Abstract
一般说来,来自一个物种的动物组织只能在两个物种相同时移植到另一物种中;否则,会发生超免疫排斥,在本发明中,供体组织被修饰,例如经基因转移,以表达或与一种或多种称为同源补体限制因子(HCRFS)的物质结合,这些物质在接受体物种中有活性以防止补体的完全活化和因之发生的排斥。本发明部分基于补体活化交替途径,而不是经典途径负责过急相异异种移植物排斥的发现。
Description
本发明涉及用于移植物中的生物相容性材料以及该材料的生产及其应用。
在过去四十年中,替换衰退的或有缺陷的动物(尤其是人)组织(包括器官)已成为临床医学中一种常用的部位疗法。这些替换疗法的例子有使用聚对苯二甲酸乙酯(商标为DACRON,由Dupont公司生产)修复有缺陷的血管、将隐静脉作为自身移植物用于旁路阻塞的动脉以及将心脏从一人移植给另一人。
由于现代免疫抑制剂能够以相对少量的成本带来较高的成功率,器官移植已取得显著发展。对器官移植的需求已迅速增加。现在全世界每年要完成20,000个以上器官移植物。但是,按照现今的依据所作的估计这一数据仅代表了需要量的约15%。以现存来源并不能满足各种类型捐献器官的供/需比。一个最好的例子可能是对心脏移植的需求。由Barnard于1967完成的第一例心脏移植受到新闻界的广泛报道。在一年的时间内,二十二个国家的六十四个不同的外科小组共完成了101个心脏移植物。由于所得结果很差,使人们的希望成为泡影,因此,到二十世纪七十年代早期,全世界每年完成的移植物还不到30个。但是,随着环孢多肽免疫抑制剂的引入使心脏移植产生革命,现在大部分中心进行的心脏移植中其成功率均在80%以上(且患者存活)。随着专门技能的获得,人们有理由期望这一存活率可进一步提高。当然,这一方法的成功增加了人们对它的需求,因此医学行业和公共大众更加意识到心脏移植提供了一种真正的替代死亡的方法,并且越来越多的患者求助于这一方法。现在每年完成2,000个以上的心脏移植物。
现在,在心脏移植中最大的死亡危险在于等待得到合适的捐献器官。虽然人工心脏为这些患者提供了短期支持装置,而长期的需求则是对于更多的心脏移植物中心和更大的捐献物供应。尽管可能从心脏移植获益的潜在个体数从未被科学地建立起来,但是已公布的要求心脏移植的估算数字则在每年每百万人50到250人的广泛范围内变化,它取决于所选样的依据、接受者的年龄以及疾病等等。不管实际数据如何,但有一点已是很明确的,即现在捐献者所提供的可选样之物不能够满足需求。对于肾移植和肝脏移植也存在类似情况,一旦胰或胰岛细胞移植成为普遍接受的治疗糖尿病的方法,则很可能使得该组织的短缺也成为主要问题。
现在移植疗法还存在许多其它缺点。捐献者的器官并不是在所有情况下都适用于移植,这不仅是因为许多器官捐献者本人是某些事故(如道路事故)的受害者,而这些事故是通过损伤正要被移植的器官之外的某些其他器官而引起死亡;对于被移植的器官还可能有某些其他损伤或与其相关的困难。
而且,由于从捐献者获得器官的不可预测性,对移植物外科手术常常不能象常规手术那样提前作出安排(包括预定手术时间)。经常是一旦捐献器官被鉴定外科小组和医院管理人员就不得不开始行动,并且工作时间不固定,从而增加了管理困难和个人困难。
在心脏、肝脏和肺移植物的情况下,如果遇到排斥反应,常常不可能进行再移植,除非在很短的时间内偶而得到另一个合适的捐献物。
除了以上医学上的困难外,现在的移植实践在某些情况下还涉及社会问题,首先,从可能的捐献者身上摘除器官可能遭到宗教上的反对(尤其是在笃信再生的国家),当然从死人身上摘取器官还会遇到其他伦理和社会问题,尤其在某些国家需要取得允许。最后,活肾脏捐献商业交易的出现正引起关注(尤其在某些第三世界国家),社会需要抑制或减少这一交易。
具有以上缺点的常规移植手术包括从一特定种属的动物个体(通常是人)向同一种类的另一个体移植,这种移植称为同种移植。由于常规的同种移植物供应存在如上所述困难,因此人们已把注意力放到了在移植中使用异种移植物的可能性上。异种移植是通用于组织(包括细胞和器官)移植的通用术语,它越过了种属的障碍。
在替换疗法中已经有几个使用异种移植物的成功例子。例如,近年已见报道使用猪组织取代主动脉瓣、用猪皮覆盖严重烧伤的患者以及用脐静脉作为替代静脉移植物。但是,所有这些异种移植物都有一个共同点:它们仅提供了一种机械取代。所用的组织是无生物学功能的,其原因是存在于人体中的免疫过程立即(在几分钟或几小时内)摧毁了大部分种属组织的细胞完整性。这类异种移植物就是所知的相异性异种移植物(discordant xenografts)。
这种极强的破坏作用在系统发育上是相关的。因此,来自黑猩猩(一种与人类密切相关的灵长类动物)的组织能够以和同种移植物相同的方式在人体中存活;这类异种移植物就是已知的一致性异种移植物(Concordont xenograft)。
尽管可能有人想到一致性异种移植物可能为同种移植物所遇到的困难提供解决办法,但实际上情况可能并非如此。黑猩猩比人小得多,因此黑猩猩的器官通常不够大,不能在人体内工作。对于肾脏,可通过移植两个黑猩猩肾脏取代受损的人肾而克服这一问题,但对于肝脏和心脏来说这显然是不可能的。而且黑猩猩在自然状态下繁植缓慢,在捕获状态下也很差,并且对于作为实验动物的黑猩猩的需要(尤其在研究爱滋病的当今时代)意味着需求再一次超过供给。另外,让公众接受使用其它灵长类作为异种移植物供体可能存在某些社会问题。
因此,注意力又重新集中到相异性异种移植物上。人们普遍相信相异性异种移植物如此迅速失败的原因是受体中存在“天然产生的”抗供体种中还未定义的抗原的抗体〔Shons等,Europ.Surq.Res.5 26-36(1973)〕这些抗体之所以称作“天然产生的”是因为在没有来自供体种的任何免疫攻击的个体中就已发现它们的存在。
