Abstract

Description
インスリン様増殖因子I受容体(IGP-IR,EC2.7.12,CD 221 抗原)が、膜貫通タンパク質チロシンキナーゼのファミリーに属する(Leroith,D.,らによるEndocrin. Rev.16(1995)143-163、およびAdams,T.EらによるCell.Mol.Life Sci.57(2000) 1050-1063)。IGP-IRが高親和性にてIGF-Iと結合し、そしてin-vivo中でこのリガンドに対して生理的応答を開始する。しかしながら、さらにIGP-IRが、わずかに低い親和性にてIGF-IIにも結合する。IGP-IRが、過剰発現すると、細胞の悪性形質転換を促進し、IGP-IRが細胞の悪性形質転換に関与することが実証され、そしてそれによりガン治療用の治療剤の生成に有益な標的である(Adams,T.E.らにより、Cell.Mol.Life Sci.57(2000)1050-1063)。
さらにIGF-IRに対する抗体が、さらに多くの刊行物に、たとえばArteaga,C.L.らによるBreast Cancer Res.Treatment22(1992)101-106、およびHailey,J.らによるMol.Cancer Ther.1(2002)1349-1353に記載されている。
抗体および抗体の特性および特徴付けの技術的状態の概要が、Adams,T.E.,らによるCell.Mol.Life Sci.57(2000)1050-1063に記載されている。技術的状態が記載されているほとんどの抗体が、マウスを起源とするものである。こうした抗体が、技術的に良く知られているように、化学化又はヒト化のようなさらに変化されることなく、ヒトの患者の治療に有益でない。
しかしながら、抗腫瘍治療が必要な患者に確実な利点を伴うIGF-IRに対する抗体の必要性がさらにある。患者のために関係する利点は、簡単な言葉で抗腫瘍形成剤の治療により生ずる腫瘍増殖の減少、および時間的進行が有意に延長することである。
本発明は、IGF-IRに結合し、且つIGF-IRへのIFG-I及びIFG-IIの結合を抑制する抗体を含み、前記抗体が、
a) IgG1のアイソタイプであり、
b) IGF-IRに対するIGF-IIの結合抑制のIC50値に対する、IGF-IRに対するIGF-Iの結合抑制のIC50値における比率が1:3から3:1を示し、
c) 0.5%の熱不活性化胎児牛血清(FCS)を含む培地中で、前記抗体を用いることなく、こうしたアッセイと比較する場合、HT29細胞を使用し細胞リン酸化アッセイにおいて、5nMのIGF-IRリン酸化濃度で、少なくとも80%、好ましくは少なくとも90%を抑制し、そして
d) 0.5%の熱不活性化胎児牛血清(FCS)を含む培地中で、前記抗体を用いることなく、こうしたアッセイと比較する場合、細胞当り400,000乃至600,000分子のIGF-IRを具備する3T3細胞を使用する細胞リン酸化アッセイにおいて10μMの濃度で、PKBリン酸化として測定されたIGF-IR刺激活性を全く示さない、
ことを特徴とする。
a) CDRsとして、配列番号:1または3のCDR1(aa31-35)、DR2(aa50-66)およびCD3(aa99-107)を含む抗体の重鎖、
b) CDRs群として、配列番号:2または4のCDR1(aa24-34)、CDR2(aa50-56)およびCD3(aa89-98)を含む抗体の軽鎖、

好ましくは抗体が、インスリン受容体へインスリンの検知可能な濃度依存性結合に抑制を全く示さないことである。

さらに本発明は、本発明によるIGF-IRに対するアンタゴニスト的抗体の有効量を、ガン罹患と診断された患者に(そしてそのため抗腫瘍治療が必要である)投与することを含む、ガン治療のための方法を提供する。抗体が、医薬組成物において単独、又はそれ以外として放射線治療あるいは細胞傷害剤又はその先駆薬剤などを組み合わせて投与することができる。
用語「抗体」は、全抗体、抗体の断片、ヒトの抗体、ヒト様抗体、および本発明による特徴的特性が保持されているかぎり、一般的な遺伝子操作がされた抗体を含むがそれに限定されない種々の形状の抗体を包含する。

