Abstract

用来防治叮咬人和/或温血动物的蚊子和其它 昆虫驱避剂组合物,它含有以重量计低于5%的,选 自以下组分的:对-氨基苯甲酸烷基酯(I),2-氨基 乙酸芳酰胺(II),2-氨基烷基芳基醚(III),对-氨基 苯甲酸2-氨基烷基酯(IV)及其混合物的局部麻醉 药。特别优选的是利度卡因和利度卡因与丙胺卡因 的混合物。

Description

本发明涉及一种昆虫驱避剂,特别是一种用于防治叮咬人和/或温血动物的蚊子或其它昆虫,尤其是一种飞行种类昆虫的驱避剂。
多年以来,许多物质曾被建议作为驱避组合物的活性成分用来防治蚊子,例如,邻苯二甲酸二甲酯和N,N-二乙基-间-甲苯甲酰胺(避蚊胺)。这些组合物施于皮肤之后,驱避剂在一段时间内通过蒸发而被释放。然而,这种过程通常很快,因而减少了它们的使用。再则,业已发现具有昆虫驱避性能的已知主要活性成分易于被皮肤吸收并可能引起皮肤过敏、皮肤刺激等。再有的缺点是,它们很容易从皮肤上自然去除。有些已知的昆虫驱避化合物具有明显的且令人很不愉快的气味。
为了延长它们的效果,曾试图将具驱虫性能的主要活性成分制成不同的制剂形式。
美国专利4,477,467公开了一种昆虫驱避剂组合物,它含有与芳香载体组合的作为活性成分的避蚊胺。这种组合物通过包括主要活性成分配合物的制剂,提供加大的阻力以防止皮肤吸收。
美国专利3,590,118号公开了一种成膜组合物,它含有与水溶性聚合物残留物混合的已知驱避剂。有效保护时间因此被延长到约24小时。然而,这种组合物在皮肤上形成的膜容易破碎,驱避效果因此大大减小。
本文中,“昆虫驱避剂”为一种药剂,它防止昆虫穿透人或动物皮肤叮咬人、哺乳动物或鸟。这表示被驱避的昆虫可以接近皮肤但却不叮咬。
本发明的目的之一是提供一种昆虫驱避剂组合物,与已知的昆虫驱避剂组合物相比,它具有优越的性能,尤其是在防治效果,持效期和被皮肤吸收和皮肤刺激方面。
根据本发明,本文提供一种昆虫驱避剂组合物,它含有选自以下的组分:对-氨基苯甲酸烷基酯(Ⅰ),2-氨基乙酸芳基酰胺(Ⅱ),2-氨基烷基芳基醚(Ⅲ),对-氨基苯甲酸2-氨基烷基酯(Ⅳ)及其混合物的局部麻醉剂作为活性成分。
优选的对一氨基苯甲酸烷基酯(Ⅰ)为苯佐卡因(R=CH2CH3)。
优选的2-氨基乙酸芳酰胺(Ⅱ)为选自以下的组分:利度卡因(Ⅱ;R=CH3,R1=H,R2,R3=C2H5),丙胺卡因(Ⅱ;R=H,R1=-CH3,R2=CH2CH2CH3,R3=H),衣铁卡因(Ⅱ;R=CH3,R1=C2H5,R2=C2H5,R3=CH2CH2CH3)和卡波卡因(Ⅱa;R=CH3,R1,R2为由(CH24构成的桥,R3=CH3)。
优选的2-氨基烷基芳基醚(Ⅲ)为选自以下的组分酮卡因(Ⅲ,R=异丙基,Ar=2-(1′-氧代丁基苯基))和Kinisocaine(Ⅲ,R=CH3,Ar=3-丁基-1-异喹啉基)。
优选的对-氨基苯甲酸2-氨基烷基酯(Ⅳ)为选自以下的组分普鲁卡因(Ⅳ,R=C2H5,R1=H)和丁卡因(Ⅳ;R=CH3,R1=n-丁基)。

