亡原因の第2である。前立腺癌はアンドロゲン感受性腫瘍であるため、進行前立
腺癌患者の治療法の1つとして例えば精巣除去によるアンドロゲン離脱が使用さ
れている。アンドロゲン離脱は、前立腺腫瘍で広範なアポトーシスを引き起こし
、従って疾患が退縮する。しかし精巣除去に誘導されたアポトーシスは完全では
なく、最終的に、生存している腫瘍細胞のアンドロゲン非依存性への進行が起き
る。この進行は、生存とクオリティオブライフの改善に対する大きな障害であり
、従ってアンドロゲン非依存性細胞を標的とする試みが行われている。これらの
試みは、アンドロゲン非依存性腫瘍細胞を標的とする非ホルモン治療法に集中し

細胞では、精巣除去後直ちにTRPM−2の発現が上昇し、ラットの前立腺細胞
では精巣除去後3〜4日でピークレベルに達し、多量の細胞死滅の開始と一致す
る。これらの結果から一部の研究者は、TRPM−2が細胞死滅のマーカーおよ
びアポトーシスのプロモーターであると結論している。一方、セルトリ細胞と一
部の上皮細胞は、細胞死滅レベルを上昇させることなく高レベルのTRPM−2
を発現するという観察結果は、この結論が正しいかどうかについて疑問を投げか
けている。
1−2437(1995)は、前立腺細胞の死滅におけるTRPM−2の役割を
より明確に解明するために行ったインビトロの実験を報告した。彼らは、TRP
M−2をコードする遺伝子でトランスフェクションしたLNCaP細胞を使用し
て、このタンパク質の発現が、腫瘍壊死因子α(TNFα)(LNCaP細胞は
、これに対して非常に感受性であり、通常約12時間以内で細胞の死滅が起きる
)の作用を改変するかどうかを観察した。トランスフェクションしたLNCaP
細胞をTNFαで処理すると、数時間の間一時的にTRPM−2レベルが上昇す
るが、細胞死滅に先行するDNA断片化が観察されるまでに、これらのレベルは
消滅することが証明された。TRPM−2配列の塩基1〜21に対応するアンチ
センス分子(しかし、他のTRPM−2アンチセンスオリゴヌクレオチドではな
い)を使用すると、TRPM−2の発現が大幅に低下し、TNFαに暴露された
LNCaP細胞のアポトーシス性細胞死が増加した。このためセンシバー(Sens
ibar)らは、TRPM−2の過剰発現はTNFの細胞障害作用から細胞を防御し
、TRPM−2の枯渇が、細胞死滅の開始の原因であるという仮説をたてた(た

究室で増殖させたLNCaP細胞中では、TRPM−2の発現レベルは非常に低

が変化する。すなわち、センシバー(Sensibar)らのデータから、TRPM−2

ビボでアンドロゲン離脱後のTRPM−2のレベルの変化が、治療的利点が有る
かどうかについて予測することは不可能である。確かに、TRPM−2は、アン
ドロゲン離脱後に種々の段階(有意なアポトーシス性細胞死が起きている段階を
含む)で前立腺腫瘍細胞中で多量に発現されるという事実は、インビボでのTR
PM−2の役割はもっと複雑かも知れないことを示唆する。すなわち、この分野
では、前立腺腫瘍細胞中のアポトーシス性細胞死のある面に関するデータを提供
するが、アンドロゲン非依存性の発症の遅延を提供する方法を教示も示唆もしな
い。

ンス療法は、前立腺癌および腎細胞癌の治療に応用することができる。 インビボで前立腺腫瘍細胞へのアンチセンスTRPM−2オリゴデオキシヌク
レオチド(ODN)の添加は、アンドロゲン非依存性の発症を遅らせるのに有効
である。すなわちある実施態様において本発明は、前立腺癌に罹っている個体の
前立腺癌の治療法であって、アンドロゲン離脱を開始して個体の前立腺腫瘍細胞
のアポトーシス性細胞死を誘導する工程と、腫瘍細胞によるTRPM−2の発現
を阻害するのに有効な組成物を個体に投与する工程とを含んでなり、こうして個
体のアンドロゲン非依存性状態への前立腺腫瘍細胞の進行を遅らせる上記方法を
提供する。さらに、アンチセンスTRPM−2と細胞障害性化学療法(例えば、
タキサン)を組合せて使用すると、ホルモン不応性の前立腺癌での化学療法感受
性を、相乗的に増強させる。本発明の別の実施態様において、TRPM−2以外
の抗アポトーシスタンパク質の発現を阻害する第2のアンチセンスODNが、ア
ンチセンスTRPM−2 ODNとともに投与される。 アンチセンスTRPM−2は、他のタイプの癌にも有効作用を有することがわ

