総ての送達可能な細胞およびナノ粒子の中で、生細胞由来の細胞外小胞、特に、30~150nmのナノサイズ範囲のエクソソームは、ここ数十年で細胞間コミュニケーションにおいて重要な役割を示されている。免疫細胞由来のエクソソームは、免疫刺激または免疫抑制の調節において文書により十分に立証されており、炎症性、自己免疫性および感染性疾患の病理を駆動する。エクソソームの形成は、細胞内小胞としてエンドソームが生成することから始まる。エクソソームは、細胞分泌のための多小胞体(MVB)に由来するという点で、他の膜由来微小小胞体とは異なる(4, 9)。従って、エクソソームは、特定のタンパク質と核酸を含み、親細胞での形成時のそれらの親細胞の状態と機能を表す。エクソソームの多くのサブタイプの中で、MHCクラスIおよびII分子と他の共刺激分子の内在性のペイロードを有する免疫原性エクソソームは免疫応答を媒介することができ、このことは、癌ワクチンに使用することができる新規な送達プラットフォームの開発、免疫療法の送達、およびイン・ビボ(in vivo)輸送に関連するその他の送達の機会を開く。

本明細書に記載されるデバイスは、流体サンプルと適合性のある任意の材料で作製することができる。いくつかの実施形態では、本明細書に記載されるデバイスを製作するための材料は、磁場が貫通し得る。いくつかの実施形態では、本明細書に記載されるデバイスを製作するための材料は、その中のサンプルがイン・サイチュ(in situ)で光切断され得、またはそれが、例えば、磁性標識されたエクソソームのイン・サイチュ分析のために、顕微鏡下で見えるように、実質的に透明であり得る。本明細書に記載されるマイクロ流体デバイスを製作するために使用することができる例示的材料には、限定されるものではないが、ガラス、コポリマー、ポリマーまたはそれらの任意の組合せが含まれ得る。例示的ポリマーには、限定されるものではないが、ポリウレタン、ゴム、成形プラスチック、ポリメチルメタクリレート(PMMA)、ポリカーボネート、ポリテトラフルオロエチレン(テフロン(商標))、ポリ塩化ビニル(PVC)、ポリジメチルシロキサン(PDMS)、ポリスルホン、およびエーテル系脂肪族ポリウレタンが含まれる。
本明細書で論じられるように、開示されるマイクロ流体デバイスは、操作された細胞外小胞および様々な小胞様生物学的構造を分離、捕捉、操作、および放出するために使用することができる。特定の実施形態では、操作された細胞外小胞は免疫原性エクソソームである。本明細書で使用する場合、「エクソソーム」という用語は、一般に、エンドソームコンパートメントまたはあらゆる細胞、例えば、腫瘍細胞(例えば、前立腺癌細胞)、および免疫細胞(例えば、抗原提示細胞、例えば、樹状細胞、マクロファージ、肥満細胞、Tリンパ球またはBリンパ球)に由来する外部に放出される小胞を指す。エクソソームは、一般に、腫瘍細胞、赤血球、血小板、リンパ球、および樹状細胞を含む様々な異なる細胞種から放出される約20~150nmのサイズの膜小胞である。エクソソームは、後期エンドソームの膜からの陥入および出芽により形成され得る。それらは、サイトゾルの多小胞体(MVB)に蓄積し、そこから原形質膜との融合によって放出され得る。特定の理論に縛られることを望むものではないが、小胞放出のプロセスは、癌細胞などの増殖細胞において特に活発であり、そこでは放出が継続的に起こり得ると考えられる。エクソソームは、腫瘍細胞から放出されると、浸潤と移動を促進し得る。従って、いくつかの実施形態では、本明細書に記載される免疫磁性粒子を使用して、癌細胞に由来するエクソソームを捕捉することができる。エクソソームは、細胞の起源に応じて、MHC分子、テトラスパニン、接着分子およびメタロプロテイナーゼを含む、原形質膜とは異なる可能性がある様々な細胞タンパク質を動員することができる。エクソソームの多くのサブタイプの中で、MHCクラスIおよびII分子と他の共刺激分子の内在性のペイロードを有する免疫原性エクソソームは、免疫応答を媒介することができ、このことは、新規な癌ワクチンの開発および免疫療法における送達の機会を開く。