本明細書に使用される用語「IGF-IRに対するIGF-IおよびIGF-IIの結合を抑制すること」は、in vitroアッセイにおけるHT29(ATCC HTB-38)腫瘍細胞の表面上に提示されるIGF-IRに対するI125-標識化IGF-I又はIGF-IIの結合を抑制することを指している。抑制することは、IC50値が2nM以下であることを意味している。

選択される投与経路の関係なく、適切な水和物形状にて使用可能な本発明による化合物、そして/又は本発明の医薬組成物が、当業者の周知の従来の方法にて医薬的に受け入れ可能な投与量の形態に処方される。本発明の医薬組成物における活性成分の実際の投与量レベルが、特定患者、組成物、患者に毒性のない投与形態に応答して、所望の治療を行うために効果的である活性成分の量を得るように、変更することができる。
たとえば、適切な流動性は、レシチンなどの被覆を用いることにより、分散性の場合必要な粒子サイズを維持することにより、そして界面活性剤を使用することにより、維持することができる。

ハイブリドーマの培養
生成されたHuMAbハイブリドーマを、37℃で2mMのL-グルタミン(BioWhittaker)および4%の原クローニング因子(Igen,France)、そして5%のCO2にて補給されたハイブリドーマ発現培地(Hybridoma Express Medium)(PAA Laboratories GmbH,Austria)で培養した。
10匹のHCo7の遺伝子導入マウス(雄4匹、雌6匹)、菌株GG2201(Medarex,San Jose,CA,USA)が、1x106のNIH 3T3細胞にてそれぞれ免疫化され、IGF-IRおよび、そしてIGF-IRの20μの可溶な細胞外ドメインに対し発現ベクターにてトランスフェクトされた。6匹の免疫化全体に行われ、3匹に,IGF-IR発現細胞にて腹腔内(IP)への免疫化、そして3匹が、組み換えタンパク質にて、尾の底部にて皮下(SC)免疫化が行われた。最初の免疫化に対し、100μの1x106 NIH 3T3 IGF-IR細胞を、100μの完全フロイントアジュバント(CFA;Difco Laboratories,Detroit,USA)と混合した。その他の全ての免疫化に対し、PBS中100μlの細胞を使用するか、又は組み換えタンパク質を、100μの不完全フロイントアジュバント(ICFA;Difco)と混合した。
免疫化されたマウスの血清中の抗-IGF-IRタイターが、抗原特異的ELISAにより決定される。PBS中1μg/mlの濃度でIGF-IR可溶な細胞外ドメインを、4℃と一昼夜又は37℃で2時間、96ウエルプレートへ被覆した。その後、そのブロックをPBSTC(0.05%Tween(商標)-20および2%のトリ血清(Gibco BRL))、1時間(h)、室温にて遮断した。最初タップ(tap)による血清を、PBSTC中で1/50に希釈し、他のすべてのタップ(tap)からの血清を、PBSTC中で1/100に前希釈(pre-diluted)し、次に1/6400に希釈した。希釈された血清をウエルに加え、そして37℃で1時間インキュベートした。前希釈(pre-diluted)の血清を負のコントロールとして使用した。200ng/mlのヤギの抗-ヒトIGF-IR(100μg/ml)を正のコントロールとして使用した。
抗原特異性ELISAにより決定することに加えて、免疫化されたマウスの血清中の抗-IGF-IRタイターが、さらにFACS]分析により決定された。NIH 3T3 IGF-IR細胞および非トランスフェクトされたNIH 3T3 IGF-IR細胞を、30分間、4℃で希釈された血清によりインキュベートした。プレ・タップ(Pre-tap)血清が負のコントロールとして使用される。最初、200ng/mlヤギの抗-ヒトIGF-IRを、正のコントロールとして使用した。細胞を1%のウシ血清アルブミンおよび0.01%アジド(azide)にて補給されたPBSにおいて、3度洗浄した。次に細胞を、FACS緩衝液中で1/100に希釈されたラット抗-ヒトIgGの光イソチオシアネイト(FITC)-接合抗原結合断片(F(ab’)2断片)にて、4℃で30分間インキュベートした。細胞を、FACS緩衝液中で2度洗浄し、そして試料をFACS Calibur(Becton Dickinson Erebodegem−Aalst,Belgium)上で分析した。