特别优选的是利度卡因。优选的还有丙胺卡因以及利度卡因和丙胺卡因的混合物。
用于皮肤的组合物中的活性成分的浓度优选的是以重量计不超过5%;更优选的是以重量计浓度为0.3至3%之间。除了活性组分外,根据本发明的具昆虫驱避性能的组合物包括一种载体,它可以是一种软膏,乳液,乳膏,饱和油或有机溶剂或溶剂混合物。根据本发明的组合物还可以包括一种喷雾制剂。
根据本发明的组合物可以和用于制备膏剂,乳液,乳膏,饱和油溶液或饱和油悬浮液,有机溶剂液剂或溶剂混合物液剂的很广范围的非活性成分结合,诸如稳定剂,尤其是抗氧化剂,着色剂,填料,抑菌和抗霉剂,乳化剂,香料,维生素,拒水剂,增湿剂,辐射吸收剂,pH调节剂,胶凝剂,表面张力改善剂等。它们还可以包括抗生剂和促进伤愈剂,尤其是用于治疗家畜如牛或马的伤口。
根据本发明的一个方面,组合物可以含有一种透明质酸盐,尤其是透明质酸钠,尤为优选的是含有利度卡因和透明质酸钠盐的组合物。
前已提到,根据本发明的活性化合物对皮肤无刺激作用。因为主要活性成分是以低浓度施用的,并且是存在于不促进皮肤吸收的载体中,也没有任何可查觉的局部麻醉效果。局部麻醉药作为驱避剂的作用机理尚不完全明了。昆虫仅被适度地阻拦落于皮肤,但一旦落下却不吸血的事实表明,其作用机理是一种不同于已知驱避剂的作用机理,例如避蚊胺在气相中象激素一样发挥它们的效果。试验表明,驱避作用是由于大范围的D级局部麻醉而拥有的,即是化合物通过神经膜受体依赖性和受体非依赖性结合发挥其麻醉作用。
本发明通过以下实施例作更详细的描述。给出的百分比为重量比。

埃及伊蚊驱避试验
试验采用黄热病蚊子在+28℃和相对湿度为70%的养虫室中进行。自愿者的手经洗净干燥。根据实施例5的乳膏剂以6.8mg/cm2的量涂于一只手。将处理过的手插入一个约有30只饥饿雌蚊子的网笼内。一分钟调节期后,记录在一分钟内飞到皮肤上,落到皮肤上和吸血的蚊虫数量。在乳膏剂施于皮肤之后0、4、6、和8小时分别进行实验。另一只手作为对照。结果总结成表1。

结果很明显,根据实施例5的乳膏剂对埃及伊蚊有良好的驱避效果。
实施例8
在养虫室中进行对比试验。在+21℃的条件下,用兔子分别测试根据实施例2的含有利度卡因,卡波卡因,衣铁卡因,丙胺卡因,丁卡因和酮卡因的成对组合物的家蚊的相对效果。通过剃除相应左右背部大小为5×5cm的正方形区域将毛去除,将此区域标记好。每个区域施用一种组合物。动物固定于大小和空间方向与剃除区域匹配的一对养虫室的窗口。由此提供了只能接近的这些区域。记载在30分钟内各自区域的叮咬试图,按效果降低获得以下效果的等级:利度卡因≈丙胺卡因>卡波卡因≈丁卡因>丁哌卡因>酮卡因>苯佐卡因。利度卡因和丙胺卡因因没有观察到叮咬的尝试,而苯佐卡因与未处理区域相应大小观察到的叮咬次数相比:降低了约60%。
实施例9
根据实施例5的软膏剂,作了牛的伤口被牛虻(Tabanus  bovinus)叮咬的田间试验。与根据实施例5的基膏相比,可见愈合时间大大缩短(40%)。
实施例10
在大田研究中调查了根据本发明的几种活性化合物的驱避作用。白卡片表面(60×60cm)分别喷上含0.3g利度卡因,酮卡因和丁哌卡因(Ⅱ,R=CH3,R1,R2为(CH24桥,R3=正丁基)的95%的乙醇(5ml)溶液。第四张的表面(空白)只用乙醇处理。让卡片表面干燥10分钟,竖立于瑞典中部有稀疏树林的湖边地区的地上,时间为六月初(2点钟开始观察30分钟,多云天气,气温18℃,湿度70%)。这个地区大量滋生伊蚊(主要是A.Punctor和A.Communis)。所有驱避剂处理的卡片表面表现出减少了的着落频率:利度卡因为空白卡片表面着落率的27%,Kinisocaine为35%,酮卡因为45%,丁哌卡因为45%。与空白区域相比,蚊子一旦着落于处理区域,逗留的时间大为缩短。

