Description
KAISERLICHES
PATENTAMT.
Das i-Phenyl-3-methyl-4-amino-5-pyrazolon eignet sich wegen seiner leichten Oxydierbarkeit
— schon durch den Sauerstoff der Luft wird es in Rubazonsäure übergeführt — nicht
zur technischen Verarbeitung.
An Stelle des sauerstoffempfindlichen I-Phe-
nyl-3-methyl-4-amino-5-pyrazolons kann man aber seine Säure- und Alkylderivate zur
Methylierung verwenden, von denen nachstehend einige beschrieben sind:
Das i-Phenyl^-methyl^-urethano-S-pyrazolon
(I) und· das i-Phenyl-3-methyl-4-urethano-5-pyrazolonmethylcarbonat
(II):

wird erhalten, wenn man auf eine alkalische Lösung des
pyrazolons 1 oder 2 Mol. Chlorameisensäuremethylester,
zweckmäßig unter Luftabschluß, einwirken läßt. Das i-Phenyl-3-methyl-4-methylurethano-5-pyrazolon
kristallisiert aus Alkohol in Blättchen oder Nädelchen vom Schmelzpunkt 198 bis 199 °. In verdünnter
Natronlauge löst es sich und wird durch Säuren wieder gefällt. i-Phenyl-3-methyl-4-urethano-5;-pyrazolonmethylcarbonat
ist in verdünnter Natronlauge unlöslich; es kristall»-.--40
siert aus Benzol in feinen Nädelchen, die bei 153 bis 1540 schmelzen.
Ein Alkylderivat des 4-Acetylaminopyrazolons
ist das i-Phenyl-3-methyl-4-aminoessigestermethylurethari-5-pyrazolon
(III):

dieses entsteht, wenn man auf i-Phenyl-3-methyl-4-urethano-5-pyrazolon
oder auf i-Phenyl^-methyl^-urethano-S-pyrazolonme- ■
thylcarbonat Chloressigsäurerhethylester bei Gegenwart von Natriummethylat einwirken
läßt. I-Phenyl-3-methyl-4-aminoessigsäuremethylestermethylurethän-5-pyrazolon
bildet aus Essigäther kristallisiert feine verfilzte Nadeln vom Schmelzpunkt 135 bis 1360. Diese Verbindung
löst sich in Säure, wie in verdünnter Natronlauge.' Durch Kochen mit verdünnter
Salzsäure läßt sich dieser Ester zur i-Phenyl-3-methyl-5-pyrazolon-4-aminoessigsäure
(IV):

ίο verseifen. Letztere kristallisiert aus heißem
Wasser in schönen Blättchen, die bei i8o° erweichen und bei 193 bis 1940 unter Aufschäumen
schmelzen.

erhält man durch Reduktion aus dem i-Phenyl-3-methyl-4-benzolazO"5
-äthoxypyrazol.
Letzteres entsteht aus 4-Benzolazo-5-chlorpyrazol durch Umsetzung mit Natriumäthylat. Das
ι - Phenyl - 3 - methyl- 4 -amino-5 -äthoxypyrazol bildet ein bis jetzt nicht kristallisierendes Öl,
das sich beim Stehen-an der Luft rot färbt.
Sein Chlorhydrat kristallisiert aus Alkohol in derben Kristallen vom Schmelzpunkt 183 bis
184 °. Die Benzoylverbindung bildet, aus Alkohol kristallisiert, Nädelchen, die bei 163
bis 1640 schmelzen. Die Acetylverbindung
kristallisiert aus Essigäther in Nadeln und schmilzt bei 99 °.
Ein 4-Alkylderivat des Aminoäthoxypyrazols
ist.das i-Phenyl^-methyl^-cyanmethylamino-5-äthoxypyrazol

man erhält letzteres aus dem 4-Amino-5-äthoxypyrazol
durch Einwirkung von Formaldehyd und Blausäure. Es bildet farblose Kristalle vom Schmelzpunkt 109 bis iio°;
es ist nicht löslich in Wasser, schwer löslich in Ligroin, leicht löslich in Alkohol, Äther
und Benzol.
Ein weiteres hierher gehöriges Derivat des i-Phenyl^-methyl^-amino-S-pyrazolons erhält
man aus dem l-Phenyl-3-methyl-4-nitro-5-pyrazolon,
wenn man auf dessen Alkalisalz Chloressigester einwirken läßt. Der so entstandene
I-Phenyl -3 -methyl-4-nitropyrazol-5-oxyessigester geht durch Reduktion in das
innere Anhydrid der 1-Phenyl-3 -methyl-4-aminopyrazol-5-oxyessigsäure

