Description
Die Erfindung betrifft neue N-Aryl-oxazolidin-2-one, Verfahren
zu ihrer Herstellung und solche Verbindungen enthaltende Arzneimittel.
Im einzelnen entsprechen die neuen Verbindungen der allgemeinen
Formel
worin R
die (2-Acetyl)-äthyloxy-Gruppe CH3CO-(CH2)2~O,
die (5-Cyano)-n-pentyl-Gruppe Cft-(CH2)5~,
den p-Chlorphenyl- oder m-Cyanophenylrest,
die (2-Cyclohexyl)-äthinyl-Gruppe /~~Λ c=
!-, oder
eine Gruppe der Struktur
-A-B-
bedeutet,
worin R1 ein Halogenatom oder die Cyanogruppe bedeutet
und die Gruppierung -A-B- eine der folgenden Bedeutungen
hat: -CH2-CH2-, -CO-CH2-, -0-CH3-, -C=C-, -CH2-CH2-O-.
Auch betrifft die Erfindung Verfahren zur Herstellung dieser Verbindungen.
A) Die Verbindung der Formel (I), in der R die Gruppierung
CH3CO-(CH2)2~0- bedeutet,wird durch saure Hydrolyse der
Verbindung der Formel
erhalten, wobei die sauren Bedingungen vorzugsweise mit
Salzsäure und Schwefelsäure eingestellt werden und die Hydrolyse in einem organischen Lösungsmittel erfolgt,
wie in THF oder Aceton.
Die Verbindung der Formel (II) wird durch Kondensation des Chlor-, Brom-, Mesyl- oder Tosylderxvats des 2-Hydroxyäthyl-2-methyl-dioxolans-1,3,
vorzugsweise in DMF-, Aceton- oder Acetonitril-Medium und in Gegenwart von
Kaliumcarbonat oder Kaliumhydrid mit der Verbindung der Formel
(HD,
hergestellt gemäß der BE-PS 876 831, erhalten.
B) Die Verbindung der Formel (I), worin R CN-(CH2)5~ bedeutet,
wird nach einer zweistufigen Synthese erhalten, die im Behandeln der Verbindung der Formel
-OCH-
(IV)
mit Mesylchlorid in einem organischen Lösungsmittel, wie Methylenchlorid, vorzugsweise in der Kälte und in Gegenwart
einer Base, wie Triäthylamin, dann in der Einwirkung von Kalium- oder Natriumcyanid in einem organischen Lösungsmittel,
wie DMSO, besteht.
Die Verbindung der Formel (IV) wird durch katalytische Debenzylierung in äthanolischem Medium und in Gegenwart
von 5 % Pd/C der Verbindung der Formel
erhalten, die ihrerseits durch Cyclisieren der Verbindung
der Formel
OCH.
(VI)
durch eine 10 %ige Phosgenlösung in Dichloräthan in Gegenwart
von Kaliumcarbonat und Triäthylamin erhalten wird, die ihrerseits durch Kondensation von 3-Methoxy-1
j 2-epoxypropan in alkoholischem Medium mit dem Anilin
der Formel
(ViI)
erhalten wird. Das Anilin der Formel (VII) wird seinerseits
durch Reduktion des Ketoanilins der Formel

(VIII)
durch Triäthylsilan (HSiEt-J in Gegenwart von Trifluoressigsäure
erhalten. Diese letztere Verbindung wird durch Reduktion des Nitroderivats der Formel

mit Eisen in Gegenwart von io%igem AmmonJumchlorid in
Wasser und Alkohol erhalten, das seinerseits durch Kondensation von 4-Benzyloxy-n-butanal in Gegenwart von
Kalilauge in methanolischem Medium und THF mit of-Phenylamino-p-nitrobenzylester
der diphenyl-phosphorigen Säure erhalten wird.
Zu bemerken ist, daß es auch möglich ist, die Verbindungen der Formel (V) durch Cyclisieren der Verbindung der
Formel

Medium zu erhalten.
C) Die Verbindungen der Formel (I), für die R p-Chlorphenyl,
m-Cyanophenyl, m-Cyanobenzoylmethyl oder m-Halogenbenzoylmethyl
ist, werden
entweder durch Cyclisieren der Verbindung der Formel
entweder durch Cyclisieren der Verbindung der Formel

worin R„ die vier speziellen Bedeutungen für R, wie oben
aufgezählt, annimmt, mit äthanolischer Kalilauge oder Natriummethylat in methanolischem Medium erhalten. Die
Verbindungen der Formel (X) werden durch Einwirkung von Phosgen auf 1-Chlor-3-methoxy-propanol-2, dann durch
Umsetzen der so hergestellten Zwischenverbindung mit einem
Anilin der Formel

