Claims (26)
1. Kimerni protein naznačen time što sadrži
(i) prvi polipeptid, koji sadrži
(a) protein faktora VIII („FVIII“),
(b) XTEN prvog polipeptida, i
(c) prvu Fc regiju,
pri čemu FVIII protein ima aktivnost FVIII; i
(ii) drugi polipeptid, koji sadrži
(a) fragment von Willebrandovog faktora („VWF“) koji sadrži sekvencu aminokiseline koja je najmanje 99% identična sekvenci aminokiseline navedenoj u aminokiselinama 764 do 1240 iz SEQ ID NO: 21,
(b) XTEN drugog polipeptida,
(c) poveznicu od 10-50 aminokiselina koja se može cijepati koja sadrži a2 regiju FVIII koja sadrži sekvencu aminokiseline od Glu720 do Arg740 od FVIII koja odgovara SEQ ID NO: 65, pri čemu a2 regija može biti cijepana trombinom, i
(d) drugu Fc regiju,
pri čemu se VWF fragment veže na FVIII protein;
pri čemu XTEN drugog polipeptida sadrži sekvencu aminokiseline kako je navedena u SEQ ID NO: 58;
pri čemu XTEN prvog polipeptida sadrži sekvencu aminokiseline kako je navedena u SEQ ID NO: 8;
pri čemu je prva Fc regija povezana s drugom Fc regijom disulfidnom vezom; i
pri čemu je poluživot kimernog proteina produljen u usporedbi s FVIII proteinom bez VWF fragmenta.
2. Kimerni protein naznačen time što sadrži
(i) prvi polipeptid, koji sadrži
(a) protein faktora VIII („FVIII“),
(b) XTEN prvog polipeptida, i
(c) prvu Fc regiju,
pri čemu FVIII protein ima aktivnost FVIII; i
(ii) drugi polipeptid, koji sadrži
(a) fragment von Willebrandovog faktora („VWF“) koji sadrži sekvencu aminokiseline koja je najmanje 99% identična sekvenci aminokiseline navedenoj u aminokiselinama 764 do 1240 iz SEQ ID NO: 21,
(b) XTEN drugog polipeptida,
(c) poveznicu od 10-50 aminokiselina koja se može cijepati koja sadrži a2 regiju FVIII koja sadrži sekvencu aminokiseline od Glu720 do Arg740 od FVIII koja odgovara SEQ ID NO: 65, pri čemu a2 regija može biti cijepana trombinom, i

pri čemu XTEN drugog polipeptida sadrži sekvencu aminokiseline kako je navedena u SEQ ID NO: 58;
pri čemu XTEN prvog polipeptida sadrži sekvencu aminokiseline kako je navedena u SEQ ID NO: 8;
pri čemu je prva Fc regija povezana s drugom Fc regijom disulfidnom vezom; i
pri čemu je poluživot kimernog proteina produljen u usporedbi s FVIII proteinom bez VWF fragmenta.
3. Kimerni protein prema patentnom zahtjevu 1 ili 2, naznačen time što je FVIII protein dvolančana FVIII izoforma.
4. Kimerni protein prema patentnom zahtjevu 3, naznačen time što dvolančana FVIII izoforma sadrži prvi lanac koji sadrži teški lanac FVIII i drugi lanac koji sadrži laki lanac FVIII, pri čemu su teški lanac i laki lanac povezani jedan s drugim pomoću metalne veze.
5. Kimerni protein prema patentnom zahtjevu 1 ili 2, naznačen time što je FVIII protein jednolančana FVIII izoforma.
6. Kimerni protein prema bilo kojem od patentnih zahtjeva 1, 2, ili 5, naznačen time što FVIII protein ima sekvencu aminokiseline koja je najmanje 99% identična aminokiselinama 1 do 1438 iz SEQ ID NO: 67, te pri čemu FVIII protein ima aktivnost zgrušavanja.
7. Kimerni protein prema patentnom zahtjevu 6, naznačen time što je XTEN prvog polipeptida umetnut neposredno nizvodno od ostatka koji odgovara ostatku 745 iz SEQ ID NO: 67.
8. Kimerni protein prema bilo kojem od patentnih zahtjeva 1-7, naznačen time što drugi polipeptid sadrži D' domenu i D3 domenu VWF kondenzirane na drugu Fc regiju pomoću XTEN drugog polipeptida između njih,
pri čemu XTEN sekvenca sadrži manje od 288 aminokiselinskih ostataka, i
pri čemu je XTEN drugog polipeptida kondenziran na drugu Fc regiju poveznicom koja se može cijepati.
9. Kimerni protein prema patentnom zahtjevu 2, naznačen time što VWF fragment sadrži alaninski ostatak supstituiran za ostatke koji odgovaraju ostacima 1099 i 1142 iz SEQ ID NO: 21.
10. Kimerni protein prema bilo kojem od patentnih zahtjeva 1-9, naznačen time što su prva Fc regija i druga Fc regija identične.
11. Kimerni protein prema patentnom zahtjevu 10, naznačen time što su prva Fc regija i druga Fc regija izvedene iz ljudskog IgG1.
12. Kimerni protein prema patentnom zahtjevu 8, naznačen time što VWF fragment nadalje sadrži D1 domenu, D2 domenu ili D1 i D2 domene VWF.
13. Kimerni protein prema patentnom zahtjevu 12, naznačen time što:
(i) prvi polipeptid sadrži sekvencu aminokiseline najmanje 95% identičnu sekvenci aminokiseline SEQ ID NO: 70; i
(ii) drugi polipeptid sadrži sekvencu aminokiseline najmanje 95% identičnu sekvenci aminokiseline SEQ ID NO: 197.
14. Kimerni protein prema patentnom zahtjevu 13, naznačen time što:
(i) prvi polipeptid sadrži sekvencu aminokiseline najmanje 99% identičnu sekvenci aminokiseline SEQ ID NO: 70; i
(ii) drugi polipeptid sadrži sekvencu aminokiseline najmanje 99% identičnu sekvenci aminokiseline SEQ ID NO: 197.
15. Kimerni protein prema bilo kojem od zahtjeva 1-14, naznačen time što se kimerni protein može dobiti postupkom izrade kimernog proteina koji uključuje
(a) transfektiranje stanice domaćina koja sadrži polinukleotid ili skup polinukleotida koji kodiraju kimerni protein, i
b) kultiviranje stanice domaćina u mediju za kultiviranje pod uvjetima prikladnim za ekspresiju kimernog proteina, pri čemu se eksprimira kimerni protein,
pri čemu polinukleotid ili skup polinukleotida koji sadrži sekvencu nukleinske kiseline prema SEQ ID NO 172 kodira prvi polipeptid.
16. Kimerni protein prema bilo kojem od zahtjeva 1-15, naznačen time što prvi polipeptid sadrži sekvencu aminokiseline iz SEQ ID NO: 173.
