- TECHNISCHES GEBIET
- Die vorliegende Erfindung betrifft die Verwendung von GABAB-Rezeptoragonisten zur Herstellung eines Medikaments zur Inhibierung von vorübergehenden Relaxationen des Schließmuskels in der unteren Speiseröhre, zur Behandlung von gastroösophagealem Rückfluß und/oder zur Behandlung von Reflux bei Kleinkindern.
- STAND DER TECHNIK
- Rückfluß
- Bei einigen Menschen neigt der Schließmuskel in der unteren Speiseröhre (der untere Ösophagussphinkter, lower esophageal sphincter, LES) dazu, sich häufiger als bei anderen Menschen zu entspannen. Als Folge kann Flüssigkeit aus dem Magen in die Speiseröhre gelangen, da die mechanische Sperre zu solchen Zeiten zeitweise aufgehoben ist – ein Ereignis, das im folgenden als „Rückfluß" bezeichnet wird.
- Beim gastroösophagealen Rückfluß (gastro-esophageal reflux disease, GERD) handelt es sich um die am häufigsten vorkommende Erkrankung des oberen Magen-Darm-Trakts. Die Therapie ist gegenwärtig darauf ausgerichtet, die Magensäuresekretion zu vermindern oder die Säureexposition der Speiseröhre durch Verbesserung der ösophagealen Clearance, des Tonus des unteren Ösophagussphinkters und der Entleerung des Magens zu reduzieren. Man nahm bislang an, daß Rückfluß mechanistisch hauptsächlich auf einen hypotonen unteren Ösophagussphinkter zurückzuführen ist. Neuere Untersuchungen (z. B. Holloway und Dent (1990) Gastroenterol. Clin. N. Amer. 19, 517–535) haben jedoch gezeigt, daß die meisten Rückflußepisoden während einer vorübergehenden Entspannung des unteren Ösophagussphinkters (transient lower esophageal sphincter relaxations, TLESR) auftreten, d. h. die Relaxationen nicht durch Schlucken ausgelöst werden. Es wurde weiterhin gezeigt, daß die Magensäuresekretion bei GERD-Patienten gewöhnlich normal ist.
- Es besteht daher ein Bedarf an Verbindungen, die die Häufigkeit des Auftretens von TLESR reduzieren und somit Rückfluß verhindern. Im Idealfall sollte die Verbindung eine Wirkungsdauer von ungefähr 12 h aufweisen, da Rückfluß meistens tagsüber und nach der Einnahme von Mahlzeiten auftritt.
- Eine ein auf die Inhibierung der Relaxation des unteren Ösophagussphinkters eingestelltes Lokalanästhetikum enthaltende pharmazeutische Zusammensetzung ist in WO 87/04077 und in
US 5 036 057 offenbart. - GABAB-Rezeptoragonisten
- GABA (4-Aminobuttersäure) ist ein endogener Neurotransmitter im zentralen und peripheren Nervensystem. Die Rezeptoren für GABA werden traditionell in GABAA- und GABAB-Rezeptorsubtypen eingeteilt. In Brain Res. Bull. (1995), Band 38, Nr. 6, S. 587–594 (Robert J. Washabau et al.) wurden GABAA-Rezeptoragonisten/- antagonisten beschrieben, die den Druck des unteren Ösophagussphinkters (lower esophageal sphincter pressure, LESP) und die Magensäuresekretion erhöhen. GABAB-Rezeptoren (Übersichtsartikel: siehe Kerr, D. I. B. und Ong, J. (1995), Pharmac. Ther. Band 67, S. 187–246) gehören zur Superfamilie der G-Protein-gekoppelten Rezeptoren. In Neurochem. Res (1996), Band 21, Nr. 2, S. 211–215 (Kyoji Nakajima et al.) wird die zelluläre Lokalisierung von GABAB-Rezeptoren im Magen-Darm-System sichtbar gemacht. Es wird beschrieben, daß sich GABAB-Rezeptoragonisten zur Behandlung von ZNS-Störungen wie der Muskelrelaxation bei Spinalspastizität, Herzkreis lauferkrankungen, Asthma, Störungen der Darmmotilität wie Reizkolon und als prokinetische Mittel und Antitussiva eignen. Es wurde weiterhin offenbart, daß sich GABAB-Rezeptoragonisten zur Behandlung von Erbrechen eignen (WO 96/11680).
