Abstract

Ensayos diagnósticos

25 La expresión de un marcador de células madre del cáncer se puede determinar por la detección del polipéptido correspondiente. El polipéptido correspondiente se puede detectar y cuantificar por cualquiera de varios medios bien conocidos por los expertos en la técnica. En algunas realizaciones, un polipéptido marcador de células madre del cáncer se detecta utilizando métodos analíticos bioquímicos tales como por ejemplo, electroforesis, electroforesis capilar, cromatografía líquida de altas prestaciones (HPLC) o cromatografía de capa fina (TLC). El polipéptido aislado se puede también secuenciar de acuerdo con técnicas convencionales. DE manera alternativa, los anticuerpos contra el marcador de células madre del cáncer se puede detectar su expresión utilizando, por ejemplo, ELISA, FACS, transferencia de Western, inmunoprecipitación o micromatrices proteicas. Por ejemplo, las células madre del cáncer
35 se pueden aislar de una biopsia de un paciente y se puede detectar la expresión de un marcador de célula madre cancerosa con anticuerpos marcados con fluorescencia utilizando FACS. En otro método, las células que expresan un marcador de célula madre cancerosa se puede detectar in vivo utilizando anticuerpos marcados en un sistema de creación de imágenes típico. Por ejemplo, los anticuerpos marcados con isótopos paramagnéticos se pueden utilizar para creación de imágenes por resonancia magnética (MRI).
Un ensayo diagnóstico comprende la determinación de si se expresa o no un marcador de células madre del cáncer en las células tumorales utilizando, por ejemplo, inmunohistoquímica, hibridación in situ, o RTPCR. En general, el polímero soluble en agua no se seleccionará de estos glucosilo de origen natural aunque estos se producen habitualmente más convenientemente por los sistemas de expresión recombinante en mamíferos. El polímero puede ser de cualquier peso molecular, y puede ser ramificado o no ramificado. Un polímero soluble en agua para su uso en el presente documento es el polietilenglicol. Como se utiliza en el presente documento, polietilenglicol significa que se engloba cualquiera de las formas de PEG que se haya utilizado para derivar otras proteínas, tal como mono (C1C10) alcoxi o ariloxipolietilenglicol.

Los polipéptidos aislados descritos en el presente documento se pueden producir por cualquier método adecuado que se conocen en la técnica. Por ejemplo, se puede obtener el ADNc explorando una biblioteca de ADNc humanos con un fragmento de ADN marcado que codifica el polipéptido de la SEQ ID NO: 1 e identificando los clones positivos por autorradiografía. Se realizan más rondas de purificación e hibridación en placas utilizando los métodos convencionales.