Claims (14)

1、昆虫驱避剂组合物,特别是一种用于防止叮咬人和/或温血动物的蚊子或其它昆虫,尤其是某些飞行种类的驱避组合物,其特征在于:含有选自以下组分的麻醉剂作为活性成分:对氨基苯甲酸烷基酯(Ⅰ)、2-氨基乙酸的芳酰胺(Ⅱ)、2-氨基烷基芳基醚(Ⅲ)、对-氨基苯甲酸2-氨基烷基酯(Ⅳ)及其混合物。
2、根据权利要求1的组合物,其特征在于:对-氨基苯甲酸烷基酯(Ⅰ)为苯佐卡因(R=CH2CH3)。
3、根据权利要求1的组合物,其特征在于:2-氨基乙酸的芳基酰胺(Ⅱ)是选自以下的组分:利度卡因(Ⅱ;R=CH3,R1=H,R2,R3=C2H5),丙胺卡因(Ⅱ;R=H;R1=-CH3,R2=CH2CH2CH3,R3=H),衣铁卡因(Ⅱ;R=-CH3,R1=C2H5,R2=C2H5,R3=CH2CH2CH3)和卡波卡因(Ⅱa;R=CH3,R1,R2=(CH24桥,R3=CH3)。
4、根据权利要求1的组合物,其特征在于:2-氨基烷基芳基醚(Ⅲ)是选自以下的组分:酮卡因(Ⅲ;R=异丙基,Ar=2-(1′-氧代丁基苯基))和Kinisocaine(Ⅲ;R=CH3,Ar=3-丁基-1-异喹啉基)。
5、根据权利要求1的组合物,其特征在于:对-氨基苯甲酸2-氨基烷基酯(Ⅳ)是选自以下的组分:普鲁卡因(Ⅳ;R=C2H5,R1=H)和丁卡因(Ⅳ;R=CH3,R1=正丁基)。
6、根据权利要求1或3的组合物,其特征在于:2-氨基脂族乙酸的芳基酰胺为利度卡因。
7、根据权利要求1的组合物,其特征在于:混合物含有利度卡因和丙胺卡因。
8、根据权利要求1至7的任一项的组合物,其特征在于,它进一步包括一种载体,尤其是一种软膏,乳液,乳膏,饱和油,有机溶剂,溶剂混合物载体或喷雾制剂载体。
9、根据权利要求1至8任意一项的组合物,其特征在于:组合物进一步包含一种或几种用于制备软膏,乳液,乳膏,饱和油溶液或饱和油悬浮液,有机溶剂溶液或溶剂混合物溶液的非活性成分,例如稳定剂,尤其是抗氧化剂,着色剂,乳化剂,香料,维生素,拒水剂,增湿剂,辐射吸收剂,填充剂,抑菌和抗霉剂,胶凝剂,粘度调节剂,pH调节剂,皮肤软化剂和表面张力改善剂。
10、根据权利要求1至9的任意一项的组合物,其特征在于:组合物进一步包含抗生剂和促进伤口愈合的药剂。
11、根据权利要求1至10任意一项的组合物,其特征在于:组合物中含有透明质酸盐,特别是透明质酸钠。
12、根据权利要求11的组合物,其特征在于:组合物中含有利度卡因和透明质酸钠盐。
13、根据权利要求1至12的任意一项皮肤保护组合物,其特征在于:组合物中含有以重量计其浓度不超过5%的活性成分,优选的是按重量计浓度在0.3%和3%之间。
14、权利要求1至13任意一项所述组合物的应用,用来驱避吸食温血动物和人的昆虫,特别是蚊子和牛虻的用途。