已有许多资料证明了器官移植物的快速排斥反应——即已知的过急抗体介异的排斥反应。在六十年代初期当(同种移植物)肾移植已成为常规治疗方法时,就已观察到被移植的肾在手术还在进行时往往受到受体的排斥。在移植手术进行过程中,作为规律肾脏将变红并在受体和供体血管缝合在一起后不久获得稳定。这种移植物常常马上产尿。在患者仍在手术台上而移植物已被破坏的排斥反应情况下(过急排斥反应),破坏过程常常在移植后的最初几分钟开始。当发生这一情况时,肾脏变为带蓝色和不调和,然后充血。该器官的相容性也发生改变。通常,移植物出现水肿并不再产尿,然后新移植的肾被立即除去。现已清楚它涉及在受体中预先形成的循环抗体与供体肾中的抗原之间形成体液介导的免疫应答。在同种移植过程中避免其发生的唯一方式就是在移植前确保受体中不存在抗供体细胞的任何抗体。随着检测这种抗体的知识的增加(即已知的定义配血),现已认识到受体抗体发挥作用抵抗供体抗原的这一判断是错误的,并且当涉及同一种属的个体之间的移植物时过急移植物破坏作用仅限于特定种类抗体的存在,这一情况被称作T-热阳性交叉配血(T-WarmPositive cross-match);而且几乎可以肯定这些抗体属于IgG亚类。还有,这些抗体的存在总是由于以前存在的免疫过程的结果,这一前期免疫过程则产生于以前的输血或妊娠,而最常产生于衰退的前期移植物。
大量的看法认为过急异种移植物排斥反应的机制与上面所述的过急同种移植物排斥反应的机制差不多相同。关于异种移植物排斥反应机制的文献是非常广泛的,可追溯到约83年前。在那时,仅有三本刊物提出了异种移植物排斥反应的机制,但不涉及抗体。该意见认为补体激活的交替途径(尽管不一定使用这一术语)卷入了异种移植物的排斥反应。该理论首次出现于1976年Schilling等人的论文中〔Surgery,Gynaecology and Obstetrics 14229-32(1976)〕。该理论在1988年和1989年由Miyagawa等再次提出(相同的数据被两次公布)〔Transplantation 46(6)825-830(1988)及Transplantation proceedings 21(1)520-521(1989)〕。但是,由于这些实验实际上存在同样的错误,因此其结果并不是令人信服的。作者声称所选用的模型为其中不存在跨种属抗体的异种移植物模型。但是,现已知道用于测定跨种属抗体的检测方法是不适当的,因此这些论文中所得出的结论是建立在不适当的数据之上的。
现在最常采用的避免或减少异种移植物排斥反应的实验方法是通过血浆取出法、用眼镜蛇毒液因子处理法、抗体的葡萄球菌蛋白A吸附法等等,干扰受体免疫系统的化学疗法,该方法主要以非特异性为基础,例如使用环孢多肽A和其它免疫抑制剂,这一方法自然来自支持同种移植物的化学疗法。
本发明采用了一种根本不同的方法:本发明不是非特异性地干扰受体的免疫系统,它能够使具有供体移植物作用的材料一起施用,而这种供体移植物则被受体免疫系统的某些成分当作为自身的一部分。在一最佳实施方案中,是将供体组织本身进行修饰,以使受体在某些免疫方面将其当作自身组织。
也已发现过急异种移植物排斥反应并不一定由抗体介导。这一结论来自两个观察结果。首先,在缺乏抗体但存在补体的情况下,观察到过急排斥反应;第二,在存在抗体而缺乏补体的情况下未观察到过急排斥反应。
本发明是基于以下发现:在异种移植物的过急破坏过程中补体活化作用非常显著,而不管这一活化作用是否得到合适抗体分子结合的帮助。补体交替途径的激活可被各种细胞产物诱导。这些产物并不限于外部侵入的细胞如细菌或异种移植物,而是存在于许多细胞之上。因此从理论上讲一个体的许多细胞能够激活补体的交替途径,引起大规模的自动免疫破坏作用。这一情况之所以没有发生是由于在血清中和细胞表面天然存在大量的补体下调蛋白。这些分子(在本文中称作“同源补体限制因子)防止了自身补体被自身细胞产物通过经典途径或交替途径完全激活,从而防止了自身的自动免疫破坏作用。这些分子机能的行使在阵发性夜间血红蛋白尿症中得到很好的证明。在该疾病中,至少一种这些分子的膜上锚(加速衰变因子)缺乏。因此,该蛋白质不能保留在红细胞膜上,而与细胞分离,从而激活了补体的交替途径,发生细胞溶解,引起血红蛋白尿症。
本发明的目的之一是提供了将动物组织移植给受体的方法,其中该组织得自与受体属于不同种属的供体。供体种属相对于受体为相异性种属,该方法包括将组织移植给受体并随该移植组织一起提供一种或多种同源补体限制因子,该因子在受体中具有活性,能够防止补体的完全激活。
本说明书中所用的“组织”一词意味着任何能够被移植的生物材料,包括器官(尤其是心、肺、肝、肾、胰和甲状腺等体内重要器官)、角膜、皮肤、血管及其它结缔组织、细胞(包括血液细胞、造血细胞、胰岛细胞、脑细胞以及得自内分泌和其它器官及体液(如PPF)的细胞),所有这些均可用来从一个物种向另一物种移植。
“相异物种”是这样一个物种,当将从其获得的异种移植物移植给受体时通常产生过急排斥反应,也就是说排斥反应将在几分钟或几小时内产生,而不是在几天内产生(Calne Transplant proc2:550,1970)。这种过急排斥反应是本领域熟练技术人员所熟知的,它会在移植后24小时内、6小时内、甚至1小时内发生。
补体及其活化现在已为人们所熟知,并且在Roitt的文章中已有论述(Essential Immunology(Fifth Edition1984),Blackwell Scientific publications,Oxford)。归功于补体(C′)的活性取决于九个蛋白质成分(C1至C9)依次发挥作用,其中的第一成分由三个分别被称作为C1q、C1r和C1s的主要亚片段组成。补体可通过经典途径和交替途径激活,下面对两种途径作一简要叙述。
在经典途径中,抗体与C1结合,其C1s亚单位获得酯酶活性,引起活化并转移到膜上的位点或免疫复合物(首先是C4,然后是2)上的位点。这一复合物具有“C3转化酶”活性,它在溶液中分裂C3从而产生一小肽片段C3a和剩余分子C3b,它们具有特殊作用。C3a具有过敏毒素活性,并在补体放大级联中不再进一步发挥作用。C3b与膜结合,并能引起抗原-抗体-C3b复合物的免疫吸附,从而促进随后的吞噬作用。
在交替途径中,C3转化酶的活性由C3bB完成,其激活可被外部试剂尤其是象内毒素等微生物多糖触发,其作用与抗体无关。转化酶是通过D因子作用于C3b和B因子的复合物而形成。以上形成一个正反馈回路其中C3降解产物(C3b)帮助形成更多的裂解酶。
在经典途径和交替途径这两种途径中,C3b的水平是通过C3b灭活性(因子I)的作用而被维持。C3b很容易与H因子结合,形成复合物,该复合物被因子I破坏,从而丧失其溶血性质和免疫粘连性质。
在C3期之后经典途径与交替途径是相同的。C5被切割为C5a和C5b片段。C5a具有过敏毒素活性,导致多形核白细胞的趋化性。C5b以复合物的形式与C6和C7结合,在膜上形成一个耐热点,充实最终成分C8和C9以产生膜攻击复合物(MAC)。这一环形结构插入到膜中并从膜上凸出,从而形成一个使电解质和水能够完全渗透的跨膜通道。由于内部胶体高渗透压使钠离子和水净流入导致细胞溶解。
