Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die reduktive Entschwefelung der Verbindung der Formel III unter Verwendung eines Alkalimetallborhydrids als Reduktionsmittel in Gegenwart eines Metallhalogenides, das aus Nickelchlorid, Ferrochlorid, Stannochlorid, Zinkchlorid und Kobaltchlorid gewählt ist, in einem Lösungsmittel ausführt. 3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass man die reduktive Entschwefelung der Verbindung der Formel III bei einer Temperatur zwischen 20 und 50"C ausführt. 4. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Alkylierung der Verbindung der Formel III oder der Verbindung der Formel V unter Verwendung eines Alkylierungsmittels, das aus niederen Alkylhalogeniden, niederen Alkyloxoniumfluoroboraten und Diniederalkylsulfaten gewählt ist, gegebenenfalls in Gegenwart eines Säureakzeptors, in einem Lösungsmittel ausführt. 5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, dass man die Alkylierung der Verbindung der Formel III oder der Verbindung der Formel V bei einer Temperatur zwischen 0 und 30"C ausführt. 6. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die reduktive Entschwefelung der Verbindung der Formel IV oder der Verbindung der Formel VI unter Verwendung eines Reduktionsmittels, das aus Alkalimetallborhydriden und Lithiumaluminiumhydrid gewählt ist, in einem Lösungsmittel ausführt. 7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, dass man die reduktive Entschwefelung der Verbindung der Formel IV oder der Verbindung der Formel VI bei einer Temperatur zwischen 0 und 30"C ausführt. Die vorliegende Erfindung bezieht sich auf ein neues Verfahren zur Herstellung von Benzylalkoholderivaten der Formel: EMI2.1 worin Rl für Wasserstoff oder Aralkyl steht, oder den pharmazeutisch unbedenklichen Säureadditionssalzen davon. Es ist bekannt, dass die Benzylalkoholderivate der Formel I oder deren pharmazeutisch unbedenkliche Säureadditionssalze als Cardiotonicum oder Antidiabeticum brauchbar sind (US-PS Nr.4032575). Aus der US-PS 4032575 ist bekannt, dass die Verbindung der Formel I hergestellt werden kann, indem man ein a-Halogen-monobenzyloxyacetophenon (A) mit 3,4-Dimethoxyphenyläthylamin (B) zu einem a-(3,4-Dimethoxyphenyl äthylamino)-monobenzyloxyacetophenon (C) kondensiert, gegebenenfalls die Verbindung (C) zu einem a-(3,4-Dimethoxyphenyläthylaminomethyl)-monobenzyloxybenzylalkohol (D) reduziert und dann die Verbindung (C) bzw. (D) der katalytischen Hydrierung unterwirft. Als Ergebnis weiterer Untersuchungen wurde nun aber ein neues Verfahren zur Herstellung von Benzylalkoholderivaten der Formel 1 gefunden. Erfindungsgemäss werden die Benzylalkoholderivate der Formel I hergestellt, indem man (a) ein Acetophenonderivat der Formel: EMI2.2 worin R2 für Wasserstoff, Alkanoyl oder Aralkyl steht, mit einem Thionylhalogenid umsetzt, (b) das resultierende Produkt mit 3,4-Dimethoxyphenyl äthylamin zu einem Thioacetamidderivat der Formel: EMI2.3 worin R2 die obige Bedeutung hat, umsetzt und (c-l) die Verbindung der Formel III der reduktiven Entschwefelung unterwirft oder (c-2) die Verbindung der Formel III zu einem Acetophenonderivat der Formel: EMI3.1 worin R3 für Alkyl steht und R2 die obigen Bedeutungen hat, alkyliert und die Verbindung der Formel IV der