Description
- Die vorliegende Erfindung betrifft eine neuartige Gruppe von Piperazin- und Di-dehydropiperidinderivaten mit interessanten pharmakologischen Eigenschaften aufgrund einer Kombination von sowohl durch partiellen Dopamin D2-Rezeptor Agonismus, als auch partiellen Serotonin 5-HT1A-Rezeptor Agonismus vermittelten Wirksamkeiten. Zusätzlich ist eine Affinität für adrenerge α1-Rezeptoren vorhanden.
- Es ist von EP-0189612 bekannt, dass Piperazinderivate, welche an einem Stickstoff mit einer Phenyl-heterocyclischen Gruppe substituiert und an dem anderen Stickstoffatom nicht substituiert sind, Psychotrope Wirksamkeit haben.
- Ferner ist von EP-0190472 bekannt, dass Benzofuran- und Benzodioxol-Piperazinderivate, welche an dem anderen Stickstoffatom der Piperazingruppe substituiert sind, ebenfalls Psychotrope Wirksamkeit haben.
- Letztendlich ist es von EP-0169148 bekannt, dass 1,3-Dihydro-4-(1-ethyl-1,2,3,6-tetrahydropyridin-4-yl)-2H-indol-2-on und ähnliche Verbindungen analgetische Eigenschaften haben.

- – S1 Wasserstoff, Halogen, Alkyl (1-3 C) , CN, CF3, OCF3, SCF3, Alkoxy (1-3 C), Amino oder mono- oder dialkyl (1-3 C) substituiertes Amino oder Hydroxy darstellt,
- – ... Z für =C oder -N steht,
- – R1 und R2 unabhängig H oder Alkyl (1-3 C) darstellen oder R1 und R2 zusammen eine Brücke aus 2 oder 3 C-Atomen bilden können,
- – R4 für Wasserstoff oder Alkyl (1-3 C) steht,
- – Q für Methyl, Ethyl, Ethyl, substituiert mit einem oder mehreren Fluoratomen oder Cyclopropylmethyl, gegebenenfalls substituiert mit einem oder mehreren Fluoratomen, steht, und Salze davon eine Kombination aus partiellen Dopamin D2-Rezeptor Agonismus und partiellen Serotonin 5-HT1A-Rezeptor Agonismus Wirksamkeiten haben.
- Bevorzugte Verbindungen gemäß der Erfindung sind Verbindungen der Formel (I), worin S1, R1, R2 und R4 Wasserstoff darstellen und ... Z und Q die obigen Bedeutungen haben, und die Salze davon.
- Besonders bevorzugt sind die Verbindungen, worin S1, R1, R2 und R4 Wasserstoff darstellen, ... Z für -N steht und Q für Methyl oder Ethyl steht, und Salze davon, wobei die am meisten bevorzugte Verbindung diejenige ist, worin Q für Methyl steht.
- Verbindungen gemäß der Erfindung zeigen Affinitäten für sowohl den Dopamin D2-Rezeptor (pKi Bereich 7,5–8,5), als auch den Serotonin 5-HT1A-Rezeptor (pKi Bereich 7,0–8,0), gemessen gemäß gut definierter Verfahren (z. B.: Creese I., Schneider R. und Snyder S. H., [1H]-Spiroperidol labels dopamine receptors in rat pituitary and brain, Eur. J. Pharmacol. 1997, 46: 377–381 und Gozlan H., El Mestikawy S., Pichat L., Glowinsky J. und Hamon M., 1983, Identification of presynaptic serotonin autoreceptors using a new ligand 3H-PAT, Nature 1983, 305: 140–142).