Formel (IXa) ebenso wie für die Herstellung der Verbindungen der Formel (IX) beschrieben hergestellt werden,
aber ausgehend von m-Halogenbenzaldehyd oder m-Cyanobenzaldehyd
anstelle von 4-Benzyloxy-n-butanal, oder durch Cyclisieren der Verbindung der Formel
(KfS),

worin R? die gleichen Bedeutungen wie in der Formel (X)
hat, mit Natriummethylat in Toluollösung, erhalten durch
Einwirkung von Chlorameisensäureäthylester in Dichloräthanlösung auf die Verbindung der Formel
(Xb),
worin R„ die gleichen Bedeutungen wie -in Formel (Xa) hat,
wobei die Verbindungen der Formel (Xb) ihrerseits durch Kondensation der Verbindung der Formel (XI) mit dem
1-Chlor-3-methoxy-propanol-2 erhalten werden.
D) Die Verbindungen der Formel (I), für die R m-Halogenphenethyl
oder m-Cyanophenetyl bedeutet, werden durch Reduktion der Verbindung der Formel (I) # für dieRdie m-Halogenbenzoylmethyl-
oder m-Cyanobenzoylmethyl-Gruppe bedeutet, vorzugsweise
mit Triäthylsilan (SiEt3H) in Gegenwart von Trifluoressigsäure
erhalten, wobei die Synthese dieser speziellen Verbindungen der Formel (I) nach den im vorigen Abschnitt
C) beschriebenen Arbeitsweisen erfolgt.
E) Die Verbindungen der Formel (I), für die R (2-Cyclohexyl)-äthinyl,
(2-m-Ualogenphenyl)-äthinyl oder (2-m-Cyanophenyl)-äthinyl bedeutet, werden durch Kondensieren der Kupfersalze
des (2-Cyclohexyl)-äthins, (2-m-rHalogenphenyl)-äthins
und (2-m-Cyanophenyl)-äthins mit 3-p-Jodphenyl-5-methoxymethyl-oxazolidin-2-on,
hergestellt durch Cyclisieren von 3-p-Jodphenylamino-i-methoxy-propanol mit Phosgen nach
einer mit der Herstellung der Verbindung der Formel (V) identischen Arbeitsvorschrift, vorzugsweise in HMPT-Medium,
erhalten.
Die vorerwähnten Kupfersalze können leicht nach den Vorschriften der Angew. Chenu Int. Edit., 9, 464 (1970),
J.C.S. (1967), 578 und JoCoSo (1969) , 2173, erhalten werden

F) Die Verbindungen der Formel (I), für die R m-Halogenphenetyloxy
oder xn-Cyanophenetyloxy bedeutet, werden durch Kondensieren
von m-Halogen- oder m-Cyanophenetylalkohol mit 3-p-Hydroxyphenyl-5-methoxymethyl-oxazolidin-2-on,
hergestellt, wie in der BE-PS 876 831 beschrieben, in THF-Mediuxn in Gegenwart
von Jithyl-azobis-ciicarboxylat und Tripheny!phosphin
erhalten.
G) Die Verbindungen der Formel (I), für die R eine m-Halogenphenoxymethyl-
oder m-Cyanophenoxymethyl-Gruppe bedeutet, werden erhalten
entweder durch Kondensieren, vorzugsweise in THF-Medium und
in Gegenwart von Äthyl-azobisdicarboxylat (EtOCO-N=N-COOEt)
und Tripheny!phosphin, von xn-Halogenphenol oder m-Cyanophenol
mit 3-p-(Hydroxymethy1)-phenyl-5-methoxymethy1-oxazolidin-2-on,
welch letztere Verbindung durch Reduzieren von 3-p-Formylphenyl-5-methoxymethyl-oxazolidin-2-oni
vorzugsweise mit Natriuinborhydrid, erhalten wird, das wiederum durch
saure Hydrolyse der Verbindung der Formel
(ΧΠ)
erhalten wird, ihrerseits erhalten durch Cyclisieren der Verbindung der Formel
[V)
mit äthanolischer Kalilauge oder Natriuiranethylat, erhallten
nach dem schon für die Synthese der Verbindungen der Formel (X) beschriebenen Verfahren;
oder durch Kondensieren des Mesylats des 3-p-(Hydroxymethyl)-phenyl-5-methoxymethyl-oxazolidin-2-ons
mit m-Halogen- oder m-Cyanophenol, vorzugsweise in DMF-Medium in
Gegenwart von Natriumhydrid.
Die folgenden Herstellungen dienen der weiteren Veranschaulichung der Erfindung.
3-p- (2-Acetyl) -äthoxyphenyl-S-methoxymethyl-oxazolidin^-on
(I), Kennziffer 238
Man rührt 2 Ii bei Raumtemperatur eine Lösung von 3,4 g der ·
Verbindung der Formel (II) und 5 ml Salzsäure in 50 ml Aceton, gibt dann 1 ml ~2n Schwefelsäure zu und läßt weitere 5 h
rühren. Man neutralisiert mit einer Natriumbicarbonatlosung, extrahiert mit Methylenchlorid, trocknet über Natriumsulfat,
filtriert, dampft das Filtrat ein, kristallisiert den Rückstand aus Alkohol, kristallisiert dann aus einem Gemisch aus
Alkohol und Isopropyläther um. So isoliert man 1 g der erwarteten
Verbindung.

