Description
- Bereich der Erfindung
- Die vorliegende Erfindung betrifft Verbindungen zur Verabreichung aktiver Mittel und insbesondere biologisch aktiver Mittel wie beispielsweise bioaktiver Peptide und dergleichen. Diese Verbindungen werden verwendet als Träger zur Erleichterung der Verabreichung einer Nutzverbindung an ein Zielorgan. Die Träger sind modifizierte Aminosäuren und sind gut geeignet zur Bildung nicht-kovalenter Mischungen mit biologisch aktiven Mitteln zur oralen Verabreichung an Tiere. Es werden auch Zusammensetzungen, die diese Verbindungen umfassen, Verfahren zur Herstellung solcher Zusammensetzungen und Dosis-Einheiten offenbart.
- Hintergrund der Erfindung
- Herkömmliche Mittel zur Verabreichung aktiver Mittel sind oft stark beschränkt durch biologische, chemische und physikalische Barrieren. Typischerweise werden diese Barrieren auferlegt durch die Umgebung, durch die hindurch eine Verabreichung erfolgt, durch die Umgebung des für die Verabreichung angesteuerten Zielorgans oder durch das Zielorgan selbst.
- Biologisch aktive Mittel sind besonders anfällig für solche Barrieren. Beispielsweise werden bei der Verabreichung pharmakologischer und therapeutischer Mittel an Tiere Barrieren durch den Körper auferlegt. Beispiele physikalischer Barrieren sind die Haut und verschiedene Membranen von Organen, die durchquert werden müssen, bevor ein Target erreicht wird. Chemische Barrieren schließen ein, sind jedoch nicht beschränkt auf Änderungen des pH-Wertes, Lipid-Doppelschichten und abbauende Enzyme.
- Diese Barrieren sind von besonderer Signifikanz beim Entwurf oraler Verabreichungssysteme. Eine orale Verabreichung vieler biologisch aktiver Mittel wäre der Weg der Wahl für eine Verabreichung an Tiere, wenn da nicht biologische, chemische und physikalische Barrieren vorhanden wären, wie beispielsweise ein schwankender pH-Wert im Gastrointestinal-(GI-)Trakt, potente Verdauungsenzyme und für aktive Mittel undurchlässige Gastrointestinal-Membranen. Von den zahlreichen Mitteln, die nicht typisch für eine orale Verabreichung zugänglich sind, sind biologisch aktive Peptide wie beispielsweise Calcitonin und Insulin, Polysaccharide und insbesondere Mucopolysaccharide, die einschließen, jedoch nicht beschränkt sind auf Heparin, Heparinoide, Antibiotika und andere organische Substanzen. Diese Mittel werden im Gastrointestinaltrakt durch saure Hydrolyse, Enzyme oder dergleichen schnell unwirksam gemacht oder werden zerstört.
- Frühere Verfahrensweisen zur oralen Verabreichung empfindlicher pharmakologischer Mittel bauten auf die Mitverabreichung von Hilfsstoffen (z. B. Resorcinole und nichtionische Tenside wie beispielsweise Polyoxyethylenoleylether und n-Hexadecylpolyethylenether), um künstlich die Permeabilität der Darmwände zu erhöhen, sowie auf die Mitverabreichung enzymatischer Inhibitoren (z. B. Pankreas-Trypsin-Inhibitoren, Diisopropylfluorphosphat (DFF) und Trasylol) zum Inhibieren eines enzymatischen Abbaus.
- Es wurden auch Liposome als Arzneimittel-Verabreichungssysteme für Insulin und Heparin beschrieben; verwiesen wird beispielsweise auf das US-Patent Nr. 4,239,754; auf die Druckschrift „Patel et al. (1976), FEBS Letters, Band 62, Seite 60“; und auf die Druckschrift "Hashimoto et al. (1979), Endocrinology Japan, Band 26, Seite 337".
- Jedoch ist ein sich über ein breites Spektrum erstreckender Gebrauch derartiger Arzneimittel-Verabreichungssysteme ausgeschlossen, da (1) die Systeme toxische Mengen an Hilfsstoffen oder Inhibitoren erfordern; (2) geeignete Nutzverbindungen mit niedrigem Molekulargewicht, d. h. aktive Mittel, nicht verfügbar sind; (3) die Systeme eine geringe Stabilität und eine inadäquate Lagerbeständigkeit zeigen; (4) die Systeme schwierig herzustellen sind; (5) die Systeme das aktive Mittel (Nutzverbindung) nicht zu schützen vermögen; (6) die Systeme in nachteiliger Weise das aktive Mittel verän dern; oder (7) Systeme eine Absorption des aktiven Mittels nicht zuzulassen oder zu fördern vermögen.
- In noch jüngerer Zeit wurden Mikrosphären aus künstlichen Polymeren gemischter Aminosäuren (Proteinoide) zur Verabreichung pharmazeutischer Substanzen verwendet. Beispielsweise beschreibt das US-Patent Nr. 4,925,673 Arzneimittel enthaltende Proteinoid-Mikrosphären-Träger sowie Verfahren zu deren Herstellung und Verwendung. Diese Proteinoid-Mikrosphären sind nützlich für die Verabreichung einer Anzahl aktiver Mittel.
- Die Druckschrift EP-A 0 226 223 betrifft Benzoesäure- und Benzoesäureester-Derivate, die entzündungshemmende und analgetische Wirksamkeit aufweisen.
- Die Druckschrift WO-A94/23767 betrifft modifizierte Aminosäuren zur Verwendung als Verabreichungssystem empfindlicher Mittel wie beispielsweise bioaktiver Produkte.
- Der STN-Datenbank-Eintrag RN=70204-54-5 offenbart 4-[(2-Chlorbenzoyl-)amino-] benzoesäure.
- Der STN-Datenbank-Eintrag RN=73548-12-6 offenbart 4-[(Phenylacetyl-)amino-] benzoesäure.
- Es besteht nach wie vor ein Bedarf in diesem technischen Bereich für einfache, preiswerte Verabreichungssysteme, die sich leicht herstellen lassen und die einen großen Bereich aktiver Mittel verabreichen können.
- Zusammenfassung der Erfindung

- Weiter werden Polyaminosäuren bereitgestellt, die aus Peptiden oder zwei oder mehreren Aminosäuren bestehen, die durch eine Ester- oder Anhydrid-Bindung verknüpft sind und wenigstens eine Verbindung enthalten, die gewählt ist aus der Gruppe, die besteht aus den Verbindungen I bis XIII, XV bis XXXIV, XXXVI bis XXXIX ODER XLI bis XLV, wie sie oben angegeben wurden, oder den nachfolgend gezeigten Verbindungen XIV, XXXV oder XL: oder deren Salze.
