Claims (1)
- 【特許請求の範囲】 1. 寄託番号VKM F-3500Dを有するクリソスポリウム・ラクノウェ ンス・ガーグ27Kの分離培養物。 2. 中性及び/又はアルカリ性セルラーゼ活性を有する組成物であ って、適正な培地中で培養されたクリソスポリウム属の野生型又は変異体の菌類 を成長させることを含む方法によって得られ、前記菌類は、クリソスポリウム・ ラクノウェンス、クリソスポリウム・パノラム、クリソスポリウム・ケラチノフ ィラム、クリソスポリウム・ロバタム、クリソスポリウム・マーダリウム、クリ ソスポリウム・クイーンズランディカム又はクリソスポリウム・トロピカムであ る組成物。 3. 前記菌類はクリソスポリウム・ラクノウェンスである請求項2 記載の組生物。 4. 中性及び/又はアルカリ性セルラーゼ活性を有し、適正な培地 中で培養されたクリソスポリウム属の変異体の菌類を成長させることを含む方法 によって得られる組成物。 5. 前記菌類は、寄託番号VKM F-3500Dのクリソスポリウム・ラク ノウェンス・ガーグ27Kである請求項2記載の組成物。 6. 前記菌類は、クリソスポリウム・ラクノウェンス・ガーグ27K の変異体株である請求項2記載の組成物。 7. 約40℃乃至約60℃の温度、約5.0乃至約12.0のペーハーで最適 セルラーゼ活性を有する請求項2乃至6のうちいずれか一項記載の組成物。 8. 約6.0乃至約7.0のペーハー、約40℃乃至約60℃の温度で最適セ ルラーゼ活性の少なくとも50%を有する請求項2乃至6のうちいずれか一項記載 の組成物。 9. 前記セルラーゼ活性は、カルボキシメチルセルラーゼ活性測定 法、RBBカルボキシメチルセルラーゼ活性測定法、細胞内粘度測定法又は濾紙活 性測定法によって測定される請求項2乃至6のうちいずれか一項記載の組成物。 10. 実質的に精製及び分離されたタンパク画分であって、請求項2 乃至4記載の組成物から得られ、カルボキシメチルセルラーゼ活性測定法、RBB カルボキシメチルセルラーゼ活性測定法、細胞内粘度測定法又は濾紙活性測定法 によって測定されると最適セルラーゼ活性の少なくとも50%を約6.0乃至約7.0の ペーハーで有するタンパク画分。 11. 請求項10記載のタンパク画分から得られ、分子量約25kD、pI約 4を有するエンドグルカナーゼ。 12. 請求項10記載のタンパタ画分から得られ、分子量約70kD、pI約 4を有するエンドグルカナーゼ。 13. 請求項10記載のタンバタ画分から得られ、分子量約60kD、pI約 3を有するエンドグルカナーゼ。 14. 請求項10記載のタンパク画分から得られ、分子量約43kD、pI約 3を有するエンドグルカナーゼ。 15. 請求項10記載のタンパク画分から得られ、分子量約60kD、pI約 4を有するセロビオヒドロラーゼ。 16. 実質的に精製及び分離された中性及び/又はアルカリ性セルラ ーゼ酵素であって、請求項10記載のタンパク画分から分離され、pI約3乃至約5. 5を有するセルラーゼ酵素。 17. 前記セルラーゼはエンドグルカナーゼ又はセロビオヒドロラー ゼ活性のいずれかを有する請求項16記載のセルラーゼ。 18. 最大セルラーゼ活性の少なくとも50%を約6.0乃至約7.0のペー ハーで有する請求項16記載のセルラーゼ。 19. 請求項10記載の画分から得られ、分子量約25kDを有するエンド グルカナーゼ。 20. 請求項10記載の画分から得られ、分子量約70kDを有するエンド グルカナーゼ。 21. 請求項10記載の画分から得られ、分子量約43kDを有するエンド グルカナーゼ。 22. 請求項10記載のタンパク画分から分離された一以上の精製酵素 を有し、界面活性剤を更に有する洗剤組成物。 23. 請求項10記載の前記タンパク画分から得られた一以上の精製酵 素を有する生地柔軟剤組成物。 24. セルロース系繊維、セルロース系生地その他のセルロース系基 質の酵素処理に供される組成物であって、クリソスポリウム属の野生型又は変異 体の菌類の核酸配列によってアミノ酸配列がコード化されているセルラーゼを有 し、ペーハー約8.0乃至約12.0を有する組成物。 25. 