Description
Ein Großteil der gegenwärtig angebotenen Immersionsöle mit
einem Brechungsindex von größer als 1,5 enthält als
Hauptbestandteile Chlorparaffine Chlorparaffine sind jedoch
als gefährliche Arbeitsstoffe eingestuft und weisen außerdem
eine schlechte Umweltverträglichkeit auf.
Neben den Immersionsölen auf Basis von Chlorparaffinen sind
auch bereits halogenfreie Immersionsöle erhältlich. Das
Hauptproblem bei der Herstellung halogenfreier Immersionsöle
ist das ungünstige Verhältnis von Brechzahl und Dispersion bei
den meisten halogenfreien Flüssigkeiten mit Brechungsindizes,
die einen Wert von größer als 1,5 aufweisen. Die die Dispersion
beschreibende Abbe-Zahl weicht bei solchen Immersionsölen
häufig deutlich von dem durch die DIN 58884 bzw. ISO 8036/1
spezifizierten Bereich für die Abbe-Zahl von 44 ± 5 (DIN) bzw.
44 ± 3 (ISO) ab. Bekannte halogenfreie Immersionsöle, deren
Abbe-Zahl größer als 41 ist, sind entweder hinsichtlich ihrer
UV-Durchlässigkeit unbefriedigend oder weisen eine zu große
Eigenfluoreszenz auf. Halogenfreie Immersionsöle mit Werten für
die Abbe-Zahl im oberen Toleranzbereich der o.g. beiden Normen,
also mit einer Abbe-Zahl νe < 45, sind nicht bekannt.
In der EP 0 209 621 sind neben halogenhaltigen auch
Ausführungsbeispiele für halogenfreie Immersionsöle angegeben.
Zwei Ausführungsbeispiele für halogenfreie Immersionsöle
enthalten Tricyclodecanol als Nebenkomponente. Bezüglich der
UV-Durchlässigkeit (bei Wellenlängen unter 400 nm) sind in der
Schrift jedoch keine Angaben enthalten. Außerdem ist die
Viskosität mit Werten von 1000 und 2000 mm²/s bei 25°C für die
meisten Anwendungen zu groß, d. h. bei der Anwendung kann es
sehr leicht zum Einschluß von Luftblasen kommen.
Es ist das Ziel der Erfindung eine Basissubstanz für die
Herstellung von Immersionsölen für die Mikroskopie anzugeben,
mit der sich Immersionsöle mit Abbe-Zahlen zwischen 40 und 50
bei gleichzeitig geringer Eigenfluoreszenz und hoher
UVA-Durchlässigkeit herstellen lassen.
Dieses Ziel wird durch ein Immersionsöl mit den Merkmalen des
Anspruches 1 gelöst. Vorteilhafte Ausgestaltungen der Erfindung
ergeben sich aus den Merkmalen der abhängigen Ansprüche.
Das erfindungsgemäße Immersionsöl enthält als Hauptbestandteile
Tricyclodecan-Derivate oder Derivate von Stoffen mit
Grundstrukturen des Tricyclodecans. Der Anteil der
Tricyclodecan-Derivate und/oder der Derivate von Stoffen mit
Grundstrukturen des Tricyclodecans beträgt dabei mindestens 50
Gewichtsprozent des gesamten Immersionsöls bei
Ausführungsbeispielen, bei denen das Immersionsöl ein
Zweistoffgemisch ist, und mindestens 40 Gewichtsprozent bei den
Ausführungsbeispielen, bei denen das Immersionsöl ein Drei-
oder Mehrstoffgemisch ist. Das Immersionsöl kann auch mehrere
Tricyclodecan-Derivate enthalten, wobei in solchen Fällen die
Summe aller Tricyclodekan-Derivate mindestens 40 Gewichsprozent
des Immersionsöls beträgt.
Die Erfindung geht von der Erkenntnis aus, daß Tricyclodecan-
Derivate oder Derivate von Stoffen mit der Grundstruktur des
Tricyclodecans aufgrund der Ringstruktur des Tricyclodecans
(TCD) einen relativ großen Brechungsindex bei gleichzeitig
großer Abbe-Zahl aufweisen und dadurch als Hauptkomponente für
Immersionsöle hervorragend geeignet sind. Ein TCD-Derivate oder
Derivate von Stoffen mit TCD-Grundstruktur als
Hauptbestandteile enthaltendes Immersionsöl kann deshalb
halogenfrei sein.
