Abstract

Description
본 발명은 "테스토스테론이 억제된 전립선 메시지-2"(testosterone-repressed prostate message-2, 이하 "TRPM-2"라 한다)에 결합하는 안티센스 올리고누클레오티드를 사용하여 암을 치료하는 안티센스 치료법에 관한 것이다.
본 출원은 1999년 2월 26일에 출원된 미국 가출원 60/121,726호에 대한 우선권주장 출원이다.
전립선암은 사람이 걸리는 암중에서 가장 흔한 암이며 서구에서는 암 사망률 2위를 차지하는 암이다. 전립선암은 안드로겐에 민감한 종양이므로 전립선암이 진행된 환자를 위한 치료법으로 안드로겐을 절제 등에 의해 추출하여 제거하는 것이 사용되고 있다. 안드로겐을 추출해내면 전립선 종양세포는 거의 사멸하여 이 질병이 퇴행될 수 있지만, 절제로 인한 세포사멸은 완전하지 않으며 결국 생존한 종양세포는 안드로겐 비의존성으로 진행된다. 이러한 진행은 삶의 질과 수명연장을 향상시키는데 있어 주된 장애가 되어 안드로겐 비의존성 세포를 표적화하기 위한 노력이 행해지고 있다. 이들 노력은 안드로겐 비의존성 종양세포에 대해 표적화된 비호르몬 치료에 초점을 맞추고 있으나 (Yagoda et al., Cancer 71 (Supp.3): 1098- 1109 (1993); Oh et al., J. Urol. 60: 1220-1229 (1998)), 지금까지 비호르몬제가 수명을 연장시키지는 못했다. 따라서 다른 접근이 시도되고 있다.
많은 단백질이 안드로겐 제거 후에도 전립선 종양세포에 의해 증가된 양으로 발현되는 것이 관찰되었다. 이러한 단백질 중 적어도 일부는 안드로겐 제거시에 관찰되는 어팝토시스성 (apoptosis) 세포사멸과 관련된다고 추정된다 (Raffo et al., Cancer Res..: 4448-4445 (1995); Krajewska et al., Am. J. TRPM-2는 다양한 범위의 제안된 활성을 갖는 보편적인 단백질이다. 전립선 상피세포에서, TRPM-2의 발현은 절제 직후에 증가하여 다량의 세포사멸 진행과 함께 절제후 3 내지 4일에 래트(rat) 전립선 세포에서 피크 수준에 도달한다. 이들 결과로부터 몇몇 연구자는 TRPM-2가 세포사멸에 대한 식별자이고 세포사멸의 촉진자라는 결론을 얻었다. 한편, 세르톨리(Sertoli) 세포와 일부 상피세포가 증가된 수준의 세포사멸 없이 높은 수준의 TRPM-2를 발현한다는 관찰은 이 결론이 정확한 것인지에 대한 의문을 야기시킨다.
전립선 세포사멸에서 TRPM-2의 역할을 보다 분명하게 밝히기 위해 시행된 시험관내 실험이 센시바 등(Sensibar et al., Cancer Research 55: 2431-2437 (1995))에 의해 보고되었다. 이들은 유전자 코딩하는 TRPM-2로 트랜스펙션된 LNCaP 세포를 사용하고 이 단백질의 발현이 종양탈저인자 α(TNF α)의 효과를 변경시키 는지를 관찰하였다. 여기서 세포사멸이 약 12 시간내에 정상적으로 발생하면서 LNCaP 세포는 매우 민감해진다. TNF α로 트랜스펙션된 LNCaP 세포 처치로 몇시간동안 TRPM-2 수준이 일시적으로 증가되나 이 수준은 세포사멸이 관찰되기 전에 DNA 분열시에 감소되었다. 다른 TRPM-2 안티센스 올리고누클레오티드가 아닌 TRPM-2 서열중 염기 1-21에 해당하는 안티센스 분자의 사용으로 TRPM-2 발현이 실질적으로 감소하고 TNFα에 노출된 LNCaP 세포에서의 어팝토시스성 세포사멸이 증가하였다. 이것을 센시바 등은 TRPM-2의 과발현이 TNF의 세포독성 효과로부터 세포를 보호할 수 있고 작용 메카니즘이 불분명하지만 TRPM-2 소모가 세포사멸 개시를 담당한다는 가설로 이끌었다.