抗IGF-IRの血清タイターが十分であると分かった時、融合4か3日前に、200μのPBS内で15μgのIGF-IR細胞外ドメインにて静注に2回追加免疫をした。
ハイブリドーマの生成
マウスを犠牲にし、腹部大動脈および大静脈を側面に位置する脾臓およびリンパ節が、収集される。
融合相手SP 2.0細胞と脾臓細胞およびリンパ節の融合が、標準的な操作手順に従って行われた。融合の結果からハイブリドーマが、ヒト抗体を生成するかどうかを決定するために、κ-ELISAを行った。ELISAプレートが、PBS中1/10000に希釈されたラット抗体-ヒトIgG-κ-軽鎖抗体(DAKO)にて、被覆し、4℃で1昼夜インキュベートした。ウエル物を破棄した後、プレートをPBSTCにより室温で1時間インキュベートにより遮断した。その後ウエルが、PBSTCにより1/2に希釈された、ハイブリドーマ培養上澄み液にて培養した。PBSTC中で1/2希釈された培養培地を、負のコントロールとして使用し、PBSTC中で1/100に希釈されたκ-軽鎖陽性マウスとして使用する。そしてウエルを2度洗浄し、そして試験物を、新たに調製されたABTS(商標)溶液(1mg/ml)中、室温(RT)にて暗い状態で30分間生育成した。吸光度を、405nmで測定した。
モノクロナール抗体18および22を調製した。

親和性を測定するために、抗ヒトFCγ抗体(ラビットから)が、IGF-IRに対する抗体を表示するため、チップ表面に結合される。IGF-IR細胞外ドメインに、種々の濃度の溶液に加えられる。3分間のIGF-IRの結合により会離が測定される。抗体18および22への親和性データが、試料表1に示される。
材料および方法:
IGF-IR表面発現に及ぼす影響を研究するために、NCl H322M細胞が、低密度又は超密集度のいずれかの光学グレイドのグラスカバースライド上のRPMI培地で、培養した。並行して、コントロール群から単離されたH322M異種移植組織が、イソペンタン中でショック凍結され、そして低温切断にて、5μmの厚さに切断した。免疫蛍光標識を、マウス-抗IGF-IRモノクロナール抗体(αIR3,5μg/ml)又は本発明による抗体を使用し、その後ヤギ抗-マウス、又はCy3(Amersham Biosciences,GB)又はAlexa Fluor(商標)488(Molecular Probes,Inc.,USA)にて標識化されたヤギ抗-マウスにて行った。試料が、Leica SP2 confocal顕微鏡上にて表示され、そしてFACSにより分析した。
高密度にて培養されたH322M細胞が、confocal顕微鏡により表示された時、IGF-IRが、特に細胞と細胞の接触部位に集合したことが、明らかになっている。In vivoにおいて、すなわち特に異種移植組織に増殖したH322M細胞と比較すると、表面IGF-I受容体の構成が関係するかぎり、in vitro における培養物に濃密に充填される顕著な類似性がある。
さらにH322M細胞の超密集培養物における表面IGF-I受容体の上限調節が、FACSにより定量化された。細胞を細胞と細胞を有意に接触することなく、低密度と比較し高密度条件下で増殖する時、IGF-IR受容体表面発現が10倍を超えて増大した。
HT29、MDA231、およびMCF-7などの他の腫瘍細胞が、細胞と細胞の接触部位の確立に基づいて、細胞表面上のIGF-I受容体の上限調節が、H322M細胞特有の特徴でなく、さらにin vivoにおいて見出されたより多い組織様機構の一般的特徴であると見られる(図1)。
H322M細胞を、プラスチック表面への接着を抑制するようポリ-HEMA(ポリ(2-ヒドロキシエチルメタクリレイト)被覆皿上でRPMI640/0.5%FCS培地で培養した。これらの条件に基づいて、H322M細胞が、3次元的に増殖する密集した楕円面(独立的な固着と称す)を形成する。これらの楕円面は、所定位置の固体腫瘍の3次元組織構造および構成を示す。楕円面の培養物を、0から240nMへ量を増大させた抗体の存在下、5日間インキュベートした。WST変換アッセイを用い、増殖抑制を測定した。H322Mの楕円面の培養物を、抗体18を異なる濃度(1乃至240nM)で処理した場合、増殖の投与量依存性抑制が観察可能である(図2)。