über, analog der Bildung des Ketophenmorpholins aus o-Nitröphenoxyessigester.
Das Anhydrid der i-Pheny'l-3-methyl-4-aminopyrazol-5-Oxyessigsäure
bildet, aus Alkohol kristallisiert, derbe Kristalle vom Schmelzpunkt 2300. In Wasser ist diese Substanz
sehr schwer löslich, leicht in starker Salzsäure und in verdünnter Natronlauge. Durch
Methylieren mit Dimethylsulfat in alkalischer Lösung erhält man daraus das Monomethylderivat
(VIII):

Nädelchen vom Schmelzpunkt 167 bis i68°,
die sich beim Erwärmen mit Alkali lösen. In Wasser ist das methylierte Anhydrid sehr
schwer löslich, leicht in heißem Alkohol. 95 s.>
Es sind dies Ausgängsprodukte, die sich zur Ausführung des Verfahrens als besonders
geeignet erwiesen haben. Durch den Einfluß der methylierenden Mittel werden die Alkyl-
und Säurereste, die am Stickstoff oder Pyrazolonsauerstoff gebunden sind, leicht abgespalten
und man gelangt so zu dem wertvollen i-Phenyl-2 · 3-dimethyl-4-dimethylami~ no-5-pyrazolon. . ■ '
Gleiche Teile i-Phenyl-3-methyl-4-urethano-5-pyrazolon
(I), Jodmethyl und Methylalkohol werden 10 bis 12 Stunden auf 125 bis 135 °
erhitzt. Nach dem AbdestiUieren des Methyl- no alkohole wird der Rückstand in Wasser gelöst
und der mit überschüssiger Natronlauge versetzten Lösung das i-Phenyl-2 · 3-dimethyl-4-dimethylamine-5-pyrazolon
durch Ausschütteln mit Benzol entzogen. An Stelle des Urethanopyrazolons kann man auch das
i-Phenyl^-methyl^-urethano-S-pyrazolonmethylcarbonat
(II), das i-Phenyl-3-me'thyl-4-aminoessigesterurethano-5-pyrazolon
(III), die l-Phcnyl-3-methyl-5-pyrazolon-4-aminoessig-
säure (IV) und das ι - Phenyl -3 -methyl-4-methylaminopyrazol
- 5 - oxyessigsäureanhydrid (VIII) verwenden.
- Beispiel II.
5 Teile ι -Phenyl-3-methyl-4-amino-5 "
äthoxypyrazol (V) werden mit 7 Teilen Dimethylsulfat allmählich vermischt. Nachdem
■ die Selbsterwärmung aufgehört hat, wird das Gemenge noch einige Stunden bei Wasserbadtemperatur
erwärmt und das Reaktionsprodukt in Wasser gelöst. Man macht hierauf
mit Natronlauge alkalisch, erwärmt kurze Zeit gelinde und schüttelt das i-Phenyl-2-3-dimethyl-4-dimethylamine-5-pyrazolon
mit Benzol aus. Etwa vorhandenes i-Phenyl-2 · 3-dirriethyl-4-amino-5-pyrazolon kann mittels
Benzaldehyd entfernt werden.
Das so erhaltene i-Phenyl-2· 3-dimethyl-4-dimethylamino-5-pyrazolon
wird aus Wasser und Alkohol umkristallisiert und bildet dann farblose Kristalle vom Schmelzpunkt io8°.

- Patent- Anspruch :Verfahren zur Darstellung von i-Phenyl-2 ·^-dimethyl^-dimethylamino-S-pyra-. zolon, dadurch gekennzeichnet, daß man die Alkyl- und Säurederivate des 1-Phenyl - 3 - methyl - 4 - amino - 5 - pyrazolons mit Methylierungsmitteln behandelt.
