Abstract
Description
本願は、米国特許法第119条(e)項に基づいて、2015年10月8日に出願した米国仮出願第62,239,241号及び2014年11月14日に出願した米国仮出願第62/080,201号の優先権の利益を主張し、その全体を参照により本明細書に援用する。
本発明は、米国国立衛生研究所により付与された助成金番号R01GM101420‐01A1の下で政府の支援によってなされた。政府は本発明の一定の権利を有する。
別段定義されない限り、本明細書で使用する全ての技術用語及び科学用語は、当業者に一般的に理解されているもの(例えば、細胞培養、分子遺伝学、及び生化学において)と同じ意味を有するものと解釈するべきである。
ミトコンドリア病は、機能不全のミトコンドリアに起因する。ミトコンドリア病は、例えば、ミトコンドリアDNA(mtDNA)またはミトコンドリア成分をコードする核遺伝子における後天性または先天性の変異によって生じ得る。疾患は、薬物、感染症、または他の環境的要因の副作用による後天性ミトコンドリア機能不全に起因し得る。ミトコンドリア病の例として、ミトコンドリア筋症;糖尿病性腎症;糖尿病及び難聴(DAD)のいくつかの形態;レーバー遺伝性視神経萎縮症(LHON);リー症候群;神経障害、運動失調、網膜色素変性症、及び眼瞼下垂(NARP);筋神経胃腸脳症(MNGIE);赤色ぼろ線維・ミオクローヌスてんかん症候群(MERRF);ミトコンドリア筋症、脳筋症、乳酸アシドーシス、脳卒中様症状(MELAS);ならびにミトコンドリア神経胃腸管脳筋症(MNGIE)が挙げられる。
本発明の態様は、標準的なまたは以前に記載した電気穿孔法のパラメータと比較して、より低い電場強度の使用、またはより短時間の電場への曝露に関する。電気穿孔法のパラメータは、多数の細胞型について報告されている。例えば、www.thermofisher.com/us/en/home/life-science/cell-culture/transfection/transfection---selection-misc/neon-transfection-system/neon-protocols-cell-line-data.htmlで入手可能なNeon(登録商標)形質移入システムに関する情報参照を参照されたい。また、Kim et al.,(2008) Biosens Bioelectron,23(9):1353‐1360参照も参照されたい。この全内容が参照により本明細書中に援用される。例えば、Neon(登録商標)形質移入システムは、0.3〜1kv/cmの電場を5ms〜50msのパルス持続時間で使用してもよい。Neon(登録商標)形質移入システムの電気穿孔法パラメータの例を、以下の表にも提供する。
DFEは、標準的または以前に開示した電気穿孔法システムと比較して、核酸のより効率的かつ迅速な送達(及びその後の機能、例えばコードされた遺伝子産物の発現)を、高い細胞生存率でもたらす。本発明は、電場のようなエネルギー場が、細胞変形狭窄部によって生じる細胞膜内の摂動を介してペイロードを輸送するか、ペイロードを細胞内のある位置から別の位置へ、例えば、細胞膜の近傍から1つ以上の細胞小器官へ輸送する方法、システム、及び装置を提供する。いくつかの例示的な実装形態は電場を用いるが、一方で他の輸送機構が有用である。輸送機構として、例えば1つ以上の電場、磁場、及び音場を利用してもよい。
癌特異的抗原を認識するキメラ抗原受容体(CAR)を発現するようにT細胞を改変することにより、免疫検出を免れた腫瘍細胞を認識させて死滅させることができる。本プロセスは、患者のT細胞を抽出し、それらにCARの遺伝子を形質移入し、その後、形質移入した細胞を患者に再注入することを含む。
本発明の態様は、細胞膜内に摂動を引き起こすためのマイクロ流体システムを提供し、該システムは、管腔を画定するとともに、緩衝液中に懸濁した細胞が管腔を通過できるように構成されたマイクロ流体チャネルを備え、マイクロ流体チャネルは細胞変形狭窄部を有し、狭窄部の直径は細胞の直径の関数であり、かつ該システムは、(a)エネルギー場;または(b)該狭窄部の下流、上流、もしくは上流及び下流に位置するエネルギー場のソースもしくはエミッタを有する。