reduktiven Entschwefelung unterwirft oder (c-3) die Verbindung der Formel III zu einem Benzylalkoholderivat der Formel: EMI3.2 worin R' die obigen Bedeutungen hat, reduziert, die Verbindung der Formel V zu einem Benzylalkoholderivat der Formel: EMI3.3 worin Rl und R3 die obigen Bedeutungen haben, alkyliert und die Verbindung der Formel VI der reduktiven Entschwefelung unterwirft. Bevorzugte Bedeutungen von R2 in den Acetophenonderivaten der Formel II, die als Ausgangsmaterial im erfindungsgemässen Verfahren verwendet werden, sind Wasserstoff, Niederalkanoyl, wie Acetyl oder Propionyl, und Phenylniederalkyl, wie Benzyl. Andererseits verwendet man als oben genanntes Thionylhalogenid vorzugsweise Thionylchlorid. Bevorzugte Bedeutungen von R3 sind Niederalkyl, wie Methyl oder Äthyl. Die Umsetzung des Acetophenonderivates der Formel II mit dem Thionylhalogenid kann ausgeführt werden, indem man die Verbindung der Formel II in dem Thionylhalogenid auflöst und die Lösung rührt. Diese Reaktion wird vorzugsweise in Gegenwart einer katalytischen Menge einer organischen Base ausgeführt. Geeignete Beispiele derartiger organischer Basen sind Pyridin, Triäthylamin, Dimethylanilin und dergleichen. Eine geeignete Menge des zu verwendenden Thionylhalogenides beträgt ca. 3 bis 10 Mol pro Mol des Acetophenonderivates der Formel II. Die Reaktion wird vorzugsweise bei einer Temperatur zwischen 10 und 50"C ausgeführt. Das so erhaltene Reaktionsprodukt, von dem man annimmt, dass es ein Gemisch der folgenden beiden Verbindungen ist: EMI3.4 worin X für Halogen steht und R2 die obigen Bedeutungen hat, wird vorzugsweise ohne Isolierung aus der Reaktionslösung und/oder Reinigung in der nächsten Stufe verwendet. Die Reaktion des oben erwähnten Produktes mit 3,4-Dimethoxyphenyläthylamin kann in Gegenwart oder Abwesenheit eines Säureakzeptors in einem geeigneten Lösungsmittel ausgeführt werden. Geeignete Beispiele von Säureakzeptoren sind organische Basen, wie Pyridin, Triäthylamin, Dimethylanilin oder N-Methylpyrrolidin, und anorganische Basen, wie Kaliumcarbonat, Natriumcarbonat, Natriumbicarbonat oder Natriumhydroxyd. Als Lösungsmittel eignen sich organische Lösungsmittel, z.B. Benzol, Chloroform, Methylenchlorid, Äthylacetat oder Dimethylformamid oder ein Gemisch eines dieser organischen Lösungsmittel mit Wasser. Vorzugsweise wird die Reaktion bei einer Temperatur zwischen 0 und 30"C ausgeführt. Die Benzylalkoholderivate der Formel I können aus den so erhaltenen Thioacetamidderivaten der Formel III nach einem der oben erwähnten Verfahren (c-l), (c-2) und (c-3) hergestellt werden. Nach Verfahren (c-l) wird das Benzylalkoholderivat der Formel I hergestellt, indem man die Verbindung der Formel III der reduktiven Entschwefelung unterwirft. Die reduktive Entschwefelung der Verbindung der Formel III kann ausge führt werden, indem man sie in Gegenwart eines Metallhalogenides in einem geeigneten Lösungsmittel mit einem Reduktionsmittel behandelt. Geeignete Beispiele von Metallhalogeniden sind Nickelchlorid, Ferrochlorid, Stannochlorid, Zinkchlorid, Kobaltchlorid und dergleichen. Geeignete Beispiele von Reduktionsmitteln sind Alkalimetallborhydride, wie Natriumborhydrid, Lithiumborhydrid und dergleichen. Alkanole, z.B. Methanol, Äthanol oder