H322M細胞を、プラスチック表面への接着を抑制するようポリ-HEMA被覆皿上でRPMI1640/10%NCS培地で培養した。これらの条件に基づいて、H322M細胞が、3次元的に増殖する密集したほぼ円形(独立的な固着と称す)を形成する。これらの楕円面は、所定位置の固体腫瘍の3次元組織構造および構成を示す。楕円面の培養物を、0から7.5μg/mlへ量を増大させた抗体の存在下、5乃至10日間インキュベートした。<HVB>モノクロナール抗体を負のコントロールとして使用した。
直径が100μより大きいコロイニーの定量分析では0.5μg/mlの抗体18にて処理された培養物において、コロニーの数が、約66%減少したことを表わしている(図4)。
本発明の抗体が、IGF-I受容体(IGF-IR)にリガンドIGF-IおよびIGF-IIの結合を遮断できることを決定するために、放射性活性に標識化されたリガンド・ペプチドによる競合的実験を行った。ヒト腫瘍細胞(HT29,NCI H322M,0.5 to 1 x 105/ml)を、2mM L-グルタミン、1x非必須アミノ酸(Gibco.Cat番号11140-035)、1mMのビルビン酸ナトリウム(gibco,Cat.番号11360-039),および10%の熱不活性化FCS(PAA,Cat.番号A15-771)により補給されたRPMI 1640培地(PAA,Cat.番号E15-039)において平板培養した。T175形式(format)の6個の瓶において各実験用のそれぞれの培地に20mlの細胞を植え付けし、そして37℃で、5%のCO2で2日間培養し単層の密集化細胞が得られた。
細胞を、2000rpm(Eppendorf,5415C)で5分間遠心分離機にて再単離し、そして上澄み液を取り出した。1mlの結合緩衝液に2度洗浄した後、シンチレーション管に移した。細胞表面受容体に結合された放射性活性のペプチド量を、シンチレーション計数機で測定した。
IGF-I受容体にIGF-IおよびIGF-IIペプチドの結合を抗体が抑制できることを実証する得られたIC50の曲線を図5および図6に示す。
抗体18に対する平均IC50値が、0.3nMである。抗体αIR3に対する結果を、図7に示す。IGF-II結合に対する検出可能な抑制が全く観察されなかった。
IGF-IRモノクロナール抗体のエピトープ・マッピングに対する、親和性測定(実施例2)に対する同様の方式が、選択されたが、IGF-IRを溶液中抗体と共に少なくとも0.5時間室温でプレ・インキュベートした。この混合物を注入し、そしてIGF-IRの結合(又は抑制)が検出された。このアッセイにより、IGF-IRに結合するモノクロナール抗体の相互抑制活性の測定が可能である。本発明の抗体が、aa 217-274(Gustafson,T.A.,and Rutter,W.J.,Biol.Chem.265(1990) 18663-18667)へ結合に周知である抗体である、αIR3と共にIGF-IRへの結合に対し競合することが、見出された。
IGF-I受容体(IGF-IR)の活性化およびリン酸化を本発明の抗体が抑制できることを決定するために、競合実験をIGF-Iペプチドにて行いそしてその後、リン酸化チロシンに対し特異的抗体にてウエスターン・ブロットテング分析を行った。
ヒト腫瘍細胞(HT29,NCI H322M,5x104/ml)を、2mM L-グルタミン、1x非必須アミノ酸(Gibco.Cat番号11140-035)、1mMのビルビン酸ナトリウム(gibco,Cat.番号11360-039),および0.5%の熱不活性化FCS(PAA,Cat.番号A15-771)により補給されたRPMI 1640培地(PAA,Cat.番号E15-039)において平板培養した。IC50の値を決定するために、12ウエルのプレートを、各実験のためのそれぞれの培地に1mlの細胞にて植え付けし、そして37℃で、5%のCO2で2日間培養した。