센시바 등은 TRPM-2의 가능한 역할에 대한 정보를 제공하면서, TRPM-2의 발현이 트랜스펙션된 유전자에 의거한다는 단지 모델 시스템으로부터의 결과도 개시하고 있다. 더욱이, TRPM-2의 발현수준은 다른 실험실에서 성장한 LNCaP 세포에는 없거나 매우 낮다. 전립선 종양세포에서 안드로겐 제거될 때 생체내에서 얻어지는 결과는 보다 복잡하며 그 결과로서 수많은 단백질의 발현 수준이 변경된다. 따라서, 센시바 등의 데이터로부터 TRPM-2가 다른 단백질과 조합되어 존재할 때 동일한 기능을 수행하는지, 또는 생체내에서 안드로겐 제거 후에 TRPM-2의 수준 변화가 어떤 치료적 이점을 제공하는지를 기대할 수 없다. 상당한 어팝토시스성 세포사멸이 발생하는 단계를 포함하는 안드로겐 제거후 다양한 단계에서 TRPM-2가 전립선 종양세포에서 실질적 양으로 발현된다는 사실은 생체내의 TRPM-2의 역할이 보다 복잡해질 수 있다는 것을 제시하고 있다. 따라서 상기 기술은 전립선 종양세포에서 어팝 토시스성 세포사멸의 일정한 양태에 관한 데이터를 제공하지만, 안드로겐 비의존성 징후를 지연시키기 위한 방법의 교시나 제안을 제공하지는 못한다.
본 발명의 목적은 그러한 방법을 제공하는 것이다.
본 발명의 다른 목적은 전립선 종양세포에서 안드로겐 비의존성 징후를 지연시키는 치료적 안티센스 분자를 제공하는 것이다.
본 발명의 다른 목적은 TRPM-2를 발현하는 암으로부터의 암 세포의 약제감수성 또는 방사선 감도를 증가시키는 방법을 제공하는 것이다.
본 발명의 다른 목적은 TRPM-2의 발현을 저해하는 치료적 안티센스 분자를 제공하는 것이다.
본 발명에 따라서, TRPM-2 발현을 감소시키는 안티센스 치료법은 암 치료에 있어 치료적 이점을 제공한다는 것이 이제 밝혀졌다. 특히 그러한 안티센스 치료법은 전립선암 및 신세포암 치료에 적용될 수 있다.
생체내에서 안티센스 TRPM-2 올리고데옥시누클레오티드 (ODN)를 전립선 종양세포에 첨가하면, 안드로겐 비의존성 징후가 효과적으로 지연된다. 따라서, 본 발명의 한 양태로서, 본 발명은 전립선암에 걸린 개체의 전립선 암 치료방법을 제공하고, 이 방법은 안드로겐 제거를 실시하여 개체에서의 전립선 종양세포의 어팝토시스성 세포사멸을 유발하는 단계 및 종양세포에 의한 TRPM-2 발현을 저해하는데 효과적인 조성물을 개체에 투여함으로써 개체에서 안드로겐 비의존 상태가 되는 전립선 종양세포의 진행을 지연시키는 단계로 이루어진다. 더욱이, 안티센스 TRPM-2 와 세포독성 화학요법제 (예, 탁산)의 조합된 사용은 동반상승작용으로 호르몬 난치료성인 전립선암에서의 약제감수성을 증가시킨다. 본 발명의 다른 양태에 있어서, TRPM-2 이외의 안티-세포사멸성 단백질의 발현을 저해하는 제2의 안티센스 ODN을 안티센스 TRPM-2 ODN과 함께 투여한다.

본 발명은 안티센스 TRPM-2 ODN과, 암 치료에의 이들 조성물의 사용에 관한 것이다. 본 발명은 암 세포가 TRPM-2를 발현시키는 암 치료에 적용될 수 있다. TRPM-2를 발현시키는 세 가지 유의한 암 세포종은 전립선 암세포, 인간 신세포암 (RCC) 및 일부 유암 세포이다.