可用于本发明的同源补体限制因子(HCRFS)通常可影响补体激活级联的任何一部分。HCRF可能仅影响组成径典途经的那一部分,或者仅影响组成交替途径的那一部分,而最常见的是影响经典途径和交替途径的共同部分。优选HCRF调节剂影响途径的共同部分。该HCRF可能与天然HCRF相同,或简单地具有适当作用。合成和半合成的HCRFS(包括通过重组DNA技术制备的HCRFS及其变异物)均包括在术语HCRF的范围之内。
如上所述,同源补体限制因子是以减少或防止其溶解活性的方式调节补体级联作用的物质。它们被动物体用来标记自身组织以避兔自动免疫反应。在本发明中,从理论上讲HCRF可能是与膜结合的,也可能游离在血清中,但在实践中优选HCRF与异种移植物组织细胞上的膜结合。按这种方式HCRF容易与移植物组织“相结合”。优选的HCRFS包括推想的细胞因子,例如C3b/C4b受体(CR1)、C3.dg受体(CR2)、加速衰变因子(DAF)、C3b灭活剂和膜辅助因子蛋白(MCP)。推想的血清HCRF包括H因子、加速衰变因子(DAF)和C4结合蛋白(C4bp)所有这些HCRFS都是通过影响C3阶段来下调补体活性。阻断C8的同源限制因子(HRF)也是推想的膜因子。
合适HCRFS的许多基因(但不是所有的)位于RCA(补体活化调节剂)位点,它位于染色体1的q32带〔Rey-Campos等,J.Exp.Med.167 664-669(1988)〕。
尽管对于HCRF的命名和定位存在某些混淆,Rey Campus等(在上述引文中)和在他们的早期研究中〔J.Exp.Med.166 246-252(1987)〕对C4Bp因子、CR1、DAF和H因子进行了鉴定。某些工作者认为膜辅助因子蛋白(MCP)是C4结合蛋白(C4bp)的同义词,且这两个因子要么相关要么相同Rother和Till〔″The ComplementSystem″,Springer-Verlog,Berlin(1988)〕在1.2.3.2节综述了C3转化酶的调节因子;他们将C4结合蛋白(C4bp)等同于加速衰变因子,将H因子等同于B1H-蛋白和C3b灭活剂加速剂。HCRFs的命名、定位和鉴定无疑将继续深入,但应当理解,本发明包括应用所有合适的HCRFs。

C8bp/HRF/MIP蛋白是通过糖脂锚状物吸附到细胞表面,如同CD59和DAF一样:已知这些蛋白质在阵发性夜间血红蛋白尿中是缺乏功能的。

SP-40.40也涉及MAC调节(Kivszbaum等EMBO8,711(1989))。
HCRF最好在C3阶段于扰补体激活。MCP和DAF均在C3激活的交替途径中阻断正反馈回路它们构成优选的HCRFs。
HCRF是与被移植的组织一起提供的。这意味着HCRF是以这样一种方式施用,即移植组织被标记为自身,而其它外源物质如侵入的细菌则没有这样明显地被标记。通过非肠道,而非局部地将一种或多种合适的HCRFs简单地施用于移植组织是可能的。但是,从理论上讲它是不可取的,因为这将为HCRF适当定位在移植组织上带来困难,进一步的困难还在于移植发生后不得不给受体反复施用HCRF,但这可通过使用专家药品输送系统来克服。
将HCRF以与供体组织的细胞膜整合的方式提供一般是非常便利的。尽管可能存在被移植组织的良性感染(它可引起适当表达),但达到这一目的的最佳途径是供体组织为转基因的,在移植到受体后它含有并表达编码一个或多个在受体物种中有活性的HCRF的核酸。这种转基因组织可能无限制地表达HCRF。HCRF可通过遗传方法得自受体物种,也可以得自一个密切相关的物种(尽管不是优选的),从它有可能获得一致性异种移植物。
尽管从理论上讲转基因供体组织可来自细胞培养,但优选的供体组织来自转基因动物。这种转基因动物最好应该至少在被移植的组织中表达(或有能力表达)HCRF。但是,甚至连这也不是必需的,因为通过某些结合剂(如杂交的单克隆抗体)(Milstein &Cuello Nature 305 537(1983))或受体也可能将HCRF结合到供体组织的细胞膜上。
受体物种主要是人,但不排除其它的。其它的灵长目及经济和论理所允许的任何其它物种都可能是合适的受体。
供体物种可以是不同于受体物种的任何合适的物种,它能够针对受体物种的生理提供合适的移植组织。对于人受体来说猪供体被认为是合适的,但任何其它物种也可能是合适的。
本发明涉及的第二个方面是提供可供移植的供体物种的动物细胞或组织,而这些细胞或组织是与一个或多个在预期受体物种中具有活性以防止补体完全激活的同源补体限制因子相结合的,相对于受体物种来说该供体物种为相异物种。
本发明涉及的第三个方面是提供具有可移植组织的转基因动物,它不会对移植进或暴露给至少一个相异物种的免疫系统的异种移植物产生排斥反应。相异物种是通常过急排斥异种移植物于动物的物种。
因此本发明包括在制各可移植进受体物种的组织的过程中应用得自供体物种的动物组织以及一个或多个在受体物种中具有活性的同源补体限制因子,其中供体物种相对于受体物种来说为相异物种。
本发明涉及的第四个方面是提供含有能表达另一物种的同源补体限制因子的细胞的转基因动物。该同源补体限制因子通常在相异于转基因动物的物种中具有活性,这些细胞可能具有某一特定组织(优选的是象本发明涉及的第一个方面所述的那样),或具有一个以上组织或所有组织,在这种情况下该动物可能成为一个以上组织的供体。这种动物可被认为是非转化(在非繁殖意义上)细胞的集合。
本发明的第五个方面是提供能够表达一个或多个同源补体限制因子的非转化动物细胞,而这些同源补体限制因子在相对于动物细胞为相异的物种中具有活性。
本发明的第六个方面是提供重组DNA,它包括编码至少一种同源补体限制因子的DNA以及保证该编码DNA被非转化动物细胞表达的一个或多个序列。该动物细胞可以是遗传上加进了该构建物的转基因动物的细胞。或者,该细胞为培养器官或其它组织,如胰岛。
本发明的第七个方面是提供适合于加到动物的遗传物质中去的遗传构建物,以产生转基因动物。该构建物包括编码至少一种同源补体限制因子的DNA以及保证该编码DNA在遗传上加进了该构建物的转基因动物的至少某些细胞中被表达的一个或多个序列。这种构建物可能是以极微染色体(已知为YAC)的形式存在。同上,同源补体限制因子将在相对于转基因动物为相异的物种中产生活性。
