Abstract
Die Erfindung betrifft ein Verfahren und ein Reagenzsystem zum Nachweis eines Analyten in einer Probe durch eine enzymatische Reaktion, umfassend die Verwendung eines Enzym-Coenzym-Komplexes als stöchiometrischem Reaktionspartner für den in der Probe vorhandenen Analyten.

Verfahren und Reagenzsystem mit nicht-regenerierbarem Enzym-Coenzym- Komplex
Beschreibung
Die Erfindung betrifft ein Verfahren und ein Reagenzsystem zum Nachweis eines Analyten in einer Probe durch eine enzymatische Reaktion, umfassend die Verwendung eines Enzym-Coenzym-Komplexes als nicht- regenerierbarem, insbesondere stöchiometrischem Reaktionspartner für den in der Probe vorhandenen Analyten.
Der Nachweis von Analyten, beispielsweise Glucose in Blut, durch enzymatische Methoden ist bekannt. Dabei wird der zu bestimmende Analyt mit einem geeigneten Enzym und einem Coenzym in Kontakt gebracht, wobei das Enzym in katalytischen Ivlengen eingesetzt wird. Die bei Reduktion bzw. Oxidation des Coenzyms entstehenden Redoxäquivalente werden auf Mediatoren übertragen, die dann in einem weiteren Schritt elektrochemisch oder photometrisch erfasst werden. Eine Kalibrierung liefert einen direkten Zusammenhang des Messwerts mit der Konzentration des zu bestimmenden Analyten.
Sierra et al. (Anal. Chem. 69 (1997), 1471 -1476) beschreiben eine Bestimmung von Blutglucose basierend auf der intrinsischen Fluoreszenz von Glucose-Oxidase. Auch bei diesen Verfahren wird das Enzym zusammen mit seinem Coenzym FAD in katalytischen Mengen eingesetzt, wobei Redoxäquivalente auf Sauerstoff als Mediator übertragen werden.
Narayanaswamy et al. (Analytical Letters 21 (7) (1988), 1 165-1 175) beschreiben eine Fluoreszenzmessung mit Glucosedehydrogenase und NAD zur Glucosebestimmung. Das Enzym wird dabei in katalytischen, d.h. nicht-
stöchiometrischen, Mengen eingesetzt. Die Fluoreszenzmessung erfasst das freie NADH in der Lösung.
Durch die für die Nachweissysteme des Standes der Technik benötigten elektrochemisch aktiven Substanzen (Mediatoren) können die zu bestimmenden Analyten nur indirekt, d.h. über mehrere chemische Reaktionen, nachgewiesen werden. Hierzu ist oftmals eine komplizierte Anpassung der Konzentrationen der beteiligten Substanzen zur Optimierung der Reaktionsgeschwindigkeit notwendig. Weiterhin besteht die Gefahr, dass die benötigten elektrochemisch aktiven Substanzen bei längerer Lagerung instabil sind.
Die Mediatoren müssen oftmals auch in großem Überschuss gegenüber dem Enzym-Coenzym-System eingesetzt werden. Das Coenzym hat eine hohe Reaktivität, so dass die Enzymaktivität bei Zerfall des Mediators selbst in geringen Mengen, z.B. < 1 % oder bei Einwirkung von Fremdsubstanzen, z. B. Ausdampfen der Substanzen aus Verpackungsmaterialien, deutlich abnimmt. Dies zu kann zu falschen Signalen bei der Analytbestimmung führen. Noch ein weiterer Nachteil besteht darin, dass die Bestimmungszeiten für den Analyten üblicherweise im Bereich von mindestens einigen Sekunden, bei Glucose z.B. im Bereich von > 4 s betragen, und die benötigten Probenvolumina groß, z.B. > 0,5 μl sind.
Die der vorliegenden Erfindung zugrunde liegende Aufgabe bestand darin, die geschilderten Nachteile des Standes der Technik mindestens teilweise zu vermeiden. Insbesondere sollte ein unempfindliches und schnelles Verfahren zum enzymatischen Nachweis von Analyten bereitgestellt werden, welches auch in Abwesenheit von Mediatoren oder/und Indikatoren zu zuverlässigen Messergebnissen führt.