한 구체예에서, 본 발명은 절제로 인한 종양세포 사멸을 증가시키고 안드로겐 비의존성이 되는 전립선 종양세포의 진행을 지연시키는 방법; 전립선암에 걸린, 인간을 포함하는 개체의 치료방법; 및 그러한 방법에 사용하는데 효과적인 치료제를 제공한다. 본 발명의 치료방법은 전립선암이 진행된 개체 치료에 가장 일반적으로 사용된다.
절제로 인한 종양세포 사멸의 증가 및 안드로겐 비의존성이 되는 안드로겐에 민감한 전립선 암세포 진행의 지연은 종양세포에 의한 TRPM-2 발현을 저해함으로써 달성된다. 실험은 세 가지 모델 시스템, 즉 생체내의 시오노기 종양 모델, 인간 TRPM-2 트랜스펙션된 LNCaP 모델 및 인간 PC-3 모델에서 시행하고, 안드로겐 비의존성의 지연이 되게 하는 그러한 저해는 안드로겐에 민감한 전립선 종양 세포를 안티센스 올리고데옥시누클레오티드 (ODN)로 처치함으로써 달성될 수 있다는 것을 함께 입증하였다.
제1실험에서, 마우스 TRPM-2 안티센스 분자(서열번호 1)의 시오노기 종양 모델에서 안드로겐 비의존성 징후를 지연시키는 능력을 평가하였다. 시오노기 종양 모델은 수컷 동계 숙주에서 피하 성장하는 안드로겐 의존성 마우스 유암의 이종이식이다. 시오노기 종양 세포는 매우 발암성이며 국부적으로 침습성이다. 이 세포는 전립선 종양세포의 관찰된 행동을 의사하는 방식으로 안드로겐 제거에 반응하도록 나타나고 인간에서 전립선암에 대한 유효한 모델로서 수용되었다 (Bruchovsky et al., Cancer Res. 50: 2275-2282 (1990); Rennie et al., Cancer Res. 48: 6309-6312 (1998); Bruchovsky et al., Cell 13: 272-280 (1978); Gleave et al., in Genitourinary Oncology, pp. 367-378, Lange et al., eds, Lippencott (1997); Gleave et al., J. Urol. 157: 1727-1730 (1997); Bruchovsky et al., The Prostate 6: 13-21 (1996)). 따라서, 안드로겐 제거는 매우 재현가능한 방식으로 세포사멸 및 종양 퇴행을 촉진한다. 더욱이, 안드로겐 비의존성으로의 진행동안과 절제후에 인간 전립선암에서 TRPM-2 및 Bcl-2의 발현 변화는 시오노기 종양 세포에서 관찰된 것과 유사하다. 따라서, 시오노기 종양 모델은 전립선 암세포의 많은 특징들을 의사한다. 또한 시오노기 종양 모델은 화합물의 안드로겐 비의존성의 징후 지연능력을 평가하는데 매우 유용한 모델을 제공한다. 절제후 완전한 종양 퇴행에도 불구하고, 급속히 성장하는 안드로겐 비의존적 시오노기 종양은 1개월 후에 일정하게 재발되는데, 이 시점은 안드로겐 비의존성으로의 진행을 지연시킬 수 있는 제제를 평가하는데 신뢰성있는 종점을 제공한다. 일반적으로, 1개월 내에 시오노기 종양 모델에서 발생하는 경우는 약 2년내에 인간 환자에서 발생한다.
