Claims (16)
- IgGクラス抗体とIL−10分子との融合タンパク質であって、2つの同一の重鎖ポリペプチドと2つの同一の軽鎖ポリペプチドとを含み、
前記重鎖ポリペプチドのそれぞれが、IgGクラス抗体重鎖とIL−10単量体とを含み、かつ
前記IL−10単量体が、そのN末端にて前記IgGクラス抗体重鎖のC末端に融合し、任意選択的にペプチドリンカーを介して融合している、融合タンパク質。 - 前記重鎖ポリペプチドがそれぞれ、IgGクラス抗体重鎖と、IL−10単量体と、任意選択的にペプチドリンカーとから本質的になる、請求項1に記載の融合タンパク質。
- 前記IL−10単量体が、天然IL−10単量体、特に天然ヒトIL−10単量体である、請求項1又は2に記載の融合タンパク質。
- 前記IL−10単量体が、配列番号1のポリペプチド配列を含む、請求項1から3の何れか一項に記載の融合タンパク質。
- 前記重鎖ポリペプチドに含まれる前記IL−10単量体が、機能的ホモ2量体IL−10分子を形成する、請求項1から3の何れか一項に記載の融合タンパク質。
- 前記IgGクラス抗体が、改変を有さない対応するIgGクラス抗体と比較して、Fc受容体との抗体の結合親和性を低減する改変を含む、請求項1から5の何れか一項に記載の融合タンパク質。
- (i)前記Fc受容体が、Fcγ受容体、特にヒトFcγ受容体である、
(ii)前記Fc受容体が、活性化Fc受容体である、
(iii)前記Fc受容体が、FcγRIIIa(CD16a)、FcγRI(CD64)、FcγRIIa(CD32)及びFcαRI(CD89)からなる群から選択される、
(iv)前記Fc受容体が、FcγIIIa、特にヒトFcγIIIaである、
(v)前記IgGクラス抗体が、抗体重鎖の329位(EU番号付け)にアミノ酸置換を含む、特に、アミノ酸置換P329Gを含む、
(vi)前記IgGクラス抗体が、抗体重鎖の234位及び235位(EU番号付け)にアミノ酸置換を含む、特に、アミノ酸置換L234A及びL235A(LALA)を含む、及び/又は
(vii)前記IgGクラス抗体が、抗体重鎖中にアミノ酸置換L234A、L235A及びP329G(EU番号付け)を含む、請求項6に記載の融合タンパク質。 - 前記IgGクラス抗体が、
(i)IgG1サブクラス抗体である、
(ii)全長抗体である、及び/又は
(iii)ヒト抗体である、請求項1から7の何れか一項に記載の融合タンパク質。 - 前記IgGクラス抗体が、線維芽細胞活性化タンパク質(FAP)と特異的に結合できる、請求項1から8の何れか一項に記載の融合タンパク質。
- 前記IgGクラス抗体が、
(i)配列番号37の重鎖CDR(HCDR)1と、配列番号41のHCDR2と、配列番号49のHCDR3と、配列番号53の軽鎖CDR(LCDR)1と、配列番号57のLCDR2と、配列番号61のLCDR3とを含む、
(ii)配列番号63の重鎖可変領域(VH)と、配列番号65の軽鎖可変領域(VL)とを含む、
(iii)配列番号37のHCDR1と、配列番号43のHCDR2と、配列番号47のHCDR3と、配列番号51のLCDR1と、配列番号55のLCDR2と、配列番号59のLCDR3とを含む、又は
(iv)配列番号67のVHと、配列番号69のVLとを含む、請求項9に記載の融合タンパク質。 - 請求項1から10の何れか一項に記載の融合タンパク質をコードするポリヌクレオチド。
- 請求項11に記載のポリヌクレオチドを含む宿主細胞。
- IgGクラス抗体とIL10分子との融合タンパク質を生成する方法であって、(i)請求項12に記載の宿主細胞を、融合タンパク質の発現に適切な条件下で培養する工程と、(ii)融合タンパク質を回収する工程とを含む方法。
- 請求項13に記載の方法により生成される、IgGクラス抗体とIL−10分子との融合タンパク質。
- 請求項1から10及び14の何れか一項に記載の融合タンパク質と、薬学的に許容される担体とを含む薬学的組成物。
- 請求項1から10及び14の何れか一項に記載の融合タンパク質又は請求項15に記載の薬学的組成物であって、
(i)医薬として用いるための、
(ii)炎症性疾患の治療又は予防に用いるための、又は
(iii)炎症性腸疾患又は関節リウマチの治療に用いるための、融合タンパク質又は薬学的組成物。