- Die Verbindungen zeigen variierende Wirksamkeiten als partielle Agonisten am Dopamin D2-Rezeptor und, überraschend, am 5-HT1A-Rezeptor. Diese Wirksamkeit wurde an der Bildung von Adenylatcyclase in Zelllinien gemessen, welche diese geklonten Rezeptoren exprimieren (z. B. humane D2-Rezeptoren und 5-HT1A-Rezeptoren, exprimiert in CHO-Zelllinien gemäß den durch Solomon Y., Landos C., Rodbell M., 1974, A highly selective adenylyl cyclase assay, Anal. Biochem. 1974, 58: 541–548 und Weiss S., Sebben M. und Bockaert J. J., 1985, Corticotropin-peptide regulation of intracellular cyclic AMP production in cortical neurons in primary culture, J. Neurochem. 1985, 45: 869–874 beschriebenen Verfahren).
- Die einzigartige Kombination aus sowohl partiellem Dopamin D2-Rezeptor Agonismus und partiellem Serotonin 5-HT1A-Rezeptor Agonismus ergibt eine überraschend breite Wirksamkeit in mehreren Tiermodellen, welche für psychiatrische und/oder neurologische Störungen vorhersehend sind.
- Die Verbindungen zeigen eine überraschend hohe Wirksamkeit in einem therapeutischen Modell für anxiolytische/antidepressive Wirksamkeit: dem konditionierten Ultraschall-Vokalisationsmodell in Ratten (siehe z. B.: Molewijk H. E., Van der Poel, A. M., Mos J., van der Heyden J. A. M. und Olivier B. (1995), Conditioned ultrasonic vocalizations in adult male rats as a paradigm for screening anti-panic drugs, Psychopharmacology 1995, 117: 32– 40). Die Wirksamkeit der Verbindungen in diesem Modell war im unteren Mikrogramm/kg Bereich, was überraschend wirksamer (um einen Faktor 100 bis 3000) im Vergleich zu den Verbindungen ist, welche vorhergehend in EP-0190472 und EP-0398413 beschrieben wurden.
- Zusätzlich zeigen diese Verbindungen ebenfalls Wirkungen in Modellen, welche für antidepressive Wirkung vorhersehend sind, bei höheren Dosen (erzwungener Schwimmtest, siehe z. B.: Porsolt R. D., Anton G., Blavet N. und Jalfre M., 1978, Behavioural despair in rats: A new model sensitive to antidepressant treatments, Eur. J. Pharmacol. 1978, 47: 379–391 und die differentielle Verstärkung von niedrigen Raten von respondierenden Modellen in Ratten, siehe z. B.: McGuire P. S. und Seiden L. S., The effects of tricyclic antidepressants on performance under a differential-reinforcement-of-low-rate schedule in rats, J. Pharmacol. Exp. Ther. 1980, 214: 635–641).
- Bei höheren Dosen wurden auch Dopamin Antagonist-ähnliche Wirkungen beobachtet (Antagonismus von Apomorphin-verursachtem Kletterverhalten in Mäusen, (A), z. B.: Costall B., Naylor R. J. und Nohria V., Differential actions of typical and atypical agents on two behavioural effects of apomorphine in the mouse, (B), Brit. J. Pharmacol. 1978, 63: 381–382; Suppression von lokomotorischer Aktivität, z. B.: File S. E. und Hyde J. R. G., A test of anxiety that distinguishes between the actions of benzodiazepines and those of other minor tranquillisers or stimulants, Pharmacol. Biochem. Behav. 1979, 11: 65–79 und Hemmung von konditionierter Vermeidungsresponse in Ratten, z. B.: Van der Heyden J. A. M., Bradford L. D., A rapidly acquired one-way conditioned avoidance procedure in rats as a primary screening test for antipsychotics: influence of shock intensity on avoidance performance, Behav. Brain. Res. 1988, 31: 61–67). von den ersten beiden Wirksamkeiten A und B ist vorhergehend für partielle Do pamin D2-Rezeptor Agonisten durch Mewshaw et al., Bioorg. Med. Chem. Lett. 8 (1998) 2675 berichtet worden.