TRPM-2의 발현을 저해하는 안티센스 ODN의 안드로겐 비의존성 징후 지연능력을 시오노기 종양모델에서 후절제 종양 크기를 측정함으로써 평가하였다. 시험동물 (n=7)을 완충염수액중의 안티센스 TRPM-2 ODN (서열번호 1) 12.5 mg/kg의 반복 투여량으로 1일 1회 복강내 처치하였다. 대조군으로서, 동물 (n=7)을 비합치 ODN (서열번호 2)으로 처치하였다. 도 1에 도시한 바와 같이, 시험군 및 대조군 둘다에서 절제직후 종양 크기의 기대한 감소를 보였으나, 안티센스 TRPM-2 ODN 처치된 마우스에서의 종양은 대조군에서 보다 신속하게 퇴행하였다. 대조군도 안드로겐 비의존성의 발전과 관련되어 기대한 종양 크기 증가를 나타냈다. 반대로, 후절제 49일째에 안티센스 TRPM-2 ODN을 사용하여 처치한 마우스에서는 종양 재성장이 거의 일어나지 않았다. 종양은 안티센스 TRPM-2 ODN 처치된 마우스에서 다시 재발하였으나, 재발에 걸리는 중위수 시간은 대조군 시간의 약 2배이다. 따라서, TRPM-2의 저해는 안드로겐 제거직후에 발생하는 세포사멸량을 증가시킬 뿐만 아니라 안드로겐 비의존성 징후를 지연시키는데 효과적이다. 안티센스 TRPM-2 ODN으로 처치된 마우스에서 보다 신속하게 종양크기가 감소한 것은 조기 진행과 대규모의 절제로 인한 세포사멸 때문이었다. 이것은 시오노기 종양 표본에서 폴리(ADP-리보스) 폴리머라제 (PARP) 절단 단편을 검출함으로써 확인하였다 (Miyake, et al., Cancer Res. 60: 170-176 (2000)).
TRPM-2 mRNA 서열에 상보적인 어떠한 인간 안티센스 ODN이 이 목적에 가장 효과적인 것을 평가하기 위해, 일련의 안티센스 포스포로티오에이트 ODN 10개를 도 2에 도시한 바와같이 다양한 mRNA 부위를 포함하여 제조하였다. 이들 10개의 ODN의 서열은 서열번호 3-12로서 첨부된 서열목록에 기재되어 있다. 10개의 인간 안티센스 ODN은 TRPM-2 mRNA의 발현을 저해하는 능력에 대해 인간 전립선암 PC-3 및 TRPM-2 트랜스펙션된 LNCaP 세포를 사용하여 측정하였다. 도 3에 도시한 바와 같이, 안티센스 ODN으로 시험하여 TRPM-2 mRNA 발현의 가변성 저해수준 결과를 얻고, 그 중 서열번호 4, 5 및 12로부터 최상의 결과를 얻었다. 서열번호 5는 LNCaP 세포 에서 TRPM-2 발현의 저해를 일으킨 센시바 등에 의해 사용된 서열에 해당하고, TRPM-2 mRNA의 처음 21개의 염기에 상보적이다. 가장 효과적인 하향조절은 서열번호 4에 의해 일어났다. 모든 효과적인 서열에 대한 공통부는 TRPM-2 mRNA의 개시 또는 종결 부위와 중복된다. 따라서 일반적 센스에서 본 발명의 방법은 번역 개시부위 또는 그 종결부위를 포함하는 TRPM-2 mRNA 부위에 상보적인 안티센스 올리고누클레오티드로 시행할 수 있다.
본 발명의 다른 양태에 따라서, 전립선암에 걸린 인간 개체를 포함한 개체의 치료는, 안드로겐 제거를 실시하여 개체에서의 전립선 종양세포의 어팝토시스성 세포사멸을 유발하고, 개체에 종양 세포에 의한 TRPM-2 발현을 저해하는데 효과적인 조성물을 투여함으로써 개체에서 전립선 안드로겐 비의존 상태가 되는 종양 세포의 진행을 지연시킴으로써 달성된다.
안드로겐 제거의 실시는 외과적 (두개의 고환 제거) 또는 의학적 (테스토스테론의 약제-유발된 억제) 절제를 통해 수행될 수 있으며, 이는 전립선암의 치료에 있어 현재 통용되고 있다. 의학적 절제는 LHRH제 또는 안티안드로겐을 포함하는 다양한 방법으로 수행될 수 있다 (Gleave et al., CMAJ 160: 225-232 (1999)). 가역적인 안드로겐 제거를 시행하는 간헐적 치료는 Gleave et al., Eur. Urol. 34 (Supp. 3): 37-41 (1998)에 기재되어 있다.
TRPM-2 발현의 저해는 일시적일 수 있으며, 이상적으로는 안드로겐 제거와 함께 일어나야 한다. 인간에 있어서, 이것은 발현의 저해가 안드로겐 제거후 1일이나 2일내로부터 약 3 내지 6개월까지 효과적인 것을 의미한다. 이를 실시하는데 있 어 다양한 투여량이 필요할 수 있다. 그러나, 절제 전부터 본 발명의 범위를 벗어나지 않는 실질적 시간까지 시간을 더욱 연장할 수 있다는 것을 이해한다.