- Der GABAB-Rezeptoragonist Baclofen (4-Amino-3-(4-chlorphenyl)buttersäure) (Schweizer Patent Nr. CH449046) ist das am besten untersuchte GABA-Analogon.
- Andere GABAB-Rezeptoragonisten oder partielle Agonisten sind in
EP 0356128 ,EP 0181833 ,EP 0399949 ,EP 0463969 und FR 2722192 offenbart. Ein Übersichtsartikel zur Chemie der GABAB-Modulatoren findet sich bei Froestl, W. und Mickel, S. J. in The GABA Receptors, S. 271–296 (Hrsg. S. J. Enna und N. G. Bowery, Humana Press Inc., Totowa, NJ, USA, 1997). - Es ist im Stand der Technik bekannt, daß sich das Screening von Arzneimitteln verbessern läßt, indem man Zellen verwendet, die mit einem geklonten Rezeptorgen transfiziert sind. Solche transfizierten Zellen können gegenüber dem traditionellen Screening eine Reihe von Vorteilen bieten, wobei der wichtigste Vorteil vermutlich die Selektivität ist. Ein weiterer Vorteil von transfizierten Zellen ist, daß sie eine einfache Abschätzung der Aktivität von Arzneimitteln an geklonten humanen Rezeptoren ermöglichen. Dadurch, daß der GABAB-Rezeptor vor kurzem kloniert wurde (Kaupmann et al., Nature 386 (6622), 239–246, 20. März 1997), bietet sich somit die Möglichkeit, spezifischere Arzneimittel mit Wirkung auf den GABAB-Rezeptor zu entwickeln. In dem besagten Artikel werden zwei Subtypen des Rezeptors aus Ratten offenbart, die als GABABR1a und GABABR1b bezeichnet werden, es wurde jedoch sehr deutlich gemacht, daß sich auch mehrere andere Subtypen isolieren lassen.
- OFFENBARUNG DER ERFINDUNG
- Überraschenderweise wurde gefunden, daß GABAB-Rezeptoragonisten zur Inhibierung von vorübergehenden Relaxationen des unteren Ösophagussphinkters und somit zur Behandlung von gastroösophagealem Rückfluß angewendet werden können.
- Infolgedessen stellt die vorliegende Erfindung die Verwendung eines GABAB-Rezeptoragonisten zur Herstellung eines Medikaments zur Inhibierung von vorübergehenden Relaxationen des Schließmuskels in der unteren Speiseröhre (transient lower esophageal sphincter relaxations, TLESR) bzw. genauer gesagt zur Behandlung von gastroösophagealem Rückfluß bereit. Für die Zwecke der Erfindung sollte der Ausdruck „Agonist" so verstanden werden, daß er sowohl volle Agonisten als auch partielle Agonisten einschließt, während „partieller Agonist" als eine Verbindung zu verstehen ist, die den GABAB-Rezeptor partiell, jedoch nicht vollständig aktivieren kann.
- Die Inhibierung von TLESR läßt auch darauf schließen, daß sich die Verbindungen zur Behandlung von Reflux bei Kleinkindern anwenden lassen. Eine wirksame Kontrolle von Reflux bei Kleinkindern wäre ein wichtiger Beitrag zur Behandlung von Lungenerkrankungen aufgrund des Einatmens von erbrochenem Mageninhalt und zur Behandlung von Entwicklungsstörungen aufgrund eines übermäßigen Verlusts aufgenommener Nährstoffe.
- In einer bevorzugten Ausführungsform der Erfindung handelt es sich bei dem GABAB-Rezeptoragonisten um ein substituiertes Aminopropylsäurederivat, bei dem die saure Kopfgruppe eine Carbonsäuregruppe, eine Phosphinsäuregruppe, eine Phosphonigsäuregruppe oder eine Sulfinsäuregruppe ist.
