Gegenstand der Erfindung sind neue ß-Lactam-Äntibiotika und ihre Verwendung zur Behandlung von bakteriellen Infektionen.
Die Erfindung beruht auf dem überraschenden Befund, daß der ß-Lactamkern biologisch durch einen Substituenten der

Die ß-Lactame mit dem Substituenten

1-Stellung und einem Acylamino-Substituenten in 3-Stellung
besitzen Wirksamkeit gegen eine Reihe von gram-negativen
und gram-positiven Keimen.

die zur Beschreibung der ß-Lactame der Erfindung benutzt werden. Diese Definitionen beziehen sich auf Bezeichnungen,
die im Anmeldetext verwendet werden (soweit sie nicht in bestimmten
Fällen besonders begrenzt sind) und zwar entweder einzeln oder als Teile von größeren Gruppen.
Die Bezeichnungen "Alkyl-"und "Alkoxyrest" beziehen sich auf verzweigte und unverzweigte Reste. Reste mit 1 bis 10 Kohlenstoffatomen
sind bevorzugt.

Die Bezeichnungen "Alkanoyl-", "Alkenyl-", "Alkenyl-" und
"Alkinylrest" betreffen verzweigte und unverzweigte Reste. Diese Reste haben vorzugsweise 2 bis 10 Kohlenstoffatome.
Der Begriff "Halogenatom" betrifft Fluor-, Chlor-, Brom-

Beispiele für Substituenren sind die Oxogruppe (=0), HaIogen,

halogensubstituierten Alkylrest bedeutet), einen aromatischen Rest (wie vorstehend durch R definiert), einen Alkyl-
oder substituierten Alkylrest bedeutet (wobei der Alkylrest mit mindestens einem Halogenatom, Cyano-, Nitro-,
Amino- oder Mercaptogrupye substituiert sein kann).
Bevorzugte [[[3-substituierte-2-Oxo-1-imidazolidinyl]-carbonyl]
-amino-arylacetylreste der vorstehenden Formeln sind
solche, in denen R eine Phenyl- oder 2-Thienylgruppe ist. Ferner sind solche Verbindungen bevorzugt, in denen R^ ein
Wasserstoffatom, eine Methylsulfonyl-, Phenylmethylenamino-
oder 2-Furylmethylenaminogruppe darstellt.

Die Bezeichnungen "Salz" und "Salze" betreffen basische Salze
mit anorganischen und organischen Basen. Solche Salze umfassen Ammoniumsalze, Alkalimetallsalze, wie Natrium- und
Kaliumsalze, die bevorzugt sind, Erdalkalimetallsalze, wie Calcium- und Magnesiumsalze, Salze mit organischen Basen,
wie Dicyclohexylamin-, Benzathin, N-Methyl-D-glucamin- und
Hydrabaminsalze, Salze mit Aminosäuren, wie Arginin und Lysin. Die nicht toxischen pharmazeutisch verträglichen Salze
sind bevorzugt, wobei aber auch andere Salze wertvoll sind, beispielsweise zum Gewinnen und Reinigen der Produkte.
Die Salze werden in üblicher Weise durch Umsetzung der Produkte in Form der freien Säure mit mindestens einem Äquivalent
einer geeigneten Base hergestellt mit der Maßgabe, daß
^ das gewünschte Kation in eirem Lösungsmittel oder Medium vorliegt,
in dem das Salz unlöslich ist, oder in Wasser, wobei das Wasser dann vorzugsweise durch Gefriertrocknung entfernt
wird. Durch Neutralisierung der Salze mit einer unlöslichen Säure, beispielsweise einem Kationenaustauscherharz
in der H -Form (beispielsweise ein Polystyrol-Sulfonsäureharz,
wie Dowex 50) oder mit einer wäßrigen Säure und Extraktion mit einem organischen Lösungsmittel, wie Essigsäureäthylester
oder Dichlormethan, kann die freie Säureform erhalten werden und gewünschtenfalls in ein anderes Salz umgewandelt
werden.
ß-Lactame mit einem Substituenten der Formel

der 3-Stellung enthalten mindestens ein Asymmetriezentrum,
nämlich das Kohlenstoffatom (in 3-Stellung des ß-Lactam-Kerns),
an das der Amino- oder Acylaminosubstituent gebun-