안티센스 TRPM-2 ODN도 인간 신세포암 (RCC)에서 약제감수성을 증강시킨다는 것을 알았다. RCC는 목적 반응률이 10 %보다 높은 활성 화학요법제가 없는 화학적 저항성 질병이다. 세포사멸을 진행하는 신장 근처의 포선형 세포에서 증가된 TRPM-2 발현이 요관폐쇄 및 아미노글리코시드를 포함하는 다양한 자극후에 관찰되었다. 그러나, RCC에서의 기능적으로 유의한 TRPM-2 발현은 증명되지 못하였다. 그러나 시험결과는 안티센스 TRPM-2 ODN이 인간 RCC CaKi-2 세포에서 약제감수성을 증강시킨다는 것을 나타낸다 (하기 실시예 6 참조).
또한 안티센스 TRPM-2 ODN이 방사선에 대한 감도를 증가시킨다는 것도 알았다 (실시예 7 및 도 8 참조).
TRPM-2 발현 저해는 안티센스 ODN, 특히 번역 개시부위 또는 그 종결부위를 포함한 TRPM-2 mRNA의 부위에 상보적인 안티센스 ODN의 투여로 수행될 수 있다. 인간의 전립선암을 치료하기 위해, 특이적으로 유효한 서열은 서열번호 4, 5 및 12에 나타낸 것이다.
사용된 ODN은 생체내에서 ODN의 안정성을 증가시키기 위해 변형될 수 있다. 예를들면, ODN은 누클레아제 분해에 대해 내성이 증가된 포스포로티오에이트 유도체(다리결합하지 않은 포스포릴 산소원자를 황원자로 치환)로서 사용될 수 있다. MOE 변형(ISIS 골격)도 유효하다.
안티센스 ODN은 단독 투여하거나 약학적으로 허용가능한 지질 담체와 함께 투여하는 본 기술분야에서 공지된 각종 메카니즘을 사용하여 투여될 수 있다. 예를들면, 안티센스 운반용 지질 담체는, 본문에 참고로 포함되는 미국특허 제 5,855,911 호 및 제 5,417,978 호에 개시되어 있다. 일반적으로, 안티센스는 정맥내, 복강내, 피하 또는 구강 경로, 또는 직접적인 국부 종양 주사로 투여된다. 시오노기 마우스 모델을 사용하여 수행된 실험으로부터, 안티센스 ODN은 우선적으로 종양세포에 활성이라는 것을 알 수 있다. 비종양 조직중에서 TRPM-2 발현은 실질적으로 영향을 받지 않으며, 안티센스 ODN 투여의 부작용은 관찰되지 않았다.
투여된 안티센스 ODN의 양은 전립선 세포에서 TRPM-2의 발현을 저해하는데 효과적인 양이다. 이 양은 사용된 안티센스 ODN의 유효성과, 사용된 어느 담체의 성질 둘다에 의해 다양해진다는 것을 이해한다. 어떤 조성물에 대한 적당한 양의 측정은 적절한 치료수준을 평가하도록 설계된 일련의 표준 시험을 통해 본 기술분야내에 있다.
본 발명에 따른 전립선암 치료방법은 화학요법제 및/또는 상이한 표적을 지향한 추가의 안티센스 ODN의 투여를 더 포함할 수 있다. 예를들면, 안티센스 TRPM-2 ODN이 탁산 (파클리탁셀 또는 도세탁셀) 및 미톡산트론과 같은 종래의 화학요법제에 대한 감도를 증가시킨다는 것을 시오노기 종양 모델을 사용하여 알았다 (도 12A 및 도 12B). 도 12A 및 도 12B에 도시한 바와 같이, 탁솔 또는 미톡산트론의 존재하에서 안티센스 TRPM-2 ODN으로 처치하여 탁솔이나 미톡산트론과 비합치(MM) ODN의 조합과 비교하여 종양 크기를 감소시켰다. 동반상승작용으로 인한 활성을 나타내는 다른 제제에는 다른 세포독성제(예, 시클로포스파미드, 토포이소머라제 저 해제), 맥관형성 저해제, 분화제 및 시그널 트랜스덕션 저해제가 있다. 유사하게, TRPM-2 안티센스와 다른 안티센스종, 즉 안티센스 Bcl-2 ODN의 조합은 ODN 단독보다 시험관내에서 시오노기 세포를 사멸시키는데 더 잘 작용하였다. 따라서, TRPM-2는 안티센스 Bcl-2, Bcl-xl 및 c-myc ODN과 같은 다른 안티센스 분자와 함께 작업하여 보다 큰 효과를 얻을 수 있다.