Propanol, Tetrahydrofuran, Dioxan, Dimethoxyäthan und dergleichen eignen sich als Lösungsmittel. Vorzugsweise wird die Reaktion bei einer Temperatur zwischen 20 und 50"C ausgeführt. Während dieses Reaktionsverfahrens finden die Entschwefelung der Verbindung der Formel III und die Reduktion der Oxogruppe in der 2-Stellung derselben gleichzeitig statt. Wenn eine Verbindung der Formel III, worin R2 für Alkanoyl steht, in dieser Reaktion verwendet wird, wird die Alkanoylgruppe gleichzeitig mit den Entschwefelungs- und Reduktionsreaktionen weiter hydrolysiert zu dem Benzylalkoholderivat der Formel I, worin R' für Wasserstoff steht. Nach dem Verfahren (c-2) werden die Benzylalkoholderivate der Formel I hergestellt, indem man die Verbindung der Formel III alkyliert und die resultierende Verbindung der Formel IV der reduktiven Entschwefelung unterwirft. Die Alkylierung der Verbindung der Formel III kann ausgeführt werden, indem man sie in Gegenwart oder Abwesenheit eines Säureakzeptors in einem geeigneten Lösungsmittel mit einem Alkylierungsmittel behandelt. Geeignete Beispiele von Alkylierungsmitteln sind Niederalkylhalogenide, wie Methyljodid oder Äthylbromid, Triniederalkyloxoniumfluoroborate, wie Trimethyloxoniumfluoroborat oder Triäthyloxoniumfluoroborat und Diniederalkylsulfate, wie Dimethylsulfat oder Diäthylsulfat. Geeignete Beispiele von Säureakzeptoren sind anorganische Basen, wie Kaliumcarbonat, Natriumcarbonat oder Natriumbicarbonat und organische Basen, wie Pyridin, Triäthylamin oder 1 ,5-Diazabicyclo-[5,4,0]-5-undecen. Methylenchlorid, Aceton, Chloroform, Methanol, Äthanol, Äther, Dimethylformamid, Tetrahydrofuran und dergleichen eignen sich als Lösungsmittel. Vorzugsweise wird die Reaktion bei einer Temperatur zwischen 0 und 30"C ausgeführt. Durch diese Reaktion wird das Acetophenonderivat der Formel IV hergestellt. Die so erhaltene Verbindung der Formel IV kann vorzugsweise ohne Isolierung aus der Reaktionslösung und/oder Reinigung in der nächsten Stufe verwendet werden. Die reduktive Entschwefelung der Verbindung der Formel IV kann ausgeführt werden, indem man sie in einem geeigneten Lösungsmittel mit einem Reduktionsmittel behandelt. Geeignete Beispiele von Reduktionsmitteln sind Alkalimetallborhydride, z.B. Natriumborhydrid oder Lithiumborhydrid, Lythiumaluminiumhydrid und dergleichen. Alkanole, z.B. Methanol, Äthanol oder Propanol, Tetrahydrofuran, Dioxan und Dimethoxyäthan eigenen sich als Lösungsmittel. Vorzugsweise wird die Reaktion bei einer Temperatur zwischen 0 und 30"C ausgeführt. In dieser Reaktionsstufe wird die Alkylthiogruppe der Verbindung der Formel IV gleichzeitig mit der Reduktion der Oxogruppe derselben entfernt. Wenn in dieser Stufe eine Verbindung der Formel IV, worin R2 für Alkanoyl steht, verwendet wird, wird die Alkanoylgruppe ebenfalls gleichzeitig mit den oben genannten Reaktionen hydrolysiert. Die Benzylalkoholderivate der Formel I können alternativ auch nach dem Verfahren (c-3) hergestellt werden, d.h. indem man die Verbindung der Formel III zu einem Benzylalkoholderivat der Formel V reduziert, die Verbindung der Formel V zu einem Benzylalkoholderivat der Formel VI alkyliert und die Verbindung der Formel VI der reduktiven Entschwefelung unterwirft. Die Reduktion der Verbindung der