den ist. Die Erfindung ist auf solche ß-Lactame gemäß vorstehender
Beschreibung gerichtet, in denen die Stereochemie am Asymmetriezentrum in 3-Stellung des ß-Lactamrings der
Konfiguration des Kohlenstoffatoms in 6-Stellung der natürlieh
vorkommenden Penicilline (beispielsweise Penicillin G) und der Konfiguration des Kohlenstoffatoms in 7-Stellung
der natürlich vorkommenden Cephamycine (beispielsweise Cephamycin C) entspricht.
Bei den bevorzugten ß-Lactamen der Formel I wurden die Strukturformeln
so gezeichnet, daß die Stereochemie am Asymmetriezentrum in 3-Stellung zu sehen ist.
Zum Gegenstand der Erfindung gehören auch Razemate, die die vorstehend beschriebenen ß-Lactame enthalten.
ß-Lactame mit einem Substiiuenten der Formel
-0-C-SO3H
(oder einem Salz davon) in 1-Stellung des ß-Lactamkerns und
einem Acylaminosubstituenten in 3-Stellung des ß-Lactamkerns sind wirksam gegen eine Reihe von gram-negativen und grampositiven
Organismen. Der Substituent der Formel
-0-C-SO3H
(oder ein Salz davon) ist von ausschlaggebender Bedeutung für die Aktivität der Verbindungen der Erfindung.
Die Verbindungen der Erfindung eignen sich als Arzneimittel zur Behandlung von bakteriellen Infektionen, einschließlich
Harnwegsinfektionen und Infektionen des Atemtrakts bei Säugern, wie Haustieren, beispielsweise Hunde, Katzen, Kühe
und Pferde, sowie beim Menschen.

Zur Behandlung von bakteriellen Infektionen bei Säugern werden
die Verbindungen der Erfindung in einer Menge von etwa 1,4 mg/kg/Tag bis etwa 350 mg/kg/Tag, vorzugsweise von etwa
14 mg/kg/Tag bis etwa 100 mg/kg/Tag gegeben. Alle Verabreichungsarten, die in der Vergangenheit zur Gabe von Penicillinen
und Cephalosporinen verwendet wurden, kommen auch für die neue Gruppe von ß-Lactamen der Erfindung in Betracht.
Hierzu gehören orale, intravenöse und intramuskuläre Verabreichung sowie die Gabe alt Suppositorium.

In einer anderen Ausführungsform kann die Cyclisierung der

Verbindung der Formel V auch dadurch erreicht werden, daß zunächst die Hydroxylgruppe in eine austretende Gruppe umgewandelt wird. Behandlung der Verbindung der Formel V (oder ein Salz davon) mit Triphenylphosphin und Diäthylazodicarboxylat oder Tetrachlorkohlenstoff und Triäthylamin ergibt eine Verbindung der Formel VII.
Verbindung der Formel V auch dadurch erreicht werden, daß zunächst die Hydroxylgruppe in eine austretende Gruppe umgewandelt wird. Behandlung der Verbindung der Formel V (oder ein Salz davon) mit Triphenylphosphin und Diäthylazodicarboxylat oder Tetrachlorkohlenstoff und Triäthylamin ergibt eine Verbindung der Formel VII.
Die beiden genannten Verfahren zum Ringschluß einer Verbindung der Formel V ergeben eine Inversion der Stereochemie an
dem an die Reste !R3 und R. gebundenen Kohlenstoffatom.
Die Abspaltung der Schutzgruppe vom 3-Aminosubstituenten der Verbindung der Formel VII kann mit auf dem Fachgebiet bekann
ten Verfahren erreicht werden. Wenn die Schutzgruppe beispielsweise eine tert.-Butoxycarbonylgruppe ist, kann Trifluoressigsäure
und Anisol zur Abspaltung der Schutzgruppe verwendet werden. Wenn die Schutzgruppe eine Benzyloxycar
bony!gruppe ist, kann katalytische Hydrierung, beispielsweise
mit Palladium auf Aktivkohle, angewandt werden. Wenn die Schutzgruppe eine o-Nitrophenylsulfenylgruppe ist,
kann p-Toluolsulfonsäure in Verbindung mit p-Thiokresol benutzt
werden. Die von der Schutzgruppe befreite Verbindung hat die Formel VIII

Spezielle Beispiele für selche Verfahren sind die Umsetzung
mit einer Carbonsäure (R1-OH) oder einem entsprechenden Carbonsäurehalogenid
oder -auhydrid. Die Umsetzung mit einer
Carbonsäure verläuft am besten in Gegenwart eines Carbodiimids,
wie Dicyclohexylcarbodiimid und eines Stoffes, der in situ ein reaktionsfähiges Zwischenprodukt erzeugen kann,

