Abstract

Los macrófagos se colocaron en placas a células 5 x 104 por pocillo en una placa de cultivo de tejidos de 24 pocillos (Falcon). Después de 24 horas, los medios se sustituyeron con IMDM libre de suero. EJEMPLO 3
Las células madre hematopoyéticas y células progenitoras CD47 regulan hacia arriba para facilitar la movilización y toma de referencia para los tejidos hematopoyéticos
[0138] Hemos demostrado que las células madre hematopoyéticas (HSC) de ratones CD47 deficientes (IAP -/-) no logran injertar los receptores de tipo silvestre. Como era de esperar, estas células se eliminan rápidamente por los macrófagos huésped, mientras que IAP +/+ HSCs no lo son. Cuando las células madre y progenitoras se ven obligadas a dividir y entrar en circulación utilizando ciclofosfamida/G-CSF o lipopolisacárido, CD47 se regula hacia arriba rápidamente en estas células. Proponemos que los niveles más altos de CD47 en las células madre durante el estrés hematopoyesis y movilización proporciona protección frente a la fagocitosis por los macrófagos activados añadidos del sistema reticuloendotelial. [0184] Inicialmente se identificó una mayor expresión de CD47 en AML LSC, pero se observó que la expresión de explosiones a granel era la misma. Debido a esto, decidimos utilizar los datos de expresión génica publicados sobre AML mayor para investigar la relación entre la expresión de CD47 y los resultados clínicos. De acuerdo con nuestra hipótesis, se encontró que el aumento de expresión CD47 era independientemente predictivo de un resultado clínico peor en los pacientes con AML con un cariotipo normal, incluyendo el subconjunto sin la mutación FLT3-ITD, que es el mayor subgrupo de pacientes con AML. Al ser este análisis dependiente de la expresión relativa de CD47 ARNm, un ensayo de PCR cuantitativo para AML pronóstico se puede basar en el nivel de expresión CD47. Dicho ensayo podría ser utilizado en el riesgo adaptado de toma de decisiones terapéuticas, particularmente en el gran subgrupo de pacientes con AML con cariotipos normales que carecen de la mutación FLT3-ITD.
[0185] Orientación de CD47 en AML LSC con moléculas de superficie celular de anticuerpos terapéuticos monoclonales expresados preferentemente en AML LSC en comparación con sus homólogos normales son candidatos para la orientación con anticuerpos monoclonales terapéuticos. Hasta el momento, varias moléculas han sido objetivo de AML incluyendo CD33, CD44, CD123, CD47 y ahora. [0186] Varias líneas de evidencia sugieren que la orientación de CD47 con un anticuerpo monoclonal probable actúa mediante la interrupción de la interacción CD47-SIRPα, evitando de este modo una señal inhibidora fagocítica.