その培地を、吸引により慎重に取り出し、そしてウエル当り100μの冷えた溶離緩衝液 (50mM Hepes pH 7.2,150 mM NaCl,1mM EGTA,10%glycerol、1% のTriton(商標)-X100、100mM NaF,10mM Na4P2O7,Complete(商標)protease inhibitor) が加えられた。
IGF-IRの免疫沈降のために、IGF-IRβに対し(C-20,Santa Cruz Biotechologies)1μlのポリクロナール抗体、又はヒトのIGF型1受容体の細胞外ドメイン(α-鎖)のアミノ酸440-586内のエピトープを認識するマウスのモノクロナール抗体(IgG1)を加える(mAb 24-55,GroPep)直前、細胞溶離物の残りの上清液にて、清浄化回転(clearifying spin)、(13000rpmおよび4℃で10分間)が行こなわれた。
観察により、IGF-IRとAkt/PKBとの両方のIGF-Iにより誘発されたリン酸化の遮断を図8に示す。
腫瘍細胞におけるIGF-I受容体(IGF-IR)の量に及ぼす本発明の抗体の効果を検出するために、時間経過による実験およびその後IGF-IR特異的抗体によるウエスターン・ブロッテング分析を行った。RPMI 1640培地(PAA,Cat.番号E15-039)においてヒト腫瘍細胞(HT29, 5x104 細胞/ml)が、2mM L-グルタミン、1x非必須アミノ酸(Gibco.Cat番号11140-035)、1mMのビルビン酸ナトリウム(gibco,Cat.番号11360-039),および10%の熱不活性化FCS(PAA,Cat.番号A15-771)により補給された。各培養期間に対し、12ウエルのプレートの1を、各実験のためのそれぞれの培地で1mlの細胞にて植え付けし、そして37℃で、5%のCO2で24時間培養した。
IGF-IRに対し特異的抗体(C-20,Santa Cruz Biotechologies)1μlを、IGF-IRのタンパク質レベルを決定するために使用した。抗体の追加より24時間以下の時間後、本発明の抗体により誘発されたIGF-IRの下限調節が観察された。
さらに本発明の抗体が、インスリン受容体(IR)にインスリンの結合を遮断するかどうかを決定するために、放射性活性にて標識化されたリガンド・ペプチドによる競合的実験を行った。ヒト腫瘍細胞(HT29,NCI H322M,0.5 to 1 x 105/ml)を、2mM L-グルタミン(Gibco,Cat.番号25030-024)、および10%の熱不活性化FCS(PAA,Cat.番号A15-771)により補給された高グルコース(PAA,Cat.番号E15-009)を伴うMEM Dulbecco培地(DMEM)において平板培養した。T175形式(format)の6個の瓶において、各実験用のそれぞれの培地に20mlの細胞を植え付けし、そして37℃で、5%のCO2で2日間培養し密集化した単層の細胞が得られた。
0.1%のCH3COOHに溶解した、I125-標識化したインシュリン・ペプチド(Amersham,Cat.番号IM166,~2000 Ci/mmol)を、結合緩衝液中で最終活性が4*105の計数値/(分 x ml)に希釈した。前希釈した25μlのI125で標識化したインシュリン・ペプチドと共に75μlの抗体を、200μlの細胞懸濁液(最終抗体濃度200nM) に加え、そして4℃で3.5時間インキュベートした。
その結果により、本発明の抗体が、インスリン受容体へインスリン・リガンドの結合を干渉しないことが実証された(図9)。