이제 본 발명은 다음의 제한되지 않는 실시예를 참조하여 더욱 자세하게 기술된다.
실시예 1
시오노기 종양 모델 실험은 이식가능한 SC-115 AD 마우스 유암의 토론토 서브라인으로부터의 세포를 사용하여 수행하였다. 생체내 연구를 위해, 약 5 ×106개 세포의 시오노기 암종을 다 자란 수컷 DD/S계 마우스에 주입하였다. 주입후 통상 2 내지 3주후에 시오노기 종양 직경이 1 내지 2 ㎝일 때 메톡시플루란 마취하에 복부 절개로 절제를 수행하였다. 마우스 유지, 종양 저장 및 수술과정의 상세함은 이미 기재되었다 (Bruchovsky et al., Cancer res. 50: 2275-2282 (1990); Rennie et al., Cancer Res. 48: 6309-6312 (1988); Bruchovsky et al., Cell 13: 272-280 (1978); Gleave et al., in Genitourinary Oncology, pp.367-378, Lange et al., eds, Lippencott (1997); Gleave et al., J. Urol. 157: 1727-1730 (1997); Bruchovsky et al., The Prostate 6: 13-21 (1996)).
마우스를 임의로 선별하여 뮤린 포스포로티오에이트 안티센스 TRPM-2 ODN ( 서열번호 1) 또는 안티센스 TRPM-2 ODN 서열과 두 개의 염기가 다른 비합치 대조군(서열번호 2)으로 처치하였다. 각 실험군은 7 마리의 마우스로 이루어졌다. 절제한지 1일 후에, 포스페이트 완충염수에 용해된 안티센스 TRPM-2 또는 비합치 대조군 ODN 12.5 mg/kg을 40일동안 각 마우스에 1일 1회 복강내 주사하였다. 종양 크기를 매주 2회 측정하고, 길이×폭×깊이×0.5236의 식으로 계산하였다. Gleave et al., Cancer Res. 52: 1598-1605 (1992). 데이터는 평균 종양크기±표준 편차로 기록하였다.
이 연구의 결과는 도 1에 도시되어 있다. 도시한 바와 같이, 안티센스 TRPM-2 ODN으로 처치한 마우스에서 시오노기 종양은 보다 신속하게 퇴행하고 완전한 퇴행은 조기에 발생하였다. 더욱이, 안티센스 TRPM-2 ODN으로의 처치는 대조군 동물에서 21일 후에 종양 크기의 증가로 반영되는 안드로겐 비의존성 징후를 실질적으로 지연시켰다. 안티센스 TRPM-2 또는 비합치 대조군과 관련된 부작용은 관찰되지 않았다.
생체내에서 TRPM-2 mRNA의 수준에 대한 ODN 처치 효과를 조사하기 위해, 노던 블럿 (Northern blot) 분석을 마우스로부터 시오노기 종양 조직상에서 수행하였다. 마우스를 절제한 지 1일 후에 복강내 주사로 안티센스 TRPM-2 ODN (n=6) 또는 비합치 대조군 (n=6) 12.5 mg/kg으로 매일 처치하였다. 절제한지 4일째에 종양 조직을 회수하고 TRPM-2 mRNA에 대해 노던 블럿으로 분석하였다. 안티센스 TRPM-2 ODN은 비합치 대조군 ODN 처치된 종양과 비교하여 시오노기 종양에서 TRPM-2 mRNA 수준을 75 % 감소시켰다 (도 3).
비교가능한 분석은 정상 마우스 기관상에서 수행하였다. 비장, 신장, 전립선 및 뇌 샘플을 동일한 처치 스케쥴하에서 안티센스 TRPM-2 ODN 및 비합치 대조군으로 처치한 시오노기 종양 마우스로부터 회수하고 노던 블럿으로 분석하였다. TRPM-2 mRNA 수준은 종양조직에서 현저하게 낮았지만, 안티센스 TRPM-2 ODN은 정상 기관에서 TRPM-2 mRNA 수준에 아무런 영향을 끼치지 않았다.