Diese Aufgabe wird dadurch gelöst, dass ein Enzym-Coenzym-Komplex als stöchiometrischer Reaktionspartner, anstatt wie üblicherweise als Katalysator, verwendet wird. Der Nachweis des Analyten erfordert nur einen einzigen Reaktionsschritt und ist daher extrem schnell. Die Verwendung von Mediatoren und Indikatoren, verbunden mit dem Einsatz komplexer Reagenzgemische, mit geringer Stabilität und hoher Störungsanfälligkeit, ist nicht mehr erforderlich.
Ein Gegenstand der Erfindung ist somit ein Verfahren zum Nachweis eines Analyten in einer Probe durch eine enzymatische Reaktion, umfassend die Schritte:
(a) Inkontaktbringen der Probe mit einem Nachweisreagenz, umfassend einen Enzym-Coenzym-Komplex, wobei keine Regeneration des Coenzγms erfolgt, und (b) Nachweisen einer Reaktion des Analyten durch Veränderung des Enzym-Coenzym-Komplexes.
Ein weiterer Gegenstand der Erfindung ist ein Reagenzsystem zum Nachweis eines Analyten in einer Probe, umfassend: (a) ein Nachweisreagenz, umfassend einen Enzym-Coenzym-Komplex, wobei keine Regeneration des Coenzyms erfolgt, und
(b) einen Träger zur Aufnahme des Nachweisreagenz.
Die vorliegende Erfindung ermöglicht eine einfache qualitative oder quantitative Bestimmung von Analyten innerhalb einer sehr kurzen Reaktionsdauer von vorzugsweise ≤ 5 s, besonders bevorzugt ≤ 1 s, am meisten bevorzugt <0,1 s . Die Reaktion wird unter Bedingungen durchgeführt, bei denen keine Regeneration des Coenzyms während der Bestimmung erfolgt. Dabei kann ein Molekülenzym-Coenzym-Komplex nur mit einem einzigen Molekül des Analyten reagieren. Zweckmäßigerweise wird die Reaktion daher in Abwesenheit von Mediatoren oder anderen
Substanzen durchgeführt, die eine Regeneration des Coenzyms bewirken können.
Das Nachweisreagenz enthält den Enzym-Coenzym-Komplex in einer ausreichenden Menge, um entsprechend dem gewünschten Testformat eine qualitative oder/und quantitative Bestimmung des Analyten zu ermöglichen. Insbesondere für eine quantitative Bestimmung des Analyten wird der Enzym-Coenzym-Komplex in einer Menge eingesetzt, so dass die Anzahl der reagierenden Moleküle des Enzym-Coenzym-Komplexes mit der in der Probe vorhandenen Analytkonzentration korreliert. Besonders bevorzugt setzt man den Enzym-Coenzym-Komplex in einer mindestens stöchiometrischen Menge bezüglich des in der Probe vorhandenen Analyten, vorzugsweise in einem stöchiometrischen Überschuss bezüglich des Analyten, ein. Dabei bedeutet die Angabe "in mindestens einer stöchiometrischen Menge", dass die Größe der Probe bezüglich der Anzahl der Moleküle des Enzym-Coenzym-Komplexes derart abgestimmt wird, dass bei den in der Probe zu erwartenden Analytkonzentrationen die Anzahl der mit dem Analyten reagierenden Moleküle des Enzym-Coenzym-Komplexes mit der in der Probe vorhandenen Analytkonzentration korreliert. Bevorzugt bedeutet "stöchiometrische Menge", dass die Anzahl der Moleküle des Enzym-Coenzym-Komplexes der maximal zu erwartenden Anzahl der Analytmoleküle in der untersuchten Probe entspricht.