本発明の抗体のIGF-IR刺激活性を除外するため、IGF-Iリガンドを存在しないが、本発明の抗体そして引用の抗体(αIR3、Oncogene,Germany)の存在にて、IGF-IRの燐酸化が決定される。これは、燐酸化状態特異的抗体を用い、ウエスターン・ブロット分析により実行される。IGF-IRにてトランスフェクトされた(5*104細胞/ml,Pietrzkowski,Z.らによる、Cell Growth Differ.4(1992)199-205)3T3細胞(ATCC CRL 1658)が、2mMnのL-グルタミン(Gibco,CatNo.25030-024)および0.5%の熱不活性FCS(PAA,Cat番号A15-771)にて補給された高グルコース(PAA,Cat番号 E15-009)によるMEM Dulbecco 培地(DMEM)に入れられるか、又はヒト腫瘍細胞(HT29,NCl H322M,5*104/ml)が、2mMのL-グルタミン、1x非必須アミノ酸(Gibco,Cat番号11140-035)、1mMのピルベイトナトリウム(Gibco,Cat番号11360-039)、および0.5%熱不活性FCS(PAA, Cat番号A15-771)にて補給されたRPMI 1640培地(PAA, Cat番号E15-039)に入れられる。C50値を判定するために12のウエル・プレートを、各実験のためのそれぞれの培地に1mlの細胞にて接種し、そして37℃および5%のCO2で2日間培養した。
腫瘍がヌードマウスにおいて誘発され、そして相違する濃度による本発明の抗体を一度で治療した。治療より24時間後、腫瘍を抽出し液体窒素下で均一化(homogenized)された。冷やした溶離緩衝液を、緩衝液の体積と腫瘍の重量比を3:1にして (50mMのHepes pH7.2, 150mMのNaCl,1mMのEGTA,10%のグリセロール,1%のTriton-X100、100mMのNaF,1mMのNa3VO4, 10mMのNa4P2O7,CompleteTM protease プロテアーゼ抑制剤,1mMのPMSF) 加え、そして解凍した腫瘍均質物を完全に混合した。
紹介
本発明による抗体が、C1qを固定できることを決定するため、ELISAが使用される。C1qが、獲得免疫系の1部で、そして免疫複合体に結合することにより、幾つかの酵素原の順次活性化を引き金とする。次にその酵素が、C3分子の分割を起こし、それが炎症反応、異種物のオプソニック作用、又は細胞膜の異常な粒子および溶離物を開始することになる。
主にELISAプレートが抗体の濃度範囲にて被覆され、そこにヒトのC1qが加えられる。C1qの結合が、ヒトC1qに対向する抗体により、その後にペルオキシダーゼ標識化結合体により検出される。
抗体18,8および23、そしてコントロール抗体を、10,5,1および0.5μg/mlの濃度にて試験した。試料表1が、試験された試料の明細を示している。負のコントロールとして、C1qの結合が非常に弱いヒトIgG4(CLB,ストック0.5μg/ml)を使用した。ヒトIgG1を正のコントロールとして組み入れた。2μg/mlの濃度のヒトC1qストック溶液を使用した。C1qを検出するために、C1qに対向するラビット抗体(Dako)および西洋ワサビペルオキシダーゼ(Sigma)に結合した抗-ラビットIgG抗体が、使用された。
計算および適合する曲線
試験されたHuMAbの最大結合(Bmax)に関する計算では、Graphpad Prism softwareを用いたを非線形適合(1部位の結合)回帰曲線使用し決定した。
本発明による抗体が、ヒトのC1qタンパク質の投与量依存を示す。405nmの光学的密度(OD405)をHuMAb濃度に対しプロットし、そしてその曲線が、非線形回帰曲線を用いて適合した。最大結合(Bmax)として最良適合値が試料表2に一覧的に示され、同様曲線(R2)の関連係数およびそれぞれの値に対する標準的偏差を示す。最も低い関係係数が、0.950(IgG4)の値であった。最大結合値が0.279の場合、ヒトIgG4(負のコントロール)が、C1qの最小結合を示す。正のコントロールIgG1および同様最大結合が、それぞれ1.729と2.223にて示される。
C1q結合ELISA(n=3)にて試験されたHuMAbの最大結合(Bmax)
ヒトのIgG4(負のコントロール、0.279のO.D.)の結合C1qと比較すると、試験された全ての抗体が、C1qを等しく固定することができる。
生成されたHuMAb 抗体を惹起できることを決定するために、免疫イフェクター機構、相補性依存細胞傷害(CDC)および抗体依存性細胞の細胞障害(ADCC)の研究が行われた。