- Verbindungen mit agonistischer oder partieller agonistischer Affinität zu GABAB-Rezeptoren, die daher für eine erfindungsgemäße Verwendung geeignet sind, sind beispielsweise:

(3-Aminopropyl)methylphosphinsäure;
(3-Amino-2-hydroxypropyl)methylphosphinsäure;
(3-Amino-2-(4-chlorphenyl)propyl)sulfinsäure;

(3-Amino-2-oxopropyl)methylphosphinsäure
4-Amino-3-(5-chlorthien-2-yl)buttersäure;
(3-Aminopropyl)phosphonigsäure. - Die Erfindung betrifft weiterhin die Verwendung von pharmazeutisch unbedenklichen Salzen von GABAB-Liganden für die besagten Zwecke. Die meisten bekannten GABAB-Liganden wie beispielsweise Baclofen, (3-Aminopropyl)methylphosphinsäure und (3-Amino-2-(S)-hydroxypropyl)-methylphosphinsäure sind amphoter und können in Form innerer Salze vorliegen. Sie können auch Säureadditionssalze und Salze mit Basen bilden. Solche Salze sind insbesondere pharmazeutisch unbedenkliche Säureadditionssalze sowie pharmazeutisch unbedenkliche Salze, die mit Basen gebildet werden. Zu den für die Bildung solcher Salze geeigneten Säuren gehören beispielsweise Mineralsäuren wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure oder Phosphorsäure, oder organische Säuren wie organische Sulfonsäuren und organische Carbonsäuren. Salze von GABAB-Liganden mit Basen sind beispielsweise Alkalisalze, z. B. Natrium- oder Kaliumsalze, oder Erdalkalisalze, z. B. Calcium- oder Magnesiumsalze, sowie Ammoniumsalze wie die mit Ammoniak oder organischen Aminen.
- Die Erfindung umfaßt weiterhin die Verwendung optischer Isomere von GABAB-Liganden für die besagten Zwecke. Viele bekannte GABAB-Liganden wie beispielsweise Baclofen und (3-Amino-2-(S)-hydroxypropyl)methylphosphinsäure sind aufgrund des Vorhandenseins eines asymmetrischen Kohlenstoffatoms chirale Verbindungen. Je nachdem, ob asymmetrische Atome vorhanden sind oder nicht, können die GABAB-Liganden in Form von Isomerenmischungen, insbesondere als Racemate, oder in Form reiner Isomere, insbesondere Enantiomere, vorliegen.
- Gemäß einem anderen Aspekt stellt die Erfindung ein Verfahren zur Inhibierung von vorübergehenden Relaxationen des Schließmuskels in der unteren Speiseröhre bereit, bei dem man einem einer solchen Behandlung bedürftigen Säugetier einschließlich des Menschen eine wirksame Menge eines wie oben definierten GABAB-Rezeptoragonisten verabreicht.
- Die Erfindung schließt auch eine Verwendung zur Herstellung einer pharmazeutischen Zusammensetzung zur Verwendung bei der Inhibierung von vorübergehenden Relaxationen des Schließmuskels in der unteren Speiseröhre ein. Genauer gesagt eignet sich die pharmazeutische Zusammensetzung zur Behandlung von gastroösophagealem Rückfluß und/oder zur Behandlung von Reflux bei Kleinkindern. Bei dem Wirkstoff in der pharmazeutischen Zusammensetzung kann es sich um einen beliebigen der oben definierten GABAB-Rezeptoragonisten handeln.
- Tagesdosis
- Bei der Verwendung als Inhibitor von TLESR und als Rückflußinhibitor kann man den GABAB-Rezeptoragonisten in Dosierungen anwenden, die sich für andere Leiden eignen, von denen man weiß, daß bei ihnen GABAB-Rezeptoragonisten von Nutzen sind. Die typische Tagesdosis des Wirkstoffs schwankt in einem weiten Bereich und hängt von verschiedenen Faktoren wie beispielsweise den individuellen Anforderungen des einzelnen Patienten und dem Verabreichungsweg ab. Im allgemeinen liegen die Dosierungen im Bereich von 1 μg bis 100 mg pro Tag und kg Körpergewicht, vorzugsweise von 10 μg bis 10 mg pro Tag und kg Körpergewicht.