Zu einer Lösung von 5,6 g S-p-Hydroxyphenyl-S-hydroxymethyloxasolidin-2-on
(III) in 100 ml DMB1 gibt man in kleinen Anteilen
1,2 g 50 Siges Watriumhydrido Dann gibt man 9 g 2-(2-Brom)-äthyl-2-methyl-1,3-dioxolan
in 20 ml DMF zu und bringt das Gemisch 3 h auf 70-8O0C, Dann gießt man auf Eiswasser, extrahiert
mit Äthylacetat, wäscht mit Wasser, trocknet über Natriumsulfat, filtriert, dampft das Filtrat ein und chromatographiert
den Rückstand an einer Kieselgelsäule„ Durch Eluiexen mit Methylchlorid/Aceton-Gemischen (zunächst 98/2,
dann 95/5) erhält man 5,6 g der erwarteten Verbindung, die man aus Isopropyläther umkristallisiert.

5-Benzyloxy -1-p-nitrophenyl-pentan-i-on (IX)
Zu einer Lösung von 105 g ^-Phenylamino-p-nitrobenzylester
der Diphenylphosphorigensäure in 1200 ml THF, auf -40°C gekühlt, gibt man eine Lösung von 12,8 g KOH in 128 ml Methanol,
dann eine Lösung von 40,6 g 4-Benzyloxy-n-butanal in 500 ml
THB1 in 30 min, läßt 2 h bei -40°C, dann 12 h bei Raumtemperatur.
Dann verdampft man die Lösungsmittel, nimmt den Rückstand wieder in Wasser auf, extrahiert mit Äthylacetat,
verdampft das Lösungsmittel und verreibt den Rückstand in konzentrierter Salzsäure. Man verdünnt mit Wasser, extrahiert
mit Äther, verdampft das Lösungsmittel und kristallisiert den Rückstand aus Isopropyläther. So erhält man 23 g der
erwarteten Verbindung.

Nach der gleichen Arbeitsweise, aber ausgehend von entsprechenden Reaktionskomponenten, erhält man die Verbindungen

Beispiel 4
5-Benzyloxy-i-p-aminophenyl-pentan-i-on (VIII)
5-Benzyloxy-i-p-aminophenyl-pentan-i-on (VIII)
Man läßt ein Gemisch aus 23 g der Verbindung der Formel (IX), im vorherigen Beispiel erhalten, und 23 g Eisenpulver in
200 ml einer wässrigen, 10 %igen Ammoniumchloridlösung und 20 ml Äthanol 1 h bei Raumtemperatur rühren. Dann extrahiert
man mit Methylenchlorid, filtriert, wäscht das Filtrat mit
Wasser, trocknet es über Natriumsulfat, filtriert, dampft das
Filtrat ein und kristallisiert den Rückstand aus einem Gemisch aus Äthylacetat und Isopropylather« So erhält man 18,6 g
der erwarteten Verbindung.