前記菌類は、クリソスポリウム・ラクノウェンス、クリソスポ リウム・パノラム、クリソスポリウム・プルイノサム、クリソスポリウム・ケラ チノフィラム、クリソスポリウム・ロバタム、クリソスポリウム・マーダリウム 、クリソスポリウム・クイーンズランディカム及びクリソスポリウム・トロピカ ムからなるグループから選択される請求項24記載の組成物。 26. 前記菌類はクリソスポリウム・ラクノウェンスである請求項25 記載の組成物。 27. 前記セルラーゼは、クリソスポリウム属の野生型又は変異体の 菌類から分離又は得られる請求項24乃至26のうちいずれか一項記載の組成物。 28. 軽石、研磨剤、柔軟剤、溶媒、保存料、漂白剤、青み剤、蛍光 染料、酸 化防止剤、可溶化剤、洗剤、界面活性剤、酵素、ビルダー、再汚染防止剤、緩衝 剤、ケーキング防止剤、セルラーゼ活性阻害因子用のマスキング剤及びセルラー ゼ活性化剤からなるグループから選択された一以上の成分を更に有する請求項24 セルロース系繊維、セルロース系生地その他のセルロース系基 質の酵素処理に供される組成物であって、細胞内粘度測定法により測定するとエ ンド-1,4-β-グルカナーゼ活性がセルラーゼの乾燥組成物1g当たり少なくとも 124単位あり、前記セルラーゼのアミノ酸配列はクリソスポリウム属の野生型又 は変異体の菌類の核酸配列によってコード化されている組成物。 41. 記セルラーゼのアミノ酸配列はクリソスポリウム属の野生型又は変異体の菌類の 核酸配列によってコード化されている組成物。 53. 前記菌類は、クリソスポリウム・ラクノウェンス、クリソスポ リウム・パノラム、クリソスポリウム・プルイノサム、クリソスポリウム・ケラ チノフィラム、クリソスポリウム・ロバタム、クリソスポリウム・マーダリウム ンド-1,4-β-グルカナーゼ活性が、セルラーゼの乾燥組成物1g当たり少なく とも964単位あり、前記セルラーゼはクリソスポリウム属の野生型又は変異体の 菌類から分離又は得られる組成物。 56. 前記菌類は、クリソスポリウム・ラタノウェンス、クリソスポ リウム・パノラム、クリソスポリウム・プルイノサム、クリソスポリウム・ケラ チノフィラム、クリソスポリウム・ロバタム、クリソスポリウム・マーダリウム 前記菌類は、クリソスポリウム・ラクノウェンス、クリソスポ リウム・パノラム、クリソスポリウム・プルイノサム、クリソスポリウム・ケラ チノフィラム、クリソスポリウム・ロバタム、クリソスポリウム・マーダリウム 、クリソスポリウム・クイーンズランディカム、クリソスポリウム・トロピカム からなるグループから選択される請求項58記載の組成物。 60. 前記菌類はクリソスポリウム・ラクノウェンスである請求項56 記載の組成物。 61. クリソスポリウム属の野生型又は変異体の菌類から分離又は得 られるセルラーゼを有し、一以上の界面活性剤を更に有する洗濯用洗剤組成物。 62. 前記菌類は、クリソスポリウム・ラクノウェンス、クリソスポ リウム・パノラム、クリソスポリウム・プルイノサム、クリソスポリウム・ケラ チノフィラム、クリソスポリウム・ロバタム、クリソスポリウム・マーダリウム 、クリソスポリウム・クイーンズランディカム、クリソスポリウム・トロピカム からなるグループから選択される請求項61記載の組成物。 63. 前記菌類はクリソスポリウム・ラクノウェンスである請求項62 記載の組成物。 64. 中性及び/又はアルカリ性ペーハーでセルラーゼ活性を有し、 クリソスポリウム・ラクノウェンスの変異体又は野生型の菌類から得られるセル ラーゼ組成物。 65. 中性及び/又はアルカリ性ペーハーでセルラーゼ活性を有し、 寄託番号VKM F-3500Dを有するクリソスポリウム・ラクノウェンス・ガーグ27Kの 変異体又は野生型の菌類から得られるセルラーゼ組成物。 66. 中性及び/又はアルカリ性ペーハーでセルラーゼ活性を有し、 クリソスポリウム属の変異体又は野生型の菌類から得られ、前記菌はクリソスポ リウム・プルイノサム、クリソスポリウム・ファーガシー又はクリソスポリウム ・サーモフィルではないセルラーゼ組成物。 67. 中性及び/又はアルカリ性ペーハーでセルラーゼ活性を有する 組成物の製造方法であって、摘正な培地中でクリソスポリウム属の野生型又は変 異体の菌類を成長させることを有する方法。 