Das Verfahren und das Nachweissystem erlauben die Verwendung von kleinsten Probenmengen, beispielsweise Probenvolumina < 1 μ\, insbesondere ≤0,1 μ\. Gegebenenfalls kann die Probe vor dem Inkontaktbringen mit dem Nachweisreagenz noch verdünnt werden.
Das erfindungsgemäße Verfahren und Nachweissystem eignet sich zur Bestimmung beliebiger Analyten, beispielsweise Parametern in
Körperflüssigkeiten, wie etwa Blut, Serum, Plasma oder Urin, aber auch in
Abwasserproben oder Lebensmitteln. Das Verfahren kann sowohl als
Nasstest, z.B. in einer Küvette, oder als Trockentest auf einem entsprechenden Reagenzträger durchgeführt werden.
Als Analyten können beliebige biologische oder chemische Substanzen bestimmt werden, die mit einem Enzym-Coenzym-Komplex eine Reaktion, insbesondere eine Redoxreaktion eingehen können, wie etwa Glucose, Milchsäure, Äpfelsäure, Glycerin, Alkohol, Cholesterin, Triglyceride, Ascorbinsäure, Cystein, Glutathion, Peptide etc.
Die enzymatische Reaktion ist vorzugsweise eine Redoxreaktion, bei der eine Reduktion oder Oxidation des Coenzyms im Enzym-Coenzym-Komplex erfolgt. Für eine derartige Reaktion wird als Enzym vorzugsweise eine Oxidoreduktase verwendet. Besonders bevorzugt verwendet man als Enzym eine Dehydrogenase, beispielsweise ausgewählt aus einer Glucose- Dehydrogenase (E.C.1.1.1.47), Lactat-Dehydrogenase (E.C.1.1.1.27, 1.1 .1.28), Malat-Dehydrogenase (E.C.1.1.1 .37), Glycerin-Dehydrogenase (E.C.1.1 .1 .6), Alkohol-Dehydrogenase (E.C.1.1 .1.1 ) oder Aminosäure- Dehydrogenase, z.B. L-Aminosäure-Dehydrogenase (E.C.1 .4.1.5). Weitere geeignete Enzyme sind Oxidasen, wie etwa Glucoseoxidase (E.C.1.1 .3.4) oder Cholesterinoxidase (E.C.1 .1.3.6).
Coenzyme im Sinne der vorliegenden Erfindung sind vorzugsweise organische Moleküle, die kovalent oder nichtkovalent an ein Enzym gebunden sind und durch die Umsetzung des Analyten verändert, beispielsweise oxidiert oder reduziert werden. Bevorzugte Beispiele für Coenzyme sind Flavin-, Nicotin- und Chinonderivate, beispielsweise Flavinnukleosidderivate, wie etwa FAD, FADH2, FMN, FMNH2, etc., Nicotinnukleosidderivate wie etwa NAD + , NADH/H+, NADP+, NADPH/H + etc. oder Ubichinone, wie etwa Coenzym Q, PQQ etc.
Die Veränderung des Coenzyms durch Reaktion mit dem Analyten kann grundsätzlich auf beliebige Art und Weise nachgewiesen werden. Hier
können grundsätzlich alle aus dem Stand der Technik bekannten Methoden zum Nachweis enzymatischer Reaktionen eingesetzt werden. Vorzugsweise wird jedoch die Veränderung des Coenzyms durch optische Methoden nachgewiesen. Optische Nachweismethoden umfassen beispielsweise die Messung von Absorption, Fluoreszenz, Circulardichroismus (CD), optische Rotationsdispersion (ORD), Refraktometrie etc. Besonders bevorzugt wird die Veränderung des Coenzyms durch Messung der Fluoreszenz nachgewiesen. Die Fluoreszenzmessung ist hochsensitiv und ermöglicht den Nachweis selbst geringer Konzentrationen des Analyten in miniaturisierten Systemen.