- Die Verbindungen sind wahrscheinlich bei der Behandlung von Affektionen oder Erkrankungen des zentralen Nervensystems von Wert, welche durch Störungen des dopaminergen und/oder serotonergen Systems verursacht werden, zum Beispiel: Angststörungen (einschließlich z. B. generalisierter Angst, Panik, obzessiven Zwangsstörungen), Depression, Autismus, Schizophrenie, Parkinson-Krankheit, Störungen von Kognition und Gedächtnis.
- Geeignete Säuren, mit welchen die Verbindungen der Erfindung annehmbare Säureadditionssalze bilden können, sind zum Beispiel Salzsäure, Schwefelsäure, Phosphorsäure, Salpetersäure und organische Säuren wie Zitronensäure, Fumarsäure, Maleinsäure, Weinsäure, Essigsäure, Benzoesäure, p-Toluolsulfonsäure, Methansulfonsäure und Naphthalinsulfonsäure.
- Proarzneien sind Derivate der Verbindungen mit der Formel (I), worin R4 für eine Gruppe steht, welche nach Verabreichung leicht entfernt wird. Geeignete Proarzneien sind zum Beispiel Verbindungen, worin N-R4 für eine der folgenden Gruppen steht: Amidin, Enamin, eine Mannich-Base, ein Hydroxymethylenderivat, ein O-(Acyloxymethylencarbamat)derivat, Carbamat oder Enaminon.
- Die Verbindungen und die Salze davon können mittels üblicher Verfahren unter Verwendung von Hilfssubstanzen wie flüssigen und festen Trägermaterialien in Formen zur Verabreichung gebracht werden.
- Die Verbindungen der Erfindung können gemäß Verfahren hergestellt werden, welche für die Synthese von analogen Verbindungen bekannt sind.
- Verbindungen mit der Formel (I) können durch Umsetzen der entsprechenden Verbindung, worin Q Wasserstoff darstellt, mit einer Verbindung Q-Hal erhalten werden, worin Q Methyl, (gegebenenfalls fluoriertes) Ethyl oder (gegebenenfalls fluoriertes) Cyclopropylmethyl darstellt und Hal für Halogen, vorzugsweise Iod steht. Diese Umsetzung kann in einem Lösungsmittel wie Acetonitril in Gegenwart einer Base, zum Beispiel Ethyldiisopropylamin oder Triethylamin durchgeführt werden.
- Die Ausgangsverbindungen, worin Q für Wasserstoff steht und ... Z für -N sind bekannt oder können wie in EP-0189612 beschrieben erhalten werden. Ausgangsverbindungen, worin Q für Wasser stoff steht und ... Z für =CH2 können wie unten beschrieben erhalten werden.
- Die Verbindungen der Erfindung, worin ... Z für -N steht, können auch durch Umsetzen einer Verbindung mit der Formel (II) mit einer Verbindung mit der Formel (III) in welchen Formeln die Symbole die obigen Bedeutungen haben, erhalten werden. Diese Umsetzung kann in einem organischen Lösungsmittel wie Chlorbenzol durchgeführt werden.

- Die Ausgangsverbindung für Schritt (i) kann gemäß dem in J. Org. Chem. 45, (1980), 4789 beschriebenen Verfahren erhalten werden und Schritt (i) selbst kann wie in J. Org. Chem. 47, (1982), 2804 beschrieben durchgeführt werden.
- Schritt (ii) wird auf eine Weise durchgeführt, welche für diesen Typ von chemischen Umsetzungen bekannt ist, und wird in Beispiel 3 veranschaulicht.
