Description

- Perindopril sowie seine pharmazeutisch annehmbaren Salze, und insbesondere sein tert.-Butylaminsalz, besitzen interessante pharmakologische Wirkungen.
- Ihre Hauptwirkung besteht darin, das Angiotensin I umwandelnde Enzym (oder Kininase II) zu inhibieren, was es einerseits ermöglicht, die Umwandlung des Decapeptids Angiotensin I in das Octapeptid Angiotensin II (gefäßverengend) zu verhindern und andererseits dem Abbau von Bradykinin (gefäßerweiternd) in ein inaktives Peptid vorzubeugen.
- Diese beiden Wirkungen tragen zu den günstigen Wirkungen von Perindopril bei kardiovaskulären Erkrankungen bei, insbesondere der arteriellen Hypertension und von Herzinsuffizienz.
- Perindopril, seine Herstellung und seine Anwendung in der Therapie sind in dem Europäischen Patent
EP 0 049 658 beschrieben. - Aufgrund des pharmazeutischen Interesses an dieser Verbindung wäre es wichtig, sie mit einem wirksamen und leicht in den technischen Maßstab überführbaren Syntheseverfahren herzustellen, welches Perindopril ausgehend von kostengünstigen Ausgangsmaterialien mit einer guten Ausbeute und einer ausgezeichneten Reinheit ergibt.
- Das Patent
EP 0 308 341 beschreibt die technische Synthese von Perindopril durch Kupplung des Benzylesters der (2S,3aS,7aS)-Octahydroindol-2-carbonsäure mit dem Ethylester von N-((S)-1-Carboxybutyl]-(S)-alanin, gefolgt von der Abspaltung der Schutzgruppe der Carboxylgruppe des Heterocyclus durch katalyti sche Hydrierung. - Die Anmelderin hat nunmehr ein neues Verfahren zur Synthese von Perindopril ausgehend von leicht zugänglichen Ausgangsmaterialien gefunden.
- Die vorliegende Erfindung betrifft insbesondere ein Verfahren zur Synthese von Perindopril und von seinen pharmazeutisch annehmbaren Salzen, welches dadurch gekennzeichnet ist, dass man die Verbindung der Formel (II) in der Konfiguration (S): in der R ein Wasserstoffatom oder eine Schutzgruppe für die Säurefunktion bedeutet,
mit einer Verbindung der Formel (III) in der Konfiguration (R): in der G ein Chlor- oder Bromatom oder eine Hydroxy-, p-Toluolsulfonyloxy-, Methansulfonyloxy- oder Trifluormethansulfonyloxy-gruppe bedeutet,
in Gegenwart einer Base umsetzt,
zur Bildung der Verbindung der Formel (IV): in der R und G die oben angegebenen Bedeutungen besitzen,
welche man einer intramolekularen Kupplungsreaktion unterwirft zur Bildung der Verbindung der Formel (V): in der R und G die oben angegebenen Bedeutungen besitzen,
welche man mit der Verbindung der Formel (VI): umsetzt zur Bildung der Verbindung der Formel (VII): in der R die oben angegebenen Bedeutungen besitzt,
welche man einer katalytischen Hydrierungsreaktion unterwirft, so dass man gegebenenfalls nach der Abspaltung der Schutzgruppe die Verbindung der Formel (I) erhält. - Als Schutzgruppe für die Säurefunktion kann man in nicht einschränkender Weise die Benzylgruppe oder geradkettige oder verzweigte (C1-C6)-Alkylgruppe nennen.