실시예 2
안티센스 TRPM-2 ODN (서열번호 1)의 서열 선택성은 안티센스 TRPM-2 ODN 또는 비합치 대조군 (서열번호 2)의 다양한 수준으로 처치한 후에 시험관내에서 유지된 시오노기 종양세포에서 TRPM-2 mRNA 의 발현수준을 비교함으로써 확인하였다. ODN의 세포로의 흡수를 촉진하기 위해, ODN을 양이온성 지질 담체 (Lipofectin™, (Life Technologies, Inc.))중에서 조제하였다. 세포를 다음 프로토콜을 사용하여 2일동안 2회 처치하였다. 세포를 혈청없는 OPTI-MEM™ (Life Technologies, Inc.)에서 리포펙틴 4㎍/ml로 20분간 예비배양한 다음에 ODN 및 리포펙틴의 선택된 농도를 함유하는 배지에서 4시간동안 배양하였다. 다음에 배지를 표준 배지로 교체하였다.
세포중의 TRPM-2 mRNA 양을 노던 블럿 분석을 사용하여 측정하였다. 도 4에 도시한 바와 같이, 시오노기 세포의 안티센스 TRPM-2 ODN으로의 처치는 투여량 의존방식으로 TRPM-2 mRNA 수준을 감소시켰다. 반대로, TRPM-2 mRNA 수준은 사용된 어느 농도에서도 비합치 ODN (서열번호 2)에 의해 영향을 받지 않았다. 따라서, 안 티센스 TRPM-2 ODN의 영향은 명백하게 서열 특이적이다.
실시예 3
시험관내에서 유지된 시오노기 세포를 다양한 양의 탁솔 단독 또는 500 nM 안티센스 TRPM-2 ODN (서열번호 1) 또는 비합치 대조군 (서열번호 2)과의 조합으로 처치하였다. 세포를 실시예 2에 기재한 대로 2회 처치하고 생존세포 잔존률을 측정하였다. 결과는 도 5에 요약되어 있다. 나타낸 바와 같이, 안티센스 TRPM-2 ODN의 포함은 투여량 반응곡선을 좌측으로 이동시키고, 인자 IC50을 5 내지 10으로 저하시켰다. 유사한 결과를 파클리탁셀 대신에 미톡산트론을 사용하여 얻었다 (도 12A 및 도 12B).
실시예 4
안티센스/비합치 TRPM-2 ODN 및 탁솔과 다양하게 조합하는 안티센스 Bcl-2 ODN (서열번호 13) 또는 비합치 Bcl-2 ODN (서열번호 14)을 첨가하여 실시예 3의 실험을 반복하였다. 결과는 도 6에 나타나 있다. 안티센스 TRPM-2 ODN과 안티센스 Bcl-2 ODN 및 탁솔과의 조합은 탁솔의 세포독성 효과를 더욱 증가시켰다. 따라서 추가의 안티-세포사멸제의 표적화는 치료적 이점을 제공하는 것으로 보인다.
실시예 5
인간 치료에 적당한 안티센스 TRPM-2 ODN 서열을 동정하기 위해, 인간 TRPM- 2 유전자중 10개의 상이한 부위에 대한 안티센스 ODN 서열(도 2, 서열번호 3-12)을 합성하고 인간 전립선암 PC-3에서의 TRPM-2 유전자 발현을 감소시키는 능력에 대해 시험하고, 실시예 2에 기재한 동일한 처치 프로토콜을 사용하여 TRPM-2를 과발현하는 LNCaP 세포를 트랜스펙션시켰다. 결과는 도 3에 요약되어 있다. 나타낸 바와 같이, 서열번호 4, 5 및 12는 TRPM-2 발현 감소에 활성이다. 이들 세 개의 서열은 번역 개시 또는 종결 부위에 바로 인접해 있거나 중복되어 있다.