概要:細胞外ナノ小胞(≦1μm)、特に、エクソソーム(30~150nm)は細胞コミュニケーションを媒介する際の新たな送達システムであり、これは親細胞シグナル伝達タンパク質または腫瘍抗原を免疫細胞に提示することにより免疫応答をプライムすることに関して認められたものである。本例では、1つのワークフローで直接、表面操作されたエクソソームの採取、抗原修飾、および光放出を行うための合理化されたマイクロ流体細胞培養プラットフォームが提供される。PDMSマイクロ流体細胞培養プラットフォームは、3D印刷型から複製される。エクソソーム表面の抗原性ペプチド(例えば、gp-100、MART-1、MEGA-A3)を操作することにより、効果的な抗原提示およびT細胞の活性化が達成され得る。このことは、分泌されたエクソソームをリアルタイムで、黒色腫腫瘍ペプチドで操作するためのヒト血液由来白血球のオンチップ培養を使用することにより実証された。2個体のPmel1トランスジェニックマウスの脾臓から精製されたgp100特異的CD8 T細胞を試験した。操作されていないエクソソームに比べて有意に高いT細胞活性化レベル(~30%)が操作されたエクソソームにより誘導されることが認められた。このマイクロ流体プラットフォームは、癌免疫療法を進展させ得る治療エクソソームの迅速かつリアルタイムの生産のための自動高集積型細胞培養デバイスとして役立つ。
3D印刷およびマイクロ流体デバイスの製作:土台、壁、および上部磁石ホルダーを含むPDMSチップ製作用の3つの型を準備した。この型はSolidWorks(登録商標)2017を使用することにより設計し、3D SystemsからのProject 1200の3Dプリンターで印刷した。これら複数の部品は50μmの精密構造を有し、チャネル高は50μmであった。細胞培養チャンバーは直径1000μm、チャンバー高500μmで設計した。総ての型にスポーツラインパラジウムで20nm厚にコーティングした。3つ総ての部品を、PDMSチップを用いて組み立てた。このPDMSを、細胞培養チャンバーにチャンバープラグのための開放端が残るように高さ500μmまで満たした。PDMSは、リンカー試薬と10:1比でキャストし、40℃の温度で6時間インキュベートした。PDMSは硬化した後、簡単に剥離することができた。チップの入口および出口は、0.75mmパンチャーを使用して穴をあけた。ピラニア処理ガラス及びPDMSを両方とも、少なくとも30秒間、高電圧プラズマであった。次に、PDMSチップを40℃の温度のホットパッド上で5分間、後接着した。これらのチップを脱イオン水で洗浄し、オートクレーブで滅菌した(121℃で30分)。
エクソソームをオンインライン採取するための3D印刷成形マイクロ流体細胞培養デバイス:3D印刷型を用いてPDMSに基づくオンチップ細胞培養マイクロ流体デバイスを作製するために簡単で低コストのアプローチを開発した。培養チップは細胞をオンチップ増殖させ、下流で培養培地からエクソソームを収集するための、直径1mmおよび高さ0.5mmのオンチップ細胞培養チャンバーを含む。細胞培養チャンバーは、培地交換のためのPDMS製のフィンガープッシュプラグを適用し、培地を下流の収集イオンチャネルに押し出すために上部が開いたままとする。細胞培養チャンバーの底部は、免疫磁性分離ビーズと混合するために(A-入口)培養培地を導入するための、幅約200μmおよび高さ200μmの出口チャネル(B-入口)を備える。C-入口は、シリンジポンプにより駆動される、洗浄バッファーを導入するために使用される。図4は、蛍光色素溶液を用いた蛍光顕微鏡観察下での、A-入口およびB-入口およびエクソソーム分離チャネル(蛇行チャネル)への出口を介する混合プロセスを説明する。図5(b)は、蛇行チャネル内で混合している免疫磁性ビーズを記録したものである。ヒト血液由来白血球を、図5(c)に示す形態を有する培養デバイスで培養した。わずかな赤血球がなおカップ形状として観察された。分泌されたエクソソームを分離し、捕捉し、チップの出口から光放出させ、図5(d)に示されるSEMイメージングにより特性評価を行った。