Das erfindungsgemäße Verfahren oder Nachweissystem können einen Flüssigtest umfassen, wobei das Reagenz z.B. in Form einer Lösung oder Suspension in einer wässrigen oder nichtwässrigen Flüssigkeit oder als Pulver oder Lyophilisat vorliegt. Vorzugsweise beinhaltet das erfindungsgemäße Verfahren und Nachweissystem jedoch einen Trockentest, wobei das Reagenz auf einem Träger aufgebracht ist. Der Träger kann beispielsweise einen Teststreifen, umfassend ein saugfähiges oder/und quellbares Material, umfassen, das von der zu untersuchenden Probeflüssigkeit benetzt wird.
In einer besonders bevorzugten Ausführungsform verwendet man als Nachweisreagenz eine Gelmatrix mit einem darin eingelagerten Enzym- Coenzym-Komplex. Die Gelmatrix weist vorzugsweise eine Schichtdicke 50 μm, insbesondere < 5 μm, auf und ist auf einem Träger, beispielsweise einem zumindest teilweise optisch transparenten Träger aufgebracht. Die Gelmatrix kann eine Matrix umfassend ein oder mehrere lösliche Polymere sein, wie bei bekannten Trockentestsystemen (z.B. AccuChek Active), und kann durch Aufrakeln und Trocknen hergestellt werden. Vorzugsweise ist die Matrix ein Polymer, das auf Basis von photopolymerisierbaren Substanzen, wie etwa acrylischen Monomeren, z.B. Acrylamid oder/und Acrylsäureestern, wie Polyethylenglykoldiacrylat, oder
vinylaromatischen Monomeren, z.B. 4-Vinylbenzolsulfonsäure, oder Kombinationen davon, aufgebaut ist. Zur Herstellung einer derartigen Gelmatrix kann eine Flüssigkeit, die das Reagenz, umfassend Enzym, photopolymerisierbares Monomer sowie gegebenenfalls Coenzym, Photoinitiator oder/und nichtreaktive Bestandteile, enthält, auf einen zumindest teilweise optisch transparenten Träger, beispielsweise auf eine Plastikfolie, aufgetragen und z.B. mit UV-Licht von der Rückseite her bestrahlt werden, so dass eine Polymerisation des Monomers oder der Monomere auf dem Träger bis zu einer vorbestimmten Schichtdicke erfolgt. Die Schichtdicke kann durch Zugabe von absorbierenden Substanzen zum Reagenz oder/und durch die Bestrahlungsdauer bzw. -intensität gesteuert werden. Überschüssiges flüssiges Reagenz kann nach der Polymerisation entfernt und erneut eingesetzt werden (siehe beispielsweise Figur 2).
Andererseits kann die Gelmatrix auch durch konventionelle Beschichtungsprozeduren hergestellt werden, wobei das flüssige Reagenz auf einen Träger aufgetragen, dort mit geeigneten Methoden, z.B. mit einem Rakel, auf die gewünschte Dicke gebracht und dann vollständig polymerisiert wird.
Nach Einpolymerisierung oder Einbetten in die Gelmatrix befindet sich das Enzym in einer geschützten Mikroumgebung. Bei ausreichender Vernetzung der polymeren Gelmatrix liegen die Enzymmoleküle in einer immobilisierten Form vor. Niedermolekulare Substanzen bzw. Glucose oder andere Analyten oder auch Coenzyme können aber frei durch das Polymernetzwerk diffundieren.
Das Enzym kann entweder zusammen mit seinem Coenzym in die Matrix einpolymerisiert werden oder die Matrix kann nach der Polymerisation mit einer Lösung des Coenzyms in Kontakt gebracht werden, so dass sich der entsprechende Enzym-Coenzym-Komplex bildet. Die Konzentration des Enzyms in der Gelmatrix wird vorzugsweise so hoch gewählt, dass eine
stöchiometrische Reaktion mit dem zu bestimmenden Analyten und eine direkte Bestimmung des sich durch die Reaktion verändernden Coenzyms möglich ist. Die Reaktion besteht nur aus einer einzigen katalytischen Reaktion, beispielsweise einer Redoxreaktion, die im Bereich von Milli- oder Mikrosekunden ablaufen kann. Das durch die Reaktion veränderte Coenzym ist durch Bindung an das aktive Zentrum des Enzyms und gegebenenfalls zusätzlich durch Einlagerung in die Gelmatrix optimal vor Störeinflüssen geschützt.