本发明的第八个方面是提供制备转基因动物的方法,该方法包括将编码至少一种同源补体限制因子的DNA以及保证该编码DNA在至少某些转基因动物细胞中表达的一个或多个序列结合到动物的遗传物质中。
总的来说生产转基因动物的方法正变得越来越广泛,所采用的具体步骤可以是本领域现今所使用的常规步骤。例如,WO-A-8800239公开了从理论上构建转基因动物所需要的步骤。
将构建物加到转基因动物的细胞中去的实际方法可以是通过微量注射、精子介导的加入法或任何其它合适的方法。前期遗传操作最好是在原核生物中进行。
编码HCRFs的DNA既可以eDNA形式得到也可用常规克隆技术推断出来。编码衰变加速因子(DAF)的DNA可能被最佳特征化并已由Medof等人所描述(PNAS 84 2007-2011(1987))。Rey-Campos等人(1988)(在上述引文中)给出了RCA基因群的物理图谱。DAF变体及其通过DNA重组技术的制备方法公开于EP-A-0244267;这类变体可用于本发明。
由于DAF较好的遗传学特征和编码DAF的cDNA的已知序列,DAF可构成一个优选同源补体限制因素。
本发明第二至第七方面的其它优选的特征同第一方面,在细节上已作必要的修正。

实施例1
异种移植物排斥在无任何抗物种抗体存在下发生
一般说来,在没有循环免疫球蛋白情况下动物无法生存。这些免疫球蛋白是在对抗原刺激应答中由淋巴细胞产生的。然而,在早期的新生儿生命中,被动转移的母体免疫球蛋白可作为这种自身产生的抗体的暂时替代物,这种被动转移的免疫球蛋白在获得早期免疫经历的同时为幼小动物提供保护。在哺乳动物中,这种母体免疫球蛋白的被动转移通常经胎盘和通过初乳发生。然而,在几个物种中,胎盘的结构使得母体抗体不能通过这一途径转移。猪就是这样的物种之一。所有母体抗体都是从初乳中获得。因此,新生哺乳前的猪从理论上讲无免疫球蛋白。
取刚出生的大白猪,在不给喂奶的情况下将它们置于用热水袋温热的木笼中。每次实验从每胎小猪中取两只猪。这些动物出生时体重约为1Kg。
重约300g的新西兰白兔幼崽用作供体。这些供体用海卜那和安定麻醉,打开胸腔并用-19号针经静脉腔插管。灌注冷(+4℃)心麻痹液(Thomas No.2)直到心脏停止跳动并充满心麻痹液。也用直接来自注射器的冷心麻痹液从外部冷却。然后采用标准外科技术取出兔的心脏并在需要前贮存于+4℃的心麻痹溶液中。已发现采取这些措施是必要的,因为已证明兔的心脏极易患局部缺血性损伤。
通过吸入氟烷/02初步麻醉受体猪。然后将静脉内蝶形(23号)针插入乳房静脉,通过静脉内氯胺酮维持麻醉。同时用静脉内盐水使猪保持水合。移植前抽取血清和EDTA血样。
在Heron的方法(Acta pathol Microbiol Scand79 366-372(1971))之后将兔的心脏移植到猪的颈部。主动脉从末端至侧面(6-0Prolene)与颈动脉交织,肺动脉与颈静脉交织。所有其它心血管都连接起来。在移去夹钳后几分钟内心脏开始跳动。在整个过程中通过透皮玻片/ECG监测器监测心率。在实验期间未缝合猪的颈部,通过加盖抗滑薄膜保持心脏湿润。
图1A至1E显示了ECG结果。图1A所示的示踪图表明移植后即刻开始正常的心脏跳动。大约20分钟后(图1B)心跳开始衰退,1小时之内(图1D)无可测的心脏跳动,表明过急排斥。
因此这一实施例说明即使在无抗体存在下也会发生相异性异种移植物的过急排斥。
实施例2
实施例1中所用的乳猪无抗物种抗体
用Davies & Howard改进的(未发表)Greenwood等人的方法(Biochemical Journal 89 114-123(1963))对兔抗猪IgG进行放射性碘标记。
将下列成分迅速连续地加到聚苯乙烯试管(LP2 6Cm×1Cm)中:
25-50μl蛋白质(浓度为1mg/ml)
3-4μl Na125I(100mCi/ml)
10μl氯胺-T(*4mg/5ml;0.5M)
磷酸钠缓冲液(PH7.5)
*必须用前新鲜配制
将这些成分连续搅拌30秒钟使之混合。然后迅速加入下列物质:
50μl DL-酪氨酸(0.5ml磷酸钠缓冲液(PH7.5)的饱和溶液)
300μl2%BSA/PBS/叠氮化物
用以PBS/叠氮化物配制的Sephadex G-25(中级)小柱(8Cm×1.0Cm)将标记蛋白与未反应的碘分离。将碘化反应混合物定量转移到制得的G25柱中并用PBS/叠氮化物洗脱。将6滴级分收集到聚苯乙烯试管(LP2)中。洗脱柱直到蛋白和(125I)碘峰均被洗脱下来,测定所有级分的放射性。
掺入蛋白质中的放射性可如此计算:
蛋白质的放射性计数=原始总计数-碘峰的计数
在对乳猪抗体的测定中使用放射性标记IgG(称之为“同位素”),见下述:
材料

方法
1.制备兔红血细胞的1%PBS悬浮液并将100μl的量加到试管中。将细胞离心成小团,弃去上清液。
2.由成年猪(阳性对照),乳猪(试样)或兔(阴性对照)在PBS/BSA中制备失活血清的系列稀释物。将0.025ml的量加到红细胞团中,一式两份。将试管在4℃下保温4小时。
3.保温后,用PBS/BSA洗涤试管3次,然后将0.05ml同位素加到各试管中并于4℃保温过夜。
4.再洗3次试管并用γ计数器进行1分钟计数。
5.将结果按计数的的数目/分对滴度绘图。
结果示于图2。兔血清用作阴性对照,成年猪血清用作阴性对照。可以看出哺乳前的猪2中猪抗体水平可与阴性对照水平相比。
实施例3
补体C3与异种移植物破坏相关联的论证
将得自于Burger等人所述的品系(Eur.J.Immunol.167-11(1986))的补体缺乏豚鼠进行心脏移植,所用技术与在实施例1中对兔至猪异种移植所描述的方法基本相同。大鼠经吸入醚而麻醉,用心麻痹冷却心脏并如前所述切除。用静脉内安定和肌肉内海卜那麻醉豚鼠供体。如前所述将心脏植入颈内。对于对照豚鼠,即补体水平正常的豚鼠,通常在几分钟之内发生移植排斥,因而不必缝合颈部,在实验动物中,将颈部缝合并每日触诊两次进行监测。在手术后数小时内观察到正常ECGs,表明无过急排斥。
实施例4
A.猪淋巴细胞和肾细胞可经交替途径激活人补体