Formel III kann ausgeführt werden, indem man sie in einem geeigneten Lösungsmittel mit einem Reduktionsmittel behandelt. Geeignete Beispiele von Reduktionsmitteln sind Alkalimetallborhydride, wie Natriumborhydrid, Lithiumborhydrid und dergleichen. Alkanole, z.B. Methanol, Äthanol oder Propanol, Tetrahydrofuran und Dioxan eignen sich als Lösungsmittel. Vorzugsweise wird die Reaktion bei einer Temperatur zwischen 0 und 30"C ausgeführt. Wenn eine Verbindung der Formel III, worin R2 für Alkanoyl steht, in dieser Reaktionsstufe verwendet wird, findet die Reduktion der Oxogruppe der Verbindung der Formel III gleichzeitig mit der Hydrolyse der Alkanoylgruppe statt. Die anschliessende Alkylierung der Verbindung der Formel V und reduktive Entschwefelung der Verbindung der Formel VI können unter den gleichen Bedingungen ausgeführt werden, wie sie bei der Alkylierung der Verbindung der Formel III bzw. der reduktiven Entschwefelung der Verbindung der Formel IV im Verfahren (c-2) angewandt werden. Die erfindungsgemäss hergestellten Benzylalkoholderivate der Formel I können erforderlichenfalls in ihre pharmazeutisch unbedenklichen Säureadditionssalze übergeführt werden. Geeignete Beispiele solcher Salze sind anorganische Säureadditionssalze, wie Hydrochloride, Hydrobromide, Perchlorate, Nitrate, Sulfate oder Phosphate und organische Säureadditionssalze, wie Acetate, Propionate, Glycolate, Lactate, Ascorbate, Maleate, Fumarate, Malonate, Succinate, Oxalate, Citrate, Methansulfonate, Benzolsulfonate, Aminobenzoate, Sulfaminate, Asparaginate, Glutamate oder Nicotinate. Das oben beschriebene erfindungsgemässe Verfahren ermöglicht es, die Benzylalkoholderivate der Formel I ohne unerwünschte Nebenreaktionen in einer hohen Ausbeute herzustellen, und ist vorteilhaft für die Herstellung der Benzylalkoholderivate der Formel I in industriellem Massstab. Da das Zwischenprodukt der Formel III im erfindungsgemässen Verfahren ziemlich beständig ist und sich leicht in herkömmlicher Weise isolieren und reinigen lässt, hat das erfindungsgemässe Verfahren auch den Vorteil, dass die Benzylalkoholderivate der Formel I in hoher Reinheit und ohne Verunreinigung mit unerwünschten Nebenprodukten erhalten werden. Die Ausdrücke Niederalkyl und Niederalkanoyl in der Beschreibung und den Ansprüchen bedeuten vorzugsweise unverzweigte oder verzweigte Alkyl- bzw. Alkanoylgruppen mit 1 bis 4 Kohlenstoffatomen. Die folgenden Beispiele erläutern praktische und zur Zeit bevorzugte Ausführungsformen der Erfindung. Beispiel (1)4,97 g 4-Benzyloxyacetophenon werden in 15 ml Thionylchlorid gelöst, worauf 0,02 ml Pyridin zugesetzt werden. Das Gemisch wird 5 Stunden lang bei Raumtemperatur gerührt. Das Gemisch wird unter vermindertem Druck eingeengt, um das überschüssige Thionylchlorid zu entfernen. Der Rückstand wird in 30 ml Benzol gelöst. Die Lösung wird zu einem Gemisch aus 4,78 g 3,4-Dimethoxyphenyläthylamin, 90 ml einer 1 0%gen wässrigen Kaliumcarbonatlösung und 50 ml Benzol gegeben, worauf das Gemisch 4 Stunden lang bei Raumtemperatur gerührt wird. Die Benzollösung wird aus dem Reaktionsgemisch abgetrennt und die wässrige Schicht mit Äthylacetat extrahiert. Die Benzollösung und der Äthylacetatextrakt werden vereinigt und nacheinander mit Wasser, 1 0%Der Salzsäure und Wasser gewaschen. Die organische Schicht wird getrocknet und unter vermindertem Druck eingedampft. Der Rückstand wird aus Äthanol umkristallisiert. 