、精巣除去後とアンドロゲン非依存性への進行中のヒト前立腺癌におけるTRP
M−2とBcl−2の発現の変化は、シオノギ(Shionogi)腫瘍細胞で観察され
たものに似ている。すなわち、シオノギ(Shionogi)腫瘍モデルは、前立腺癌細
胞の特徴の多くを模倣している。さらにシオノギ(Shionogi)腫瘍モデルは、ア
ンドロゲン非依存性の発症を遅らせる化合物の能力を評価するための、非常に有
用なモデルを提供する。精巣除去後の完全な腫瘍退縮にもかかわらず、急速に増
殖するアンドロゲン非依存性シオノギ(Shionogi)腫瘍は、1ヶ月後に必ず再発
し、これは、アンドロゲン非依存性への進行を遅らせることができる物質を評価
するための信頼できる終点を提供する。1ヶ月以内にシオノギ(Shionogi)腫瘍
モデル中で起きる事象は、一般に約2年以内にヒト患者で起きる。
センスODNの能力を、シオノギ(Shionogi)腫瘍モデルで精巣除去後の腫瘍容
量を測定することにより評価した。試験動物(n=7)を、緩衝化食塩水中のア
ンチセンスTRPM−2 ODN(配列番号1)の12.5mg/kgの繰り返し用
量で、1日1回腹腔内処理した。対照として、動物(n=7)をミスマッチOD
N(配列番号2)で処理した。図1に示すように、試験群および対照群の両方と
も、精巣除去直後に腫瘍容量の予測された減少を示したが、アンチセンスTRP
M−2 ODN処理したマウスでは対照より速く腫瘍が退縮した。対照群はまた
、アンドロゲン非依存性の進展に関連する腫瘍容量の予測された増加を示した。
これに対して、精巣除去の49日後には、アンチセンスTRPM−2 ODNを
使用して処理したマウスでは、腫瘍の再増殖はほとんど起きなかった。アンチセ
ンスTRPM−2 ODN処理したマウスでは腫瘍は結局再発したが、再発まで
の中央時間は、対照群の約2倍であった。すなわちTRPM−2の阻害は、アン
ドロゲン離脱直後に起きる細胞死滅の量を上昇させるだけでなく、アンドロゲン
非依存性の発症を遅らせるのにも有効である。アンチセンスTRPM−2 OD
Nで処理したマウス中の腫瘍容量のより急速な減少は、早期の発症とより広範な
精巣除去誘導性のアポトーシスが原因であった。これは、シオノギ(Shionogi)
腫瘍検体中のポリ(ADPリボース)ポリメラーゼ(PARP)切断断片を検出
して確認した(ミヤケ(Miyake)ら、Cancer Res.60:170−1

に最も有効であるかを評価するために、図2に示す種々のmRNA領域をつない
で一連の10個のアンチセンスホスホロチオエートODNを調製した。これらの
10個のODNの配列を、添付の配列リストに配列3〜12として示す。この1
0個のヒトアンチセンスODNを、TRPM−2トランスフェクションLNCa
P細胞とヒト前立腺癌PC−3細胞を使用して、TRPM−2 mRNAの発現
を阻害する能力について評価した。図3に示すように、試験したアンチセンスO
DNは、種々のレベルのTRPM−2 mRNA発現の阻害を示し、配列番号4
、5、および12で最適な結果が得られた。配列番号5は、センシバー(Sensib
ar)らが使用した、LNCaP細胞中のTRPM−2発現を阻害した配列に対応
し、TRPM−2 mRNAの最初の21塩基に相補的である。最も有効なダウ
ンレギュレーションは、配列番号4で起きた。TRPM−2 mRNAの開始ま
たは停止部位のいずれかとの重複は、有効な配列のすべてに共通であった。すな
わち一般的な意味で、本発明の方法は、翻訳開始部位または停止部位に広がるT
RPM−2 mRNAの領域に相補的なアンチセンスオリゴヌクレオチドを用い
て行うことができる。

ンスTRPM−2 ODNが、ヒトRCC CaKi−2細胞(例6を参照、後
述)の化学療法感受性を増強させることを示している。

観察されなかった。

した。相乗作用を示す可能性のある他の物質には、他の細胞障害性物質(例えば
、シクロホスファミド、トポイソメラーゼインヒビター)、血管形成インヒビタ
ー、分化物質および神経伝達インヒビターがある。同様に、TRPM−2アンチ
センスと他のアンチセンス種(例えば、アンチセンスBcl−2ODN)との組
合せは、ODN単独よりインビトロでシオノギ(Shionogi)細胞を死滅させるの
に優れていた。すなわちTRPM−2は、他のアンチセンス分子(例えば、アン
チセンスBcl−2、Bcl−xlおよびc−myc ODN)と共同して作用
して、より大きな効果を示すことができる。