- Als Basen, die für die Umsetzung der Verbindungen der Formeln (II) und (III) geeignet sind, kann man in nicht einschränkender Weise organische Amine, wie Triethylamin, Pyridin oder Diisopropylethylamin, und anorganische Basen, wie NaOH, KOH, Na2CO3, K2CO3, NaHCO3 oder KHCO3 nennen.
- Die intramolekulare Kupplungsreaktion wird vorzugsweise entweder in Gegenwart einer Base und eines Katalysators auf der Grundlage von Palladium oder mit Hilfe von Natriumhydrid und Kupfer(I)-iodid oder Kupfer(I)-bromid durchgeführt. Die für diese Kupplungsreaktion bevorzugt verwendeten Katalysatoren auf der Basis von Palladium sind Katalysatoren auf der Grundlage von Palladium und eines Arylphosphins oder eines Bisphosphins.
- Als Katalysatoren dieser Art kann man in nicht einschränkender Weise nennen Pd(0)/PPh3, Pd(0)/P(o-Tolyl)3, Pd(0)/P(1-Naphthyl)3, Pd(0)/P(o-Methoxyphenyl)3, Pd2(dba)3/PPh3, Pd2(dba)3/P(o-Tolyl)3, Pd2(dba)3/P(1-Naphthyl)3, Pd2(dba)3/P(o-Methoxyphenyl)3, Pd2(dba)3/P(2-Furyl)3, Pd2(dba)3/dppp, Pd2(dba)3/(±)-BINAP und (DPPF)PdCl2·CH2Cl2/DPPF,
mit der Maßgabe, dass man unter BINAP 2,2'-Bis(diphenylphosphino)-1,1'-binaphthyl,
unter dba Dibenzylidenaceton,
unter DPPF 1,1'-Bis(diphenylphosphino)-ferrocen und
unter dppp 1,3-Bis(diphenylphosphino)-propan versteht. - Als für die Kupplungsreaktion in Gegenwart eines Katalysators auf der Grundlage von Palladium verwendete Basen kann man in nicht einschränkender Weise nennen Cs2CO3, NaOtBu, Na2CO3, NaOAc und KOAc.
- Wenn G ein Chlor- oder Bromatom oder eine p-Toluolsulfonyloxygruppe, Methansulfonyloxygruppe oder Trifluormethansulfonyloxygruppe bedeutet, wird die Reaktion zwischen den Verbindungen der Formeln (V) und (VI) vorzugsweise in Gegenwart einer Base, vorzugsweise eines organischen Amins, wie Triethylamin, Pyridin oder Diisopropylethylamin, oder einer anorganischen Base, wie Na2CO3, K2CO3, NaHCO3 oder KHCO3 durchgeführt.
- Wenn G eine Hydroxygruppe darstellt, wird die Reaktion zwischen den Verbindungen der Formeln (V) und (VI) vorzugsweise in Gegenwart eines Aktivierungsreagens, wie N-Methyl-N-phenyl-aminotriphenylphosphoniumiodid oder, wenn R von Wasserstoff verschieden ist, durch die Mitsunobu-Reaktion bewirkt.
- Die Verbindungen der Formel (IV) sind neue Produkte, die als Synthesezwischenprodukte in der chemischen oder pharmazeutischen Industrie eingesetzt werden können, insbesondere bei der Synthese von Perindopril und sind demzufolge integraler Bestandteil der vorliegenden Erfindung.
- Die Verbindungen der Formel (II) können nach dem in der Veröffentlichung J. Am. Chem. Soc. 116 (1994), 10847-10848 beschriebenen Verfahren hergestellt werden.
- BEISPIEL 1: tert.-Butylaminsalz der (2S,2aS,7aS)-1-{(2S)-2-[(1S)-1-(Ethoxycarbonyl)-butylamino]-propionyl}-octahydro-1H-indol-2-carbonsäure