Weiterhin soll die Erfindung durch die nachfolgenden Abbildungen und Beispiele erläutert werden.
Figuren
Figur 1 zeigt eine erste Ausführungsform des erfindungsgemäßen Nachweissystems. Auf einem optisch transparenten Träger (1 ) ist eine Reagenzschicht (2), z.B. eine Gelmatrix mit einem Enzym-Coenzym- Komplex, aufgebracht. Der Enzym-Coenzym-Komplex liegt in einer solchen Form vor, dass keine Regeneration des Coenzyms während der Analytbestimmung erfolgen kann. Auf die Reagenzschicht wird eine Probe (3), z.B. Blut, gegeben. Die Bestimmung der enzymatischen Reaktion zwischen dem in der Probe (3) enthaltenen Analyten und dem in der Reagenzschicht (2) enthaltenen Enzym-Coenzym-Komplex erfolgt durch optische Methoden. Licht aus einer Lichtquelle (4), z.B. einem Laser oder einer LED, wird von hinten (durch den Träger) auf die Reagenzschicht (2) eingestrahlt. Von der Probe zurück gestrahltes Absorptions- oder Fluoreszenzlicht wird in einem Detektor (5) nachgewiesen. Gegebenenfalls wird - insbesondere zum Nachweis von Fluoreszenzlicht - ein optisches Filterelement (6) vor den Detektor geschaltet, um ein Übersprechen des Fluoreszenzanregungslichts zu blockieren.
Figur 2 zeigt die Herstellung eines erfindungsgemäßen Nachweissystems. Auf einen optisch transparenten Träger (11 ), z.B. einer Plastikfolie, wird ein flüssiges Reagenz (12) beispielsweise an einer ersten Position (13) aufgebracht. Das flüssige Reagenz (12) wird an einer zweiten Position von unten durch den Träger (1 1 ) mit Licht aus einer Lichtquelle (14) bestrahlt. Gleichzeitig wird der Träger in die mit Pfeil gekennzeichnete Richtung (15) bewegt. Es wird unmittelbar auf dem Träger (1 1 ) eine polymerisierte Reagenzschicht (16) gebildet. Über der Polymerschicht (16) befindet sich überschüssiges flüssiges Reagenz. Die Dicke der polymerisierten Reagenzschicht (16) kann durch die Reagenzzusammensetzung, die Dauer und Intensität der Lichteinstrahlung sowie durch die Eigenschaften des Trägers (1 1 ) gesteuert werden.
Figur 3 zeigt eine Ausführungsform eines Fluoreszenz-basierenden Sensors von unten. Eine, beispielsweise durch das kontinuierliche Verfahren in Figur 2 hergestellte polymerisierte Reagenzschicht, kann geschnitten und unter Verwendung bekannter Techniken auf einem Träger (21 ) aufgebracht werden. Nach Aufbringen der Probe auf die Oberseite wird aus einer Lichtquelle von unten Anregungslicht (23), z.B. UV-Licht, eingestrahlt. Die durch die Reaktion des Analyten mit dem Enzym-Coenzym-Komplex in der Reagenzschicht (22) erzeugte Fluoreszenz (24), z.B. blaues Licht, wird mit einem Detektor nachgewiesen.
Auch mehrere (gleiche oder verschiedene) Reagenzien können auf einem Träger aufgebracht werden. Ein Beispiel für eine solche Ausführungsform in Form einer Scheibe ist in Figur 4 gezeigt. Auf dem optisch transparenten Träger (31 ) sind mehrere Reagenzspots (32) angeordnet.
Die Figuren 5A und 5B zeigen die Fluoreszenz eines erfindungsgemäßen Nachweissystems (Glucose-Dehydrogenase und NAD+) bei zunehmender Glucosekonzentration unter einer CCD-Kamera.