Bei den Derivaten sowohl des Tricyclodecans als auch der Stoffe
mit Tricyclodecan-Grundstruktur handelt es sich vorzugsweise um
Ester- oder Etherverbindungen. Dabei kommen sowohl Ester und
Ether des Tricyclodecans als auch von Tricyclodecan-Oligomeren
und Tricyclodecan-Polymeren in Betracht. Neben den Estern und
Ethern von Monomeren, Oligomeren und Polymeren des
Tricyclodecans können auch ein oder mehrere Wasserstoffe des
Tricyclodecan-Gerüstes durch anderweitige organische Reste
substituiert sein.
Bei den bevorzugten Ausführungsbeispielen bestehen die
Hauptbestandteile des Immersionsöls aus Tricyclodecan-
Methylolester oder Tricyclodecan-Methylolether, da diese
Flüssigkeiten aufgrund der Molekülstruktur vakuumdestillierbar
und damit mit hoher Reinheit darstellbar sind. Aus
Tricyclodecan-Methylolestern und Tricyclodecan-Methylolethern
lassen sich damit Immersionsöle mit minimaler Eigenfluoreszenz
herstellen. Die Herstellung der Immersionsöle erfolgt dabei
einfach durch Mischen des TCD-Methylolesters oder des
TCD-Methylolethers mit geeigneten hochsiedenden Flüssigkeiten,
beispielsweise Weichmachern, Paraffinölen, Polypropylenglycolen
usw., auf die normgerechten Werte für den Brechungsindex.
Für die Herstellung der TCD-Ester und TCD-Ether kann auf die
großtechnisch verfügbaren TCD-Alkohole TCD-Alkohol M (= 8(9)-
Hydroxymethyltricyclo [5.2.1.0 2,6] decan) und TCD-Alkohol DM
(= 3(4), 8 (9)-Dihydroxymethyltricyclo [5.2.1.0 2,6] decan)
zurückgegriffen werden, die von der Fa. Hoechst AG, Frankfurt
(Main), Germany, angeboten werden. Durch Veresterung dieser
Alkohole mit Dicarbonsäuren, wie Phthalsäure, Isophthalsäure,
Terephthalsäure, Malonsäure, Bernsteinsäure, Maleinsäure,
Glutarsäure, Adipinsäure oder Sebacinsäure nach den üblichen
Veresterungsverfahren lassen sich die Tricyclodecanmethylol-
Ester oder Di-(Tricyclodecanmethylol)-Ester synthetisieren.
Als Hauptbestandteil für Immersionsöle besonders geeignet hat
sich das Di-(TCD-Methylol)adipat erwiesen. Dabei beträgt der
Gewichtsanteil des Di-(TCD-Methylol)adipats mindestens 60%.
Als weitere Komponenten zur Einstellung des Brechungsindexes
können beispielsweise Butylbenzylphthalat und/oder Di-
(propylenglykol-1,2)dibenzoat hinzugegeben werden. Das Di-(TCD-
Methylol)adipat ist ebenso wie die anderen Di-(TCD-
Methylol)ester im Gegensatz zum Tricyclodecanol geruchlos und
weist keine nennenswerte Reizwirkung auf die menschliche Haut
auf.
Es sind viele Substanzen mit einer Grundstruktur des
Tricyclodecans möglich, deren Ester- oder Etherderivate für das
erfindungsgemäße Immersionsöl in Betracht kommen. Die
Strukturformel solcher Substanzen sind in den Fig. 1a bis 1c
dargestellt. Dabei kann es sich um ein TCD-Monomer (Fig. 1a
und 1b) oder um ein TCD-Oligomer oder -polymer (siehe Fig. 1c)
handeln, bei denen die von den eckigen Klammern der Fig. 1c
eingeschlossene Ringstruktur n-fach wiederholt auftritt. Ein
oder mehrere der Wasserstoffatome des TCD-Gerüstes können
außerdem, wie in den Fig. 1a und 1b angedeutet, jeweils
durch einen Rest R substituiert sein. Beispiele für geeignete
Reste R sind in der Tabelle III angegeben. Die Substitution der
Wasserstoffe durch Reste R hat nur einen untergeordneten
Einfluß auf die für Immersionsöle wichtigen physikalischen
Eigenschaften.