- Die vorliegende Erfindung stellt weiter eine Zusammensetzung bereit, die umfaßt: (a) ein aktives Mittel; und (b) irgendeine der Verbindungen I bis XLV, wie sie oben definiert wurden, oder deren Salze. Die vorliegende Erfindung stellt auch eine Zusammensetzung bereit, die umfaßt: (a) ein aktives Mittel; und (b) eine Polyaminosäure, wie sie oben definiert wurde. In einer bevorzugten Ausführungsform umfaßt die Polyaminosäure ein Peptid. In einer bevorzugten Ausführungsform umfaßt das aktive Mittel ein biologisch aktives Mittel. In einer anderen bevorzugten Ausführungsform ist das biologisch aktive Mittel der Zusammensetzung, wie sie vorstehend erklärt wurde, gewählt aus der Gruppe, die besteht aus einem Peptid, einem Mucopolysaccharid, einem Kohlenhydrat, einem Lipid, einem Pestizid oder irgendeiner Kombination daraus. Noch mehr bevorzugt ist das biologisch aktive Mittel gewählt aus der Gruppe, die besteht aus menschlichem Wachstumshormon, Rinderwachstumshormon, Wachstumshormon freisetzendem Hormon (growth hormone-releasing hormone), einem Interferon, Interleukin-II, Insulin, Heparin, Calcitonin, Erythropoietin, atrionatriuretischem Faktor, einem Antigen, einem monoklonalen Antikörper, Somatostatin, Adrenocorticotropin, Gonadotropin freisetzendem Hormon (gonadotropin releasing hormone), Oxytocin, Vasopressin, Cromolynnatrium, Vancomycin, Desferrioxamin (DFO), antimikrobiellen Mitteln oder irgendeiner Kombination daraus.
- Ebenfalls bereitgestellt wird eine Dosiseinheitsform, die umfaßt: (A) eine Zusammensetzung, wie sie oben erläutert wurde; und (B)(a) einen Träger, (b) ein Verdünnungsmittel, (c) ein Zerfallsmittel, (d) ein Gleitmittel, (e) einen Weichmacher, (f) ein Färbemittel, (g) ein Dosier-Bindemittel, oder (h) irgendeine Kombination daraus. Vorzugsweise umfaßt die Dosiseinheit eine Tablette, eine Kapsel oder eine Flüssigkeit.
- Die vorliegende Erfindung stellt weiter die Verwendung der wie oben definierten Verbindung und einer Verbindung bereit, die gewählt ist aus der Gruppe, die besteht aus den Verbindungen XIV, XXXV und XL, und deren Salzen, und eines biologischen aktiven Mittels, für die Herstellung einer Zusammensetzung für die orale Verabreichung.
- Auch bereitgestellt wird die Verwendung der Polyaminosäure, wie sie oben erklärt wurde, und eines biologisch aktiven Mittels zur Herstellung einer Zusammensetzung für die orale Verabreichung. Weiter stellt die vorliegende Erfindung die Verwendung der Polyaminosäure, wie sie oben erläutert wurde, und die ein Peptid und ein biologisch aktives Mittel umfaßt, zur Herstellung einer Zusammensetzung für eine orale Verabreichung bereit.
- Von der vorliegenden Erfindung ebenfalls umfaßt ist ein Verfahren zur Herstellung einer Zusammensetzung, wobei das Verfahren umfaßt ein Mischen von (A) wenigstens einem biologisch aktiven Mittel; (B) wenigstens einer Verbindung, wie sie oben definiert wurde, oder einer Verbindung, die gewählt ist aus der Gruppe, die besteht aus den Verbindungen XIV, XXXV und XL und deren Salzen; und (C) gegebenenfalls eines Dosier-Bindemittels.
- Weiter wird ein Verfahren zur Herstellung einer Zusammensetzung bereitgestellt, wobei das Verfahren umfaßt ein Mischen (A) wenigstens eines biologisch aktiven Mittels; (B) wenigstens einer Polyaminosäure, wie sie oben definiert wurde; und (C) gegebenenfalls eines Dosier-Bindemittels.
- Letzten Endes wird auch ein Verfahren zur Herstellung einer Zusammensetzung bereitgestellt, wobei das Verfahren umfaßt ein Mischen (A) wenigstens eines biologisch aktiven Mittels; (B) wenigstens eines Peptids, wie es oben definiert wurde; und (C) gegebenenfalls eines Dosier-Bindemittels.
- Detaillierte Beschreibung der Erfindung
- Die speziellen Verbindungen der vorliegenden Erfindung oder deren Salze wie beispielsweise Natriumsalze, können verwendet werden, um verschiedene aktive Mittel durch verschiedene biologische, chemische und physikalische Barrieren hindurch zu verabreichen. Diese Verbindungen sind besonders geeignet zum Verabreichen aktiver Mittel, die einem Abbau durch Umgebungseinflüsse unterliegen. Die Verbindungen und Zusammensetzungen der vorliegenden Erfindung sind besonders nützlich zum Zuleiten oder Verabreichen biologisch aktiver Mittel an Tiere wie beispielsweise Vögel, Säuger wie beispielsweise Primaten und insbesondere Menschen und Insekten.
- Andere Vorteile der vorliegenden Erfindung schließen die Verwendung von leicht herzustellenden, preiswerten Ausgangsstoffen ein. Die Zusammensetzungen und die Formulierungsverfahren der vorliegenden Erfindung sind kostensparend, einfach durchzuführen und sind zugänglich für eine Umstellung auf die Herstellung im industriellen Maßstab zur kommerziellen Produktion.
- Aminosäuren, Polyaminosäuren und Peptide in modifizierter Form können zum Zuleiten aktiver Mittel verwendet werden, die einschließen, jedoch nicht beschränkt sind auf biologisch aktive Mittel wie beispielsweise pharmakologische und therapeutische Mittel.
- Eine Aminosäure ist irgendeine Carbonsäure, die wenigstens eine freie Aminogruppe aufweist, und der Begriff schließt natürlich vorkommende und synthetische Aminosäuren ein.
- Polyaminosäuren sind entweder Peptide oder zwei oder mehrere Aminosäuren, die über eine Bindung verknüpft sind, die von anderen Gruppen gebildet wird, die verknüpft werden können, beispielsweise ein Ester, ein Anhydrid oder eine Anhydrid-Bindung. Speziell erwähnt werden nicht-natürlich vorkommende Polyaminosäuren und speziell nicht-natürlich vorkommende Heteropolyaminosäuren, d. h. Polymere gemischter Aminosäuren.
- Peptide sind zwei oder mehr Aminosäuren, die über eine Peptid-Bindung verbunden sind. Peptide können hinsichtlich ihrer Länge von Dipeptiden mit zwei Aminosäuren zu Polypeptiden mit einigen hundert Aminosäuren schwanken. Verwiesen wird auf die Druckschrift "Chambers Biological Dictionary, Herausgeber Peter M. B. Walker, Cambridge, England: Chambers Cambridge (1989), Seite 215". Speziell erwähnt werden Dipeptide, Tripeptide, Tetrapeptide und Pentapeptide.
- Die Begriffe "modifizierte Aminosäuren", "modifizierte Polyaminosäuren" und "modifizierte Peptide" sollen – so ist es beabsichtigt – Aminosäuren einschließen, die modifi ziert wurden, oder Polyaminosäuren und Peptide einschließen, in denen wenigstens eine Aminosäure modifiziert wurde durch Acylieren wenigstens einer freien Aminogruppe mit einem Acylierungsmittel, das mit wenigstens einer der freien Aminogruppen, die im Molekül vorhanden sind, reagiert.
- Die modifizierten Aminosäuren der Formeln I bis XLV können hergestellt werden durch Umsetzen einzelner Aminosäuren, Mischungen von zwei oder mehr Aminosäuren, Aminosäureestern oder Aminosäureamiden mit einem ein Amin modifizierenden Mittel, das mit freien Amino-Einheiten, die in den Aminosäuren zugegen sind, unter Bildung von Amiden reagiert.