ODNで処理したマウス中で、シオノギ(Shionogi)腫瘍はより速く退縮し、完
全な退縮が早期に起きた。さらに、アンチセンスTRPM−2 ODNで処理す
ると、実質的にアンドロゲン非依存性の発症を遅らせ、これは、対照動物での2
1日後の腫瘍容量の増加に反映される。アンチセンスTRPM−2またはミスマ
ッチ対照に関連する副作用は観察されなかった。

で解析した。TRPM−2 mRNAレベルは、腫瘍組織で有意に低かったが、
アンチセンスTRPM−2 ODNは、正常臓器中のTRPM−2 mRNAに
対して何の作用もなかった。

存性曲線が左にシフトし、IC50が5〜10倍低下した。パクリタキセルの代わ
りにミトキサントロンを使用して同様の結果が得られた(図12Aと図12B)

−2ODN(配列番号14)を添加して、例3の実験を繰り返した。結果を図6
に示す。アンチセンスTRPM−2 ODNとアンチセンスBcl−2ODNお
よびタキソールとの組合せは、タキソールの細胞障害性作用をさらに増強した。

チセンスODN配列(図2、配列番号3〜12)を合成し、ヒト前立腺癌PC−
3とトランスフェクションLNCaP細胞(TRPM−2を過剰発現する)中の
TRPM−2遺伝子発現を低下させる能力を、例2と同じ処理プロトコールを使
用して試験した。結果を図3に要約する。図示したように、配列番号4、5およ
び12は、TRPM−2発現の低下について活性である。これらの3つの配列は
重複するか、または翻訳開始または停止部位にすぐ隣接している。

株ACHN、CaKi−1、およびCaKi−2中のTRPM−2発現を、ノー

現の変化を評価するために行った。アンチセンスTRPM−2 ODNとタキソ
ールの組合せがCaKi−2細胞増殖に及ぼす作用を、MTT測定法を使用して
調べた。

られなかった。TRPM−2 mRNAとタンパク質発現の両方とも、すべての
3つのヒトRCC細胞株で検出され、CaKi−2について最も高いレベルであ
った。アンチセンスTRPM−2 ODN(配列番号1)は、用量依存性かつ配
列特異的にCaKi−2細胞でのTRPM−2発現を阻害したが、ミスマッチ対
照ODN(配列番号2)は阻害しなかった(図7A)。さらにアンチセンスTR
PM−2 ODNはタキソールの化学療法感受性を実質的に上昇させ、タキソー
ルのIC50をミスマッチ対照ODNと比較して1 log(500nM対50
nM)低下させた(図7B)。これらのデータは、TRPM−2とそのタンパク
質(クラステリン)は、正常な腎組織と比較してRCCでより高レベルで発現さ
れることを示し、アンチセンスTRPM−2 ODNは、進行性RCCの従来の
化学療法の細胞障害性を増強するのに有用かも知れないことを示している。

に上昇させることを見いだした(図8)。TRPM−2の過剰発現は、放射線誘
導性の細胞死滅に対する耐性を上昇させる。TRPM−2を過剰発現するヒト前
立腺LNCaP細胞(LNCaP/T1)は、放射線療法に対してより耐性であ
る(図9AとB)。ヒト前立腺癌PC−3細胞を100と500nMアンチセン
スTRPM−2 ODN(配列番号1)で処理すると、ミスマッチODN(配列
番号2)処理と比較して、4Gy放射線療法による単回処理後に細胞生存率が有
意に低下する(図10)。図11AとBは、インビトロで10、50、100n
MのアンチセンスTRPM−2オリゴで処理後に、ヒト前立腺癌PC−3の用量
依存性放射線増感を示す。
細胞株中の化学療法感受性を増強させるかどうかを調べるために、PC3腫瘍担
持マウスを、アンチセンスヒトTRPM−2 ODN+ミセル状パクリタキセル
またはミトキサントロン、およびミスマッチ対照ODN+でミセル状パクリタキ
セルもしくはミトキサントロンで処理した(図12Aと12B)。ODNを28
日間投与し、0.5mgのミセル状タキソールまたは0.3mgのミトキサント
ロンを2回投与(10日から14日、および24から28日)した。ミスマッチ
対照ODNで処理したものと比較して、アンチセンスODN処理動物では、腫瘍
サイズの有意な低下が観察された。この作用は、ミセル状パクリタキセルまたは
ミトキサントロンの2回目の投与後に、より顕著であった。