Die Umsetzungsteilnehmer und Verfahren der N-Chlorierung von
Amiden sind auf dem Fachgebiet bekannt. Beispiele für Umsetzungsteilnehmer
sind tert.-Butylhypochlorit, Natriumhypochlo rit und Chlor. Die Umsetzung kann in einem organischen Lö-
sungsmittel, beispielsweise in einem niederen Alkanol, wie

Die Umsetzung kann in einem organischen Lösungsmittel, beispielsweise
einem polaren organischen Lösungsmittel, wie Tetrahydrofuran, bei verminderter Temperatur durchgeführt
werden.
In einer anderen Ausführungsform kann eine Verbindung der Formel VII, in der A1 eine Benzyloxycarbonylgruppe ist, unter
Anwendung eines einstufigen Verfahrens in eine Verbindung der Formel XI umgewandelt werden. Das Methoxylierungsmittel
kann zunächst mit einer Verbindung der Formel VII gemischt und das N-Chlorierungsreagenz zu dem Reaktionsgemisch zugegeben
werden.
Die Umwandlung einer Verbindung der Formel XI in das gewünschte
Produkt der Formel I kann nach den vorstehend für die Umwandlung eines Zwischenprodukts der Formel VII in eine
Verbindung der Erfindung durchgeführt werden.

Dikaliumsalz von [3S-[3a(Z),4ß]]-2-[[[1-(2-Amino-4-thiazolyl)-2-[[4-methyl-2-oxo-1-(sulfomethoxy)-3-azetidinyl]-
amino]-2-oxoäthyliden]-amino]-oxy]-2-methy!propionsäure
A) Aminoxymethansulfonsäure
Eine Suspension von 0,8 σ (20 mMol) 60prozentige Mineralöldispersion
von Natriumhydrid in 16 ml wasserfreies Dimethylsulfoxid
wird mit 1,46 g (20 mMol) Acetonoxim versetzt. Danach werden absatzweise 3,94 g (2OmMoI) Natriumbrommethansulfonat
zugegeben. Das Reaktionsgemisch wird 4 Stunden unter Stickstoff auf 90 bis 95 C erhitzt, dann
abgekühlt und zweimal mit 250 ml Diäthyläther gewaschen.
Das Produkt verfestigt sich, wird mit 100 ml Dichlormethan gewaschen, filtriert und über P2 0R 9etrocknet. Ausbeute:
15,3 g Rohprodukt, das in 20 ml Wasser gelöst und mit 7,5 g (22 mMol) Tetrabutylammoniumhydrogensulfat versetzt wird.
Das erhaltene ionengepaarte Produkt wird zweimal mit 200 ml Dichlormethan extrahiert. Die Dichlormethanlösung wird
über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Sodann wird durch 4 Stunden Erhitzen in 120 ml
2N HCl bei 130°C das Acetonoxim hydrolysiert. Die erhaltene Lösung wird unter vermindertem Druck zweimal aus Wasser
und dann aus Acetonitril eingedampft. Das Produkt wird durch Zugabe von Dichlormethan ausgefällt, filtriert und
unter vermindertem Druck getrocknet. Ausbeute: 2,5 g der Titelverbindung.