- Modifizierte Aminosäuren werden typischerweise hergestellt durch Modifizieren der Aminosäuren oder eines Esters davon. Viele dieser Verbindungen werden hergestellt durch Acylierung mit Acylierungsmitteln, die die folgende Formel aufweisen: worin R10 der passende Rest zum Erhalt der Modifikation ist, die in dem Endprodukt angegeben ist, wie es innerhalb des Fachwissens ist, das auf der vorliegenden detaillierten Offenbarung basiert, und Y die Abgangsgruppe ist.
- Typische Abgangsgruppen schließen ein, sind jedoch nicht beschränkt auf Halogene, wie beispielsweise Chlor, Brom und Iod. Darüber hinaus sind die entsprechenden Anhydride geeignete Acylierungsmittel.
- Viele der Verbindungen der vorliegenden Erfindung können in einfacher Weise hergestellt und durch Verfahren modifiziert werden, die innerhalb des Fachwissens eines Fachmanns in diesem technischen Bereich liegen und auf der vorliegenden Offenbarung aufbauen. Beispielsweise können die oben angegebenen modifizierten Aminosäure-Verbindungen hergestellt werden durch Umsetzen der einzelnen Aminosäuren mit dem passenden Acylierungsmittel oder einem ein Amin modifizierenden Mittel, das mit der freien Amino-Einheit, die in den Aminosäuren zugegen ist, unter Bildung von Amiden reagiert. Schutzgruppen können verwendet werden, um unerwünschte Nebenreaktionen zu vermeiden, wie es für Fachleute mit Sachverstand in diesem technischen Bereich bekannt ist.
- Beispielsweise kann die Aminosäure in einer wäßrigen alkalischen Lösung eines Metallhydroxids, z. B. Natrium- oder Kaliumhydroxid, gelöst werden und auf eine Temperatur erhitzt werden, die im Bereich zwischen etwa 5°C und etwa 70°C liegt, vorzugsweise zwischen etwa 10°C und etwa 40°C, und zwar für eine Zeitdauer, die im Bereich zwischen etwa 1 h und etwa 4 h liegt, vorzugsweise etwa 2,5 h. Die Menge an eingesetztem Alkali pro Äquivalent pro NH2-Gruppen in der Aminosäure liegt allgemein zwischen etwa 1,25 und etwa 3 mMol, vorzugsweise zwischen etwa 1,5 und etwa 2,25 mMol, pro Äquivalent NH2. Der pH-Wert der Lösung liegt allgemein im Bereich zwischen etwa 8 und etwa 13, vorzugsweise im Bereich zwischen etwa 10 und etwa 12.
- Danach wird das passende, ein Amin modifizierende Mittel der Aminosäure-Lösung unter Rühren zugesetzt. Die Temperatur der Mischung wird bei einer Temperatur gehalten, die allgemein im Bereich zwischen etwa 5°C und etwa 70°C liegt, vorzugsweise zwischen etwa 10°C und etwa 40°C, und zwar für einen Zeitraum, der im Bereich zwischen etwa 1 und etwa 4 h liegt. Die Menge an Aminosäure modifizierendem Mittel, die in Relation zu der Menge an Aminosäure verwendet wird, basiert auf der Zahl der Mole an gesamtem freiem NH2 in der Aminosäure. Allgemein wird das ein Amin modifizierende Mittel eingesetzt in einer Menge, die im Bereich zwischen etwa 0,5 und etwa 2,5 Moläquivalenten liegt, vorzugsweise zwischen etwa 0,75 und etwa 1,25 Äquivalenten, pro molarem Äquivalent an Gesamt-NH2-Gruppen in der Aminosäure.
- Die Reaktion wird abgeschreckt durch Einstellen des pH-Wertes der Mischung mit einer geeigneten Säure, z. B. konzentrierter Chlorwasserstoffsäure, bis der pH-Wert einen Wert zwischen etwa 2 und etwa 3 erreicht. Die Mischung trennt sich bei Stehen bei Raumtemperatur unter Bildung einer transparenten oberen Schicht und eines weißen oder weißlichen Niederschlages. Die obere Schicht wird verworfen, und die modifizierte Aminosäure wird aus der unteren Schicht durch Filtration oder Dekantieren gewonnen. Die rohe modifizierte Aminosäure wird dann in Wasser bei einem pH-Wert gelöst, der zwischen etwa 9 und etwa 13 liegt, vorzugsweise zwischen etwa 11 und etwa 13. Unlösliche Materialien werden durch Filtration entfernt, und das Filtrat wird in Vakuum getrocknet. Die Ausbeute an modifizierter Aminosäure liegt allgemein im Bereich zwischen etwa 30 und etwa 60% und üblicherweise bei etwa 45%.
- Sofern erwünscht, können Aminosäureester, wie beispielsweise Methyl- oder Ethylester von Aminosäure-Verbindungen zur Herstellung der modifizierten Aminosäuren gemäß der Erfindung verwendet werden. Der Aminosäureester, gelöst in einem geeigneten organischen Lösungsmittel wie beispielsweise Dimethylformamid oder Pyridin, wird mit dem passenden ein Amin modifizierenden Mittel bei einer Temperatur umgesetzt, die im Bereich zwischen etwa 5°C und etwa 70°C liegt, vorzugsweise bei etwa 25°C, und zwar für eine Zeitdauer, die im Bereich zwischen etwa 7 und etwa 24 h liegt. Die Menge an ein Amin modifizierendem Mittel, das relativ zu dem Aminosäureester verwendet wird, ist dieselbe wie diejenige, die oben für Aminosäuren beschrieben wurde.
- Danach wird das Reaktionslösungsmittels unter negativem Druck entfernt, und die funktionelle Ester-Gruppe wird entfernt durch Hydrolysieren des modifizierten Aminosäure-Esters mit einer geeigneten alkalischen Lösung, z. B. 1N Natriumhydroxid, und zwar bei einer Temperatur, die im Bereich zwischen etwa 50°C und etwa 80°C liegt, vorzugsweise bei etwa 70°C, und für eine Zeitdauer, die ausreichend ist zum Abhydrolysieren der Estergruppe und Bilden der modifizierten Aminosäure, die eine freie Carboxylgruppe aufweist. Die Hydrolyse-Mischung wird dann auf Raumtemperatur abgekühlt und angesäuert, z. B. mit wäßriger 25%-iger Chlorwasserstoffsäure-Lösung, bis auf einen pH-Wert, der im Bereich zwischen etwa 2 und etwa 2,5 liegt. Die modifizierte Aminosäure fällt aus der Lösung aus und wird durch herkömmliche Mittel wie beispielsweise Filtration oder Dekantieren gewonnen.
- Die modifizierte Aminosäure kann gereinigt werden durch Umkristallisieren oder durch Fraktionieren an festen Säulenträgern. Geeignete Umkristallisations-Lösungsmittel-Systeme schließen Acetonitril, Methanol und Tetrahydrofuran ein. Ein Fraktionieren kann durchgeführt werden an einem geeigneten festen Säulenträger, wie beispielsweise Aluminiumoxid, wobei man Mischungen aus Methanol/n-Propanol als mobile Phase verwendet, Umkehrphasen-Säulenträgern unter Verwendung von Trifluoressigsäure/Acetonitril-Mischungen als mobile Phase und Ionenaustauschchromatographie unter Verwendung von Wasser als mobile Phase. Wenn eine Anionenaustauschchromatographie durchgeführt wird, wird vorzugsweise ein anschließender Natriumchlorid-Gradient im Konzentrationsbereich von 0 bis 500 mM eingesetzt.
