Description
βNGFを製造するための様々な方法が先行技術において記載されている。しかしながら、現在利用可能な製造プロセスは、不均質なβ−NGF生成物およびβ−NGFの低い収率などのいくつかの欠点を有している。
a.proNGF変異体を変性溶液に溶解すること;
b.前記proNGF変異体を、前記変性されたproNGFが生物学的に活性な立体配座を取るリフォールディング溶液に移すこと;
c.前記proNGF変異体を前記リフォールディング溶液から精製すること;
d.前記proNGF変異体のプロ配列を、前記活性なβ−NGFを得るために切断すること。
本発明を下記で詳しく記載する前に、本発明は、本明細書中に記載される特定の方法論、プロトコルおよび試薬に限定されないことを理解しなければならない。なぜならば、これらは変化する場合があるからである。本明細書中で使用される用語は、特定の実施形態を記載するという目的のためだけであり、また、添付されている請求項によってのみ限定されるであろう本発明の範囲を限定するために意図されないこともまた理解しなければならない。別途定義される場合を除き、本明細書中で使用されるすべての技術的用語および科学的用語は、本発明が属する技術分野の当業者によって一般に理解されるのと同じ意味を有する。

本発明のproNGF変異体
本発明の第1の実施形態において、本発明は、プロテアーゼ切断部位R1SK3R4が、非塩基性アミノ酸およびヒスチジンから選択されるアミノ酸によって、少なくともヒト野生型proNGF配列(配列番号1)の101位および103位に対応するR1位およびK3位で置換されるproNGF変異体を提供する。言い換えれば、少なくともヒト野生型proNGF配列(配列番号1)の101位〜104位における生来的なプロテアーゼ切断部位R1SK3R4の101位でのアルギニンR1および103位でのリシンK3が、アルギニンまたはリシンでないいずれかのアミノ酸によって置換される。

工程a)は、生来的なプロテアーゼ切断部位R1SK3R4において、ヒト野生型proNGF配列(配列番号1)の101位および103位に対応するR1位およびK3位が置換されるproNGF変異体を、変性溶液における封入体の可溶化によって溶解することに対応する。工程a)における本発明のproNGF変異体は通常、原核生物宿主細胞におけるその発現に起因して、その不活性な不溶性形態の形態であることは特筆される。不良な溶解性を示す不活性なproNGFが、原核生物の細胞質ゾルにおけるタンパク質の過剰発現の期間中に形成される。この場合、組換えによって調製されるproNGFは、不溶性の凝集形態で細胞質に留まる。これらのタンパク質凝集物、その単離、同様にまた、それらの精製が、例えば、Marston,F.A.、Biochem.J.、240(1986)に記載される。

proNGF変異体を封入体から可溶化した後、タンパク質をその生来的な立体配座にリフォールディングすることが必要である。リフォールディングプロセスのために、競合反応である誤った折り畳みおよび凝集を最小限に抑えることが重要である。凝集を防止するために、リフォールディングは、非常に低いタンパク質濃度で行われる。これは、タンパク質の凝集が、高いタンパク質濃度で優勢であるからである。工程b)において、proNGF変異体を、変性されたproNGFが生物学的に活性な立体配座を取るリフォールディング緩衝液に移すことが行われる。生物学的に活性な立体配座が、天然のβ−NGFに存在するジスルフィド架橋の存在によって決定される可能性がある。
i. シャペロン、好ましくはアルギニン、0.5M〜1.0M、好ましくは0.75M;
ii. 金属キレーター、好ましくはEDTA、1mM〜10mM、好ましくは5mM;
iii. レドックスシャフリング系、0.1mM〜10mM、好ましくは、1mMのL−シスチンおよび5mMのL−システイン、または、1mMのGSSG(酸化型グルタチオン)および5mMのGSH(還元型グルタチオン);
iv. pH8.0〜pH11.0、好ましくはpH9.5。