Beispiele
Beispiel 1 : Stöchiometrischer Nachweis von Glucose im System Glucose- Dehydrogenase (GlucDH)/NAD+ in einer Küvette
100 mg/ml GlucDH werden in Puffer pH 7 gelöst und mit der entsprechenden Menge NAD+ versetzt. Bei Zugabe von ansteigenden Mengen Glucose lässt sich visuell ein Anstieg der Fluoreszenz unter einer UV-Lampe (Anregungswellenlänge 366 nm) erkennen (Figuren 5A und 5B).
Die Lösung mit dem Enzymsytem fluoresziert ohne Glucose nicht. Auch Glucose und NAD+ ergeben keine Fluoreszenz.
Beispiel 2: Nachweis von Glucose in dem System GlucDH/NAD + in einem Polymerfilm
Eine Suspension folgender Substanzen wurde in einem Plastikreagenzglas vermischt.
Rezeptur 1
0,5 ml dieser Suspension wurden mit 0,5 ml einer Lösung von GlucDH (100 mg/ml) versetzt und die Mischung im Ultraschallbad luftblasenfrei homogenisiert.
Die klare Lösung wurde auf eine 1 25 mm dicke coronabehandelte Polycarbonatfolie gegossen und 20 min mit einer herkömmlichen Belichtungsapparatur (Isel-UV-Belichtungsgerät 2) belichtet. Die Folie wurde kurz mit Wasser gewaschen und anschließend an der Luft getrocknet.
Die sich ergebende Schichtdicke war < 2 μm. Eine frisch angesetzte Glucose/NAD+-Lösung (GKL-3-Lösung, 300 mg/dl Glucose, 1 ml/6,4 mg NAD+) wurde auf den Film aufgetüpfelt. Sofort war unter der UV-Lampe eine starke Fluoreszenz sichtbar.
Beispiel 3: Beeinflussung der Schichtdicke durch Zugabe eines UV- Adsorbers
Es wurde eine Polymerschicht hergestellt, die einen blauen Farbstoff (Absorptionsmaximum ~ 650 nm) zur besseren Erkennung enthielt (Rezeptur 2). In einem weiteren Versuch wurde der Ausgangsrezeptur ein gelber Farbstoff als UV-Absorber beigemischt (Rezeptur 3).
Rezeptur 2
Die Mischung wurde durch Rühren und durch Ultraschallbad-Behandlung homogenisiert, auf eine 140 μm Pokalonfolie (coronabehandelt, Stufe 4) mit der Pipette verteilt und auf einem UV-Belichtungsgerät (Actina U4, W. Lemmen GmbH) 1 min belichtet.
Die sich ergebende Schichtdicke wurde mit einer Mikrometerschraube gemessen und betrug 240,5 μm.
Rezeptur 3
Die Mischung wurde, wie zuvor beschrieben, auf einer Folie verteilt und dann polymerisiert. Die sich ergebende Schichtdicke wurde mit einer Mikrometerschraube gemessen und betrug 79,3 μm.
Dieses Experiment zeigt, dass sich eine Schichtdickenbeeinflussung realisieren lässt: Ohne UV-Adsorber beträgt die Schichtdicke bei ansonsten gleichen Reaktionsbedingungen 240, 5μm (s.o.); mit UV-Adsorber (Mordant Yellow 7) lediglich 79,3 μm.
Claims
1. Verfahren zum Nachweis eines Analyten in einer Probe durch eine enzymatische Reaktion, umfassend die Schritte:
(a) Inkontaktbringen der Probe mit einem Nachweisreagenz, umfassend einen Enzym-Coenzym-Komplex, unter Bedingungen, bei denen keine Regeneration des Coenzyms erfolgt, und (b) Nachweisen einer Reaktion des Analyten durch Veränderung des Enzym-Coenzym-Komplexes.
2. Verfahren nach Anspruch 1 , dadurch gekennzeichnet, dass die enzymatische Reaktion eine Redoxreaktion umfasst.
3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass man als Enzym eine Oxidoreduktase verwendet und eine Veränderung des Coenzyms durch Oxidation oder Reduktion nachweist.
