Die vorliegende Erfindung bezieht sich auf die Herstellung von 5-Hydroxy-5-aminoalkyl-di-benzo-[a,d]-cycloheptanen
der allgemeinen Formel I

bei der A eine 1,2- oder 1,3-Alkylengruppe mit nicht mehr als 5 Kohlenstoffatomen und R1 und R2 entweder
jeweils ein Wasserstoffatom, eine niedere Alkyl-, niedere Cycloalkyl- oder Benzylgruppe oder
zusammen mit dem Stickstoffatom einen heterocyclischen Ring bedeuten, wobei R1 und R'2 in
beiden Fällen zusammen nicht mehr als 6 Kohlenstoffatome enthalten.
Verbindungen der Formel I, deren Aminogruppe sekundär oder tertiär ist, sind bereits bekannt. Verbindungen,
deren Aminogruppe nicht substituiert ist, wurden bisher in der Literatur nicht beschrieben.
Nach dem bisher angewendeten Verfahren werden Verbindungen mit einer tertiären Aminogruppe aus
Di-benzo-[a,d]-cycIoheptan-5-on (Dibenzosuberon) dadurch erhalten, daß man das letztere mit einem
Grignard-Reagenz behandelt, das die einzuführende Gruppe enthält. Für die Herstellung von Verbindungen
mit einei sekundären Aminogruppe setzt man das Dibenzosuberon mit einem Grignard-Reagenz
um, das eine tertiäre Aminoalkylgruppe enthält, deren einer Substituent an dem Stickstoffatom
dann in einer nachfolgenden Umsetzung durch Hydrogenolyse abgespalten wird.
Die Herstellung eines tertiären Aminoalkylreagens nach Grignard ist immer mit Schwierigkeiten
verbunden, da ein außerordentlich aktives Magnesiummetall erforderlich ist und die Umsetzung mit
Methyl- oder Äthylbromid eingeleitet werden muß. Im allgemeinen ist die Umsetzungszeit sehr lang,
und es gibt Beispiele für Umsetzungen der vorstehend beschriebenen Art, die etwa 20 Stunden
erfordern, um zum Abschluß zu kommen. Grignard-Reagenzien sind ferner so reaktionsfähig, daß sie
bereits in der Atmosphäre zu brennen beginnen können, so daß scharfe Sicherheitsvorkehrungen in
Anlagen vorgesehen werden müssen, in denen eine Verfahren zur Herstellung von 5-Hydroxy-5-aminoalkyl-di-benzo-[a,d]-cycloheptanen

derartige Umsetzung nach Grignard durchgeführt werden soll.
Es wurde nun gefunden, daß die vorstehend beschriebene Ketoverbindung (die die nachstehende
Formel II hat) leicht 2 Alkalimetallatome a η Ligert und so ein verhältnismäßig sicheres Reagens bildet,

umgesetzt werden kann, bei der R1, R2 und A die vorstehende Bedeutung haben und X ein Halogenatom
ist. Diese Umsetzung geht mit zufriedenstellender Geschwindigkeit vonstatten und ergibt bei
der Hydrolyse in guter Ausbeute ein Produkt von ausgezeichneter Reinheit. Die Gefahren, die z. B.
mit der Verwendung von metallischem Natrium verbunden sind, werden durch die große Geschwindigkeit,
mit der die Anlagerung stattfindet, wesentlich verringert.
In diesem Zusammenhang soll darauf hingewiesen werden, daß es unter Anwendung der Grignard-Methode
überhaupt nicht möglich war. Verbindungen der vorstehenden Formel I herzustellen,
deren Aminogruppe nicht substituiert ist.
Die vorliegende Erfindung betrifft demnach ein Verfahren zur Herstellung von 5-Hydroxy-5-amino-

alkyl-di-benzo-[a,d]-cycloheptanen der Formel I, sie ist dadurch gekennzeichnet, daß man das Di-benzo-[a,d]-cycloheptan-5-on
der Formel II

