Description
Die Erfindung betrifft Chlorderivate der Sucrose.
Erfindungsgemäß werden Chlorderivate der Sucrose bereit gestellt, das tetrachlorsubstituierte Derivat, welches Chlorsubstituenten in 1´, 4, 6, 6´-Stellung aufweist, z. B. 4,6-Dichlor-4,6-didesoxy-kleines Alpha-D-galactopyranosyl-1,6-dichlor-1,6-didesoxy-kleines Alpha-D-fructofuranosid, d. h.
-dichlor-1,6-didesoxy-kleines Beta-D-fructofuranosid, d. h.
Die Erfindung erstreckt sich auch auf die entsprechenden Ester und Acetate dieser Chlorderivate.
Diese Chlorderivate der Sucrose sind geeignete Zwischenprodukte und haben den zusätzlichen Vorteil, daß sie einen sehr süßen Geschmack besitzen, wie in der schwebenden Patentanmeldung 616/76 (Tate und Lyle) beschrieben wird, so daß sie potentielle Anwendung als Zuckersubstitut finden.
Das 1´, 6´-Dichlorderivat kann durch Umsetzung der Sucrose mit Mesitylensulfonylchlorid, Abtrennen der gewünschten Dimesitylensulfonate von dem Gemisch der substituierten Sucrosen, zum Beispiel durch Chromatographie, und Ersetzen der Mesitylensulfonyloxysubstituenten durch Chloratome unter Reaktion mit Lithiumchlorid, nachdem die restlichen freien Hydroxylgruppen durch Veresterung geschützt worden sind, hergestellt werden. Diese Veresterung kann beispielsweise mit Acetanhydrid erfolgen.
Aus den oben erwähnten Gemischen der Mesitylensulfonat-substituierten Sucrosen kann das 1´, 6, 6´-trisubstituierte Sucrosederivat, das in verhältnismäßig großem Anteil zugegen ist, abgetrennt werden. Dieses trisubstituierte Derivat kann in analoger Weise, wie oben angegeben, in das 1´, 6, 6´-trichlorsubstituierte Derivat umgewandelt werden. Aus diesem trichlorsubstituierten Derivat kann das gewünschte Tetrachlor-substituierte Derivat in guten Ausbeuten nach Esterspaltung und nachfolgender Umsetzung mit Sulfurylchlorid bei tiefen Temperaturen hergestellt werden.
Andere mögliche Wege der Herstellung der 1´, 6´-Dichlorsucrosederivate der Erfindung umfassen die Chlorierung der 4,6-Benzylidenderivate der Sucrose, weitere Herstellungswege für die 1´, 4, 6, 6´-Tetrachlorsucrosederivate schließen die Umsetzung von Sucrose mit Mesitylensulfonylchlorid unter schärferen Bedingungen ein, um die Bildung des tetrasubstituierten Derivats zu begünstigen, wonach die vier Mesitylensulfonylsubstituenten durch Chloratome ersetzt werden.
Die Herstellung der erfindungsgemäßen Chlorderivate der Sucrose wird anhand der folgenden Beispiele erläutert.
Beispiel 1
Sucrose wurde 6 Tage bei -5° C mit Mesitylensulfonylchlorid (3 Mole) in Pyridin behandelt, um als Hauptprodukt 1´, 6, 6´-Tri-O-mesitylensulfonylsucrose zu erhalten, wie L. Hough, S. P. Phadnis und E-Tarelli, Carbohydrate Research, 1975, 44, C 12 und C 13 beschrieben (auf diese Literaturstelle wird verwiesen). Die Konzentrierung des Reaktionsgemisches und übliche Extraktion des Rückstandes mit Chloroform ergab 6-O-Mesitylensulfonyl-kleines Alpha-D-glucopyranosyl-1,6-di-O-mesitylen-sulfonyl-kleines Beta-D-fructofuranosid (Schmelzp. 135° C unter Zersetzung). Weitere Verbindung wurde aus den Mutterlaugen nach Chromatographie auf Kieselgel erhalten. Diese Verbindung wurde dann
in 6-Chlor-6-desoxy-kleines Alpha-D-glucopyranosyl-1,6-dichlor-1,6-didesoxy-kleines Beta-D-fructofuranosid durch Behandlung des Pentaacetats mit Lithiumchlorid in N, N-Dimethylformamid, das eine Spur Jod enthält, 18 Stunden bei 140° C umgewandelt (Schmelzp. 127° C, aus Äthanol, [kleines Alpha][tief]D + 58°; C l Chloroform).
Beispiel 2
Die Mutterlaugen aus dem Konzentrierungs- und Extraktionsschritt des Beispiels 1 wurden der Chromatographie auf Kieselgel unterworfen, um ein Gemisch von isomerem Disulfonat zu erhalten. Nach Zugabe von 2-Propanol oder Äthanol kristallisierte 1´, 6´-Dimesitylensulfonat aus, d. h.