- Aktive Mittel
- Aktive Mittel, die zur Verwendung im Rahmen der vorliegenden Erfindung geeignet sind, schließen biologisch aktive Mittel, chemisch aktive Mittel, die einschließen, jedoch nicht beschränkt sind auf Duftstoffe, sowie andere aktive Mittel wie beispielsweise kosmetische Mittel ein.
- Biologisch aktive Mittel schließen ein, sind jedoch nicht beschränkt auf Pestizide, pharmakologische Mittel und therapeutische Mittel. Beispielsweise schließen biologisch aktive Mittel, die zur Verwendung im Rahmen der vorliegenden Erfindung geeignet sind, ein, sind jedoch nicht beschränkt auf Peptide und insbesondere kleine Peptide; Hormone und insbesondere Hormone, die selbst gar nicht oder nur sehr langsam durch die Gastrointestinal-Schleimhaut hindurchtreten und/oder einer chemischen Spaltung durch Säuren und Enzyme im Gastrointestinaltrakt unterliegen; Polysaccharide und insbesondere Mischungen aus Mucopolysacchariden; Kohlenhydrate; Lipide oder irgendeine Kombination daraus. Weitere Beispiele schließen ein, sind jedoch nicht beschränkt auf menschliche Wachstumshormone; Rinderwachstumshormone, Wachstumshormon freisetzende Hormone, Interferone, Interleukin-II, Insulin, Heparin und insbesondere Heparin mit niedrigem Molekulargewicht, Calcitonin, Erythropoietin, atrionatriuretischer Faktor, Antigene, monoklonale Antikörper, Somatostatin, Adrenocorticotropin, Gonadotropin freisetzendes Hormon, Oxytocin, Vasopressin, Cromolynnatrium (Natri um- oder Dinatrium-Chromoglycat), Vancomycin, Desferrioxamin (DFO), antimikrobielle Mittel, die einschließen, jedoch nicht beschränkt sind auf Anti-Pilz-Mittel, oder irgendeine Kombination daraus.
- Verabreichungssysteme Die Zusammensetzungen der vorliegenden Erfindung können ein oder mehrere aktives) Mittel einschließen.
- Die Verbindungen I bis XIII, XV bis XXXIV, XXXVI bis XXXIX und XLI bis XLV oder Polyaminosäuren oder Peptide, die wenigstens eine der Verbindungen I bis XLV einschließen, können direkt als Arzneimittel-Verabreichungsträger verwendet werden durch einfaches Mischen einer oder mehrerer Verbindung(en), Polyaminosäure(n) oder Peptid(e) mit dem aktiven Bestandteil vor der Verabreichung.
- Die Verbindungen, Polyaminosäuren oder Peptide können zur Ausbildung von Mikrosphären verwendet werden, die das aktive Mittel enthalten. Diese Verbindungen, Polyaminosäuren oder Peptide sind besonders nützlich für die orale Verabreichung bestimmte, biologisch aktiver Mittel, z. B. kleiner Peptid-Hormone, die für sich allein gar nicht oder nur langsam durch die Schleimhaut des Gastrointestinaltraktes hindurchtreten und/oder einer chemischen Spaltung durch Säuren und Enzymen im Gastrointestinaltrakt unterliegen.
- Wenn die modifizierten Aminosäuren, Polyaminosäuren oder Peptide in Mikrosphären umgewandelt werden sollen, wird die Mischung gegebenenfalls auf eine Temperatur erhitzt, die im Bereich zwischen etwa 20 und etwa 50°C liegt, vorzugsweise bei etwa 40°C liegt, bis sich die modifizierte(n) Aminosäure(n) löst/lösen. Die letztlich erhaltene Lösung enthält zwischen etwa 1 mg und etwa 2000 mg an Verbindung, Polyaminosäure oder Peptid pro ml Lösung, vorzugsweise zwischen etwa 1 und etwa 500 mg pro ml. Die Konzentration an aktivem Mittel in der End-Lösung schwankt und ist abhängig von der für eine Behandlung erforderlichen Dosierung. Sofern nötig, kann die exakte Konzentration bestimmt werden durch – beispielsweise – Umkehrphasen-HPLC-Analyse.
- Wenn die Verbindungen, Polyaminosäuren oder Peptide zum Herstellen von Mikrosphären verwendet werden, ist ein anderes nützliches Verfahren das folgende: Verbindungen, Polyaminosäuren oder Peptide werden in entionisiertem Wasser in einer Konzentration gelöst, die im Bereich zwischen etwa 75 und etwa 200 mg/ml liegt, vorzugsweise bei 100 mg/ml, und zwar bei einer Temperatur zwischen etwa 25°C und etwa 60°C, vorzugsweise etwa 40°C. Teilchenförmiges Material, das in der Lösung verbleibt, kann mit herkömmlichen Mitteln, wie beispielsweise Filtration, entfernt werden.
- Danach wird die die Verbindung, die Polyaminosäure oder das Peptid umfassende Lösung, die bei einer Temperatur von etwa 40°C gehalten wird, im Verhältnis 1 : 1 (V/V) mit einer wäßrigen Säure-Lösung gemischt (auch bei etwa 40°C), die eine Säure-Konzentration aufweist, die im Bereich zwischen etwa 0,05 N und etwa 2 N liegt, vorzugsweise bei etwa 1,7 N. Die resultierende Mischung wird weiter bei 40°C für eine Zeitdauer inkubiert, die wirksam für die Bildung von Mikrosphären ist, wie durch Licht-Mikroskopie beobachtet wird. Bei der praktischen Durchführung der Erfindung ist die bevorzugte Reihenfolge der Zugabe diejenige, die die Verbindung, die Polyaminosäure oder das Peptid umfassende Lösung der wäßrigen Säure-Lösung zuzusetzen.
- Geeignete Säuren für die Bildung von Mikrosphären schließen jede beliebige Säure ein, die nicht
-
- (a) nachteilig die modifizierten Aminosäuren, Polyaminosäuren oder Peptide beeinträchtigt, z. B. eine chemische Abbau-Reaktion initiiert oder fördert;
- (b) die Mikrosphären-Bildung stört;
- (c) die Einarbeitung der Nutzverbindung in die Mikrosphären stört; und
- (d) in nachteiliger Weise mit der Nutzverbindung in Wechselwirkung tritt.
- Bevorzugte Säuren zur Verwendung in diesem Aspekt schließen Essigsäure, Zitronensäure, Chlorwasserstoffsäure, Phosphorsäure, Äpfelsäure und Maleinsäure ein.
- Ein Mikrosphären stabilisierendes Additiv kann in die wäßrige Säure-Lösung oder in die die Verbindung oder der Nutzverbindung umfassende Lösung vor dem Prozeß der Mikrosphären-Bildung eingearbeitet werden. Bei einigen Arzneimitteln fördert die Gegenwart derartiger Additive die Stabilität und/oder Dispergierbarkeit der Mikrosphären in Lösung.
- Die stabilisierenden Additive können in einer Konzentration verwendet werden, die im Bereich zwischen etwa 0,1 und 5% (w/v) liegt, vorzugsweise in einer Konzentration von etwa 0,5% (w/v).
- Geeignete, nicht jedoch beschränkende Beispiele von Mikrosphären stabilisierenden Additiven schließen Gummi arabicum, Gelatine, Methylcellulose, Polyethylenglycol und Polylysin ein. Die bevorzugten Stabilisierungs-Additive sind Gummi arabicum, Gelatine und Methylcellulose.