変性および続くリフォールディングを伴う本発明による方法を行うことによって、折り畳まれたpro−NGFムテインの水溶液が得られる。折り畳まれたpro−NGFムテインは続いて、知られている方法によってさらに精製することができる。
proNGFはβ−NGFの前駆体である。したがって、本発明のproNGF変異体からβ−NGFを製造する方法の工程d)において、proNGF変異体のプロ配列が、活性なβ−NGFを得るために切断される。
本発明のproNGF変異体から製造されるβ−NGFはさらに、例えば、いくつかのクロマトグラフィー方法によって精製される。さらなる精製工程が、トリプシンおよびトリプシン消化の生成物関連不純物をβ−NGFから分離するために要求される。精製工程は、HCP、エンドトキシンおよびDNAを減少させなければならない。タンパク質精製のためにこの技術分野で知られている方法はどれも使用することができる。最も好ましいものがクロマトグラフィー精製であり、例えば、Sepharoseカラム(例えば、SP Sepharose HP、Q Sepharose FF)を用いたクロマトグラフィー精製である。
a)置換されたプロテアーゼ切断部位を有する組換えpro−NGF変異体の原核生物細胞における発現;
b)pro−NGFムテイン含有封入体の単離;
c)封入体を、(i)カオトロピック物質、(ii)キレーター、(iii)緩衝剤および(iv)還元剤を少なくとも含む好適な変性緩衝液と混合すること;
d)シャペロン、金属キレーターおよびレドックスシャフリング系を少なくとも含むリフォールディング溶液におけるリフォールディング;
e)リフォールディングされたpro−NGF変異体の精製;
f)プロテアーゼ(例えば、トリプシンなど)を用いた、β−NGFから活性形態への切断;
g)β−NGFの単離および精製。
第3の局面において、本発明は、ヒトβ−NGFを製造するための本発明のproNGF変異体の使用に関する。
さらなる局面において、本発明は、生来的なプロテアーゼ切断部位R1SK3R4において、上記で記載されるようなヒト野生型proNGF配列(配列番号1)の101位および103位(K3およびR1)が置換されるproNGF変異体から得られるβNGFと、医薬的に許容されるキャリアとを含む医薬組成物に関する。
ヒトpro−NGF(配列番号1)の101位および103位に対応するアルギニンR1およびリシンK3の置換を、当業者に知られている方法によって合成遺伝子を使用してDNAレベルで実現した。102位におけるセリンは未変化のままであったか、または、ヒトpro−NGF(配列番号1)の102位の置換もまた、当業者に知られているような方法によって合成遺伝子を使用してDNAレベルで実現した。104位に対応するリシンK4は置換されなかった。配列を図1に示す。
rh−proNGFの発現のために使用される細菌宿主の大腸菌JM108(DSMZ5585;F− thi Δ(lac−proAB) endAI gyrA96、relA1 phx hsdR17 supE44 recA)はプロリン要求性であり、これは、pSCIL101の名称を有するプラスミドの使用によって中和された。プラスミドpSCIL101はプラスミドpSCIL008に基づく(国際公開第05061716号パンフレットを参照のこと)。この菌株はチアミンを合成することができない(Vieira&Messing、1982、Gene、Oct;19(3):259〜68)。配列番号5で示されるpro−NGF変異体が、pSCIL101に置かれるtacプロモーターの制御のもとで発現させられる。この場合に使用されるベクターpSCIL101は、カナマイシン抵抗性を有する高コピー型プラスミドである。発現を、規定された無機塩培地で行い、IPTGの添加によって誘導する。pro−NGF変異体が封入体(IB)の形態で細胞質ゾルに堆積する。

この発酵の目的は、その後のプロセス工程のための生成物およびバイオマスを得ることであった。発酵プロセス期間中の標的タンパク質の過剰発現をモニターするために、サンプルを誘導の前後でSDS−PAGEによって分析した。