B) Kaliumsalz von O-Sulfomethyl-a-N-t-butoxycarbonyl-L-threonin-hydroxamat
Eine Lösunq vor 1,96 g (8,9 mMol) tert.-Butoxycarbonyl-L-threonin
in 16 ml Wasser und 4 ml Tetrahydrofuran wird bei 0 C mit 1,14 g (8,9 mMol) Aminoxymethansulfonsäure versetzt.
Der pH-Wert wird mit 1F KOH auf 4,5 eingestellt und danach werden 1,87 g (9,7 mMol) 1-Äthyl-2-(3-dimethylaminopropyl)-carbodiimid-hydrochlorid
in 8 ml Wasser tropfenweise zugegeben. Das Reaktionsgemisch wird 2 Stunden bei Raumtemperatur gerührt, wobex der pH-Wert durch gelegentliche
Zugabe von 1N H3SO. auf 4 bis 4,5 gehalten wird.
Sodann wird mit dem Produkt lurch Zugabe von 3,05 g (8 mMol) Tetrabutylammoniumhydrogensulfat bei pH 2,8 die Ionenpaarung
durchgeführt und das Erodukt aus wäßriger Lösung mit 4 χ 100 ml Dichlormethan extrahiert. Die erhaltene Dichlormethanlösung
wird über Natriumsulfat getrocknet und unter vermindertem Druck eingedampft. Ausbeute: 4,5 g Produkt
als Tetrabutylammoniumsalz. Dieses wird durch Ionenaustausch an 150 ml Dowex 50 X (0,7 mäq K /ml) in das Kaliumsalz
umgewandelt. Nach Lyophilisierung werden 2,63 g der Titelverbindung erhalten.
C) Tetrabutylammoniumsalz von O-Sulfomethyl-a-N-t-butoxycarbonyl-L-(O-methansulfonylthreonin)-hydroxamat
Eine teilweise Lösung von 2,?7 g (6,5 mMol Kaliumsalz von
O-Sulfomethyl-a-N-tert.-butoxycarbonyl-L-threonin-hydroxamat
in 50 ml wasserfreies Pyridin wird bei 0 bis 5 C unter Stickstoff tropfenweise mit 0,8 ml (Überschuß) Methansulfonylchlorid
versetzt. Das Reaktionsgemisch wird 4 Stunden bei Raumtemperatur gerührt und dann unter vermindertem Druck
eingedampft. Der Rückstand wird in 10 ml Wasser gelöst und
bei pH 2,8 mit 2,0 g (6 mMol) Tetrabutylammoniumhydrogensulfat
versetzt. Das ionengepaarte Produkt wird mit Chloroform extrahiert. Die Chlorofcrmlösung wird getrocknet und
unter vermindertem Druck eingedampft. Es werden 2,6 g Rohprodukt
erhalten.
D) Kciliumsal'/. von [3S- (3α, 4ß ) ] -3- [ Π 1 ,W)imoJLhyJ. üt hoxy) carbonyl]-amino]-4-methyl-2-oxo-1-(sulfomethoxy)-azetidin
2,6 g (4f0 mMol) Tetrabutylammoniumsalz von =-Sulfomethyl-a-N-tert.-butoxycarbonyl-L-^O-methansulfonylthreonin)-hydroxamat
werden in 5 ml Aceton gelöst und tropfenweise zu einer unter Rückfluß siedenden Suspension von 2,2 g Kaliumcarbonat
in 65 ml Aceton gegeben. Das Gemisch wird weitere 3,5 Stunden unter Rückfluß gekocht, dann abgekühlt, filtriert und
unter vermindertem Druck eingedampft- Der Rückstand wird in 10 ml 0,5 M KH2PO. vom pH-Wert 5,5 gelöst und der pH-Wert
auf 2,8 eingestellt. Das Produkt wird viermal mit 100 ml Dichlormethan extrahiert, die vereinigten Extrakte werden
getrocknet und unter vermindertem Druck eingedampft.
Ausbeute: 1,52 g rohes ionengepaartes Tetrabutylammonium-ßlactamsalz.
Das Kaliumsalz wird durch Ionenaustausch an 50 ml Dowex 50 X (0,7 mäq K /ml) erhalten. Nach der Lyophilisierung
werden 0,53 g Rohprodukt erhalten, das durch Chromatographie an 100 ml HP-20 mit Wasser als Laufmittel weiter
gereinigt wird. Die entsprechenden Fraktionen werden vereinigt und lyophilisiert. Ausbeute: 0,245 g.

