Description
La présente invention se rapporte à des terpénoïdes contenant deux groupes fonctionnels et représentés par la formule générale (I):

dans laquelle n est un nombre entier de 1 à 5, R représente un groupe hydroxyméthyle, un groupe formyle ou un groupe carboxyle, et A représente un groupe 2-tétrahydropyrannyle, un groupe benzyle, un groupe méthoxyméthyle ou un groupe méthoxyéthoxyméthyle. Cette invention concerne également un procédé pour la _ préparation de ces terpénoïdes par oxydation d'un composé représenté par la formule (I) dans laquelle R est un groupe méthyle avec un micro-organisme.
Les demandes de brevet japonais publiées respectivement N05 54(1979)-70430 et 54(1979)-76513 décrivent que certains terpénoïdes de type à structure en chaîne (acyclique) contenant un groupe hydroxyle à-une extrémité et les esters de tels terpénoïdes présentent une activité hypotensive.
Les composés de formule générale (I) représentent des intermédiaires utiles pour la préparation de composés actifs du point de vue pharmaceutique, tels qu'un alcool polyprénylique.
L'alcool polyprénylique a de la 'valeur, d'une part, comme agent hypotensif et, d'autre part, pour la formation de la chaîne latérale du coenzyme Q actif du point de vue pharmaceutique. L'alcool polyprénylique peut être préparé à partir d'un composé de formule (I) dans lequel R est un groupe hydroxyméthyle, et A est un groupe de protection pour le groupe hydroxyle tel qu'un groupe 2-tétrahydropyrannyle, un groupe benzyle, un groupe méthoxyméthyle ou un groupe méthoxyéthoxyméthyle, via la réaction de prolongation de la chaîne carbonnée illustrée ci-dessous:

Dans les équations ci-dessus, ■£ et m sont des nombres entiers et Y représente le groupe de protection pour le groupe hydroxyle, R1 est un groupe alkyle en Cj-Q ou un groupe aryle tel que benzène ou toluène.
En conséquence, un composé de formule générale (I) peut être utilisé, per se, comme un intermédiaire pour la préparation de l'alcool polyprénylique, pour autant que, dans le composé de départ de formule (I), R représente le groupe hydroxyméthyle et A représente un groupe 2-tétrahydropyrannyle, un groupe benzyle, un groupe méthoxyméthyle ou un groupe méthoxyéthoxyméthyle.
Comme dans le cas d'un procédé pour l'oxydation d'un groupe terminal d'un terpénoïde de type structure à chaîne ayant un groupe fonctionnel à l'autre extrémité, il est divulgué, dans la publication de la demande de brevet japonais N° 53(19783-103445, qu'un terpénoïde à structure en chaîne ayant à l'une de ses extrémités un groupe hydroxyle protégé par un groupe de protection est oxydé en utilisant un oxyde de sélénium pour changer l'autre groupe terminal en un groupe formyle, puis est réduit. Ce procédé, toutefois, présente un rendement faible de l'étape d'oxydation. Cette tendance est plus particulièrement marquée dans la réaction d'un composé ayant une plus longue chaîne d'atomes de carbone. En outre, il est à noter que l'étape d'oxydation fournit de façon inhérente un composé organique du sélénium comme sous-produit. La séparation de ce sous-produit ainsi formé du composé désiré est très difficile même par une méthode telle que la Chromatographie sur colonne ou la distillation. Le composé organique du sélénium est connu pour être toxique vis-à-vis des êtres humains. C'est pourquoi le procédé mentionné ci-dessus n'est pas approprié comme procédé pour la préparation d'un intermédiaire pour la synthèse d'un composé actif du point de vue pharmaceutique.
Au vu du problème décrit précédemment, les présents inventeurs ont étudié un procédé utilisant un micro-organisme et ont mis en évidence que l'oxydation par un micro-organisme, en utilisant un micro-organisme appartenant au genre Nocardia, peut être employée pour la préparation du composé désiré, et ainsi à l'élimination du problème précité. Le présent procédé utilisant le micro-organisme Nocardia permet la production en masse du composé désiré par l'agrandissement de la dimension du système de culture. En outre, le réactif n'ayant pas réagi peut être facilement récupéré et réutilisé comme réactif de départ.
Une souche représentative parmi celles appartenant au genre Nocardia et qui est employée avantageusement dans le procédé selon la présente invention est celle dénommée BPM 1613, qui a été déposée au Fermentation Research Institute, Agency of Industriai Science and Technology, à 1-1-3, Higashi, Tsukuba-.Yatabe-machi, Ibaraki-préfecture, Japon, et qui a été enregistrée à cette collection de microorganismes sous la désignation FERM-P N° 1609 (date de dépôt:

18.9.72). Les caractéristiques mycologiques de la souche représentative dénommée BPM 1613 sont données ci-après. La couleur est exprimée selon le Color Standard publié par Nippon Shikisai Ken-kyusho ( Japan Color Research Center), Japon.
A. Forme des cellules
La présente souche montre une couleur caractéristique orange à rose dans presque tous les milieux de culture, ainsi qu'il ressort des caractéristiques de culture suivantes. Une jeune cellule végétative croit sous une forme mycéloïde et une ramification est rarement observée. Dans un système cultivé âgé, Fhyphe est divisée pour former un bacille (0,4-0,6 x 1,8-2,4 mm). Gram positive. Pas de flagelle. Négative sur la souche résistant aux acides selon la méthode de Ziehl-Neelsen. Aucun mycélium aérien n'est observé.
B. Caractéristiques de culture sur différents milieux 1 ) Milieu sucrose/nitrate agar (30e C)

D. Assimilabilitè pour différentes sources de carbone (Milieu Prid-ham-Gottlieb agar, 30e C, 7 d):
L-arabinose (+), D-xylose (+), D-glucose ( + + ), D-fructose ( + + ), sucrose ( + + ), inositol (+), L-rhamnose (—), raffi-nose ( +), D-mannitol (+).
Ci-dessus, ( + + ) signifie croissance modérée, (+) croissance faible, et (—) pas de croissance.
La souche identifiée ci-dessus ayant été cultivée sur le milieu glycérol Keiner Morton selon la méthode de Arai et al. décrite dans le «Journal of General Applied Microbiology», 9, 119 (1963): «The Actionomycetables», The Jena International Symposium on Taxo-nomy, 273 (1968) donne des bandes d'absorption caractéristiques du genre Nocardia sur le spectre IR, c'est-à-dire I : types C & E, II:
type C, III: type C, IV: type D.
En étudiant les caractéristiques décrites précédemment de la souche en référence au «Bergey's Manual of Determinative Bacte-riology», 7e éd., et le «Waksman's The Actinomycetes», vol. 2, la souche est également déterminée comme appartenant au genre Nocardia.
Un procédé pour la préparation du composé I selon la présente invention est caractérisé par le fait qu'on cultive un micro-organisme appartenant au genre Nocardia et présentant une activité oxydante pour un composé de formule générale (II):