按照Grabar和Williams的技术(Biochem BiophysActa 10 193(1953)),将琼脂糖凝胶1倒在8×8Cm的玻璃板上(图3)。需要10ml凝胶混合物,该混合物由5ml2%琼脂糖和5ml巴比妥缓冲液(VB)组成。(VB是75mM巴比妥钠,10mM EDTA,10mM NaN3,PH8.5。)恰在使用前于60℃下将琼脂糖和VB混在一起。将凝胶倒在一水平台面上并冷却。凝固时,凝胶由1%琼脂糖组成,其深度约为1.5mm。
从离凝胶一端若1Cm处挖直径为3mm的孔3。每孔能容纳大约8μl待测样品。样品除加入足够溴酚兰使之着色外未进行特殊制备。点样后,将凝胶小心放到电泳槽的平台上。然后沿最靠近孔的凝胶一边轻轻压上浸在VB(操作缓冲液)中的棉纱布条,将另一纱布条压到琼脂糖的对边。(确保纱布条的两端浸泡在缓冲液槽内是重要的。)然后使25-30mA的电流通过凝胶直到白蛋白(用结合溴酚兰显现)到达正极纱布条。该过程需花大约2.5~3小时。若同时平行操作两块或多块凝胶,则所加电流必须相应增加,以致两块凝胶需50mA,三块需75mA等等。
当电泳完全时,如通过用溴酚兰显色的白蛋白标志9的移动所示,从电泳槽中取出凝胶。
B.二维交叉免疫电泳(2D-火箭电泳)
切下得自(A)的含有电泳后的蛋白质的凝胶条并放在一新玻璃板13的一端。然后将含有约1%抗待显色蛋白质的抗血清的2%琼脂糖∶VB的1∶1混合物15倒在玻璃板上并使之凝固。当其冷却至大约50℃时将抗血清加到琼脂糖/VBS混合物中。
然后如上所述用火箭平板进行电泳,含有第一维条带的凝胶一端通过棉纱布条与负极17相连,另一端与正极19相连。在取决于凝胶大小的电流下使凝胶电泳过夜;每8Cm长度的凝胶需要10mA,因此16Cm长的凝胶需20mA电流等等。
通过第一维电泳分离蛋白质,通过第二维电泳对蛋白质进行定量和显色。下面将描述为进行显色的染色法。
C.碾压和染色凝胶
这一程序与常规免疫电泳法或火箭电泳法相同。将待染色的凝胶盖上一层预先用水润湿的无纤维POSTLIP(商标)纸(AdlardEvans & Co)。然后盖上6层吸水纸。将该组合件碾压1小时,然后除去所有的纸并重复这一过程。
在第二次碾压后,在温热的气流下使凝胶干燥,然后将凝胶在PBS中浸泡至少1小时以除去不沉淀的蛋白质。然后再使凝胶干燥,并在0.5%W/V考马斯亮兰G250,45%H2O,45%甲醇,10%乙酸的溶液中染色10分钟。
通过在20%甲醇,6%乙酸中连续漂洗而使凝胶脱色直到背景清晰为止。然后在温热的空气中最后干燥。
图5是干凝胶的再制。火箭1是含有50μl正常人血清(HHS)+25μl包括10m MEGTA在内的VBS的阴性对照。EGTA是一种除去钙的螯合剂;钙是经典途径补体活化所必需的,因此EGTA的存在可确保补体只能经交替途径激活。左手侧(较大的)峰是C3,右手侧(较小的)峰是C3bi,即活化C3的分解产物。在对照中,少量的C3bi表明只有少量的补体活化。
在火箭2中,将75%的猪红血球(V/V)加到鸡尾酒式缓冲液中。C3bi水平稍有增加,但可能不明显,从而表明猪红血球只能通过交替途径勉强激活人补体。反应差的原因还不清楚。
在火箭3和4中,分别将75%猪淋巴细胞(V/V)或75%猪肾细胞(V/V)加到鸡尾酒式缓冲液中。在每种情形下,C3bi水平都有明显提高,这表明人补体被猪淋巴细胞活化。
实施例5
猪淋巴细胞在猪补体存在下不被人抗体溶解,但在兔或人补体存在下被溶解
用铬释放测定法监测在猪补体、幼兔补体或人补体存在下由人血介导的细胞溶解。
材料
淋巴细胞分离介质-Flowlabs
RPMI1640+10%失活FCS
PBS(无叠氮化物)-OXOid
具V型孔的平板-Sterilin
幼兔补体淋巴-血清-凝乳酶-成人或猪补体(用RPMI稀释1+7)
热失活的血清(56℃30分钟)
方法
1.在等体积的Ficoll Hypaque淋巴细胞分离介质中加一层用PBS以1∶1稀释的去纤维化的猪金血。在1200g下于20℃将试管离心30分钟。
2.移出交界处产生的猪淋巴细胞,在PBS中漂洗一次,将细胞团重新悬浮于RPMI1640中并将细胞计数调节到2×107/ml。
3.将200uCi的51Cr加到2×107细胞团中并于室温下保温1.5小时。
4.在900g下用5分钟将标记细胞洗涤两次并用RPMI将最终细胞计数调节到1×106/ml。
5.将0.05ml量的以系列稀释物形式用作试验的失活血清一式两份以及对照一起进行制板。将稀释的补体以0.05ml量加到相应的孔中,然后加入0.05ml标记细胞。于30℃在CO2烘箱中将平板保温1小时。
6.保温后,将平板在900g下离心15分钟以使细胞沉降。将100ul上清液移入标记试管并用γ计数器进行1分钟计数。
7.将结果以原始标记细胞计数%对滴度绘图。
对照

结果
结果示于图6。可以看出猪淋巴细胞只在非猪(即兔或人)补体存在下被人血清溶解,但在猪抗体存在下不被溶解。结论是存在于猪细胞上的一种或多种同源补体限制因子可成功地抑制猪补体的作用而不能抑制人或兔补体的作用。
实施例6
本实施例的目的是证明抗体可引起过急排斥。由于观察到过急排斥可在无抗移植物抗体存在下发生,但需要功能性补体,从而产生了这一作为本申请基础的概念。因为这是一个新发现,所以在证实抗体可引起异种移植物排斥的文献中没有给出实验。由于在天然存在抗体存在下难于测定这些抗体是否起某种作用,所以这种实验不易进行。在本实施例中,通过输入具有适宜特异性的抗体将同相的异种移植物转变成相异的异种移植物已证明了抗体的作用。在这一研究中所用的接受体是3至6月龄,重250-300g的PVG品系的雄性大鼠(RTIC)(Banting&Kingdom,Bicester,Oxon.,UK)。心脏供体是Syrian仓鼠,也得自Banting & Kingdom,重100-150g。按照Heron的方法进行心脏移植(实施例1中所述文献)。将仓鼠心脏移植到大鼠的颈内,用套管技术使主动脉与颈动脉连接,肺动脉与颈静脉连接。所有其它血管都连接起来。打开血管钳后数分钟心脏开始跳动,通过外部触诊进行监测。为经吸入氟烷和氧气而麻醉的动物施行全部手术。