DNA形質移入のための多くの技術が開発されている。リポフェクションのような担体に基づく方法は、担体と細胞膜との相互作用、及び生物学的に活性なプロセスであるDNAの細胞内輸送に大きく依存する。マイクロインジェクションは、転写のためにDNAを核に直接送達するために使用されてきたが、スループットにより制限される。電気穿孔法はDNA形質移入に広く使用されているが、そのメカニズムに依然として議論の余地があり、また電気パルス後の原形質膜から核への能動的DNA輸送に依存する点でも制限がある。ここでは、破壊及び場による送達(DFE;本用語は本実施例の実施形態に限定されない)と名付けた細胞内送達の概念を説明する。DFEでは、まず破壊によって原形質膜内の摂動部が開き、次いでこれらの間隙を介した細胞質及び核へのDNA送達が可能になる。本戦略は、機械的破壊と電場との組合せに関し、GFPプラスミドDNAのような積荷分子またはその混合物を核に直接送達し、処理後1時間以内に発現させるものであり、これは電気穿孔法のような他の方法よりも迅速である。この新規戦略は、形質移入が困難な細胞に対する細胞内遺伝子送達、及び広範な物質の同時送達に有用である。
本明細書において、破壊及び場による送達(DFE)の概念の展開例を開示する。本アプローチでは、まず機械的プロセスによって細胞膜を破壊した後に、細胞を場に曝露して物質を標的細胞に輸送する。ハイスループットでDNAを細胞の核に直接送達することができるマイクロ流体装置が開発されている。DFE概念の1つの実装形態は、細胞圧搾を用いて細胞膜を一時的に破壊した後に細胞を電場に曝露し、それにより、例えば核膜またはミトコンドリア膜などの膜破壊を介して負に荷電しているDNAを輸送し、かつカート(carto)を細胞の細胞質ゾルへ送達した後に細胞核内へ輸送することを含む。CellSqueeze技術は、様々な細胞型にわたって多様な物質を送達する頑強な能力を実証したが、単独ではDNAの核送達を促進するには効果的ではない(Shalek et al.,Proc.Natl.Acad.Sci.U.S.A.107,1870‐5(2010))。DNA分子が原形質膜に向かって電気泳動的に移動して原形質膜上に凝集する電気穿孔法とは異なり、DFEプロセスでは、DNA分子は、機械的破壊によって生じる細胞膜内の摂動または間隙を介して細胞内へかつ/または細胞内部において移動する。例えば、図9A参照。本組合せプロセス(DFE)は、積荷、例えば荷電化合物、例えばDNAの、細胞質ゾル及び細胞下構造への送達において相乗効果をもたらす。相乗効果は、送達する積荷の機能、例えば、送達するDNAによる遺伝子発現を測定することによって実証される。
シリコンウェーハをPyrexウェーハに結合させて、DFEマイクロ流体装置を形成した。下記の2つの主要な工程、すなわち(1)シリコンウェーハ上へのマイクロ流体チャネルの製造、及び(2)Pyrexウェーハ上へのマイクロ流体電極の製造が製造に関与していた。入口及び出口リザーバを備えたホルダに装置を取り付けた。発振器(Agilent E4422B)から電気パルスを発生させ、増幅器を介して増幅し、導電性エポキシを用いて電極パッドに結合したワイヤを介して装置を駆動した。所望の送達物質(積荷化合物または組成物)と混合した細胞の溶液を、入口リザーバ内に配置する。次いでこのリザーバを、調整器によって制御される圧縮空気ラインに接続する。圧力(0〜20psi)を利用して流体を輸送して装置に通過させるとともに、細胞が通過する際に装置に電気パルスを印加する。出口リザーバから細胞を回収してさらに処理する。

- 細胞膜に摂動を引き起こすためのマイクロ流体システムであって、
管腔を画定するとともに、緩衝液中に懸濁した細胞が前記管腔を通過できるように構成されたマイクロ流体チャネルであって、細胞変形狭窄部を有し、前記狭窄部の直径が細胞の直径の関数である、前記マイクロ流体チャネル、
(a)エネルギー場、または、
(b)前記狭窄部の下流、上流、または上流及び下流に位置するエネルギー場のソースまたはエミッタ、
を備える、前記マイクロ流体システム。 - ペイロードを細胞に送達するためのマイクロ流体システムであって、

(o)前記マイクロ流体チャネルが、直列の複数の細胞変形狭窄部を有し;
(p)前記マイクロ流体チャネルが、単一の細胞変形狭窄部を有し;
(q)前記細胞が複数の細胞であり、前記電場に通した後に、前記細胞の約80、85、90、91、92、93、94、95、96、97、98、99、90〜95、または80〜100%が生存可能であり;
(r)前記電場の強度またはパルス強度が、約10〜2000kV/m、または100kV/m未満であり;