- Pharmazeutische Formulierungen
- Für den klinischen Gebrauch werden die erfindungsgemäßen Verbindungen zu pharmazeutischen Formulierungen für die orale, rektale, parenterale oder eine andersartige Verabreichung formuliert. Die pharmazeutische Formulierung enthält eine erfindungsgemäße Verbindung in Kombination mit einem oder mehreren pharmazeutisch unbedenklichen Bestandteilen. Der Träger kann in Form eines festen, halbfesten oder flüssigen Verdünnungsmittels oder einer Kapsel vorliegen. Diese pharmazeutischen Zubereitungen sind ein weiterer Gegenstand der Erfindung. Gewöhnlich beträgt die Menge an Wirkstoff zwischen 0,1 und 95 Gew.-% der Zubereitung, vorzugsweise zwischen 0,2 und 20 Gew.-% bei Zubereitungen zur parenteralen Verabreichung und vorzugsweise zwischen 1 und 50 Gew.-% bei Zubereitungen zur oralen Verabreichung.
- Bei der Herstellung von pharmazeutischen Formulierungen, die eine Verbindung der vorliegenden Erfindung in Form von Dosierungseinheiten zur oralen Verabreichung enthalten, kann die ausgewählte Verbindung mit festen, gepulverten Bestandteilen wie Lactose, Saccharose, Sorbit, Mannit, Stärke, Amylopektin, Cellulosederivaten, Gelatine oder einem anderen geeigneten Bestandteil sowie mit Sprengmitteln und Gleitmitteln wie Magnesiumstearat, Calciumstearat, Natriumstearylfumarat und Polyethylenglykolwachsen gemischt werden. Die Mischung wird dann zu einem Granulat verarbeitet oder zu Tabletten verpreßt.
- Man kann weiche Gelatinekapseln herstellen, wobei die Kapseln eine Mischung aus erfindungsgemäßem Wirkstoff bzw. erfindungsgemäßen Wirkstoffen, Pflanzenöl, Fett oder einem anderen für weiche Gelatinekapseln geeigneten Vehikel enthalten. Harte Gelatinekapseln können ein Wirkstoffgranulat enthalten. Es ist auch möglich, daß harte Gelatinekapseln den Wirkstoff in Kombination mit festen, gepulverten Bestandteilen wie Lactose, Saccharose, Sorbit, Mannit, Kartoffelstärke, Maisstärke, Amylopektin, Cellulosederivaten oder Gelatine enthalten.
- Dosierungseinheiten für eine rektale Verabreichung lassen sich (i) in Form von Zäpfchen, die die Wirksubstanz gemischt mit einer neutralen Fettgrundlage enthalten, (ii) in Form von Gelatine-Rektalkapseln, die die Wirksubstanz in einer Mischung mit einem Pflanzenöl, Paraffinöl oder einem anderen für Gelatine-Rektalkapseln geeigneten Vehikel enthalten, (iii) in Form eines fertigen Mikroeinlaufs oder (iv) in Form einer trockenen Mikroeinlaufsformulierung, die unmittelbar vor der Verabreichung in einem geeigneten Lösungsmittel rekonstituiert wird, herstellen.
- Flüssige Zubereitungen für die orale Verabreichung können in Form von Sirupen oder Suspensionen hergestellt werden, z. B. Lösungen oder Suspensionen, die von 0,2 Gew.-% bis 20 Gew.-% Wirkstoff enthalten, wobei der Rest aus Zucker oder Zuckeralkoholen und einer Mischung von Ethanol, Wasser, Glycerin, Propylenglykol und Polyethylenglykol besteht. Falls gewünscht können solche flüssigen Zubereitungen Farbstoffe, Geschmackstoffe, Saccharin und Carboxymethylcellulose oder ein anderes Verdickungsmittel enthalten. Flüssige Zubereitungen für die orale Verabreichung können auch in Form eines Trockenpulvers hergestellt werden, das vor der Anwendung mit einem geeigneten Lösungsmittel rekonstituiert wird.
- Lösungen für eine parenterale Verabreichung können als eine Lösung einer erfindungsgemäßen Verbindung in einem pharmazeutisch unbedenklichen Lösungsmittel hergestellt werden, vorzugsweise in einer Konzentration von 0,1 Gew.-% bis 10 Gew.-%. Diese Lösungen können weiterhin stabilisierende Bestandteile und/oder puffernde Bestandteile enthalten und werden in Einheitsdosen in Form von Ampullen bzw. Vials abgegeben. Lösungen zur parenteralen Verabreichung können auch als Trockenzubereitungen hergestellt werden, die vor der Anwendung ad hoc mit einem geeigneten Lösungsmittel rekonstituiert werden.