실시예 6
면역조직화학적 염색은 본래 신장절제 표본으로부터 얻은 정상 신장 조직 및 17가지의 RCC에서 클러스터린 발현을 특정화하는데 사용하였다. 인간 신암 세포주 ACHN, CaKi-1 및 CaKi-2에서 TRPM-2 발현은 노던 및 웨스턴 블럿 분석으로 측정하였다. 노던 블럿 분석은 안티센스 TRPM-2 ODN 처치후에 TRPM-2 mRNA 발현에서의 변화를 평가하는데 사용하였다. 조합된 안티센스 TRPM-2 ODN 및 탁솔 처치의 CaKi-2 세포 성장에 대한 효과는 MTT 분석을 사용하여 조사하였다.
면역염색은 인접한 정상 신장조직과 비교하여 11가지의 RCC 표본에서 증가된 클러스터린 발현을 나타냈다. 잔존한 6가지 경우에, 악성 및 정상 조직간의 차이점은 없었다. TRPM-2 mRNA 및 단백질 발현 둘다는 모든 세가지 인간 RCC 세포주에서 검출가능하며, CaKi-2에 대해서 최고 수준이었다. 비합치 대조군 ODN (서열번호 2)이 아닌 안티센스 TRPM-2 ODN (서열번호 1)이 투여량 의존 및 서열 특이적 방식으로 CaKi-2 세포에서 TRPM-2 발현을 저해하였다 (도 7A). 더욱이, 안티센스 TRPM-2 ODN은 탁솔 약제감수성을 실질적으로 증가시키고, 탁솔의 IC50을 비합치 대조군 ODN과 비교하여 1 log (500 nM 내지 50 nM) 만큼 감소시켰다 (도 7B). 이들 데이터는 TRPM-2 및 그 단백질, 클러스터린이 정상 신장 조직보다 RCC에서 더 높은 수준으로 발현되고, 안티센스 TRPM-2 ODN은 진행된 RCC에서 종래의 화학요법의 세포독성 효과를 증가시키는데 유효하다는 것을 입증한다.
실시예 7
안티센스 TRPM-2 ODN은 TRPM-2를 발현하는 암 세포의 방사선 감도를 증강시킨다. 노던 분석을 사용하여, 본 발명자는 방사선 치료가 인간 전립선 암 PC-3 세포에서 TRPM-2 유전자 발현을 투여량 및 시간 의존적으로 증가시킨다는 것을 알아내었다 (도 8). TRPM-2의 과발현은 방사선 유발된 세포사멸에 대한 저항을 증가시켰다. TRPM-2를 과발현하는 인간 전립선 LNCaP 세포 (LNCaP/T1)는 방사선 치료에 더 저항성이 있다 (도 9A 및 도 9B). 인간 전립선 암 PC-3 세포의 100 및 500 nM 안티센스 TRPM-2 ODN (서열번호 1)으로의 처치는 비합치 ODN (서열번호 2) 처치와 비교하여 4 Gy 방사선 치료의 단일 치료후에 세포 생존을 현저하게 감소시킨다 (도 10). 도 11A 및 도 11B는 시험관내에서 10, 50 및 100 nM 안티센스 TRPM-2 올리고로 처치한 후에 인간 전립선 암 PC-3 세포의 투여량 의존성 방사선 감도를 나타낸다.
실시예 8
인간 안티센스 TRPM-2 ODN으로의 처치가 PC-3 인간 전립선 암 세포주에서 약제감수성을 증가시키는지를 측정하기 위해, PC-3 종양이 있는 마우스를 (안티센스 인간 TRPM-2 ODN) + (미셀 파클리탁셀(micellar paclitaxel) 또는 미톡산트론(mitoxantrone)) 및 (비합치 대조군(mismatch control) ODN) + (미셀 파클리탁셀 또는 미톡산트론)으로 처치하였다 (도 12A 및 도 12B). ODN을 28일동안 투여하고 0.5 mg 미셀 탁솔 또는 0.3 mg 미톡산트론을 두가지 경우, 즉 10일 내지 14일 및 24일 내지 28일에 투여하였다. 종양 크기의 현저한 감소는 비합치 대조군 ODN으로 처치한 바와 비교하여 안티센스 ODN 처치한 동물에서 관찰되었다. 이 효과는 미셀 파클리탁셀 또는 미톡산트론의 두 번째 투여후에 보다 더욱 분명하게 나타났다.