- Unter den obigen Bedingungen bilden die Verbindungs-Moleküle, Polyaminosäuren oder Peptide hohle oder feste Mikrosphären des Matrix-Typs, worin die Nutzverbindung in einer Träger-Matrix verteilt ist, oder Mikrosphären des Kapsel-Typs, die eine flüssige oder feste Nutzverbindung einkapseln. Wenn die die Verbindung, die Polyaminosäure oder das Polypeptid umfassenden Mikrosphären in Gegenwart eines löslichen Materials gebildet werden, z. B. eines pharmazeutischen Mittels in der wie oben beschrieben zusammengesetzten wäßrigen Säure-Lösung, wird dieses Material innerhalb der Mikrosphären eingekapselt. Auf diese Weise kann man pharmakologisch aktive Materialien wie beispielsweise Peptide, Proteine und Polysaccharide sowie auch geladene organische Moleküle, z. B. antimikrobielle Mittel, die normalerweise eine schlechte Bioverfügbarkeit bei Verabreichung auf oralem Wege haben, verkapseln. Die Menge an pharmazeutischem Mittel, die durch die Mikrosphären inkorporiert werden kann, ist abhängig von einer Anzahl Faktoren, die die Konzentration des Mittels in der Lösung sowie die Affinität der Nutzverbindung gegenüber dem Träger einschließen. Die die Verbindung, die Polyaminosäure oder das Peptid umfassenden Mikrosphären verändern die physiologischen und biologischen Eigenschaften des aktiven Mittels nicht. Weiter verändert der Verkapselungsprozeß die pharmakologischen Eigenschaften des aktiven Mittels nicht. Irgendein pharmakologisches Mittel kann in die Mikrosphären eingearbeitet werden. Das System ist besonders vorteilhaft zum Verabreichen chemischer oder biologischer Mittel, die sonst durch Bedingungen zerstört oder weniger wirksam gemacht würden, die innerhalb des Körpers des Tiers angetroffen werden, an den es verabreicht wird, bevor die Mikrosphären die Ziel-Zone erreichen (d. h. den Bereich, in dem der Gehalt der Mikrosphären freigesetzt werden soll); das System ist auch besonders vorteilhaft zum Verabreichen pharmakologischer Mittel, die im Gastrointestinaltrakt schlecht absorbiert werden. Die Ziel-Zonen können in Abhängigkeit von dem verwendeten Arzneimittel schwanken.
- Die Teilchengröße der Mikrosphären spielt eine wichtige Rolle bei der Bestimmung der Freisetzung des aktiven Mittels in dem Zielbereich des Gastrointestinaltrakts. Die bevorzugten Mikrosphären haben Durchmesser zwischen etwa ≤ 0,1 Micron und etwa 10 Micron, vorzugsweise zwischen etwa 0,5 Micron und etwa 5 Micron. Die Mikrosphären sind ausreichend klein, um wirksam das aktive Mittel in dem Zielbereich innerhalb des Gastrointestinaltrakts freizusetzen, wie beispielsweise zwischen Magen und Jejunum. Kleine Mikrosphären können auch parenteral verabreicht werden, indem sie in einem passenden Träger-Fluid (z. B. isotonischer Kochsalzlösung) suspendiert und direkt in das zirkulatorische System, intramuskulär oder subkutan injiziert werden. Die gewählte Verabreichungsart schwankt natürlich in Abhängigkeit von den Erfordernissen des zu verabreichenden aktiven Mittels. Große Aminosäure-Mikrosphären (> 50 Micron) neigen dazu, als orale Verabreichungssysteme weniger effizient zu sein.
- Die Größe der Mikrosphären, die durch In-Kontakt-Bringen der Verbindungen, Polyaminosäuren oder Peptide mit Wasser oder einer wäßrigen Lösung gebildet werden, die aktive Mittel enthält, kann dadurch gesteuert werden, daß man eine Vielzahl physikalischer oder chemischer Parameter einstellt, wie beispielsweise pH-Wert, Osmolarität oder Innenstärke der verkapselnden Lösung, Größe der Ionen in Lösung und durch die Wahl der bei dem Verkapselungsprozeß verwendeten Säure.
- Die Verabreichungs-Mischungen werden hergestellt durch Mischen einer wäßrigen Lösung des Trägers mit einer wäßrigen Lösung der aktiven Komponente unmittelbar vor einer Verabreichung. Alternativ dazu können der Träger und die biologisch aktive Komponente während des Herstellungsprozesses gemischt werden. Die Lösungen können gegebenenfalls Additive wie beispielsweise Phosphatpuffer-Salze, Citronensäure, Essigsäure, Gelatine und Gummi arabicum enthalten.
- Stabilisierende Additive können in die Trägerlösung eingearbeitet werden. Bei einigen Arzneimitteln fördert die Gegenwart solcher Additive die Stabilität und Dispergierbarkeit des Mittels in Lösung.
- Die stabilisierenden Additive können in einer Konzentration verwendet werden, die im Bereich zwischen etwa 0,1 und 5% (w/v) liegt, vorzugsweise bei etwa 0,5% (w/v). Geeignete, nicht jedoch beschränkende Beispiele von stabilisierenden Additiven schließen Gummi arabicum, Gelatine, Methylcellulose, Polyethylenglycol und Polylysin ein. Die bevorzugten stabilisierenden Additive sind Gummi arabicum, Gelatine und Methylcellulose.
- Die Menge des aktiven Mittels ist eine Menge, die wirksam ist, um den Zweck des speziellen aktiven Mittels zu erreichen. Die Menge in der Zusammensetzung ist typischerweise eine pharmakologisch oder biologisch wirksame Menge. Jedoch kann die Menge geringer sein als eine pharmakologisch oder biologisch wirksame Menge, wenn die Zusammensetzung in einer Dosiseinheitsform, wie beispielsweise einer Kapsel, einer Tablette oder einer Flüssigkeit verwendet wird, da die Dosiseinheitsform eine Mehrzahl von Zusammensetzungen aus Träger und biologisch aktivem Mittel enthalten kann oder eine geteilte pharmakologisch oder biologisch wirksame Menge enthalten kann. Die effektiven Gesamtmengen können dann in kumulativen Einheiten verabreicht werden, die insgesamt pharmakologisch oder biologisch aktive Mengen eines biologisch oder pharmakologisch aktiven Mittels enthalten.
- Die Gesamtmenge an aktivem Mittel und insbesondere an biologisch aktivem Mittel, das zu verwenden ist, kann von Fachleuten in diesem technischen Bereich bestimmt werden. Es wurde jedoch überraschenderweise gefunden, daß bei einigen biologisch aktiven Mitteln die Verwendung der vorliegend offenbarten Träger zu einer extrem wirksamen Verabreichung führt. Daher können geringere Mengen an biologisch aktivem Mittel als in den Fällen, wie sie bei früheren Dosis-Einheitsformen oder Verabreichungssystemen verwendet wurden, an den Empfänger verabreicht werden, wobei man immer noch dieselben Blutwerte und therapeutischen Wirkungen erreicht.
- Die Menge an Träger in der vorliegenden Zusammensetzung ist eine für die Verabreichung wirksame Menge und kann für irgendeinen speziellen Träger oder ein biologisch aktives Mittel nach Verfahren bestimmt werden, die Fachleuten mit Sachverstand in diesem technischen Bereich bekannt sind. Dosiseinheitsformen können auch irgendeinen der Zusätze, Arzneistoffträger bzw. Vehikel, Verdünnungsmittel, Zerfallsmittel, Gleitmittel, Weichmacher, Färbemittel und Dosier-Bindemittel einschließen, die einschließen, jedoch nicht beschränkt sind auf Wasser, 1,2-Propandiol, Ethanol, Olivenöl oder irgendeine Kombination daraus.