細菌細胞中に、組換えタンパク質が凝集物の形態で存在する。pro−NGFムテインの発現がIBの形態で生じた。細胞破壊およびIB調製を標準的プロトコルに従って行った。細胞破壊およびIB調製は、およそ200gまでのバイオマスを処理することであれば実験室スケールで行うことができる。proNGFムテインを含有するこれらの「封入体」の調製を、Rudolph,R.他(1987);Folding proteins(Creighton,T.E.(編):Protein Function:A Practical Approach、Oxford University Press、57頁〜99頁)に従って、また、欧州特許第0994188B1号明細書に従って行った。細胞破砕のために、細胞ペレットを好適な緩衝液に再懸濁し、続いて、細胞を、50mMリン酸ナトリウム(pH7.0)、1mM EDTAにおける高圧均質化を使用して破砕した。
封入体を、(i)カオトロピック剤、(ii)キレーター、(iii)緩衝剤および(iv)還元剤の溶液を含む変性溶液において可溶化した。可溶化のために、グアニジウムHCl(GuaHCl)を4.0M〜6.0Mの濃度範囲で試験した。可溶化緩衝液を封入体スラリー(IBスラリー)と種々の比率で混合した。すべての実験が5mMの最終システイン濃度を有し、室温で行った。結果をSDS−PAGEによって分析した(データは示されず)。実験により、4MのGuaHClの濃度が封入体の完全な可溶化のために十分であったことが明らかにされた。封入体スラリー対緩衝液の比率は1+1.25(v/v)(IBスラリー:緩衝液)である。封入体を可溶化するための変性溶液の最終的条件は下記の通りであった:
i. 4MのグアニジウムHCl
ii. 0.1MのTris
iii. 10mMのEDTA
iv. 5mMのシステイン
v. pH8.0
可溶化後、タンパク質をその生来的な立体配座でリフォールディングし、それにより、誤った折り畳みおよび凝集を最小限に抑えることが必要である。本発明による生物学的に活性なproNGFムテインを可溶化された材料から調製するために、これらを、proNGFが生物学的に活性な立体配座を取るリフォールディング溶液に希釈した。
i. 0.75Mのアルギニン
ii. 5mMのEDTA
iii. 1mMのL−シスチンおよび5mMのL−システイン
iv. pH9.5
合成された親和性リガンド、すなわち、4−メルカプト−エチル−ピリジン(MEP)を有するカラムを使用した。溶出を、pH値を変化させることによって行った。さらに、溶出を、効率的なプロセス設計のために有利である低い塩濃度により行った。
β−NGFへのproNGF変異体のトリプシン消化のために、プロテアーゼの活性を阻害しないリン酸塩緩衝液を使用した。リン酸ナトリウム緩衝液を25mMリン酸ナトリウムの最終濃度にMEP溶出液に加えた。pH値をpH6.5に調節した。タンパク質分解のために、トリプシン(Roche、GMP規格)を1:10,000(w/w)(トリプシン:proNGF)の比率で加えた。タンパク質分解を、室温での18時間のインキュベーション時間を使用して行った。トリプシン消化の成績および収率を、SDS−PAGE、rp−HPLCおよびUV/VIS280nmによって分析した。図5は、トリプシン切断の分画物のSDS−PAGEを示す。4%〜12%のBis/Tris−Gel(1mm、泳動緩衝液としての1×MES(Invitrogen))を使用した。レーン5〜レーン7が、トリプシン切断生成物を、非切断のproNGF変異体(rhproNGF*、レーン3を参照のこと)および成熟型β−NGF(NGF、レーン8を参照のこと)と比較して示す。図は、精製されたproNGF変異体が1つだけの切断生成物(β−NGF)をもたらすことを明瞭に示す。β−NGFへのproNGF変異体の完全な消化を認めることができた。
トリプシン消化後、β−NGFを、トリプシン、切断の副生成物およびさらなる不純物を取り除くために、SP Sepharose HPカラムにロードした。SP Sepharose HPによる精製を図6に示す。
本手順をproNGF変異体SP174−101(配列番号5)およびヒト野生型proNGF(配列番号1;rhProNGF)の両方について並行して適用した。