68. リソスポリウム属の野生型又は変異体の菌類の核酸配列によってアミノ酸配列が コード化されたセルラーゼにより前記デニム生地又はデニム地ジーンズを処理す ることを有する方法。 74. 織物をバイオ磨き、デフリビレート、漂白、染色又は糊抜きす る方法であって、クリソスポリウム属の野生型又は変異体の菌類の核酸配列によ ってアミノ酸配列がコード化されたセルラーゼにより前記織物を処理することを 有する方法。 75. 紙又はパルプのインク除去又はバイオ漂白を行う方法であって 、クリソスポリウム属の野生型又は変異体の菌類の核酸配列によってアミノ酸配 列がコード化されたセルラーゼにより前記紙又はパルプを処理することを有する 方法。 76. セルロース又は混綿生地の柔軟性又は感触を強める方法であっ て、クリ ソスポリウム属の野生型又は変異体の菌類の核酸配列によってアミノ酸配列がコ ード化されたセルラーゼにより前記生地を処理することを有する方法。 77. 前記セルラーゼはクリソスポリウム属の野生型又は変異体の菌 類から分離又は得られる請求項73乃至76のうちいずれか一項記載の方法。 78. 前記菌類はクリソスポリウム・ラクノウェンスである請求項73 乃至76のうちいずれか一項記載の方法。 79. 前記菌類はクリソスポリウム・ラクノウェンスである請求項77 記載の方法。 80. 中性及び/又はアルカリ性ペーハーで高められたセルラーゼ活 性を作るクリソスポリウム属の変異体株を生成する方法であって、 (a) クリソスポリウム属の菌類の胞子を変異させ、 (b) 工程(a)で得られた前記胞子を培養し、 (c) 中性及び/又はアルカリ性セルラーゼ活性の高められたレベルのために 工程(b)で得られた前記培養物をスクリーニングする方法。 81. 前記変異工程は、前記胞子に紫外線照射又は化学的変異原を行 うことを有する請求項80記載の方法。 82. 前記化学的変異原は、亜硝酸、N-メチル-N'-ニトロ-N-ニトロ ソグアニジン、または4-ニトロキノロン-N-オキシドである請求項81記載の方法 。 83. 請求項80乃至82のうちいずれか一項記載の方法により得られた クリソスポリウム属の変異体株。 84. クリソスポリウムのセルラーゼ酵素をコード化する遺伝子を分 離する方法であって、 (a)野生型又は変異体のクリソスポリウムによって作られた中性及び/又はア ルカリ性セルラーゼ組成物からタンパク質を分離し、 (b) 工程(a)で分離されたタンパク質の全部又は一部を配列し、 (c) 工程(b)の前記配列に由来する核酸プローブを生成し、 (d) 工程(c)の前記核酸プローブにより野生型又は変異体のクリソスポリウム ライブラリをスクリーニングし、 (e) 前記プローブによって認懺された核酸配列を分離し、 (f) 工程(e)で分離された核酸配列を配列することを有する方法。 85. 請求項84記載の方法によって得られる核酸配列。 86. 配列がセルラーゼ酵素をコード化する分離核酸であって、前記 セルラーゼ酵素はクリソスポリウム属の野生型又は変異体の菌類から分離又は得 られる分離核酸。 87. 配列が請求項11乃至21のうちいずれか一項のセルラーゼをコー ド化する分離核酸。 88. 請求項85乃至87のうちいずれか一項記載の核酸配列を有する組 換え発現ベタター。 89. 請求項88記載の組換え発現ベタターを含む宿主細胞。 90. 酵母細胞、真菌類細胞、植物細胞及び細菌類細胞からなるグル ープから選択される請求項89記載の宿主細胞。 91. 宿主細胞は、トリコダーマ、アスペルギルス、ヒュミコラ、ベ ニシリウム、クリソスポリウム及びノイロスポラからなるグループから選択され る真菌類細胞である請求項90記載の宿主細胞。 92. クリソスポリウム属の菌類を培養する方法であって、前記菌類 は、ペーハー約5乃至8で、無機塩類、炭素源及び有機窒素源を有する培養液で 成長される方法。 93. ペーハーが約6.5乃至7.5である請求項92記載のクリソスポリウ ム属の菌類を培養する方法。 94. ペーハーが約6.9乃至7.1である請求項92記載のクリソスポリウ ム属の菌類を培養する方法。
