Description
- Hintergrund der Erfindung
- Die vorliegende Erfindung betrifft ein Verfahren zur Verringerung des brennenden Nachgeschmacks von Propionsäure-Zusammensetzungen, genauer gesagt Ibuprofen-Zusammensetzungen.
- Viele Aromastoffe und Süßungsmittel wurden zur Medikation hinzugefügt, um sie wohlschmeckender zu machen und das Brennen und den Nachgeschmack zu maskieren, der bei vielen Medikationen verbreitet ist. Trotz zahlreicher Bemühungen, ein effektives Mittel zu finden, um dieses Brennen zu beseitigen, besteht ein andauernder Bedarf an einem Verfahren, um effektiv das brennende Gefühl mit Medikationen zu eliminieren, bevorzugterweise kann das Brennen auf ein Niveau verringert werden, so daß eine kaubare Zusammensetzung zur Verfügung gestellt werden kann.
- Ibuprofen ist eine gut bekannte Medikation, die nach der Einnahme ein unangenehmes brennendes Gefühl im Mund und dem Hals besitzt.
- EP-A-O 753 296 von American Home Products Corporation versucht, den unangenehmen Nachgeschmack durch zur Verfügung stellen von nur einem Enantiomer zu eliminieren. Die Patentanmeldung beschreibt die Auftrennung von Ibuprofen aus seinem razemischen Gemisch, um eine oral verabreichte Wirkstoffzusammensetzung zu bilden, die nur das S (+)-Ibuprofen und im wesentlichen kein R (-)-Ibuprofen enthält. Während dieser Ansatz eine genießbarere Form von Ibuprofen zur Verfügung stellen kann, ist die Auftrennung und Isolierung der Enantiomere schwierig.
- US-A-4,762,702 beschreibt Ibuprofen-Partikel, die durch eine Beschichtung aus Hydro-Kolloid und Fumarsäure umschlossen sind. Die bevorzugte Hydro-Kolloid-Zusammensetzung umfaßt Xanthan-Gummi und/oder Maltodextrin. Das sich ergebende Produkt ist ein geschäumtes Gemisch, das Vakuum-getrocknet wird. Das Patent beschreibt dieses Verfahren als die Nachteile von Präparationen des Stands der Technik aufgrund des Umhüllens der Ibuprofen-Kristalle durch das Hydro-Kolloid in der Anwesenheit der Fumarsäure überwindend.
- Trotz der Beschreibung der oben genannten Patente ist das zur Verfügung stellen einer Geschmacks-maskierten Ibuprofen-Zusammensetzung ein einfacheres und weniger kostenträchtiges Verfahren.
- Zusammenfassung der Erfindung
- Die vorliegende Erfindung stellt ein Verfahren zur Verringerung des brennenden Nachgeschmacks eines Propionsäure-Derivats, wie in den Ansprüchen 1 bis 4 der beigefügten Ansprüche definiert, zur Verfügung. Die vorliegende Erfindung stellt ausreichend Fumarsäure zur Verfügung, um den brennenden Nachgeschmack von Propionsäure bei Fehlen eines Hydro-Kolloidmittels zur Verfügung zu stellen. In einer bevorzugten Ausführungsform wird die Propionsäure-Derivat-Fumar-Zusammensetzung, die so hergestellt wird, in einer kaubaren Form zur Verfügung gestellt, kann jedoch auch eine flüssige Formulierung sein.
- Genaue Beschreibung der Erfindung
- Propionsäure-Derivate sind eine gut bekannte Klasse von analgetischen Verbindungen. Wie hier verwendet wird von Propionsäure-Derivaten verstanden werden, daß sie einschließen, jedoch nicht begrenzt sind auf, Ibuprofen, Naproxenbenoxaprofen, Naproxennatrium, Flurbiprofen, Fenoprofen, Fenbuprofen, Ketoprofen, Indoprofen, Pirprofen, Carprofen, Oxaprofen, Pranoprofen, Mikroprofen, Tioxaprofen, Suproprofen, Alminoprofen, Tiaprofensäure, Fluprofen und Bucloxinsäure. Die Strukturformel ist in US-Patent 4,923,898 angegeben. Propionsäure-Derivate, wie hier definiert, werden als pharmazeutisch akzeptable Analgetika/nicht-steroidale anti-inflammatorische Wirkstoffe definiert, die ein freies -CH(CH3)COOH oder -CH2CH2COOH oder eine pharmazeutisch akzeptable Salzgruppe, wie zum Beispiel – CH(CH3)COOO-Na+ oder -CH2CH2COO-Na+ aufweisen, die typischerweise direkt oder über eine Carbonyl-Funktionalität an ein aromatisches Ringsystem angebracht sind.