mit einem Alkalimetall M in geeigneten Lösungsmitteln umsetzt und dieses Reaktionsgemisch oder
die daraus erhaltene Dialkalimetallverbindung mit einer Verbindung der Formel III

umsetzt, bei der A, X, R1 und R- die vorstehende Bedeutung haben, wobei eine Verbindung der
Formel IV

gebildet wird, bei der A, R1 und R2 die vorstehende Bedeutung haben und M ein Alkalimetallatom ist
und die erhaltene Verbindung zur Verbindung I hydrolysiert.
Man kann die bei der Hydrolyse erhaltene Verbindung der Formel I in ein therapeutisch wertvolles
Säureadditionssalz nach bekannten Verfahren umwandeln.

Die Umsetzung mit dem Alkalimetall kann gesondert von der Umsetzung mit der Verbindung der
Formel III durchgeführt werden. Die Umsetzung kann unter Verwendung des in flüssigem Ammoniak
gelösten Metalls und/oder in Gegenwart eines inerten Reaktionsmediums durchgeführt werden, in dem
das Metall dispergierbar ist, besonders in Benzol, Toluol, Xylol, Diäthyläther oder Fraktionen von
leichten Kohlenwasserstoffen, z. B. Extraktionsbenzine oder Petroläther.
Verbindungen der Formel I, bei denen R1 und R2 jeweils Wasserstoff bedeuten, wurden bisher in der
Literatur nicht beschrieben. Es hat sich gezeigt, daß die Verbindungen spasmolytische Eigenschaften
haben. Die Verbindungen der Formel I können mit wasserabspaltenden Mitteln behandelt werden, um
sie in die entsprechenden Alkylidenverbindungen umzuwandeln, die wertvolle Mittel zur Einwirkung
auf das zentrale Nervensystem sind.
5-Hydroxy-5-(3-N,N-dimethylamino-propyl)-di-benzo- [a,d]-cycloheptan
4,8 g metallisches Natrium werden in 250 ecm flüssigem Ammoniak gelöst. Dann wird zu der
erhaltenen Lösung im Verlauf von 20 Minuten eine Lösung von 20,8 g Di-benzo-[a,d]-cycloheptan-5-on
in 50 ecm getrocknetem Diäthyläther gegeben. Anschließend wird das Reaktionsgemisch tropfenweise
mit einer Lösung von 12,2 g I-N5N-Dimethylamino1 3-chlorpropan in 50 ecm Diäthyläther versetzt. Dann
gibt man zu dem gesamten Gemisch 150 ecm getrockneten Äther und läßt das Gemisch 18 Stunden
stehen. Danach wird es in IOOccm 5 n-Essigsäure gegossen. Die saure Phase wird mit 5 n-Natriumhydroxyd
alkalisch gemacht, um die Verbindung als Kristalle zu fällen, die dann getrocknet werden. Die
Ausbeute an 5-Hydroxy-5-(3-N,N-dimethylaminopropyl)-di-benzo-[a,d]-cycloheptan betrug 23,5 g
(79,8%), und der Schmelzpunkt lag bei 112 bis 115 C. Nach Umkristallisation lag der Schmelzpunkt
bei 116 bis 118°C. Der Schmelzpunkt des Hydrochlorids lag bei 218 bis 218,5°C

Auf gleiche Weise wie im Beispiel 1 erhält man 5-Hydroxy - 5 - (2 - methyl - 3 - Ν,Ν - dimethylaminopropyl)-di-benzo-[a,d]-cycloheptan
aus 4,8 g metallischem Natrium, 250 ecm flüssigem Ammoniak, 20,8 g Di-benzo-[a,d]-cycloheptan-5-on und 13,6 g
1 - Dimethylamino - 2 - methyl - 3 - chlorpropan. Nach Umkristallisation erhält man 22 g (73,5%) der Verbindung,
deren Schmelzpunkt bei 148 bis 149,5°C liegt. Der Schmelzpunkt des Hydrochlorids liegt bei
209,5 bis 211,5 C.