- BEISPIELE
- Materialien und Methoden
- Es wurden ausgewachsene Labrador-Retriever beiderlei Geschlechts (5 männlich, 3 weiblich) mit einem Gewicht von 20–30 kg verwendet. Um eine Intubierung zu ermöglichen, wurde eine zervikale Ösophagostomie gebildet. Nach einer Erholungszeit wurden die Hunde Kontrollexperimenten unterzogen, bis eine stabile und verläßliche Kontrollreaktion erzielt wurde. Die Hunde wurden in anderen Experimenten eingesetzt, bevor ihnen die GABAB-Rezeptoragonisten verabreicht wurden, es wurde jedoch im jedem Fall eine Auswaschphase von wenigstens zwei Tagen eingehalten, während der keine Arzneimittel verabreicht wurden. Bei jedem vierten Experiment mit einem Hund handelte es sich um ein Kontrollexperiment, wodurch die Stabilität und Reproduzierbarkeit der Ergebnisse sichergestellt wurde.
- Mit den Experimenten wurde um etwa 8 Uhr morgens begonnen, wobei die Tiere zu diesem Zeitpunkt ungefähr 17 Stunden lang keine Nahrung zu sich genommen hatten. Der Hund wurde in einen Pawlow-Stand gestellt, an den er zuvor gewöhnt worden war. Zum Aufzeichnen des Schluckens wurde ein dünner Silikonkatheter mit einer distalen Öffnung retrograd durch die Ösophagostomie in den Rachen eingeführt. Der Katheter wurde mit Luft (ungefähr 2 ml/min) perfundiert. Eine Multilumenanordnung wurde so positioniert, daß die Drücke im proximalen Magen, im LES und an vier Stellen im distalen Ösophagus aufgezeichnet werden konnten. Die Anordnung wurde mit einer 6 cm langen Hülle umgeben, um eine verläßliche Messung des Drucks im LES zu ermöglichen. Magen- und LES-Kanäle sowie die Kanäle in der Speiseröhre wurden mit destilliertem, entgasten Wasser perfundiert (0,45 ml/min bzw. 0,1 ml/min). Optimale Perfusionsbedingungen ließen sich mit einer „low compliance" pneumohydraulischen Pumpe erzielen. Die Drücke im Lumen wurden mit externen Druckaufnehmern gemessen. Die Signale wurden verstärkt und mit LabWindows/CVI-Software (Version 3.1) aufgezeichnet. Druckeichungen wurden bei 0 und 100 mm Hg durchgeführt, wobei die Multilumenanordnungen auf einer Ebene etwas unterhalb der Druckaufnehmer zu liegen kamen. Der Hund wurde so positioniert, daß der durchschnittliche Mageninnendruck ungefähr 0 mm Hg betrug. 3 cm oberhalb der oberen Grenze des LES wurde eine Antimon-pH-Elektrode angebracht, und die Signale wurden wie oben beschrieben aufgenommen. Die aufgenommenen Signale wurden mit LabWindows-Software ausgewertet.
- Nach einer wenigstens 10minütigen Baseline-Messung wurde Vehikel (0,9% NaCl; 0,5 ml/kg) bzw. ein GABAB-Rezeptoragonist i. v. im Verlauf von 2 min verabreicht. Zehn min nach Ende der Verabreichung wurde über die Multilumenanordnung mit einer Geschwindigkeit von 100 ml/min (30 ml/kg) Nährstoff infundiert. GABA wurde als i. v.-Dauerinfusion verabreicht, mit der 10 min vor der Nährstoffverabreichung begonnen wurde; und in einigen Fällen wurde 30 min vor dem Nährstoff intragastrisch RS-Baclofen gegeben. Der Nährstoff enthielt 10% Pepton (w/v), 5% Intralipid (v/v) und 5% D-Glucose (w/v) und wurde mit HCl auf einen pH-Wert von 3,0 angesäuert. Unmittelbar nach der Infusion wurde Luft zu 40 ml/min bis zu einer Gesamtzeitspanne von 90 min vom Beginn der Nährstoffinfusion eingeblasen. Dann wurde der Hund extubiert und eine Baseline-Messung durchgeführt, um sicherzustellen, daß es zu keiner Verschiebung gekommen war.