- Propionsäure-Derivate werden typischerweise auf einer täglichen Basis verabreicht, die von ungefähr 50 bis ungefähr 2000 Milligramm, bevorzugterweise von ungefähr 100 bis 1600, und am meisten bevorzugt von ungefähr 200 bis ungefähr 1200 Milligramm reicht.
- Ibuprofen ist ein weit verwendetes gut bekanntes nicht-steroidales anti-inflammatorisches Proprionsäure-Derivat. Ibuprofen ist chemisch als 2-(4-Isobutylphenyl)propionsäure bekannt. Wie hier verwendet wird Ibuprofen als 2-(4-Isobutylphenyl)propionsäure sowie die pharmazeutisch akzeptablen Salze einschließend verstanden. Geeignete Ibuprofensalze schließen Arginin, Lysin, Histidin sowie andere Salze, beschrieben in US-Patent Nrn. 4,279,926, 4,873,231, 5,424,075 und 5,510,385 ein.
- Fumarsäure ist eine weit erhältliche pharmazeutisch akzeptable Säure. Die Konzentration von Fumarsäure, die vorhanden ist, um das Brennen des Propionsäure-Derivats zu inhibieren, wird mit dem Ausmaß der gewünschten Verringerung des Brennens variieren. Im allgemeinen ist der Spiegel von Fumarsäure von ungefähr 50 bis ungefähr 150Gew.% der Propionsäure-Derivatdosierung. Typischerweise liegt der Spiegel von Fumarsäure von ungefähr 60 bis 100Gew.% des Spiegels von Propionsäure-Derivat und am meisten bevorzugt von ungefähr 70 bis ungefähr 90Gew.% des Spiegels der Propionsäure-Derivatdosierung.
- Im Gegensatz zu der Lehre von vorangegangenen Beschreibungen erfordert die vorliegende Erfindung nicht die Inkorporation eines Hydro-Kolloidmaterials, um effektiv zu sein. Die vorliegende Erfindung kann in die folgenden Ausführungsformen inkorporiert sein.
- Die einfachste und bevorzugteste Ausführungsform ist die Inkorporierung von Fumarsäure in eine Matrix, d.h. frei und zufällig in einem Gemisch zur Verfügung gestellt. In dieser Ausführungsform werden das Propionsäure-Derivat, bevorzugterweise Ibuprofen und die Fumarsäure in einer Matrix innerhalb der Caplet-, Tabletten- oder Kapselform zur Verfügung gestellt. In einer anderen Ausführungsform der Erfindung werden das Ibuprofen und die Fumarsäure in einer Granulierung zur Verfügung gestellt. Typischerweise schließt dies das Mischen des Propionsäure-Derivats, der Fumarsäure, sowie Zuckern, Bindemitteln, Wasser und anderen Inhaltsstoffen zusammen unter der Verwendung von gut im Stand der Technik bekannter Ausrüstung ein. Zum Beispiel beschreibt US-Patent Nr. 5,429,825 Roto-Schmelzgranulierungsverfahren. Das Gemisch wird dann getrocknet und gemahlen. Das gemahlene Produkt liegt dann in einer Form vor, die dazu geeignet ist, in eine Tablette komprimiert zu werden. Bevorzugte Sprengmittel werden zu dem Gemisch hinzugefügt, um bei der Freisetzung der aktiven Inhaltsstoffe an den Verbraucher zu helfen. In einer hoch bevorzugten Ausführungsform wird die Propionsäure und Fumarsäure auf eine solche Weise zur Verfü gung gestellt, daß eine kaubare Tablette für Diejenigen zur Verfügung gestellt wird, die Schwierigkeiten beim Schlucken einer Tablette haben.
- In einer anderen Ausführungsform der vorliegenden Erfindung wird die Fumarsäure/Propionsäure in der Anwesenheit eines nicht-hydrokolloidalen Bindemittels zur Verfügung gestellt. Bevorzugterweise ist das nicht-hydrokolloidale Bindemittel ein pharmazeutisch akzeptables Wachs oder Fett. Geeignete Wachse und Fette schließen ein, Glycerylmonostearat, hydrogenierter Talk, Myristylalkohol, Myristinsäure, Stearylalkohol, substituierte Monoglyceride, substituierte Diglyceride, substituierte Triglyceride, Bienenwachs, Carnuba-Wachs, Japan-Wachs, Acetylatmonoglyceride und ähnliches. Kombinationen von zwei oder mehr der nicht-kolloidalen Bindemittel können verwendet werden. Bevorzugterweise weist der Schmelzpunkt der nicht-kolloidalen Bindemittel der Erfindung eine Schmelztemperatur von ungefähr 30 bis ungefähr 100°C auf, am meisten bevorzugt von ungefähr 40 bis ungefähr 85°C.