抗仓鼠溶解性抗体水平如下测定:将50ul 1%仓鼠红血球溶液加到50ul已进行系列稀释的试验血清中。加入50ul 1/7幼兔抗体稀释液(Sera Lab,Crawleg Down,Sussex)并于37℃保温1小时。加入750ul补体结合稀释剂并离心(Beckman MICROFUGE,13000rPm,4分钟),然后测定上清液的OD415。(MICROFUGE是商标)。阴性和阳性对照分别为加到1%细胞溶液中的CFD和蒸馏水。将OD415读数结果对X轴上的血清滴度绘图。从图7可以看出,将仓鼠心脏移植到大鼠体内使大鼠产生极高滴度的溶解性抗仓鼠抗体。使用标准柱层析技术(“The use of SEPHADEX in theSeparation,Purification and Characerisationof biological materials”,Curling in Exp inPhysiol and Biochem 3(1970)417-484(G.A.Kerkut,Ed.)Academic Press,London andNew York,1970)在SEPHADEXG 200(PharmaciaGB Ltd,London)上进行柱层析将取自某些大鼠的血清分离成其成分蛋白级分。(SEPHADEX是商标)。从柱上收集7ml级分,如上所述对溶解性抗仓鼠活性进行测定。图8表明尽管这些抗体是由心脏移植所诱导的,但抗物种活性几乎全部存在于IgM级分中,测定活性之后,用C×10超滤器(Pharmacia)将级分浓缩至0.5mg/ml的浓度并在用前贮存于-70℃下。
为了检验它们与只溶解红细胞相反的破坏异种移植物的能力,将仓鼠心脏移植到首次用来作实验的大鼠中。仓鼠心脏跳动一经建立,就将2ml纯净的血清成0.5mg含有溶解性抗仓鼠抗体的纯化兔疫球蛋白静脉输注到大鼠体内。未分离的血清和0.5mg IgM二者都一致引起仓鼠心脏移植物在15分钟内被破坏。输注IgG的结果与某些使移植物丧失功能的制剂不一致,即输注IgG后心脏移植物继续跳动。当将来自G200柱的白蛋白作为对照输入时,心脏移植物一直存活并在该模型的正常时间(3天)内发生排斥。这说明该抗体与移植物的结合会引起过急破坏。
实施例7
到目前为止在本申请中所示的数据已证明异种移植物的破坏可包括通过交替途径或抗体介导的补体活化(经典途径)。此外,在异种移植物靶表面上的补体调节剂可使之不被同源而不是异源补体破坏。两种补体活化途径所共有的关键活化步骤是补体C3成分的裂解。这一裂解是由C3转化酶,C4b2a(经典途径C3转化酶)或转化酶C3bBb(交替途径C3转化酶)引起的。这些酶可裂解C3为C3b,反过来C3b又可保证交替途径形成更多的C3转化酶(反馈回路)。结果补体系统能够迅速扩大C3b在“外来”靶上的沉积。然而许多C3b不能成功地与外来靶相互作用而残留在液相,因而可不加选择地与宿主细胞结合。为了使这些细胞兔受补体无选择结合的进攻,对照蛋白已发展到使液相中或结合于自身组织上的补体成分失活。
在控制C3过程中所涉及的糖蛋白都是在人染色体1中被称作RCA(补体活化调节剂)位点的一个区域内在遗传上相关。在本实施例中,发明人证明已通过融合技术获得了人染色体1并可表达其表面上RCA部位的蛋白质的小鼠细胞在异种移植物破坏的体外测定中表现得好象它们是人细胞而不是小鼠细胞。
细胞系
T5是用Bird等人的技术(Nature289 300-301(1981))生产的EPsfein Barr病毒转化扁桃体B-细胞系。B10是产生人抗破伤风单克隆抗体的杂交瘤,它是由人B淋巴母细胞系(BLL)与小鼠骨髓瘤细胞系X63-AG8.653融合而衍生的(Kierneyet al.(J.Immunol。123.1548-1550(1979))。T5和B10细胞系可得自MSC Carter and Dr NC Hughes-Jones of theMRC MITI Group at Babraham,Cambridge。DB3是可产生抗黄体酮单克隆抗体的小鼠杂交瘤细胞系(Wright et alNature 295 415-417(1982))。获得了下列特异于人染色体1的寡核苷酸引物:(5′-CCACAGGTGTAACATTGTGT-3′)和(5′-GAGATAGTGTGATCTGAGGC-3′);这些引物分别在人抗凝血酶2(AT3)基因的上游和下游,已知该基因位于人染色体1上(Wuet alNucl Acids Res 17 6433(1989))。可用本领域技术人员熟知的技术合成该寡核苷酸。
用Harrmann和Frischauf的方法(MethodsEnzymol 152 180-183(1987))制备高分子量基因组DNA。简言之,用5ml TNE(100mm Tris pH7.5,100mM NaCl,100mM EDTA1%Sarkosyl)溶解来自各培养细胞系的100×106细胞并用新鲜蛋白酶K(100ug/ml)进行处理。用苯酚(用水饱和并用0.1M Tris,PH8平衡)、苯酚∶氯仿(1∶1,V/V)、然后氯仿∶异戊醇(24∶1,V/V)提取该制剂。通过乙醇沉淀获得DNA,用NaCl调至100mM的TE(100m MTris PH8.0,1mM EDTA)和单独用TE在4℃下透析。用0.5%琼脂糖凝胶分析所分离的DNA并由在260nm处的光密度确定其浓度。如Saiki等人所述(Science 239 487-491(1988))对每种细胞系进行聚合酶链反应(PCR)。在含有500ng基因组DNA、1、2ng各种引物及2.5单位TagDNA多聚酶(水生物热菌3型)(Camkio Ltd,Camkzidgeuk)的100ul体积中使用缓冲液提供该酶。核苷酸(dNTPs)(Boehringer Mannheim Diagnosticsand Biochemicals Lal,Lewis,East Sussex,uK)的浓度各自为2mM。使用程控温度调节器(Genetic ResearchInstrunmentation Ltd,Dunmow,Essex UK)93℃下变性1分钟:55℃下退火1分钟:和72℃下伸展2分钟扩增DNA30次。用在Tris硼酸EDTA缓冲液中操作的2%琼脂糖凝胶直接分析10ul反应产物,通过与HincII消化的噬菌体X-174-rf DNA(Pharmacia LKB Biotechnology,Upsala,Sweolen)比较确定产物的大小。
