Claims (8)
1. Förfarande för effektivering av t-PA-artade proteiners produktion ur kulturer av transformerade CHO-celler, k ä n -netecknat därav, att tili kulturen sätter i en kon-centration av 0,005 - 500 mM tioglykolsyra, tiodiglykolsyra, L-cystein, glutation, butyrylkolinklorid, butylkolinbromid, nonaktinsyra, furanfettsyra, askorbinsyra, afidikolin, 6-hydroxi-4,6-dimetyl-3-hepten-2-on, D-oi-hydroxisubstituerad (C3- eller C4-) alifatisk mono- eller dikarboxylsyra, C3-C4-alifatisk monokarboxylsyra eller deras salter.
2. Förfarande enligt patentkravet 1, kännetecknat därav, att proteinet är t-Pa eller dess mutant.
3. Förfarande enligt patentkravet 1 eller 2, kännetecknat därav, att som protein inducerande ämne används smörsyra eller dess sait.
4. Förfarande enligt nägot av patentkraven 1-3, kännetecknat därav, att kultiveringen utförs i ett serumhal-tigt medium.
5. Förfarande enligt nägot av patentkraven 1-4, kännetecknat därav, att den C3-C4-alifatiska monokarboxyl- *. syran eller dess sait tillsätts i en koncentration av 1 mM - 10 mM.
6. Förfarande enligt nägot av patentkraven 1-5, kännetecknat därav, att cellerna kultiveras i närvaro av nägot av följande ämnen: tioglykolsyra, tiodiglykolsyra, » L-cystein, glutation, butylkolinbromid, butyrylkolinklorid, nonaktinsyra, furanfettsyra, askorbinsyra, afidikolin, 6-hydroxi-4,6-dimetyl-3-hepten-2-on, D-a-hydroxisubstituerad (C3- eller C4-) alifatisk mono- eller dikarboxylsyra eller deras sait, cellerna separeras ur kulturen och därefter sätts 41 94963 till ett nytt kultiveringsmedium och kultiveras i närvaro av nägon C3- eller C4-alifatisk monokarboxylsyra eller dess sait.
7. Förfarande enligt patentkravet 6, kännetecknat därav, att afidkolin är det förstnämnda ämnet och smörsyra, propionsyra eller en C3- eller C4-alifatisk monokarboxylsyra eller dess sait är det andra ämnet.
8. Förfarande enligt patentkravet 6, kännetecknat därav, att smörsyra eller tioglykolsyra eller deras sait an-vänds. - i • ·