- Die vorliegenden Zusammensetzungen oder Dosiseinheitsformen sind geeignet für eine orale Verabreichung oder eine intraduodenale Injektion.
- Die Verabreichungs-Zusammensetzungen der vorliegenden Erfindung können auch einen oder mehrere Enzyminhibitoren einschließen. Solche Enzyminhibitoren schließen ein, sind jedoch nicht beschränkt auf Verbindungen wie beispielsweise Actinonin oder Epiactinonin und Derivate davon. Diese Verbindungen haben die folgenden Formeln:
- Derivate dieser Verbindungen sind offenbart in dem US-Patent Nr. 5,206,384. Actinonin-Derivate haben die folgende Formel: worin R12 für Sulfoxymethyl oder Carboxyl oder eine substituierte Carboxyl-Gruppe steht, die gewählt ist aus Carboxamid-Gruppe, Hydroxyaminocarbonyl-Gruppen und Alkoxycarbonyl-Gruppen; und R13 für eine Hydroxyl-Gruppe, Alkoxy-Gruppe, Hydroxyamino-Gruppe oder Sulfoxyamino-Gruppe steht. Andere Enzym-Inhibitoren schließen ein, sind jedoch nicht beschränkt auf Aprotinin (Trasylol) und den Bowman-Birk-Inhibitor.
- Die Verbindungen und Zusammensetzungen der vorliegenden Erfindung sind nützlich zum Verabreichen biologisch aktiver Mittel an irgendwelche Tiere wie beispielsweise Vögel, Säuger wie beispielsweise Primaten und insbesondere Menschen und Insekten. Das System ist besonders vorteilhaft zum Verabreichen chemisch oder biologisch aktiver Mittel, die ansonsten durch Bedingungen zerstört oder weniger wirksam gemacht würden, wie man sie antrifft, bevor das aktive Mittel seine Ziel-Zone erreicht (d. h. den Bereich, in dem das aktive Mittel der Verabreichungs-Zusammensetzung freigesetzt werden soll) und innerhalb des Körpers des Tiers antrifft, an den die Zusammensetzungen verabreicht werden. Insbesondere sind die Verbindungen und Zusammensetzungen der vorliegenden Erfindung nützlich bei der oralen Verabreichung aktiver Mittel, insbesondere solcher, die normalerweise nicht oral verabreichbar sind.
- Beschreibung der bevorzugten Ausführungsformen
- Die folgenden Beispiele veranschaulichen die Erfindung ohne Beschränkung.
- Beispiel 1
- Verbindung VI wurde wie folgt hergestellt:
Acetylsalicyloylchlorid (47,0 g; 0,24 mol; Äquivalent) wurde portionsweise einer Mischung von 4-(4-Aminophenyl-)buttersäure (50,0 g; 0,28 mol; 1,2 Äquivalente) in wäßrigem Natriumhydroxid (2M; 300 ml) zugesetzt. Die Reaktionsmischung wurde bei 25°C 2 h lang gerührt, und die resultierende Lösung wurde mit wäßriger Chlorwasserstoffsäure (1M) auf einen pH-Wert von 2,1 angesäuert. Der resultierende Niederschlag wurde filtriert und wurde mit wäßriger Chlorwasserstoffsäure (1M; 3 × 100 ml) und Wasser gewaschen und ergab Verbindung VI in Form eines blaß-pink-farbenen Feststoffs (31,89 g; 52%). - Die Eigenschaften werden nachfolgend aufgelistet:

- Verbindung IX wurde wie folgt hergestellt: eine Lösung von 4-Phenylbutyrylchlorid (10,20 g; 56 mMol) in Tetrahydrofuran (30 ml) wurde tropfenweise zu einer Mischung von 4-(4-Aminophenyl-)buttersäure (10,00 g, 56 mMol; 1,0 Äquivalente), Triethylamin (8,50 ml; 62 mMol; 1,1 Äquivalente) und Tetrahydrofuran (100 ml) bei 10°C gegeben. Die Reaktion wurde bei 10°C 1 h lang und bei 25°C 3 h lang gerührt. Das Lösungsmittel wurde dann verdampft, und der Rückstand wurde in Ethylacetat (150 ml) gelöst. Nach Waschen der Ethylacetat-Schicht mit wäßriger Chlorwasserstoffsäure (1M; 3 × 100 ml) und Wasser (2 × 100 ml) wurde die organische Schicht getrocknet und verdampft. Der resultierende Rückstand wurde aus Acetonitril-Wasser umkristallisiert und ergab Verbindung IX in Form eines blaß-gelben Feststoffes (11,69 g; 65%).

- Verbindung XXIV wurde wie folgt hergestellt: N-Hydroxysuccinamid (8,86 g; 77,00 mMol; 1,1 Äquivalente) und Dicyclohexylcarbodiimid (15,88 g; 77,0 mMol, 1,1 Äquivalente) wurden zu einer Lösung von 3-(4-Fluorbenzoyl-)propionsäure (13,73 g; 70,0 mMol; 1 Äquivalent) in Dimethylformamid (250 ml) gegeben. Die Reaktionsmischung wurde bei 25°C unter Stickstoff 12 h lang gerührt. Die Lösung wurde mit Wasser (500 ml) verdünnt und mit Chloroform (250 ml) extrahiert. Die organische Schicht wurde dann getrocknet und filtriert. Eisessig (5 ml) wurde dem Filtrat zugesetzt, und diese Mischung wurde für 1 h gerührt. Die resultierende Chloroform-Lösung wurde mit Natriumbicarbonat (250 ml) und Wasser (250 ml) gewaschen und über Magnesiumsulfat getrocknet. Nach Filtration wurden 4-(4-Aminophenyl-)buttersäure (12,5 g; 70,0 mMol; 1 Äquivalent) und Triethylamin (16 ml) dem Filtrat zugesetzt. Die resultierende Mischung wurde über Nacht bei 25°C gerührt, und sie wurde dann mit Chlorwasserstoffsäure (250 ml) angesäuert und lyophilisiert, und man erhielt Verbindung XXIV in Form eines weißen Feststoffs (3,50 g; 14%).

- Verbindung XXXII wurde wie folgt hergestellt: 1-Oxaspiro(2.5)-octan (3,76 g; 33,48 mMol, 1,2 Äquivalente) und Aluminiumchlorid (0,36 g; 2,70 mMol; 0,1 Äquivalente) wurden einer Suspension von 4-(4-Aminophenyl-)buttersäure (5,00 g; 27,90 mMol; 1 Äquivalent) in Toluol (100 ml) zugesetzt. Die Mischung wurde unter Argon über Nacht am Rückfluß behandelt. Nach Abkühlen auf Raumtemperatur wurde das Toluol abfiltriert, und der Rückstand wurde mit Ethylacetat (ca. 100 ml) gewaschen. Das kombinierte Filtrat wurde eingedampft, und man erhielt einen braunen Gummi. Der Gummi wurde in Ethylacetat (250 ml) gelöst. Er wurde dann mit Wasser (3 × 100 ml) gewaschen und getrocknet. Nach Entfernen des Lösungsmittels wurde der Rückstand durch Säulenchromatographie (30% bis 70% Ethylacetat/Hexane) gereinigt, und das gewonnene Produkt wurde aus Ethylacetat-Hexanen unter Erhalt von Verbindung XXXII in Form eines gelben Feststoffs (0,8 g; 10%) umkristallisiert.