用抗-DAF(衰变加速因子)单克隆抗体(Kinoshita etal(J.Exp.Med 162 75-92(1985))和荧光素结合第二抗体处理培养的T5,B10和DB3细胞。使细胞(1×106)与小鼠抗-DAF单克隆抗体1A10(IgG2a10ug/ml,于100ul 10%FCS0.1%叠氮化物中)。从美国纽约纽约大学医疗中心M Davitz博士那里获得了1A10(Kinoshita et al(J.Immunol.136 3390-3395(1986)))。空白对照是单独的缓冲液。在冰上保温2小时后,在含有1/100FITC结合山羊F(ab′)2抗小鼠IG(亲合纯化的重链和轻链和所吸附的人IG)(TagoImmunochemicals Inc,Burlingame,California,USA)的100ul缓冲液中将细胞洗涤3次,重新悬浮并在冰上保温1小时。某些细胞只与第二抗体一起保温以作为染色对照。由于DB3是分泌IgG1的小鼠细胞系,所以为了消除在染色DB3细胞时出现抗IgG1反应性还使用预先用等体积DB3细胞吸附的山羊抗小鼠IG或FITC结合绵羊抗小鼠IgG2A(1/40)将所有细胞用200ul缓冲液广泛洗涤并重新悬浮。使用用于荧光激活细胞分类(FACS)分析的Beckton Dickinson FACS-STAR仪检测DAF阳性细胞。(FACS-STAR是商标。)
用PCR法测定人染色体1是否存在于三种不同的培养细胞系,T5(人),B10(人-小鼠)和DB3(小鼠-小鼠)中。图9示出扩增后T5和B10二者都有495碱基对的谱带,而DB3(即小鼠-小鼠杂交细胞)根本无谱带。根据导用AT3引物得到的PCR产物由2个等位基因组成,大小为572碱基对(等位基因1)和496碱基对(等位基因2)Wu et al,在上述引文中)。见于T5和B10基因组DNA的谱带相当于等位基因2。这表明人-小鼠杂交细胞系B10含有人染色体1。
对DAF存在的FACS分析表明大多数人T5细胞(85.7%)用抗-DAF单克隆抗体染色呈阳性。类似水平(83.1%)的阳性细胞见于小鼠/人杂交B10细胞。小鼠-小鼠杂交OB3细胞对抗-DAF处理的制剂和未处理的制剂显示出相同的染色图形。然而,若(1)使用FITC结合绵羊抗小鼠IgG2a或(2)用DB3细胞预先吸附上述山羊抗小鼠IgG,则可消除这种抗小鼠IgG1反应性。结果表明人-小鼠杂交细胞系B10可表达细胞膜表面上的人DAF,这可用特异性抗-DAF单克隆抗体加以检测。其表达水平与人细胞系T5的表达水平相同。小鼠-小鼠杂交瘤细胞系不能表达人DAF。
如上面实施例5所述,对这些细胞系进行铬释放胞毒细胞杀伤研究。图10表明,当将猪抗人抗体与T5人细胞系一起保温时,加入兔补体后使细胞溶解,而当加入人补体时不发生细胞溶解,当然,这是因为T5细胞系会具有人HCRFS。这是对实施例5结果的验证。当将人抗体用于人细胞系时,用人补体或用兔补体都不发生细胞溶解,这表明没有自动抗体。铬释放技术不允许保温持续足够长的时间以检测任何显著水平的人细胞对兔补体的交替途径活化(图11)。然而,当将人抗体与DB3小鼠-小鼠杂交瘤细胞系一起保温(图12)时,用兔补体和人补体均可杀伤细胞,这表明人补体确实可在这种测定中发挥功能。当使用具有人染色体1并已知至少可表达DAF的B10人-小鼠杂交细胞时,兔补体引起细胞系溶解,而人补体不引起细胞系溶解(图13)。对此的解释是由于小鼠-人杂交细胞上具有染色体1而得以表达的人HCRFS抑制了人补体的活性。
实施例8
前一实施例表明具有染色体1可防止异种移植细胞破坏。这是强有力的间接证据以证明可使小鼠细胞免受异种移植物破坏的是CRA部位。本实施例将提供正式证据。在本实施例中,可证实用人MCP转染非人细胞系并使之经受人或兔补体作用的效果。
按Lublin等人所详述的方法(J.EXP.Med.168181-194(1988))生产MCP的CDNA。用表达质粒SFFV neo,按Fuhlbrigge等人所述的技术(ProcNatl.Acad.Sci.85 5649-5653(1988))进行转染细胞系的构建。其包括形成Friend脾病灶病毒5′长末端重复(SFFV.LTR)(Clark and nak(Nucl.Acids.Res 10 3315-3330(1982))和(Proc Natl Acad.Sci.80 5037-5041(1983)))。细胞系得自美国典型培养物保藏中心(12301Parklanin Drive,Rockville,Maryland,USA)。所用细胞系是CHO-K1(ATCC CCL61)和NIH/3T3(ATCCRL1658)。用抗MCP的单克隆抗体(Andrews et al Ann Hum Genet 49 31-39(1985))和已述的FACS分析证实其基因的表达在某些情况下,用标准技术通过FACS的细胞分类选出高水平表达MCP的细胞系。
本实施例说明用MCP转染CHO细胞的效果。因为这些细胞系长成单层,所以如de Bono等人所述(Immunology 32221-226(1977))通过末端 腺嘌呤摄取测定法对细胞杀伤情况进行估价。简言之,该测定法包括在平底无菌96孔平孔平板中将细胞培养物与补体和抗体一起保温。在实验保温期末,通过培养物摄取放射性腺嘌呤的能力估价细胞存活性。活性细胞将摄取腺嘌呤,死细胞则不行;因此活细胞计数高,死细胞计数低。
同许多转化细胞一样,CHO对天然存在的抗体和补体交替途径的作用不敏感。然而,这些细胞对已证明可引起实施例6所述的仓鼠心脏异种移植物破坏的抗体是敏感的。由于CHO细胞得自于仓鼠,所以这些抗体杀伤具有人和兔补体的CHO细胞(图14)。当用人MCP转染CHO细胞时,细胞只能在兔补体存在下溶解。由于人MCP存在于仓鼠细胞系的表面而抑制了人补体(图15)。细胞未被杀伤实际上是由于C3转化酶的缺乏这一看法的证据是在CHO细胞保温之后通过如上面实施例4所述的火箭免疫电泳分析人C3的分解所提供的。可以看出,仅在对照水平上补体未发生裂解(图16)。
这些资料进一步证实在非人细胞表面上的遗传工程补体抑制蛋白质可使那些细胞不受过急异种移植物破坏机制的影响,该机制的共同特征是需要补体C3成分的裂解。
实施例9
接实施例8的步骤,用编码MCP(MCP克隆K5·23)的CDNA转染3T3鼠成纤维细胞。如实施5所述向细胞中加入51Cr,然后将1份体积的该细胞与1份体积的热灭活的人补体和1份体积的于4℃被鼠脾细胞预先吸收以从人补体中除去抗鼠抗体的人补体一起保温,将该混合物及其补体系列稀释液铺板,铬释放检测的一般条件和特点如图5所述。从克隆5·23得到的结果示于图17,它也表明,作为对照MCP CDNA的作用从反方向介入(在此情况下它不被转录)。正确转录的MCP CDNA保护细胞免受杀伤,这一点由相对低水平的51Cr释放所证实,而非转录CDNA不能提供显著的保护作用,这一点可由相对较高水平的51Cr释放所证实。