7,67 g 2-(4-Benzyloxyphenyl)-2-oxo-N-(3 ,4-dimeth- oxyphenyläthyl)-thioacetamid werden als blassgelbe Nadeln erhalten. Ausbeute: 80,1%. Schmelzpunkt 132 bis 133"C. (2) 500 mg 2-(4-Benzyloxyphenyl)-2-oxo-N-(3 ,4-dimeth- oxyphenyläthyl)-thioacetamid und 610 mg Nickelchloridhexahydrat werden in 30 ml Methanol gelöst, worauf bei Raumtemperatur 700 mg Natriumborhydrid zugegeben werden. Das Gemisch wird bei der gleichen Temperatur 1,5 Stunden lang gerührt. Unlösliche Substanzen werden abfiltriert, und das Filtrat wird unter vermindertem Druck eingeengt. Der Rückstand wird in Chloroform gelöst, und die Lösung wird mit Wasser gewaschen, getrocknet und dann unter vermindertem Druck eingedampft. Der Rückstand wird mit äthanolischem Chlorwasserstoff behandelt, und der kristalline Niederschlag wird durch Filtration isoliert. 360 mg a-(3,4-Dimethoxyphenyläthylaminomethyl)-4-benzyloxy- benzylalkohol-hydrochlorid werden erhalten. Ausbeute: 70%. Schmelzpunkt 168 bis 170"C. Beispiel 2 2,0 g 2-(4-Benzyloxyphenyl)-2-oxo-N-(3,4-dimethoxyphenyläthyl)-thioacetamid werden in 40 ml Methylenchlorid gelöst, worauf 1,05 g Triäthyloxoniumfluoroborat hinzugegeben werden. Nachdem das Gemisch 1,5 Stunden lang bei Raumtemperatur gerührt worden ist, wird es unter vermindertem Druck eingeengt. Der Rückstand wird in 30 ml Äthanol gelöst, worauf 2,1 g Natriumborhydrid zugesetzt werden. Das Gemisch wird 1 Stunde lang bei Raumtempe- ratur gerührt. Wasser wird in das Reaktionsgemisch gegossen, worauf das Gemisch unter vermindertem Druck eingeengt wird. Die kristallinen Niederschläge werden durch Filtration isoliert, worauf die Kristalle mit Wasser gewaschen und dann aus Isopropanol umkristallisiert werden. 1,45 g a-(3,4-Dimethoxyphenyläthylaminomethyl)-4-benzyloxybenzylalkohol (freie Base) werden als farblose Nadeln erhalten. Ausbeute: 77,5%. Schmelzpunkt 114,5 bis 116"C. Beispiel3 2,2 g 2-(4-Benzyloxyphenyl)-2-oxo-N-(3,4-dimethoxyphe- nyläthyl)-thioacetamid werden in 30 ml Aceton gelöst, worauf 850 mg Kaliumcarbonat und 1,0 g Methyljodid zugesetzt werden. Das Gemisch wird über Nacht bei Raumtemperatur gerührt. Die unlöslichen Substanzen werden abfiltriert, worauf das Filtrat unter vermindertem Druck eingeengt wird. Der Rückstand wird in 50 ml Methanol gelöst, worauf 300 mg Natriumborhydrid zugesetzt werden. Das Gemisch wird 30 Minuten lang bei Raumtemperatur gerührt. Dann wird das Reaktionsgemisch in der gleichen Weise, wie in Beispiel 2 beschrieben, behandelt. 1,65 g a-(3,4-Dimethoxyphenyläthylaminomethyl)-4-benzyloxybenzylalkohol (freie Base) werden als farblose Nadeln erhalten. Ausbeute: 80%. Die physikalisch-chemischen Eigenschaften dieses Produktes sind identisch mit denjenigen der in Beispiel 2 erhaltenen Probe. Beispiel4 (1)8,0 g 2-(4-Benzyloxyphenyl)-2-oxo-N-(3 ,4-dimethoxy- phenyläthyl)-thioacetamid werden in 150 ml Methanol gelöst, worauf 500 mg Natriumborhydrid zugesetzt werden. Das Gemisch wird 1 Stunde lang bei Raumtemperatur gerührt. Das Gemisch wird unter vermindertem Druck eingeengt. Der Rückstand wird in Chloroform gelöst und die Lösung mit Wasser gewaschen, getrocknet und unter vermindertem Druck eingedampft. Der Rückstand wird aus Isopropanol umkristallisiert. 