- Das nicht-hydrokolloidale Bindemittel wird durch zuerst Schmelzen des Wachses oder Fettes und dann Vermischen der Ibuprofen- und Fumarsäurekombination hergestellt. Die Kombination wird dann auf die geeignete Größe gemahlen und dann unter der Verwendung von dem Fachmann im Stand der Technik gut bekannten Techniken in Tabletten gepreßt.
- Die Formulierung der vorliegenden Erfindung kann auch pharmazeutisch akzeptable Hilfsstoffe, Füllstoffe, Aromastoffe, Verdünnungsmittel, Gleitmittel, Sprengmittel, suspendierende Mittel, Stabilisatoren, Bindemittel, Farbstoffe, Träger und ähnliches enthalten.
- Zum Beispiel schließen geeignete Träger Lactose, Stärke, Disclaimphosphat, Calciumsulfat, Kaolin, Mannitol und Puderzucker ein. Typische Bindemittel schließen Stärke, Gelatine, Zucker, wie zum Beispiel Dextrose, Mannitol, Xylitol, Sorbitol, Maltodextrin, Fructose, Saccharose, Melasse, Lactose; natürliche und synthetische Gummis, Carboxymethylzellulose, Methylzellulose, Polyvinylpyrrolidon, Polyethylenglycol, Ethylzellulose und Wachse ein. Gleitmittel schließen Borsäure, Natriumbenzoat, Natriumacetat, Natriumchlorid, Leucin, Polyethylenglycol und ähnliches ein. Typische Sprengmittel schließen ein, Stärke abgeleitet von Holz, Mais, Kartoffeln und Reis, Methylzellulose, Magnesiumsilicate, Aluminiumsilicate, Saccharose, Dextrose, Maltodextrose, Agar, Bentonid, Algininsäure, Holzprodukte, Guar-Gummi, Zitronenpulpe, Natriumlaurylsulfat und ähnliches.
- Die vorliegende Erfindung kann in flüssiger Form, z.B. einem Elixier, Suspension oder Sirup zur Verfügung gestellt werden. Die flüssigen Formulierungen werden unter der Verwendung von Herstellungsverfahren und pharmazeutisch akzeptablen Oberflächenmitteln, dispergierenden Mitteln und Verdünnungsmitteln hergestellt, die im Stand der Technik bekannt sind. Bevorzugterweise wird die vorliegende Erfindung in Caplets, Kapseln, Tabletten und am meisten bevorzugt in einer kaubaren Form zur Verfügung gestellt.
- Wie innerhalb dieser Beschreibung verwendet, wird Brennen als den Nachgeschmack bedeutend verstanden, der gewöhnlich als metallisch bemerkt wird, wenn Ibuprofen eingenommen wird. Dieser Nachgeschmack unterscheidet sich von bitteren Geschmack insoweit, als die Zugabe eines Süßungsmittels bei der Verringerung des Nachgeschmacks nicht effektiv ist.
- Alternativ kann die Ibuprofen, Fumarsäure-Zusammensetzung in geeigneten Spiegeln, die wünschenswert sein können, zu Getränken, Nahrungsmitteln und anderen eßbaren Zusammensetzungen hinzugefügt werden. Es ist auch vorgesehen, daß die Ibuprofen/Fumarsäure-Zusammensetzungen der vorliegenden Erfindung in Veterinäranwendungen verwendet werden können.
- Ohne durch irgendeine Theorie gebunden sein zu wollen, verringert die Inkorporation von Fumarsäure zu dem Propionsäure-Derivat das charakteristische Nachgeschmacks-Brennen durch Ansäuern des Speichels in ausreichendem Maße, um die protonierte Form des Propionsäure-Derivats beizubehalten. Die protonierte Form des Propionsäure-Derivats weist eine geringe Löslichkeit auf und daher eine geringere Irritation der Hals-Schleimhaut. Anders als andere Azidulate, löst sich Fumarsäure langsam, so daß sein saurer Geschmack im Mund minimal ist, jedoch ausreichend, um den Hals anzusäuern. Andere pharmazeutisch akzeptable Säuren, wie zum Beispiel die Zitronen-, Äpfel- und Tartarsäuren sind viel löslicher als Fumarsäure. Diese und andere Säuren bewirken sehr schnell einen unakzeptablen sauren Geschmack im Mund. Die Lösung ist so schnell, daß der saure Geschmack weit vor einer ausreichenden Ansäuerung des Speichels wahrgenommen wird.