- Ein TLESR wurde gemäß der folgenden Kriterien identifiziert: der Unterschied zwischen LES- und i. g.-Drücken betrug weniger als 2 mm Hg, die Dauer betrug mehr als 0,5 s und die Entspannung wurde nicht durch primäre Peristalsis (d. h. durch ein pharyngeales Signal) ausgelöst. Die Geschwindigkeit des Druckabfalls betrug mehr als 10 mm Hg/s. Die meisten, jedoch nicht alle, TLESR konnten auch akustisch durch ein charakteristisches Geräusch aus der Speiseröhre (d. h. Rülpsen) wahrgenommen werden. Es wurde die Gesamtzahl an TLESR für die erste 45-Minuten-Periode und für das gesamte Experiment (90 min) berechnet. Die Wirkung der GABAB-Rezeptoragonisten wurde relativ zu den einzelnen Kontrolldaten (n ≥ 5) ausgedrückt. Jeder Agonist wurde in wenigstens zwei verschiedenen Hunden getestet.
- Ergebnisse und Diskussion
- TLESR trat praktisch nie in nüchternem Zustand auf, sondern immer nach Infusion von Nährstoffen und Einblasen von Luft. Die Häufigkeit des Auftretens von TLESR schwankte zwischen den einzelnen Hunden beträchtlich, die intraindividuelle Variation war jedoch gering.
- GABAB-Rezeptoragonisten reduzierten die Häufigkeit des Auftretens von TLESR in dosisabhängiger Weise. Die Inhibierung bei 45 min lag über der für die gesamte Dauer des Experiments (90 min) berechneten Inhibierung. Da eine Ausdehnung des Magens der Hauptstimulus für TLESR ist, ist der 45-min-Wert eindeutig der bei der Verabreichung von inhibierenden Verbindungen relevanteste Meßwert: eine Reduktion in TLESR führt zu einer erhöhten gasinduzierten Ausdehnung des Magens und somit zu einem neuen Schwellenwert. Dieser verwirrende Effekt ist zu Beginn des Experiments weniger stark ausgeprägt (siehe unten).
- Die hemmende Wirkung von GABAB-Rezeptoragonisten auf TLESR wurde festgestellt, ohne daß es zu verhaltensbezogenen Nebenwirkungen kam, außer bei der hohen R,S-Baclofendosis, bei der es zu einer gewissen Sedation kam, die ungefähr eine Stunde nach der Verabreichung wieder verschwand.
- In Kontrollexperimenten stieg der i. g.-Druck während der Infusion von Nährstoffen/des Einblasens von Luft von 0 auf etwa 4 mm Hg an. Dosen an GABAB-Rezeptoragonisten, bei denen es zu einer nahezu vollständigen TLESR-Inhibierung kam, wurden von größeren Zunahmen im i. g.-Druck begleitet (10–13 mm Hg). Bei solch hohen i. g.-Drücken kam es gelegentlich zu einem Erbrechen am Ende des Experiments. Dies rührt davon her, daß die Hunde bei Eindämmung von TLESR nicht dazu in der Lage sind, Gas aus dem Magen abzulassen.
- Aus den Ergebnissen (Tabelle 1) geht hervor, daß GABAB-Rezeptoragonisten das Auftreten von TLESR nach einer flüssigen Mahlzeit mit darauf folgendem Einblasen von Luft inhibieren. Dieser Effekt ist nicht sekundär zu Sedierung oder Somnolenz. Die Schlußfolgerung ist, daß Verbindungen mit Affinität zu GABAB-Rezeptoren sich als therapeutische Mittel bei der Behandlung von gastroösophagealem Rückfluß eignen können.

- Verwendung nach einem der Ansprüche 1 bis 3, wobei es sich bei dem GABAB-Rezeptoragonisten um 4-Amino-3-(4-chlorphenyl)buttersäure (Baclofen) handelt.
- Verwendung nach einem der Ansprüche 1 bis 3, wobei es sich bei dem GABAB-Rezeptoragonisten um (3-Aminopropyl)methylphosphinsäure handelt.
- Verwendung nach einem der Ansprüche 1 bis 3, wobei es sich bei dem GABAB-Rezeptoragonisten um (3-Amino-2-hydroxypropyl)methylphosphinsäure handelt.
- Verwendung nach einem der Ansprüche 1 bis 3, wobei es sich bei dem GABAB-Rezeptoragonisten um (3-Amino-2-(4-chlorphenyl)propyl)sulfinsäure handelt.