CDC(National Cancer Institute,lung adenocarcinoma cell line),H322M,H460およびNIH 3T3細胞(2-6x106)を、45乃至120分100μのCi51Cr(Amersham Pharmacia Biotech,UK,Cat CJS11)にて標識化した。標識かした後、細胞を40mlのPBSにて2回洗浄し、そして1500rpmにて3分間回転(spun)した。次に細胞は、丸底プレート中で、1容量50μlにてウエル当り5,000を入れた。
ADCCにおける抗体の効果を研究するために、H322M,H460およびNIH 3T3又は他の適切なIGF-IR発現細胞(2-6x106)を、45乃至120分100μのCi51Cr (Amersham Phamacia Biotech,UK,Cat CJS11) にて標識化し、40mlのPBSにて2回洗浄し、そして1500rpmにて3分間回転(spun)した。その細胞が、丸底プレート中で、1容量50μlにてウエル当り5,000入れられた。HuMAb抗体を、容量が50μlの培養培地に、25乃至0.1μg/mlの範囲の最終濃度で、50μlの細胞懸濁液へ加え、そして30乃至60分インキュベートした。
CDC又はADCCにより最大の細胞溶離が、各標的細胞から放射能活性の自然放出として補正された洗浄剤により溶離された標的細胞から放射能の最大放出の%として表現される。
確立された効果により無胸腺症(athymic)ヌード・マウスにおける腫瘍を誘発することにより、in vivoにおける抗体の効果を研究した。ヒトH322M NSCLC細胞を、6週間経過した無胸腺症(athymic) nuマウス(nu/nu)に、マトリゲル(Matrigel)と共に皮下に共注入した。その目的のために、5x106H322M細胞を、100μl培養培地に集中させ、そして100μlのマトリゲル(Matrigel)にて今後した。この混合物200μlを、マウスの右側腹へ注入した。腫瘍の体積を、ノギス(Vernier calipers)を用い週2回腫瘍に直径を測定することにより、Geranらにより最初に公開された公式により計算し、Geranら (「Protocols for screening chemical agents and natural products against animal tumors and other biological systems」、Cancer Chemother. Rep.11.301,1972) が、腫瘍体積[mg = (長さx(幅)2)を公開している。
ヒトのIGF-IRを過剰発現するマウスの3t3繊維芽細胞が使用されたのを除ぞいて、腫瘍が、実施例15記載のように必ずヌードマウスに誘発される。約180mgの確立された腫瘍を有するマウスが、抗体18を18,6又は0.6mg/kgで7回として週1度腹腔内にて治療した。再度治療が、負荷用量として(36,12および1.2mg/kg)として与えられる抗体の二重化された用量で開始さた。抗体による治療により、週1度18および6mg/kgにて投与される場合、腫瘍の成長が遅れる可能性があることが、実験により実証された。

Claims (19)
- IGF-IRに結合し、且つIGF-IRへのIFG-I及びIFG-IIの結合を抑制する抗体において、
a) IgG1のアイソタイプであり、
b) IGF-IRへIGF-IIの結合の抑制におけるIC50値に対する、IGF-IRへIGF-Iの結合の抑制におけるIC50値の比率が1:3から3:1を示し、
c) 0.5%の熱不活性化胎児牛血清(FCS)を含む培地中で、HT29細胞を使用する細胞リン酸化アッセイにおいて、前記抗体を用いない前記のアッセイと比較した場合、5nMの濃度で少なくとも80%にてIGF-IRのリン酸化を抑制し、そして
d) 0.5%の熱不活性化胎児牛血清(FCS)を含む培地中で、細胞当り400,000乃至600,000分子のIGF-IRを備えた3T3細胞を使用する細胞リン酸化アッセイにおいて、前記抗体を用いない前記のアッセイと比較する場合、10μMの濃度でIGF-IRのリン酸化として測定されたIGF-IR刺激活性を全く示さない、