Nach der gleichen Arbeitsweise, aber ausgehend von Verbindun gen (IXa)„ erhält man die Verbindungen der Formel (XIa)<>

Zu einem Gemisch aus 16 g der Verbindung der Formel (VIII) des vorherigen Beispiels und 21 g Triäthylsilan gibt man
30,1 ml Trifluoressigsäure und läßt 20 h bei Raumtemperatur rühren. Dann verdünnt man mit Wasser, macht mit konzentrierter
Natronlauge basisch, extrahiertmit Methylenchlorid, wäscht mit Wasser, trocknet über Natriumsulfat, filtriert, dampft
das Filtrat ein und chromatographlert den Rückstand über eine. Kieselgelsäule (Elutionsmittels Gemisch aus Methylenchlorid
(99) und Methanol (1)).. So erhält man 14 g der erwarteten Verbindung in Form eines Öls (Ausbeute 92 %)»

Nach der gleichen Arbeitsweise, aber ausgehend von entsprechenden
Reaktionskomponenten,erhält man die Verbindungen der Formel (I) und der Kennziffern 271 und 272 der folgenden

Zu einer auf Rückfluß gebrachten Lösung von 13,9 g der Verbindung der Formel (VII) des vorherigen Beispiels in
80 ml Äthanol fügt man in 4 h eine Lösung von 4,61 g
3-Methoxy-1,2-epoxy-propan in 80 ml Äthanol. Man läßt noch
5 h unter Rückfluß, verdampft dann das.Lösungsmittel und
chromatographiert den Rückstand an einer Kieselgelsäule (Elutionsmittel: Methylenchlorid (98,9 %)/Methanol (1 %)/
Ammoniak (0,1 %)). So isoliert man 11 g der erwarteten Verbindung in Form eines Öls.

Zu einer Lösung.von 10,3 g der Verbindung der Formel (VI) des
vorherigen Beispiels und 8,3 g Kaliumcarbonat in 150 ml Di-' chloräthan gibt man langsam unter Beibehaltung einer Temperatur
von200C 36ml einer 10 %igen Phosgenlösung in Dichloräthan,
läßt 15 min rühren und bringt 5 h auf 50 C. Dann wäscht man mit einer Natriumbicarbonatiösung, mit Wasser, trocknet über
Natriumsulfat, filtriert, dampft das Filtrat ein und chromatographiert
den Rückstand an einer Kieselgelsäule (Elutions-

mittels Äthylacetat (40 %)/Hexan (60 %)). So erhält man 10,3 g
der erwarteten Verbindung in Form eines Öls«

Man hydrogenolysiert bei Normaldruck und Raumtemperatur 2 h im Autoklaven eine Suspension von 10 g der Verbindung der Formel
(V) des vorhergehenden Beispiels und 2 g 5 % Pd/C in 300 ml 96 %igem Äthanol» Dann filtriert man, dampft das FiI-trat
ein und erhält ein öl, das kristallisiert.

Zu einer auf O0C gekühlten Lösung von 5,.7 g der Verbindung der
Formel (IV) des vorherigen Beispiels und 7 ml Triäthylamin in 180 ml Methylenchlorid gibt man langsam 3,2 g Mesylchlorid.
Dann läßt man 1 h bei Raumtemperatur rühren, wäscht mit Wasser, trocknet über Natriumsulfat, filtriert, und dampft
den Rückstand (7,9 g) ein, den man in 60 ml DMSO löst und zu einer Lösung von 1,4 g Kaliumcyanid in 70 ml DMSO gibt. Man
bringt das Gemisch 7 h auf 50-6O0C, verdünnt dann mit Wasser,
extrahiert mit Methylenchlorid, wäscht mit Wasser, trocknet über Natriumsulfat, filtriert, dampft das Filtrat ein, chromatographiert
den Rückstand an einer Kieselgelsäule (Elutionsmittel: Hexan/Äthylacetat (60/40) und (50/5O)). So
erhält man 5,6 g der erwarteten Verbindung (Öl).
Ausbeute: 96 %

(R„ = p-Chlorphenyl) in 70 ml Äthanol gibt man eine Lösung
von 0,17 g KOH in 1 ml Äthanol und läßt 45 min rühren. Dann gießt man auf Eiswasser, extrahiert mit Äthylacetat,
wäscht mit Wasser, mit einer In Salzsäurelösung, mit Wasser, trocknet über Natriumsulfat, filtriert, dampft den Rückstand
ein und kristallisiert aus Isopropylalkohol um.

