Claims (26)
- ヒトIL-8に結合する単離された抗IL-8抗体であって、以下(a)から(f)の少なくとも一つにおいて、少なくとも一つのアミノ酸置換を含む、pH依存的にIL-8に結合する、抗IL-8抗体:
(a)配列番号:67のアミノ酸配列を含むHVR-H1、
(b)配列番号:68のアミノ酸配列を含むHVR-H2、
(c)配列番号:69のアミノ酸配列を含むHVR-H3、
(d)配列番号:70のアミノ酸配列を含むHVR-L1、
(e)配列番号:71のアミノ酸配列を含むHVR-L2、および
(f)配列番号:72のアミノ酸配列を含むHVR-L3。 - 配列番号:68のアミノ酸配列の9位のチロシン、配列番号:68のアミノ酸配列の11位のアルギニン、および配列番号:69のアミノ酸配列の3位のチロシンのアミノ酸置換を含む、請求項1に記載の抗IL-8抗体。
- 更に、配列番号:68のアミノ酸配列の6位のアラニンおよび配列番号:68のアミノ酸配列の8位のグリシンのアミノ酸置換を含む、請求項1又は2に記載の抗IL-8抗体。
- 配列番号:71のアミノ酸配列の1位のアスパラギン、配列番号:71のアミノ酸配列の5位のロイシン、および配列番号:72のアミノ酸配列の1位のグルタミンのアミノ酸置換を含む、請求項1から3のいずれか1つに記載の抗IL-8抗体。
- 以下の(a)から(f)の性質から選択される少なくとも一つの性質を有するFc領域を含む、請求項1から4のいずれか1つに記載の抗IL-8抗体:
(a)酸性pHでの、当該Fc領域のFcRnに対する結合親和性が、天然型のFc領域のFcRnに対する結合親和性よりも増大している、
(b)既存のADAに対する当該Fc領域の結合親和性が、天然型のFc領域の既存のADAに対する結合親和性よりも低下している、
(c)当該Fc領域の血漿中半減期が、天然型のFc領域の血漿中半減期よりも増大している、
(d)当該Fc領域の血漿中クリアランスが、天然型のFc領域の血漿中クリアランスよりも減少している、
(e)エフェクター受容体に対する当該Fc領域の結合親和性が、天然型のFc領域のエフェクター受容体に対する結合親和性よりも低下している、および
(f)細胞外マトリックスとの結合が増大している。 - Fc領域が、EUナンバリングで表される235位、236位、239位、327位、330位、331位、428位、434位、436位、438位、及び440位からなる群から選択される一以上の位置にアミノ酸置換を含む、請求項5に記載の抗IL-8抗体。
- L235R、G236R、S239K、A327G、A330S、P331S、M428L、N434A、Y436T、Q438R、及びS440Eからなる群から選択されるアミノ酸置換を一つ以上含むFc領域を含む、請求項6に記載の抗IL-8抗体。
- Fc領域が、L235R、G236R、S239K、M428L、N434A、Y436T、Q438R、及びS440Eのアミノ酸置換を含む、請求項7に記載の抗IL-8抗体。
- Fc領域が、L235R、G236R、A327G、A330S、P331S、M428L、N434A、Y436T、Q438R、及びS440Eのアミノ酸置換を含む、請求項7に記載の抗IL-8抗体。
- 請求項1から9のいずれか1つに記載の抗IL-8抗体をコードする単離した核酸。
- 請求項10の核酸を含むベクター。
- 請求項11のベクターを含む宿主細胞。
- 請求項12に記載の宿主を培養する工程を含む、抗IL-8抗体の製造方法。
- 抗体を培養上清から単離する工程を含む、請求項13に記載の抗IL-8抗体の製造方法。
- 請求項1から9のいずれか1つに記載の抗IL-8抗体と、薬学的に許容される担体とを含む医薬組成物。
- IL-8への結合活性がpH依存的である可変領域を含む抗IL-8抗体の製造方法であって、
(a)抗IL-8抗体と細胞外マトリックスとの結合を評価する工程、
(b)細胞外マトリックスとの結合が高い抗IL-8抗体を選択する工程、
(c)当該抗体をコードする核酸を含むベクターを含む宿主を培養する工程、および
(d)培養液から当該抗体を単離する工程、
を含む、抗IL-8抗体の製造方法。 - EUナンバリングで表される235位、236位、239位、327位、330位、331位、428位、434位、436位、438位、および440位からなる群から選択される一以上の位置にアミノ酸置換を含むFc領域を含む、抗IL-8抗体。
- 以下の(a)から(f)の性質から選択される少なくとも一つの性質を有するFc領域を含む、請求項17に記載の抗IL-8抗体:
(a)酸性pHでの、当該Fc領域のFcRnに対する結合親和性が、天然型のFc領域のFcRnに対する結合親和性よりも増大している、
(b)既存のADAに対する当該Fc領域の結合親和性が、天然型のFc領域の既存のADAに対する結合親和性よりも低下している、
(c)当該Fc領域の血漿中半減期が、天然型のFc領域の血漿中半減期よりも増大している、
(d)当該Fc領域の血漿中クリアランスが、天然型のFc領域の血漿中クリアランスよりも減少している、
(e)エフェクター受容体に対する当該Fc領域の結合親和性が、天然型のFc領域のエフェクター受容体に対する結合親和性よりも低下している、および
(f)細胞外マトリックスとの結合が増大している。 - EUナンバリングで表されるL235R、G236R、S239K、A327G、A330S、P331S、M428L、N434A、Y436T、Q438R、及びS440Eからなる群から選択されるアミノ酸置換を一つ以上含むFc領域を含む、請求項17または18に記載の抗IL-8抗体。
- EUナンバリングで表される、(a)L235R、G236R、S239K、M428L、N434A、Y436T、Q438R、及びS440E;または(b)L235R、G236R、A327G、A330S、P331S、M428L、N434A、Y436T、Q438R、及びS440Eからなる群から選択されるアミノ酸置換を一つ以上含むFc領域を含む、請求項19に記載の抗IL-8抗体。
- 請求項17から20のいずれか1つに記載の抗IL-8抗体をコードする単離した核酸。
- 請求項21に記載の核酸を含むベクター。
- 請求項22に記載のベクターを含む宿主細胞。
- 請求項23に記載の宿主細胞を培養する工程を含む、抗IL-8抗体の製造方法。
- 請求項17から20のいずれか1つに記載の抗IL-8抗体を宿主細胞培養物から単離する工程をさらに含む、請求項24に記載の抗IL-8抗体の製造方法。
- 請求項17から20のいずれか1つに記載の抗IL-8抗体と、薬学的に許容される担体とを含む医薬組成物。