-azetidin werden in 0,5 ml Dichlormethan und 0,5 ml Anisol suspendiert. Das Gemisch wird dann auf 0 C abgekühlt
und unter Stickstoff mit 1,0 ml Trifluoressigsäure versetzt.
■j Das Reaktionsgemisch wird 1 Stunde gerührt und dann unter
vermindertem Druck eingedampft. Der Rückstand wird zweimal aus Benzol eingedampft. Danach wird der Rückstand mit Diäthyläther
digeriert. Der Äther wird abdekantiert, wobei das gewünschte Produkt als weußer Feststoff erhalten wird.
"F) Kaliumsalz von [3S-[3a(Z),4ß]]-2-[[[1-(2-Amino-4-thiazolyl) -2-[[4-methyl-2-oxo-1-(sulfomethoxy)-3-azetidinyl]-amino]-2-oxoäthyliden]-amino]-oxy]-2-methylpropionsäurediphenylmethy!ester
. 0,30 g (0,68 mMol) (Z)-2-Amino-a-[[2-(diphenylmethoxy)-1,1 dimethyl-2-oxoäthoxy]-imino]-4-thiazolessigsäure
und 0,10 g (0,68 mMol) 1-Hydroxybenzotriazolhydrat werden unter Stickstoff
in 4 ml wasserfreies Dimethylformamid gelöst. Die Lösung wird auf 0 C abgekühlt und absatzweise mit 0,14 g
(0,68 mMol) Ν,Ν'-Dicyclohexylaarbodiimid versetzt. Nach
der Zugabe wird das Reaktionsgemisch 1 Stunde bei 0 C gerührt. Sodann wird das Gemisch mit einer Lösung von etwa
0,68 mMol des vorstehend hergestellten rohen 3-Amino-1-(sulfomethoxy)-azetidins
in 10 ml Dimethylformamid und 0,5 ml Ν,Ν-Diisopropyläthylamin bei 00C versetzt. Das Reaktionsgemisch
wird 1 Stunde bei 0 C und dann etwa 15 Stunden bei Raumtemperatur gerührt. Sodann wird die Lösung filtriert
und das Filtrat unter vermindertem Druck eingedampft. Der Rückstand wird in 50 ml Dichlormethan gelöst und mit 2 ml
Wasser gewaschen. Nach dem Verdampfen des Dichlormethans werden 0,372 g Rohprodukt erhalten. Dieses wird durch 30 ml
Dowex 50 (0,7 mäq K /ml) mit Wasser als Laufmittel geführt.
Nach der Lyophilisierung werden 0,211 g Rohprodukt erhalten, das mit Hydroxybenzotriazol verunreinigt ist.

G) Dikaliumsalz von [3S-[3α(Ζ),4ß]]-2-[[[1-(2-Amino-4- thiazolyl)-2-[[4-methyi-2-oxo-1-(sulfomethoxy)-3-azetidinyl]-amino]-2-oxoäthyliden]-amino]-oxy]-2-methylpropionsäure
0,211 g Kaliumsalz von [3S-[3α(Ζ) ,4ß]]-2-[[[1 -(2-Amino-4-thiazolyl)-2-[[4-methyl-2-oxo-1-(sulfomethoxy)-3-azetidinyl]
amino]-2-oxoäthyliden]-amino]-oxy]-2-methylpropionsäurediphenylmethylester
werden in 1,8 ml Dichlormethan, 0,5 ml
Anisol und 1,5 ml Trifluoressigsäure gelöst und unter Stickstoff bei 0°C 2 Stunden gerührt. Dann wird das Reaktionsgemisch
unter vermindertem Druck eingedampft und anschließend zweimal aus Benzol eingedampft. Der Rückstand
wird mit Diäthyläther und Essigsaureäthylester im Verhältnis
1 : 1 und dann mit Diäthyläther und Acetonitril im Verhältnis 1 : 1 gewaschen, wobei ein weißer Feststoff erhalten
wird. Dieser wird in 1,0 ml 0,5 M KH„PO.-Lösung vom
pH-Wert 5,5 gelöst, mit 1 N KOH auf den pH-Wert 6,5 eingestellt und durch 40 ml HP-20 mit Wasser als Laufmittel
chromatographiert. Ausbeute: 53 mg der Titelverbindung vom F. 200°C (Zers.).

A) Erythro-3-hydroxy-DL-aspartinsäure
Erythro-3-hydroxy-DL-aspartinsäure wird aus trans-epoxybernsteinsäure
nach der Beschreibung von C.W. Jones und Mitarb., Can. J. Chem. Bd. 47 (1969), S. 4363 hergestellt.
Trans-Epoxybernsteinsäure wird aus Fumarsäure nach der Beschreibung
von G.B. Payne und Mitarb., J. Org. Chem. Bd. 24 (1959), S. 54 erhalten.
B) ß-Methyl-erythro-3-hydroxy~DL-aspartat
Eine Suspension von 5,0 g (33,5 mMol) Erythro-3-hydroxy-DL-aspartinsäure
in 50 ml wasserfreies Methanol wird mit 6 ml konzentrierte Salzsäure versetzt. Das Gemisch wird
3 Stunden unter Rückfluß gekocht, dann auf Raumtemperatur abgekühlt und unter vermindertem Druck eingedampft. Der
Rückstand wird in 95 % Äthanol aufgenommen und der pH-Wert durch Zugabe von Pyridin auf 8,0 eingestellt. Der erhaltene
weiße Feststoff wird abfiltriert und getrocknet.

