Abstract

本发明涉及从一种药用植物大叶蒟Piper laetispicum C.DC.中提取、纯化有效部位提取物的制备方法,以及该有效部位提取物在治疗、预防抑郁症等情感障碍性疾病的医疗、保健等领域的应用。大叶蒟粗提取物经大孔树脂吸附等制备工艺处理,得到酰胺类生物碱含量不低于30%的有效部位提取物。

Description

大叶蒟有效部位的制备及其在医疗、保健领域的应用
技术领域
本发明涉及一种有效部位的提取方法及其在抑郁症等情感障碍性精神疾病领域的医疗或保健等方面应用。
背景技术
随着人类社会的飞速发展,各方面竞争也越发激烈,使得抑郁症等情感障碍性疾病成为一种常见病、高发病,且发病率正在快速攀升。调查表明世界十大疾病中,抑郁症的医疗总负担现在已超过心脑血管、恶性肿瘤而跃居首位。高发且难以根治的抑郁症已经形成了巨大的抗抑郁药市场。现在临床中一般采用合成抗抑郁药,但合成抗抑郁药存在着抗抑郁谱窄、副作用大、药价高以及停药后易复发造成“停药综合症”等缺陷。因而国内外在抗抑郁药的研究开发方面越来越注意天然药物的应用,特别是有2000多年历史的中草药。
发明内容
本发明旨在提供未在国内上市的从药用植物大叶蒟Piper laetispicumC.DC.中获得的有效部位提取物及其制备方法,及其在治疗、预防抑郁症等情感障碍性精神疾病等医疗、保健领域的应用。
本发明公开的有效部位提取物制备方法为:
1、取大叶蒟的干燥地下部分打成粗粉。
2、所得粗粉加30-95%乙醇溶液浸渍,水浴回流两次,每次约1小时,过滤。
3、合并滤液,减压浓缩至原滤液体积的10-30%,并加乙醇得澄清不透明乙醇稀溶液。
4、所得溶液以大孔吸附树脂柱色谱法分离、精制,用不同浓度的乙醇溶液洗脱2-4次。
5、收集80-95%乙醇洗脱液,合并、浓缩干燥后得该提取物。
该提取物经HPLC检测,含酰胺类生物碱为30-70%。
本发明公开的有效部位提取物,其主要成分为酰胺类生物碱、β-谷甾醇、甘油酯类及酯类化合物,其中酰胺类生物碱主要化合物为:
(1)5’-甲氧基-3’,4’亚甲基二氧桂皮酸异丁基酰胺;
(2)3’,4’-亚甲基二氧桂皮酸异丁基酰胺;
(3)N-异丁基-9(3’,4’-亚甲基二氧苯基)-2E,8E-壬二烯酰胺;
(4)1-[(2E,4E)-2,4-癸二烯酰]四氢吡咯;
(5)N-异丁基癸-反-2-反-4-二烯酰胺;
(6)1-[(2E)-5-(3,4-亚甲基二氧苯基)]四氢吡咯;
(7)5’-甲氧基-3’,4’-亚甲基二氧桂皮酸四氢吡咯。
用本发明有效部位提取物制成制剂,进行有关的药效实验,其结果如下:药物以0.1% Tween80水溶液配制,阳性对照为百优解(Prozac,礼来苏州制药有限公司,胶囊剂,生产批号203181)空白对照为0.1%0.1% Tween80水溶液。
实验动物:KM种清洁级小白鼠,雄性,20-25克,每组8-10只。
实验方法:强迫性游泳实验(Force,Swimming Test)。Porsol等(1977,1978)提出行为绝望实验可作为抗抑郁药作用的评价模型,Alpermann等(1991)制作了改进型小鼠行为绝望实验。此实验模型将小鼠或大鼠放在一个局限的无法逃出的空间里游泳,可诱导实验动物出现不动状态,这种状态反映了动物的绝望行为,可以被某些有效的抗抑郁药所抑制。具体操作为小鼠随机分组,适应环境至少1天,给药前12小时禁食不禁水。灌胃给受试药物后60分钟,将动物置于直径约10厘米、高约20厘米、水深8-10厘米玻璃圆筒内,水温22-24℃。自第2分钟开始,纪录4分钟内不动累计时间。实验数据采用t检验法统计处理。

C  大孔吸附树脂80%-95%乙醇洗脱部分;
实施例
1.取大叶蒟的干燥地下部分打成粗粉,加60%乙醇溶液浸渍,水浴回流两次,每次约1小时,过滤,合并滤液,减压浓缩至原滤液体积的14%,并加30%乙醇得澄清不透明乙醇稀溶液。随后以大孔吸附树脂柱色谱法分离、精制,先后以40%、55%和80%乙醇溶液洗脱,收集80%乙醇洗脱液,浓缩干燥后得有效部位提取物。经HPLC检测,该提取物中酰胺类生物碱含量为68%。
2.取大叶蒟的干燥地下部分打成粗粉,加85%乙醇溶液浸渍,水浴回流两次,每次约1小时,过滤,合并滤液,减压浓缩至原滤液体积的18%,并加30%乙醇得澄清不透明乙醇稀溶液。随后以大孔吸附树脂柱色谱法分离、精制,先后以25%、45%、75%和95%乙醇溶液洗脱,收集95%乙醇洗脱液,浓缩干燥后经HPLC检测,提取物酰胺类生物碱含量为34%。

Claims (1)

1、大叶蒟有效部位提取物的制备方法,其特征在于包括如下步骤:取大叶蒟的地下部分的药材粗粉加乙醇以回流提取法获得粗提物,进而以大孔吸附树脂法进行有效成分的富集、纯化和精制,获得以酰胺类生物碱为主的有效部位提取物,其中酰胺类生物碱含量30-70%。