(s)前記電場を、約0.1、0.1〜2、または0.1〜2000msの持続時間、1〜20、0.1〜2000、または1〜200msの周期でパルスし;
(t)前記細胞を、約100、170、300、100〜300、200〜700、250〜400、100〜1000mm/s、または1〜1000mm/sの速度で電場に通し;
(u)前記細胞膜の前記摂動が、約1〜20、1〜600、4、5、6、7、8、9、10、12、14、16、18、20、25、50、75、100、150、200、250、300、350、400、450、500、または600nmの最大直径を有し;かつ/または、
(v)約1〜20、1〜600、4、5、6、7、8、9、10、12、14、16、18、20、25、50、75、100、150、200、250、300、350、400、450、500、または600nmの最大直径を有する前記細胞膜の前記摂動が、前記細胞膜上において、少なくとも約1、2、3、4、5、6、7、8、9、10、または1〜10分間持続する、請求項25〜41のいずれか一項に記載の方法。 - 前記細胞が原核細胞または真核細胞である、請求項1〜25のいずれか一項に記載のマイクロ流体システムまたは請求項25〜42のいずれか一項に記載の方法。
- 導入遺伝子をコードする発現ベクターの細胞への送達方法であって:
前記細胞及び前記発現ベクターを含有する溶液を細胞変形狭窄部に通し、それによって前記細胞に圧力を加え、前記発現ベクターが通過するのに十分なほど大きい前記細胞膜の摂動を引き起こすこと;
前記発現ベクターを前記細胞内に輸送するために、少なくとも1つの電極によって生成された電場に前記溶液を通すことを含み、
前記細胞中に前記導入遺伝子が、細胞変形狭窄部を通過することなく電場を通過した細胞中の前記導入遺伝子の発現に比べて、より速い速度で発現する、前記送達方法。 - 前記細胞変形狭窄部を通過することなく前記電場に接触した対応する細胞に比べて0.1、1.5、1、1.5、2、2.5、3、3.5、4、または0.1〜4時間早く、前記細胞中で前記導入遺伝子が発現する、請求項44に記載の方法。
- 導入遺伝子をコードする発現ベクターの細胞への送達方法であって:
前記細胞及び前記発現ベクターを含有する溶液を細胞変形狭窄部に通し、それによって前記細胞に圧力を加え、前記発現ベクターが通過するのに十分なほど大きい前記細胞膜の摂動を引き起こすこと;
前記発現ベクターを前記細胞内に輸送するために、少なくとも1つの電極によって生成された電場に前記溶液を通すことを含み、
前記細胞中の前記導入遺伝子の最大発現を、細胞変形狭窄部を通過することなく電場を通過した細胞中の前記発現に比べて、より速い速度で達成する、前記送達方法。 - 前記細胞中の前記導入遺伝子の発現を、細胞変形狭窄部を通過することなく電場、磁場、または音場と接触した対応する細胞中での前記発現よりも約0.1、1.5、1、1.5、2、2.5、3、3.5、4、または0.1〜4時間早く達成する、請求項46に記載の方法。
- 導入遺伝子をコードする発現ベクターの細胞への送達方法であって:
前記細胞及び前記発現ベクターを含有する溶液を細胞変形狭窄部に通し、それによって前記細胞に圧力を加え、前記発現ベクターが通過するのに十分なほど大きい前記細胞膜の摂動を引き起こすこと;
前記発現ベクターを前記細胞内に輸送するために、少なくとも1つの電極によって生成された電場に前記溶液を通すことを含み、
前記細胞中に前記導入遺伝子が、細胞変形狭窄部を通過することなく電場を通過した細胞中の前記導入遺伝子の発現に比べて、より広範囲に発現する、前記送達方法。 - 前記細胞中での導入遺伝子の発現が、細胞変形狭窄部を通過することなく電場に接触した対応する細胞中での前記導入遺伝子の発現に比べて、少なくとも約5、10、15、20、25、30、35、40、45、50、55、60、65、70、75、80、85、90、95、もしくは100%大きいか、または2倍、5倍、8倍、10倍、20倍もしくはそれ以上大きい、請求項48に記載の方法。
- 前記細胞が前記狭窄部を通過してから約0.1、1.5、1、1.5、2、2.5、3、3.5、4、または0.1〜4時間以内における前記細胞内での導入遺伝子の発現が、細胞変形狭窄部を通過することなく電場と接触した対応する細胞内での発現よりも、少なくとも約5、10、15、20、25、30、35、40、45、50、55、60、65、70、75、80、85、90、95、もしくは100%大きいか、または2倍、5倍、8倍、10倍、20倍もしくはそれ以上大きい、請求項48に記載の方法。