- Die Erfindung wird in den folgenden Beispielen veranschaulicht:
- Beispiel 1
- 1,28 g (5 mmol) I-H·HCl wurden in 25 ml Acetonitril suspendiert und 0,34 ml (4,4 mmol) Ethyliodid wurden zusammen mit 5 ml Di-isopropylethylamin zugegeben. Das sich ergebende Umsetzungsgemisch wurde gerührt und für 18 Std. unter einer Stickstoffatmosphäre refluxiert. Das Umsetzungsgemisch durfte Raumtemperatur erreichen, wonach eine kleine Menge an SiO2 zugegeben wurde. Die sich ergebende Suspension wurde in vacuo konzentriert, was ein Pulver hinterließ, welches auf eine chromatographische Säule gegeben wurde, wonach ein Chromatographie-Durchlauf durchgeführt wurde (SiO2, Eluent CH2Cl2/MeOH 95/5), was 0,55 g weißen Feststoff ergab. Letzterer wurde aus EtOAc/EtOH (ca. 1/1) umkristallisiert, zu welchem 1,1 Äquivalent von 1 M HCl/EtOH zugegeben wurde. Die Kristalle wurden durch Filtration gesammelt, Waschen mit EtOAc bzw. Diethylether ergab nach Trocknen 0,5 g (42%) des gewünschten HCl-Salzes der Verbindung, worin S1, R1, R2 und R4 Wasserstoff darstellen, ... Z für -N steht und Q für Ethyl steht, Fp. 280–2°C (Zers.).
- Beispiel 2
- 6,0 g (40 mmol) der Verbindung mit der Formel (II) (worin S1 und R4 für Wasserstoff stehen und X Sauerstoff darstellt) wurden in 150 ml Chlorbenzol gelöst, wonach 8,47 g (44 mmol) N-Methyl-bis(chlor-ethyl)amin-monohydrochlorid zugegeben wurden. Das sich ergebende Umsetzungsgemisch wurde gerührt und zu Rückfluss gebracht. Das in den Ausgangsmaterialien vorhandene Wasser wurde mittels einer Dean-Stark-Vorrichtung abgetrennt. Nach 44 Std. hatte sich ein Feststoff gebildet und das Umsetzungsgemisch durfte Raumtemperatur erreichen. Die Flüssigkeit wurde abgetrennt, der Rückstand wurde mit Toluol gewaschen, wonach er in Ethanol refluxiert wurde. Nach Kühlen wurde der Feststoff filtriert und nachfolgend durch Blitz-Säulenchromatographie (SiO2, Eluent: CH2Cl2/MeOH/NH4OH = 97/2,5/0,5) gereinigt. Dieses Verfahren ergab 4,5 g Feststoff, welcher in 96% EtOH (ca. 300 ml) gelöst wurde, wonach unter Rühren 2 Äquivalente von 1 M HCl/MeOH zugegeben wurden. Kristallisation begann und schließlich konnten nach Filtration und Trocknen 4,15 g (38%) des Hydrochlorids der gewünschten Verbindung isoliert werden, worin S1, R1, R2 und R4 für Wasserstoff stehen, ... Z für -N steht und Q Methyl darstellt, Fp. 301,5–302,5°C.
- Beispiel 3
- Unter einer inerten Atmosphäre wurden 16,5 g (78,2 mmol) N-( tert.-Butyloxycarbonyl)-meta-fluoranilin in 230 ml trockenem Tetrahydrofuran (THF) gelöst, wonach die Lösung auf –75°C gekühlt wurde (Trockeneis, Aceton). Unter Rühren wurde eine im Handel erhältliche Lösung aus 1,5 M tert.-Butyllithium in Heptan (ca. 156 mmol, 2 Moläquivalente) langsam zugegeben, wonach das Umsetzungsgemisch für 0,5 Std. bei –70°C und nachfolgend für 2 zusätzliche Stunden bei –25°C gerührt wurde. Wieder wurde das Umsetzungsgemisch auf –75°C gebracht und eine Lösung aus 9,6 ml N-Methylpiperidon (78,2 mmol, 1 Moläquivalent) in ca. 25 ml trockenem THF. Das Umsetzungsgemisch durfte Raumtemperatur erreichen und wurde für zusätzliche 16 Std. gerührt. Nachfolgend wurde eine Lösung aus 1,5 ml (83 mmol) H2O in 50 ml MeOH langsam zum Umsetzungsgemisch