Ausgangspunkt für die Estersynthese können sowohl TCD-Alkohole,
bei denen X einen Alkoholrest, bevorzugt OH oder CH₂OH,
bezeichnet, oder TCD-Carbonsäuren sein, bei denen X einen
Carbonsäurerest, bevorzugt (COOH)₂, darstellt. Beispiele für
literaturbekannte TCD-Alkohole sind in der Tabelle IV
angegeben. Dabei können sowohl TCD-Mono-Alkohole, TCD-Di-
Alkohole als auch TCD-Trialkohole zum Einsatz kommen. Der
TCD-Alkohol DM (Dialkohol) und der TCD-Alkohol M (Monoalkohol) sind
allerdings aufgrund ihrer großtechnischen Verfügbarkeit
bevorzugt. Durch Umsetzung der TCD-Alkohole mit einer
Carbonsäure lassen sich nach den üblichen Syntheseverfahren die
entsprechenden TCD-Ester herstellen. Geeignete Mono-
Carbonsäuren, Dicarbonsäuren oder Tricarbonsäuren sind in der
Tabelle V angegeben. Die nachfolgende Reinigung der TCD-Ester
erfolgt durch Vakuum- oder Hochvakuum-Destillation. Von
besonderer Bedeutung ist dabei die Temperaturstabilität und der
hohe Siedepunkt der Di-(Tricyclodecanmethylol)ester (Siedepunkt
über 200°C bei einem Druck von 10-5 mbar), woraus eine gute
Vakuumdestilierbarkeit resultiert.
Alternativ zur Estersynthese aus TCD-Alkoholen kann auch von
TCD-Carbonsäuren ausgegangen werden. Geeignete
literaturbekannte TCD-Monocarbonsäuren und TCD-Dicarbonsäuren
sind in der Tabelle VI angegeben. Die Veresterung erfolgt in
diesem Fall durch Umsetzung mit einem monofunktionellem oder
einem höheren Alkohol. Geeignete Alkohole sind in der Tabelle
VII angegeben.
Analog zur Synthese der TCD-Ester sind durch Umsetzung der in
der Tabelle IV angegebenen TCD-Monoalkohole nach den gängigen
Ethersyntheseverfahren die entsprechenden Ether herstellbar.
In der Fig. 2 ist die Struktur von Di-(TCD-Methylol)estern
allgemein und in der Fig. 3 die Struktur von Di-(TCD-
Methylol)adipat dargestellt. Die TCD-Gruppen können dabei
selbst die in den Fig. 1a bis 1c dargestellten Strukturen
mit den in der Tabelle III angegebenen Resten R aufweisen.
In der Tabelle Ia sind die für Immersionsöle wichtigen
physikalischen Eigenschaften einiger bevorzugter Di-(TCD-
Methylol)ester angegeben. Wesentlich für die hervorragende
Eignung von aliphatischen Di-(TCD-Methylol)estern als
Hauptbestandteil von Immersionsölen ist der Brechungsindex
ne < 1,5 und die gleichzeitig hohe Abbe-Zahl νe < = 46 (νe = 47
für das Di-(TCD-Methylol)maleinat) und sogar Abbe-Zahlen
νe < 50 für die anderen angegebenen Di-(TCD-Methylol)ester.
Weiterhin von Bedeutung ist die gute UV-Durchlässigkeit der
Di-(TCD-Methylol)ester, die erst bei Wellenlängen unter 320 nm
für eine 10 mm Schichtdicke bei unter 10% Transmission liegt.
Die Tabelle Ib zeigt einen Vergleich der für Immersionöle
wichtigen physikalischen Eigenschaften eines Immersionsöls nach
der Erfindung und bekannten halogenhaltigen und halogenfreien
Immersionsölen. Die Zusammensetzung des erfindungsgemäßen
Immersionsöls ist unter Nr. 11 in der Tabelle II angegeben.