实施例10
对于用DAF的CDNA转染的L1/210细胞(小鼠白血病细胞系)可以获得与上面实施例8所述相似的结果。DAF的CDNA生产方法如Lublin & Atkinson所述(Ann.Rev Immunol.7 35-58(1989))。
实施例11
如上面实施例8所述,制备MCP的CDNA并与SFFV.neo连接。
使用这种DNA制剂,按Manipulating the MouseEmbryo,A Laboratory Manual by B.Hogan etal Colcl Spring Harbour Laboratory(1986)中所述的方法生产基因转移小鼠。通过腹膜内注射5单位得自妊娠马的血清促性腺激素(市售产品为Folligon),48小时后,腹膜内注射5单位来自人妊娠尿的绒膜促性腺激素(市售产品为Chorulon)使10~15只3-4周龄雌性小鼠(CBA×B10)f1超数排卵。在注射Chorulon当天,让雌鼠与(CBA×B10)F1雄鼠交配,次日用颈部脱位法处死塞有阴道栓的雌鼠并从其输卵管中分离出受精卵。
用这种方法分离出的300-400个卵含有两个在放大400倍的Nomarski差示干扰对照镜片下清晰可见的原核。向两个原核之一中注射大约2000拷贝浓度为0.5~2ng/ml含有MCPCDNA转移基因的DNA制剂。
将微量注射后存活的卵再植入(CBA×B10)F1雌鼠的输卵管中,该雌鼠在前一夜已与切除了输精管的雄鼠交配,因此出现假孕(即,它们已排卵并且其激素状态呈妊娠状态,但其卵母细胞并未受精)。在微量注射的当天或当卵处于二细胞期时的第二天,将约30个经微量注射的卵转移到每只麻醉的假孕雌鼠的输卵管中,结果是正常妊娠并由10个母体内生出17只小鼠。
通过狭槽吸印法和/或Southern吸印法(见实施例8),以及PCR,来自尾部皮肤细胞的DNA分析,采用32P-标记的探针和识别转移基因的引物筛选后代。后代之一(雄鼠)证明发生了MCP DNA序列的基因转移。
实施例12
重复实施例11的步骤,所不同的是用如实施例10所述的DAF的CDNA代替MCP的CDNA。从10个母体内生出23个后代。其中3只小鼠(两只雌鼠,一只雄鼠)发生了DAF基因转移,如Southern吸印法所示。
实施例13
将得自实例11含有人MCP CDNA转移基因的雄性鼠培养至成年并与(CBA×B10)F1雄鼠交配。生出11个子鼠。使用标记的人MCP CDNA作为探针通过slot-blot分析法对每个后代的尾部细胞DNA进行筛选,以测定转移基因是否被遗传。所得结果示于图18上半部分。可以看出子代0、1、5、7、8和10已接受遗传。(使用四个对照:人DNA(H);鼠DNA(M);与10pg人MCP标记的CDNA混合的鼠DNA;与100pg人MCP标记的CDNA混合的鼠DNA〕。
实施例14.
将得自实例12含有人DAF CDNA转移基因的雄性小鼠培养至成年,并与(CBA×B10)F1雄鼠交配,使用标记的人DAF CDNA作为探针通过slot-blot分析法对每一个产生的子代进行尾部细胞DNA进行筛选,以测定转移基因是否被遗传。所得结果示于图18的下半部分。可以看出,子代13·3(雌性)接受遗传〔采用四个对照:人DNA(H);鼠DNA(M);与10pg人DAF标记的CDNA混合的鼠DNA;与100pg人DAF标记的CDNA混合的鼠DNA〕
Claims (18)
1.一种降低移植组织的免疫排斥反应的方法,其中该组织得自于不同于接受体物种的供体,供体物种是与接受体相异的物种,该方法包括向待移植组织提供和/或使其表达一种或多种在受体物种中有活性的同源补体限制因子(HCRF),从而防止供体组织在移植到受体中时发生补体的完全活化。
2.根据权利要求1的方法,其中组织是一种器官。
3.权利要求2所述的方法,其中器官是心、肺、肝、肾、胰或甲状腺。
4.权利要求1所述的方法,其中组织包括血液或造血细胞,胰岛,脑细胞或来自内分泌器官的细胞。
5.权利要求1-4任一项所述的方法,其中HCRF可干扰经典和交替途径二者所共有的补体活化级联的那部分。
6.权利要求1-5任一项所述的方法,其中HCRF是天然HCRF。
7.权利要求5所述的方法,其中HCRF可调控C3处的补体活化。
8.权利要求7所述的方法,其中HCRF是或具有下列物质的活性:
因子I(以前也称为C3b失活剂或KAF),
因子H;
C4结合蛋白;
DAF(也称为CD55);
膜辅助因子蛋白(MCP,也称为CD46,首次描述为gp45-70,进而称为gp66/56);
CR1(也称为C36/C46受体或CD35);
和/或
CR2(也称为CD21,C3.dg受体,3d/EBU受体和p140)。
9.权利要求1-8任一项所述的方法,其中HCRF具有天然HCRF的活性,天然HCRF的基因位于RCA(补体活化调节剂剂)部位,即染色体1图谱的g32带处。
10.权利要求5所述的方法,其中HCRF可调控C8和/或C9处的补体活化。
11.权利要求10所述的方法,其中HCRF是或具有下列物质的活性:
C8bP(也称为HRF或MIP);
P-18(也称为HRF-20,CD59或MIRL);或
SP40.40。
12.权利要求1-11任一项所述的方法,其中HCRF是与膜结合的。
13.权利要求1-12任一项所述的方法,其中以将其与供体组织上的细胞膜相结合的方式提供HCRF。
14.权利要求13所述的方法,其中供体组织是发生基因转移的,它含有并当植入接受体时,表达编码一种或多种在受体物种中有活性的HCRF的核酸。
15.权利要求1-14任一项所述的方法,其中接受体物种是人。
16.权利要求1-14任一项所述的方法,其中供体物种是猪。
17.得自于猪、兔、大鼠、仓鼠和小鼠等供体物种的心脏和一种或多种在猪、兔、大鼠、仓鼠和小鼠等受体物种中有活性的DAF或MCP在制备可植入受体物种的组织中应用,其中供体物种是与接受体物种相异的物种。
18.一种制备基因转移动物的方法,该方法包括将编码至少一种同源补体限制因子的DNA和一种或多种能使编码DNA得以在基因转移动物的至少某些细胞中表达的序列掺入某些动物的遗传物质中。