上記の免疫原性試験では、開示されている特許方法を用いて操作されたエクソソームを注射したトランスジェニックマウスを使用した。エクソソームを、上記の合理化された/連続的なマイクロ流体プロセスを用いて表面にgp-100またはBRSV M187-196ペプチドで操作し、尾からのin vivo腹腔内注射向けにPBSバッファーに懸濁させた。本発明者らは、注射部位の反応も注射を受けたマウスからの有害応答(注射部位の腫脹、刺激作用など)は認めなかった。マウスは、注射後最初の72時間は1日2回観察し、有害反応は認められず、操作されたエクソソームおよび関連組成物の全般的なin vivo安全性を示す。
Claims (50)
- マイクロ流体デバイスにおいて生物学的標的を操作するための方法であって、
前記マイクロ流体デバイスのマイクロ流体混合チャネル内で、前記生物学的標的を含むと疑われる生物学的サンプルを流体中の複数の免疫磁性粒子と混合して混合物を形成すること;
前記生物学的標的が存在する場合、前記生物学的標的が前記混合物中の前記免疫磁性粒子と反応および結合して粒子/標的複合体を形成することを可能にすること;
前記粒子/標的複合体を、マイクロ流体デバイスのチャンバー内に磁場を印加することにより前記チャンバー内に固定すること;
前記粒子/標的複合体中の前記標的を、前記標的を複数の活性部分または活性薬剤と接触させることにより操作すること(ここで、前記標的の表面は前記活性部分で表面修飾されてまたは前記複数の活性薬剤は前記標的内に搭載されて、操作された標的を生成する);および
前記操作された標的を前記粒子/標的複合体から光分解的に放出すること(ここで、前記粒子は前記チャンバー内に固定されたままであり、かつ、前記操作された標的は前記マイクロ流体デバイスの出口に向かって下流で洗浄される)、
を含んでなる、方法。 - 前記生物学的標的が、細胞、細胞外小胞、および小胞様細胞画分からなる群から選択される、請求項1に記載の方法。
- 前記細胞外小胞がエクソソームまたはミクロソームである、請求項2に記載の方法。
- 前記マイクロ流体デバイスの細胞培養チャンバーに細胞を導入すること;
前記細胞を前記エクソソームの放出を可能にする条件下で培養すること;および
前記エクソソームを前記複数の免疫磁性粒子と混合するために前記混合チャネルに導入すること
をさらに含んでなる、請求項3に記載の方法。 - 前記細胞が、樹状細胞、幹細胞、免疫細胞、巨核球前駆細胞、およびマクロファージからなる群から選択される、請求項4に記載の方法。
- 前記活性部分が生物学的標的上の表面タンパク質を認識し、それらと特異的に結合する、請求項1に記載の方法。
- 前記標的が前記活性部分で実質的にコーティングされるように、前記操作工程後に前記表面タンパク質の実質的に総てが活性部分と結合する、請求項6に記載の方法。
- 前記免疫磁性粒子が500nm以上の直径を有する、請求項1に記載の方法。
- 前記粒子/標的複合体が、単一の中心粒子に結合した複数の前記生物学的標的を含んでなるように、前記免疫磁性粒子のそれぞれが前記サンプル中の複数の前記生物学的標的と反応する、請求項1に記載の方法。
- 同じタイプの生物学的標的が各粒子に結合するように、各粒子が単一タイプの生物学的標的に一度に結合する、請求項9に記載の方法。
- 前記光分解的に放出することが、前記粒子/標的複合体を前記マクロ流体チャンバー内で露光することを含んでなる、請求項1に記載の方法。
- 前記標的が前記露光後約15分以内に放出される、請求項11に記載の方法。
- 前記マイクロ流体デバイスにおいて連続的なプロセスで、好ましくは前記混合から前記出口での前記操作された標的の収集まで90分以内に実施される、請求項1に記載の方法。
- 前記活性部分が抗原性ペプチドである、請求項1に記載の方法。
- 前記活性薬剤が薬物、ヌクレオチド、CRISPR Cas9システム、小分子化合物、化学療法薬などである、請求項1に記載の方法。