Man bringt ein Gemisch aus 4,8 g Kupfersalz des m-Chlorphenyläthins
und 6,7 g S-p-Jodphenyl-S-methoxymethyl-oxazolidin-^-
on in 100 ml HMPT unter einem Stickstoffstrom 1,5 h auf 220-23O°C.
Dann gießt man in Wasser, extrahiert mit Äthylacetat, wäscht mit Wasser, dann mit einer Kaliumjodidlosung, dann
mit Wasser, trocknet über Natriumsulfat, filtriert, dampft das Filtrat ein, kristallisiert den Rückstand aus Methanol.
Ausbeute 27 %

1,5 g m-Cyanophenethylalkohol in 50 ml THF gießt man unter
einem Stickstoffstrom 2,1 ml Äthylaaobisdicarboxylat. Dann
läßt man 14 h bei 20°C rühren, dampf das Lösungsmittel ab
und Chromatographiert den Rückstand an einer Kieselgelsäule
(Elutionsmittöl% Heptan/Üthylacetat„ 50/50). So isoliert man
die erwartete Verbindung, die man aus Methanol umkristallisiert. '

Zu einer Lösung von 37,4 g 3-p-Formylphenyl-5-methoxymethyl
oxazolidin-2-on in 200 ml Äthanol gibt man langsam 12 g
Natriumborhydrid, bringt dann 2 h auf Rückfluß, verdampft
das Lösungsmittel, nimmt den Rückstand in Wasser auf, extrahiert mit Äthylacetat, wäscht mit Wasser, trocknet
über Natriumsulfat, filtriert und verdampft das Lösungsmittel. Man erhält das erwartete Produkt in einer Ausbeute
von 54 %.

Zu einer Suspension von 3,5 g 3-Cyanophenol in 50 ml DMF
gibt man 1,4 g Natriumhydrid (50 %ig), dann 6 g 3-p-(Hydroxymethyl)-phenyl-S-methoxymethyl-oxazolidin-^-on-mesylat,
hergestellt durch Einwirkung von Mesylchlorid in Gegenwart von Triäthylamin. Man bringt das Gemisch 3 h auf 1000C. Dann
gießt man in Wasser, extrahiert mit Äthylacetat, wäscht mit Wasser, trocknet über Natriumsulfat, filtriert, dampft das
Filtrat ein und kristallisiert den Rückstand aus absolutem Alkohol. Man erhält das erwartete Produkt in einer Ausbeute
von 59 %.

Die Verbindungen der Formel (I) wurden am Labortier untersucht und haben auf psychotropem Gebiet Aktivitäten als
leistungsfähige Antidepressiva gezeigt.
Diese Aktivitäten treten in den folgenden Tests in Erscheinung :
Test A: Potenzierung des allgemeinen Zitterns bei der Maus,
hervorgerufen durch eine intraperitoneale Injektion (200 mg/kg) von dl-5-Hydroxytryptophan, nach dem Arbeitsprotokoll von
C. Gouret und G. Raynaud in J. Pharmacol- (Paris) (1974), ^,
Test B; Antagonismus gegenüber der Ptosis 1 h nach einer
intravenösen Injektion (2 mg/kg) von Reserpin bei der Maus nach dem Arbeitsprotokoll von C. Gouret und J. Thomas in J.
Pharmacol. (Paris), (1973), _4, 401.
Die Ergebnisse dieser beiden Tests sowie die einer Vergleichssubstanz, von Toloxaton, sind in der folgenden Tabelle II zusammengestellt.
Aus den in Tabelle II zusammengestellten Ergebnissen wird
man den Schluß ziehen, daß die erfindungsgemäßen Verbindungen
weitaus aktiver sind als Toloxaton, die notorisch bekannte Vergleichsverbindung.
Die erfindungsgemäßen Verbindungen stellen also Arzneimittel dar, die besonders zur Behandlung von endogenen und exogenen
depressiven Zuständen angezeigt sind.
Gegenstand der Erfindung sind ebenso auch Arzneimittel, die wenigstens eine Verbindung der Formel (I), gegebenenfalls
in Verbindung mit einem pharmazeutisch annehmbaren Träger, umfassen. Diese Mittel können verabreicht werden
entweder oral in Form von Tabletten,Dragees oder Gelatinekapseln
bei einer mittleren Gabe des wirksamen Prinzips von 50 - 500 mg/Tag
oder in Form injizierbarer Lösung bei einer Gabe des wirksamen Prinzips von 5-50 mg/Tag; das verwendete Lösungsmittel
besteht aus binären und ternären Gemischen mit z.B. Wasser, Polypropylenglykol, Polyäthylenglykol 300 oder 400
oder jedem anderen physiologischen Lösungsmittel, wobei die relativen Anteile der verschiedenen Bestandteile in Abhängigkeit
von der verabreichten Dosis eingestellt werden.