7,5 g 2-(4-Benzyloxyphenyl)-2 hydroxy-N-(3 ,4-dimethoxyphenyläthyl)-thioacetamid werden als farblose Nadeln erhalten. Ausbeute: 93%. Schmelzpunkt 82 bis 84"C. (2) 1,0 g 2-(4-Benzyloxyphenyl)-2-hydroxy-N-(3,4-dimethoxyphenyläthyl)-thioacetamid werden in 20 ml Methylenchlorid gelöst, worauf 460 mg Triäthyloxoniumfluoroborat zugesetzt werden. Nachdem das Gemisch 30 Minuten lang bei Raumtemperatur gerührt worden ist, wird es unter vermindertem Druck eingeengt. Der Rückstand wird in einem Gemisch aus 20 ml Methanol und 20 ml Chloroform gelöst, worauf 440 mg Natriumborhydrid zugesetzt werden. Das Gemisch wird 1 Stunde lang bei Raumtemperatur gerührt. Dann wird das Reaktionsgemisch in gleicher Weise, wie in Beispiel 2 beschrieben, behandelt. 780 mg a-(3,4-Dimethoxyphenyläthylaminomethyl)-4-benzyloxybenzylalkohol (freie Base) werden als farblose Nadeln erhalten. Ausbeute 84%. Die physikalisch-chemischen Eigenschaften dieses Produktes sind identisch mit denjenigen der in Beispiel 2 erhaltenen Probe. Beispiel 5 (1) 5,34 g 4-Acetoxyacetophenon, 20 ml Thionylchlorid, 0,03 ml Pyridin und 6,51 g 3,4-Dimethoxyphenyläthylamin werden in gleicher Weise behandelt, wie in Beispiel 1-(1) beschrieben. 8,5 g 2-(4-Acetoxyphenyl)-2-oxo-N-(3,4-dimethoxyphenyläthyl)-thioacetamid werden als blassgelbes Öl erhalten. Ausbeute: 73%. IR, Vom,, (cm-'): 1755, 1665 Massenspektrum, m/e: 387 (M+) NMR (CDC13), 6:2,30(s, 3H, CH3COO 3,00 (t, 2H, J=7,5 Hzi-CH2-CH2-), 3,84 (s, 6H,-OCHl x 2) (2) 1,94 g 2-(4-Acetoxyphenyl)-2-oxo-N-(3 ,4-dimethoxy- phenyläthyl)-thioacetamid, 1,14 g Triäthyloxoniumfluoroborat und 1,0 g Natriumborhydrid werden in der gleichen Weise, wie in Beispiel 2 beschrieben, behandelt. 1,47 g a-(3,4 Dimethoxyphenyläthylaminomethyl)-4-hydroxybenzylalkohol werden als farblose Prismen erhalten. Ausbeute: 92%. Schmelzpunkt 151 bis 153"C. Beispiel 6 (1) 4,08 g 4-Hydroxyacetophenon, 12 ml Thionylchlorid, 0,06 ml Pyridin und 6,51 g 3,4-Dimethoxyphenyläthylamin werden in gleicher Weise, wie in Beispiel 1-(1) beschrieben, behandelt. 3,1 g 2-(4-Hydroxyphenyl)-2-oxo-N-(3,4-dimethoxyphenyläthyl)-thioacetamid werden als blassgelbe Nadeln erhalten. Ausbeute: 30%. Schmelzpunkt 179 bis 1800C (umkristallisiert aus Äthanol). (2) 1,0 g 2-(4-Hydroxyphenyl)-2-oxo-N-,4-dimethoxy- phenyläthyl)-thioacetamid, 1,37 g Nickelchloridhexahydrat und 1,1 g Natriumborhydrid werden in gleicher Weise, wie in Beispiel 1-(2) beschrieben, behandelt. 230 ml a-(3,4-Dimeth oxyphenyläthylaminomethyl)-Shydroxybenzylalkohol werden als farblose Prismen erhalten. Ausbeute: 25%. Die physikalisch-chemischen Eigenschaften dieses Produktes sind identisch mit denjenigen der in Beispiel 5-(2) erhaltenen Probe. Beispiel 7 1,58 g 2-(4-Hydroxyphenyl)-2-oxo-N-(3 P-dimethoxyphe- nyläthyl)-thioacetamid, 1,13 g Triäthyloxoniumfluoroborat und 870 mg Natriumborhydrid werden in gleicher Weise, wie in Beispiel 2 beschrieben, behandelt. 940 mg a-(3,4-Dimetho- xyphenyläthylaminomethyl)-4-hydroxybenzylalkohol werden als farblose Prismen erhalten. Ausbeute: 65%. Die physikalisch-chemischen Eigenschaften dieses Produktes sind identisch mit denjenigen der in Beispiel 5-(2) erhaltenen Probe. Beispiel8 (1)6,78 g 2-Benzyloxyacetophenon, 21 ml Thionylchlorid, 0,05 ml Pyridin und 6,54 g 3,4-Dimethoxyphenyläthylamin werden in gleicher Weise, wie in Beispiel 1-(1) beschrieben, behandelt. 