- 導入遺伝子をコードする発現ベクターの細胞集団への送達方法であって:
前記細胞及び前記発現ベクターを含有する溶液を細胞変形狭窄部に通し、それによって前記細胞に圧力を加え、前記発現ベクターが通過するのに十分なほど大きい前記細胞膜の摂動を引き起こすこと;
前記発現ベクターを前記細胞内に輸送するために、少なくとも1つの電極によって生成された電場に前記溶液を通すことを含み、
前記集団中の前記導入遺伝子を発現する細胞の割合は、細胞変形狭窄部を通過することなく電場を通過した細胞集団中の前記導入遺伝子を発現する細胞の割合よりも大きい、前記送達方法。 - 前記集団中の前記導入遺伝子を発現する細胞の割合が、細胞変形狭窄部を通過することなく電場と接触した対応する細胞集団中における導入遺伝子を発現する細胞の割合よりも、少なくとも約5、10、15、20、25、30、35、40、45、50、55、60、65、70、75、80、85、90、95、もしくは100%大きいか、または2倍、5倍、8倍、10倍、20倍もしくはそれ以上大きい、請求項51に記載の方法。
- 前記細胞が前記狭窄部を通過してから約0.1、1.5、1、1.5、2、2.5、3、3.5、4、または0.1〜4時間以内における前記集団中の前記導入遺伝子を発現する細胞の割合が、細胞変形狭窄部を通過することなく電場と接触した対応する細胞集団中における前記導入遺伝子を発現する細胞の割合よりも、少なくとも約5、10、15、20、25、30、35、40、45、50、55、60、65、70、75、80、85、90、95、もしくは100%大きいか、または2倍、5倍、8倍、10倍、20倍もしくはそれ以上大きい、請求項51または52に記載の方法。
- 導入遺伝子をコードする発現ベクターの細胞集団への送達方法であって:
前記細胞及び前記発現ベクターを含有する溶液を細胞変形狭窄部に通し、それによって前記細胞に圧力を加え、前記発現ベクターが通過するのに十分なほど大きい前記細胞の摂動を引き起こすこと;
前記発現ベクターを前記細胞内に輸送するために、少なくとも1つの電極によって生成された電場に前記溶液を通すことを含み、
前記集団中の前記導入遺伝子を高レベルで発現する細胞の割合が、細胞変形狭窄部を通過することなく電場を通過した細胞集団中の前記導入遺伝子を高レベルで発現する細胞の割合よりも大きい、前記送達方法。 - 前記集団中の前記導入遺伝子を高レベルで発現する細胞の割合が、細胞変形狭窄部を通過することなく電場と接触した対応する細胞集団中における前記導入遺伝子を発現する細胞の割合よりも、少なくとも約5、10、15、20、25、30、35、40、45、50、55、60、65、70、75、80、85、90、95、もしくは100%大きいか、または2倍、5倍、8倍、10倍、20倍もしくはそれ以上大きい、請求項54に記載の方法。
- 前記細胞が前記狭窄部を通過してから約0.1、1.5、1、1.5、2、2.5、3、3.5、4、または0.1〜4時間以内における前記集団中の前記導入遺伝子を高レベルで発現する細胞の割合が、細胞変形狭窄部を通過することなく電場と接触した対応する細胞集団中における前記導入遺伝子を発現する細胞の割合よりも、少なくとも約5、10、15、20、25、30、35、40、45、50、55、60、65、70、75、80、85、90、95、もしくは100%大きいか、または2倍、5倍、8倍、10倍、20倍もしくはそれ以上大きい、請求項54または55に記載の方法。
- 導入遺伝子をコードする発現ベクターの細胞への送達方法であって:
前記細胞及び前記発現ベクターを含有する溶液を細胞変形狭窄部に通し、それによって前記細胞に圧力を加えて、前記発現ベクターが通過するのに十分なほど大きい前記細胞膜の摂動を引き起こすこと;
前記発現ベクターを前記細胞に輸送するために、少なくとも1つの電極によって生成された電場に前記溶液を通すことを含み、
前記細胞内で前記導入遺伝子が、細胞変形狭窄部を通過することなく電場を通過した細胞内の前記導入遺伝子の発現よりも、より速く発現する、前記送達方法。 - 前記導入遺伝子が、細胞変形狭窄部を通過することなく電場と接触した対応する細胞よりも、約0.1、1.5、1、1.5、2、2.5、3、3.5、4、または0.1〜4時間早く前記細胞中で発現する、請求項57に記載の方法。