- 前記マイクロ流体デバイス平面基板が、入口とチャンバーとの間に延伸するマイクロ流体チャネルを介して前記チャンバーと流体連通する入口を含んでなり、前記チャンバーは前記入口の下流にあり、出口と流体連通し、前記出口は前記チャンバーの下流にある、請求項1に記載の方法。
- 前記マイクロ流体デバイスが、前記サンプル、前記免疫磁性粒子、および洗浄バッファーを前記デバイスに導入するための複数の入口を含んでなる、請求項16に記載の方法。
- 前記マイクロ流体チャネルが、前記免疫磁性粒子と前記サンプルの混合を促進するための蛇行流路を有する、請求項16に記載の方法。
- 前記デバイスが、前記マイクロ流体チャネルと流体連通する細胞培養チャンバーをさらに含んでなる、請求項16に記載の方法。
- 前記免疫磁性粒子はそれぞれ、前記生物学的標的を捕捉するための、その粒子表面から延伸する複数の光切断可能なリンカーを含んでなり、それぞれはその末端にそれぞれの親和性プローブを有する、請求項1~19のいずれか一項に記載の方法。
- 前記親和性プローブが抗原性ペプチドまたはその抗原性エピトープを含んでなる、請求項20に記載の方法。
- 前記親和性プローブが、MAGE-A3、gp-100、HER-2、p53、PSA-1、およびMART-1から選択される抗原性ペプチドを含む、請求項21に記載の方法。
- 前記親和性プローブと前記生物学的標的が結合して免疫刺激性複合体を形成し、これが前記粒子から放出されると免疫細胞の抗原提示および活性化ならびに免疫応答のプライミングを増強する、請求項20に記載の方法。
- 前記親和性プローブが、MHCクラスI分子、MHCクラスII分子、インターロイキン、TNFα、IFNγ、RANTES、G-CSF、M-CSF、IFNα、CTAPIII、ENA-78、GRO、I-309、PF-4、IP-10、LD-78、MGSA、MIP-1α、MIP-1β、およびそれらの組合せからなる群から選択される免疫刺激性分子に対する特異性について選択される、請求項20に記載の方法。

前記混合チャネルから分離出口までの流体の流れの経路を規定する分離チャネル;および
収集チャンバー内で磁場を生成するための、前記収集チャンバーに作動可能に連結された磁石を含んでなる、前記分離出口に流体接続された収集チャンバー;
とを含んでなる、デバイス。 - 生物学的標的の操作において使用するためのマイクロ流体デバイスであって、
細胞培養入口および細胞培養出口を含んでなる細胞培養チャンバー;
流体入口チャネルおよび粒子入口チャネル(ここで、前記細胞培養出口、前記流体入口チャネル、および前記粒子入口チャネルは混合交差点で流体収束する);
前記混合交差点から混合出口までの流体の流れの経路を規定する、前記混合交差点に流体接続された混合チャネル(ここで、前記細胞培養チャンバーの幅と前記混合チャネルの最大断面寸法の比は少なくとも5:1である);および
収集チャンバー内で磁場を生成するための、前記収集チャンバーに作動可能に連結された磁石を含んでなる、前記混合出口に流体接続された収集チャンバー;

- 前記分離チャネルが、局所的な渦流プロフィールを作出するための複数のチャネル狭窄領域、好ましくは少なくとも5つのチャネル狭窄領域を含んでなる、請求項45に記載のマイクロ流体デバイス。
- 前記細胞培養チャンバー幅と前記混合チャネルの最大断面寸法の比が5:1~500:1、5:1~20:1、または6:1~12:1である、請求項41~46のいずれか一項に記載のマイクロ流体デバイス。
- 前記細胞培養チャンバーが、約200マイクロリットル以上、好ましくは約200マイクロリットル~約1ミリリットルの容積を有する、請求項41~47のいずれか一項に記載のマイクロ流体デバイス。
- 前記混合チャネルが、高さおよび幅を有し、前記高さおよび前記幅のそれぞれが少なくとも50ミクロン、好ましくは50~500ミクロンである、請求項41~48のいずれか一項に記載のマイクロ流体デバイス。
- マイクロ流体デバイスに作動可能に連結されたポンプをさらに含んでなる、請求項41~49のいずれか一項に記載のマイクロ流体デバイス。