10,3 g 2-(2-Benzyloxyphenyl)-2-oxo-N-(3,4- dimethoxyphenyläthyl)-thioacetamid werden als blassgelbe Prismen erhalten. Ausbeute: 79%. Schmelzpuntk 112 bis 114"C. (2) 500 mg 2-(2-Benzyloxyphenyl)-2-oxo-N-(3,4-dimetho xyphenyläthyl)-thioacetamid, 900 mg Nickelchloridhexahydrat und 700 ml Natriumborhydrid werden in der gleichen Weise, wie in Beispiel 1-(2) beschrieben, behandelt. 165 mg a-(3,4-Dimethoxyphenyläthylaminomethyl)-2-benzyloxy- benzylalkohol-hydrochlorid werden als farblose Nadeln erhalten. Ausbeut: 32%. Schmelzpunkt 159 bis 160"C. Beispiel 9 2,0 g 2-(2-Benzyloxyphenyl)-2-oxo-N-(3,4-dimethoxyphe nyläthyl)-thioacetamid, 1,13 g Triäthyloxoniumfluoroborat und 870 mg Natriumborhydrid werden in der gleichen Weise, wie in Beispiel 2 beschrieben, behandelt. 1,65 g a-(3,4-Di methoxyphenyläthylaminomethyl)-2-benzyloxybenzylalkohol (freie Base) werden als farblose Nadeln erhalten. Ausbeute: 82%. Schmelzpunkt 95 bis 97"C (umkristallisiert aus Isopropanol). Beispiel 10 2,2 g 2-(2-Benzyloxyphenyl)-2-oxo-N-(3,4-dimethoxyphe nyläthyl)-thioacetamid, 850 mg Kaliumcarbonat, 1,0 g Methyljodid und 300 mg Natriumborhydrid werden in gleicher Weise, wie in Beispiel 3 beschrieben, behandelt. 1,1 g a-(3,4- Dimethoxyphenyläthylaminomethyl)-2-benzyloxybenzylal- kohol (freie Base) werden als farblose Nadeln erhalten. Ausbeute: 53%. Die physikalisch-chemischen Eigenschaften dieses Produktes sind identisch mit denjenigen der in Beispiel 9 erhaltenen Probe. Beispiel 11 (1)2,73 g 2-Hydroxyacetophenon, 8 ml Thionylchlorid, 0,03 ml Pyridin und 2,53 g 3,4-Dimethoxyphenyläthylamin werden in der gleichen Weise, wie in Beispiel 1-(1) beschrieben, behandelt. 4,0 g 2-(2-Hydroxyphenyl)-2-oxo-N (3,4-dimethoxyphenyläthyl)-thioacetamid werden als blassrotbraune Nadeln erhalten. Ausbeute 58%. Schmelzpunkt 166 bis 168"C (umkristallisiert aus Äthanol). (2) 1,58 g 2-(2-Hydroxyphenyl)-2-oxo-N-(3,4-dimethoxy phenyläthyl)-thioacetamid, 1,13 g Thriäthyloxoniumfluoroborat und 1,4 g Natriumborhydrid werden in der gleichen Weise, wie in Beispiel 2 beschrieben, behandelt. 850 mg a-(3,4-Dimethoxyphenyläthylaminomethyl)-2-hydroxyben- zylalkohol werden als farbloses Öl erhalten. Ausbeute: 58%. Oxalat dieses Produktes: Schmelzpunkt 175 bis 176"C. Beispiel 12 (1) 1,78 g 2-Acetoxyacetophenon, 6 ml Thionylchlorid, 0,02 ml Pyridin und 1,27 g 3,4-Dimethoxyphenyläthylamin werden in der gleichen Weise, wie in Beispiel 1-(1) beschrieben, behandelt. 1,37 g 2-(2-Acetoxyphenyl)-2-oxo-N (3,4-dimethoxyphenyläthyl)-thioacetamid werden als blassgelbes Öl erhalten. Ausbeute: 35%. IR, v maHCl3 (cm- ): 1760,1670 Massenspektrum, m/e: 387 (M+) NMR (CDCl3), 6:2,20(s, 3H, CH3COO-), 2,94 (t,2H, J = 7,5 Hz, -CH2-CH2-), 3,85 (s, 6H,-OCH3 x 2) (2) 2-(2-Acetoxyphenyl)-2-oxo-N-(3,4-dimethoxyphenyl- äthyl)-thioacetamid, Triäthyloxoniumfluoroborat und Natriumborhydrid werden in der gleichen Weise, wie in Beispiel 2 beschrieben, behandelt. a-(3,4-Dimethoxyphenyläthylaminomethyl)-2-hydroxybenzylalkohol wird erhalten. Die physikalisch-chemischen Eigenschaften dieses Produktes sind identisch mit denjenigen der in Beispiel 11-(2) erhaltenen Probe.
