約0.65倍濃度の工程(a)の変性剤と約100mM〜約1Mのアルカリ金属塩および/またはアルカリ金属塩とを含む第1のリフォールド緩衝液を用いて、可溶化された該MYC含有ポリペプチドに対して第1のリフォールディング工程を少なくとも約30〜180分間実施する工程;
(c)第2のポリペプチド混合物を提供するために、約0.10〜約0.30倍濃度の工程(b)の変性剤と約100mM〜1Mのアルカリ金属塩および/またはアルカリ金属塩とを含む第2のリフォールド緩衝液を用いて、該第1のポリペプチド混合物に対して第2のリフォールディング工程を少なくとも約30〜180分間実施する工程;ならびに
(e)約100mM〜1Mのアルカリ金属塩および/またはアルカリ金属塩を含む第3のリフォールド緩衝液を用いて、該第2のポリペプチド混合物に対して第3のリフォールディング工程を少なくとも約30〜180分間実施する工程;
(f)約80nm〜約150nmの数平均直径を有する生物活性ナノ粒子を作製するために十分な時間、該第3のリフォールド緩衝液中に該MYC含有ポリペプチドを維持する工程
を含み、
T細胞増殖に適した条件下で抗CD3活性化または抗CD28活性化T細胞を該生物活性ナノ粒子と接触させることによって、該T細胞の活性化、生存、または増殖のうちの一つまたは複数が、該生物活性ナノ粒子と接触していない抗CD3活性化または抗CD28活性化T細胞と比較して増大する、該方法。
タンパク質形質導入ドメイン−X−MYC配列
を有するMYC融合ペプチドを含み、
ここで、−X−が、該タンパク質形質導入ドメインと該MYC配列とを連結する分子である、請求項19〜54のいずれか一項に記載の方法。
(a)変性したMYC含有ポリペプチドを提供するために、6〜8M尿素を含む緩衝可溶化溶液中でMYC含有ポリペプチドを変性させる工程;
(b)第1のポリペプチド混合物を提供するために、約3M尿素と約500mM NaClとを含む第1のリフォールド緩衝液を用いて、変性した該MYC含有ポリペプチドに対して第1のリフォールディング工程を少なくとも約120分間実施する工程;
(c)第2のポリペプチド混合物を提供するために、約1.5M尿素と約500mM NaClとを含む第2のリフォールド緩衝液を用いる緩衝液交換によって、該第1のポリペプチド混合物に対して第2のリフォールディング工程を少なくとも約120分間実施する工程;
(d)約500mM NaClを含む第3のリフォールド緩衝液を用いる緩衝液交換によって、該第2のポリペプチド混合物に対して第3のリフォールディング工程を少なくとも約120分間実施する工程;ならびに
(f)約80nm〜約150nmの数平均直径を有する生物活性ナノ粒子を作製するために十分な時間、該第3のリフォールド緩衝液中に該MYC含有ポリペプチドを維持する工程
を含み、
T細胞増殖に適した条件下で抗CD3活性化または抗CD28活性化T細胞を該生物活性ナノ粒子と接触させることによって、該T細胞の活性化、生存、または増殖のうちの一つまたは複数が、該生物活性ナノ粒子と接触していない抗CD3活性化または抗CD28活性化T細胞と比較して増大する、該方法。