- Die Erfindung wird nun durch die folgenden Beispiele beschrieben werden, ist jedoch nicht gedacht, als dadurch begrenzt zu sein. In diesen Beispielen soll es verstanden sein, daß, wenn nicht anders angegeben, alle Teile in Gewichtsprozent vorliegen.
- Beispiel 1
- Kaubare Tabletten, die 100 Milligramm (mg) an Ibuprofen enthalten, wurden mit entweder 70 mg Fumarsäure oder mit keiner Fumarsäure präpariert. Die kaubaren Tabletten, die keine Fumarsäure enthalten, wurden als eine Kontrolle verwendet. Ein Geschmacks-Panel von 18 Subjekten wurde gebeten, zwei Kontrolltabletten zu kauen und das Halsbrennen auf einer Skala von 1 bis 9 (höchster Spiegel) zu bewerten. Nach einer Stunde wurden Tabletten, die die Ibuprofen/Fumarsäurekombination enthielten, gekaut und die Subjekte wurden gebeten, die Ergebnisse unter der Verwendung derselben 1-9-Skala zu bewerten.
- Die Subjekte bewerteten die Kontrolltabletten als moderat hoch, als eine 7 auf einer 9-Punkteskala, wohingegen die Tabletten, die die Ibuprofen/Fumarsäurekombination enthielten, als moderat niedrig, eine 3 oder 4 auf der 9-Punkteskala, bewertet wurden.
- Beispiel 2
- Die folgende Formulierung wurde als effektiv bei der Eliminierung des Nachbrennens gefunden. Ibuprofen 100 Milligramm (mg), Farbstoff 1,76 mg, mikrokristalline Zellulose 84 mg, Süßungsmittel 11 mg, zweites Süßungsmittel 4 mg, Aromastoff 2 mg, Gleitmittel 6 mg, Hilfsstoff 465 mg, Fumarsäure 65 mg.
- Beispiel 3
- Zwei Subjekte wurden verwendet, um den effektiven Spiegel von Fumarsäure für eine 100 mg-Dosierung an Ibuprofen zu bestimmen. Die Subjekte bewerteten die Ibuprofen/Fumarsäure-Zusammensetzung mit der folgenden Skala. Brennintensität: extrem hoch 9, sehr hoch 8, moderat hoch 7, leicht hoch 6, weder hoch noch niedrig 5, leicht niedrig 4, moderat niedrig 3, sehr niedrig 2, extrem niedrig 1, keine Bitterkeit oder Brennen 0. Die Durchschnittswerte der Testergebnisse sind unten angegeben:
30 mg Fumarsäure 7
40 mg Fumarsäure 6
50 mg Fumarsäure 3
60 mg Fumarsäure 1 - Dieses Beispiel zeigt, daß 50 mg Fumarsäure pro Gewicht pro 100 mg an Ibuprofen bei der Verringerung des brennenden Nachgeschmacks von Ibuprofen effektiv waren.

- Verfahren zur Verringerung des brennenden Nachgeschmacks eines Propionsäurederivats, umfassend: Vermischen einer analgetisch effektiven Menge eines racemischen Propionsäurederivats und von 50 bis 150 Gewichts% von Fumarsäure, basierend auf dem Gewicht des Propionsäurederivats; wobei die Fumarsäure und das Propionsäurederivat in der Abwesenheit eines Hydrokolloids vermischt werden.
- Verfahren nach Anspruch 1, wobei das Propionsäurederivat Ibuprofen ist.
- Verfahren nach Anspruch 1 oder 2, wobei die Fumarsäure und das Propionsäurederivat in einem Granulierungsverfahren vermischt werden.
- Verfahren nach Anspruch 3, wobei das Granulierungsverfahren mit einem nichthydrokolloidalen Bindemittel durchgeführt wird.
- Zusammensetzung zur oralen Verabreichung, erhältlich durch das Verfahren nach einem der Ansprüche 1 bis 4, in Form einer Kautablette.
- Zusammensetzung zur oralen Verabreichung, erhältlich durch das Verfahren nach einem der Ansprüche 1 bis 4, in Form einer flüssigen Formulierung.