Claims (7)
- PATENTANSPRÜCHE 1. Verfahren zur Herstellung von Benzylalkoholderivaten der Formel: EMI1.1 worin R1 für Wasserstoff oder Aralkyl steht, oder pharmazeutisch unbedenklichen Säureadditionssalzen davon, dadurch gekennzeichnet, dass man (a) ein Acetophenonderivat der Formel: EMI1.2 worin R2 für Wasserstoff, Alkanoyl oder Aralkyl steht, mit einem Thionylhalogenid umsetzt, (b) das resultierende Produkt mit 3,4-Dimethoxy-phenyläthylamin zu einem Thioacetamidderivat der Formel: EMI1.3 worin R2 die obige Bedeutung hat, umsetzt und (c)-i) die Verbindung der Formel III der reduktiven Entschwefelung unterwirft oder (c)-ii) die Verbindung der Formel III zu einem Acetophenonderivat der Formel:: EMI1.4 worin R3 für Alkyl steht und R2 die obigen Bedeutungen hat, alkyliert und die Verbindung der Formel IV der reduktiven Entschwefelung unterwirft oder (c)-iii) die Verbindung der Formel III zu einem Benzylalkoholderivat der Formel: EMI1.5 worin R1 die obigen Bedeutungen hat, reduziert, die Verbindung der Formel V zu einem Benzylalkoholderivat der Formel: EMI1.6 worin R' und R3 die obigen Bedeutungen haben, alkyliert und die Verbindung der Formel VI der reduktiven Entschwefelung unterwirft und dann (d) erforderlichenfalls das so erhaltene Benzylalkoholderivat der Formel I weiter in ein pharmazeutisch unbedenkliches Säureadditionssalz davon überführt.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die reduktive Entschwefelung der Verbindung der Formel III unter Verwendung eines Alkalimetallborhydrids als Reduktionsmittel in Gegenwart eines Metallhalogenides, das aus Nickelchlorid, Ferrochlorid, Stannochlorid, Zinkchlorid und Kobaltchlorid gewählt ist, in einem Lösungsmittel ausführt.
- 3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, dass man die reduktive Entschwefelung der Verbindung der Formel III bei einer Temperatur zwischen 20 und 50"C ausführt.
- 4. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die Alkylierung der Verbindung der Formel III oder der Verbindung der Formel V unter Verwendung eines Alkylierungsmittels, das aus niederen Alkylhalogeniden, niederen Alkyloxoniumfluoroboraten und Diniederalkylsulfaten gewählt ist, gegebenenfalls in Gegenwart eines Säureakzeptors, in einem Lösungsmittel ausführt.
- 5. Verfahren nach Anspruch 4, dadurch gekennzeichnet, dass man die Alkylierung der Verbindung der Formel III oder der Verbindung der Formel V bei einer Temperatur zwischen 0 und 30"C ausführt.
- 6. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass man die reduktive Entschwefelung der Verbindung der Formel IV oder der Verbindung der Formel VI unter Verwendung eines Reduktionsmittels, das aus Alkalimetallborhydriden und Lithiumaluminiumhydrid gewählt ist, in einem Lösungsmittel ausführt.
- 7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, dass man die reduktive Entschwefelung der Verbindung der Formel IV oder der Verbindung der Formel VI bei einer Temperatur zwischen 0 und 30"C ausführt.Die vorliegende Erfindung bezieht sich auf ein neues Verfahren zur Herstellung von Benzylalkoholderivaten der Formel: EMI2.1 worin Rl für Wasserstoff oder Aralkyl steht, oder den pharmazeutisch unbedenklichen Säureadditionssalzen davon.Es ist bekannt, dass die Benzylalkoholderivate der Formel I oder deren pharmazeutisch unbedenkliche Säureadditionssalze als Cardiotonicum oder Antidiabeticum brauchbar sind (US-PS Nr.4032575).Aus der US-PS 4032575 ist bekannt, dass die Verbindung der Formel I hergestellt werden kann, indem man ein a-Halogen-monobenzyloxyacetophenon (A) mit 3,4-Dimethoxyphenyläthylamin (B) zu einem a-(3,4-Dimethoxyphenyl äthylamino)-monobenzyloxyacetophenon (C) kondensiert, gegebenenfalls die Verbindung (C) zu einem a-(3,4-Dimethoxyphenyläthylaminomethyl)-monobenzyloxybenzylalkohol (D) reduziert und dann die Verbindung (C) bzw. (D) der katalytischen Hydrierung unterwirft.Als Ergebnis weiterer Untersuchungen wurde nun aber ein neues Verfahren zur Herstellung von Benzylalkoholderivaten der Formel 1 gefunden.Erfindungsgemäss werden die Benzylalkoholderivate der Formel I hergestellt, indem man (a) ein Acetophenonderivat der Formel: EMI2.2 worin R2 für Wasserstoff, Alkanoyl oder Aralkyl steht, mit einem Thionylhalogenid umsetzt, (b) das resultierende Produkt mit 3,4-Dimethoxyphenyl äthylamin zu einem Thioacetamidderivat der Formel: EMI2.3 worin R2 die obige Bedeutung hat, umsetzt und (c-l) die Verbindung der Formel III der reduktiven Entschwefelung unterwirft oder (c-2) die Verbindung der Formel III zu einem Acetophenonderivat der Formel: **WARNUNG** Ende CLMS Feld konnte Anfang DESC uberlappen**.