4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, dass man als Enzym eine Dehydrogenase verwendet, ausgewählt aus einer Glucose-Dehydrogenase (E.C.1 .1 .1 .47), Lactat- Dehydrogenase (E.C.1 .1 .1 .27, 1 .1 .1 .28), Malat-Dehydrogenase (E.C.1 .1 .1 .37), Glycerin-Dehydrogenase (E.C.1 .1 .1 .6), Alkohol- Dehydrogenase (E.C 1 .1 .1 .1 ) oder Aminosäure-Dehydrogenase, z.B. L-Aminosäure-Dehydrogenase (E.C.1 .4.1 .5).
5. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass man ein Coenzym ausgewählt aus Flavin-, Nicotin- und Chinonderivaten verwendet.
6. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass man das Coenzym auswählt aus FAD, FADH2, FMN, FMNH2, Coenzym Q, PQQ, NAD + , NADH/H + , NADP+ und NADPH/H + .
7. Verfahren nach Anspruch 1 bis 6, dadurch gekennzeichnet, dass man die Veränderung des Coenzyms durch optische Methoden nachweist.
8. Verfahren nach Anspruch 7, dadurch gekennzeichnet, dass man die Veränderung des Coenzyms durch Messung von Absorption, Fluoreszenz, CD, ORD oder Refraktometrie nachweist.
9. Verfahren nach Anspruch 8, dadurch gekennzeichnet, dass man die Veränderung des Coenzyms durch Messung der Fluoreszenz nachweist.
10. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass man als Nachweisreagenz eine Gelmatrix mit einem darin eingelagerten Enzym-Coenzym-Komplex verwendet.
1 1 . Verfahren nach Anspruch 10, dadurch gekennzeichnet, dass die Gelmatrix eine Schichtdicke von < 50 μm, insbesondere von < 5 m, aufweist.
12. Verfahren nach Anspruch 10 oder 1 1 , dadurch gekennzeichnet, dass man eine Gelmatrix auf Basis von photopolymerisierbaren Substanzen verwendet.
13. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass man einen Analyten in einer Körperflüssigkeit bestimmt.
14. Verfahren nach Anspruch 13, dadurch gekenzeichnet, dass man eine Bestimmung von Glucose im Blut durchführt.
1 5. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass die Reaktion des Analyten eine Dauer von < 5 s aufweist.
16. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass die Reaktion in Abwesenheit von Mediatoren, die mit dem
Coenzym reagieren können, durchgeführt wird.
17. Verfahren nach einem der vorhergehenden Ansprüche, dadurch gekennzeichnet, dass man den Enzym-Coenzym-Komplex als stöchiometrischen
Reaktionspartner bezüglich des Analyten einsetzt.
18. Reagenzsystem zum Nachweis eines Analyten in einer Probe, umfassend:
(a) ein Nachweisreagenz, umfassend einen Enzym-Coenzym- Komplex, in einer Form, dass keine Regeneration des Coenzyms erfolgt, und
(b) einen Träger zur Aufnahme des Nachweisreagenz.
19. Reagenzsystem nach Anspruch 18, dadurch gekennzeichnet, dass der Enzym-Coenzym-Komplex in einer Gelmatrix eingelagert ist.
20. Reagenzsystem nach Anspruch 18 oder 19, dadurch gekennzeichnet, dass der Träger zumindest teilweise optisch transparent ist.
21 . Reagenzsystem nach einem der Ansprüche 18 bis 20, dadurch gekennzeichnet, dass der Träger eine im Wesentlichen planare Struktur umfasst.
22. Reagenzsystem nach einem der Ansprüche 18 bis 20, dadurch gekennzeichnet, dass der Träger mehrere unterschiedliche Nachweisreagenzien umfasst.
23. Verwendung eines Reagenzsystems nach einem der Ansprüche 18 bis 22 in einem Verfahren nach einem der Ansprüche 1 bis 17.