Es werden 5,2 g (95 % Ausbeate) des gewünschten Methylesters vom F. 210 C erhalten.
C) Erythro-3-hydroxy-DL-asparagin

5
4,0 g (24,5 mMol) des vorstehend erhaltenen ß-Methylaspartats
werden in 40 ml konzentriertes Ammoniak gelöst und etwa 15 Stunden bei Raumtemperatur gerührt. Dann wird
das Reaktionsgemisch unter vermindertem Druck eingedampft und der reste Rückstand in heißem Wasser gelöst. Der
pH-Wert wird mit 6 N HCl auf 5,0 eingestellt, die Lösung unter vermindertem Druck auf etwa 20 ml eingeengt und
dann etwa 15 Stunden bei 5 C stehengelassen. Die erhaltene
weiße kristalline Masse wird abgetrennt und getrocknet.
Ausbeute: 3,4 g (95 %) der Titelverbindung vom F. 233°C.
D) Kaliumsalz von n-tert.-Butoxycarbpnyl-erythro-3-hydroxy-DL-asparagin
7,0 g (47 mMol) Erythro-3-hydroxy-DL-asparagin werden in 50 ml Wasser suspendiert und durch Zugabe von 3N KOH in
Lösung gebracht. Diese Lösung wird auf den pH-Wert 10,0
eingestellt und auf diesem pH gehalten, während tropfenweise eine Lösung von 15,5 g (71 mMol) Di-tert.-butyldicarbonat
in 20 ml tert.-Butanol zugegeben wird. Das Reaktionsgemisch wird etwa 15 Stunden bei Raumtemperatur und pH 10,0
gerührt. Dann wird das tert.-Butanol unter vermindertem Druck abdestilliert, der wäßrige Rückstand wird mit 6N HCl
auf pH 5,0 eingestellt und d~e Lösung unter vermindertem
Druck auf ein kleines Volumen eingeengt. Sodann wird der pH-Wert mit 6 N HCl auf 3,0 eingestellt und das Lösungsmittel
unter vermindertem Druck entfernt. Es werden 20 g Feststoff erhalten, die das gewünschte Produkt in Form der
freien Säure und in etwa quantitativer Ausbeute zusammen mit anorganischen Salzen enthält. Ein Anteil von 2,9g
dieses Feststoffs wird in Wasser aufgenommen und mit ver-

dünnter KOH auf pH 6,5 eingestellt. Die Lösung wird dann durch 100 ml HP-20-Harz unter Verwendung von Wasser und
dann von Aceton/Wasser im Verhältnis 1 : 9 als Laufmittel chromatographiert. Es wird das gewünschte Kaliumsalz als
Rückstand (1,8 g) erhalten.
E) Tetrabutylammoniumsalz von Aminoxymethansulfonsäure
Eine Lösung von 0,635 g (5 mMol) Aminoxymethansulfonsäure in 2 ml Wasser wird mit 1,02 g (3 mMol) Tetrabutylammoniumhydrogensulfat
versetzt und der pH-Wert der Lösung mit verdünnter KOH auf 10,0 eingestellt. Dann wird das Wasser
unter vermindertem Druck abdestilliert und der Rückstand mit Methylenchlorid digeriert und filtriert. Das Filtrat
wird über Natriumsulfat getrocknet und eingedampft. Ausbeute: 1,06 g (2,88 mMol) der Titelverbindung.
F) Kaliumsalz von O-Sulfomethyl-a-n-tert.-butoxycarbonylerythro-3-hydroxy-DL-asparagin-hydroxamat
0,792 g (2,77 mMol) Kaliumsalz von n-tert.-Butoxycarbonylerythro-3-hydroxy-DL-asparagin
werden in 3 ml Wasser gelöst und mit verdünnter K„SO auf pH 2,4 eingestellt. Dann
wird die Lösung unter vermindertem Druck zu einem Rückstand konzentriert, der zweimal aus Wasser und dann zweimal aus
Benzol eingedampft wird. Dieser Rückstand wird in 4 ml wasserfreies
Acetonitril und 8 ml wasserfreies Tetrahydrofuran gelöst und auf 0°C abgekühlt. Sodann wird die Lösung
unter Rühren mit 0,424 g (2,77 mMol) 1-Hydroxybenzotriazolmonohydrat
und hierauf mit 0,572 g (2,77 mMol) N,N1-Dicyclohexylcarbodiimid
versetzt. Das Gemisch wird 2 Stunden bei 0°C gerührt und danach tropfenweise mit 1,06 g
(2?S8 mMol) des vorstehend hergestellten Tetrabutylammoniuidsalzes
von Aminoxymethansulfonsäure in 5 ml Acetonitril versetzt. Nach der Zugabe wird das Reaktionsgemisch
4 Stunden bei 0°C gerührt, dann filtriert und unter ver-

