Abstract

Předkládaný vynález se týká rekombinantních glykoproteinů, které byly připraveny v rostlinách nebo buňkách rostlin, ve kterých je utlumena nebo zcela potlačena endogenní produkce GlcNAc-αl,3-fukosyltransferázy a jejichž peptidová sekvence má méně než 50 % αl,3-vázaných fukózových zbytků ve srovnání se zbytky přítomnými v glykoproteinech exprimovaných v rostlinných systémech s neredukovanou fukosyltransferázovou aktivitou, přičemž rekombinantní glykoproteiny mají neredukovaný GlcNAc-konec molekuly a způsob jejich výroby.

Claims (6)

1. Způsob produkce rekombinantních glykoproteinů vyznačující se tím, že zahrnuje produkci rekombinantního glykoproteinů v rostlinách nebo rostlinných buňkách, ve kterých byla produkce GlcNAc-al,3-fukosyltransferázy potlačena nebo zcela vyloučena a pro které platí, že jejich endogenní aktivita GlcNAc-al,3-fukosyltransferázy je menší než 50 % aktivity GlcNAc-al,3-fukosyltransferázy přítomné v přírodních rostlinách nebo rostlinných buňkách, přičemž jejich GlcNAc-al,3-fukosyltransferáza je kódována molekulou DNA zahrnující sekvenci podle sekvence ID č. 1 s otevřeným čtecím rámcem od páru bází 211 do páru bází 1740 nebo sekvenci, která je alespoň z 50 % homologní s výše uvedenou sekvencí nebo která hybridizuje za stringentních podmínek s výše uvedenou sekvencí nebo která zahrnuje sekvenci, která degeneruje s výše uvedenou DNA sekvencí kvůli genetickému kódu.
2. Způsob podle nároku 1, vyznačující se tím, že rekombinantní glykoproteiny jsou lidské proteiny, zejména proteiny pro medicinální použití.
3. Způsob podle nároku 1 nebo 2, vyznačující se tím, že endogenní aktivita GlcNAc-al,3- fukosyltransferázy je menší než 20 % aktivity GlcNAc-al,3-fukosyltransferázy přítomné v přírodních rostlinách nebo rostlinných buňkách.
4. Způsob podle kteréhokoli z nároků 1 až 3, vyznačující se tím, že endogenní aktivita GlcNAc-al,3fukosyltransferázy je 0 % aktivity GlcNAc-al,3-fukosyltransferázy přítomné v přírodních rostlinách nebo rostlinných buňkách.
5. Způsob podle kteréhokoli z nároků 1 až 4, vyznačující se tím, že snížení aktivity GlcNAc-al,3fukosyltransferázy je díky antisense inhibici, přičemž je použit polynukleotid, který je alespoň částečně komplementární k sekvenci molekuly DNA.
6. Způsob podle kteréhokoli z nároků 1 až 4, vyznačující se tím, že snížení aktivity GlcNAc-al,3fukosyltransferázy je díky knock-out mutaci sekvence molekuly DNA.