dans laquelle A et n sont comme défini précédemment, dans un milieu de culture contenant un composé de formule générale (II) comme source de carbone, puis par le fait qu'on récupère le composé I ainsi obtenu.
Il n'y a pas de limitation spécifique en ce qui concerne le microorganisme du genre Nocardia utilisé dans le procédé selon l'invention, pour autant qu'il présente une activité oxydante pour un composé de formule générale (II). Des exemples de tels micro-orga-nismes comprennent la souche BPM 1613, FERM-P N° 1609, telle que spécifiée précédemment.
Des détails des techniques de culture seront maintenant mentionnés comme suit:
En plus du composé de formule générale (II) incorporé comme source de carbone, des sources d'autres éléments nutritifs pour la culture peuvent être choisis parmi les sources traditionnelles.
Comme source d'azote, on peut mentionner les nitrates tels que le nitrate de potassium, le nitrate de sodium et le nitrate d'ammonium, des sels d'ammonium et le phosphate d'ammonium, l'ammoniaque et l'urée. D'autres sels inorganiques tels que le phosphate de potassium, le phosphate de sodium, le sulfate de magnésium, le sulfate ferrique et le sulfate de manganèse, ainsi que d'autres sources nutritives organiques telles que des vitamines, des amino-acides et des extraits de levures, des liqueurs de maïs macéré et des extraits de malt contenant ces éléments nutritifs peuvent être en outre incorporés dans le milieu, si nécessaire. Le milieu est de préférence ajusté de manière à être alcalin, par exemple dans un domaine de pH de 7-10. La culture est effectuée de façon appropriée entre 20 et 40° C, pendant 3 à 5 d et dans des conditions aérobies, par exemple en effectuant la culture sous aération et agitation.
Après la fin de la culture, le milieu cultivé est extrait "au moyen d'un solvant organique pour récupérer le composé de formule générale (I). Comme solvant d'extraction, on peut employer l'éther éthy-lique, le benzène, le chloroforme, etc. Ce composé de formule générale (I) peut être séparé et purifié sur une colonne de Silicagel.
Les réactifs n'ayant pas réagi peuvent être récupérés par la technique d'extraction mentionnée précédemment et par Chromatographie sur colonne, avec un rendement approximatif de 80 à 90%, et employés à nouveau comme réactifs pour un autre cycle.
Le composé ayant un groupe hydroxyméthyle, formyle ou carboxyle à son extrémité peut être obtenu selon l'importance de l'oxydation microbiologique. En outre, la configuration du produit obtenu peut être variée en modifiant les conditions de culture, le groupe de protection du groupe hydroxyle du composé de départ.
La présente invention sera en outre décrite en référence aux exemples illustratifs suivants:

1) Ether tétrahydropyrannilique (A = tétrahydropyrannyle)
Dans du chlorure de méthylène ont été dissous 15 g de 3,7,11,15-tétramêthyl-2,6,10,14-hexadécatétraène-l-ol et 1,5 g d'acide p-toluènesulfonique, et 8,7 g de 2,3-dihydropyranne ont été ajoutés goutte à goutte à cette solution sous agitation, à une température de 0-5° C et sur une période de 30 min. La solution obtenue a été agitée à 0-5 C pendant 30 min, puis lavée avec une solution de carbonate de sodium aqueuse dans un entonnoir à séparation. La solution a été concentrée après le lavage et la solution concentrée a été purifiée sur une colonne de Silicagel pour donner 13 g de l-(2-tétrahydro-pyrannyl)oxy-3,7,11,15-tétraméthyl-2,6,10,14-hexadécatétraène,
avec un rendement de 68%.
D'autres éthers tétrahydropyrannyliques ont été préparés de la même manière que celle décrite ci-dessus.
2) Ether benzylique (A = benzyle)
5 A 100 ml de chlorure de benzène ont été ajoutés 15 g de 3,7,ll,15-tétraméthyl-2,6,10,14-hexadécatétraène-l-ol et 6,0 g d'hydroxyde de potassium finement divisé, et le mélange obtenu a été chauffé au reflux sous agitation pendant 2 h. Le mélange obtenu a été refroidi, puis 11 d'hexane a été ajouté à celui-ci. La solution i° dans l'hexane a été ensuite lavée avec de l'eau et concentrée. Le concentrât a été purifié sur une colonne de Silicagel afin de donner 12 g de l-benzyloxy-3,7,11,15-tétraméthyl-2,6,10,14-hexadécatétraène, avec un rendement de 61 %.
D'autres éthers benzyliques ont été préparés de la même manière 15 que celle décrite ci-dessus.
Les rendements et données spectrales RMN des éthers tétra-hydropyrannylique et benzylique obtenus sont résumés dans le tableau I.

2. Acide 8-(2-tétrahydropyrannyl)oxy-2,6-diméthyl-2,6-octa-diénoïque selon la revendication 1.
3. Acide 12-(2-tétrahydropyrannyl)oxy-2,6,10-triméthyl-2,6,10-dodécatriénoïque selon la revendication 1.

10. Procédé pour la préparation d'un composé de formule (I) selon la revendication 1, caractérisé par le fait qu'on cultive un mi-cro-organisme appartenant au genre Nocardia et présentant une activité oxydante pour un composé de formule (II):