TF1細胞(ATCC、カタログ番号CRL2003)を標準的手順に従って培養した。試験培地(90%の培地RPMI1640、10%のウシ胎児血清FBS、50U/mlのペニシリンおよび50μg/mlのストレプトマイシン)を細胞に加え、遠心分離した。ペレットを、37℃で、試験培地に1.5・105細胞/mlの密度で再懸濁した。細胞懸濁物をproNGFタンパク質の種々の濃度物(10−10M、3・10−10M、10−9M、3・10−9M、10−8M、3・10−8M、10−7M、3・10−7M、10−6M、3・10−6M、10−5Mおよび3・10−5M)と混合し、96ウエルプレートで分析した。37℃で48時間のインキュベーションの後、細胞増殖試薬(例えば、WST−1、Roche Applied Science、カタログ番号1644807)を加え、プレートを、37℃で4時間、再びインキュベーションした。吸収を450nmで測定し、EC50値を、好適なプログラム(例えば、Sigma−Plot 2000)を使用することによって求めた。
Claims (36)
- プロテアーゼ切断部位R1SK3R4が、非塩基性アミノ酸およびヒスチジンから選択されるいずれかのアミノ酸によって、少なくともヒト野生型proNGF配列(配列番号1)の101位および103位に対応するR1位およびK3位で置換される、proNGF変異体。
- 前記プロテアーゼ切断部位の101位および103位が、アルギニンまたはリシンでないいずれかのアミノ酸によって置換される、請求項1に記載のproNGF変異体。
- 前記プロテアーゼ切断部位の101位および103位が、アラニン、グリシン、バリン、セリン、トレオニン、メチオニン、チロシン、ヒスチジン、アスパラギン、アスパラギン酸、グルタミン、グルタミン酸、フェニルアラニン、イソロイシン、ロイシン、トリプトファン、システインおよびプロリンから選択されるいずれかのアミノ酸によって置換される、請求項1に記載のproNGF変異体。
- 前記プロテアーゼ切断部位が、アラニン、バリン、グリシン、セリン、トレオニン、メチオニン、チロシン、ヒスチジン、アスパラギン、アスパラギン酸、グルタミン、グルタミン酸から選択されるいずれかのアミノ酸によって101位および103位で置換される、請求項1に記載のproNGF変異体。
- 生来的な前記プロテアーゼ切断部位の101位および103位が、アラニンおよびバリンから選択されるいずれかのアミノ酸によって置換される、請求項1に記載のproNGF変異体。
- 前記プロテアーゼ切断部位の101位がバリンによって置換される、請求項1〜3に記載のproNGF変異体。
- 前記プロテアーゼ切断部位の103位がアラニンによって置換される、請求項1〜6に記載のproNGF変異体。
- ヒト野生型proNGF配列(配列番号1)の104位に対応するR4位におけるアミノ酸がアルギニンまたはリシンから選択される、請求項1〜7に記載のproNGF変異体。
- ヒト野生型proNGF配列(配列番号1)の102位を置換するアミノ酸が、セリン、グリシン、システイン、アスパラギン、チロシン、トレオニン、アスパラギン酸、グルタミン、アラニン、バリン、グルタミン酸、ヒスチジン、イソロイシン、ロイシン、フェニルアラニン、プロリン、トリプトファン、メチオニン、のいずれかから選択される、請求項1〜8に記載のproNGF変異体。
- ヒト野生型proNGF配列(配列番号1)の101位におけるアミノ酸がバリンによって置換され、102位におけるアミノ酸がセリンによって置換され、103位におけるアミノ酸がアラニンによって置換され、かつ、104位におけるアミノ酸がアルギニンである、請求項1〜9に記載のproNGF変異体。
- 配列番号5を有する、請求項1〜10に記載のproNGF変異体。
- 原核細胞における組換え発現によって得られる、請求項1〜11に記載のproNGF変異体。
- 大腸菌における組換え発現によって得られる、請求項12に記載のproNGF変異体。
- (i)請求項1〜13のいずれかに記載されるproNGF変異体を提供すること、および
(ii)活性なヒトβ−NGFを得るために前記proNGF変異体を切断すること
を含む、生物学的に活性なヒトβ−NGFを調製する方法。 - 下記の工程:
a.請求項1〜13のいずれかに記載されるproNGF変異体を変性溶液における封入体の可溶化によって溶解する工程、
b.前記proNGF変異体を、前記変性されたproNGFが生物学的に活性な立体配座を取るリフォールディング溶液に移す工程、
c.前記リフォールディングされたproNGF変異体を精製する工程、
d.前記proNGF変異体のプロ配列を、前記活性なβ−NGFを得るために切断する工程
を含む、請求項14に記載の方法。 - 前記変性溶液が、(i)カオトロピック物質、(ii)キレーター、(iii)緩衝剤および(iv)還元剤を含有する溶液を含む、請求項15に記載の方法。
- 前記変性溶液が、
i. 1M〜8MのグアニジウムHCl(好ましくは4M〜6M)、
ii. 0.01M〜1MのTris、
iii. 1mM〜50mMのEDTA、
iv. グルチオン(GSH)またはシステインから選択される1mM〜100mMを含み、
v. pH7.0〜10.0
である、請求項16に記載の方法。 - 前記変性溶液が、
i. 4MのグアニジウムHCl、
ii. 0.1MのTris、
iii. 10mMのEDTA
iv. 5mMのGSHまたはシステインを含み、
v. pH8.0
である、請求項17に記載の方法。 - 前記リフォールディング溶液が、
i. 0.5M〜1.0Mのシャペロン、
ii. 1mM〜10mMの金属キレーター、
iii. 0.1mM〜10mMのレドックスシャフリング系を含み、
iv. pH8.0〜pH11.0
である、請求項15に記載の方法。 - 前記リフォールディング溶液が、
i. 0.75Mのアルギニン、
ii. 5mMのEDTA
iii. 1mMのL−シスチンおよび5mMのL−システイン、または、1mMのGSSG(酸化型グルタチオン)および5mMのGSH(還元型グルタチオン)を含み、
iv. pH9.5
である、請求項18に記載の方法。 - 前記リフォールディングがパルス再生として行われる、請求項15および17〜20に記載の方法。
- パルス再生期間中の最終的リフォールディング体積に関して、グアニジウムHClの濃度が0.3Mを超えず、かつ、パルスあたりのタンパク質濃度が50μg/mlを超えない、請求項21に記載の方法。
- 前記proNGF変異体が混合モードのクロマトグラフィーにより精製される、請求項15に記載の方法。
- 前記クロマトグラフィーカラムが、合成された親和性リガンドを有する混合モード物質カラムである、請求項23に記載の方法。
- 混合モード物質カラムが、4−メルカプト−エチル−ピリジン(MEP)、ヘキシルアミノ(HEA)、フェニルプロピルアミノ(PPA)、2−メルカプト−5ベンゾイミダゾールスルホ酸(MBI)、Capto MMC(GEHC)、N−ベンジル−N−メチルエタノールアミン(GEHC)、CHTヒドロキシアパタイトまたはCHTフルオロアパタイトを有するカラムである、請求項24に記載の方法。
- 混合モード物質カラムが、4−メルカプト−エチル−ピリジン(MEP)を有するカラムである、請求項25に記載の方法。
- 前記proNGF変異体のプロ形態がプロテアーゼによって切断される、請求項14〜26に記載の方法。
- 前記proNGF変異体の前記プロ形態がセリンプロテアーゼによって切断される、請求項27に記載の方法。
- 前記proNGF変異体の前記プロ形態がトリプシンによって切断される、請求項28に記載の方法。
- トリプシン対proNGF変異体の比率が1:200〜1:100,000である、請求項27〜29に記載の方法。
- トリプシン対proNGF変異体の前記比率が重量あたり1:5,000〜1:20,000である、請求項30に記載の方法。
- トリプシン対proNGF変異体の前記比率が1:10,000(w/w)である、請求項31に記載の方法。
- β−NGFを精製するさらなる工程をさらに含む、請求項14〜32に記載の方法。
- β−NGFをカラムクロマトグラフィーによって精製する工程をさらに含む、請求項33に記載の方法。
- β−NGFを、SP Sepharose HPカラムによって精製する工程をさらに含む、請求項34に記載の方法。