- Stufe A: (2S)-3-(2-Bromphenyl)-2-{((2R)-2-brompropanoyl]-amino}-propansäurebenzylester
- Man beschickt ein Reaktionsgefäß mit 25,7 g (S)-2-Bromphenylalaninbenzylester und 150 ml Dichlormethan, bringt die Temperatur der Reaktionsmischung auf 0°C und gibt 20 ml Diisopropylethylamin und dann 13,2 g (2R)-2-Brompropionylchlorid zu. Man bringt die Temperatur der Mischung anschließend auf Raumtemperatur, rührt während 1 Stunde bei dieser Temperatur, wäscht die Mischung mit Wasser und dann mit einer verdünnten Essigsäurelösung, verdampft die Lösungsmittel und erhält die Titelverbindung.

- Stufe B: (2S)-1-[(2R)-2-Brompropanoyl]-2-indolincarbonsäurebenzylester
- Man beschickt ein Reaktionsgefäß mit 15,5 g der in der vorhergehenden Stufe erhaltenen Verbindung in Lösung in Toluol, 1,57 g Pd2(dba)3, 1,83 g P(o-Tolyl)3 und 21,5 g Cs2CO3. Man erhitzt die Reaktionsmischung auf 100°C, rührt während 15 Stunden bei dieser Temperatur, bringt die Mischung dann auf Raumtemperatur und reinigt durch Chromatographie über Siliciumdioxid unter Erhalt der Titelverbindung.

- Stufe C: (2S)-1-((2S)-2-{[(1S)-1-(Ethoxycarbonyl)-butyl]-amino}-propanoyl)-2-indolincarbonsäurebenzylester
- Man beschickt ein Reaktionsgefäß mit 12,3 g (2S)-2-Aminopentansäureethylester, 16 ml Triethylamin und 16 ml Acetonitril, bringt die Temperatur der Mischung auf 60°C und gibt langsam eine Lösung von 19,4 g der in der vorhergehenden Stufe erhaltenen Verbindung in Dichlormethan zu und erhitzt während 4 Stunden zum Sieden am Rückfluss. Nach dem Abkühlen auf Raumtemperatur wäscht man die Mischung mit Wasser und mit einer verdünnten Essigsäurelösung, verdampft die Lösungsmittel und erhält die Titelverbindung.

- Stufe D: (2S,3aS,7aS)-1-{(2S)-2-((1S)-1-(Ethoxycarbonyl)-butylamino]-propionyl}-octahydro-1H-indol-2-carbonsäure
- Man beschickt ein Hydriergefäß mit 20 g der in der vorhergehenden Stufe erhaltenen Verbindung in Lösung in Essigsäure mit 0,5 g 10 % Pd/C. Man hydriert bei einem Druck von 0,5 bar zwischen 15 und 30°C bis zur Absorption der theoretischen Wasserstoffmenge.
- Man entfernt den Katalysator durch Filtration, kühlt auf 0 bis 5°C ab und gewinnt den erhaltenen Feststoff durch Filtration. Man wäscht den Filterkuchen, trocknet ihn bis zur Gewichtskonstanz und erhält die Titelverbindung mit einer Enantiomerenreinheit von 99 %.

- Stufe E: tert.-Butylaminsalz der (2S,3aS,7aS)-1-{(2S)-2-((1S)-1-(Ethoxycarbonyl)-butylamino]-propionyl}-octahydro-1H-indol-2-carbonsäure
- Man löst den in der vorhergehenden Stufe erhaltenen Niederschlag (20 g) in 280 ml Ethylacetat, gibt 4 g tert.-Butylamin und 40 ml Ethylacetat zu.
- Man erhitzt die erhaltene Suspension anschließend bis zur vollständigen Auflösung zum Sieden am Rückfluss, filtriert die erhaltene Lösung in der Wärme und kühlt unter Rühren bis auf eine Temperatur von 15–20°C ab.
- Man filtriert den erhaltenen Niederschlag ab, teigt ihn erneut mit Ethylacetat an, trocknet ihn, verreibt ihn und erhält das erwartete Produkt mit einer Ausbeute von 95 %.
- BEISPIEL 2: tert.-Butylaminsalz der (2S,3a,7aS)-1-{(2S)-2-[(1S)-1-(Ethoxycarbonyl)-butylamino]-propionyl}-octahydro-1H-indol-2-carbonsäure