In der Tabelle II sind die Zusammensetzungen und die
physikalischen Eigenschaften von 14 bevorzugten Gemischen aus
einem Di-(TCD-Methylol)ester und einer oder mehreren
hochsiedenden Flüssigkeit(en) angegeben. Der Gewichtsanteil der
Di-(TCD-Methylol)ester beträgt dabei bei den zweikomponentigen
Gemischen zwischen 68 und 98,5 Gewichtsprozent und bei den
dreikomponentigen Gemischen zwischen 51 und 70 Gewichtsprozent.
Wie die Zahlenwerte für die Abbe-Zahl und für die Viskosität
zeigen, lassen sich nahezu beliebige Viskositätswerte zwischen
100 mm²/s und 6000 mm²/s für Abbe-Zahlen im Bereich 39 bis 50
einstellen. Für die häufigsten Anwendungen sind Gemische mit
Viskositäten im Bereich 400 mm²/s bis 600 mm²/s und damit die
Ausführungsbeispiele 6, 11 und 12 besonders bevorzugt.
Prinzipiell kann das Immersionsöl aus mehreren Di-(TCD-
Methylol)-Estern als Hauptbestandteil und mehreren
hochsiedenden Flüssigkeiten zum Einstellen des Brechungsindex
als Nebenbestandteil zusammengesetzt sein. Da jedoch die
Ausgangssubstanzen für eine geringe Restfluoreszenz in hoher
Reinheit bereitgestellt werden müssen und die Verbrauchsmengen
relativ klein sind, sind unter dem Gesichtspunkt geringer
Herstellkosten die Zweistoffgemische besonders bevorzugt.

2. Immersionsöl nach Anspruch 1, wobei die Stoffe mit
Grundstrukturen des Tricyclodecans Tricyclodecanpolymere
oder Tricyclodecanoligomere sind.
3. Immersionsöl nach Anspruch 1, wobei die Tricyclodecan-
Derivate Ester oder Ether des Tricyclodecans oder die
Derivate Ester oder Ether von Stoffen mit Grundstrukturen
des Tricyclodecans sind.
4. Immersionsöl nach Anspruch 1, wobei die Hauptbestandteile
Tricyclodecan-Methylolester oder Tricyclodecan-
Methylolether sind.
5. Immersionsöl nach Anspruch 4, wobei die Hauptbestandteile
Di-(Tricyclodecanmethylol)ester oder Di-
(Tricyclodecanmethylol)ether sind.
6. Immersionsöl nach Anspruch 5, wobei die Hauptbestandteile
Di-(Tricyclodecanmethylol)adipat, Di-(Tricyclodecan
methylol)phthalat, Di-(Tricyclodecanmethylol)malonat, Di-
(Tricyclodecanmethylol)succinat, Di-(Tricyclodecan
methylol)maleinat, Di-(Tricyclodecanmethylol)glutarat
und/oder Di-(Tricyclodecanmethylol)sebazat sind.
7. Immersionsöl nach einem der Ansprüche 1-6, das als
Nebenbestandteile eine oder mehrere hochsiedende
Flüssigkeit(en) enthält.
8. Immersionsöl nach Anspruch 7, wobei die hochsiedenden
Flüssigkeiten Paraffinöl, Weichmacher,
Polypropylenglycole, Butylbenzylphthalat, Dioctylphthalat,
Dioctylsebazat und/oder Di-(propylenglycol-1,2)dibenzoat
sind.
9. Immersionsöl nach einem der Ansprüche 1-8, wobei der
Anteil der Tricyclodecan-Derivate oder Derivate von
Stoffen mit Grundstruktur des Tricyclodecans mehr als 50
Gewichtsprozent, vorzugsweise mehr als 60 Gewichtsprozent
beträgt.
10. Di-(Tricyclodecanmethylol)adipat.
11. Di-(Tricyclodecanmethylol)adipat hergestellt durch
Veresterung von 8(9)-Hydroxymethyltricyclo [5.2.1.0 2,6]
decan und Adipinsäure durch Veresterung und nachfolgende
Vakuumdestillation des gewonnenen Esters.
