Abstract

发明概述

储库凝胶的制备

布比卡因碱的制备

实施例11
PLGA聚合物的差示扫描量热(DSC)测定
[0113]使用差示扫描量热计(Perkin Elmer Pyris 1,Shelton,CT)测定本发明中使用的不同低分子量PLGA聚合物的玻璃转化温度。在Mettler PJ3000顶装式天平上将DSC样品配衡。将至少20mg聚合物样品放在平盘中,记录样品的重量。将DSC平盘的盖子放到平盘上,使用加压器密封平盘。从-50℃到90℃以10℃的增量进行扫描。

实施例12
PLGA聚合物的体外降解
[0115]37℃在PBS缓冲液中将测定的PLGA聚合物的质量损失速率作为时间的函数对在本发明中使用的低分子量PLGA聚合物的体外降解曲线进行研究。每种共聚物包括一个样本集。使用13mm的不锈钢冲模压制约25个圆片(每个为100±5mg)。使用Carver压制机用10吨的力将样品压制约10分钟。在室温和25mmHg条件下将圆片放入在真空干燥箱中的玻璃瓶中直到为在降解水浴中使用做好准备。对于每个待测聚合物重复进行该过程。制备含有叠氮化钠(0.1N)的磷酸盐缓冲盐水(PBS)溶液(50mM,pH7.4)。将一个样品圆片称量后放入配衡的玻璃瓶中,记录初始重量(Minitial)。用移液管向每个瓶中移入PBS(10mL)。将玻璃瓶密封,在37℃振荡水浴中放置。每周更换两次缓冲液,在更换前记录溶液的pH。在预先指定的时间点,从缓冲液浴中取出样品,用去离子的Milli-Q水洗涤,表面干燥,称重。将样品重量记为湿重(Mwet)。将样品放置10mL的冻干瓶中,在冻干之前在冰箱(-20℃)中放置。在冻干后,对样品再次进行称量,记为干重(Mlyophilized)。质量损失百分数定义为{(Mlyoplilized-Minitial)/Minitial}×100%。
[0116]图15表示在上述制剂中使用的三种PLGA的质量损失曲线。由此,可观察到使用的三种聚合物分别具有明显不同的降解速率。具有酯末端基团或羧基末端基团的低分子量PLGA的降解速率明显要比高分子量的PLGA快。这表明对于短期间储库凝胶而言优选活性剂一从储库制剂中释放就降解的聚合物更为有利。根据本发明的不同方面,可以获得一个或多个显著的优点。更具体地说,使用简单的加工步骤,就能够获得通过常规注射针使用低配出力就可注射到动物一定部位而无需手术的储库凝胶组合物。一旦到达原位,这些组合物可很快恢复其原始粘度,可快速固化以致于基本上避免了突释效应和提供需要的有益试剂释放曲线,另外,一旦有益试剂施用完全,不需要去除该组合物,因为该组合物可完全生物降解。作为另外的优点,本发明可避免使用使某些有益试剂如基于多肽和核酸的药物降解的微粒化或微囊化技术,并且微粒和微囊很难从使用环境取出。由于形成粘性凝胶不需要水、极限温度或其它溶剂,因此有益试剂的悬浮颗粒保持干燥和其原始的构型,这对于其稳定性有利。另外,由于形成块状物,如果需要可从使用环境中将可注射储库凝胶组合物取出。