- Verbindung XXXIX wurde wie folgt hergestellt: N-Hydroxysuccinimid (7,72 g; 67,50 mMol; 1,1 Äquivalente) und Dicyclohexylcarbodiimid (13,96 g, 67,50 mMol; 1,1 Äquivalente) wurden einer Lösung von N-(2-Phenylbutyryl-)4-(aminophenyl)buttersäure (20,00 g; 61,40 mMol; 1,0 Äquivalent) in Tetrahydrofuran (400 ml) zugesetzt. Die Reaktionsmischung wurde über Nacht bei 25°C gerührt. Der gebildete Harnstoff wurde durch Filtration entfernt. Eisessig (5 ml) wurde dem Filtrat zugesetzt, die Mischung wurde für 2 h gerührt. Das Lösungsmittel wurde dann verdampft, und man erhielt ein Öl. Das Öl wurde erneut in Chloroform (300 ml) gelöst, und die resultierende Lösung wurde aufeinanderfolgend mit gesättigtem Natriumbicarbonat (2 × 200 ml) und Wasser (200 ml) gewaschen. Die vereinigten wäßrigen Schichten wurden mit Chloroform (100 ml) extrahiert, und man erhielt ein Filtrat (Gesamtvolumen 500 ml), das den Osu-(O-Succinyl-)ester von N-(2-Phenylbutyryl-)4-(4-aminophenyl-)buttersäure enthielt.
- Eine Mischung aus Phenylglycin-0-methylester-hydrochlorid (12,40 g; 61,40 mMol; 1,0 Äquivalent) und Triethylamin (35 ml) in Chloroform (100 ml) wurde in einen Zugabetrichter gegeben. Die Mischung wurde tropfenweise der Chloroform-Lösung des Osu-Esters zugesetzt, der wie oben beschrieben hergestellt worden war. Die Reaktionsmischung wurde bei 25°C 24 h lang gerührt. Die resultierende Lösung wurde mit wäßriger Chlorwasserstoffsäure (2 × 500 ml) und Wasser (500 ml) gewaschen. Die wäßrige Schicht wurde mit Chloroform zurück-extrahiert (50 ml). Die vereinigten Chloroform-Schichten wurden getrocknet und eingedampft, wobei man ein Öl erhielt. Wäßriges Natriumhydroxid (2M, 200 ml) wurde dem Öl zugesetzt, und die Mischung wurde für 2 h auf 100°C erhitzt. Nach Abkühlen auf Raumtemperatur wurde die Lösung mit Chlorwasserstoffsäure (2M) auf einen pH-Wert von 2,5 angesäuert. Der Niederschlag wurde filtriert, mit Chlorwasserstoffsäure (100 ml) und Wasser (100 ml) gewaschen und ergab dann Verbindung XXXIX in Form eines weißlichen Feststoffs (15,2 g; 54%).

- In vivo-Bewertung von Interferon bei Ratten
- Es wurden Dosierungs-Zusammensetzungen hergestellt durch Mischen der modifizierten Aminosäure-Verbindungen und Interferon α2 in den in der nachfolgenden Tabelle 1 aufgelisteten Mengen in einer Trizma®-Hydrochlorid-Pufferlösung (Tris-HCl) bei einem pH-Wert von etwa 7 bis B. Propylenglycol (0 bis 25%) wurde als solubilisierendes Mittel zugegeben, sofern dies erforderlich war.
- Die Dosierungs-Zubereitungen wurden oral oder intraduodenal (ID) an Ratten verabreicht, und die Verabreichung wurde über einen ELISA-Assay für Human-Interferon α-2b bewertet. Die Ergebnisse sind in der nachfolgenden Tabelle 1 angegeben.
- Beispiel 7
- In-vivo-Bewertung von Lachs-Calcitonin in Ratten
- Es wurden Dosierungs-Zusammensetzungen hergestellt und unter Verwendung der modifizierten Aminosäure-Verbindungs-Träger und Lachs-Calcitonin gemäß den Angaben der nachfolgenden Tabelle 2 dosiert. Die Konzentration an Calcitonin in jeder Zusammensetzung betrug 2,5 μg/ml. Jeder Ratte wurden 2 ml/kg der Dosierungs-Zusammensetzung verabreicht.
- Blutproben wurden in Reihen-Tests von der Schwanz-Arterie abgenommen. Der Serum-Calcium-Wert wurde bestimmt durch Test mit einem Calcium-Keto (Firma Sigma Chemical Co., St. Louis, MO.).
- Die Ergebnisse sind in der nachfolgenden Tabelle 2 angegeben.
- Beispiel 8
- In-vivo-Bewertung der Lachs-Calcitonin-Konzentration in Ratten
- Es wurde eine Dosierungs-Zusammensetzung hergestellt unter Verwendung von 400 mg der Verbindung VI mit 2,9 ml 25%-igen wäßrigen Propylenglycols. Die resultierende Lösung wurde gerührt, und der pH-Wert wurde mit Natriumhydrochlorid (1,01 N) auf 7,2 eingestellt. Wasser wurde zugesetzt, um das Gesamtvolumen auf 2,0 ml und eine Endkonzentration an der modifizierten Aminosäure von 200 mg/ml zu bringen. Lachs-Calcitonin (10 mg) wurde zugesetzt. Diese Zusammensetzung wurde wie im obigen Beispiel 7 beschrieben dosiert.
- Die Ergebnisse sind in der nachfolgenden Tabelle 2 angegeben.
- Beispiel 9
- In-vivo-Bewertung von rekombinantem menschlichem Wachstumshormon (rhGh) in Ratten
- Es wurden Dosierungs-Zusammensetzungen hergestellt mit modifizierten Aminosäuren in einem Phosphatpuffer bei einem pH-Wert von etwa 7 bis 8 und mit rhGh gemäß den Angaben in der nachfolgenden Tabelle 3.
- Ratten wurden die Zusammensetzungen durch orale Gabe, intraduodenale Verabreichung (ID) oder Dickdarm-Verabreichung (IC) verabreicht. Die Ergebnisse sind in der nachfolgenden Tabelle 3 veranschaulicht.
- Beispiel 10
- In-vivo-Bewertung von Heparin in Ratten
- 900 mg einer modifizierten Aminosäure wurden in 3 ml Propylenglycol gelöst, und 0,299 g Natriumheparin wurden in 3 ml Wasser gelöst. Die beiden Lösungen wurden durch Vortex-Mischen miteinander gemischt. Natriumhydrochlorid wurde der resultierenden Mischung zugesetzt, bis eine Lösung erhalten wurde. Der pH-Wert wurde dann auf 7,4 ± 0,5 mit konzentrierter Chlorwasserstoffsäure eingestellt. Die End-Lösung wurde bei 40°C 30 min lang mit Ultraschall behandelt und so eine Dosierungslösung erhalten.
- Die Dosierungslösung wurde durch orale Gabe an auf Fastendiät gesetzte Ratten verabreicht. Blutproben wurden durch Herzpunktieren im Anschluß an die Verabreichung von Ketamin (44 mg/kg) gewonnen. Die Heparin-Aktivität wurde bestimmt durch Anwenden des Verfahrens der aktivierten partiellen Thromboplastin-Zeit (activated partial thromboplastin time; APTT) gemäß dem Verfahren von J. B. Henry, Clinical Diagnosis and Management by Laboratory Methods; Philadelphia, PA; WB Saunders (1979). Die Ergebnisse sind in der nachfolgenden Tabelle 4 angegeben.