Das Dimesitylensulfonat wurde durch Behandlung mit Acetanhydrid acetyliert und dann bei 140° C 18 Stunden mit Lithiumchlorid in N,N-Dimethylformamid, das eine Spur Jod enthält, umgesetzt, um das 1´, 6´-Dichlorid in hoher Ausbeute zu erhalten. Dieses Dichlorid wurde durch [hoch]1H-NMR-, Massenspektroskopie und durch Überführung in die entsprechenden 1´, 6´-Didesoxyverbindung charakterisiert.
Beispiel 3
Die 1´, 6, 6´-Tri-O-mesitylensulfonylsucrose, hergestellt nach Beispiel 1, wurde acetyliert durch Behandlung mit Acetanhy-
drid und Umsetzen 18 Stunden bei 140° C mit Lithiumchlorid in N,N-Dimethylformamid, das eine Spur Jod enthält, um das 1´, 6, 6´-Trichlorid zu erhalten, d. h.
Dieses Trichlorid wurde dann des-O-acetyliert durch Behandeln mit methanolischem Natriummethoxid; das Produkt (1 Mol) wurde in Pyridin gelöst, auf -40° C gekühlt und tropfenweise mit einer Lösung von Sulfurylchlorid (10 Mole) in Chloroform versetzt. Das Reaktionsgemisch wurde auf 0° C erwärmen gelassen, 24 Stunden bei dieser Temperatur gehalten, wonach es in eiskalte 10%ige Schwefelsäure gegossen wurde. Das Gemisch wurde mit Chloroform extrahiert, der Extrakt wurde mit wässrigem Natriumbicarbonat gewaschen, dann mit Wasser gewaschen und mit MgSO[tief]4 getrocknet. Verdampfung des Chloroforms und folgende Dechlorsulfatierung in Methanol unter Verwendung von Natriumjodid und Natriumcarbonat, wie von L. Hough, S.P.Phadnis und E.Tarelli in Carbohydrate Research 1975, 44, 37-44, beschrieben wird, ergab das rohe 1´, 4, 6, 6´-Tetrachlorgalactosucrose-Produkt. Dieses wurde durch Umwandlung in das Tetra-O-acetat und nachfolgende Säulenchromatographie sowie Des-O-acetylierung gereinigt.
Die gewünschte Tetrachlorverbindung wurde anhand der [hoch]13C-NMR-Spektroskopie und Massenspektroskopie und durch Überführung in das Tetramethansulfonat [Schmelzp. 120-121° C;[kleines Alpha][tief]D + 65,5° (c l Chloroform)] charakterisiert.
Claims (8)
1. Chlorderivate der Sucrose, dadurch gekennzeichnet, daß es das tetrachlorsubstituierte Derivat, in welchem Chlorsubstituenten in 1´-, 4-, 6- und 6´-Stellung stehen, das dichlorsubstituierte Derivat, in welchem Chlorsubstituenten in 1´- und 6´-Stellung stehen, oder das trichlorsubstituierte Derivat, in welchem Chlorsubstituenten in 1´-, 6- und 6´-Stellung stehen, ist.
2. 4,6-Dichlor-4,6-didesoxy-kleines Alpha-D-galactopyranosyl-1,6-dichlor-1,6-didesoxy-kleines Beta-D-fructofuranosid.
3. kleines Alpha-D-Glucopyranosyl-1,6-dichlor-1,6-didesoxy-kleines Beta-D-fructofuranosid.
4. 6-Chlor-6-desoxy-kleines Alpha-D-glucopyranosyl-1,6-dichlor-1,6-didesoxy-kleines Beta-D-fructofuranosid.
5. Ester eines der Chlorderivate der Sucrose nach einem der Ansprüche 1 bis 4.
6. Verfahren zur Herstellung der Verbindung nach Anspruch 3, dadurch gekennzeichnet, daß man Sucrose mit Mesitylensulfonylchlorid umsetzt, die gewünschten Dimesitylensulfonate von dem Gemisch der substituierten Sucrosen abtrennt und die Mesitylensulfonyloxy-Substituenten durch Chloratome mittels Umsetzung mit Lithiumchlorid ersetzt, nachdem die restlichen freien Hydroxylgruppen durch Veresterung geschützt worden sind.
7. Verfahren zur Herstellung der Verbindung nach Anspruch 4, dadurch gekennzeichnet, daß man Sucrose mit Mesitylensulfonylchlorid umsetzt, das 1´, 6, 6´-trisubstituierte Derivat aus dem Gemisch der substituierten Sucrosen abtrennt und die Mesitylensulfonyloxy-Substituenten durch Chloratome mittels Umsetzung mit Lithiumchlorid ersetzt,
nachdem die restlichen freien Hydroxylgruppen durch Veresterung geschützt worden sind.
8. Verfahren zur Herstellung der Verbindung nach Anspruch 2, dadurch gekennzeichnet, daß man die Verbindung nach Anspruch 4 entestert (deesterifiziert) und dann bei niedrigen Temperaturen mit Sulfurylchlorid umsetzt.