- Stufe A: (2S)-3-(2-Bromphenyl)-2-{((2R)-2-brompropanoyl]-amino}-propansäure
- Man beschickt ein Reaktionsgefäß mit 28,8 g (S)-2-Bromphenylalanin, 7,5 ml Wasser und 15 ml Toluol, bringt die Mischung auf eine Temperatur zwischen 0 und 5°C und gibt 25 ml 5M Natriumhydroxidlösung und dann eine Lösung von 20,2 g (2R)-2-Propionylchlorid in Toluol zu, wobei man die Temperatur unterhalb 10°C und den pH-Wert des Mediums durch Zugabe von 5M Natriumhydroxid bei 10 hält. Nach dem Rühren während einer weiteren Stunde bei 10°C gibt man konzentrierte Chlorwasserstoffsäure zu, um den pH-Wert der Mischung auf 6 zu bringen. Man trennt die Toluolphase ab, gibt konzentrierte Chlorwasserstoffsäure zu der wässrigen Phase, um den pH-Wert auf 2 zu bringen, filtriert den in dieser Weise gebildeten Niederschlag ab, trocknet ihn und erhält die Titelverbindung.

- Stufe B: Identisch mit der Stufe B des Beispiels 1.

- Stufe C: (2S)-1-((2S)-2-{((1S)-1-(Ethoxycarbonyl)-butyl]-amino}-propanoyl)-2-indolincarbonsäure
- Man beschickt ein Reaktionsgefäß mit 10,5 g (2S)-2-Aminopentansäureethylester, 13,5 ml Triethylamin und 13,5 ml Acetonitril, bringt die Mischung auf 60°C und gibt langsam eine Lösung von 19,3 g der in der vorhergehenden Stufe erhaltenen Verbindung in 130 ml Dichlormethan zu und erhitzt während 4 Stunden zum Sieden am Rückfluss. Nach dem Abkühlen auf Raumtemperatur wäscht man die Mischung mit Wasser und mit einer verdünnten Essigsäurelösung, verdampft die Lösungsmittel und erhält die Titelverbindung.

- Stufen D und E: Identisch mit den Stufen D und E des Beispiels 1.
- BEISPIEL 3: tert.-Butylaminsalz der (2S,3aS,7aS)-1-{(2S)-2-[(1S)-1-(Ethoxycarbonyl)-butylamino]-propionyl}-octahydro-1H-indol-2-carbonsäure

- Stufe A: (2S)-3-(2-Bromphenyl)-2-{((2R)-2-(p-toluolsulfonyloxy)-propanoyl]-amino}-propansäurebenzylester
- Man beschickt ein Reaktionsgefäß mit 25,7 g (R)-2-Bromphenylalaninbenzylester und 150 ml Dichlormethan, bringt dann die Reaktionsmischung auf 0°C und gibt 20 ml Diisopropylethylamin und dann 20,2 g (1R)-2-Chlor-1-methyl-2-oxoethyl-p-toluolsulfonatchlorid zu, bringt dann die Mischung auf Raumtemperatur, rührt während 1 Stunde bei dieser Temperatur, wäscht die Mischung mit Wasser, verdampft die Lösungsmittel und erhält die Titelverbindung.