ことを特徴とする抗体。 - 前記抗体が、ADCCにより抗体100nMの前記抗体の濃度で、24時間後の調製したIGF-IR発現細胞のうち、20%以上の細胞死を誘発することを特徴とする、請求項1記載の抗体。
- 前記抗体が、CDCにより、100nMの抗体濃度で、4時間後、IGF-IR発現細胞の調製物のうち、20%以上の細胞死を誘発することを特徴とする、請求項1又は2のいずれか1項記載の抗体。
- ヒト抗体又はヒト型化抗体であることを特徴とする請求項1乃至3のいずれか1項記載の抗体。
- 約10-13乃至10-9M(KD)の親和性を特徴とする請求項1乃至4のいずれか1項記載の抗体。
- a) CDRsとして、配列番号1または3のCDR1(aa31-35)、CDR2(aa50-66)およびCD3(aa99-107)を含む抗体の重鎖、
b) CDRsとして、配列番号2または4のCDR1(aa24-34)、CDR2(aa50-56)およびCD3(aa89-98)を含む抗体の軽鎖、
の配列を有する相補決定領域(CDRs)として含むことを特徴とする請求項1乃至5のいずれか1項記載の抗体。 - ハイブリドーマ細胞株<IGF-IR>HUMAB Clone18(DSM ACC2587)又は<IGF-IR>HUMAB Clone22 (DSM ACC2594)から得ることができる、請求項1乃至6の何れか1項記載の抗体。
- 医薬組成物生成のための請求項1乃至7のいずれか1項記載の抗体の使用。
- 医薬的有効量にて請求項1乃至7のいずれか1項記載の抗体を含む医薬組成物。
- <IGF-IR>HUMAB Clone18(DSM ACC2587)および<IGF-IR>HUMAB Clone22(DSM ACC2594)のハイブリドーマ細胞株。
- 請求項1乃至7記載の抗体の医薬的に有効量を含む医薬組成物の生成方法。
- 下記定義のそれぞれ別な抗体と共に構築できるポリペプチドをコードする核酸において、
ここで前記ポリペプチドが、
a) CDRsとして、配列番号1または3のCDR1(aa31-35)、CDR2(aa50-66)およびCD3(aa99-107)を含む抗体の重鎖、
b) CDRsとして、配列番号2または4のCDR1(aa24-34)、CDR2(aa50-56)およびCD3(aa89-98)を含む抗体の軽鎖、
のいずれである核酸。 - 原核又は真核宿主細胞中で前記核酸を発現できる請求項12記載の核酸を含む発現ベクター。
- 請求項13記載のベクターを含む原核又は真核宿主細胞。
- IGF-IRに結合し、且つIGF-IRに対するIGF-I及びIGF-IIの結合を抑制するポリペプチドの生成方法において、原核又は真核宿主細胞中で、請求項11記載の重鎖をコードする核酸、および軽鎖をコードする核酸を発現し、そして前記細胞から前記ポリペプチドを取り出すことを特徴とする方法。
- 請求項1乃至7のいずれか1項記載の抗体の治療としての有効量を患者に投与することを特徴とする、抗腫瘍治療が必要な患者の治療方法。
- 抗体が、細胞障害剤、その前駆薬剤、又は細胞障害性放射線治療を組み合わせて投与されることを特徴とする請求項16記載の方法。
- 0.5%の熱不活性胎児牛血清(FCS)を含む培地中で、細胞当り400,000乃至600,000分子のIGF-IRを備えた3T3細胞を使用した細胞リン酸化アッセイを、前記抗体にて行い、そして前記抗体を用いることなくこうしたアッセイと比較した場合、10μMの濃度でPKBリン酸化として測定されたIGF-IR刺激活性を全く示さない前記抗体が、選択されることを、特徴とするIGF-IRに対し複数の抗体からIGF-IRに対する抗体を選択する方法。
- 前記抗体を伴う0.5%の熱不活性胎児牛血清(FCS)を含む培地中で、細胞当り400,000乃至600,000分子のIGF-IRを備えた3T3細胞を使用した細胞リン酸化アッセイを行うIGF-IRに対し複数の抗体からIGF-IRに1の抗体を選択し、組み換え発現の手段により前記抗体を生成し、前記抗体を取り出し、そして前記抗体と医薬的に受け入れ可能な緩衝液、そして/又はアジュバントとを組み合わせることを特徴とする医薬的組成物の調製方法。