zugegeben, wonach 100 ml SiO2 zugegeben wurden. Die Suspension wurde zu Trockenheit eingedampft, wonach der sich ergebende pulverige Rückstand auf eine Chromatographiesäule gegeben wurde, wonach ein Blitz-Chromatographiedurchlauf durchgeführt wurde (SiO2, erster Eluent: EtOAc, zweiter Eluent: MeOH/EtOAc/Triethylamin 15/85/1), was 12,4 g eines dunkelgelben Öls ergab. Unter Rühren wurden 4,7 g (ca. 15,5 mmol) des erhaltenen Produkts in 100 ml Dioxan gelöst, wonach 100 ml konzentrierte HCl zugegeben wurden und das sich ergebende Gemisch wurde für 1 Std. refluxiert. Das Umsetzungsgemisch durfte Raumtemperatur erreichen, wonach es in vacuo konzentriert wurde, was einen festen Rückstand ergab. Der Rückstand wurde suspendiert und in i-Propanol gerührt, wonach der Feststoff filtriert und nachfolgend mit EtOAc, Diethylether bzw. Hexan gewaschen wurde. Nach Trocknen blieben 3,1 g Rückstand übrig, von welchen 1,5 g in EtOH suspendiert wurden, wobei die letztere Suspension für 1 Std. refluxiert wurde. Das Gemisch durfte Raumtemperatur erreichen, wonach es filtriert wurde, was einen Rückstand ergab, welcher mit absolutem EtOH bzw. Di(i-propyl)ether gewaschen wurde. Nach Trocknen wurden 1,1 g (53%) der gewünschten Verbindung erhalten, worin S1, R1, R2 und R4 Wasserstoff darstellen, ... Claims (11)
- Verbindungen mit der Formel (I) worin – S1 Wasserstoff, Halogen, Alkyl (1-3 C) , CN, CF3, OCF3, SCF3, Alkoxy (1-3 C), Amino oder mono- oder dialkyl (1-3 C) substituiertes Amino oder Hydroxy darstellt, – ... Z für =C oder -N steht, – R1 und R2 unabhängig H oder Alkyl (1-3 C) darstellen oder R1 und R2 zusammen eine Brücke aus 2 oder 3 C-Atomen bilden können, – R4 für Wasserstoff oder Alkyl (1-3 C) steht, – Q für Methyl, Ethyl, Ethyl substituiert mit einem oder mehreren Fluoratomen, oder Cyclopropylmethyl gegebenenfalls substituiert mit einem oder mehreren Fluoratomen, steht, und Salze davon.
- Verbindungen wie in Anspruch 1 beansprucht, worin S1, R1, R2 und R4 Wasserstoff darstellen, Q für Methyl oder Ethyl steht und ... Z die in Anspruch 1 gegebene Bedeutung hat.
- Verbindungen wie in Anspruch 2 beansprucht, worin ... Z für -N steht.
- Verbindung wie in Anspruch 3 beansprucht, worin Q für Methyl steht.
- Verfahren zur Herstellung der in Anspruch 1 beanspruchten Verbindungen durch Umsetzen einer Verbindung mit der Formel (I), worin Q Wasserstoff darstellt, mit einer Verbindung der Formel Q-Hal, worin Q Methyl oder (gegebenenfalls fluoriertes) Ethyl, (gegebenenfalls fluoriertes) Cyclopropylmethyl darstellt und Hal für Halogen steht.
- Pharmazeutische Zusammensetzungen, welche mindestens eine Verbindung wie in Anspruch 1 beansprucht als wirksamen Bestand teil enthalten.
- Verfahren zum Herstellen einer pharmazeutischen Zusammensetzung, dadurch gekennzeichnet, dass eine Verbindung wie in Anspruch 1 beansprucht in eine zur Verabreichung geeignete Form gebracht wird.
- Verwendung einer Verbindung wie in Anspruch 1 beansprucht zur Herstellung einer Zusammensetzung zur Behandlung von CNS-Störungen.
- Verwendung einer Verbindung wie in Anspruch 1 beansprucht zur Herstellung einer Zusammensetzung zur Behandlung von Angstzuständen und/oder Depression.