- Beispiel 11
- Heparin mit niedrigem Molekulargewicht wurde nach dem Verfahren von Beispiel 10 dosiert.
Claims (27)
- Polyaminosäure, wie sie in Patentanspruch 2 definiert ist, umfassend ein Peptid.
- Zusammensetzung, umfassend (a) ein aktives Mittel; und (b) eine Polyaminosäure, wie sie in Patentanspruch 2 definiert ist.
- Zusammensetzung nach Anspruch 5, worin die Polyaminosäure ein Peptid umfaßt.
- Zusammensetzung nach Anspruch 4, worin das aktive Mittel ein biologisch aktives Mittel umfaßt.
- Zusammensetzung nach Anspruch 7, worin das biologisch aktive Mittel gewählt ist aus der Gruppe, die besteht aus einem Peptid, einem Mucopolysaccharid, einem Kohlenhydrat, einem Lipid, einem Pestizid oder irgendeiner Kombination daraus.
- Zusammensetzung nach Anspruch 8, worin das biologisch aktive Mittel gewählt ist aus der Gruppe, die besteht aus menschlichem Wachstumshormon, Rinder-Wachstumshormon, Wachstumshormon-freisetzendem Hormon (growth hormonereleasing hormone), einem Interferon, Interleukin-II, Insulin, Heparin, Calcitonin, Erythropoietin, Atrionatriuretischem Faktor, einem Antigen, einem monoklonalen Antikörper, Somatostatin, Adrenocorticotropin, Gonadotropin-freisetzendem Hormon (gonadotropin releasing hormone), Oxytocin, Vasopressin, Cromolyn-Natrium, Vancomycin, Desferrioxamine (DFO), antimikrobiellen Mitteln oder irgendeiner Kombination daraus.
- Zusammensetzung nach Anspruch 5, worin das aktive Mittel ein biologisch aktives Mittel umfaßt.
- Zusammensetzung nach Anspruch 10, worin das biologisch aktive Mittel gewählt ist aus der Gruppe, die besteht aus einem Peptid, einem Mucopolysaccharid, einem Kohlenhydrat, einem Lipid, einem Pestizid oder irgendeiner Kombination daraus.
- Zusammensetzung nach Anspruch 11, worin das biologisch aktive Mittel gewählt ist aus der Gruppe, die besteht aus menschlichem Wachstumshormon, Rinder-Wachstumshormon, Wachstumshormon-freisetzendem Hormon (growth hormonereleasing hormone), einem Interferon, Interleukin-II, Insulin, Heparin, Calcitonin, Erythropoietin, Atrionatriuretischem Faktor, einem Antigen, einem monoklonalen Antikörper, Somatostatin, Adrenocorticotropin, Gonadotropin-freisetzendem Hormon (gonadotropin releasing hormone), Oxytocin, Vasopressin, Cromolyn-Natrium, Vancomycin, Desferrioxamine (DFO), antimikrobiellen Mitteln oder irgendeiner Kombination daraus.
- Zusammensetzung nach Anspruch 6, worin das aktive Mittel ein biologisch aktives Mittel umfaßt.
- Zusammensetzung nach Anspruch 13, worin das biologisch aktive Mittel gewählt ist aus der Gruppe, die besteht aus einem Peptid, einem Mucopolysaccharid, einem Kohlenhydrat, einem Lipid, einem Pestizid oder irgendeiner Kombination daraus.
- Zusammensetzung nach Anspruch 14, worin das biologisch aktive Mittel gewählt ist aus der Gruppe, die besteht aus menschlichem Wachstumshormon, Rinder-Wachstumshormon, Wachstumshormon-freisetzendem Hormon (growth hormonereleasing hormone), einem Interferon, Interleukin-II, Insulin, Heparin, Calcitonin, Erythropoietin, Atrionatriuretischem Faktor, einem Antigen, einem monoklonalen Antikörper, Somatostatin, Adrenocorticotropin, Gonadotropin-freisetzendem Hormon (gonadotropin releasing hormone), Oxytocin, Vasopressin, Cromolyn-Natrium, Vancomycin, Desferrioxamine (DFO), antimikrobiellen Mitteln oder irgendeiner Kombination daraus.
- Dosiseinheitsform, umfassend (A) eine Zusammensetzung nach Anspruch 4; und (B) (a) einen Träger, (b) ein Verdünnungsmittel, (c) ein Zerfallsmittel, (d) ein Gleitmittel, (e) einen Weichmacher, (f) ein Färbemittel, (g) ein Dosier-Bindemittel, oder (h) irgendeine Kombination daraus.
- Dosiseinheitsform nach Anspruch 16, umfassend eine Tablette, eine Kapsel oder eine Flüssigkeit.
- Dosiseinheitsform, umfassend (A) eine Zusammensetzung nach Anspruch 5; und (B) (a) einen Träger, (b) ein Verdünnungsmittel, (c) ein Zerfallsmittel, (d) ein Gleitmittel, (e) einen Weichmacher, (f) ein Färbemittel, (g) ein Dosier-Bindemittel, oder (h) irgendeine Kombination daraus.
- Dosiseinheitsform nach Anspruch 18, umfassend eine Tablette, eine Kapsel oder eine Flüssigkeit.
- Dosiseinheitsform, umfassend (A) eine Zusammensetzung nach Anspruch 6; und (B) (a) einen Träger, (b) ein Verdünnungsmittel, (c) ein Zerfallsmittel, (d) ein Gleitmittel, (e) einen Weichmacher, (f) ein Färbemittel, (g) ein Dosier-Bindemittel, oder (h) irgendeine Kombination daraus.
- Dosiseinheitsform nach Anspruch 20, umfassend eine Tablette, eine Kapsel oder eine Flüssigkeit.
- Verwendung der Polyaminosäure von Patentanspruch 2 und eines biologisch aktiven Mittels für die Herstellung einer Zusammensetzung für eine orale Verabreichung.
- Verwendung der Polyaminosäure von Patentanspruch 2, umfassend ein Peptid und ein biologisch aktives Mittel, für die Herstellung einer Zusammensetzung für eine orale Verabreichung.
- Verfahren zur Herstellung einer Zusammensetzung, wobei das Verfahren umfaßt ein Mischen von (A) wenigstens einem biologisch aktiven Mittel; (B) wenigstens einer Verbindung, wie sie im Patentanspruch 1 definiert ist, oder einer Verbindung, die gewählt ist aus der Gruppe, die besteht aus und deren Salzen; und (C) gegebenenfalls eines Dosier-Bindemittels.
- Verfahren zur Herstellung einer Zusammensetzung, wobei das Verfahren umfaßt ein Mischen von (A) wenigstens einem biologisch aktiven Mittel; (B) wenigstens einer Polyaminosäure, wie sie in Patentanspruch 2 definiert ist; und (C) gegebenenfalls einem Dosier-Bindemittel.
- Verfahren zur Herstellung einer Zusammensetzung, wobei das Verfahren umfaßt das Mischen von (A) wenigstens einem biologisch aktiven Mittel; (B) wenigstens einem Peptid, wie es in Patentanspruch 3 definiert ist; und (C) gegebenenfalls einem Dosier-Bindemittel.