Claims (12)
- Verfahren zur Synthese von Perindopril der Formel (I): und von seinen pharmazeutisch annehmbaren Salzen, dadurch gekennzeichnet, dass man die Verbindung der Formel (II) in der Konfiguration (S): in der R ein Wasserstoffatom oder eine Schutzgruppe für die Säurefunktion bedeutet, mit einer Verbindung der Formel (III) in der Konfiguration (R): in der G ein Chlor- oder Bromatom oder eine Hydroxy-, p-Toluolsulfonyloxy-, Methansulfonyloxy- oder Trifluormethansulfonyloxy-gruppe bedeutet, in Gegenwart einer Base umsetzt, zur Bildung der Verbindung der Formel (IV): in der R und G die oben angegebenen Bedeutungen besitzen, welche man einer intramolekularen Kupplungsreaktion unterwirft zur Bildung der Verbindung der Formel (V): in der R und G die oben angegebenen Bedeutungen besitzen, welche man mit der Verbindung der Formel (VI): umsetzt zur Bildung der Verbindung der Formel (VII): in der R die oben angegebenen Bedeutungen besitzt, welche man einer katalytischen Hydrierungsreaktion unterwirft, so dass man gegebenenfalls nach der Abspaltung der Schutzgruppe die Verbindung der Formel (I) erhält.
- Syntheseverfahren nach Anspruch 1, dadurch gekennzeichnet, dass R eine Benzylgruppe oder eine geradkettige oder verzweigte (C1-C6)-Alkylgruppe bedeutet.
- Syntheseverfahren nach Anspruch 1, dadurch gekennzeichnet, dass die intramolekulare Kupplungsreaktion entweder in Gegenwart einer Base und eines Katalysators auf der Grundlage von Palladium oder mit Hilfe von Natriumhydrid und Kupfer(I)-iodid oder Kupfer(I)-bromid durchgeführt wird.
- Syntheseverfahren nach Anspruch 3, dadurch gekennzeichnet, dass die intramolekulare Kupplungsreaktion in Gegenwart einer Base, eines Katalysators auf der Grundlage von Palladium und eines Arylphosphins oder eines Bisphosphins durchgeführt wird.
- Syntheseverfahren nach Anspruch 4, dadurch gekennzeichnet, dass die für die intramolekulare Kupplungsreaktion verwendete Base ausgewählt wird aus Cs2CO3, NaOtBu, Na2CO3, NaOAc und KOAc.
- Syntheseverfahren nach Anspruch 4 oder 5, dadurch gekennzeichnet, dass der Katalysator auf der Grundlage von Palladium und das Arylphosphin oder Bisphosphin ausgewählt werden aus Pd(0)/PPh3, Pd(0)/P(o-Tolyl)3, Pd(0)/P(1-Naphthyl)3, Pd(0)/P(o-Methoxyphenyl)3, Pd2(dba)3/PPh3, Pd2(dba)3/P(o-Tolyl)3, Pd2(dba)3/P(1-Naphthyl)3, Pd2(dba)3/P(o-Methoxyphenyl)3, Pd2(dba)3/P(2-Furyl)3, Pd2(dba)3/dppp, Pd2(dba)3/(±)-BINAP und (DPPF)PdCl2·CH2Cl2/DPPF, mit der Maßgabe, daß man unter BINAP 2,2'-Bis(diphenylphosphino)-1,1'-binaphthyl, unter dba Dibenzylidenaceton, unter DPPF 1,1'-Bis(diphenylphosphino)-ferrocen und unter dppp 1,3-Bis(diphenylphosphino)-propan versteht.
- Syntheseverfahren nach Anspruch 1, dadurch gekennzeichnet, dass G ein Chlor- oder Bromatom oder eine p-Toluolsulfonyloxygruppe, Methansulfonyloxygruppe oder Trifluormethansulfonyloxygruppe bedeutet.
- Syntheseverfahren nach Anspruch 7, dadurch gekennzeichnet, dass die Reaktion der Verbindungen der Formeln (V) und (VI) in Gegenwart eines organischen Amins ausgewählt aus Triethylamin, Pyridin und Diisopropylethylamin oder einer anorganischen Base ausgewählt aus Na2CO3, K2CO3, NaHO3 und KHCO3 durchgeführt wird.
- Syntheseverfahren nach Anspruch 1, dadurch gekennzeichnet, dass G eine Hydroxygruppe bedeutet.
- Syntheseverfahren nach Anspruch 9, dadurch gekennzeichnet, dass die Reaktion der Verbindungen der Formel (V) und (VI) in Gegenwart von N-Methyl-N-phenyl-aminotriphenylphosphoniumiodid oder, wenn R von einem Wasserstoffatom verschieden ist, durch eine Mitsunobu-Reaktion erfolgt.
- Syntheseverfahren nach einem der Ansprüche 1 bis 10 für die Herstellung von Perindopril in Form seines tert.-Butylaminsalzes.