mindertem Druck zu einem Rücketand eingeengt. Dieser wird mit 10 ml Wasser versetzt und bei 00C bis zur Verfestigung
digeriert. Der erhaltene Feststoff wird abfiltriert, das wäßrige Filtrat auf etwa 3 ml eingeengt, sein pH-Wert auf
4,0 eingestellt und eine Fraktionierung über 80 ml Dowex 50 (K+)-Ionenaustauscherharz durchgeführt. Die produkthaltigen
Fraktionen werden gesammelt und zu einem kleinen Volumen eingeengt. Der pH-Wert wird a if 3,4 eingestellt und die Lösung
über HP-20-Harz chromatoyraphiert, wobei nach der Lyophilisierung 502 mg (46 %) der Titelverbindung als Feststoff
vom F. 145°C erhalten werden.

G) Tetrabutylammoniumsalz von O-Sulfomethyl-a-n-tert.-butoxycarbonyl-erythro-3-hyaroxy-DIi-asparagin-hydroxamat
Eine Lösung von 104 mg (0,263 mMol) des vorstehend erhalte
nen Kaliumsalzes von Asparaginhydroxamat in 1 ml Wasser wird mit 2,63 ml 0,1 M Tetrabutylammoniumhydrogensulfat in
0,5 M KH0PO4-Pufferlösung vom pH-Wert 5,5 versetzt. Die Lö
sung wird dann zur Trockene eingedampft und der Rückstand mit Methylenchlorid digeriert. Filtrieren und Eindampfen
des Methylenchlorids ergibt 156 mg (0,26 mMol) der Titelverbindung als Rückstand.
H) Kaliumsalz von (2a,3a)-i-Sulfomethoxy-2-(aminocarbonyl).
3-[[(1,1-dimethyläthoxy)-carbonyl]-amino]-4-oxoazetidin
140 lag (0,234 mMol) des vorstehend erhaltenen Tetrabutyl-..
ammor.iumsalzes von O-Sulfomethylhydroxamat werden in 1,6 ml
wasserfreies Methylenchlorid gelöst und etwa 15 Stunden mit
1 g trockenes 3A Molekularsieb gerührt. Die, Lösung wird mit

einer Spritze entfernt und mit 0,8 ml wasserfreies Methylen
chlorid verdünnt. Dann wird die Lösung unter Rühren bei -50°C unter Argon mit 0,12 ml (0,85 mMol) Triäthylamin und
dann tropfenweise mit 0,08 ml (0,42 mMol) Trifluormethansulfonsäureanhydrid
versetzt. Das Reaktionsgemisch wird innerhalb von 1 Stunde auf -30°C erwärmt und dann mit
0,06 ml Triäthylamin versatzt. Nach 5 Minuten wird das Reaktionsgemisch unter vermindertem Druck eingedampft und der
Rückstand in 2 ml Aceton gelöst. Die Lösung wird auf 0 C ab gekühlt und mit einer Lösung von 79 mg Kaliumperfluorbutansulfonat
in 1 ml Aceton versetzt. Sodann wird Diäthyläther zugegeben und die Feststoffe abzentrifugiert. Behandlung
der Feststoffe durch Rühren mit 2 ml Dowex 50 (K )-Harz in Wasser, Filtration und Entfernung des Wassers unter vermindertera
Druck ergibt das rohe Kaliumsalz, das keine Amine enthält. Durch Chromatographie dieses rohen Kaliumsalzes
an HP-20-Harz unter Verwendung von Wasser als Laufmittel werden 15 mg (20 %) der Titelverbindung erhalten, die nach
Lyophilisierung den F. 149 C aufweist.
IR-Spektrum (KBr): 1786 cm (ß-Lactam-carbonyl) und
1696 cm (Breit; Amid- urd Carbamat-carbonyl).
